A strain of Zymomonas mobilis (ZYM-TS 1) was isolated from fermenting palm wine (toddy). In addition to glucose, sucrose, and fructose, the organism utilized hydrolysates of corn ( Zea mays ) flour, corn starch and ragi ( Eleusine coracana ) flour. Amounts of ethanol produced in media (adjusted to 10% (w/v) total carbohydrates) fortified with wheat bran extract only, were comparable with those obtained from the defined media containing yeast extract, (NH4)2SO4, KH2PO4, and MgSO4.7H2O. The results indicate that wheat bran extract can supply all the necessary nutrients required for ethanol production by Zymomonas mobilis .  相似文献

A membrane separator/bioreactor system was developed for rapid detection of Escherichia coli O157:H7. The system consisted of a membrane separator/bioreactor (0.45 μm of the pore size) to separate the-complexes of E. coli O157:H7 and alkaline phosphatase-conjugated anti-E. coli O157:H7 antibodies from the sample and to produce p-nitrophenol through the enzymatic reaction (p-nitrophenyl phosphate hydrolysis), and an optical detector for measuring the p-nitrophenol absorbance at 400 nm. The membrane material and the flow rate of the substrate for the enzymatic hydrolysis had great effects on the absorbance of p-nitrophenol. The optimum conditions for the enzymatic reaction were determined as 1.0 M Tris buffer, pH 8.0, and 0.1 M MgCl2 for this system. The detection range was 104± 107 CFU/mL with a relative standard deviation of 4.3 ± 14.2%, and whole procedure could be completed in 50 min without any enrichment and culture. Other bacteria such as Salmonella typhimurium, Campylobacter jejuni and Listeria monocytogenes had no significant interference with the detection of E. coli O157:H7.  相似文献

The B798 light-harvesting baseplate of the chlorosome antenna complex of the thermophilic, filamentous anoxygenic phototrophic bacterium Chloroflexus aurantiacus has been isolated and characterized. Isolation was performed by using a hexanol-detergent treatment of freeze-thawed chlorosomes. The isolated baseplate consists of Bchl a, beta-carotene, and the 5.7 kDa CsmA protein with a ratio of 1.0 CsmA protein/1.6 Bchl a/4.2 beta-carotenes. The baseplate has characteristic absorbance at 798 nm as well as carotenoid absorbance maxima at 519, 489, and 462 nm. The energy transfer efficiency from the carotenoids to the Bchl a is 30% as measured by steady-state and ultrafast time-resolved fluorescence and absorption spectroscopies. Energy equilibration within the Bchl a absorbing regions exhibits ultrafast kinetics. Circular dichroism spectroscopy shows no evidence for excitonically coupled Bchl a pools within the 798 nm region.  相似文献

Ultraviolet (UV) radiation has recently been demonstrated to drive an aerobic production of methane (CH4) from plant tissues and pectins, as do agents that generate reactive oxygen species (ROS) in vivo independently of UV. As the major building-blocks of pectin do not absorb solar UV found at the earth's surface (i.e. >280 nm), we explored the hypothesis that UV radiation affects pectin indirectly via generation of ROS which themselves release CH4 from pectin. Decreasing the UV absorbance of commercial pectin by ethanol washing diminished UV-dependent CH4 production, and this was restored by the addition of the UV photosensitizer tryptophan. Certain ROS scavengers [mannitol, a hydroxyl radical (OH) scavenger; 1,4-diazabicyclo[2.2.2] octane; and iodide] strongly inhibited UV-induced CH4 production from dry pectin. Furthermore, pectin solutions emitted CH4 in darkness upon the addition of OH, but not superoxide or H2O2. Model carbohydrates reacted similarly if they possessed —CH3 groups [e.g. methyl esters or (more weakly) acetyl esters but not rhamnose]. We conclude that UV evokes CH4 production from pectic methyl groups by interacting with UV photosensitizers to generate OH. We suggest that diverse processes generating OH could contribute to CH4 emissions independently of UV irradiation, and that environmental stresses and constitutive physiological processes generating ROS require careful evaluation in studies of CH4 formation from foliage.  相似文献

