古代岩画和壁画是人类文明发展历史进程的重要记录,具有极高的历史价值、艺术价值及科学价值,但他们无时无刻不受到环境因素引起的物理、化学及生物的劣化作用。近年,有关微生物对古代岩/壁画的危害逐渐受到关注。阐述了世界一些著名的岩/壁画文物的微生物的研究进展,如法国拉斯科洞穴岩画(Lascaux Cave)、西班牙阿尔塔米拉洞穴岩画(Altamira Cave)、敦煌石窟壁画、嘉峪关魏晋墓壁画以及我国东北地区公元5世纪的墓葬壁画等。通过对以上所述岩画和壁画微生物群落的分析比较,发现主要的细菌群落是变形菌门和放线菌门,放线菌门以假诺卡氏菌为主。真菌群落主要以虫生真菌为主。进一步分析了岩画和壁画微生物病害的共性和区别,揭示了洞穴、墓室等特殊环境下的微生物群落特点,为防治古代岩/壁画微生物病害提供了以下借鉴。  相似文献

Advances in microscopic analysis and molecular genetics research methods promoted the acquisition of evidence that natural bacteria populations exist predominately as substrate attached biofilms. Bacteria in biofilms are able to exchange signals and display coordinated activity that is inherent to multicellular organisms. Formation of biofilm communities turned out to be one of the main survival strategies of bacteria in their ecological niche. Bacteria in attached condition in biofilm are protected from the environmental damaging factors and effects of antibacterial substances in the environment and host organism during infection. According to contemporary conception, biofilm is a continuous layer of bacterial cells that are attached to a surface and each other, and contained in a biopolymer matrix. Such bacterial communities may be composed of bacteria of one or several species, and composed of actively functioning cells as well as latent and uncultured forms. Particular attention has recently been paid to the role of biofilms in the environment and host organism. Microorganisms form biofilm on any biotic and abiotic surfaces which creates serious problems in medicine and various areas of economic activity. Currently, it is established that biofilms are one of the pathogenetic factors of chronic inflection process formation. The review presents data on ubiquity of bacteria existence as biofilms, contemporary methods of microbial community analysis, structural-functional features of bacterial biofilms. Particular attention is paid to the role of biofilm in chronic infection process formation, heightened resistance to antibiotics of bacteria in biofilms and possible mechanisms of resistance. Screening approaches for agents against biofilms in chronic infections are discussed.  相似文献

麋鹿的采食、躺卧和践踏行为均会对栖息地土壤环境造成影响,进而影响土壤微生物群落结构。利用高通量测序技术,分析江苏大丰麋鹿国家级自然保护区禁牧点和补饲点土壤细菌和真菌群落结构差异,并结合土壤理化性质探究禁牧对土壤微生物群落结构的影响。结果表明细菌群落的优势菌门为变形菌门,真菌群落的优势菌门为子囊菌门。禁牧改变了土壤微生物群落结构,在门水平上提高了变形菌门、放线菌门和担子菌门的相对丰度,降低了绿弯菌门、厚壁菌门和子囊菌门的相对丰度,禁牧点与补饲点土壤微生物群落多样性的相似性较低。冗余分析中,细菌受土壤环境因子的影响大于真菌,其中土壤pH是影响细菌和真菌群落最大的土壤环境因子。研究揭示了禁牧对土壤微生物群落结构的影响,为保护区制定麋鹿生境恢复方案提供参考。  相似文献

Protistan grazing on biofilms is potentially an important conduit enabling energy flow between microbial trophic levels. Contrary to the widely held assumption that protistan feeding primarily involves ingestion of biofilm cells, with negative consequences for the biofilm, this study demonstrated preferential grazing on the noncellular biofilm matrix by a ciliate, with selective ingestion of yeast and bacterial cells of planktonic origin over attached and biofilm-derived planktonic cells. Introducing a ciliate to two biofilm-forming Cryptococcus species, as well as two bacterial species in a model biofilm system, fluorescent probes were applied to determine ingestion of cellular and noncellular biofilm fractions. Fluoromicroscopy, as well as photometric quantification, confirmed that protistan grazing enhanced yeast biofilm metabolism, and an increase in biofilm biomass and viability. We propose that the extracellular polymeric matrix of biofilms may act as an interface regulating interaction between predator and prey, while serving as source of nutrients and energy for protists.  相似文献

