The complete ITS (internal transcribed spacer) region coding the ITS1, the ITS2 and the 5.8S rDNA was amplified by polymerase chain reaction from two strains of Gilbertella persicaria, six strains in the Mucoraceae (Mucor piriformis, M. rouxii, M. circinelloides, Rhizomucor miehei, R. pusillus and R. tauricus) and four strains representing three species of the Choanephoraceae (Blakeslea trispora, Choanephora infundibulifera and Poitrasia circinans). Sequences of the amplified DNA fragments were determined and analysed. G. persicaria belongs to the monogeneric family (Gilbertellaceae), however, originally it was described as Choanephora persicaria. The goal of this study was to reveal the phylogenetic relationship among fungi belonging to Gilbertellaceae, Choanephoraceae and Mucoraceae. Our results support that the "intermediate" position of this family is between Choanephoraceae and Mucoraceae.  相似文献

关于睑虎属Goniurosaurus物种的研究主要集中在新种描述和系统进化等方面,对种群密度量化的研究较少。为研究海南吊罗山国家级自然保护区海南睑虎Goniurosaurus hainanensis种群的资源现状,2019年7—9月,在保护区内选取2个不同海拔梯度的样区(A样区551~688 m和B样区289~448 m),采用标记重捕法对该物种的种群密度进行了调查,并分别采用Schnabel法和Lincoln指数法进行种群密度估算。结果显示:采用Schnabel法估算的种群密度A样区为501只/hm2,B样区为1 999只/hm2;采用Lincoln指数法估算的种群密度A样区为583只/hm2,B样区为1 403只/hm2。结果表明:该物种的种群密度随海拔升高呈下降趋势。  相似文献

Barley and white persicaria ( Polygonum lapathifolium L.) were grown in pure and mixed populations of varying density in sand culture in a glasshouse. The leaf areas and dry weights of leaves, stems and roots were determined at intervals. During the phase of vegetative growth the addition of dry matter by barley was unaffected, although tillering was reduced, by competition from dense stands of white persicaria, whereas the growth of white persicaria was reduced by low densities of barley. The decline in leaf area and reduction in root growth of barley with the onset of flowering was accompanied by increased growth of white persicaria with the production of branches from the upper axillary buds. These flowered and set abundant seed. This behaviour probably accounts for the persistence of this weed in arable rotations.
The greater competitive ability of barley may be attributed to its larger embryo, giving much larger plants at emergence. The intrinsic relative growth rate of white persicaria is as high as, or higher than, that of barley and falls more slowly with time. This advantage is not sufficient to offset the initial differences in size while barley is vegetative. The larger embryo size of barley also determines the more intense competition that exists between plants of this species than between plants of white persicaria at similar densities and times from germination. With equal weights of produce per pot (or unit area) the intensity of competition within each species appears to be similar.
It is emphasized that the course of growth with time, rather than weight-density relationships at given points of time, must be followed in order to understand the influence of density. Two functions of the relative growth rate with time are examined.  相似文献

Sexuality has been established for a culture of Gonyaulax tarnarensis Lebour (strain NEPCC–71). The addition of a thick inoculum to a nitrogen–deprived medium results in the occurrence of anisogamous sexual fusion within the first three days in the new culture. Planozygotes, large “lumpy” cells recognizable by their four flagella, may persist up to 2 wk before forming a smooth–walled, oval hypnozygote. The latter resembles cysts released asexually by ecdysis but has a slightly thicker wall. Viable cysts resembling hypnozygotes (zygotic cysts), but with reduced photosynthetic pigmentation, have been isolated from natural murine sediments in Hidden Basin, British Columbia, and a culture (strain NEPCC–254) was initiated from excysted individuals. Zygotic cysts of NEPCC–71 remained encysted in the light at 17 C for 8 wk before excysting. The presence of a ventral pen with toxicity in the latter strain indicates that the taxonomy of G. tamarensis-like organisms is still in a stale of flux and the criteria for recognition of G. excavata (Braarud) Balech as a separate species are not satisfactory as presently formulated.  相似文献

