Our objective was to understand the influence of dietary gluconeogenic amino acids on hepatic glucose metabolism in rainbow trout (Oncorhynchus mykiss). We analyzed the effects of partial substitution of dietary protein by a single gluconeogenic dispensable amino acid (DAA: alanine, aspartic acid or glutamic acid), on the regulation of hepatic glycolytic and gluconeogenic enzymes. We fed juvenile rainbow trout with isonitrogenous and isoenergetic diets in which part of nitrogen from fishmeal was replaced by nitrogen from one of the three DAA. Fish were fed over 9 weeks and samples withdrawn 6 h after feeding or 5 days after food deprivation. Our data did not show a clear effect of an excess of DAA on activities of glycolytic enzymes (glucokinase and pyruvate kinase) compared to the control diet. In contrast, feeding caused a significant repression of gluconeogenic enzyme activities (glucose-6-phosphatase, fructose-1,6-bisphosphatase and mitochondrial phosphoenolpyruvate carboxykinase) only in fish fed the three DAA substituted diets. However, these differences were insufficient to affect postprandial glycemia significantly. In conclusion, an excess of dietary DAA tested does not seem to modify glycemia or to have a negative impact on dietary carbohydrate utilization in rainbow trout.  相似文献

Eighteen groups of 40 juvenile tilapia (Oreochromis niloticus) were fed five experimental diets in which crude protein (30% dry matter) was replaced at a level of 25% by dispensable amino acids (DAA) in the free form, given singly (alanine, glutamic acid or glycine) or as a mixture and at a level of 50% by DAA mixture. At the 25% substitution level, irrespective of the nature of the DAA, growth was slightly reduced (10%), while glycaemia, nitrogen retention and excretion were unaffected. A 50% substitution with the DAA mixture resulted in a marked reduction in growth (50%), a modified pattern of glycaemia and a lower nitrogen retention associated with a higher nitrogen excretion. Results suggest that DAA should not account for more than 60% of the total amino acid supply in tilapia diets.  相似文献

Summary Kittens fed diets containing 0.75 × the NRC (1986) essential amino acid requirement (EAArq) and 210 to 560g crude protein(CP)/kg diet exhibited, with increasing CP: 1) decreasing weight gain, 2) decreasing plasma arginine concentrations, 3) increasing urinary orotic acid excretion, 4) increasing plasma glutamic acid concentrations, and 5) plasma isoleucine concentrations at levels that suggest a marginal isoleucine deficiency. Kittens fed a control diet (CD) containing 1.5 × EAArq and 350 g CP/kg diet had maximal weight gains and no orotic aciduria. It was concluded that the decreased weight gain and adverse metabolic effects were caused by arginine deficiency and possibly glutamic acid toxicity induced by high dietary dispensable amino acids. Kittens fed the diets containing 1.0 × EAArq and 350 and 560 g CP/kg diet had depressed plasma arginine and elevated glutamic acid concentrations and orotic aciduria. These results indicate that 10 g arg/kg diet is not adequate at CP concentrations above 280 g/kg and the calculated requirement of arginine is (0.02 g arginine/g CP) × (Y g CP/kg diet) + (4.0 g arginine/kg diet) where Y is the dietary CP level.Abbreviations CD control diet - CP crude protein (g CP/kg diet = g nitrogen/kg diet × 6.25) - DAA dispensable amino acids - EAA essential amino acids - EAArq essential amino acid requirement  相似文献

When the amino-acid mixture of an effective chemically defined diet was replaced by single amino acids, keeping the total nitrogen at the same level, the egg production of Dacus oleae was minimal with all the 19 amino acids tested. Male survival was adversely affected by the amino acids : alanine, aspartic acid, glutamic acid, glycine, hydroxyproline, lysine, serine and tyrosine, while female survival was shortened when the amino acids : glycine, hydroxyproline and lysine were added. The creation of amino-acid imbalances, by deleting the 19 amino acids individually, from the complete amino-acid mixture, showed that the amino acids : arginine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, serine, threonine, tryptophane and valine were indispensable to the adult Dacus oleae flies, as far as egg production is concerned. Survival of the male flies was significantly shortened when the amino acids : alanine, hydroxyproline and tryptophane were omitted. Significant differences in longevity between males and females were scored, when the amino acids : alanine, aspartic acid, cystine, glycine and tryptophane, were omitted.  相似文献

Chicks were fed on the purified diets of which amino acid pattern was modeled after whole egg protein and crude protein content was 21.1%, changing the dietary ratio of indispensable amino acid nitrogen to dispensable amino acid nitrogen (I/D ratio) from 1/1.5 to 3/1 at regular intervals. The balances among amino acids in each indispensable and dispensable group of test diets were kept the same pattern as that of the whole egg, respectively. Optimum I/D ratio for normal chick growth was estimated to be in the range of between 1/1 and 1.5/1, because feed efficiency was the highest at the I/D ratio 1/1 and growth rate was the highest at the I/D ratio 1.5/1.

