Abstract. The use of three techniques for determining yield turgor in excised Salix leaves was investigated. These were the osmotic-solutions technique, the psychrometer technique, and the pressure-chamber technique. The application of the osmotic-solutions technique to a range of leaf types was discussed and the appropriate corrections for volume changes and the contribution of apoplastic water were detailed. It was concluded that the osmotic-solutions technique is not satisfactory for use with slowly growing and/or very elastic leaves. The psychrometer and pressure-chamber techniques were both simple compared with the osmotic-solutions technique, and gave values for yield turgor in the range of 0·3–0·5 MPa. A disadvantage of the psychrometer technique for field applications is that it requires one psychrometer chamber per sample. The pressure-chamber technique was modified for use as a field technique where multiple sampling could be easily and inexpensively achieved. Particular care was required with this technique to prevent water loss from the leaf during stress relaxation, but simple and effective procedures for doing so were found. The modified pressure-chamber technique described here, is recommended as the preferred technique for measuring the yield turgor of leaves in experiments where many simultaneous estimates of yield turgor are to be made.  相似文献

A simple and rapid technique for assaying toxicants in aphids   总被引:2,自引:0,他引:2  
A simple bioassay technique has been developed in order to study the effect of toxic compounds on aphids. The technique involves dipping of leaves of a particular aphid host in a solution of the desired toxic compound contained in a vacuum flask. The flask is then connected to a vacuum source in order to remove the air from the intercellular spaces of the palisade and the spongy parenchymas of the leaves. Breakage of the vacuum and return to atmospheric pressure forces the test solution into the now empty intercellular spaces which are thus filled with the solution. The treated leaves exhibited a typical water-soaked appearance. The infiltrated leaves were then offered to the aphids. The above treatment did not affect either the structure of the leaves or the development of the aphid colony on them. Aphids grown on leaves infiltrated with water lived longer in comparison with controls. In addition, insects grown on leaves infiltrated with 27.5, 13.7 and 6.9 p.p.m. of the systemic insecticide heptenophos died after 1.5, 3.0 and 5.0 h, respectively. The bioassay technique described in this report has certain advantages: it is simple, fast and inexpensive and is especially suitable for short-term studies involving the effects of such chemicals as toxins, insecticides and other toxicants on aphids. It may also be used to study the effects of growth factors, hormones and special nutrients on aphid growth and possibly on other sucking insects.  相似文献

The presence of microbial biofilms in the phyllosphere of terrestrial plants has recently been demonstrated, but few techniques to study biofilms associated with living plant tissues are available. Here we report a technique to estimate the proportion of the bacterial population on leaves that is assembled in biofilms and to quantitatively isolate bacteria from the biofilm and nonbiofilm (solitary) components of phyllosphere microbial communities. This technique is based on removal of bacteria from leaves by gentle washing, separation of biofilm and solitary bacteria by filtration, and disintegration of biofilms by ultrasonication. The filters used for this technique were evaluated for their nonspecific retention rates of solitary bacteria and for the efficiency of filtration for different concentrations of solitary bacteria in the presence of biofilms and other particles. The lethality and efficiency of disintegration of the sonication conditions used here were also evaluated. Isolation and quantification of bacteria by this technique is based on use of culture media. However, oligonucleotide probes, sera, or epifluorescent stains could also be used for direct characterization of the biofilm and solitary bacteria in the suspensions generated by this technique. Preliminary results from estimates of biofilm abundance in phyllosphere communities show that bacteria in biofilms constitute between about 10 and 40% of the total bacterial population on broad-leaf endive and parsley leaves.  相似文献

Background and Aims

The hydraulic architecture and water relations of fruits and leaves of Capsicum frutescens were measured before and during the fruiting phase in order to estimate the eventual impact of xylem cavitation and embolism on the hydraulic isolation of fruits and leaves before maturation/abscission.

Methods

Measurements were performed at three different growth stages: (1) actively growing plants with some flowers before anthesis (GS1), (2) plants with about 50 % fully expanded leaves and immature fruits (GS2) and (3) plants with mature fruits and senescing basal leaves (GS3). Leaf conductance to water vapour as well as leaf and fruit water potential were measured. Hydraulic measurements were made using both the high-pressure flow meter (HPFM) and the vacuum chamber (VC) technique.

