Annualization of woody perennials has the potential to revolutionize the breeding and production of fruit crops and rapidly improve horticultural species. Kiwifruit (Actinidia chinensis) is a recently domesticated fruit crop with a short history of breeding and tremendous potential for improvement. Previously, multiple kiwifruit CENTRORADIALIS (CEN)‐like genes have been identified as potential repressors of flowering. In this study, CRISPR/Cas9‐ mediated manipulation enabled functional analysis of kiwifruit CEN‐like genes AcCEN4 and AcCEN. Mutation of these genes transformed a climbing woody perennial, which develops axillary inflorescences after many years of juvenility, into a compact plant with rapid terminal flower and fruit development. The number of affected genes and alleles and severity of detected mutations correlated with the precocity and change in plant stature, suggesting that a bi‐allelic mutation of either AcCEN4 or AcCEN may be sufficient for early flowering, whereas mutations affecting both genes further contributed to precocity and enhanced the compact growth habit. CRISPR/Cas9‐mediated mutagenesis of AcCEN4 and AcCEN may be a valuable means to engineer Actinidia amenable for accelerated breeding, indoor farming and cultivation as an annual crop.  相似文献

Plants defend themselves against the attack of natural enemies by using an array of both constitutively expressed and induced defenses. Long-lived woody perennials are overrepresented among plant species that show strong induced defense responses, whereas annual plants and crop species are underrepresented. However, most studies of plant defense genes have been performed on annual or short-lived perennial weeds or crop species. Here I use molecular population genetic methods to survey six wound-inducible protease inhibitors (PIs) in a long-lived woody, perennial plant species, the European aspen (Populus tremula), to evaluate the likelihood of either recurrent selective sweeps or balancing selection maintaining amino acid polymorphisms in these genes. The results show that none of the six PI genes have reduced diversities at synonymous sites, as would be expected in the presence of recurrent selective sweeps. However, several genes show some evidence of nonneutral evolution such as enhanced linkage disequilibrium and a large number of high-frequency-derived mutations. A group of at least four Kunitz trypsin inhibitor genes appear to have experienced elevated levels of nonsynonymous substitutions, indicating allelic turnover on an evolutionary timescale. One gene, TI1, has enhanced levels of intraspecific polymorphism at nonsynonymous sites and also has an unusual haplotype structure characterized by two divergent haplotypes occurring at roughly equal frequencies in the sample. One haplotype has very low levels of intraallelic nucleotide diversity, whereas the other haplotype has levels of diversity comparable to other genes in P. tremula. Patterns of sequence diversity at TI1 do not fit a simple model of either balancing selection or recurrent selective sweeps. This suggests that selection at TI1 is more complex, possibly involving allelic cycling.  相似文献

ABSTRACT: BACKGROUND: The resistance of plants to pathogens relies on two lines of defense: a basal defense response and a pathogen-specific system, in which resistance (R) genes induce defense reactions after detection of pathogen-associated molecular patterns (PAMPS). In the specific system, a so-called arms race has developed in which the emergence of new races of a pathogen leads to the diversification of plant resistance genes to counteract the pathogens' effect. The mechanism of resistance gene diversification has been elucidated well for short-lived annual species, but data are mostly lacking for long-lived perennial and clonally propagated plants, such as roses. We analyzed the rose black spot resistance gene, Rdr1, in five members of the Rosaceae: Rosa multiflora, Rosa rugosa, Fragaria vesca (strawberry), Malus x domestica (apple) and Prunus persica (peach), and we present the deduced possible mechanism of R-gene diversification. RESULTS: We sequenced a 340.4-kb region from R. rugosa orthologous to the Rdr1 locus in R. multiflora. Apart from some deletions and rearrangements, the two loci display a high degree of synteny. Additionally, less pronounced synteny is found with an orthologous locus in strawberry but is absent in peach and apple, where genes from the Rdr1 locus are distributed on two different chromosomes. An analysis of 20 TIR-NBS-LRR (TNL) genes obtained from R. rugosa and R. multiflora revealed illegitimate recombination, gene conversion, unequal crossing over, indels, point mutations and transposable elements as mechanisms of diversification.A phylogenetic analysis of 53 complete TNL genes from the five Rosaceae species revealed that with the exception of some genes from apple and peach, most of the genes occur in species-specific clusters, indicating that recent TNL gene diversification began prior to the split of Rosa from Fragaria in the Rosoideae and peach from apple in the Spiraeoideae and continued after the split in individual species. Sequence similarity of up to 99% is obtained between two R. multiflora TNL paralogs, indicating a very recent duplication. CONCLUSIONS: The mechanisms by which TNL genes from perennial Rosaceae diversify are mainly similar to those from annual plant species. However, most TNL genes appear to be of recent origin, likely due to recent duplications, supporting the hypothesis that TNL genes in woody perennials are generally younger than those from annuals. This recent origin might facilitate the development of new resistance specificities, compensating for longer generation times in woody perennials.  相似文献

