Male and female reproductive functions have been proposed as possibly sensitive targets for the biological effects of 60-Hz (power frequency) magnetic fields (MF). However, experimental data relevant to this hypothesized association are very limited. In the present study, the "reproductive assessment by continuous breeding" design was used to identify possible effects of MF exposure on reproductive performance, fetal development, and early postnatal growth in rats. Groups of age-matched Sprague-Dawley rats (40 breeding pairs/group) were exposed continuously (18.5 hr per day) to linearly polarized, transient-free 60-Hz MF at field strengths of 0 Gauss (G; sham control), 0.02 G, 2.0 G, or 10.0 G. An additional group of 40 breeding pairs received intermittent (1 hr on/1 hr off) exposure to 10.0 G fields. F0 breeding pairs were exposed to MF or sham fields for 1 week prior to mating, during a 14-week period of cohabitation, and during a 3-week holding period after cohabitation. The duration of the cohabitation period was selected to be sufficient for the delivery of five litters in the sham control group. Pups from the final F1 litter from each breeding pair were exposed to MF or sham fields until sexual maturity, were cohabitated in MF or sham fields for 7 days with nonsiblings from the same exposure group, and were held in the MF or sham fields for 22 days to permit delivery of F2 pups for evaluation. No evidence of exposure-related toxicity was identified in any rat in the F0, F1, or F2 generations. Fetal viability and body weights in all litters of groups exposed to MF were comparable to those of sham controls. No significant differences between sham controls and MF-exposed groups were seen in any measure of reproductive performance (litters/breeding pair, percent fertile pairs, latency to parturition, litter size, or sex ratio) in either the F0 or F1 generation. Exposure of Sprague-Dawley rats to 60-Hz MF strengths of up to 10.0 G either during their peak reproductive period (F0) or during gestation and throughout their life span (F1) has no biologically significant effects on reproductive performance. These results do not support the hypothesis that exposure to pure, linearly polarized 60-Hz MF is a significant reproductive or developmental toxicant.  相似文献

Nicotine exposure has been associated with an increased likelihood of developing attention deficit hyperactivity disorder (ADHD) in offspring of mothers who smoked during pregnancy. The goal of this study was to determine if exposure to E-cigarette nicotine vapors during late prenatal and early postnatal life altered behavior in adult mice.

Methods

Timed-pregnant C57BL/6J mice were exposed to 2.4% nicotine in propylene glycol (PG) or 0% nicotine /PG once a day from gestational day 15 until delivery. After delivery, offspring and mothers were exposed to E-cigarette vapors for an additional 14 days from postnatal day 2 through 16. Following their last exposure serum cotinine levels were measured in female juvenile mice. Male mice underwent behavioral testing at 14 weeks of age to assess sensorimotor, affective, and cognitive functional domains.

Results

Adult male mice exposed to 2.4% nicotine/PG E-cigarette vapors had significantly more head dips in the zero maze test and higher levels of rearing activity in the open field test compared to 0% nicotine/PG exposed mice and untreated controls. In the water maze test after reversal training, the 2.4% nicotine/PG mice spent more than 25% of time in the new location whereas the other groups did not.

Conclusion

Adult male mice exhibited increased levels of activity in the zero maze and open field tests when exposed to E-cigarette vapor containing nicotine during late prenatal and early postnatal life. These findings indicate that nicotine exposure from E-cigarettes may cause persistent behavioral changes when exposure occurs during a period of rapid brain growth.  相似文献