Abstract Glucose-fructose oxidoreductase (GFOR) is a periplasmic enzyme of the ethanologenic, Gram-negative bacterium Zymomonas mobilis . It contains tightly bound NADP+ as cofactor. In Z. mobilis GFOR-recombinant strains, a precursor form of GFOR was accumulated. To assay the preGFOR for its NADP(H) content and enzymatic activity, it was purified from an overproducing strain. Using SDS-PAGE, the precursor subunit size was determined to approximately 45 kDa (compared with a 40 kDa subunit size for the mature GFOR subunit). The N-terminal amino acid sequence of the precursor was determined. The N-terminal residues of the GFOR matched with the signal sequence from the DNA sequence of the gene gfo . The precursor form of GFOR was enzymatically active and contained the cofactor NADP(H).  相似文献

The effect of six long-chained aliphatic amines on 14CO2-reduction, electron transport and the 515 nm absorbance change and shrinkage in isolated intact and broken chloroplasts from spinach ( Spinacia oleracea L. cv. Weibulls Medania) was investigated. Five of the six investigated amines affected photosynthesis in intact chloroplasts by inhibiting 14CO2-reduction. In broken chloroplasts the same amines uncoupled electron transport. When added to intact chloroplasts the five amines induced a light-dependent oxygen uptake leading to (he formation of hydrogen peroxide. The oxygen uptake was not due to the amines acting as Mehler reactants. The mode of action, different from that of simple aliphatic amines, was an effect on membrane integrity, first affecting the membrane potential. At higher amine concentrations a more general effect on the ion conditions in the thylakoids was evident.  相似文献

A method was developed which allows the isolation and purification of cytoplasmic membranes and chlorosomes from cells of Chloroflexus aurantiacus grown under different light conditions. The dipolar ionic detergent Deriphat (0.08%) and a sodium iodide gradient centrifugation were used in isolating cytoplasmic membranes. Chlorosomes were prepared with 0.16% of the dipolar ionic detergent Miranol and purified by a sucrose gradient centrifugation. Cytoplasmic membrane fractions prepared from either high- (3,000 W m-2), medium-(200 W m-2) or low- (7 W m-2) light-grown cells had near infrared absorption bands at 866, 808, and 755 nm in a constant characteristic absorbance ratio of 6:3.8:1. In all cytoplasmic membrane preparations, the amount of bacteriochlorophyll a (Bchl a) per cytochrome, the amount of Bchl a per reaction center, and reaction center per milligram of cytoplasmic membrane protein was found to be constant. No Bchl c was present. Five respiratory enzyme activities have been measured in the cytoplasmic membrane fraction. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of denatured cytoplasmic membrane showed many bands, but a major polypeptide with an apparent molecular weight of 8,000. In contrast, sodium dodecyl sulfate-polyacrylamide gel electrophoresis of purified chlorosomes did not contain the 8,000-molecular-weight band but revealed only three distinct protein bands with molecular weights of 15,000, 12,000, and 6,000. Isolated chlorosomes contained Bchl c and a small, yet constant, amount of Bchl a (absorbing at 790 nm) in a molar ratio of 25:1. The data indicated that the components of the photosynthetic apparatus in the cytoplasmic membrane of Chloroflexus aurantiacus remained constant and only the amount of antenna Bchl c varied with light conditions.  相似文献

