To elucidate the basic food requirement of spiders, the important polyphagous predators of rice-plant insect pests, an attempt was made to measure the respiratory energy loss of fasting spiders, Lycosa pseudoannulata. Relationship between fresh (y) and dry (x) weights of spiders inhabiting the bottom layer of the rice-plant community was represented by the following allometric equation:y=0.428x0.872. The carbon dioxide production by previously fed and unfed females under the dark at 29°C 100% R. H. was measured by a titration technique. The relationship between fresh body weight and CO2 production by unfed animals could be represented by the equation M=aWb, M being the CO2 output per individual per day and W the fresh body weight. The constant b, which determines the slope of curve, was 0.808. Respiration of the adult female with 100 mg fresh weight was 1.155±0.250 mg CO2/100 g fresh weight/day or 48.69 mg CO2/g dry weight/day. This value corresponds to 35.81 cal/g fresh weight/day or 150.94 cal/g dry weight/day. Supposing the calorific content of spiders to be 5820 cal/g dry weight, rate of the respiratory energy loss to total energy of the body was estimated to be 2.60%. This rate did not strongly contradict with the loss of fresh body weight before and after the measurement. The metabolic rate showed remarkable fluctuation with changing food supply. The CO2 production of starved individuals decreased to 83.63±16.34% as compared with individuals which were fed before the measurement.  相似文献

Chlordecone greatly potentiates carbon tetrachloride (CC14) hepatotoxicity. In order to quantitate the degree of this potentiation, the effects of a range of doses of CC14 on two microsomal enzymatic functions and liver enzyme release were examined in chlordecone-treated and control rats. Male Sprague-Dawley rats were pretreated with 15 mg chlordecone per kilogram body weight (BW) intragastrically or with vehicle. After 48 hours, 0 to 250 μ1 CC14 per 100 g body weight were given intraperitoneally (IP), and the rats were killed 24 hours later. Chlordecone treatment produced approximately a 17-fold potentiation of the CC14 dependent loss of cytochrome P-450 and glucose-6-phosphatase activity, so that a dose of 6 μ1 CC14 per 100 g body weight in the chlordecone-treated animals resulted in a similar amount of damage as observed with 100 μ1 CC14 per 100 g body weight in controls. A similar potentiation by chlordecone was seen with CC14- induced increases in serum glutamic-oxaloacetic transaminase (SGOT) levels. Chlordecone treatment also increased hepatic cytochrome P-450 levels by 67% and resulted in an increase in the covalent binding of [14-C]-CC14-derived metabolites to microsomal protein and lipid in vivo.  相似文献

It was shown that rRNA and HnRNA synthesis in rat liver nuclei does not change-within 30 min after intraperitoneal injection of acetylcholine (0.005 mg per 100 g of body weight) but decreases after injection of norepinephrine and epinephrine (0.05 mg per 100 g of body weight). The synthesis of rRNA (but not of HnRNA) increases after injection of hydrocortisone (2,5 mg per 100 g of body weight). The synthesis of HnRNA (but not of rRNA) increases after injection of ACTH1-24 (3 ME per 100 g of body weight) and oxytocin (1 ME per 100 g of body weight). The synthesis of rRNA decreases after injection of propranolol and atropine (0.5 mg per 100 g of body weight). At the same time, the synthesis of HnRNA does not change thereby. The inhibitory effect of propranolol and atropine was corrected by electrostimulation of hypothalamus. The content of cAMP and Ca2+ and the phosphorylation degree of nuclear proteins are increased after stimulation of hypothalamus. The phosphorylation of nuclear proteins is increased by 10(-8)-10(-6) M cAMP. The synthesis of RNA in liver nuclei is increased by 10(-6) M cAMP only after addition of cytosol. In this case the activity of RNA-polymerase II increases in a greater degree than that of RNA-polymerase I + III. It is assumed that the regulatory mechanisms of rRNA and HnRNA synthesis are different. The role of hypothalamus electrostimulation, neurotransmitters, hormones, and cAMP in the mechanisms of RNA synthesis in rat liver nuclei is discussed.  相似文献

Administration of 100 μg of testosterone (T) daily for 14 and 28 days to 7-day castrate rats restored the weight of the ventral prostate to a level which slightly exceeded that of the controls. Ventral prostate weight in groups receiving estradiol-17β (E2) doses of 10, 50, 100, 200, or 500 μg administered simultaneously with 100 μg of T did not differ significantly from intact controls, although the weights were lower at E2 levels greater than 100 μg. Body weights of the castrated rats receiving 100 μg of T did not differ from those of sham castrated controls. However, mean body weights of all groups which received E2 (10 to 500 μg) simultaneously with 100 μg of T were significantly less than (p< .025 or less) those of the sham castrated controls. Analysis of normalized ventral prostate weights, i.e., mg ventral prostate/100 gm body weight, showed that E2 does not antagonize T and revealed a trend which suggested that low levels of E2 (10, 50 and 100 μg) may have enhanced the restorative effects of 100 μg T. Our data indicate that 100 μg of T approaches a physiological dosage for castrated rats and that in contrast to the possible enhancement of its restorative effects on the ventral prostate by low leve E2, its body weight stimulating effects are clearly impaired by E2.  相似文献