Background

Despite continued preventive efforts, dental caries remains the most common disease of man. Organic acids produced by microorganisms in dental plaque play a crucial role for the development of carious lesions. During early stages of the pathogenetic process, repeated pH drops induce changes in microbial composition and favour the establishment of an increasingly acidogenic and aciduric microflora. The complex structure of dental biofilms, allowing for a multitude of different ecological environments in close proximity, remains largely unexplored. In this study, we designed a laboratory biofilm model that mimics the bacterial community present during early acidogenic stages of the caries process. We then performed a time-resolved microscopic analysis of the extracellular pH landscape at the interface between bacterial biofilm and underlying substrate.

Methodology/Principal Findings

Strains of Streptococcus oralis, Streptococcus sanguinis, Streptococcus mitis, Streptococcus downei and Actinomyces naeslundii were employed in the model. Biofilms were grown in flow channels that allowed for direct microscopic analysis of the biofilms in situ. The architecture and composition of the biofilms were analysed using fluorescence in situ hybridization and confocal laser scanning microscopy. Both biofilm structure and composition were highly reproducible and showed similarity to in-vivo-grown dental plaque. We employed the pH-sensitive ratiometric probe C-SNARF-4 to perform real-time microscopic analyses of the biofilm pH in response to salivary solutions containing glucose. Anaerobic glycolysis in the model biofilms created a mildly acidic environment. Decrease in pH in different areas of the biofilms varied, and distinct extracellular pH-microenvironments were conserved over several hours.

Conclusions/Significance

The designed biofilm model represents a promising tool to determine the effect of potential therapeutic agents on biofilm growth, composition and extracellular pH. Ratiometric pH analysis using C-SNARF-4 gives detailed insight into the pH landscape of living biofilms and contributes to our general understanding of metabolic processes in in-vivo-grown bacterial biofilms.  相似文献

Understanding the evolution of microbial diversity is an important and current problem in evolutionary ecology. In this paper, we investigated the role of two established biochemical trade-offs in microbial diversification using a model that connects ecological and evolutionary processes with fundamental aspects of biochemistry. The trade-offs that we investigated are as follows:(1) a trade-off between the rate and affinity of substrate transport; and (2) a trade-off between the rate and yield of ATP production. Our model shows that these biochemical trade-offs can drive evolutionary diversification under the simplest possible ecological conditions: a homogeneous environment containing a single limiting resource. We argue that the results of a number of microbial selection experiments are consistent with the predictions of our model.  相似文献

Microbial biofilms on surface of sandstone is detrimental to the integrity of the substratum material and they are biodeteriogens responsible for the damage of sandstone over time. We observed that fungi formed extensive biofilms on areas previously colonized by autotrophic and heterotrophic microbial biofilms causing darkening of the stone surface. Appearance of fungi on these biofilms has resulted in removal of the preformed biofilm through extensive examination of sandstone surfaces in Angkor Thom, Cambodia. One fungus, isolated from the surface with capability of removing biofilms, was purified and identified as Aspergillus allahabadii during our survey and sampling of microbial biofilms at Bayon temple, Angkor Thom, Cambodia in 2008. Ribosomal RNA (ITS and 5.8S) and β-tubulin gene sequences were phylogenetically analyzed to confirm the taxonomy of this strain. In addition, its protein profile and enzyme assays were also carried out and β-galactosidase was the highest among 7 enzymes tested. Our results suggest that fungi may have an important role in removing microbial biofilms on surfaces of stone and potential mechanisms and applications are discussed.  相似文献

