Paints containing mercuric oxide or thiophanate-methyl were applied to either scraped or unscraped cankers caused by Nectria galligena on apple trees at two sites. Three gel formulations (carbendazim/triadimefon in alginate or xanthan and carbendazim/imazalil in xanthan) and a solvent-based PP969 formulation were applied to unscraped cankers only. Assessments were made 9 and 21 months after treatment. Mercuric oxide was more effective than thiophanate-methyl after 21 months on both scraped and unscraped cankers. All gel formulations reduced spore production and fungitoxicant could still be detected in bark and wood after 21 months. The solvent-based PP969 formulation did not perform as well as the gels.  相似文献

Four paints containing mercuric oxide, octhilinone, thiophanate-methyl or triad-imefon and five gel formulations (carbendazim/triadimefon in sodium alginate or xanthan gum, carbendazim/triadimefon/hypophosphorous acid and thiophanate-methyl/oxycarboxin in xanthan or PP969/alginate) were tested on natural cankers caused by Nectria galligena on apple trees. Scraping cankers decreased their size 7–9 months later due to stimulation of callus formation, even where no fungicide was applied, and decreased perithecial, but not conidial production. Only the paints were applied to scraped cankers and that containing mercuric oxide was the most effective in decreasing canker size and restricting new canker growth. All formulations except the octhilinone paint were applied to unscraped cankers. Only mercuric oxide paint and acidic carbendazim/triadimefon/xanthan gel stimulated callus formation and only the former caused a decrease in canker size. Every treatment, except triadimefon paint and PP969/alginate, reduced conidial production. Bioassay with TV. galligena showed that the carbendazim and thiophanate-methyl gels achieved some movement of fungitoxicant into plant tissue.  相似文献

Plum decline was associated with Pseudomonas syringae pathovars syringae and morsprunorum in Baden‐Württemberg. The trunks of affected plum trees (Prunus domestica) were girdled by bacterial cankers resulting in sudden death of infected trees. Copper compounds that were applied extensively during leaf fall and bud burst, were not effective. A minority of P. syringae strains isolated from cankers on plum trees were moderately resistant, while most strains were sensitive to cupric ions. Invasions through blossoms, leaves and wounds during the vegetation period were limited to the infection sites and plum trees coped effectively with both P. syringae pathovars eliminating them eventually. Infections after dormancy including very rare leaf scar infections did not induce cankers on the trunk. However, infections of dormant trees through frost injuries, (pruning) wounds or non‐injurious ingress by freezing and thawing were serious, because they led to cankers girdling the trunk. Control strategies to manage plum decline have to be adapted to the disease cycle. They should concentrate on the dormant period beginning with early frosts in autumn and ending with bud burst.  相似文献

Trapping of ascospores and conidia of Nectria galligena Bres. released from cankers on Bramley's Seedling apple trees over a period of three years showed that ascospores were most prevalent in the spring and early summer, and conidia from early summer to late autumn. Few ascospores were released in late summer or mid-winter but a minor period of discharge occurred in autumn. The distribution of cankers within trees in various orchards was also examined, and most were found to occur at the junctions of different seasons' growth. Possible reasons for this are discussed.  相似文献

Aims: To evaluate a sodium hypochlorite and hydrogen peroxide solution (Ox‐B7) as a potential decontaminant of Bacillus subtilis spore‐contaminated surface materials (porous and nonporous). Methods and Results: Test materials were contaminated with B. subtilis spores to a final concentration in the range of 5·7–6·6 log CFU cm?2. Ox‐B7 reduced spore counts by 99·999% (5 log) for both porous and nonporous surfaces within a 5‐min contact. Treatment with equivalent concentrations of only sodium hypochlorite reduced spore counts by 99% (2 log) on porous materials and by 99·99% (4 log) on nonporous materials. Hydrogen peroxide treatments reduced spores by less than 90% (<1 log) on both porous and nonporous materials when compared with untreated samples. Conclusions: A combination of sodium hypochlorite and hydrogen peroxide (Ox‐B7) effectively killed B. subtilis spores on both porous and nonporous surface materials. Significance and Impact of the Study: The combination of sodium hypochlorite and hydrogen peroxide can be used as an alternative disinfectant of spore‐contaminated surface materials, as it is more effective than when hydrogen peroxide or sodium hypochlorite are used separately.  相似文献

