Cytomixis is a spontaneous and normal process occurring through the formation of plasma channels between neighbouring pollen mother cells (PMCs). This phenomenon was observed in the PMCs of 2 variaties of Secale cereale which had been collected from Gausu and Shanxi Provinces. The process had led to the formation of up to 6.37% of PMCs with chromosome numbers deviating from the normal haploid number n=7. The abnormal PMCs contained chromosome numbers ranging from 5–9 in metaphase Ⅰ. Abnormal PMCs with decreased and increased chromosome numbers could be the result of cytomixis. The cytological consequences and the evolutionary significance of cytomixis are discussed.  相似文献

应用普通电镜和DGD去包埋技术 ,研究了烟草花粉母细胞中的细胞融合现象及细胞融合过程中细胞骨架的变化。观察发现 ,处于凝线期的花粉母细胞 ,其内含物 ,包括细胞器和染色质 ,主要通过胞质通道向相邻细胞发生转移。DGD去包埋观察发现 ,花粉母细胞中核骨架与细胞质内及胞间连丝和胞质通道内胞质骨架连接成一个整体。在整个细胞融合过程中 ,均有核骨架纤维与染色质相连。本文讨论了细胞骨架在细胞融合过程中的作用  相似文献

黄瓜花粉母细胞减数分裂行为的研究   总被引:17,自引:1,他引:17  
研究了华北型、华南型和西南型黄瓜花粉母细胞的减数分裂行为,发现黄瓜细胞核减数分裂的同步性较高,细胞质是同时型分裂。在细胞核分裂的过程中,核仁在前期Ⅰ到中期Ⅰ逐渐消失,在前期Ⅰ再次出现,随后消失;染色体在前期Ⅰ到中期Ⅰ逐渐收缩,变得清晰,至末期Ⅰ解螺旋,变得模糊,在前期I再次清晰。不同生态型黄瓜终变期的染色体构型均以环状二价体为主。在前期Ⅰ和前期Ⅰ,西双版纳黄瓜的核仁都相应地比另外两种生态型黄瓜品种的多,在后期Ⅰ还偶尔出现染色体桥,显示了西双版纳黄瓜变种的特殊性。研究还发现寒冷的气候条件下栽培黄瓜都能够形成高频率的多分体,推测其形成很可能与低温逆境有关。  相似文献

Standard lead precipitation procedures have been used to examine the localization of ATPase activity during cytomixis in pollen mother cells of Lilium davidii var. willmottiae (Wilson) Roffill. Before cytomixis, cells at this stage of development show ATPase activity on plasma membrane, in the endoplasmic reticulum, dictyosomes, plastids, plasmodesmata, and in part of the groundplasm; however, there is no ATPase activity on the chromatin and nucleolus. During cytomixis, the chromatin substance begin to transfer from one cell to an adjacent cell, reaction product indicating ATPase activity is observed associated with the chromatin and nucleolus. ATPase activity is also found with the cistenae of both endoplasmic reticulum and dictyosomes, and some plastids. There is no deposition of ATPase reaction product associated with the plasm membrane and intercellular spaces. After cytomixis, the chromatin is little or no deposition of enzyme reaction product. ATPase activity, however, is consistenlly found within the intercellular space and on the plasm membrane, and also occur in the endoplasmic reticulum, dictyosome and plastid. The presence or absence of ATPase activity in the cell structure of pollen mother cells before, during or after eytomixis is discussed in relation to the active uptake or export of water for short-distance transport. It is also suggested that the intensive ATPase activity in the nucleus during cytomixis of pollen mother cells is evidence for a transport system involved in the active movement of the intercellular migrating ebromatin substance.  相似文献