A study was conducted to evaluate the effect of short-term relocation and male exposure on receptivity rate, kindling rate and total born per litter in lactating does under an artificial insemination (AI) programme. Thirty-two, 2-month-old New Zealand White rabbits were randomly allocated to one of four treatments: (1) relocation and male exposure; (2) relocation without male exposure; (3) no relocation with male exposure; (4) no relocation without male exposure (control). Relocation and male exposure were done 8-10 h before the time of service. First insemination was when does reached 3200 g body weight and does were bred 4-13 days after parturition across parities during a 6 month reproduction period. Of all breeding records, 125 inseminations and 91 kindlings were from nursing does. The mean interval from parturition to insemination for nursing does was 10.3 days. Relocation of lactating does resulted in greater (P<0.01) receptivity rate at service (74.8%) as compared with no relocation (55%). Receptivity rate was not influenced by male exposure. However, the interaction of relocationxmale exposure tended to be significant (P=0.07). Receptivity rate in relocated does exposed to males was 62.8 and 86.7% without exposure while in non-relocated does male exposure showed no effect. Kindling rate was not influenced by relocation or male exposure. The mean total born per litter in relocated and non-relocated does was 8.05 +/- 0.33 and 7.39 +/- 0.36, respectively, but no significant difference was observed. There was no effect of male exposure on total born per litter (7.85 +/- 0.34 versus 7.59 +/- 0.34 without male exposure). However, interaction of relocationxmale exposure on this variable was significant (P=0.009). Male exposure in relocated does decreased the size of the litter (7.52 +/- 0.46 versus 8.58 +/- 0.47 without male exposure) whereas mean values in non-relocated does increased when they were exposed to males (8.18 +/- 0.52 versus 6.60 +/- 0.49). Short-term relocation improved receptivity rate and reproduction in lactating does under an artificial insemination programme. Preliminary results indicated that male exposure in non-relocated does improves the total born per litter at a similar level than relocated does without male exposure. Relocation combined with male exposure decreased receptivity rate and total born per litter as compared with relocated does without male exposure, but the reproductive performance in the former was greater as compared with those does where no relocation occurred without male exposure.  相似文献

Food quantity and especially food quality are thought to be key factors driving reproductive changes in the house mouse, Mus domesticus, leading to outbreaks of house mouse populations in the Australian grain-growing region. Characteristic changes during an incipient mouse plague are an early start of breeding, a high proportion of females breeding at a young age and a prolonged breeding season. We conducted a large-scale food manipulation during an incipient mouse plague, which started with early breeding and relatively high spring numbers of mice. We measured background food availability in four farms throughout the study and conducted a food manipulation experiment from November to March in two of them. After harvest in December 100-200 kg/ha spilled grain remained in the stubble. This was depleted by March. In two treatment farms we added high-protein food pellets on a weekly basis between November and March and two farms served as controls. We measured changes in mouse numbers by capture-mark-recapture trappings and changes in reproduction by scoring embryos and recent placental scars at necropsy. Mouse numbers did not differ between treatments and controls. There were no differences in the litter size or the proportion of females breeding between treatments and controls. We observed the normal pattern of high litter size in spring and decreasing litter size towards the end of summer in treatments and controls. In all farms reproduction stopped in March. Mouse numbers were high but not at plague densities. Contrary to our prediction we did not observe food constraint affecting the reproduction of female mice. Our field experiment seems to rule out food quality as the driving factor for improved reproduction and formation of an outbreak of mice. We suggest that physiological mechanisms in mice might not enable them to take advantage of food with a high protein content in arid summers in southeastern Australian grain fields because of the lack of free-standing water.  相似文献

Single trait selection of mice for either large body size or large litter size resulted in an increased ovulation rate because of possible enhanced ovarian sensitivity to gonadotrophins. There was no difference in pre-implantation embryonic survival in either of the selected lines when compared to control mice. Selection for body weight did not alter post-implantation embryo survival, but fewer fetuses were lost after implantation in the litter size line compared to the control line. Index selection for large body size and small litter size did not change ovulation rate but increased pre- and post-implantation embryonic mortality. Selection for small body size and large litter size increased ovulation rate and decreased early embryonic death.  相似文献