Host ecological traits may limit exposure to infectious disease, thereby generating the wide variation in disease incidence observed between host populations or species. The exclusion of disease by ecological traits may then allow selection to act against physiological defenses when they are costly to maintain in the absence of disease. This study investigates ecological resistance in the Silene-Microbotryum pathosystem. An estimated 80% of perennial Silene species host the anther-smut disease while no annuals harbor the disease in nature. Artificial inoculations of annual and perennial Silene plants, obtained from both natural and horticultural populations, demonstrate that the absence of disease in annuals is not explained by elevated physiological resistance. The annual habit is thus a powerful form of ecological defense against anther smut. Moreover, the higher susceptibility of annual species to anther smut relative to perennials supports the hypothesis of a loss of costly physiological resistance under ecological protection. The observation in annuals that physiological susceptibility is correlated with lower rates of flowering (i.e., lower fitness) suggests that variation in physiological resistance is costly in the absence of disease, even in a naїve Silene species. The absence of disease in natural populations of annuals combined with their high physiological susceptibility attest to the strength of host ecology in shaping the distribution of disease and to the dynamic nature of disease resistance.  相似文献

Specific leaf area (SLA) is a key trait to screen plants for ecological performance and productivity; however, the relationship between SLA and photosynthesis is not always up-scalable to growth when comparing multiple species with different life cycles. We explored leaf anatomy in annual and perennial species of Physaria, and related it to photosynthesis and water loss. The annual Physaria gracilis had higher SLA, thinner leaves, and lower investment in protective tissues, than perennial P. mendocina. Physaria angustifolia (annual), and P. pinetorum (perennial) showed intermediate values. Both perennials had a thicker palisade and high photosynthesis, relative to annuals. The larger leaf veins of perennials should allow high water availability to the mesophyll. The thicker palisade should determine high resistance to water flow and help explain their high water-use efficiency. These leaf functions reflect the construction of long-lived leaves that efficiently use resources under environmental limitations of arid environments.

Background  

Rates of molecular evolution vary widely among species. While significant deviations from molecular clock have been found in many taxa, effects of life histories on molecular evolution are not fully understood. In plants, annual/perennial life history traits have long been suspected to influence the evolutionary rates at the molecular level. To date, however, the number of genes investigated on this subject is limited and the conclusions are mixed. To evaluate the possible heterogeneity in evolutionary rates between annual and perennial plants at the genomic level, we investigated 85 nuclear housekeeping genes, 10 non-housekeeping families, and 34 chloroplast genes using the genomic data from model plants including Arabidopsis thaliana and Medicago truncatula for annuals and grape (Vitis vinifera) and popular (Populus trichocarpa) for perennials.  相似文献

