1 Carex arenaria forms large clonal fragments (up to 12 m long) in environments in which soil-bound resources limit growth.
2 We hypothesized that extensive integration of C. arenaria would facilitate the exploitation of scarce and patchily distributed soil resources and that the continued functioning of old roots would enable exploitation of resources that are temporally variable.
3 We used labelling with 14C and acid fuchsin to study the degree and extent of physiological integration and root function of intact clonal systems of C. arenaria in a sand dune area in south Sweden.
4 The uptake and translocation of dye suggests that old roots remain capable of taking up water and nutrients, in contrast with negative reports from previous studies. Water was translocated both acropetally and basipetally.
5 Thirty per cent of the assimilated carbon was found to be translocated towards the growing apex. Smaller, but significant, amounts of carbon were translocated basipetally throughout the fragments (17–74 ramet generations).
6 The results are discussed in terms of the source–sink relations of large clonal systems in the field. The translocation patterns are considered in relation to soil moisture and nutrient availability.  相似文献

Cyst and root-knot nematodes show high levels of gross morphological similarity. This presents difficulties for the study of their ecology in natural ecosystems. In this study, cyst and root-knot nematode species, as well as some ectoparasitic nematode species, were identified using the second internal transcribed spacer (ITS2) sequence variation detected by polymerase chain reaction-single-strand conformational polymorphism (PCR-SSCP). The ITS2 region was sufficiently variable within the taxa investigated to allow species to be separated on the basis of minor sequence variation. The PCR primers used in this study were effective for 12 species with three genera within the Heteroderinae (Globodera pallida, G. rostochiensis, Heterodera arenaria/avenae, H. ciceri, H. daverti, H. hordecalis, H. mani, H. schachtii, H. trifolii, Meloidogyne ardenensis, M. duytsi and M. maritima). However, pathotypes of Globodera pallida and G. rostochiensis could not be distinguished. The method was tested at two coastal dune locations in The Netherlands (one in the lime-poor dunes of the north and one in calcareous dunes of the south) to determine the population structure of cyst nematodes. At each site, cyst nematodes were associated with three plant species: two plant species on the foredune (Elymus farctus and Ammophila arenaria) and one plant species occurring further inland (Calamagrostis epigejos). Two species of cyst nematodes, H. arenaria and H. hordecalis, were found. H. arenaria associated with vigorous A. arenaria and H. hordecalis in association with degenerating A. arenaria and C. epigejos. The field survey demonstrated that in coastal dunes abiotic factors may be the important affecting the distribution of cyst nematodes.  相似文献

Meloidogyne polycephannulata n. sp. is described from specimens collected from an area cultivated with carrot cv. Brasilia, in the city of Rio Paranaíba, in the region of Alto Paranaíba, Minas Gerais State, Brazil. The perineal pattern of the female is circular to ovoid with a high dorsal arch that has widely spaced, coarse annulations. The lateral field may have a deep furrow separating the dorsal and ventral arches. The medial lips are short and wide, whereas the lateral lips are large and triangular. The female stylet is 15-16 μm long with wide knobs, distinctly divided by an indentation in the center. Its tip is slightly curved dorsally. The excretory pore opens 34-65 μm from the anterior end. Females retain eggs and second-stage juveniles in their body cavity, similar to that of the cyst-forming nematodes. Males are 1.3-1.7 mm long and have a high head cap that is rounded and slopes posteriorly. The labial disc is fused to the medial lips. The head region has several irregular annulations that are similar in appearance to the first or second body annules that are likewise irregular, making the head region appear to be extremely large. The stylet of the male is 21-24 μm long; it is slender, and has small, rounded knobs, that are distinctly indented medially and appear heart-shaped. The shaft has several tiny projections throughout its length. Mean second-stage juvenile length is 411.7 μm. The juvenile head cap is elevated, the medial lips are small, and the lateral lips are elongate to triangular-shaped. The head region has several short, incomplete and irregular transverse annulations. The juvenile stylet is 14-23 μm long with small, rounded, and sloping knobs. The thin tail ends with a short hyaline portion that is variable in size (16-26 μm) and with a small, rounded tip. Isozyme profiles of esterases from Meloidogyne javanica show 3 strong bands (SB) at Rm 46, 59, and 66; profiles of M. polycephannulata n. sp. show a SB at Rm 47 and a weak band (WB) at Rm 52; M. petuniae has two SB at Rm 44 and 53; M. phaseoli has a SB at 53, 58, and 64 Rm; M. brasilensis has three SB at Rm 40, 58, and 66 and a WB at Rm 71; M. pisi has a SB at Rm 40, 60, and 64 and two WB at 46 and 50 Rm. Data from sequencing the 18S rDNA region of M. polycephannulata n. sp. confirms that it is different from M. arabicida, M. arenaria, M. ethiopica, M. incognita, M. javanica, M. paranaensis, and M. thailandica. Sequence identity among these eight species ranged between 85 to 93.4%. Meloidogyne polycephannulata n. sp. reproduces very well on carrot and tomato; poorly on pepper; and not at all on cotton, peanut, tobacco, watermelon, and sweet corn.  相似文献