In greenhouse tests, Alternaria crassa (Sacc.) Rands killed > 80% of inoculated jimsonweed (Datura stramonium L.) seedlings within 14 days following a 9-h dew period at 25°C with 1 × 105 spores/ml, and after 8 h of dew at 1 × 106 spores/ml. At least 10 h of dew with 1 × 105 spores/ml and 9 h of dew with 1 × 106 spores/ml were required to obtain 100% mortality of fungus-inoculated plants. Growth stage and inoculum concentration studies revealed that higher concentrations of inoculum were required to obtain 100% mortality of larger plants. Weed control was significantly reduced by day/night air temperatures of 35°C and 24°C, respectively, at all inoculum concentrations as compared to the controls at lower air and dew temperature regimes. The results of these studies indicate that A. crassa has potential as a biological herbicide for the control of jimsonweed.  相似文献

During the genesis of microspores and male gametes, the percentages of cells with an increas- ed and decreased chromosome number at metaphase Ⅱ, mitotic metaphase of microspores and generative cells approximated to the one at previous stage (metaphase Ⅰ, metaphase Il, and microspores) respectively, and the one at metaphase 1 again approximated to the percentages of chromatin intercellular migration of type 2.3→1 and 1→2.3 respectively, which had no significant diversity statistically. According to this fact, it was considered that the changes of chromosome numbers were related to the chromatin intercellular migration at synizesis stage and that pollen mother cells with an abnormal chromosome number at metaphase Ⅰ basically were able to continue developing untill the formation of male gametes.  相似文献

Datura roots were pressure-infiltrated with 400 µg ml–115N-nitrate feeding solutions with and without the additionof 7 mM L-methionine-DL-sulphoximine (MSO), a glutamine synthetaseinhibitor. Over a 30 min time course the main diversion of newlyreduced 15N in MSO untreated roots was to glutamine. In MSO-treatedroots ammonia assimilation into amino compounds was completelysuppressed, with resultant accumulation of a large 15N ammoniapool. This treatment also caused marked concentrational changesin the free amino compound pools, suggesting that conditionsof nitrogen stress had been induced. Glutamate dehydrogenaseactivity was unaffected by the MSO treatment. The results are consistent with the concept that the glutaminesynthetase/glutamate synthase pathway is the major route ofnewly reduced nitrogen assimilation in Datura roots.  相似文献

The lectin of Jimson-weed seeds (Datura stramonium L.) was isolated by affinity chromatography on a polysaccharide mixture from mycelium of Aspergillus niger. The lectin yields two bands on disc electrophoresis, it has sedimentation coefficient s20,w = 3.8 S and its apparent molecular weight estimated by thin layer gel chromatography is 120,000. The lectin reduced with mercaptoethanol yields on polyacrylamide gel electrophoresis in the presence of dodecyl sulfate three zones corresponding to subunits of molecular weight 72,000, 45,000 and 25,000. The lectin contains large amounts of cystine, glycine, 6.3% of hydroxyproline residues, 4.5% glucosamine and 28% of neutral sugar, predominantly arabinose. The lectin is nonspecific in human erythrocyte ABO system, it is not inhibited by simple sugars but is inhibited by a partial hydrolysate of chitin-containing mixture of polysaccharides from Aspergillus niger.  相似文献

采用压片法,对麦蓝菜的花粉母细胞减数分裂及体细胞核型进行了研究。结果表明,麦蓝菜的绝大多数花粉母细胞减数分裂中染色体的行为正常,在终变期同源染色体配对后可形成15个二价体;在少数花粉母细胞减数分裂中观察到落后染色体、染色体桥等异常行为;其减数分裂为同时型,其花粉粒育性为88.04%。麦蓝菜的染色体数目为2n=2X=30,核型公式为K(2n)=2X=30=22m(2SAT)+8 sm,染色体相对长度组成为2n=16M2+14M1,其核型为"1A"型。  相似文献

