The phenomenon of cytomixis is a general occurenee in the pollen mother cells (PMCs) of certain angiosperms. Recently, we found this phenomenon also occurred in jimsonweed. According to our observations and a test of chi-square, it has been shown that the discrepancies between the patterns of intercellular chromatin migration and the variation of chromosome numbers in the PMCs of jimsonweed, as well as rye, are not significant. It means that the patterns of intercellulr chromatin migration and the variation chromosome numbers are related to each other.  相似文献

The types of opaline silica-bodies (opal phytoliths) which occurin the mature prophylls, radical and culm leaves, culms, andinflorescence bracts of rye (Secale cereale L.) are describedand figured. Silica-bodies are absent from the coleoptile, andthe adaxial epidermis of the prophylls, leaf sheaths, and inflorescencebracts. The stages of silica-body formation in young radicalleaf sheaths are also described. Alternative hypotheses forthe origin of silica-bodies are discussed.  相似文献

The pattern of opaline silica deposition in the leaves and internodesof rye (Secale cereale L.) has been studied by means of countsof silica-bodies in cleared epidermal preparations. Silica depositionoccurs during the maturation process when the leaves are fullyexpanded. The increase in total silica content and changes inthe ‘free’ and residual silica fractions of theleaves during their growth period have been determined usinga colorimetric estimation.  相似文献

Structural investigation and morphometry of meiotic chromosomes by scanning electron microscopy (in comparison to light microscopy) of all stages of condensation of meiosis I + II show remarkable differences during chromosome condensation in mitosis and meiosis I of rye (Secale cereale) with respect to initiation, mode and degree of condensation. Mitotic chromosomes condense in a linear fashion, shorten in length and increase moderately in diameter. In contrast, in meiosis I, condensation of chromosomes in length and diameter is a sigmoidal process with a retardation in zygotene and pachytene and an acceleration from diplotene to diakinesis. The basic structural components of mitotic chromosomes of rye are "parallel fibers" and "chromomeres" which become highly compacted in metaphase. Although chromosome architecture in early prophase of meiosis seems similar to mitosis in principle, there is no equivalent stage during transition to metaphase I when chromosomes condense to a much higher degree and show a characteristic "smooth" surface. No indication was found for helical winding of chromosomes either in mitosis or in meiosis. Based on measurements, we propose a mechanism for chromosome dynamics in mitosis and meiosis, which involves three individual processes: (i) aggregation of chromatin subdomains into a chromosome filament, (ii) condensation in length, which involves a progressive increase in diameter and (iii) separation of chromatids.  相似文献

The technique of mieromanipulaion has been used to establish a system of single chromosome mierodissection. According to the standard karyotype of rye ( Secale cereale L. ), 1R chromosome carrying disease-resistant gene has been identified, microdissected and transferred into Ep-pendorf tube using mieromanipulator. The results showed that IR chromosome in cells pretreated with α-bromonaphthalene could be identified quickly and the efficiency of microdissection was greatly improved when the technique of wall degradation and hypotonic treatment was applied.  相似文献

Apoplastic extracts of cold-acclimated winter rye (Secale cereale L. cv Musketeer) leaves were previously shown to exhibit antifreeze activity. The objectives of the present study were to identify and characterize individual antifreeze proteins present in the apoplastic extracts. The highest protein concentrations and antifreeze activity were obtained when the leaf apoplast was extracted with ascorbic acid and either CaCl2 or MgSO4. Seven major polypeptides were purified from these extracts by one-dimensional sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis under nonreducing conditions. The five larger polypeptides, of 19, 26, 32, 34, and 36 kD, exhibited significant levels of antifreeze activity, whereas the 11- and 13-kD polypeptides showed only weak activity. Five of these polypeptides migrated with higher apparent molecular masses on SDS gels after treatment with 0.1 M dithiothreitol, which indicated the presence of intramolecular disulfide bonds. The apparent reduction of the disulfide bonds did not eliminate antifreeze activity in four of the polypeptides that contained intramolecular disulfide bonds and exhibited significant levels of antifreeze activity. The amino acid compositions of these polypeptides were similar in that they were all relatively enriched in the residues Asp/Asn, Glu/Gln, Ser, Thr, Gly, and Ala; they all lacked His, except for the 26-kD polypeptide, and they contained up to 5% Cys residues. These polypeptides were examined with antisera to other cystine-containing antifreeze proteins from fish and insects, and no common epitopes were detected. We conclude that cold-acclimated winter rye leaves produce multiple polypeptides with antifreeze activity that appear to be distinct from antifreezes produced by fish and insects.  相似文献