C57BL/6 female mice impregnated by C3H males mice to produce B6C3F1 fetuses were exposed daily for six hr to atmospheres containing 0, 250, 500, or 750 ppm methyl chloride, from gestation day 6 to gestation day 18. There were 74 to 77 females with copulation plugs per exposure concentration. Females exposed to 750 ppm ethyl chloride exhibited ataxia commencing on the seventh day of exposure (gestation day 12). They also showed hypersensitivity to touch or sound, tremors and convulsions. Six females in the 750 ppm group died and one was euthanized in extremis prior to scheduled sacrifice. On gestation day 18, all other females were euthanized for evaluation. Only dams exposed to 750 ppm exhibited significant decrease in body weight by gestation day 18, weight gain during the gestation period, and absolute weight gain (weight gain minus gravid uterine weight) versus controls. There were no treatment related-effects on these parameters in the other exposure groups. None of the groups exhibited exposure-related differences in pregnancy rate, gravid uterine weight, or maternal liver weight. There were no differences in the numbers of implantations, resorption, dead fetuses, nonlive (dead plus resorbed) fetuses, live fetuses, sex-ratio, or mean fetal body weight per litter. There was a significant exposure-related increase in the number and percentage of affected (nonlive plus malformed) fetuses per litter with the incidence of affected fetuses in the 750 ppm group significantly higher than controls. There was a statistically significant increase in the incidence of heart defects in the 500 and 750 ppm group relative to controls. Of the 37 fetuses in the study with heart defects, 23 were females, 14 were males. The heart defects observed included: absent or abnormal tricuspid valve, reduced number of papillary muscles and/or chordae tendineae on the right side, small right ventricle, globular heart, and white spots in the left ventricular wall. Multiple malformations were observed in one fetus from the 500 ppm group and in three fetuses in the 750 ppm group. It is concluded that methyl chloride inhalation exposure to pregnant C57BL/6 mice from gestation day 6 through gestation day 17 resulted in maternal toxicity only at the 750 ppm exposure concentration and was teratogenic to B6C3F1 conceptuses at exposure concentrations of 750 and 500 ppm, leading to fetal heart malformations. No evidence of embryo or fetotoxicity other than teratogenicity was seen at any of the exposure concentrations employed. No maternal, embryo or fetotoxicity or teratogenicity was associated with exposure of mice, during critical periods of embryo and fetal development, to 250 ppm of methyl chloride.  相似文献

The influence of a pulsed magnetic field (PMF; sawtooth with 45-μs linear rise time and 5-μs decay time, peak strength of 15 μT, and frequency 20 pps) on the embryogenesis of CBA/S mice was investigated in five experiments based on a total of 707 exposed and 543 unexposed primigravidas. Sham and PMF exposures began on day 1 of gestation (experiments 1 and 2), on day 2 (experiment 3), on day 5 (experiment 4). and on day 7 (experiment 5): all exposures continued until day 19 post conception (p.c.) when they were terminated, at which time the following variables were measured: number of implants; number of placental resorptions; number of living fetuses; number of dead fetuses; number of malformations in living and dead fetuses; and length and body mass of living fetuses. Control dams were sham-exposed concurrently with corresponding. PMF-exposed dams. With the exception of experiment 5, in which exposure to PMF started on day 7 p.c., all groups of exposed mice had significantly more placental resorptions when compared with concurrent controls. The increased resorption rate was not reflected in a reduction in litter size or in the number of litters. A significant increase in malformed fetuses was not seen in any of the exposed groups, or when groups were pooled. Only in experiment 1 was the number of dead fetuses affected by exposure to PMF. The effect of PMF on the implantation rate was not significant. Body mass and length of exposed fetuses were significantly reduced only when the PMF treatment began on day 7 p.c. That PMF-treated mice had significantly more placental resorptions when exposure began on day 5 p.c. or earlier (before implantation), but not when exposure began on day 7 (after implantation), may indicate a causative pre-implantation effect. Because a PMF-induced increase in the number of resorptions has not been observed in other strains of mice, the effect might be strain-related. © 1993 Wiley-Liss, Inc.  相似文献

