The use of proton and carbon-13 magnetic resonance spectroscopy for the determination of the geometry and the stoichiometry of the actinomycin D-deoxyguanosine 5'-monophosphate complex is outlined. The dimerization of actinomycin D has been reexamined by recording the proton magnetic resonance spectrum of actinomycin D to much lower concentrations through the use of Fourier transform nuclear magnetic resonance techniques. The effect of the actinomycin D dimerization on the observed chemical shifts that results from the additon of nucleotides to an actinomycin D solution is directly demonstrated by comparing the actinomycin D-nucleotide titrations at both low (approximately 0.3 mM) and high (approximately 12 mM) concentrations of actinomycin D. In the presence of excess nucleotide the chemical shifts of the actinomycin D groups were essentially the same for both the low and high concentration titrations. The complexes of actinomycin D with pdG-dC, dG-dC, deoxyguanosine 3'-monophosphate, G-C, C-G, dIMP(5'), 2, 6-diaminopurine deoxyribose, and other nucleotides were also investigated by proton magnetic resonance and visible spectral titrations. These data were interpreted in terms of the molecular geometry of the complexes and in terms of the effect of the structure of the nucleotide base on the relative binding affinity of the nucleotides for the two nucleotide binding sites of actinomycin D. The carbon-13 chemical shifts of dGMP(5') were measured as a function of concentration over the concentration range of 0.5-0.025 M. The infinite dilution carbon-13 chemical shifts were graphically estimated from the dilution curves. These values were used to calculate the changes in the chemical shifts of the dGMP carbons that result from the formation of an actinomycin D-(dGMP)2 complex. It was not possible to interpret these carbon-13 chemical shift changes in terms of only ring current effects, which thus rules out the use of carbon-13 spectroscopy in the determination of the geometries of the actinomycin D complexes with the mono- and dinucleotides. The induced chemical shifts in the proton spectra may be used in the determination of the geometries of the complexes. A consideration of these data for the above nucleotide series shows that the predominant complex formed is one in which the guanine rings in the two nucleotide binding sites of actinomycin D are oriented in a manner very similar to that observed in the cocrystalline complex of actinomycin D with deoxyguanosine.  相似文献

A new metabolite of vitamin D was isolated from the blood plasma of chicks given large doses of vitamin D3. The isolation involved methanol-chloroform extraction and four column chromatographic steps. The metabolite was identified by high- and low-resolution mass spectroscopy, chemical derivatization, an comigration with authentic standard as 3 beta, 24(R)-dihydroxy-9,10-seco-5,7,10(19)-cholestatriene [24(R)-hydroxyvitamin D3]. No detectable 24-(R)-hydroxyvitamin D3 was recovered from 16 L of plasma from chicks receiving physiologic levels of vitamin D3.  相似文献

基于中药材产地适宜性分析地理信息系统(TCM-GIS)平台,获得了采样区域11个生态因子值.应用偏最小二乘回归法(PLS)分析了石斛中的化学成分及其与生态因子的相关性.结果表明: 同种石斛不同产地的化学成分含量差异显著.土壤类型与铁皮石斛中多糖含量呈极显著正相关;年降水量与金钗石斛中石斛碱积累呈极显著正相关;温度是影响鼓槌石斛中毛兰素含量的主要因素.主成分分析表明,浙江是铁皮石斛的最佳产地,贵州是金钗石斛的最佳产地,而云南是鼓槌石斛的最佳产地.