To effectively use elite genes on the long arm of rye chromosome 6(the 6RL arm) in wheat breeding programs,precise and fast identification of 6RL chromatin in wheat backgrounds is necessary.PCR-based 6RL-specific markers can facilitate the detection of elite genes on 6RL in wheat breeding.However,only a limited number of 6RL-specific markers have been developed.In the present study.300 new PCR-based 6RL-specific markers were identified using specific length amplified fragment sequencing(SLAF-seq) technology,and were further physically mapped to four regions on the 6RL arm using 6R and 6RL deletion lines.Interestingly,127 of the 300 markers were physically localized to a region from the site between 2.3 and 2.5 to the telomere,the same region where the powdery mildew resistance gene was mapped.In addition,95 of the 300 markers exhibit polymorphisms,which can be used to investigate the diversity of rye 6RL arms.The markers developed in this study can be used to identify given segments of 6RL in wheat backgrounds and accelerate the utilization of elite genes on 6RL in wheat breeding.  相似文献

After mechanical spraygun inoculation barley mild mosaic virus (BaMMV) was detected in barley cv. ‘Gerbel’ (control) as well as in rye cv. ‘Somro’, but not in wheat cv. ‘Kanzler’ and oat cv. ‘Alfred’. ELISA values of infected barley and rye were similar. Furthermore, infected rye plants developed symptoms typical for barley yellow mosaic virus infection.  相似文献

Two groups of polypeptides, which together represented over90% of the secalin fraction, were purified by ion exchange chromatographyand gel filtration. These fractions had molecular weights of40 000 and 75 000 by SDS-PAGE and 33 000 and 54 000 by sedimentationequilibrium ultracentrifugation respectively. Isoelectric focusingshowed that each fraction contained a number of polypeptides.Amino acid analysis showed that the two groups had similar compositionswith high glutamate + glutamine and proline and low lysine.The N-terminal sequences of the two groups were identical at17 of the first 20 positions and were similar to those reportedfor 2 and 3-gliadins of wheat. Both groups had C-terminal histidine.These results are discussed in relation to prolamin evolutionand homology in barley, rye and wheat.  相似文献

Summary When rye embryos imbibe water they rapidly return to a condition of biochemical and structural complexity. Three stages of imbibition can be recognised: Phase I a short period (10 min) of physical wetting; Phase II a longer period (1 h) when little further imbibition occurs, followed by Phase III a continuous phase of active water uptake. The latter coincides with an increase in respiration rate and an increase both in the number of mitochondria and of cristae within them. Changes in fine structure become evident in all organelles in Phase III, after 2 h of imbibition. In the unimbibed embryo endoplasmic reticulum is present only as short crescents associated with electron lucent bodies, but in Phase III the endoplasmic reticulum proliferates to form many surrounding cirlets. After 6 h these circlets become fewer and instead the endoplasmic reticulum is seen in close association with the nuclear membrane. Concurrently incorporation of radioactive uridine and thymidine is first detectible. This suggests that the large increase in protein synthesis occurs on new ribosomes present on the reticulum associated with the nuclear membrane. For the first 6 h protein synthesis must occur either on polysomes within the dense packing of ribosomes or on these circlets of endoplasmic reticulum associated with electron lucent bodies.  相似文献

Summary Fine structural investigations of non-viable rye grains indicate recognisible abnormalities in the plasmalemma and mitochondrial membranes of the unimbibed embryo. Once such grains are wetted there is rapid and progressive disorganisation of the tissue. Biochemical studies show a reduced uptake of water, lack of respiratory activity and a failure in nucleic acid and protein synthesis. Whereas total DNA, RNA and protein levels are unchanged on loss of viability, the integrity of DNA and RNA is impaired and ribosomal RNA and soluble protein levels are reduced.  相似文献

The fragments of MADS-box genes belonging to the agamous and agamous-like structural classes were isolated by direct amplification of genomic DNA from annual rye (Secale cereale L.) and perennial rye (Secale montanum Guss.). The characterized fragments (deposited in the Genbank as the accession nos. AF332885–AF332887 and AF346894) comprise the complete sequences of exons 1 to 5 and lack corresponding introns. Their nearest homologs are the maize genes zag1 and zag5 (the Genbank accession nos. L18924 and L46398). One more agamous-like fragment isolated from annual rye (AF362364) is similar to theTaMADS12 wheat gene (AB007505). The fragment comprises exons 3–5 and contains a 105-bp insert between the exons 3 and 4; this insert does not resemble any MADS-box introns presently known. We assume that all these fragments of MADS-box genes are retropseudogenes.  相似文献