Abstract An assessment of the impact of the soil-borne pathogenic fungus, Phytophthora cinnamomi (Oomycetes, Peronosporales), on the plant communities of the Stirling Range National Park was carried out between December 1988 and April 1989. A total of 541 plant species were collected, with the Proteaceae and the Myrtaceae the two largest families in the study region. Of the 330 species assessed for susceptibility to P. cinnamomi, 118 (36%) were recorded as having at least some individuals in a population judged to have been killed by the fungus and 33 (10%) were highly sensitive to the pathogen (more than 80% of plants in a population killed). Several families had large numbers of susceptible species, while others were apparently unaffected by the pathogen. Notably, 85% of proteaceous species assessed were rated as susceptible to P. cinnamomi. Proteaceous elements had a mean projective foliage cover of 40% in healthy plant communities, but had a mean cover of only 10% at sites that had a long history of infestation with the fungus. In contrast, some species with low levels of susceptibility to the pathogen, such as some monocotyledons, were found to be more abundant at old-infested sites than at healthy sites. Growth form may also influence susceptibility, with herbaceous perennials, annuals and geophytes assessed in this survey apparently unaffected by the fungus whereas 48% of woody perennials surveyed were susceptible. Changes in the floristic structure of plant communities may influence the composition of associated animal communities. In particular, vertebrate flower visitors may be vulnerable since 59% of the species with vertebrate-pollinated flowers were found to be susceptible to the pathogen. This research highlights the serious ecological impact of P. cinnamomi on native plant communities and suggests that significant components of the flora and associated fauna of the southwest of Western Australia are endangered by this virulent pathogen.  相似文献

Protein-protein interactions have emerged as key determinants of whether plant encounters with pathogens result in disease or successful plant defense. Genetic interactions between plant resistance genes and pathogen avirulence genes enable pathogen recognition by plants and activate plant defense. These gene-for-gene interactions in some cases have been shown to involve direct interactions of the products of the genes, and have indicated plant intracellular localization for certain avirulence proteins. Incomplete specificity of some of the interactions in laboratory assays suggests that additional proteins might be required to confer specificity in the plant. In many cases, resistance and avirulence protein interactions have not been demonstrable, and in some cases, other plant components that interact with avirulence proteins have been found. Investigation to date has relied heavily on biochemical and cytological methods including in vitrobinding assays and immunoprecipitation, as well as genetic tools such as the yeast two-hybrid system. Observations so far, however, point to the likely requirement for multiple, interdependent protein associations in pathogen recognition, for which these techniques can be insufficient. This article reviews the protein-protein interactions that have been described in pathogen recognition by plants, and provides examples of how rapid future progress will hinge on the adoption of new and developing technologies.  相似文献

Plants are under continuous threat of infection by pathogens endowed with diverse strategies to colonize their host. Comprehensive biochemical and genetic approaches are now starting to reveal the complex signaling pathways that mediate plant disease resistance. Initiation of defense signaling often involves specific recognition of invading pathogens by the products of specialized host resistance (R) genes. Potential resistance signaling components have been identified by mutational analyses to be required for specific resistance in the model Arabidopsis and some crop species. Strikingly, many of the components share similarity to that of innate immune systems in animals. Evidence is also accumulating that plant pathogens have a number of ways to evade host defenses during the early stages of infection, similar to animal pathogens. These strategies are becoming much better understood in a number of plant–pathogen interactions. In this review, we focus on the current knowledge of host factors that control plant resistance and susceptibility to fungal pathogens. The knowledge accumulated in these studies will serve a fundamental basis for combating diseases in strategic molecular agriculture.  相似文献

We studied the ecological characteristics of the plants of Italian cultivated land that cause allergies in sensitized individuals. Differences between the allergenic flora of annual and perennial cultivations were found. Prevalence of annual therophytes was found in seasonal and annual crops subjected to soil tillage and to strong human disturbance. Plants growing in stable, moderately disturbed cultivations (citrus and olive groves, vineyards) were more often perennials. Allergenic species dispersed by wind and those endowed with multiple dispersal strategies (polychory) were well represented in seasonal and annual crops. The moderate height of these cultivations facilitates circulation of air, favouring the diffusion of anemochoric plants, which explains the abundance of the Poaceae and the Asteraceae families. The spreading of some allochthonous invasive allergenic species was ascertained. The entrance of allergenic plants from surrounding natural ecosystems increases the overall allergenicity of Italian cultivations. The results obtained pointed out a strong relationship between allergenic flora, human impact, and the structure of the cultivations. They point out the importance of knowledge about the ecological characteristics of the allergenic flora infesting the cultivations, which enables efficacious control of the diffusion of the most dangerous species.  相似文献