The morphological state of the internal organs as well the changes of microsomal oxidation in liver of white rats exposed to the action of sodium sulfate in doses 200 and 600 mg per 100 g of body weight have been studied. The sodium sulfate in dose 600 mg per 100 g of body weight has been found to decrease the cytochrome P450 content, increase its inactivation rate and have an injurious effect on the membranes of hepatocytes. Sodium sulfate administration through the gastroenteric tract causes the destructive and pathological histochemical changes in liver, stomach, duodenum and small intestine. The alternative changes are expressed most of all in duodenum and small intestine.  相似文献

The widespread use of sodium nitrite (NaNO2) for various industrial purposes has increased human exposure to alarmingly high levels of nitrate/nitrite. Because NaNO 2 is a strong oxidizing agent, induction of oxidative stress is one of the mechanisms by which it can exert toxicity in humans and animals. We have investigated the possible protection offered by carnosine (CAR) and N-acetylcysteine (NAC) against NaNO 2-induced nephrotoxicity in rats. Animals orally received CAR at 100 mg/kg body weight/d for seven days or NAC at 100 mg/kg body weight/d for five days followed by a single oral dose of NaNO 2 at 60 mg/kg body weight. The rats were killed after 24 hours, and the kidneys were removed and processed for various analyses. NaNO 2 induced oxidative stress in kidneys, as shown by the decreased activities of antioxidant defense, brush border membrane, and metabolic enzymes. DNA-protein crosslinking and DNA fragmentation were also observed. CAR/NAC pretreatment significantly protected the kidney against these biochemical alterations. Histological studies supported these findings, showing kidney damage in NaNO 2-treated animals and reduced tissue impairment in the combination groups. The protection offered by CAR and NAC against NaNO 2-induced damage, and their nontoxic nature, makes them potential therapeutic agents against nitrite-induced nephrotoxicity.  相似文献

Four rumen fistulated Suffolk wethers were allocated to a 4×4 Latin square designed experiment. High nitrate (1.5 g NaNO3 kg−0.75 body weight), high nitrate with high L-cysteine (0.55 g sulphur equivalent kg−0.75 body weight), low nitrate (0.75 g NaNO3 kg−0.75 body weight), and low nitrate with low L-cysteine (0.275 g sulphur equivalent kg−0.75 body weight) were administered into the rumen through fistulae as a single dose after a morning meal. Gaseous exchanges were monitored by an open circuit respiratory system using a hood over the animal's head. High or low L-cysteine remarkably decreased nitrite production from ruminal reduction of high or low nitrate. Consequently, methaemoglobin formation was suppressed by L-cysteine in both levels of nitrate. Oxygen consumption, carbon dioxide production and metabolic rate were depressed as methaemoglobin was formed. L-cysteine suppressed the pulmonary dysfunction induced by methaemoglobin. L-cysteine equivalent to 60% of the upper allowance of dietary sulphur appeared to be useful as a prophylactic for acute poisoning of nitrate. Thus, dosage of L-cysteine can be adjusted to correspond with the nitrate content in feeds.  相似文献

After crude protein of the marine yeast strains maintained in this laboratory was estimated by the method of Kjehldahl, we found that the G7a strain which was identified to be a strain of Cryptococcus aureus according to the routine identification and molecular methods contained high level of protein and could grow on a wide range of carbon sources. The optimal medium for single-cell protein production was seawater containing 6.0 g of wet weight of Jerusalem artichoke extract per 100 ml of medium and 4.0 g of the hydrolysate of soybean meal per 100 ml of medium, while the optimal conditions for single-cell protein production were pH 5.0 and 28.0°C. After fermentation for 56 h, 10.1 g of cell dry weight per liter of medium and 53.0 g of crude protein per 100 g of cell dry weight (5.4 g/l of medium) were achieved, leaving 0.05 g of reducing sugar per 100 ml of medium and 0.072 g of total sugar per 100 ml of medium total sugar in the fermented medium. The yeast strain only contained 2.1 g of nucleic acid per 100 g of cell dry weight, but its cells contained a large amount of C16:0 (19.0%), C18:0 (46.3%), and C18:1 (33.3%) fatty acids and had a large amount of essential amino acids, especially lysine (12.6%) and leucine (9.1%), and vitamin C (2.2 mg per 100 g of cell dry weight). These results show that the new marine yeast strain was suitable for single-cell protein production.  相似文献