Abstract.— Although insect herbivory is frequently assumed to be responsible for the maintenance of plant secondary metabolites such as alkaloids, the assumption is controversial and experimental evidence for this assumption is sparse. We examined natural selection on the two major alkaloids present in the leaves of Datura stramonium and found that both alkaloids came under active selection. We found negative directional selection for scopolamine (natural selection acting to reduce scopolamine levels) and stabilizing selection for hyoscyamine (natural selection acting to maintain an intermediate level of hyoscyamine). We also present evidence that insect herbivores act as the agents of selection on these alkaloids. Finally, we show that there were no trade-offs in resistance to different species of insects.  相似文献

The metabolic relationship between littorine and hyoscyamine has been monitored in transformed roots of Datura stramonium. Quantification by GC of unlabelled littorine and by GCMS of 13C-labelled littorine demonstrated that exogenously added littorine (0.1 mm) was significantly metabolised (35%) to hyoscyamine. In contrast, exogenously added hyoscyamine was not metabolised to littorine, indicating that this conversion is irreversible. The conversion of littorine to hyoscyamine was suppressed by P-450 oxidase inhibitors (particularly clotrimazole), implicating the involvement, at least in part of a cytochrome P-450 activity operating hyoscyamine biosynthesis. 以欧洲黑杨的雄花枝为材料,采用温室切枝水培和醋酸洋红染色制片法,观察花粉母细胞减数分裂进程及其与花芽发育外部形态的关系。结果表明:(1)欧洲黑杨花粉母细胞的减数分裂进程快慢与温度高低关系密切;(2)减数分裂发生时其花芽的外部形态发育没有明显变化;(3)花粉母细胞减数分裂不同步性突出,同一花药内的花粉母细胞处于不同的减数分裂时期,且同一花芽尖端发育明显滞后于中部和基部;(4)减数分裂过程中有少量的核仁异常现象,虽观察到较多平行纺锤体现象,但未发现天然2n花粉。该研究结果揭示了欧洲黑杨减数分裂的遗传变异规律,为诱变育种、倍性育种以及分子水平的进一步研究奠定了细胞学基础。  相似文献

以石油醚为溶剂提取曼陀罗种子中的脂肪酸,采用GC-MS技术测定其化学成分,并通过饲料混毒法测定了曼陀罗种子脂肪酸对小鼠的毒性.结果表明,曼陀罗种子中含有23.2%的脂肪酸,其主要化学成分为1,20-二十碳二酸(34.55%)、Crinan-3-ol,1,2-didehydro-,(3à)(13.41%)、8,11-十八碳二烯酸(4.56%)、8,11,14-二十碳三烯酸(Z,Z,Z)(4.39%)、2,3,3-三甲基辛烷(4.35%)、7,10-十八碳二烯酸(3.61%)、亚油酸乙脂(2.64%)、α-亚麻酸(2.34%)、cis-乙酸环己脂-2-丙酸(2.34%)等.采用饲料混毒法对小鼠的毒性测定结果表明,曼陀罗种子脂肪油对小鼠无胃毒作用.研究认为,曼陀罗种子中含有多种对人体有益的活性成分,可作为脂肪酸食用油来源或从中提取对人体有益的活性成分开发利用.  相似文献

大麦种子经5种不同剂量γ射线辐照后, M1植株的花粉母细胞出现多种染色体畸变类型,例如后Ⅰ、后Ⅱ中出现桥、落后,后Ⅰ中出现不对称分裂,后Ⅱ中出现不同步分裂,中期Ⅰ出现环状四价体,末期Ⅰ中有三极分裂和微核,四分体中出现微核且有的微核独立于四分体,从而形成多分体。花粉母细胞的染色体畸变率随剂量的增高而升高,二者呈极显著直线相关关系。 M1花粉母细胞的染色体畸变率与M2的叶绿素突变率及农艺性状变异率呈显著或极显著指数函数关系。 M2花粉母细胞的染色体畸变率低于M1。 Abstract:Barley seeds were irradiated with five different doses of 60 Co-γ ray.There were many kinds of chromosome aberration in pollen mother cells,such as bridge,laggards in anaphase I and anaphase II,asymmetrical division in anaphase I and asynchronous division in anaphase II,ring quadrivalent in metaphase I,three pole division and micro-nuclei in telophase I,micro-nuclei in tetrad and some micro-nucleus separates from the tetrad to form polyad.The frequency of chromosome aberration in pollen mother cells of M1 increased with the increase of dose.There was a significant linear relationship between them.The frequencies of agronomic character mutation and chlorophyll mutation in M2 significantly correlated with the frequencies of chromosome aberration in pollen mother cells of M1 and showed an exponential function.The chromosome aberration frequency in pollen mother cells of M2 was lower than that of M1.  相似文献