Interactions among endophytic bacteria and fungi: Effects and potentials   总被引:5,自引:0,他引:5  
Plants benefit extensively by harbouring endophytic microbes. They promote plant growth and confer enhanced resistance to various pathogens. However, the way the interactions among endophytes influence the plant productivity has not been explained. Present study experimentally showed that endophytes isolated from rice (Oryza sativa) used as the test plant produced two types of interactions; biofilms (bacteria attached to mycelia) and mixed cultures with no such attachments. Acidity, as measured by pH in cultures with biofilms was higher than that of fungi alone, bacteria alone or the mixed cultures. Production of indoleacetic acid like substances (IAAS) of biofilms was higher than that of mixed cultures, fungi or bacteria. Bacteria and fungi produced higher quantities of IAAS than mixed cultures. In mixed cultures, the potential of IAAS production of resident microbes was reduced considerably. There was a negative relationship between IAAS and pH of the biofilms, indicating that IAAS was the main contributor to the acidity. However, such a relationship was not observed in mixed cultures. Microbial acid production is important for suppressing plant pathogens. Thus the biofilm formation in endophytic environment seems to be very important for healthy and improved plant growth. However, it is unlikely that an interaction among endophytes takes place naturally in the endophytic environment, due to physical barriers of plant tissues. Further, critical cell density dependant quorum sensing that leads to biofilm formation may not occur in the endophytic environment as there is a limited space. As suchin vitro production and application of beneficial biofilmed inocula of endophytes are important for improved plant production in any agro-ecosystem. The conventional practice of plant inoculation with monocultures or mixed cultures of effective microbes may not give the highest microbial effect, which may only be achieved by biofilm formation.  相似文献

It is generally predicted that grazers enhance soil microbial activity and nutrient availability and promote soil bacteria in fertile ecosystems, but retard microbial activity and nutrient availability and promote soil fungi in infertile ecosystems. We tested these predictions in tundra by comparing grazing effects between fertile and infertile habitats and with/without nutrient manipulation by fertilization. Grazing decreased soil N content in fertile and in fertilized plots in infertile habitats while increased it in infertile tundra habitats, which directly opposed our prediction. We conclude that this unpredicted outcome probably resulted from nutrient transport between habitats. Also contrasting with our hypothesis, grazing increased fungal rather than bacterial abundance in fertilized plots at both habitats. In support with predictions, grazing increased microbial activity for soil C decomposition in fertile but decreased it in infertile habitats. The effect of grazing on soil C decomposition followed same patterns as grazer‐induced changes in the activity of β‐glucosidase, which is an extracellular enzyme synthesized by soil microorganisms for degrading soil cellulose. We suggest that the theoretical framework on grazer–soil interactions should incorporate microbial potential for extracellular enzyme production (‘microscale’ grazer effects) and nutrient translocation by grazers among habitats (‘macroscale’ grazer effects) as important mechanisms by which grazers influence soil processes and nutrient availability for plants at contrasting levels of habitat fertility.  相似文献

A primary goal of ecological restoration is often to return processes and functions to degraded ecosystems. Soil, while often ignored in restoration, supports diverse communities of organisms and is a fundamental actor in providing ecosystem processes and services. We investigated the impact of seeding and livestock grazing on plant communities, soil microorganisms, and soil fertility 3 years after the restoration of a disturbed pipeline corridor in southeastern Arizona. The initial soil disturbance and topsoil treatment, regardless of seeding or grazing, was the most influential factor in determining differences in both plant and microbial communities. Compared with the control, the disturbed and restored sites had greater plant species richness, greater total herbaceous plant cover, greater soil organic matter, higher pH, and differed in soil nutrients. Bacteria and fungi appeared to generally correlate with micro‐environment and soil physiochemical properties rather than specific plant species. The undisturbed control had a smaller proportion of bacterial functional groups associated with the breakdown of plant biomass (polysaccharide decomposition) and a smaller proportion of arbuscular mycorrhizal fungi (AMF) compared with disturbed and restored sites. The ability of the unseeded disturbed site to recover robust vegetation may be due in part to the high presence of AMF. These differences show selection for soil microorganisms that thrive in disturbed and restored sites and may contribute to increased plant productivity. Restoration of specific plant species or ecological processes and services would both benefit from better understanding of the impacts of disturbance on soil microorganisms and soil fertility.  相似文献