The fossil leaf flora from the Shanwang Formation, Linqu County, Shandong Province, eastern China, is diverse, abundant, well preserved and discretely spaced, all of which make it suitable for the recognition and application of leaf physiognomic characters. This article is the first study of Chinese Tertiary floras using the CLAMP (Climate Leaf Analysis Multivariate Program) approach. Numerous bedding planes yield leaf assemblages through over 20 m thickness of diatomaceous shales within the Middle Miocene Shanwang Formation. The diatomaceous shales, containing abundant fossils, have been divided into 19 units. A working platform (3×3 m) in the Diatomite Quarry of Shanwang was studied for about 3 months in 1998 and as many fossils as possible were recorded layer by layer. This collecting method, used for the first time in the history of Shanwang research, enabled us to record the presence or absence of megafossils in distinct units so that we could investigate leaf physiognomic characteristics and palaeoclimate for each time interval. Leaf fossils from six units (5, 6, 7, 13, 15 and 16) have been used to estimate palaeoclimate parameters of the Shanwang area using the CLAMP 3B dataset. The predicted climate is that of a lakeside setting, at over 1000 m above sea level and within the 17–15-Ma time interval. Meteorological factors have been estimated using the CLAMP approach, including mean annual temperature (MAT; 9.5–11.2°C), warmest month mean temperature (20.2–22.5°C), growing season length (5.9–6.8 months), mean growing season precipitation (83–165 cm) and relative humidity (73–79%). MAT estimates from leaf margin analysis (LMA) range from 12.5 to 15.2°C, which are a maximum of 4° higher than those from our CLAMP analyses and from previous results of LMA studies of the Shanwang flora (10–13°C). Nearest living relatives (NLR) of other biota (pollen, fish, insects, diatoms, microthyriaceous fungi, etc.) indicate warm and moist temperate to subtropical conditions in the Shanwang area during the Miocene. CLAMP predictions are similar to the climate predicted from NLRs although the MAT prediction is a few degrees cooler. The fact that the Shanwang leaf flora is from a lacustrine sequence may have biased the CLAMP analysis towards cooler (and possibly wetter) climates. The modern Yangtze River valley is a reasonable environmental analogue for the Shanwang Middle Miocene, though Shanwang may have had cooler summers, cooler MAT and less seasonality in rainfall. The climate was essentially constant during deposition of the sequence studied, with possible minor variations in precipitation.  相似文献

中国新近系的年代地层单位山旺阶与哺乳动物分期的山旺期相对应 ,其定义的生物带相当于欧洲的MN3~MN5 ,下界年龄应为 2 0 .5Ma。按照现代地层学的建阶原则对山东临朐的山旺剖面进行综合研究 ,得出山旺组的底界年龄约为 1 8Ma ,山旺动物群的地质时代也由原来认为的与欧洲MN5相当提早到与MN4相当。尽管如此 ,研究结果证实山旺组仍然缺失定义的山旺阶下部沉积和MN3动物群。《国际地层指南》提倡将重点放在选择下界的界线层型来确定年代地层单位 ,其上界应该由后续单位的下界来确定。甘肃兰州盆地和秦安地区有发育的渐新世至中新世沉积出露 ,并有山旺阶最底部的化石和适合于作古地磁分析的沉积物 ,因此 ,是有可能建立山旺阶下界界线层型的有利地点。  相似文献