In a factorial experiment on canker control the efficiency of phenylmercurie nitrate (PMN) applied at leaf fall, before bud burst or both was compared with the application of dodine, dithianon, triforine, thiabendazole, benomyl or carbendazim in May and June. Of the total number of cankers which developed in unsprayed trees 76–78% resulted from infections in April to August of each year of the experiment. Infection was reduced significantly by all of the fungicides applied in summer. Of these carbendazim was outstanding, controlling both summer and autumn infections. Sporulation throughout the summer was suppressed by carbendazim and to a lesser extent benomyl, but whereas the suppressant effect of carbendazim persisted until long after leaf fall that of benomyl was evident only until August. Dodine, dithianon, triforine and thiabendazole had no significant effect on spore production. Dithianon and dodine showed highest toxicity to the germination of Nectria galligena spores of all fungicides used in summer. All of the fungicides controlled apple scab (Venturia inaequalis) although thiabendazole was the least effective. Autumn applications of PMN reduced canker incidence but their value alone was not as great as carbendazim, dithianon, benomyl or dodine applied in summer. PMN applied in spring reduced the number of cankers in trees receiving no other fungicides but tended to increase the incidence of infections, particularly in autumn, when used in conjunction with fungicides applied in May and June. PMN applied before bud burst reduced sporulation of N. galligena for a few weeks, after which production resumed and in late summer and autumn actually exceeded the controls. None of the fungicides had any direct effect on leaf fall. Infection of the crotch and basal leaf scars was more common in summer than in autumn whereas infection of leaf scars above the basal region was more common in autumn than in summer. The rootstocks of many of the trees became infected via callus tissue associated with adventitious root development and this was controlled by the carbendazim treatment.  相似文献

Injury or inoculation of apparently healthy citrus trunk bark and wood with Fusarium solani immediately prior to and after the December 1983 freeze resulted in cankers similar to natural branch and trunk cankers which formed on trees following the freeze. Fusarium solani was consistently isolated from active lesions of natural and induced cankered bark. Phytophora spp. were not isolated from cankered tissue. Earliest cankers appeared as a water-soaked area under the bark around the point of inoculation, later F. solani sporodochia formed on the bark surface. Older cankers became dry and cracked but produced no gum. Canker size ranged from 10–90 cm vertical diameter in spring 1984 inoculations but were only several cm diameter following late summer inoculations. Active cankers developed again after inoculation following the January 1985 freeze. Similar cankers were produced on Hamlin sweet orange stems inoculated with F. solani.  相似文献

A field trial to assess the ability of Chondrostereum purpureum to limit the resprouting of cut red alder (Alnus rubra) was established in British Columbia, Canada. Overall, 92% of stumps inoculated with C. purpureum died in the first year and 100% were dead by the second year. C. purpureum was thus as effective as the herbicide treatments Carbopaste and liquid glyphosate (12% Vision) and reduced resprouts to zero. Naturally occurring C. purpureum spores colonized about 40% of untreated cut stumps and about 20% of the stumps treated with chemical herbicides. The peak production of C. purpureum basidiocarps occurred about 22 months after the trial was established, with C. purpureum on 66% of treated stumps. The following year, the basidiocarps persisted on 23% of treated stumps. About 600 young red alder trees were cut to act as spore traps 2 years after trial establishment, when the most C. purpureum basidiocarps were present on treated stumps in the trial. These spore-trap trees were subsequently sampled, and PCR markers were used to verify and characterize 43 dikaryotic C. purpureum individuals, none of which were identified as the released isolate 2139. There was little difference in average band-sharing with the applied isolate among pre-trial (37% average) and spore-trap (38% average) populations. There was therefore no evidence that the C. purpureum strain applied inundatively to stumps in this trial was detectable on nontarget trees in the local area or had any measurable impact on the local C. purpureum population.  相似文献