A method that combines NMR spectral and structural information into a constructed three-dimensional (3D)-connectivity matrix is developed for modeling biological binding activity of small molecules. The 3D-connectivity matrix for a molecule is defined by associating the distances between all possible carbon-to-carbon connections with their assigned carbon NMR chemical shifts. In this project we selected from the total 3D-connectivity matrix a subset, the two-dimensional (2D) (13)C-(13)C COSY and a theoretical long range 2D (13)C-(13)C distance connectivity spectral plane. Patterns of (13)C chemical shifts observed at these two relative distances for 50 steroids were used to produce a mathematical relationship for the steroids' relative binding affinity (pK(i)) to the aromatase enzyme. We call this technique comparative structural connectivity spectra analysis (CoSCoSA) modeling. Using combinations of the 2D COSY and 2D long-range distance spectra as modeling parameters, we built four CoSCoSA models. One model was made from the 2D COSY spectra alone and another was developed using only the 2D long-range distance spectra. Then the COSY and long-distance spectra were combined in two different ways: starting with the combined principal components (PCs) from the separately calculated COSY and distance spectra or using the combined raw spectra (3D). The best CoSCoSA model was based on the combined PCs from COSY and distance spectra. This model had an r(2) of 0.96 and a leave-one-out cross-validation (q(2)) of 0.92. In general CoSCoSA modeling combines the quantum mechanical information inherent in NMR chemical shifts with internal molecular atom-to-atom distances to give a reliable and straightforward basis for predictive modeling. The technique has the flexibility and accuracy to outperform not only the cross-validated variance q(2) of previously published quantitative structure-activity relationships (QSAR) but also those obtained by related quantitative spectral data-activity relationships (QSDARs) lacking connectivity dimensions.  相似文献

A five-dimensional (5D) APSY (automated projection spectroscopy) HCNCH experiment is presented, which allows unambiguous correlation of sugar to base nuclei in nucleic acids. The pulse sequence uses multiple quantum (MQ) evolution which enables long constant-time evolution periods in all dimensions, an improvement that can also benefit non-APSY applications. Applied with an RNA with 23 nucleotides the 5D APSY-HCNCH experiment produced a complete and highly precise 5D chemical shift list within 1.5 h. Alternatively, and for molecules where the out-and-stay 5D experiment sensitivity is not sufficient, a set of out-and-back 3D APSY-HCN experiments is proposed: an intra-base (3D APSY-b-HCN) experiment in an MQ or in a TROSY version, and an MQ sugar-to-base (3D APSY-s-HCN) experiment. The two 3D peak lists require subsequent matching via the N1/9 chemical shift values to one 5D peak list. Optimization of the 3D APSY experiments for maximal precision in the N1/9 dimension allowed matching of all 15N chemical shift values contained in both 3D peak lists. The precise 5D chemical shift correlation lists resulting from the 5D experiment or a pair of 3D experiments also provide a valuable basis for subsequent connection to chemical shifts derived with other experiments.  相似文献

Myrtucomvalones D–F, three new triketone‐phloroglucinol‐triketone adducts, and three known ones (myrtucommulone E, myrtucommulone D and callistenone D) were obtained from Myrtus communis ‘Variegata’. Myrtucomvalone D is a pair of enantiomers which was further resolved into (+)‐myrtucomvalone D and (?)‐myrtucomvalone D by chiral HPLC. Their structures and complete stereochemistry were established from interpretation of NMR and crystallographic data and chemical calculations. Myrtucomvalone F, myrtucommulone D and callistenone D showed significant antibacterial activities.  相似文献

Evolutionary theory proposes that signals used in sexual selection can only be stable if they are honest and condition dependent. However, despite the fact that chemical signals are used by many animals, empirical research has mainly focused on visual and acoustic signals. Vitamin D is an essential nutrient for lizards, but in some lizards its precursor (cholesta-5,7-dien-3-ol=provitamin D) is found in femoral gland secretions, which males use for scent marking and intraspecific communication. By allocating provitamin D to secretions, males might need to divert vitamin D from metabolism. This might be costly and condition dependent. We tested whether diet quality affected chemical signals of male Iberian rock lizards (Lacerta monticola) and its consequences for sexual selection. After experimental supplementation of dietary vitamin D, males increased the proportion of provitamin D in femoral secretions. Further experiments showed that females detected these changes in males' signals by chemosensory cues, and discriminated provitamin D, and changes in its concentration, from similar steroids (i.e. cholesterol) found in secretions. Moreover, females preferred areas scent marked by males with more provitamin D in their secretions. This mechanism would confer honesty to chemical signals of male lizards, and, thus, females may rely on it to select high-quality males. We suggest that the allocation of vitamins and other essential nutrients to either visual (e.g. carotenoids) or chemical ornaments might be the common basis of honest sexual displays in many animals.  相似文献