Total protein variation (up to ninety-five different positions) was revealed by two-dimensional electrophoresis (2-DE) in 18 isolates from populations of M. arenaria (6 isolates), M. incognita (10), M. javanica (1) plus an unclassified isolate in a previously reported study. Isolates of M. arenaria, M. javanica, Meloidogyne sp., and M. incognita formed two separate groups defined on the basis of two sets of protein positions that could be considered as diagnostic characters, but we could not identify these proteins by MALDI-TOF. To identify these marker positions, nano-liquid chromatography as peptides separation method was coupled to an ion-trap mass spectrometer for induced real-time fragmentation of eluted peptides. Group diagnostic proteins for M. incognita and M. arenaria were in-gel digested and on line analyzed by tandem mass spectrometry (LC-MS/MS). Six proteins out of seven selected spots were unambiguously identified by the analysis of the corresponding MS/MS (MS2) spectrum from parent ions fragmentation: Actin, Enolase, CG3752-PA protein similar to Aldehyde Dehydrogenase, HSP-60 and Translation initiation factor elF-4A. In M. incognita sample, de novo sequencing experiment of doubly charged ion at m/z=936.9 Da in spot 29 identified as enolase, reveals three residue substitutions (K to T, N to T, and D to E) when tentative sequence was compared with that of Anisakis simplex and Onchocerca volvulus enolase, thus three SNPs (single nucleotide polymorphisms) were also possibly identified.  相似文献

Within a 7 kb segment of the mtDNA molecule of the root knot nematode, Meloidogyne javanica, that lacks standard mitochondrial genes, are three sets of strictly tandemly arranged, direct repeat sequences: approximately 36 copies of a 102 ntp sequence that contains a TaqI site; 11 copies of a 63 ntp sequence, and 5 copies of an 8 ntp sequence. The 7 kb repeat-containing segment is bounded by putative tRNAasp and tRNAf-met genes and the arrangement of sequences within this segment is: the tRNAasp gene; a unique 1,528 ntp segment that contains two highly stable hairpin-forming sequences; the 102 ntp repeat set; the 8 ntp repeat set; a unique 1,068 ntp segment; the 63 ntp repeat set; and the tRNAf-met gene. The nucleotide sequences of the 102 ntp copies and the 63 ntp copies have been conserved among the species examined. Data from Southern hybridization experiments indicate that 102 ntp and 63 ntp repeats occur in the mtDNAs of three, two and two races of M.incognita, M.hapla and M.arenaria, respectively. Nucleotide sequences of the M.incognita Race-3 102 ntp repeat were found to be either identical or highly similar to those of the M.javanica 102 ntp repeat. Differences in migration distance and number of 102 ntp repeat-containing bands seen in Southern hybridization autoradiographs of restriction-digested mtDNAs of M.javanica and the different host races of M.incognita, M.hapla and M.arenaria are sufficient to distinguish the different host races of each species.  相似文献

Amplified fragment length polymorphism (AFLP) analysis has been used to characterize 15 root-knot nematode populations belonging to the three parthenogenetic species Meloidogyne arenaria , M. incognita and M. javanica. Sixteen primer combinations were used to generate AFLP patterns, with a total number of amplified fragments ranging from 872 to 1087, depending on the population tested. Two kinds of polymorphic DNA fragments could be distinguished: bands amplified in a single genotype, and bands polymorphic between genotypes (i.e. amplified in not all but at least two genotypes). Based on presence/absence of amplified bands and pairwise similarity values, all the populations tested were clustered according to their specific status. Significant intraspecific variation was revealed by AFLP, with DNA fragments polymorphic among populations within each of the three species tested. M. arenaria appeared as the most variable species, while M. javanica was the least polymorphic. Within each specific cluster, no general correlation could be found between genomic similarity and geographical origin of the populations. The results reported here showed the ability of the AFLP procedure to generate markers useful for genetic analysis in root-knot nematodes.  相似文献