Device-related infections are often associated with biofilms (microbial communities encased within polysaccharide-rich extracellular matrix) formed by pathogens on surfaces of these devices. Candida species are the most common fungi isolated from infections associated with catheters and dentures, and both Candida and Fusarium are commonly isolated from contact lens–related infections such as fungal keratitis. These biofilms exhibit decreased susceptibility to most antimicrobial agents, which contributes to the persistence of infection. Drug resistance in fungal biofilms is multifactorial and phase-dependent; for example, efflux pumps mediate resistance in biofilms during early phase, whereas altered membrane sterol composition contributes to resistance in mature phase. Both substrate type and surface coatings play an important role in the pathogenesis of device-related fungal biofilms. Host immune cells influence the ability of Candida to form biofilms in vitro. This review summarizes recent advances in research on fungal biofilms and discusses their clinical relevance.  相似文献

Biofilms are interface micro-habitats formed by microbes that differ markedly from those of the ambient environment. The term 'subaerial biofilm' (SAB) was coined for microbial communities that develop on solid mineral surfaces exposed to the atmosphere. Subaerial biofilms are ubiquitous, self-sufficient, miniature microbial ecosystems that are found on buildings, bare rocks in deserts, mountains, and at all latitudes where direct contact with the atmosphere and solar radiation occurs. Subaerial biofilms on exposed terrestrial surfaces are characterized by patchy growth that is dominated by associations of fungi, algae, cyanobacteria and heterotrophic bacteria. Inherent subaerial settlers include specialized actinobacteria (e.g. Geodermatophilus), cyanobacteria and microcolonial fungi. Individuals within SAB communities avoid sexual reproduction, but cooperate extensively with one another especially to avoid loss of energy and nutrients. Subaerial biofilm metabolic activity centres on retention of water, protecting the cells from fluctuating environmental conditions and solar radiation as well as prolonging their vegetative life. Atmospheric aerosols, gases and propagatory particles serve as sources of nutrients and inoculum for these open communities. Subaerial biofilms induce chemical and physical changes to rock materials, and they penetrate the mineral substrate contributing to rock and mineral decay, which manifests itself as bio-weathering of rock surfaces. Given their characteristic slow and sensitive growth, SAB may also serve as bioindicators of atmospheric and/or climate change.  相似文献

Bacterial biofilms are complex microbial depositions on immersed interfaces that form wherever the environmental conditions sustain microbial growth. Despite their name, biofilms can develop in highly irregular structures. Recently several mathematical concepts have been introduced to model these spatially structured microbial populations. Regardless of the type of model, they all have, even for microbially relatively simple systems, many parameters which generally are known at most approximately. We investigate the effect of uncertainties in model parameters on four morphological and four ecological output parameters using a nonlinear diffusion model for a biofilm in which two species compete for a shared nutrient. To this end we conduct an extensive computer simulation experiment for two different levels of data uncertainty, three different hydrodynamic conditions, and two different scenarios of bulk substrate availability. Our results indicate that input model parameter uncertainties have a much larger effect on ecological than on morphological output parameters.  相似文献

Bacteria and fungi are of uttermost importance in determining environmental and host functioning. Despite close interactions between animals, plants, their associated microbiomes, and the environment they inhabit, the distribution and role of bacteria and especially fungi across host and environments as well as the cross-habitat determinants of their community compositions remain little investigated. Using a uniquely broad global dataset of 13 483 metagenomes, we analysed the microbiome structure and function of 25 host-associated and environmental habitats, focusing on potential interactions between bacteria and fungi. We found that the metagenomic relative abundance ratio of bacteria-to-fungi is a distinctive microbial feature of habitats. Compared with fungi, the cross-habitat distribution pattern of bacteria was more strongly driven by habitat type. Fungal diversity was depleted in host-associated communities compared with those in the environment, particularly terrestrial habitats, whereas this diversity pattern was less pronounced for bacteria. The relative gene functional potential of bacteria or fungi reflected their diversity patterns and appeared to depend on a balance between substrate availability and biotic interactions. Alongside helping to identify hotspots and sources of microbial diversity, our study provides support for differences in assembly patterns and processes between bacterial and fungal communities across different habitats.  相似文献