Abstract. This paper reports on vegetation development on permanent experimental plots during five years of succession. Nine (1 m2) plots were filled with three typical substrates from man-made habitats of urban and industrial areas in the region of Berlin. The three substrates (a commercial ‘topsoil’, a ruderal ‘landfill’ soil and a sandy soil), differ in organic matter and nutrient contents. Relevés of species composition and percent cover of each species present were made monthly during the growing season from the start of vegetation development. This paper describes the different successional pathways on topsoil and ruderal soil and the colonization process on sandy soil. On topsoil, ruderal annuals are dominant in the first year and are replaced by short-lived perennials from the second year. Those species were replaced by long-lived perennial herbs (Ballota nigra or Urtica dioica) from the third year of succession onwards. On the ruderal land-fill soil the early successional stages are less sharp and the perennial Solidago canadensis is able to dominate within one year after the succession was initiated. On sandy soil there is still an ongoing colonization process, where pioneer tree species like Betula pendula and Populus nigra play a main role. The importance of ‘initial floristic composition’, the role of substrate for community structure and the peculiarities of successional sequences on anthropogenic soils in the context of primary and secondary successions are discussed.  相似文献

Soil-borne fungal pathogen, Fusarium oxysporum causes major economic losses by inducing necrosis and wilting symptoms in many crop plants. Management of fusarium wilt is achieved mainly by the use of chemical fungicides which affect the soil health and their efficiency is often limited by pathogenic variability. Hence understanding the nature of interaction between pathogen and host may help to select and improve better cultivars. Current research evidences highlight the role of oxidative burst and antioxidant enzymes indicating that ROS act as an important signaling molecule in banana defense response against Fusarium oxysporum f.sp. cubense. The role of jasmonic acid signaling in plant defense against necrotrophic pathogens is well recognized. But recent studies show that the role of salicylic acid is complex and ambiguous against necrotrophic pathogens like Fusarium oxysporum, leading to many intriguing questions about its relationship between other signaling compounds. In case of banana, a major challenge is to identify specific receptors for effector proteins like SIX proteins and also the components of various signal transduction pathways. Significant progress has been made to uncover the role of defense genes but is limited to only model plants such as Arabidopsis and tomato. Keeping this in view, we review the host response, pathogen diversity, current understanding of biochemical and molecular changes that occur during host and pathogen interaction. Developing resistant cultivars through mutation, breeding, transgenic and cisgenic approaches have been discussed. This would help us to understand host defenses against Fusarium oxysporum and to formulate strategies to develop tolerant cultivars.  相似文献

Prolamin and resistance gene families are important in wheat food use and in defense against pathogen attacks, respectively. To better understand the evolution of these multi‐gene families, the DNA sequence of a 2.8‐Mb genomic region, representing an 8.8 cM genetic interval and harboring multiple prolamin and resistance‐like gene families, was analyzed in the diploid grass Aegilops tauschii, the D‐genome donor of bread wheat. Comparison with orthologous regions from rice, Brachypodium, and sorghum showed that the Ae. tauschii region has undergone dramatic changes; it has acquired more than 80 non‐syntenic genes and only 13 ancestral genes are shared among these grass species. These non‐syntenic genes, including prolamin and resistance‐like genes, originated from various genomic regions and likely moved to their present locations via sequence evolution processes involving gene duplication and translocation. Local duplication of non‐syntenic genes contributed significantly to the expansion of gene families. Our analysis indicates that the insertion of prolamin‐related genes occurred prior to the separation of the Brachypodieae and Triticeae lineages. Unlike in Brachypodium, inserted prolamin genes have rapidly evolved and expanded to encode different classes of major seed storage proteins in Triticeae species. Phylogenetic analyses also showed that the multiple insertions of resistance‐like genes and subsequent differential expansion of each R gene family. The high frequency of non‐syntenic genes and rapid local gene evolution correlate with the high recombination rate in the 2.8‐Mb region with nine‐fold higher than the genome‐wide average. Our results demonstrate complex evolutionary dynamics in this agronomically important region of Triticeae species.  相似文献