Advantage was taken of a specific and sensitive bioassay for rat plasma adrenocorticotropin (ACTH) based on the dispersion of rat adrenal cells with trysin, to investigate the relationship between plasma corticosterone concentration and inhibition of ACTH release under steady-state conditions achieved by graded rates (0-5.12 mug/min per 100 g body weight) of intravenous infusion of the steroid for 45 min in 28-day adrenalectomized rats. In contrast to prior reports involving suppression of stress-induced ACTH release, the inhibitory effect of corticosterone was shown, under our experimental conditions, to be exerted also on the basal rate of ACTH secretion. Indeed, a slight though not significant decrease of plasma ACTH concentration was observed with the corticosterone infusion rate of 0.64 mg/min per 100 g body weight, and further progressive and highly significant drops in concentration were recorded for infusion rates of 2.56 and 5.12 mg/min per 100 g body weight. An increase of the metabolic clearance rate of corticosterone, observed as a function of the infusion rate, was ascribed to saturation by the steroid of the plasma transcortin binding sites.  相似文献

Studies were undertaken on the turnover of ribosomal RNA and on ribonuclease activity in the liver of the pregnant rat in an attempt to explain the accumulation of liver RNA which occurs during the latter half of pregnancy. Between the 15th and 20th day of gestation the rate constant of degradation, biological half-life and daily rate of synthesis of ribosomal RNA were calculated to be 0.0887, 7.81 days and 6.21 mg per liver per 100g body weight respectively. Corresponding values in non-pregnant rats were 0.123, 5.68 days and 3.47 mg per liver per 100g body weight. The increase in RNA was therefore associated with an increase in its rate of synthesis and a decrease in its rate of breakdown. From the 14th day of pregnancy there was a decrease in alkaline ribonuclease activity and a marked increase in the level of alkaline ribonuclease inhibitor. The activity of acid ribonuclease was found to increase and that of acid phosphatase to decrease during this period.  相似文献

On a restricted food supply of Tubifex tubifex (2h/day) the fish consumed only 69.5 mg dry food/g live fish/day and grew only to 25.6 mg live body weight by the 30th day of it's age, while those fed ad libitum consumed 94.7 mg/g/day and attained a body weight of 125.9 mg live weight by the 30th day. Conversion efficiency (K1) averaged to 8.3% in the former and 14.1% in the latter series, throughout the 30 day experimental period.Feeding rate of the test series fed individually and at ad libitum, decreased from 780 mg dry food/g dry fish/day for a fish weighing 4 mg dry weight to 180 mg/g/ day for one weighing 41 mg, exhibiting a log-log relationship to the body weight (Y = 2.20140–0.5639 X). The effect of chlordecone (CD) on hepatic repair, measured either as recovery of microsomal enzymatic functions or as the induction of cytosolic thymidine kinase (TK) activity, was evaluated in rats given carbon tetrachloride (CC14). Carbon tetrachloride was administered to CD-potentiated and control animals using doses of this hepatotoxin which produce similar degrees of damage at 24 hours in both groups of animals (6 and 100 μ1 CC14 per 100 g body weight, respectively). Chlordecone had no significant effect on the time course of recovery of microsomal cytochrome P-450 content or glucose-6-phosphatase activity following CC14 administration. Hepatic TK activity was measured 48 hours after CC14 administration as a biochemical index of the hepatic regenerative response. Thymidine kinase activity was increased eightfold in CD-treated rats receiving 6 μ1 CC14 per 100 g body weight, whereas in controls a similar induction of TK activity was produced by 100 μ1 CC14 per 100 g body weight. Therefore, the TK response in CD-treated rats receiving CC14 is appropriate for the amount of damage produced, suggesting that CD does not inhibit the hepatic regenerative response to CC14-induced injury. The effect of CD on hepatic repair was also examined in rats receiving a two-thirds partial hepatectomy. Pretreatment of animals with CD had no significant effect on the increase in TK activity produced 24 hours after partial hepatectomy. These results offer no support for the idea that CD impairs hepatic repair after either partial hepatectomy or CC14 administration.  相似文献

The influence of cortisone (1.5 mg per 100 g of body weight, daily, for 10 days) on the neuro-muscular system was studied in rats in situ. The action potentials of the nerve and muscle were recorded with the extracellular electrodes. The rest potentials (RP) of the muscle fibers and the miniature end-plate potentials (MEPP) were recorded with the intracellular glass microelectrodes. A decrease of the RP and the MEPP amplitude, and an increase of the MEPP frequency and prolongation of the neuromuscular transmission time were revealed in rats given daily doses of cortisone, 1.5 mg/100 g of body weight, during 10 days; reliability of the neuro-muscular transmission (acceleration of the fall of the muscle action potential amplitude during tetanus) proved to decrease under the action of cortisone.  相似文献