Among root knot nematodes of the genus Meloidogyne, the polyploid obligate mitotic parthenogens M. arenaria, M. javanica, and M. incognita are widespread and common agricultural pests. Although these named forms are distinguishable by closely related mitochondrial DNA (mtDNA) haplotypes, detailed sequence analyses of internal transcribed spacers (ITSs) of nuclear ribosomal genes reveal extremely high diversity, even within individual nematodes. This ITS diversity is broadly structured into two very different groups that are 12%-18% divergent: one with low diversity (< 1.0%) and one with high diversity (6%-7%). In both of these groups, identical sequences can be found within individual nematodes of different mtDNA haplotypes (i.e., among species). Analysis of genetic variance indicates that more than 90% of ITS diversity can be found within an individual nematode, with small but statistically significant (5%-10%; P < 0.05) variance distributed among mtDNA lineages. The evolutionarily distinct parthenogen M. hapla shows a similar pattern of ITS diversity, with two divergent groups of ITSs within each individual. In contrast, two diploid amphimictic species have only one lineage of ITSs with low diversity (< 0.2%). The presence of divergent lineages of rDNA in the apomictic taxa is unlikely to be due to differences among pseudogenes. Instead, we suggest that the diversity of ITSs in M. arenaria, M. javanica, and M. incognita is due to hybrid origins from closely related females (as inferred from mtDNA) and combinations of more diverse paternal lineages.  相似文献

The endospore-forming bacterium Pasteuria penetrans is an obligate parasite of root-knot nematodes (Meloidogyne spp.). The primary objective of this study was to determine the effect of crop sequence on abundance of P. penetrans. The experiment was conducted from 2000 to 2008 at a field site naturally infested with both the bacterium and its host Meloidogyne arenaria and included the following crop sequences: continuous peanut (Arachis hypogaea) (P-P-P) and peanut rotated with either 2 years of corn (Zea mays) (C-C-P), 1 year each of cotton (Gossypium hirsutum) and corn (Ct-C-P), or 1 year each of corn and a vegetable (V-C-P). The vegetable was a double crop of sweet corn and eggplant (Solanum melongena). A bioassay with second-stage juveniles (J2) of M. arenaria from a greenhouse (GH) population was used to estimate endospore abundance under the different crop sequences. A greater numerical increase in endospore densities was expected in the P-P-P and V-C-P sequences than in the other sequences because both peanut and eggplant are good hosts for M. arenaria. However, endospore densities, as determined by bioassay, did not substantially increase in any of the sequences during the 9-year experiment. To determine whether the nematode population had developed resistance to the resident P. penetrans, five single egg-mass (SEM) lines from the field population of M. arenaria were tested alongside the GH population for acquisition of endospores from the field soil. Four of the five SEM lines acquired 9 to 14 spores/J2 whereas the GH population and one of the SEM lines acquired 3.5 and 1.8 spores/J2, respectively. Endospore densities estimated with the four receptive SEM lines were highest in the P-P-P plots (14-20 spores/J2), intermediate in the V-C-P plots (6-7 spores/J2), and lowest in the Ct-C-P plots (< 1 spore/J2). These results indicate that the field population of M. arenaria is heterogeneous for attachment of P. penetrans endospores. Moreover, spore densities increased under intensive cropping of hosts for M. arenaria, but the GH population of the nematode was not receptive to spore attachment. However, previously, the GH population was very receptive to spore acquisition from this field site. One explanation for this inconsistency is that the M. arenaria population in the field became resistant to the dominant subpopulation of P. penetrans that had been present, and this led to the selection of a different subpopulation of the bacterium that is incompatible with the GH population.  相似文献