Benzene is an important industrial chemical. At certain levels, benzene has been found to produce aplastic anemia, pancytopenia, myeloblastic anemia and genotoxic effects in humans. Metabolism by cytochrome P450 monooxygenases and myeloperoxidase to hydroquinone, phenol, and other metabolites contributes to benzene toxicity. Other xenobiotic substrates for cytochrome P450 can alter benzene metabolism. At high concentrations, toluene has been shown to inhibit benzene metabolism and benzene-induced toxicities. The present study investigated the genotoxicity of exposure to benzene and toluene at lower and intermittent co-exposures. Mice were exposed via whole-body inhalation for 6h/day for 8 days (over a 15-day time period) to air, 50 ppm benzene, 100 ppm toluene, 50 ppm benzene and 50 ppm toluene, or 50 ppm benzene and 100 ppm toluene. Mice exposed to 50 ppm benzene exhibited an increased frequency (2.4-fold) of micronucleated polychromatic erythrocytes (PCE) and increased levels of urinary metabolites (t,t-muconic acid, hydroquinone, and s-phenylmercapturic acid) vs. air-exposed controls. Benzene co-exposure with 100 ppm toluene resulted in similar urinary metabolite levels but a 3.7-fold increase in frequency of micronucleated PCE. Benzene co-exposure with 50 ppm toluene resulted in a similar elevation of micronuclei frequency as with 100 ppm toluene which did not differ significantly from 50 ppm benzene exposure alone. Both co-exposures - 50 ppm benzene with 50 or 100 ppm toluene - resulted in significantly elevated CYP2E1 activities that did not occur following benzene or toluene exposure alone. Whole blood glutathione (GSH) levels were similarly decreased following exposure to 50 ppm benzene and/or 100 ppm toluene, while co-exposure to 50 ppm benzene and 100 ppm toluene significantly decreased GSSG levels and increased the GSH/GSSG ratio. The higher frequency of micronucleated PCE following benzene and toluene co-exposure when compared with mice exposed to benzene or toluene alone suggests that, at the doses used in this study, toluene can enhance benzene-induced clastogenic or aneugenic bone marrow injury. These findings exemplify the importance of studying the effects of binary chemical interactions in animals exposed to lower exposure concentrations of benzene and toluene on benzene metabolism and clastogenicity. The relevance of these data on interactions for humans exposed at low benzene concentrations can be best assessed only when the mechanism of interaction is understood at a quantitative level and incorporated within a biologically based modeling framework.  相似文献

Background

The present study elucidates the protective potential of bromelain against dichlorvos intoxication in mice brains. Dichlorvos induces the oxidative stress by disproportionating the balance between free radicals generation and their scavenging in neurons which leads to neuronal degeneration.

Methods

In this study, mice were divided into four groups-group I (control), group II (dichlorvos treated), group III (bromelain treated) and group IV (exposed to both bromelain and dichlorvos both).

Results

Dichlorvos treatment increased the levels of thiobarbituric acid reactive substances (TBARS) and protein carbonyl content (PCC) which indicate the increased oxidative stress. Meanwhile, brain endogenous antioxidants and cholinesterases level was decreased after dichlorvos exposure. Levels of TBARS and PCC decreased whereas cholinesterases level was recorded to be elevated after bromelain exposure.

Conclusion

Bromelain offered neuroprotection by decreasing oxidative stress and augmenting cholinesterases in mice brains. This study highlights the invulnerability of bromelain against oxidative and cholinergic deficits in mice brains.

The least weaselMustela nivalis nivalis Linnaeus, 1766 is unique among carnivores because of its small body size and capacity for fast reproduction. It has been suggested to be the main agent for maintaining cyclic fluctuations in northern vole populations, largely based on its high reproductive potential and dependence on small rodents. This study describes basic reproductive data on the least weasel obtained during a captive breeding program. In the experiments, food availability was manipulated at the onset of breeding. Altogether, 65 litters were born during the 5-year study. The mean litter size was 5.1 (in 53 litters of known litter size), the most common litter size being six. The sex ratio of weaned young was not biased from 1∶1. The median date of birth was June 4. Food manipulations did not affect mating frequency suggesting that the observation of failure in breeding in years of low abundance of small mammals is due to mortality of embryos or young before weaning, but not due to avoidance of breeding at low food availability. In this respect, weasels differ from other main vole predators, owls and raptors, which often skip breeding if small mammals are scarce.  相似文献

Female mice were exposed to lead in utero by intravenous injection of lead chloride into the mothers at different stages of pregnancy. At a mature age the mice exposed as fetuses (F1 generation) conceived at a normal rate, but the litter size and fetal survival varied significantly. Small litters and increased numbers of fetal deaths were observed in mice exposed to lead on day 8 of intrauterine life. The live fetuses in this group were normal with respect to weight and morphological appearance. Serum levels of estradiol and progesterone, measured on day 17 of pregnancy, did not differ significantly between F1 mice of a control, unexposed group and of the group exposed to lead on day 8 of intrauterine life. Ovarian follicle counts revealed a significantly smaller number of primordial follicles in the latter group. It is suggested that the exposure to lead at a time of early organogenesis caused the observed fertility decrease by interfering with the development of the female germ cells.  相似文献