Species with close associations to a specific host species, such as parasites and phytophages, make immense contributions to biodiversity. Hence, factors determining the variation in species richness among hosts are a main focus of ecological research. Investigations of determining factors of fungivorous species among host species are still scarce. Based on ecological patterns of parasites and phytophages, we hypothesized that the species richness of tree‐fungus beetles of the family Ciidae (Coleoptera) would increase with increasing basidiome size, niche diversity of the growth form, durability, increasing abundance and decreasing phylogenetic isolation of the host fungus. Our generalized least‐squares model, controlled by host phylogeny, revealed that Ciidae species richness increases with host abundance, but decreases with host phylogenetic isolation. In contrast with our prediction, Ciidae species richness was higher in annual basidiomes than in perennials. Pileate basidiomes revealed higher species richness than resupinate and stipitate basidiomes, which may be interpreted as being a result of their higher host niche diversity. The importance of host abundance, measured on the landscape scale, corroborates that fungivore species richness among macrofungal hosts is determined by factors similar to those that determine parasite and phytophage species richness among their hosts. © 2015 The Linnean Society of London, Biological Journal of the Linnean Society, 2015, 114 , 699–708.  相似文献

For a species to be able to respond to environmental change, it must either succeed in following its optimal environmental conditions or in persisting under suboptimal conditions, but we know very little about what controls these capacities. We parameterized species distribution models (SDMs) for 135 plant species from the Algerian steppes. We interpreted low false‐positive rates as reflecting a high capacity to follow optimal environmental conditions and high false‐negative rates as a high capacity to persist under suboptimal environmental conditions. We also measured functional traits in the field and built a unique plant trait database for the North‐African steppe. For both perennial and annual species, we explored how these two capacities can be explained by species traits and whether relevant trait values reflect species strategies or biases in SDMs. We found low false‐positive rates in species with small seeds, flowers attracting specialist pollinators, and specialized distributions (among annuals and perennials), low root:shoot ratios, wide root‐systems, and large leaves (perennials only) (R2 = .52–58). We found high false‐negative rates in species with marginal environmental distribution (among annuals and perennials), small seeds, relatively deep roots, and specialized distributions (annuals) or large leaves, wide root‐systems, and monocarpic life cycle (perennials) (R2 = .38 for annuals and 0.65 for perennials). Overall, relevant traits are rarely indicative of the possible biases of SDMs, but rather reflect the species' reproductive strategy, dispersal ability, stress tolerance, and pollination strategies. Our results suggest that wide undirected dispersal in annual species and efficient resource acquisition in perennial species favor both capacities, whereas short life spans in perennial species favor persistence in suboptimal environmental conditions and flowers attracting specialist pollinators in perennial and annual species favor following optimal environmental conditions. Species that neither follow nor persist will be at risk under future environmental change.  相似文献

Fire is an integral ecological factor in African savanna ecosystems, but its effects on savanna productivity are highly variable and less understood. We conducted a field experiment to quantify changes in herbaceous phytomass and nutrient composition in a Sudanian savanna woodland subjected to annual early fire from 1993 to 2004. Fire effects were also assessed on two perennial and two annual grass species during the following growing season. Early fire significantly reduced above‐ground phytomass of the studied species (P = 0.03), their crude protein (P = 0.022), neutral detergent insoluble crude protein (P = 0.016) and concentrations of Ca, Fe and Mn (P < 0.05). Perennial grasses had higher above‐ground phytomass but lower total crude protein and fat than annual grasses. Nonstructural carbohydrates tended to be higher for annuals, while fibre and lignin contents were high for perennials. Except Na and Fe, the concentration of mineral elements varied between species. Fire did not affect measures of digestibility and metabolizable energy, but its effect differed significantly among species. In conclusion, the results illustrate that long‐term frequent fire will counterbalance the short‐term increase in soil fertility and plant nutrient concentrations claimed to be accrued from single or less frequent fire.  相似文献