Key Results

The hydraulic architecture of hot pepper plants during the fruiting phase was clearly addressed to favour water supply to growing fruits. Hydraulic measurements revealed that leaves of GS1 plants as well as leaves and fruit peduncles of GS2 plants were free from significant xylem embolism. Substantial increases in leaf petiole and fruit peduncle resistivity were recorded in GS3 plants irrespective of the hydraulic technique used. The higher fraction of resistivity measured using the VC technique compared with the HPFM technique was apparently due to conduit embolism.

Conclusions

The present study is the first to look at the hydraulics of leaves and fruits during growth and maturation through direct, simultaneous measurements of water status and xylem efficiency of both plant regions at different hours of the day.  相似文献

A microscale technique has been developed for routine quantifications of picogram amounts of indole-3-acetic acid (IAA) in plant tissues by combined gas chromatography-mass spectrometry. Low- and high-resolution selected-ion-monitoring and selected-reaction-monitoring mass spectrometry techniques were compared for selectivity and precision. The best selectivity was obtained with selected-reaction-monitoring analysis, and 1-mg samples containing 500 fg of IAA could be analyzed accurately with this method. This technique was used to investigate the IAA distribution pattern along the longitudinal axis of tobacco (Nicotiana tabacum [L.]) leaves. In young, developing leaves an increase of endogenous IAA from the leaf tip to the base of the leaf was observed, whereas the level of IAA was uniform along this axis in mature leaves.  相似文献

Accurate information on the optical properties (reflectance and transmittance spectra) of single leaves is important for an ecophysiological understanding of light use by leaves, radiative transfer models and remote sensing of terrestrial ecosystems. In general, leaf optical properties are measured with an integrating sphere and a spectroradiometer. However, this method is usually difficult to use with grass leaves and conifer needles because they are too narrow to cover the sample port of a typical integrating sphere. Although ways to measure the optical properties of narrow leaves have been suggested, they have problems. We propose a new measurement protocol and calculation algorithms. The protocol does not damage sample leaves and is valid for various types of leaves, including green and senescent. We tested our technique with leaves of Aucuba japonica, an evergreen broadleaved shrub, and compared the spectral data of whole leaves and narrow strips of the leaves. The reflectance and transmittance of the strips matched those of the whole leaves, indicating that our technique can accurately estimate the optical properties of narrow leaves. Tests of conifer needles confirmed the applicability.  相似文献

When intact plants of Xanthium strumarium L. were water stressed, the youngest leaves accumulated the highest levels of abscisic acid (ABA). On the other hand, when leaves of different ages were detached and then stressed, the capacity to produce ABA was highest in the mature leaves. Radioactive ABA was transported from mature leaves to the shoot tips and young leaves, as well as to the roots, as evidenced by the presence of radioactive ABA and phaseic acid in the xylem exudate coming from the roots. Thus, ABA was recirculated in the plant, moving down the stem in the phloem and back up in the transpiration stream to the mature leaves. Phloem exudate collected by the use of the EDTA technique had a high concentration of ABA and phaseic acid which increased several-fold after water stress. The high ABA levels in immature leaves and apical buds are, therefore, mainly due to import from older leaves, rather than to in situ synthesis.  相似文献

Hydraulic resistances for whole sunflower plants (Helianthusannuus L.) and sunflower leaves, stems, petioles and roots weremeasured. Whole plant hydraulic resistance was shown to declinewith an increase in transpiration. Leaf hydraulic resistancewas shown, with one technique employing transpiring leaves,to vary with transpiration and with another technique, employingpressure-induced flow in leaves, to be constant over a widerange of transpiration. Stem and petiole hydraulic resistanceswere constant over a wide range of exudation. Pressure-inducedflow through root systems was shown to be an inappropriate methodfor characterizing their hydraulic properties because flow mayoccur through unnatural paths. The technique employing measuredtranspiration rates and water potentials of non-growing leavesand soil is suggested to be better. 利用叶绿素荧光分析技术和高效液相色谱研究了链霉素(SM,叶绿体基因编码蛋白的抑制剂)处理玉米叶片的叶黄素循环及依赖叶黄素循环的热耗散。与对照相比,强光下SM处理叶片的最大光化学效率(Fv/Fm)降低且不能完全恢复,同时电子传递速率(ETR)显著下降。而且,SM处理叶片的非光化学淬灭(NPQ)和叶黄素循环的脱环氧化水平增加。但是,NPQ的主要组分高能态(qE)淬灭减小。因此,推测qE的降低可能与电子传递速率降低有关。  相似文献