The distribution of divalent cation stimulated ATPase activity in relation to the distribution of other enzyme activities was studied for membrane fractions from wheat roots ( Tritium aestivum L . cv. Svenno). A homogenate from dark grown plants was fractionated by differential centrifugation at 1000 g , 10,000 g , 30,000 g and 60,000 g (1, 10, 30 and 60 KP fractions), followed by partition in an aqueous polymer two-phase system, using polyethylene glycol 4000/dextran T500 concentrations of 5.7/5.7, 5.9/5.9, 6.1/6.1, 6.3/6.3 and 6.5/6.5% (w/w). The 30 KP fraction was also separated by counter-current distribution id a 6.3/6.3% two-phase system. Protein and activities of Ca2+, Mg2+, and Mn2+ stimulated ATPases. cytochrome oxidase, light induced absorbance change (LIAC) related to cyt b reductions, inosine diphosphatase and NADH dependent antimycin A insensitive cytochrome c reductase were measured.
The partition of ATPase activities stimulated by Ca2+, Mg2+ or Mn2+ was similar at all polymer concentrations tested, indicating: a low cation specificity of the dominating ATPases. The distribution of ATPases. agreed with different marker enzymes in different centrifuge fractions. Divalent cation stimulated ATPases were evidently related to several of the organelles. In the different fractions the distribution of ATPase activity should then follow that of the marker enzyme of the dominant organelle. From studies with different polymer concentrations the 6.3/6.3-system was selected for further separation of the membranes in the 30 KP fraction by counter-current distribution. By this method one fraction was obtained, which probably consisted of plasmalemma and was free from mitochondrial material. Indications for plasmalemma in this fraction were a) similar partition as protoplasts and b) high LIAC activity.  相似文献

The visible absorption spectrum of bacteriochlorophyll a (Bchl) in the 560–620 nm (yellow) region can be used to establish the coordination state of the central Mg atom. Five-coordinated Mg species absorb near 580 nm, whereas 6-coordinated Mg species are red-shifted to about 610 nm. Five-coordination is confirmed to be the principal coordination state of Mg in self-aggregated bacteriochlorophyll. The equilibrium constant for the reaction Bchl · Py + Py ai Bchl · Py2 has been determined from computer-assisted analyses of spectral data, where Py represents pyridine. The spectral criteria for the coordination state of Mg in bacteriochlorophyll advanced here are shown to be applicable to both in vitro and in vivo systems. Similar spectral behavior is exhibited by chlorophylls a and b, and a band at 633 nm is shown to be associated with the presence of 6-coordinated Mg in chlorophyll a.  相似文献

The size and de-epoxidation state of the xanthophyll cycle pool was measured in cultures of Nitzschia palea grown at six fluence rates in continuous light or with a 12 h photoperiod. In both series the size of the pool increased with increasing irradiance. The de-epoxidized form, diatoxanthin, was only present at fluence rates saturating for growth. The portion of diadinoxanthin, which was not readily de-epoxidized in saturating light, was constant and not related to the size of the pool. In the culture grown in a light-dark cycle at 300 μmol photons m-2 s-1 (PAR) increasing de-epoxidation took place in the latter half of the photoperiod, when the rate of photosynthesis was decreasing. A rapid, spectrophotometric method for measuring the extent of de-epoxidation of the xanthophyll cycle pool in a culture of diatoms is described. Upon addition of a small volume of hydrochloric acid to an extract of pigments in 90% acetone, the absorbance at 480 nm is reduced. The size of the reduction is a measure of the state of the xanthophyll cycle pool, since the absorbance of diatoxanthin is reduced by 5%, but the absorbance of diadinoxanthin by 87% due to an epoxide-furanoid rearrangement, which causes the absorption spectrum to be shifted by ca 20 nm towards shorter wavelengths.  相似文献

Abstract. The half time (t1/2) of the reduction of P-700+ in the millisecond time frame is known to be limited by the reaction between plastoquinol and the cytochrome cytb6f complex. This is considered to be the rate limiting reaction of thylakoid electron transport and measurements of it provide a means of analysing how thylakoid election transport is regulated in vivo. The half time for the reduction of photochemically oxidized P-700 has been measured in vivo using absorbance changes around 820 nm. The results showed that t1/2 is independent of irradiance and decreases as photosynthetic induction progresses. Even with a constant t1/2 the quantum efficiency of PSI declined as irradiance increased. The significance of the concept of photosynthetic control of electron transport is discussed in the light of these observations.  相似文献

Isoprenoid biosynthesis in bacteria: Two different pathways?   总被引:4,自引:0,他引:4  
Abstract The biosynthesis of isopentenylpyrophosphate, a central intermediate of isoprenoid formation, was investigated in six different bacterial organisms. Cell-free extracts of Myxococcus fulvus, Staphylococcus carnosus, Lactobacillus plantarum and Halobacterium cutirubrum converted [14C]acetyl-CoA or [14C]hydroxymethylglutaryl-CoA to [14C]mevalonic acid. Furthermore, [14C]mevalonic acid, [14C]mevalonate-5-phosphate and [14C]mevalonate-5-pyrophosphate were metabolized to [14C]isopentenylpyrophosphate in bacteria. In contrast, no intermediates of this reaction sequence could be detected using cell-free extracts of Zymomonas mobilis and Escherichia coli . These results indicate that at least two different pathways for the biosynthesis of isopentenylpyrophosphate are present in bacteria.  相似文献