To study the chemical constituents from traditional Chinese herb Ophiopogon japonicus (Thunb.) Ker-Gawl., a new steroidal glycoside, named ophiopojaponin C (1), together with two known ones, was isolated by column chromatography. Spectroscopic and chemical evidence revealed the structures to be ophiopogenin 3-O-[α-L-rhamnopyranosyl(1→2)]-β-D-xylopyranosyl(1→4)-β-D-glucopyranoside (1), diosgenin 3-O-[2-O-acetyl-α-L-rhamnopyranosyl(1→2)]-β-D-xylopyranosyl(1→3)-β-D-glucopyranoside (2), and ruscogenin 1-O-[2-O-acetyl-α-L-rhamnopyranosyl(1→2)]-β-D-xylopyranosyl(1→3)-β-D-fucopyranoside (3).  相似文献

A new metabolite of vitamin D3 was produced in vitro by perfusing rat kidneys with 1,25-dihydroxyvitamin D3 (4 X 10(-6) M). It was isolated and purified from the lipid extract of the kidney perfusate by high-performance liquid chromatography. By means of ultraviolet absorption spectrophotometry, mass spectrometry, chemical derivatization, and chemical synthesis, the new metabolite was identified as 1,23-dihydroxy-24,25,26,27-tetranorvitamin D3. Along with the new metabolite, three other previously identified metabolites, namely, 1,24,25-trihydroxyvitamin D3, 1,25-dihydroxy-24-oxovitamin D3, and 1,23,25-trihydroxy-24-oxovitamin D3, were also isolated. The new metabolite was also formed when 1,23,25-trihydroxy-24-oxovitamin D3 was used as the substrate. Thus, the new metabolite fits into the following metabolic pathway: 1,25-dihydroxyvitamin D3----1,24(R),25-trihydroxyvitamin D3----1,25-dihydroxy-24-oxovitamin D3----1,23,25-trihydroxy-24-oxovitamin D3----1,23-dihydroxy-24,25,26,27-tetranorvitamin D3. Further, we used 1 alpha,25-dihydroxy[1 beta-3H]vitamin D3 in the kidney perfusion system and demonstrated 1,23-dihydroxy-24,25,26,27-tetranorvitamin D3 as the major further metabolite of 1,25-dihydroxyvitamin D3, circulating in the final perfusate when kidneys were perfused with 1,25-dihydroxyvitamin D3 (6 X 10(-10) M) for 4 h. The biological activity of 1,23-dihydroxy-24,25,26,27-tetranorvitamin D3 (C-3 alcohol) and its metabolic relationship to 1-hydroxy-23-carboxy-24,25,26,27-tetranorvitamin D3 (calcitroic acid or C-23 acid), the other previously identified side-chain cleavage metabolite of 1,25-dihydroxyvitamin D3, are unknown and are presently undergoing investigation.  相似文献

Here we report the variation of diterpenes from three populations of the brown alga Dictyota mertensii. The populations were analyzed by 1H NMR and GC-MS techniques. Both chemical GC-MS and 1H NMR profiles of D. mertensii presented diterpenes as major constituents. In all populations, we detected the prenylated guaiane diterpenes, dictyoxide, pachydictyol A, isopachydictyol A, dictyol B and its acetate, and dictyol C. Although dictyol H has been described as a taxonomic marker, this diterpene was found in only two of the three studied populations. This result reinforces the need for studies of temporal and geographic variation for the establishment of the taxonomic limits of the species chemical patterns. It was also possible to observe mass spectra fragmentation and proton chemical shift in the NMR spectra characteristic from xeniane diterpenes, for the first time in this species, which led to the detection of dictyodial, suggesting that D. mertensii belongs to the chemotaxonomic group of species D. ciliolata, D. jamaicensis, and D. menstrualis, all of which are producers of diterpene from chemical groups I and III.  相似文献