Chicks were killed and the serum was collected at the end of the experiment. It was shown that the I/D ratios of free amino acid in the serum of chicks were strongly influenced by that of diet.

White Leghorn chicks fed on the Scott’s reference amino acid diet grew as well as those fed on a conventional chick starter. Nitrogen retention of the former was a little less than that of the latter, but the amount of carcass fat of the former was almost twice as much as the latter.

Growth rate of chick was considerably reduced, when glutamic acid which is the only dispensable amino acid in the Scott’s diet was replaced by a mixture of glutamic acid, aspartic acid, alanine and serine, nitrogen content being kept at constant. Sufficient amount of glutamic acid in the Scott’s diet seems to be essential for the maximum growth of chick.  相似文献

The intention of this experiment was to assess the effects of different sources of dietary lipid on the fatty acid composition of the fillet and liver and the flesh quality traits of rainbow trout (Oncorhynchus mykiss) after a 70‐day feeding period. Four iso‐nitrogenous (approx. 51% crude protein) and iso‐lipidic (approx. 14% crude lipid) experimental diets were formulated. The control diet contained only fish oil (FO) as the primary lipid source. In the other three dietary treatments, fish oil was replaced by 100% (LO30/SO35/SFO35) and 70% (FO30/LO35/SO35 or FO30/SO35/SFO35) sesame oil (SO), linseed oil (LO), or sunflower oil (SFO). Triplicate groups of 40 rainbow trout (~46 g) held under similar culture conditions were hand‐fed daily to apparent satiation for 70 days. At the end of the feeding trials, no difference in growth performance among experimental groups was noted (P > 0.05). There were some differences in the proximate composition of fish fillets (P < 0.05): the eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) levels were highest in fish fed the control diet (P < 0.05); and EPA and DHA levels in fish fed the FO30/LO35/SO35 diet were closest to the control diet (P < 0.05). In contrast, fish fed the diet containing 100% plant oils (LO30/SO35/SFO35) had the highest level of total n‐6 fatty acids in the fillet and liver. In a 12‐day refrigerated storage at 1°C the thiobarbituric acid (TBA), trimethylamin nitrogen (TMA‐N) and pH values gradually increased in all dietary groups (P < 0.05). The chemical indicators of spoilage, TBA, TMA‐N, and pH values were within the limit of acceptability for human consumption.  相似文献

Dried tomato pomace (DTP), as an alternative for other ingredients in laying hen as well as broiler diets, has shown to produce comparable or even superior performance. However, there is no report on the digestibility coefficients of DTP in poultry. The coefficient of total tract apparent digestibility (CTTAD) of dry matter, nitrogen and ash as well as metabolisable energy values of DTP alone and with enzyme were determined in laying and broiler cockerels, using the force feeding method. In two consecutive experiments, 15 adult laying-type cockerels (Hy-Line W36, 35-week-old) and 21 meat-type cockerels (Ross 308, 8-week-old) were randomly grouped into three groups of five and seven replicates, respectively. Each bird received glucose solution (30 g/50 ml), DTP alone (30 g) and DTP with Avizyme 1502 (30 g + 20 mg) and the voided excreta were collected quantitatively, for 48 h.Adult laying-type cockerels and broilers were similar in total output of endogenous dry matter, nitrogen, ash and energy. DTP increased (P<0.001) total dry matter, nitrogen, ash and energy output of experimental birds. Enzyme neither reduced the output of dry matter, nitrogen, ash and energy from adult cockerels, nor improved the CTTAD of dry matter, nitrogen, ash and metabolisable energy values of DTP (P>0.05). However, the enzyme reduced the loss of dry matter, nitrogen and energy from broiler cockerels fed on DTP by 17%, 16% and 14%, respectively. Peptostreptococcus elsdenii, a strict anaerobe from the rumen, was grown on a medium containing yeast extract and [1-(14)C]- or [2-(14)C]-lactate. Radioisotope from lactate was found in all cell fractions, but mainly in the protein. The label in the protein fraction was largely confined to a few amino acids: alanine, serine, aspartic acid, glutamic acid and diaminopimelic acid. The alanine, serine, aspartic acid and glutamic acid were separated, purified and degraded to establish the distribution of (14)C from lactate within the amino acid molecules. The labelling patterns in alanine and serine suggested their formation from lactate without cleavage of the carbon chain. The pattern in aspartic acid suggested formation by condensation of a C(3) unit derived directly from lactate with a C(1) unit, probably carbon dioxide. The distribution in glutamic acid was consistent with two possible pathways of formation: (a) by the reactions of the tricarboxylic acid cycle leading from oxaloacetate to 2-oxoglutarate, followed by transamination; (b) by a pathway involving the reaction sequence 2 acetyl-CoA-->crotonyl-CoA-->glutaconate-->glutamate.  相似文献