Uredospore production of Puccinia striiformis in single wheat seedlings was assessed by weighing spores on an electrical microbalance, by counting on a haemocytometer or by measuring turbidity of spore suspensions with a spectrophotometer and compared with production from groups of seedlings determined by weighing spores on an analytical balance. The data were used to assess differential interaction of wheat cultivars Hybrid 46 and Joss Cambier with two isolates of race 104 E137 of P. striiformis and cvs Maris Templar and Joss Cambier with two isolates of race 41 E136. A significant differential interaction was shown in both experiments by each method but most rapidly and with the minimum of materials by the single-plant microbalance technique. Measurements of spore production demonstrated differences between isolates within races more clearly than the conventional visual assessment of yellow rust symptoms.  相似文献

Rhizoctonia disease, caused by Rhizoctonia solani is one of the most important fungal diseases in bean fields in Isfahan, Iran. Bean plants showing stem and root cankers were collected and Rhizoctonia-like fungi obtained from the samples were identified by anastomosis. Pure cultures of bean isolates of R. solani were identified as AG-4. There were also AG-4 isolates from tomato, potato, cucumber, alfalfa and sugar beet in the areas sampled. A total of 163 isolates of R. solani AG-4 originating from stem and root cankers of beans were examined using pectic zymogram electrophoresis. Polygalacturonase (PG) and pectin estrase isozymes were observed in all AG-4 isolates tested. One (PG) and one pectic esterase (PE) band was found in common between all isolates examined. The electrophoretic patterns were grouped into seven zymogram groups (ZGs) according to the diagnostic PG and PE bands. One ZG occurred in a high frequency throughout the areas sampled. A pathogenicity test was conducted and representative isolates of each ZG were used to inoculate healthy bean plants. The results showed that each ZG caused different symptoms with varying severity. Isolates belonging to two ZGs were highly pathogenic causing root, stem and hypocotyl cankers whereas isolates of the other ZGs produced weak or no symptoms.  相似文献

The objective of this study was to determine the remarkable role of the microwave power density of argon plasma in the inactivation of Bacillus subtilis, Bacillus stearothermophilus and Bacillus pumilus spores deposited on polypropylene bio‐indicator carriers. In particular, spore survival by argon plasma was determined as a function of the initial spore density of the bio‐indicators. The microwave induced argon plasmas were generated at 1.47, 2.63 and 4.21 w/cm3 microwave power densities under a low gas pressure of 50 Pa at an ambient temperature of 15 °C to reach low temperature distribution of 31, 35 and 43 °C, respectively. Our results indicate that the different Bacillus spores showed distinct degrees of argon plasma sensitivity, and spore survival was significantly reduced when the microwave power density of the plasma treatments was increased. Among the three Bacillus strains, Bacillus subtilis was the most argon plasma resistant, whereas Bacillus stearothermophilus was the most sensitive. However, spore survival was not affected by the initial spore density of the bio‐indicators. Only a certain degree of the spore inactivation log (No/N) from 1.67 to 1.95 was observed despite the 4‐order differences in the initial spore density of the Bacillus pumilus bio‐indicators.  相似文献

A severe leaf spot disease of cucumber caused by a pathotype of Alternaria alternata (Fr.) Keissler was recorded recently in plastic houses in Crete. Lesions ranged in size of a pin point to over 5 cm in diameter, with necrotic tissue on most of their area and a surrounding yellow zone. The pathogen grew satisfactorily on PDA at temperatures between 5 °C–40 °C and spore germination occurred in the range less than 10 °C to over 37 °C. Optimum temperature in both cases was near 26 °C. Of,13 fungicides tested in vitro, sodium omadine, etem, dichlofluanid, captan and folpet were the most inhibitory on spore germination, and iprodione, sodium omadine and dichlofluanid on mycelial growth. Of 25 fungicides applied on two leaf cucumber plants 24 h before inoculation, maneb, etem, dichlofluanid and chlorothalonil were the most effective. When the last fungicides, plus mancozeb, were applied 24 h after inoculation only maneb was effective. In greenhouse experiments, iprodione, prochloraz-manganese-complex, chlorothalonil, dichlofluanid, guazatine, maneb and etem were the most effective for disease control, while mancozeb was less effective. The local cucumber cv. Knossos and the Dutch F1 hybrids Evadan, Frella, Herta, Malfa, Mazourka, Pepinex 69 and Renova were all susceptible to infection.  相似文献