Bovine mitochondrial (mt) phenylalanine tRNA (tRNA(Phe)), which lacks the 'conserved' GG and T psi YCG sequences, was efficiently purified by the selective hybridization method using a solid phase DNA probe. The entire nucleotide sequence of the tRNA, including modified nucleotides, was determined and its higher-order structure was investigated using RNaseT2 and chemical reagents as structural probes. The D and T loop regions as well as the anticodon loop region were accessible to RNaseT2, and the N-3 positions of cytidines present in the D and T loops were easily modified under the native conditions in the presence of 10mM Mg2+. On the other hand, the nucleotides present in the extra loop were protected from the chemical modification under the native conditions. From the results of these probing analyses and a comparison of the sequences of mitochondrial tRNA(Phe) genes from various organisms, it was inferred that bovine mt tRNA(Phe) lacks the D loop/T loop tertiary interactions, but does have the canonical extra loop/D stem interactions, which seem to be the main factor for bovine mt tRNA(Phe) to preserve its L-shaped higher-order structure.  相似文献

Nineteen secondary metabolites of the brown alga Dictyota dichotoma (Huds.) Lam. and fifteen metabolites of the brown alga D. linearis (Ag.) Grev. were isolated and their chemical structures were elucidated on the basis of their NMR and mass spectral data. The diterpenes isopachydictyolal (1) from D. dichotoma and 4alpha-acetyldictyodial (2) from D. linearis are new natural products. The antiviral activity of metabolites isolated in adequate amounts was evaluated in laboratory assays against Herpes simplex virus I (HSV I) and Poliomyelitis Virus I, using Vero cells as hosts.  相似文献

31P NMR spectra of isolated rabbit bladder and uterus were obtained under steady-state arterial perfusion in vitro at rest and while stimulated. The spectra contained seven major peaks: phosphoethanolamine, sn-glycero(3)phosphocholine, inorganic phosphate (Pi), phosphocreatine, and the gamma, alpha, and beta peaks of ATP. Chemical analyses, high-pressure liquid chromatography, and NMR spectroscopy of aqueous extracts of bladders identified a number of other components that also made contributions to, but were not resolved in, the spectra of the intact tissues: UTP, GTP, UDP-Glc, NAD+, phosphocholine, and sn-glycero(3)phosphoethanolamine. Intracellular pH of unstimulated bladders and uteri, measured from the chemical shift of the Pi peak, was 7.10 +/- 0.09 S.D. and 7.01 +/- 0.12 S.D., respectively. The chemical shift of the beta-ATP peak in the smooth muscles was significantly upfield (-0.3 ppm) compared to the chemical shift observed in striated muscles (cat biceps and rat myocardium). An ADP peak was identified in stimulated and ischemic bladders. The chemical shifts of the nucleotides observed in perfused bladders were calibrated as a function of free Mg2+ concentration in solutions containing phosphocreatine, Pi, ADP, and ATP at an ionic strength of 180 mM. We derived the following estimates for the intracellular free Mg2+ concentration: uterus, 0.40 mM; unstimulated bladder, 0.46 mM; stimulated and ischemic bladder, 0.50 mM (from the ATP chemical shift) and 0.45 (from the ADP chemical shift); cat biceps, 1.5 mM; and rat myocardium, 1.4 mM.  相似文献

利用紫花曼陀罗细胞悬浮培养转化外源对羟基苯甲醛合成天麻素,并应用多种色谱技术进行分离纯化,根据转化产物的理化性质和光谱数据分析鉴定结构。实验表明,紫花曼陀罗细胞成功将对羟基苯甲醛转化为天麻素(Ⅱ),同时也得到了由对羟基苯甲醛生成天麻素的转化中间体对羟基苯甲醇(Ⅰ)。在培养基中添加0.1mg/L的水杨酸能显著提高细胞对外源对羟基苯甲醛的糖基化率,而保持气升式发酵罐(25-L)罐内压力为低压(0.001MPa)也能提高细胞对外源对羟基苯甲醛的糖基化率。实验证明,紫花曼陀罗细胞悬浮培养能有效转化对羟基苯甲醛合成天麻素。  相似文献