文章从减数分裂过程、小孢子发育两方面,探讨了木立芦荟(Aloe arboresens Mill.)花粉败育的原因。木立芦荟花粉母细胞染色体数目为2n=14,由四对长染色体和三对短染色体组成,属二型性核型。其减数分裂异常,发现存在单价体和多价体、染色体桥、落后染色体、不均等分离、微核等。同时观察到木立芦荟染色体具有极度的粘质性,使减数分裂各阶段的染色体不易散开。统计各种异常现象出现的频率并分析了这些异常现象形成的可能机制及对正常小孢子形成的影响,推测染色体间的丝状粘连可能是木立芦荟小孢子母细胞减数分裂异常并导致败育花粉产生的主要因素。成熟花粉粒中90%以上为败育花粉,属碘败型。  相似文献

The relative contributions made by the l-arginine/agmatine/N-carbamoylputrescine/putrescine and the l-ornithine/putrescine pathways to hyoscyamine formation have been investigated in a transformed root culture of Datura stramonium. The activity of either arginine decarboxylase (EC 4.1.1.19) or ornithine decarboxylase (EC 4.1.1.17) was suppressed in vivo by using the specific irreversible inhibitors of these activities, dl--difluoromethylarginine or dl--difluoromethylornithine, respectively. It was found that suppression of arginine decarboxylase resulted in a severe decrease in free and conjugated putrescine and in the putrescine-derived intermediates of hyoscyamine biosynthesis. In contrast, the suppression of ornithine decarboxylase activity stimulated an elevation of arginine decarboxylase and minimal loss of metabolites from the amine and alkaloid pools. The stimulation of arginine decarboxylase was not, however, sufficient to maintain the same potential rate of putrescine biosynthesis as in control tissue. It is concluded that (i) in Datura the two routes by which putrescine may be formed do not act in isolation from one another, (ii) arginine decarboxylase is the more important activity for hyoscyamine formation, and (iii) the formation of polyamines is favoured over the biosynthesis of tropane alkaloids. Arachidonic acid, cellulase, CuSO4, a sonicate of Phytophthora infestans mycelium and a spore suspension of Penicillium chrysogenum all elicited the formation of the sesquiterpenoid phytoalexins lubimin, 3-hydroxylubimin and rishitin in fruit cavities of Datura stramonium. 3-Hydroxylubimin was the predominant phytoalexin formed after treatment of the fruits with arachidonic acid, cellulase and the P. infestans preparation. Copper sulphate was a potent elicitor of lubimin but not 3-hydroxylubimin. The fungus P. chrysogenum metabolized lubimin and 3-hydroxylubimin to 15-dihydrolubimin and 3-hydroxy-15-dihydrolubimin respectively, both in fruit cavities inoculated with spores of this fungus and in pure culture. The 15-dihydrolubimin formed in the fruits by the fungus was further metabolized (by the fruits) to both isolubimin and 3-hydroxy-15-dihydrolubimin. The precursor-product relationships between all of the subject compounds was investigated by feeding experiments with 3H-labelled compounds. 2-Dehydro-[15-3H1]lubimin was rapidly and efficiently incorporated into lubimin and may be the direct precursor of lubimin in planta. 3-Hydroxy[2-3H1]lubimin was incorporated into the nor-eudesmane rishitin but 10-epi-3-hydroxy[2-3H1]lubimin was not.