A filamentous phage was isolated from carrier strain AI-1841 of Vibrio cholerae O139 Bengal and thus was termed fs phage. The phage was measured to be approximately 1 μm in length and 6 nm in width. One end of the phage was slightly tapered and had a fibrous appendage. The plaques developed on strain AI-4450 of V. cholerae O139 were small and turbid. The phage grew in strain AI-4450 and reached a size of 108 to 109 pfu/ml at 5 hr after infection without inducing any lysis of the host bacteria. The group of phages attached on rod-shaped materials like fimbriae of this bacteria, with their fibrous appendages at the pointed end, were often found in the phage-infected culture. The anti-fimbrial serum effectively inhibited the infection of fs phage to the host strain AI-4450. We thus concluded that the phage can be adsorbed on fimbriae with a fibrous appendage on the pointed end of the phage filament.  相似文献

The genus Pectinatus is currently composed by two species, Pectinatus cerevisiiphilus and Pectinatus frisingensis , both asociated with beer spoilage. This study describes a novel isolate (strain B6) retrieved from a wastewater treatment plant collecting residues from a large number of wineries. Based on similarity analysis of 16S rRNA gene sequences, strain B6 belongs to the genus Pectinatus . Strain B6 is a strict anaerobe like other Pectinatus species and it presents non-motile, coccoid cells showing a slight oval shape. Strain B6 shows marked physiological differences with other Pectinatus species both in fatty acid composition and carbon source utilization. The most abundant fatty acids found in strain B6 were 18:1 (42.8%) and 16:0 (18.3%) representing a total of over 61% of fatty acids in this microorganism while these fatty acids represented 41.3% in P. cerevisiiphilusT and 2.4% in P. frisingensisT of their total. Fatty acid 15:0 was not significant in strain B6 and represented 28.6% and 13.3% for P. cerevisiiphilusT and P. frisingensisT, respectively. Strain B6 showed a faster growth rate and higher optimum temperature than its relatives P. cerevisiiphilus and P. frisingensis . Strain B6, P. cerevisiiphilus and P. frisingensis could be clearly differentiated by acid production tests from substrates such as esculine and gluconate, and the lack of acid production from rhamnose and fucose among others. G+C mol% content in strain B6 is 36.5%. Based on genotypic and phenotypic differences, strain B6 is proposed as a novel Pectinatus species, P. portalensis nov. sp. Both strain B6 and the two described species of Pectinatus grow on beers and wines. These results provide insights about the origin and reservoirs of Pectinatus species and spoiling alcoholic beverages.  相似文献

A strain of Xylella fastidiosa was isolated from an invasive alternative host species, porcelain berry. Its genetic relationship with strains isolated from a native alternative host, wild grape; a nonnative alternative host, mulberry; and other economically important hosts including cultivated grape, peach, plum, oak, maple and oleander was determined by using sequence analysis of the 16S–23S rRNA intergenic spacer region. Our phylogenetic analysis revealed that the porcelain berry strain is most closely related to the wild grape strain. These two strains are more closely related to the oak, peach, and plum strains than to the mulberry and oleander strains. They are separated from the maple and cultivated grape strains. Our data suggest that suppression of porcelain berry, wild grape, and mulberry in the vicinity of susceptible economically important hosts such as oak, peach, and plum may provide an important control measure for diseases caused by X. fastidiosa.  相似文献

By combined gas chromatography-mass spectrometry the gibberellin present in suspensors of heart-shaped embryos of Phaseolus coccineus has been identified as Gibberellin A1 (GA1). The amount of GA1 in 2000 suspensors (452 mg), as estimated by gas chromatography. was 4g. 海南粗榧内生真菌抗肿瘤抗菌活性的筛选   总被引:3,自引:0,他引:3  
对72株海南粗榧(Cephalotaxus hainanensis Li)内生真菌进行了抗肿瘤和抗菌活性筛选。结果显示, 有9株内生真菌至少对一种指示瘤株具有细胞毒活性, 5株内生真菌对金黄色葡萄球菌有较强的抑菌活性, 1株内生真菌对辣椒疫霉有抑制作用。这表明海南粗榧内生真菌是寻找有价值的生物活性成分的潜在资源, 其生物活性成分值得进一步研究。  相似文献