Differential scanning calorimetry (DSC) provides authentic and accurate value of DeltaC(p)(X), the constant-pressure heat capacity change associated with the N (native state)<-->X (heat denatured state), the heat-induced denaturation equilibrium of the protein in the absence of a chemical denaturant. If X retains native-like buried hydrophobic interaction, DeltaC(p)(X) must be less than DeltaC(p)(D), the constant-pressure heat capacity change associated with the transition, N<-->D, where the state D is not only more unfolded than X but it also has its all groups exposed to water. One problem is that for most proteins D is observed only in the presence of chemical denaturants such as guanidinium chloride (GdmCl) and urea. Another problem is that DSC cannot yield authentic DeltaC(p)(D), for its measurement invokes the existence of putative specific binding sites for the chemical denaturants on N and D. We have developed a non-calorimetric method for the measurements of DeltaC(p)(D), which uses thermodynamic data obtained from the isothermal GdmCl (or urea)-induced denaturation and heat-induced denaturation in the presence of the chemical denaturant concentration at which significant concentrations of both N and D exist. We show that for each of the proteins (ribonuclease-A, lysozyme, alpha-lactalbumin and chymotrypsinogen) DeltaC(p)(D) is significantly higher than DeltaC(p)(X). DeltaC(p)(D) of the protein is also compared with that estimated using the known heat capacities of amino acid residues and their fractional area exposed on denaturation.  相似文献

As a result of the surveying of 13 taxa of the genus Diplotaxis, it was found that their leaf flavonoids are flavonol glycosides derived from either kaempferol, quercetin and isorhamnetin. Multivariate analysis of these flavonoid data of taxa suggest a close chemotaxonomic affinity. On the basis of their chemical composition the taxa relationships are discussed. The allopolyploid origin of D. muralis with D. tenuifolia and D. viminea as parentals is strongly supported by chemical evidence.  相似文献

There is currently a significant need to improve our understanding of the factors that control a number of critical soil processes by integrating physical, chemical and biological measurements on soils at microscopic scales to help produce 3D maps of the related properties. Because of technological limitations, most chemical and biological measurements can be carried out only on exposed soil surfaces or 2-dimensional cuts through soil samples. Methods need to be developed to produce 3D maps of soil properties based on spatial sequences of 2D maps. In this general context, the objective of the research described here was to develop a method to generate 3D maps of soil chemical properties at the microscale by combining 2D SEM-EDX data with 3D X-ray computed tomography images. A statistical approach using the regression tree method and ordinary kriging applied to the residuals was developed and applied to predict the 3D spatial distribution of carbon, silicon, iron, and oxygen at the microscale. The spatial correlation between the X-ray grayscale intensities and the chemical maps made it possible to use a regression-tree model as an initial step to predict the 3D chemical composition. For chemical elements, e.g., iron, that are sparsely distributed in a soil sample, the regression-tree model provides a good prediction, explaining as much as 90% of the variability in some of the data. However, for chemical elements that are more homogenously distributed, such as carbon, silicon, or oxygen, the additional kriging of the regression tree residuals improved significantly the prediction with an increase in the R2 value from 0.221 to 0.324 for carbon, 0.312 to 0.423 for silicon, and 0.218 to 0.374 for oxygen, respectively. The present research develops for the first time an integrated experimental and theoretical framework, which combines geostatistical methods with imaging techniques to unveil the 3-D chemical structure of soil at very fine scales. The methodology presented in this study can be easily adapted and applied to other types of data such as bacterial or fungal population densities for the 3D characterization of microbial distribution.  相似文献