The barley (Hordeum vulgare L.) low temperature responsive geneblt14 was used as a probe, to isolate two different cognateclones (rlt1412; rlt1421) from a rye (Secale cereale L.) cDNAlibrary prepared from low temperature-treated (6°C day/2°C night) shoot meristems of the cultivar, Puma. Northernblot analysis revealed that low temperature expression of rlt1412is highest in root tissues whereas, rlt1421 shows greatest mRNAaccumulation in mature leaf tissues. There is a relationshipbetween the steady-state levels of these mRNA species and thefrost hardiness of Puma (North American cultivar) and Rhayader(UK cultivar) such that the expression ofboth genes is higherin the more frost hardy cultivar, Puma, compared with Rhayader. DNA and predicted amino acid sequence analysis indicated thatthe rye and barley clones encode small proteins with consensusN-terminal signal sequences whose biological function is atpresent unknown. A test system is presented allowing the quantitative measurement of two resistance components, number of mildew infections per cm2 and spore production. The infection frequency is obtained from segments of the first leaf. The spore production is determined spectrophotometrically using suspensions from the infected second leaf of the same plant. The assessment of resistance can be extended to later growth stages of the genotypes, thus providing information about the dynamics of mildew development and the existence of adult plant resistance. Multiple measurements of resistance (two components, several growth stages) increase the reliability of single plant evaluation and enhance the response to selection. This point is especially important in population breeding of rye.  相似文献

The development and growth of rye (Secale cereale L. cv. Rheidol)was studied in seedlings grown hydroponically in complete nutrientsolutions containing between 10 and 600 µM K+. The phyllochron(defined as the interval between the appearance of successiveleaves) was used as a developmental timescale to compare plants.The pattern of both shoot and root development was strictlyordered on a phyllochron basis and was unaffected by solutionK+ concentration, with the exception that tillers in plantsgrown at the lowest K+ concentrations were occasionally eithernot initiated or aborted at an early stage of development. However,both the rate of leaf appearance on the main stem and successivetillers and the rate of tiller appearance were slower in plantsgrown at lower K+ concentrations. The rate of leaf appearanceon the main stem was reduced to below 90% of its maximal valueat solution concentrations below about 50 µM K+. Plantrelative growth rate (RGR) was also reduced by lowering theK+ concentration of the nutrient solution and fell to below90% of its maximal value at solution concentrations below about200 µM K+. There was a complex relationship between tissueK+ concentration and the K+ concentration of the nutrient solution,which differed between leaves and root. Leaf K+ concentrationincreased steadily from about 50 µmol g-1 f. wt to about200 µmol g-1 f. wt as solution K+ concentration was increasedfrom 10 to 400 µM. In contrast, root K+ concentrationexhibited a sigmoidal dependence on solution K+ concentration,maintaining a minimal value of approximately 20 µmol g-1f. wt at concentration below 100 µM K+, then increasingprogressively to about 120 µmol g-1 f. wt at a solutionconcentration of 600 µM K+. The 'critical' leaf K+ concentration,i.e. the concentration at which either plant RGR or plant developmentwas reduced 90% of its maximal value, was 86 µmol g-1f. wt for plant RGR and 150 µmol g-1 f. wt for plant development.The 'critical' root K+ concentration was 24 µmol g-1 f.wt K+ for both RGR and development. A decline in tissue K+ concentrationbelow these thresholds reduced plant growth considerably. Molecular linkage mapping in rye (Secale cereale L.)   总被引:3,自引:0,他引:3  
A rye linkage map containing clones from rye, wheat, barley, oat and rice genomic and cDNA libraries, known-function genes and microsatellite markers, was created using an F2 population consisting of 110 F2-derived F3 families. Both co-dominant and dominant markers were added to the map. Of all probes screened, 30.8% were polymorphic, and of those polymorphic 79.3% were mapped. The current map contains 184 markers present in all seven linkage groups covering only 727.3 cM. This places a marker about every 3.96 cM on average throughout the map; however, large gaps are still present. The map contains 60 markers that have been integrated from previous rye maps. Surprisingly, no markers were placed between the centromere and C1–1RS in the short arm of 1R. The short arm of chromosome 4 also lacked an adequate number of polymorphic markers. The population showed a remarkable degree of segregation distortion (72.8%).
