On the mechanism of anaphase spindle elongation in Diatoma vulgare   总被引:19,自引:19,他引:0       下载免费PDF全文
Central spindles from five dividing cells (one metaphase, three anaphase, and one telophase) of Diatoma vulgare were reconstructed from serial sections. Each spindle is made up of two half-spindles that are composed almost entirely of polar microtubules. A small percentage of continuous microtubules and free microtubules were present in every stage except telophase. The half-spindles interdigitate at the midregion of the central spindle, forming a zone of overlap where the microtubules from one pole intermingle with those of the other. At metaphase the overlap zone is fairly extensive, but as elongation proceeds, the spindle poles move apart and the length of the overlap decreases because fewer microtubules are sufficiently long to reach from the pole to the zone of interdigitation. At telophase, only a few tubules are long enough to overlap at the midregion. Concurrent with the decrease in the length of the overlap zone is an increase in the staining density of the intermicrotubule matrix at the same region. These changes in morphology can most easily be explained by assuming zone mechanochemical interaction between microtubules in the overlap zone which results in a sliding apart of the two half-spindles.  相似文献

试验以刚开花的3年生巨峰葡萄实生苗为材料,分析其童区、转变区及成年区叶片中核酸、半乳糖醛酸和可溶性糖含量的变化.结果表明:每克鲜重叶片中RNA含量由童区至转变区急剧上升达最大值,在转变区保持在较高水平上,开花以上节位其含量降低,总核酸含量的变化也是如此;RNA/DNA比值在刚进入转变区达峰值,然后逐渐降低;半乳糖醛酸含量在童区和成年区均较低,在转变区出现最大值;非还原糖含量随阶段发育的提高而增加,在成年区大量开花部位降低,近植株顶端无花序部位复又增加;还原糖含量在进入转变区时达最大值,然后迅速降低,在转变区及成年区处于较低水平.  相似文献

The all black carrion crow ( Corvus corone corone ) and the grey and black hooded crow ( Corvus   corone cornix ) meet in a narrow hybrid zone across Europe. To evaluate the degree of genetic differentiation over the hybrid zone, we genotyped crows from the centre and edges of the zone, and from allopatric populations in northern (Scotland–Denmark–Sweden) and southern Europe (western–central northern Italy), at 18 microsatellites and at a plumage candidate gene, the MC1R gene. Allopatric and edge populations were significantly differentiated on microsatellites, and populations were isolated by distance over the hybrid zone in Italy. Single-locus analyses showed that one locus, CmeH9, differentiated populations on different sides of the zone at the same time as showing only weak separation of populations on the same side of the zone. Within the hybrid zone there was no differentiation of phenotypes at CmeH9 or at the set of microsatellites, no excess of heterozygotes among hybrids and low levels of linkage disequilibrium between markers. We did not detect any association between phenotypes and nucleotide variation at MC1R , and the two most common haplotypes occurred in very similar frequencies in carrion and hooded crows. That we found a similar degree of genetic differentiation between allopatric and edge populations irrespectively of their location in relation to the hybrid zone, no differentiation between phenotypes within the hybrid zone, and neither heterozygote excess nor consistent linkage disequilibrium in the hybrid zone, is striking considering that carrion and hooded crows are phenotypically distinct and sometimes recognised as separate species.  相似文献

This study was designed to study the time course of the incorporation of 55Fe into the ameloblasts of maturation in rat incisors. Male Sherman rats (100 +/- 5 gm) were injected intravenously with 0.9 mCi of 55Fe and sacrificed in pairs by perfusion at various time intervals from 5 min to 7 days after injection. The incisors were demineralized in 4.13% disodium EDTA, postfixed in 1% osmium tetroxide in veronal acetate buffer, and embedded in Epon. Incisors from control rats injected with only physiological saline were treated in the same way. Sections from blocks of tissue in the zone of maturation were prepared for light microscope radioautographic observations. The greatest incorporation of iron occurred at 9 mm within the zone; at this site the ameloblasts contained few pigment granules. About 5 mm deeper into the zone the activity fell off to zero, as observed at 2.5 hr after injection of 55Fe. Between 1 day and 7 days after injection the 55Fe labeling was found over the cells containing many pigment granules, while the initial labeling over the cells within 9 mm of the zone had diminished. These data have shown that at any given time, from 30 min to 4 hr, the iron enters the maturation ameloblasts over a wide extent of the zone, reaching a maximum at about 9 mm from the onset of maturation. However, at longer times (1 day to 7 days) the labeling curve shifts and shows the greatest activity beyond 9 mm within the zone.  相似文献

Specimens of articular cartilage from the superficial and mid-depth zones of the human femoral head and the talus of the ankle joint were tested in tension in planes parallel to the articular surface and parallel to the predominant orientation of the superficial collagen fibrils. The tensile fracture stress of cartilage from both the superficial and mid-depth zones of the femoral head decreased considerably with age. The superficial zone decreased from 33 MPa at 7 years to 10 MPa by the age of 90 years, while the mid-depth zone decreased from 32 MPa at 7 years to 2 MPa by the age of 85 years. In contrast the fracture stress of both levels of cartilage from the talus of the ankle did not decrease significantly with increasing age. The tensile stiffness at 10 MPa of both the superficial and mid-depth zones of the femoral head decreased with age. That of the superficial zone decreased from 150 MPa at 7 years to 80 MPa at 90 years, while the mid-depth zone decreased from 60 MPa at 7 years to 10 MPa at 60 years. The stiffness of talar cartilage from the superficial zone decreased by 20%, while that of the mid-depth zone showed a slight increase in stiffness at 10 MPa with increasing age. There was no significant decrease in the tensile stiffness at 1 MPa with age for either the femoral head or talar cartilage. Based on the results of previous studies it is possible to conclude that the decrease in tensile properties seen in the femoral head results from a deterioration in the tensile properties of the network of collagen fibrils. It is suggested that progressive fatigue failure, perhaps with associated changes in the structure of cartilage due to altered chondrocyte metabolism, causes the reduction in tensile properties with age. The results offer a potential explanation for the observation that osteoarthritis commonly occurs in the hip and knee joints at an increasing incidence as age increases, while the condition only rarely occurs in the ankle joint except as a secondary event to trauma.  相似文献