The nutritional values of 16 D-amino acids in chick growth were studied on the purified diets containing crystalline amino acids as a sole source of nitrogen. Growth rate, feed consumption and nitrogen retention were measured. The nutritional values of D-amino acids were studied by comparing individually with the control groups fed on the diet containing all L-amino acids and negative control groups fed with the diet omitted the corresponding L-isomer. The following results were obtained. Essential amino acids: 1. Equal or almost equal nutritional value to the corresponding L-isomer; methionine, phenylalanine, leucine, proline. 2. Half nutritional value compared with L-isomer; valine. 3. Small nutritional value compared with L-isomer; tryptophan, isoleucine, histidine. 4. No nutritional value; lysine, threonine, arginine. Non-essential amino acid: 1. Equal or almost equal nutritional value to the corresponding L-isomer; serine, tyrosine, cystine. 2. There is a possibility that it has a slight growth retardation effect; alanine. 3. The growth retardation effect was found; aspartic acid.  相似文献

Ten holidic diets, varying in amino-acid concentration or composition, were fed to larvae of Tenebrio molitor for four weeks at 27 +/- 0.25 degrees C and 65 +/- 5% r.h. Effects of diet on growth, food utilization and energy utilization were recorded for individual larvae. Differences in gains in fresh weight or in dry matter among larvae fed diets containing 0% to 5% of the amino-acid mixture were not demonstrated. However, larvae fed 10% or 20% of this mixture gained more than the former, but less than larvae fed a diet of ground whole wheat and brewer's yeast (9:1, w/w). When the amino-acid mixture was supplemented with alanine, aspartic acid, and serine, or with these three and asparagine, and was fed to larvae at the 10% level, growth was slower than with the unsupplemented mixture. Supplementation of the amino-acid mixture with the first three amino acids did not reduce larval growth when fed at the 20% level. Amounts of food and of energy utilized were positively correlated with larval fresh-weight and dry-matter gains. Energy utilization was negatively correlated with dietary amino-acid level. Food and energy utilization and larval growth were influenced by dietary amino acids, either metabolically or through phagostimulation.  相似文献

1. Proline was found to be the major component of CTC-12 (44%) and FSS II (45%) strain.2. The cypermethrin treatment resulted in an increase in most of the amino acids of sixth instar larvae and all amino acids of adult beetles of CTC 12 strain.3. In the susceptible strain (FSS II), however, the tyrosine, phenylalanine and arginine increased, whereas serine, proline, glycine, alanine, valine, isoleucine, leucine and lysine were decreased significantly in the sixth instar larvae.4. Field and laboratory studies were conducted to evaluate the intestinal responses to partial replacement of fish meal with rice protein concentrate (RPC) in practical diets for blackspot seabream Pagellus bogaraveo . Two experimental diets were formulated to be isoproteic and isoenergetic with an increasing level of RPC (20 and 35%, respectively) and were tested against a fish meal-based control diet (RPC0). The diets showed similar features for growth performances and both intestinal histology and digestive enzymes. This study confirmed that RPC does not induce intestinal mucosa alterations in this fish. The dietary RPC supplement caused a significant increase in trypsin activity, whereas lipase activity was reduced.  相似文献