Abstract:  An experimental study on the effect of isolation and crowding of the desert locust, Schistocerca gregaria (Forskål) on haemolymph pigment composition (as measured by absorbance ratios at 460 and 680 nm) was carried out at the International Centre of Insect Physiology and Ecology, Nairobi. The results showed that haemolymph absorbance ratios of gregarious and solitarious locusts were different (P < 0.05) and that there was no phase-specific sex differentiation with regard to this measure (P < 0.05). The haemolymph pigment composition changes during isolation and grouping showed contrasting trends. It changed rapidly in nymphs, but slowly and erratically in adults both resulting from isolation and grouping at hopper stage. Significant shifts in locust phase status were recorded in both situations in nymphs in F0 generation, and in the adults by the end of the F2 and F3 generations of solitary and gregarious conditions, respectively. Shifting of fledglings did not induce significant changes in their mean absorbance ratios in both situations, i.e. isolation and crowding within F0 generation. The ratio of length of posterior femur (F) to greatest width of the head capsule (C) changed either at a similar rate or more often slower than the haemolymph pigment ratio and both required more than a generation to transform fully. The study has shown that haemolymph pigment composition could be a suitable measure for monitoring phase changes at nymphal stages of the desert locust.  相似文献

Unusually for a deep-sea fish, the retina of the myctophid (lanternfish) Myctophum nitidulum was found to contain two visual pigments, shown by extract spectrophotometry to be maximally sensitive at 468 and 522 nm, respectively, giving this species one of the broadest spectral ranges of all deep-sea fishes. High performance liquid chromatography (HPLC) indicated that the retina contained both A1- and A2-based chromophores. Surprisingly, the maximum absorbance (λmax) values of the two visual pigments were too far apart to form a rhodopsin–porphyropsin 'pigment pair', suggesting they were based on distinct opsins each linked to a different chromophore. This might be an adaptation to the detection of both long-wave bioluminescence and residual shorter-wave surface illumination, and could be related to this animal's tendency to migrate towards surface waters at night.  相似文献

Antenna components in the energy transfer processes of a green photosynthetic bacterium Chloroflexus aurantiacus were spectrally investigated by time-resolved fluorescence spectroscopy at −196°C on intact cells. Besides major antenna components so far reported, three minor components were resolved; those were Bchl c located at 785 nm, the baseplate Bchl a at 819 nm and Bchl a in the B808-866 complex at 910 nm. The last component was assigned to a longer wavelength antenna closely associated with a reaction center. An additional Bchl c fluorescence component was kinetically suggested to be present, which can be an energy donor to a major Bchl c. Presence of these minor components was signified in terms of (1) increase in the spectral overlap integral and (2) adjustment of the direction of dipole moments in the energy transfer sequence of intact cells.  相似文献

Predominantly, Hoplias malabaricus inhabits stagnant O2 poor environments, whereas Hoplias lacerdae occurs in well-aerated streams. The present study evaluates the influence of mode of life on O2 uptake and gill ventilation in equally-sized (300 g) specimens of this genus at 25° C. Comparing the species, H. lacerdae was characterized by the highest O2 uptake and gill ventilation combined with a relatively higher cost of breathing and a lower O2 extraction. Both species substantially increased ventilation in response to hypoxia with the difference that H. malabaricus exclusively augmented tidal volume, whereas H. lacerdae also increased breathing frequency.  相似文献