An effective and simple technique to obtain conidial inoculum of Peronosclerospora sorghi has been devised, which confers many advantages over the conventional method. Systemically infected sorghum leaves were packed into folds of paper-towel. The end of folded paper-towel containing cut-ends of leaves was dipped in 3?cm deep water in a container. The container was exposed to moist field conditions at night and sporulated leaves were harvested with chilled distilled water in the morning to obtain the inoculum. Advantages of the technique include lower cost and effort, rapid harvesting and daily production of inoculum for up to seven consecutive days from same leaves. Though conventional method of plating leaf pieces in Petri plates proves superior in terms of the amount of sporulation, paper-towel method has the edge under working conditions.  相似文献

The phloem exudation technique using ethylenediaminetetraacetic acid (EDTA) was evaluated in studies of amino acid translocation in Pisum sativum L. seedlings. Exudation of phloem sap from cut petioles of fully expanded leaves was enhanced by EDTA (20 millimolar disodium salt [pH 7.0]). Amino acids (mainly asparagine, homoserine, glutamate, and also aspartate and serine) were present in petiole exudates from EDTA-treated leaves at levels which were commonly 5- to 10-fold (or more) higher compared with water-treated controls. Exudation was greater from darkened leaves, and the pattern of amino acids was markedly different from the more uniform mixture leaking from water-treated controls.  相似文献

A new imaging technique is described which uses nuclear magnetic resonance (NMR) to create a water profile of plant leaves. The water profile shows the average distribution of water as a function of depth into a leaf along a line perpendicular to the leaf surface; it can be used to measure the thickness of cell layers and the quantity of water in each layer. Two-dimensional NMR methods were used to avoid chemical shift distortion which degrades the resolution in leaf images made by conventional NMR techniques; image resolution was improved further by deconvolution analysis. To illustrate its application, the technique was used to follow changes in the internal structure of developing leaves.  相似文献

We evaluated a new, two-dimensional (2-D) nuclear magnetic resonance (NMR) imaging technique as a method for measuring the distribution of chloroplasts in leaves. NMR images that showed the distribution of chloroplast water and of total water as a function of depth into Acer platanoides sun and shade leaves were compared with the distribution of chlorophyll in the same leaf types (as measured by fluorescence microscopy), with the cellular structure (by scanning electron microscopy), and with published information. Results showed that the volume fraction of chloroplast water was much larger in shade than in sun leaves, and that it averaged about one-third larger in the palisade than in the spongy parenchyma region of both leaf types. Chlorophyll fluorescence was more intense in shade than in sun leaves. In sun leaves, fluorescence was maximal in the palisade region near the junction with the spongy parenchyma, while in shade leaves, fluorescence was maximal in the upper part of the spongy layer. We concluded that 2-D NMR imaging reliably indicates the location of chloroplast water.  相似文献

Analysis of virus spread using co-expressed reporter proteins has provided important details on cell-to-cell and long-distance movement of viruses in plants. However, most viruses cannot tolerate insertion of large non-viral segments or loss of any open-reading frames, procedures required to detect viruses non-evasively. A technique used to localize mRNAs intracellularly in yeast has been modified for detection of viral RNAs in whole plants. The technique makes use of the binding of the coat protein of MS2 bacteriophage (CPMS2) to a 19 base hairpin (hp). A fusion protein, consisting of the CPMS2, green fluorescent protein (GFP), and a nuclear localization signal (NLS), was nuclear-localized upon transient expression in protoplasts. However, addition of the hp to the 3' untranslated region of Turnip crinkle virus (TCV-hp) and co-transfection of the virus and fusion protein construct into protoplasts resulted in the re-location of GFP to the cytoplasm. Neither the insertion of the hp nor the interaction with the fusion protein impaired any viral functions. Transgenic plants expressing the GFP-NLS-CPMS2 fusion protein were generated, and GFP was detected in nuclei of young plant cells. Foci of GFP cytoplasmic fluorescence were detected in TCV-hp-inoculated leaves at 2 days post-inoculation. Later, GFP was detected in young leaves near the midvein and in the base (support) cells of trichomes in the vicinity of secondary and tertiary veins. In older leaves, cytoplasmic GFP could be visualized throughout many of the leaves. This technique should be amenable for detection of any virus with a transformable plant (or animal) host and may also prove useful for localizing properly engineered host RNAs.  相似文献