The current experiment aimed to study whether interactions with lipid metabolism possibly might explain the relative increased liver weight obtained in fish fed sub-optimal methionine levels. A basal diet based on a blend of plant proteins which is low in methionine (1.6 g Met/16 g N) was compared to a methionine adequate diet (2.2 g Met/16 g N) prepared by adding dl-methionine (2.4 g/kg) to the basal diet in the expense of wheat grain. Fish oil was used as the lipid source. The diets were balanced in all nutrients except methionine. The diets were fed to Atlantic salmon (500 g BW) for a period of 3 months. Feed intake did not differ, rendering the intake of all nutrients except methionine equal. Fish fed the low methionine diet had an increased liver size relative to body weight, indicating fat deposition in the liver. Fish given the sub-optimal methionine diet showed about six times higher fatty acid synthase (FAS) activity as compared to the fish fed the adequate methionine diet, indicating a higher de novo lipogenesis. A significant rise in the liver 18:1 to 18:0 fatty acid ratios also supported storage of lipids over fatty acid oxidation. Indeed, methionine limitation resulted in significantly higher TAG concentrations in the liver. Sub-optimal dietary methionine also resulted in lower hepatic taurine concentrations and the total bile acids concentrations were reduced in faeces and tended to be reduced in plasma. Taken together, our data show that salmon fed sub-optimal methionine levels had increased relative liver weight and developed signs commonly described in the early stage of non-alcoholic fatty liver disease in rodent models (increased FAS activity, changed fatty acid ratios and TAG accumulation).  相似文献

A study was undertaken to examine the effect of different amounts of dietary lysine (13 and 21 g kg?1 diet), lipid (80 and 160 g kg?1 diet) and L ‐carnitine (0.2 and 1.0 g kg?1 diet) on growth performance, proximate composition and amino acid metabolism of the African catfish (Clarias gariepinus). Juvenile African catfish (23 ± 1.5 g/fish) were stocked into 70‐L aquaria (16 aquaria, 28 fish/aquarium) connected to a recirculation system during a maximum period of 74 days. All groups were fed at a level of 24 g kg?0.8 day?1 in an experiment run at pair feeding. Animals receiving 1.0 g carnitine accumulated up to six times more carnitine in their tissues than animals receiving 0.2 g (P < 0.05). Acyl‐carnitine and free L ‐carnitine levels increased in the whole body and in tissues. Dietary L ‐carnitine supplements increased protein‐to‐fat ratios in the body, but did not affect growth rate. Protein‐to‐fat ratios were only affected when the biosynthesis capacity of L ‐carnitine was restricted due to low lysine levels and when there was a shortage of dietary fat. When lysine was offered at 21 g kg?1 feed, dietary L ‐carnitine supplements did not affect the amino acid concentrations of body tissues. Dietary L ‐carnitine supplements raised the concentration of glutamic acid > aspartic acid > glycine > alanine > arginine > serine > threonine in skeletal muscle tissue (P < 0.05). Total amino acid concentration in muscle and liver tissues (dry‐matter basis) increased from 506 to 564 and from 138 to 166 mg g?1, respectively, when diets were offered with high L ‐carnitine, low lysine and low fat levels. These data suggest that dietary L ‐carnitine supplementation may increase fatty acid oxidation and possibly decrease amino acid combustion for energy.  相似文献

The skeletal muscle of Ophicephalus punctatus contains nine essential free amino acids, arginine, histidine, isoleucine, leucine, methionine, phenylalanine, threonine, valine and lysine, and eight non-essential amino acids, alanine, aspartic acid, cystine, glutamic acid, glycine, tyrosine, proline and serine. Histidine and lysine dominated the free amino acids pool. Seasonal variation was detected in the levels of histidine, arginine, leucine, phenylalanine, glycine, cystine and serine with highest values occurring in April and again in November. Changes were also detected in the concentrations of certain amino acids as the fish grew in size. Levels of free amino acids did not significantly differ between sexes. Factors effecting variation are discussed.  相似文献