Freshwater wetlands constitute important ecosystems, and their benthic, attached microbial communities, including biofilms, represent key habitats that contribute to primary productivity, nutrient cycling, and substrate stabilization. In many wetland biofilms, algae constitute significant parts of the microbial population, yet little is known about their activities in these communities. An analysis of wetland biofilms from the Adirondack region of New York (USA) was performed with special emphasis on desmids, a group of evolutionarily advanced green algae commonly found in these habitats. Desmids constituted as much as 23.7% of the total algal and cyanobacterial flora of the biofilms during the July and August study periods. These algae represented some of the first eukaryotes to colonize new substrates, and during July their numbers correlated with fluctuations in general biofilm parameters such as biofilm thickness and dry weight as well as total carbohydrate. Significant numbers of bacteria were associated with both the EPS sheaths and cell wall surfaces of the desmids. Colonization of new substrates and development of biofilms were rapid and were followed by various fluctuations in microbial community structure over the short- and long-term observations. In addition to desmids, diatoms, filamentous green algae and transient non-motile phases of flagellates represented the photosynthetic eukaryotes of these biofilms.  相似文献

Microbial biogeography studies expend much effort in determining whether environmental selection or stochastic processes related to dispersal are more important in shaping community composition. While both types of factors are possibly influential, it is tacitly assumed that protists, or microbial eukaryotes in general, behave biogeographically as prokaryotes because of their small physical size. However, direct evidence for this in exactly the same environment and at the same phylogenetic depth is lacking. In this study, we compared the structure of both prokaryotic and eukaryotic components of microbial communities forming biofilms on mineral substrates in different geographic locations at the level of small-subunit (SSU) rRNA-based operational taxonomic units (OTUs). These microbial communities are subjected to strong environmental selection and contain significant proportions of extremophilic microorganisms adapted to desiccation and UV radiation. We find that the nature of the substrate as well as climatic variables and geography influences microbial community structure. However, constrained correspondence analyses and distance-decay curves showed that, whereas the substrate type was the most significant factor structuring bacterial communities, geographic location was the most influential factor for microbial eukaryote communities. Biological explanations implying a higher dispersal success for bacteria combined with more mobile lifestyles for predatory protists may underlie these different prokaryote versus microbial eukaryote biogeographic patterns.  相似文献

Biofilm formation is controlled by an array of coupled physical, chemical, and biotic processes. Despite the ecological relevance of microbial biofilms, their community formation and succession remain poorly understood. We investigated the effect of flow velocity, as the major physical force in stream ecosystems, on biofilm community succession (as continuous shifts in community composition) in microcosms under laminar, intermediate, and turbulent flow. Flow clearly shaped the development of biofilm architecture and community composition, as revealed by microscopic investigation, denaturing gradient gel electrophoresis (DGGE) analysis, and sequencing. While biofilm growth patterns were undirected under laminar flow, they were clearly directed into ridges and conspicuous streamers under turbulent flow. A total of 51 biofilm DGGE bands were detected; the average number ranged from 13 to 16. Successional trajectories diverged from an initial community that was common in all flow treatments and increasingly converged as biofilms matured. We suggest that this developmental pattern was primarily driven by algae, which, as “ecosystem engineers,” modulate their microenvironment to create similar architectures and flow conditions in all treatments and thereby reduce the physical effect of flow on biofilms. Our results thus suggest a shift from a predominantly physical control to coupled biophysical controls on bacterial community succession in stream biofilms.  相似文献

Phenol oxidase from blood as well as from soft cuticle of different crayfish was highly activated by cell walls of a number of fungi and other plants, but the enzymes from other crustaceans tested and from some insects were not. Attachment of the phenol oxidase on blood and soft cuticle to the cell wall surface of certain plants and subsequent heavy melanization of the walls with natural or added substrate may be a unique property of crayfish since it did not occur in any other arthropods tested. Interestingly, blood and cuticle of crayfish showed the same specificity in these processes and the mechanisms might therefore be similar, if not the same, in both. The activity of the nonadsorbed enzyme dispersed in the blood was higher in a crayfish species resistant to fungal attack than in susceptible ones, but the relative degree of activation by adding cell walls was greater in the latter.The natural substrate for the phenol-oxidase activity in crayfish blood was released or mobilized by the blood cells. The substrate for the activity in cuticle was not available in the intact cuticle itself. It appeared only after a “signal” had been transmitted through the cuticle to the cells inside. The possibility that these mechanisms are involved in the cuticular as well as the blood defense system against fungal and other microbial parasites is discussed.  相似文献