Aims: To obtain data on the efficacy of various liquid and foam decontamination technologies to inactivate Bacillus anthracis Ames and Bacillus subtilis spores on building and outdoor materials. Methods and Results: Spores were inoculated onto test coupons and positive control coupons of nine different materials. Six different sporicidal liquids were spray‐applied to the test coupons and remained in contact for exposure times ranging from 10 to 70 min. Following decontamination, spores were recovered from the coupons and efficacy was quantified in terms of log reduction. Conclusions: The hydrogen peroxide/peracetic acid products were the most effective, followed by decontaminants utilizing hypochlorous acid chemistry. Decontamination efficacy varied by material type. Significance and Impact of the Study: The study results may be useful in the selection of technologies to decontaminate buildings and outdoor areas in the event of contamination with B. anthracis spores. These results may also facilitate selection of decontaminant liquids for the inactivation of other spore‐forming infectious disease agents.  相似文献

Some lignivorous hymenomycete fungi are capable of causing both cankers and decay in stemwood of adult trees. Recently in Tuscany (Italy), Platanus x acerifolia trees were found colonized by Sarcodontia pachydon (Polyporales, Meruliaceae), a fungus associated with white rot and stem cankers on different host tree species. Because the relationship S. pachyodon‐plane‐tree was only preliminary studied, we decided to investigate whether isolates obtained from this host are distinct from those commonly collected from oaks. For this purpose, isolates obtained from plane‐tree and from holm oak (Quercus ilex) were compared by in vitro test and molecular markers. Results showed that fungal isolates did not differ in growth nor in wood degradation, also molecular tests revealed relative similarity among fungal samples.  相似文献

Summary The effect of different physical and biological factors like soil sterilization, incubation period of soil, spore age, amendment of certain fungal species and their metabolites on soil fungistasis has been investigated.Different degree of sterilization affected the fugistasis differently. Soil heating above 80°C completely annulled the fungistasis. No fungistasis was recorded in soil samples steamed for 15 mts in an autoclave.Incubation of soil samples to longer duration resulted in increased fungistasis. Maximum fungistatic value was noted in samples incubated for 15 days at 25±1°C.Spore age also played important role in fungistasis. A positive relation was noted in the spore age and fungistasis upto 30 days of age and thereafter the increase in fungistasis was not well marked.Varying inhibitory effect was noted on the spore germination of the test fungi in relation to amendment of certain fungi individually and in different combinations to the soil.Aspergillus flavus alone and in combination ofAspergillus niger proved most inhibitory. The filtrate of the different fungi also induced fungistasis in soil. In this case alsoA. flavus was most effective.  相似文献

Little research has been carried out in London concerning fungal spore prevalence yet this information may help to elucidate geographical patterns of asthma and hay fever. Although many types of spore reach peak concentrations outdoors in late-summer, the incidences in the indoor environment may be more important through the winter because of heating and poor ventilation. Daily average concentrations of fungal spores in the ambient atmosphere were monitored with a Burkard volumetric spore trap on an exposed roof in North London from autumn 1991 until the summer of 1992. Indoor spore measurements were taken in 19 homes in the vicinity through the winter months, both by direct air sampling using a portable Burkard sampler and by dust culture. Trends in the occurrence and concentrations of fungal spores indoors and outdoors were examined. Relationships between the abundance of selected allergenic fungi and features of the houses were analysed including age of dwelling, dampness, cleanliness and presence of pets.Aspergillus andPenicillium were the most frequently occurring spore types in the homes. Overall, high spore incidence was associated with dampness and dust accumulation. The outdoor spore samples revealed generally low concentrations through the winter until March when concentrations of many types includingCladosporium, Epicoccum andAlternaria increased in abundance in response to the warmer weather. Even during the late-spring and early-summer, concentrations of most fungal spores were notably below those reported for rural sites.  相似文献