A 15N kinetic-analysis of the assimilation of nitrate nitrogenin the roots of rice seedlings indicated that (1) nitrate wasrapidly reduced to ammonia in the roots, where it was incorporatedinto glutamine and glutamic acids; (2) the pattern of nitrateassimilation into amino acids was very similar to that of ammoniumassimilation; and (3) the pattern of nitrogen incorporationinto protein was also similar to that of the incorporation withNH4-feeding. In the shoots, alanine, serine, glutamic acid, -amino butyricacid and aspartic acid were relatively strongly labelled with15N as compared with the other amino acids. Steady-states of growth of Acer pseudoplatanus L. cells were established in a chemostat in a medium containing, successively, two different concentrations of urea alone as a limiting nitrogen source. These cells had different growth characteristics from those grown in the presence of urea and nitrate together as limiting nitrogen sources. The urea medium produced fewer, larger cells which contained larger amino acid pools and converted nitrogen to protein more efficiently. In addition, certain amino-N components in the pools occurred in far higher concentrations than in cells grown in the presence of urea and nitrate together, notably asparagine, aspartic and glutamic acids, alanine, glycine, serine and threonine.When amino acid pools were examined in detail, it was found that steady-states of some constituents were not achieved until several hundred hours after stability had occurred in cell dry weight, cell number, and cell volume.  相似文献

This trial studied the effect of including mannanoligosaccharides (MOS, Bio-Mos®, Alltech Inc., USA) in the diet on the caecal volatile fatty acids (VFA) and pH of rabbits from 34 to 90 days of age. Three experimental diets were compared: control diet, zinc bacitracin (ZnBac) diet (control diet with 0.1 g ZnBac/kg feed) and MOS diet (control diet with 2.0 g MOS/kg feed). Rabbits were slaughtered at 34, 48, 69 and 90 days of age and caecal contents were collected and analyzed for dry matter, pH and VFA concentration. The empty caecum and caecal contents weights relative to live weight were also determined.Age affected (P<0.0001) VFA concentration and pH values in the caecum. The pH decreased with age whereas VFA concentration increased. Rabbits fed MOS had higher (P<0.05) VFA and tended (P=0.098) to had lower pH in the caecum than rabbits fed ZnBac and control diets. Acetic, propionic and butyric acids concentrations in the caecum increased with MOS but its molar proportions were similar among diets. Diet had no effect on empty caecum weight and caecal contents weight and dry matter concentration. There was no interaction effect between diet and age. From 34 to 90 days of age, VFA production was higher (P<0.05) in the caecum of rabbits fed MOS than in those fed ZnBac diet and control diet.The addition of MOS to the diet increased the VFA concentration in the caecum of growing rabbits from 34 to 90 days of age.  相似文献

The dry matter content of 18 samples of fish silage prepared from fish species or prawn offal varied from 190 to 274 g/kg (mean 239). Chemical composition (mean and range, g/kg dry matter) was: crude protein 686 (487–882); ether extract 100 (53–211); ash 163 (105–319); and total mercury 1.05 mg/kg (0.24–4.05 mg/kg).The nutritional value of fish silage, prepared from Ocean perch and Nannygai, was investigated for grower pigs. Wheat-based diets were formulated in which fish silage partially or completely replaced soya been meal as the protein supplement. The diets were evaluated over the 20 to 45 kg growth phase. Half the pigs were slaughtered at 45 kg live weight; the remainder were fed on the wheat and soya bean meal diet to 80 kg live weight in order to observe the effect of withdrawal of fish silage at 45 kg on subsequent growth performance and mercury content in the lean tissue.The addition of fish silage improved growth rate (P < 0.05) and food conversion ratio (P < 0.01) during the 20 to 45 kg growth phase. No adverse effects on carcass quality were observed. During the 45 to 80 kg growth phase, when all pigs were fed on the wheat and soya bean meal diet, all produced similar performance. Over the 20 to 80 kg growth phase, the beneficial effects of fish silage on growth rate and feed conversion were maintained.Methyl mercury content in the semi-membraneous muscle of pigs fed on fish silage and slaughtered at 45 kg live weight was 0.085 mg/kg on a wet weight basis. This decreased to 0.018 mg/kg in pigs that had the fish silage withdrawn from the diet during the 45 to 80 kg growth phase.  相似文献

Summary The depth distribution and composition of residual amino acids was determined in December 1980 in the Bransfield Strait. The major components were aspartic and glutamic acids together with alanine, glycine and serine. Non-protein amino acids were not found in significant amounts. On the average, the amino acids accounted for 25% of the total organic carbon and 50% of the organic nitrogen. Amino acid nitrogen/chlorophyll ratios indicated that in most euphotic zone samples phytoplankton in good physiological condition was present whereas heavily degraded material was found in deeper water layers. Statistical treatment and cluster analysis show distinct differences between euphotic and aphotic zone samples which could be used to characterize phytoplankton communities.  相似文献