We isolated eight strains of denitrifying bacteria that reduce nitrate and nitrous oxide at pH 10 from Lake Magadi (Kenya). Despite certain differences between the strains, they are similar in terms of G+C content (66.1-68.1 mol %) and DNA-DNA homology (75-92%) and represent different morphotypes of the same species. Based on the results of partial 16S rRNA sequencing, strain Z-7398-2 was most closely related to the Halomonas campisalis isolate from Alkali Lake (USA). The DNA-DNA homology level between the tested strain and the type strain of H. campisalis 4A was 88%. These two strains were also similar phenotypically. However, the culture isolated by us was characterized by peculiar properties, such as obligate alkaliphily, which manifested itself in the culture dependence on carbonates and lack of growth at pH values below 7, a nitrous oxide-reducing capacity, and an unusual nitrate reductase that lacked molybdenum and a Mo cofactor.  相似文献

A new thermophilic sulfate-reducing bacterium isolated from the high-temperature White Tiger oil field (Vietnam) is described. Cells of the bacterium are oval (0.4-0.6 by 0.6-1.8 microns), nonmotile, non-spore-forming, and gram-negative. Growth occurs at 45 to 65 degrees C (with an optimum at 60 degrees C) at NaCl concentrations of 0 to 50 g/l. In the course of sulfate reduction, the organism can utilize lactate, pyruvate, malate, fumarate, ethanol, salts of fatty acids (formate, acetate, propionate, butyrate, caproate, palmitate), yeast extract, alanine, serine, cysteine, and H2 + CO2 (autotrophically). In addition to sulfate, the bacterium can use sulfite, thiosulfate, and elemental sulfur as electron acceptors. In the absence of electron acceptors, the bacterium can ferment pyruvate and yeast extract (a yet unrecognized capacity of sulfate reducers) with the formation of acetate and H2. The G + C content of DNA is 60.8 mol %. The level of DNA-DNA hybridization of the isolate (strain 101T) and Desulfacinum infernum (strain B alpha G1T) is as low as 34%. Analysis of the nucleotide sequence of 16S rDNA places strain 101T in the phylogenetic cluster of the Desulfacinum species within the sulfate reducer subdivision of the delta subclass of Proteobacteria. All these results allowed the bacterium studied to be described as a new species, Desulfacinum subterraneum sp. nov., with strain 101 as the type strain.  相似文献

We have studied 43 strains of the species Alcaligenes dentrificans, A. odorans, and A. faecalis. Twenty-five of them were isolated by enrichment culture on minimal medium containing an organic acid (L-malate, succinate, tartrate, adipate, or itaconate) and N2O as a respiratory electron acceptor. These constitute a single phenon with the A. dentrificans strain type and 9 other strains isolated from clinical specimens. However, strain 4 differs from the other 34 strains in 12 nutritional characters, in its ability to effect a meta cleavage of diphenols, and by the absence of tetrathionate reductase. The percentages of G + C are the following: strains isolated from soil, 66.4 +/- 1.1; collection strains, 67.0 +/- 1.3. The 5 strains of A. odorans differ from the 34 strains of A. denitrificans (not including strain 4) in their inability to denitrify nitrate and use D-saccharate, adipate, pimelate, suberate, beta-hydroxy-beta-methylglutarate meso-tartrate, azelate, and itaconate. Their percentage of G + C is much lower: 56.1 +/- 0.4. From the nutritional point of view the 3 strains of A. faecalis resemble A. dentrificans. However, they differ from the latter by their inability to grow anaerobically on NO3-, NO2-, N2O, and by a slightly lower percentage of G+ C: 64.3 +/- 0.0. The 43 strains synthesize poly-beta-hydroxybutyric acid. None of them is chemolithotrophic.  相似文献

Strains of Gracilaria foliifera (Forsskål) Børgesen isolated into culture from Ireland (where the species would appear to be introduced), England and the Atlantic coast of France were interfertile. Strains of G. foliifera from Texas and North Carolina, U.S.A. were similarly interfertile but did not cross with the European strains. The North Carolina strain was distinctly flattened in culture and superficially resembled G. foliifera from Europe whereas the Texas strain was terete or slightly flattened. Both of the American strains were interfertile with an isolate of G. tikvahiae McLa-chlan from the type locality of this species in northern Nova Scotia, Canada. It seems likely from these data that G. foliifera is not represented on the coast of North America and that G. tikvahiae is a highly morphologically variable species widely distributed from Nova Scotia south to the Caribbean. Examination of type material of Gracilaria foliifera and G. multipartita (Clemente) Harvey, together with phyto-geographical considerations, suggests that the species known as G. foliifera on eastem Atlantic masts should be known as G. rnultipartita , and that G. foliifera s. str . is probably confined to the Red, Arabian, and Indian Seas.  相似文献