Berberine and santonin were isolated from rhizomes ofBerberis aristata and unexpanded flower buds ofArtemisia maritima, respectively. Efficacy of these two chemicals individually as well as of their mixtures, was tested against spore germination of some saprophytic and obligate fungi. While berberine individually was effective against most of the fungi,Helminthosporium spp. were least affected even at the highest dose (1500 ppm). Santonin was equally effective against several fungi. Mixture of both alkaloids found to be more effective than individual ones. Keeping the dose of berberine constant and santonin at two different concentrations (viz. 250 and 500 ppm) the spore germination ofHelminthosporium, oryzae was stimulated. Increasing concentration of santonin inhibited the spore germination of all other fungi tested,Colletotrichum capsici being affected only by 20 and 5% (at berberine concentration of 250 and 500 ppm, respectively). On the other hand, santonin being constant and berberine at different concentrations, the mixture was effective against all the fungi.  相似文献

Six selected antioxidants (catechin, quercetin-3-galactoside, cyanidin-3-glucoside, pelargonidin-3-glucoside, ellagic and gallic acids) were evaluated in vitro for their antifungal activities and mode of action on Botrytis cinerea Pers., one of the most important pathogens of strawberries. Inhibitory effects were found for all the tested antioxidants, but varied at different fungal developmental stages. Catechin and quercetin-3-galactoside showed linear inhibitory effects on germ tube elongation, with the highest suppression ratios of 54.8% and 58.8% respectively. No significant effect was found on spore germination between treatments and control. Gallic acid showed very strong and linear inhibition on spore germination (r = ?0.95), but the effect diminished after spore germination. Cyanidin-3-glucoside and pelargonidin-3-glucoside provided effective control on the fungi as concentrations increased. The arresting effect of ellagic acid on development of B. cinerea was quadratic. Ellagic acid inhibited germ tube elongation and mycelial growth at its highest and lowest concentrations, while no effects were observed at its medium concentration used in this study.  相似文献

An effective formulated biopesticide for controlling sheath blight in rice was developed using three microbial antagonists (Bacillus megaterium, Bacillus subtilis and Aspergillus niger) isolated from the rice sheath. The efficiency of spore-based formulations of the above microbial antagonists was investigated and their effectiveness in controlling sheath blight was demonstrated. Application of talc-based formulations of individual antagonists and mixtures of the three antagonists as spray treatments or soil applications were effective in reducing the incidence by up to 45% at 27 days after inoculation of the pathogen of sheath blight and increased rice yield. The use of spores of a fungal antagonist (A. niger), in comparison to commonly used bacterial antagonists, is a novel feature of the present study. Optimum sporulation conditions of the antagonists for preparation of spore-based formulations and their commercially desirable features such as the ability to maintain spore viability in storage were also determined. Culturing in the synthetic replacement sporulation medium (SRSM-2) for 72 hours was the most effective for sporulation of the two bacterial antagonists while culturing in potato dextrose broth (PDB) for 7 days was the most effective for sporulation of the fungal antagonist. It was demonstrated that talc-based formulations of all antagonists, either in refrigerated storage (4°C) or at room temperature (28±2°C), were able to maintain greater spore viability over a longer period (>6 months) than spore suspensions. In view of the relatively shorter life spans of formulations based on vegetative cells, spore-based formulations have a distinct advantage in achieving longer-lasting control, especially under harsh field conditions.  相似文献

The fungus Eutypa lata was isolated from diseased walnut trees (Juglans regia) exhibiting small cankers. The morphological characteristics of the culture and the pathogenicity were compared with those of known isolates of the fungus from other hosts. Inoculation tests with walnut isolates on grape, walnut, almond and apricot yielded characteristic cankers. Furthermore, pathogenicity tests on walnut with isolates from other different hosts resulted in differences in virulence. Similar differences in virulence were observed between 13 single ascospore isolates of the fungus ex apricot inoculated on walnut.  相似文献