Sequence variability and distribution of a newly characterized MPA2 satellite DNA family are described in five root-knot nematode species of the genus Meloidogyne, the mitotic parthenogens M. paranaensis, M. incognita, M. arenaria and M. javanica, and the meiotic/mitotic M. hapla (isolates A and B, respectively). The lack of distinctive mutations and the considerable contribution (40.8%) of ancestral changes disclose an ancient satellite DNA which existed in the common ancestor of extant parthenogenetic species in the same or similar form and remained preserved for a period of at least 43 My. Nonuniformly distributed polymorphic sites along the satellite monomer suggest differences in constraints acting on particular sequence segments. Sequence diversity is clearly unaffected by significant differences in genomic abundance of the MPA2 satellite DNA in the examined species. Observed results suggest that the dynamics of this satellite DNA family might be in the first instance a consequence of characteristics of its nucleotide sequence and possible constraints imposed on it. Under conditions of mitotic and meiotic parthenogenesis, slow accumulation of mutations and slow replacement of old MPA2 sequence variants with new ones may be equivalent to the dynamics of some satellite DNA sequences conserved for extremely long evolutionary periods in sexual species.  相似文献

Steroidogenesis, which plays a major role in the reproductive cycle of vertebrates, is still for the most part, unknown in invertebrates. The aim of this study was to examine the link between progesterone and the reproductive cycle in Mya arenaria. The soft-shell clams, Mya arenaria were collected in Anse à l'Orignal (Parc Provincial du Bic, Québec, Canada) from July to November 1998. Histological data have shown that female gonads of M. arenaria were in the spawning stage in August and September, while the male gonads were in the ripe stage. This period of active gametogenesis was associated with a depletion of lipid reserves. These lipids could be used as a source of energy and as a substrate for steroidogenesis. Liquid Chromatography coupled to Mass Spectroscopy (LC-MS) and quantified by Enzyme Linked ImmunoSorbent Assay (ELISA) determined progesterone. Progesterone levels in the gonad were increased during the ripe stage in the male and during the spawning stage in the female. These results indicate, for the first time, that progesterone, as in vertebrates, may play a role in the reproductive cycle of M. arenaria.  相似文献

Shells of small (< 7 mm) Littorina striata are frequently nodulose, but shells of larger individuals are striate. Nodulose L. striata dominated the littoral fringe of a black basalt Azorean shore where daytime rock temperatures rose significantly higher than nearby shores of different rock composition or colour. There was no evidence of intraspecific size-partitioning on the latter shores, where the numbers of striate and nodulose L. striata were approximately equal between high eulittoral (low-shore) and high littoral fringe (high-shore) localities. The prevalence of small L. striata in the littoral fringe is opposite to that usually characteristic of the Littorinidae, where the largest individuals usually occupy the higher positions on the shore. It is hypothesized that small L. striata attain a resting posture better able to minimize heat absorption from the substratum than attained by larger individuals. Smaller individuals also take advantage of both posture and a nodulose shell surface to re-radiate absorbed incident radiant thermal energy more effectively to the atmosphere by convection. Thus, small, nodulose L. striata are especially well adapted to occupy geologically young basaltic rocks commonly found fringing islands of the mid-Atlantic. The rate of evaporative water loss was determined for Melarhaphe neritoides and striate and nodulose L. striata for approximately 11 days emersion. All three groups are exceptionally capable of controlling evaporative water loss. Total percent evaporative water loss by nodulose L. striata (17.9%) was significantly greater than that lost by either striate L. striata (14.1%) or M. neritoides (13.5%) but, among 15 species for which evaporative water loss has been determined by similar methodology, M. neritoides and striate L. striata are the most capable of conserving body water during 11 days of emersion.  相似文献

A strict consensus tree based on chloroplast and nuclear sequences (rbcL, matK, trnL, FLint2) from 46 Amorphophallus species, two Pseudodracontium species and six outgroups is used to develop a hypothesis for the evolution of ornamentation and ectexine ultrastructure in the pollen of Amorphophallus. There are four main clades: an exclusively African, largely psilate clade (‘African clade’), an Asian, largely psilate clade (‘Asian psilate clade’) and an Asian, largely striate clade consisting of a mainly continental SE Asian clade (‘continental SE Asian striate clade’) and one centred in Malesia (‘Malesian striate clade’). Ultrastructure provides a valuable contribution towards understanding pollen ornamentation in Amorphophallus. Pollen with a thin psilate ectexine without dark granules might be plesiomorphic in Amorphophallus. Then the diverse striate type would be derived. Within both striate clades, reversals to the psilate type occur. Striate pollen with psilate caps, which is nested in the continental SE Asian striate clade, is a synapomorphy of Pseudodracontium. The fossulate type is also diverse, and its distribution in the tree indicates a polyphyletic origin. Areolate, echinate and verrucate ornamentation, occur in single species in the tree, but are found also in species not included in the molecular analysis. All three are heterogeneous and probably polyphyletic too. Reticulate, scabrate and striate/scabrate ornamentation are autapomorphies, of which the reticulate type and the striate/scabrate type may derive from psilate and striate ornamentation, respectively. Of the four main clades, the Asian psilate and African clade seem to be basal, while both striate clades might have evolved from the Asian psilate clade via a species like A. rhizomatosus. Dark granules evolved more than once, which might explain their diverse size, shape and distribution.  相似文献