A polyspecific antiserum against protein extracted from PSTV-infected tomato leaves was prepared and the IgGs were separated by affinity chromatography on a beaded cellulose adsorbent with an immobilized “healthy” antigen. The antibody not adsorbed entered into a preferential reaction with the antigen from PSTV-infected leaves as estimated by an enzyme-linked immunosorbent assay. The immunochemical reactions did not significantly exceed the control background, if antigens from tomato leaves infected with potato viruses X, Y and M were analyzed. By immunoblot technique we revealed, however, that several antigens not detected in healthy leaves appeared in the leaves infected either with PSTV or with viruses X and M. An accumulation of a major antigen having a molecular mass of about 70 kDa was observed in viroid-infected leaves only, suggesting the specificity for viroid infection. The antigen was found not to be an alkaline endoproteinase - the pathogenesis-related protein P-69. Some antigens with molecular masses approximately 38.0, 23.7 and 22 kDa, which occurred in PSTV-infected leaves and in healthy calluses, were not detectable in PSTV-infected calluses. No reaction exceeding the control level was observed using enzyme-linked immunosorbent assay for antigens from silver nitrate-treated tomato leaves, although such leaves showed symptoms similar to that caused by viroids.  相似文献

金叶马褂木是从当地金边马褂木中发现的春季至初夏间全叶呈金黄色至淡黄色的芽变材料。对该芽变种质的叶片特性进行了连续8年的观察与调查,同时结合RAPD技术进行了初步的分子鉴定。叶片特性观察与调查结果表明,金叶马褂木叶片大小与金边马褂木、北美马褂木差异不大,但叶片叶绿素含量显著少于金边马褂木和北美马褂木,叶片色泽也与金边马褂木有显著性差异。结合RAPD分子鉴定结果,认为金叶马褂木是一个发生变异的新种质。  相似文献

以口红吊兰叶片为试材,用生物超微弱发光探测技术研究衰老过程中整体叶片与部分被遮挡叶片延迟发光的变化.结果表明:光照整体叶片与光照部分叶片延迟发光的曲线平行,而整体叶片与被遮挡部分叶片延迟发光的规律不同;幼叶与老叶整体延迟发光衰减快,被遮挡部分延迟发光衰减慢;成熟叶片整体延迟发光衰减慢,而被遮挡部分的延迟发光衰减快.研究认为:在同一外部条件下,整体叶片与被遮挡部分叶片延迟发光变化规律不同,植物叶片的这种发光现象可能在生物系统内部有自己独特的规律.  相似文献

P700 absorption change signals were measured at 800 nm for plant leaves and plant leaf extracts by illuminating with far-red light. The ratio of the signal amplitudes for the two sample types shows the degree of optical signal intensification. The intensification is due to optical path-length elongation, itself caused by scattering. The intensification was found to be between 6 and 15 for different leaves. For spinach leaves, the intensification varied less than 10% among samples. By replacing the air in the spongy layer of a leaf with an isotonic solution it was shown that nearly half of the intensification is due to the scattering at air-tissue interfaces. The comparison of the P700 signals of a leaf with its extract would seem to be a new technique for determining the optical scattering effects of leaves.  相似文献

Physiological and biochemical studies on the leaf apoplast have been facilitated by the use of the infiltration-centrifugation technique to collect intercellular washing fluid (IWF). However, this technique has been difficult to implement in rice (Oryza sativa L.) for various reasons. We compared the collection efficiency of leaf IWF between two types of rice varieties (Indica and Japonica), as well as between rice and other species (spinach, snap bean and wheat). Although the extraction of IWF in most species took only 2-3 min, it took up to 35 min in rice. The difficulty in infiltration with rice was ascribed to the small stomatal aperture and hydrophobicity of the leaves. In this study, we have established an improved method for collecting IWF and determining the apoplastic air and water volumes in rice leaves. We have shortened the infiltration time to 8 min via the following improvements: (i) infiltration under outdoor shade in the daytime to prevent stomatal closure and a rise in temperature of the infiltration medium; (ii) soaking of leaves in a surfactant solution to decrease the leaf hydrophobicity; and (iii) continuous pressurization using a sealant injector to facilitate the infiltration. The rapid collection of IWF achieved using this technique will facilitate study of the leaf apoplast in rice.  相似文献