The experiments were performed on Wistar rats with weight of 150-200 g. Antibodies were prepared by immunization of rabbits with pure surfactants of rat lungs and were intravenously injected into rats three times within 3 days intervals. These antibodies were shown to influence the superficial activity of lung surfactants and the alveolar lung cells activity. The low doses of antibodies (0.06 micrograms of protein per 100 g of body mass) stimulated the superficial activity of lung surfactants, while higher doses (3 mg of protein per 100 g of body mass) inhibited it.  相似文献

White clover plants were grown for 97 days under two temperature regimes (20/15°C and 8/5°C day/night temperatures) and were supplied with either small amounts (a total of 80 mg N pot–1) of ammonium (NH 4 + ) or nitrate (NO 3 ) nitrogen, or received no mineral N and relied on N2 fixation. Greatest growth and total leaf area of clover plants occurred in N2 fixing and NO 3 -fed plants grown at 20/15°C and poorest growth occurred in NH 4 + -fed plants grown at 8/5°C. Nodule mass per plant was greater at 8/5°C due to increased nodule numbers rather than increased dry weight per nodule. This compensated to some extent for the reduced N2-fixing activity per unit dry weight of nodule tissue found at the low growth temperature up to 116 d after sowing, but thereafter both activity per nodule dry weight and activity per plant were greater at the low temperature. Highest nitrate reductase activity (NRA) per g fresh weight and total activity per leaf, petiole or root occurred in NO 3 -fed plants at 8/5°C. Low growth temperature resulted in a greater partitioning of total plant NRA to the roots of NO 3 -fed plants. The results are considered in relation to the use of N fertiliser in the spring under field conditions.  相似文献

The effect of commercial purified soybean saponin on renin activity and blood pressure was investigated. Soybean saponin significantly inhibited human renin in vitro with IC50=59.9 μg/ml. Orally administered soybean saponin at 80 mg/kg of body weight per day to spontaneously hypertensive rats for 8 weeks significantly decreased the blood pressure.  相似文献

Abstract

Coffee senna (Senna occidentalis L.) plants were subjected to five phosphorus levels: 0, 25, 50, 75 and 100 mg P per kg soil (P0, P1, P2, P3 and P4, respectively). A pot culture experiment was conducted in a net house, AMU, Aligarh, India, under phosphorus-deficient soil. The present data indicates that soil-applied phosphorus significantly ameliorates most of the attributes studied. Out of five phosphorus levels, 75 mg P per kg soil (P3) proved best and enhanced fresh and dry weights, total chlorophyll and carotenoid content, nitrate reductase activity and leaf-NPK and Ca content, analyzed at 120, 270 and 300 days after sowing (DAS). The number of pods, seed-yield per plant and seed-protein content (330 DAS) were significantly enhanced by the P3 level, except the number of seeds per pod, 100-seed weight and total anthraquinone glycosides content, respectively. Transpiration rate, stomatal conductance and net photosynthetic rate were also enhanced by this treatment.  相似文献

NAD-dependent 15-hydroxy-prostaglandin dehydrogenase (PGDH) activity was measured in homogenates of 25 human placentae obtained between 7 and 17 weeks of gestation. PGDH activity, expressed in nanomoles PGF metabolized per min, ranged from 0.2 to 5.4 nmoles per mg placental protein and from 1.5 to 80 nmoles per g wet weight. PGDH activity per mg protein and per g weight increased significantly in function of gestational age (p<0.001). Between 7–8 weeks' gestation and 15–16 weeks mean values increased tenfold from 0.4 to 3.0 nmoles per mg protein and from 2.7 to 36.6 nmoles per g wet weight. Per unit of weight these early placentae contained less PGDH activity than term controls, but this related mainly to their high water content. Per mg placental protein PGDH activities already equalled values found at term before the end of the first trimester. The data indicate that the development of terminal villi and the migration of trophoblast into the maternal spiral arteries is associated with a substantial increase in the placental capacity for prostaglandin metabolism.  相似文献

Cytokinin activity based on two bioassays was at least 100-fold higher in Psychotria punctata leaf discs with bacterial nodules than in discs without them. Nodulated discs from young leaves yielded 0.4 to 6 μg of cytokinin (zeatin equivalents) per g fresh weight of leaf tissue, whereas non-nodulated discs from the same leaves yielded 0 to 0.003 μg per g fresh weight. These estimates probably include free-base cytokinins and, if present, any nucleoside cytokinins precipitable by acidic silver nitrate. Cytokinin concentrations in Psychotria leaf nodules appear to be higher than normally found in green leaves of other plants. In l-butanol-acetic acid-water (12:3:5, v/v), the one peak of activity chromatographed with an RF similar to zeatin's, but both number and identity of the active substance(s) remain unknown. These findings suggest that a cytokinin is produced by bacteria in leaf nodules of P. punctata and that it is involved in the symbiosis.  相似文献