Establishment of arbuscular mycorrhizal (AM) germplasm collections is complex because of the obligate biotrophic nature of AM fungi. Only a few AM species are routinely maintained in monoxenic culture with Ri T-DNA transformed roots as host. Incorporation of new AM species into this culture system is important for molecular, physiological, and taxonomical studies. Here we report for the first time the successful monoxenic culture of Gigaspora decipiens (JA2 strain) with transformed carrot (Daucus carota) roots. In vitro cultures were established from field-collected spores; sub-culture of newly in vitro formed spores was established over five successive generations for a period of 6 y. Although initial culture of field-collected spores was difficult successive sub-cultures appeared to be adapted to the in vitro growing conditions. The JA2 strain of G. decipiens completed its life cycle while maintaining its morphological characteristics, stability, and propagule viability under the monoxenic conditions over several generations. This stable and homogeneous monoxenic material obtained for G. decipiens is part of the Banco de Glomeromycota In Vitro (BGIV, http://www.bgiv.com.ar), and could facilitate morphological, physiological, and molecular analysis of this AM species.  相似文献

Anaerobic oxidation of alkanes by newly isolated denitrifying bacteria   总被引:11,自引:0,他引:11  
The capacity of denitrifying bacteria for anaerobic utilization of saturated hydrocarbons (alkanes) was investigated with n-alkanes of various chain lengths and with crude oil in enrichment cultures containing nitrate as electron acceptor. Three distinct types of denitrifying bacteria were isolated in pure culture. A strain (HxN1) with oval-shaped, nonmotile cells originated from a denitrifying enrichment culture with crude oil and was isolated with n-hexane (C6H14). Another strain (OcN1) with slender, rod-shaped, motile cells was isolated from an enrichment culture with n-octane (C8H18). A third strain (HdN1) with oval, somewhat pleomorphic, partly motile cells originated from an enrichment culture with aliphatic mineral oil and was isolated with n-hexadecane (C16H34). Cells of hexane-utilizing strain HxN1 grew homogeneously in the growth medium and did not adhere to the alkane phase, in contrast to the two other strains. Quantification of substrate consumption and cell growth revealed the capacity for complete oxidation of alkanes under strictly anoxic conditions, with nitrate being reduced to dinitrogen. A novel, aerobic, Gram-stain-negative, non-motile, non-spore forming, rod-shaped bacterium, designated strain Dol 15-39T, was isolated from a seawater sample near Geoje Island in the South Sea, Republic of Korea. The strain was found to be oxidase-negative and catalase-positive. The isolate was observed to grow at temperatures from 4 to 37°C, at salinities of up to 7%, and at pH levels from 6 to 9; moreover, it was not able to degrade starch, DNA, esculin, or tyrosine. Phylogenetic analysis based on 16S rRNA gene sequences showed that Dol 15-39T was most closely related to Flavobacterium jumunjinense HME7102T with a sequence similarity of 97.3%. However, the levels of DNA-DNA relatedness between Dol 15-39T and the most closely related species were much lower than 70%, confirming that they represented distinct genomic species. The genomic DNA G + C content of Dol 15-39T was calculated to be 32.6 mol%. MK-6 was the predominant respiratory quinine, while iso-C15:0 (25.0%), iso-C15:1 G (17.0%), and iso-C17:0 3-OH (10.4%) were the major cellular fatty acids. Phosphatidylethanolamine was identified as a major polar lipid, while various unidentified aminolipids and polar lipids were also detected. Based on polyphasic taxonomic data, Dol 15-39T represents a novel species of the genus Flavobacterium, for which the name F. aquimarinum sp. nov. is proposed. The type strain is accessible under the culture collection numbers (KEMB 9005-617T = JCM 31930T).  相似文献