The present research describes the histology of the midgut, gastric caeca, and pyloric valve of Tropidacris collaris (Stoll, 1813), (Orthopetera: Romaleidae). We used light microscopy, staining (Gomori's trichrome and periodic acid-Schiff (PAS)), and a routine histological analysis method (hematoxilin-eosin). The insects were obtained from, and also bred in, the Laboratory of Entomology, Department of Biology, of the Rural Federal University of Pernambuco (UFRPE). The collected material was fixed in alcoholic Botüin and embedded in paraplast. The results demonstrated that the midgut wall is composed of an inner epithelial layer and two outer layers of striate muscles: one internal (circular) and the other external (longitudinal), with connective tissue between the muscle fibers. The epithelium is single-layered, with two cell types: regenerative and elongated columnar. The gastric caeca presents muscle layers similar to those of the midgut. Simple columnar epithelium lines the gastric caeca, which presents villi and projects towards the lumen. The pyloric valve is of striate muscle tissue, covered by a single epithelial-cell layer.  相似文献

五十多年前胡骕在研究山东临朐县山旺组的植物化石中,发现有花的化石,仅保存了5个花瓣或仅5个萼片,均难给予确切的分类位置;六十年代我们两次去山旺野外工作,发现为数较多,同样仅保存花瓣或萼片的化石。本文研究的是80年代发现并保存较为完好的花化石。新近李凤麟详细论述了各门类化石,均认为山旺组的地质时代属中中新世。近十余年来,国际上被子植物的花化石的研究进展较快。每次花化石的发现,对研究被子植物种系发生、系统位置及在地史中的演化及演变速率等均是极重要  相似文献

Root knot nematodes are causing serious losses in protected cultivation fields in the West Mediterranean region of Turkey. Correct and confident identification of the plant parasitic nematodes is important for vegetable growing and breeding. Therefore, ninety-five populations of plant parasitic nematodes were collected from regional greenhouses. Previously described species-specific primers were used to identify Meloidogyne populations. The present study indicated that SEC-1F/SEC-1R and INCK14F-INCK14R primers for identifying of M. incognita, Fjav/Rjav and DJF/DJR primers for M. javanica and Far/Rar for M. arenaria primers can be effective tools to identify the Turkish root-knot nematode species. Dissemination ratios of the population were 64.2%, 28.4% and 7.3% for Meloidogyne incognita, M. javanica and M. arenaria, respectively. The results showed that M. incognita was the prominent root-knot nematode species in the West Mediterranean coastal areas of Turkey.  相似文献

A structure is proposed for the type II tRNA molecules containing the long variable loop and the tertiary base interactions here are compared with type I tRNAs having the short variable loop. The type II tRNAs are similar to the type I tRNAs in their tertiary base pairing interactions but differ from them generally by not having the tertiary base triples. The long variable loop, which is comprised of a helical stem and a loop at the end of it, emerges from the deep groove side of the dihydrouridine helix, and is tilted roughly 30° to the plane formed by the amino acid-pseudouridine and anticodon-dihydrouridine helices found in yeast tRNAPhe. The fact that many of the type I tRNAs also lack the full compliment of base triples suggests that the tertiary base pairs may alone suffice to sustain the tRNA fold required for its biological function. The base triples and the variable loop appear to have little functional significance. The base type at position 9 is correlated with the number of base triples and G-C base pairs in the dihydrouridine stem.  相似文献