In the LH2 proteins from Rhodobacter (Rb.) sphaeroides, the hydrogen bonds between the bacteriochlorophyll (Bchl) molecules and their proteic binding sites exhibit a strong variance with respect to carotenoid content and type. In the absence of the carotenoid molecule, such as in the LH2 from Rb. sphaeroides R26.1, the void in the protein structure induces a significant reorganization of the binding site of both Bchl molecules responsible for the 850 nm absorption, which is not observed when the 800 nm absorbing Bchl is selectively removed from these complexes. FT Raman spectra of LH2 complexes from Rb. sphaeroides show that the strength of the hydrogen bond between the 850 nm absorbing Bchl bound to the alpha polypeptide and the tyrosine alpha(45) depends precisely on the chemical nature of the bound carotenoid. These results suggest that the variable extremity of the carotenoid is embedded in these LH2 complexes, lying close to the interacting Bchl molecules. In the LH2 from Rhodopseudomonas acidophila, the equivalent part of the rhodopin glucoside, which bears the glucose group, lies close to the amino terminal of the antenna polypeptide. This contrast suggests that the structure of the carotenoid binding site in LH2 complexes strongly depends on the bacterial species and/or on the chemical nature of the bound carotenoid.  相似文献

Background. The mouse model using a human isolate of Helicobacter pylori is being widely accepted as an economical means of studying gastric infection. A noninvasive monitoring method would be useful for repeated testing to establish the time course of infection and the efficacy of treatments. In this study, we describe factors that affect interpretation of 13C urea breath test results for the assessment of H. pylori infection status in this model.
Materials and Methods. Female C57Bl/6 mice that underwent gavage with H. pylori or saline were breath-tested using 50 μg of 13C urea at intervals up to 2 months after inoculation. The generation of 13CO2 (excess δ13CO2) by infected mice was compared to that of uninfected controls. The effects of diet, fasting, and coprophagy on the reliability of the 13C urea breath test were quantitated.
Results. Both commercial and synthetic mouse diets exhibited marked in vitro urease activity. A minimum fasting time of 13 hours prior to breath testing significantly reduced this dietary contribution to excess δ13CO2 values. The coprophagic tendency of the mice caused spuriously high excess δ13CO2 counts in the breath of both control and H. pylori –infected mice.
Conclusions. Although the dietary contribution to spuriously high values of excess δ13CO2 in mice breath-tested for H. pylori infection was reduced by fasting, the high nonspecific urease activity generated by coprophagy severely limited the reliability of the urea breath test in the assessment of H. pylori infection status.  相似文献

Abstract In Zymomonas mobilis a novel pathway for the formation of glycerol 3-phosphate was identified by enzymatic studies and nuclear magnetic resonance spectroscopy. This pathway branches off from the Entner-Doudoroff pathway at the intermediate glyceraldehyde 3-phosphate and proceedes via dihydroxyacetone phosphate, dihydroxyacetone, glycerol to glycerol 3-phosphate. The reaction sequence is catalyzed by the enzymes triosephosphate isomerase (0.4 U (mg protein)−1), dihydroxyacetone phosphatase (0.31 U (mg protein)−1), dihydroxyacetone reductase (0.25 U (mg protein)−1), and glycerokinase (0.08 mU (mg protein)−1), respectively. The action of a postulated aldolase catalyzing the cleavage of fructose 6-phosphate to dihydroxyacetone and glyceraldehyde 3-phosphate could be excluded.  相似文献

The epidermis of Argenteum mutant of Pisum sativum L. and Vicia faba L. was shown to be effective in protecting mesophyll photosynthesis from UV-C irradiation (peak 254 nm; 1.5 W m−2). These plants were chosen because it is easy to peel the epidermis from both sides of the leaf in Argenteum and the abaxial side in Vicia . Chlorophyll a fluorescence induction was decreased to the same extent by UV-C radiation in both leaf sides when the epidermis was removed. The fluorescence from the leaf with the epidermis inact was not affected. With irradiation of leaves with higher intensity (3.4 W m−2) of UV-C, the variable fluorescence was decreased by UV-C impinging on the abaxial side, but not the adaxial side in either of the plant species. A methanol extract from the mutant Pisum epidermis had a high absorbance in the UV region, and the absorbance was more than two-fold larger in adaxial epidermis than in abaxial epidermis. These results indicate that the epidermal layer, which contains substances that have high UV absorbance, protects mesophyll photosynthesis against UV radiation.  相似文献