The adoral ciliary zone of Cycloposthium spp., inhabiting the large intestine of the horse, was studied by scanning electron microscopy. It could be divided into four parts: outer, inner, left, and right zones. The outer zone, extending on the anterior periphery of the apical cone of the body, had 20 tuft-like syncilia arranged radially around the longitudinal axis. Each ciliary tuft consisted of about 170 cilia, and in cross section it had a rectangular shape. The cilia of the inner zone, situated at the top of the apical cone, were aggregated irregularly to form shorter bundles than the tufts of the outer zone. The innermost cilia of this zone were shorter than the outermost. There was a distinct non-ciliated border between the outer and inner zones. A horseshoe-like operculum having no cilia was present at the center of the adoral ciliary zone, and the opening of the vestibulum was situated as a cleft crossing from the center to the right periphery of this zone. No cilia extended onto the vestibular wall. The left ciliary zone was situated beneath the outer zone and consisted of five short rows of barren kinetosomes of which only the central row possessed very short cilia. The right ciliary zone, consisting of a few rows of cilia situated at the bottom of the inner adoral lip, was also easily distinguished from the other ciliary zones. This zone was interpreted as an extension of the outer adoral zone passing along the right side of the apical cone. These ciliary zones were considered to be highly differentiated and useful for both movement of and ingestion of food.  相似文献

Sediment core samples were obtained at a groundwater study site in Oklahoma in January and June 1985. Most-probable-number estimates showed that protozoan numbers declined steeply with depth in subsoil. Flagellates and amoebae dominated the protozoan population, which declined to a most probable number of 28 . g (dry weight) in a clay loam layer at the bottom of the unsaturated zone. Samples from a texturally variable interface zone between 3 and 4 m down also were variable in their content of protozoa. Four contiguous clay loam samples in a single core from this zone contained variable numbers of amoebae ranging from 0.2 to 44 . g (dry weight). However, a sandy clay loam layer at the bottom of the core contained a mixture of flagellates and amoebae with a combined population density of 67 . g (dry weight). A slow-growing filose amoeba was isolated from interface zone samples and was tentatively classified in a new family in the order Aconchulinida. Protozoa were not detected in the saturated zone except in a very permeable gravelly, loamy sand layer at a depth of approximately 7.5 m. Low numbers (4 to 6 . g [dry weight]) of surface-type flagellates and amoebae, as well as the filose amoeba seen in the interface zone, were observed in this layer. Acid-treated and untreated samples contained equivalent numbers of protozoa, showing that the majority of protozoa in the layer at 7.5 m and the interface zone samples were encysted. Increased numbers of bacteria also were found in the layer at 7.5 m, indicating that it was biologically more active than other saturated-zone layers. Cyanobacteria grew in illuminated samples from this layer, suggesting that it may be connected hydrologically to a nearby river.  相似文献

The development of the stratified squamous epithelium of the tympanic membrane and external auditory canal was studied in serial sections of 124 mouse ears aged from 11 gestational days to 100 days. A fold developed from the edge of the fundus of the primary canal. It possessed two regions: firstly the meatal plate, which produced the pars tensa-covering epithelium (zone 2) and most of the deep ear canal epithelium (zone 3), and secondly the fundal extension plate, which grew from that part of the fundus not forming the meatal plate. The fundal extension plate gave rise to the pars flaccida-covering epithelium (zone 1) and also to the adjacent deep canal epithelium (zone 3). A difference from human development was that zone 3 in the mouse, in both the meatal plate- and the fundal extension plate-derived areas, formed adnexal structures. In the early development of the meatal plate, zone 3, at its tip, was swollen and actively mitotic and extended always for a short distance on to the zone 2 side. Zone 2, first perceived two days after zone 3, became progressively attenuated, and by the fourth day after its formation was a single thin layer. It is suggested that the proximal part of zone 3, situated in the mature ear around the periphery of the tympanic membrane, is a generation center for unidirectional outward flux of epithelium which terminates in the mouse at the first adnexal structure. It may cause the whole of zone 2 to move in the same direction by negative contact inhibition.  相似文献

The pollen analytical investigations of hanging peat bogs in the summit zone of the Polish east Carpathians show that the absence of the upper montane spruce zone in the western Bieszczady mountains is a phenomenon that has been historically stable at least for the last millennium and probably also in earlier times. At altitudes corresponding to the spruce zone in other parts of the Carpathians, dwarf beech woods form here a low-lying upper tree limit. The montane grassland zone occurring above this tree line did not result from human activities, although it has been extended by human activity. Besides, it is shown how far it is possible to reconstruct from a pollen diagram the differentiation of vegetation surrounding the site of profile collection located in the upper montane zone when the present-day local plant communities are well known, in spite of pollen from long distance transport always being present at the summits of mountain ranges.  相似文献