Does light exposure make plant litter more degradable?   总被引:4,自引:0,他引:4  
Many field experiments have indicated that litter decomposition in semi-arid areas may be partly or fully controlled by photodegradation. We devised a study to test our hypothesis that light exposure makes plant litter more degradable. Dry, senescent, aboveground plant litter from Miscanthus x giganteus was exposed to light including ultraviolet (UV) radiation for various lengths of time from 0 to 289 days. Weight loss was measured after exposure and appeared to be modest and did not increase with time of exposure. The litter of the longest and shortest exposure time as well as controls were then incubated with soil and moisture for 35 days and CO2 and N2O production were measured. The longest exposed litter degraded much faster than any other treatment during incubation with moisture, about twice as fast as the unexposed control. The shortest exposed however, degraded only slightly faster than the unexposed control. This suggests that increasing litter degradability is a more important mechanism for photodegradation than direct light-induced mass loss. N2O production from decomposition of the exposed litter was high in the beginning, suggesting that nitrogen may be released quickly. The mechanism is probably that light exposure leaves the nitrogen in plant litter easily available to microbial utilisation upon wetting. Such a mechanism might play an important role for nutrient cycling in semi-arid areas.  相似文献

Age at puberty, fertility and litter size of ewe lambs of synthetic sire and dam strains raised under different photoregimens were determined. The lambs were bred during January, May or September at 30 to 32 weeks of age. Irrespective of birth date, the lambs were reared under continuous light from birth to 5 weeks of age. From 5 to 20 weeks of age, they were kept under 16 hours of light dairy (16L:8D; Treatment A), 8 hours of light daily (8L:16D; Treatment B), or a split photoperiod of 8 hours total light daily (7L:9D:1L:7D; Treatment C). Subsequently, all lambs were exposed to 9 hours of light daily until after breeding. Lambs were exposed to rams for two estrous periods after treatment with fluorogestone acetate-impregnated intravaginal sponges and pregnant mares' serum gonadotropin (PMSG) to induce synchronized estrus. Although the age at puberty (174 days) was similar among treatments, the incidence of puberty prior to progestagen sponge treatment was higher ( approximately 50%) for lambs reared under Treatments A and C than under Treatment B. Fertility and litter size of lambs were not influenced by the previous photoperiod history or by sexual maturity, i.e., puberal or prepuberal, at the start of the sponge treatment. However, strain, age and weight of lambs at breeding influenced significantly the reproductive outcome.  相似文献

Scientific research has yet to conclusively determine the optimal cage size for mice. The authors examined the effect of cage size on mouse breeding performance and on offspring behavior, which can serve as indications of overall well-being. They housed breeding trios of C57BL/6Tac mice in standard or large individually ventilated cages and measured four reproductive parameters: litter size; litter survival to weaning age; average pup weight at 7, 14 and 21 days; and the number of days between litter births. They investigated the behavior of a subset of male and female pups from parents housed in cages of each size in the elevated plus maze test, the open field assay and the acoustic startle test. Cage size had no significant effect on any of the reproductive parameters measured and few or inconsistent effects on behavior in weaned pups.  相似文献

Germline mutation induction at mouse repeat DNA loci by chemical mutagens   总被引:4,自引:0,他引:4  
Mutation rates at two expanded simple tandem repeat (ESTR) loci were studied in the germline of male mice exposed to two monofunctional alkylating agents, ethylnitrosourea (ENU) and isopropyl methanesulfonate (iPMS), and a topoisomerase II inhibitor, etoposide. Pre-meiotic exposure to the alkylating agents resulted in a highly significant increase in ESTR mutation rate, but did not alter post-meiotically exposed cells. Pre-meiotic mutation induction by ENU and iPMS was linear within the interval of doses from 12.5 to 25mg/kg and reached a plateau at higher concentrations. Paternal exposure to etoposide resulted in ESTR mutation induction at meiotic stages but did not affect post- or pre-meiotic cells. The pattern of ESTR mutation induction after pre-meiotic and meiotic exposure to chemical mutagens was similar to that previously obtained by various traditional approaches for monitoring germline mutation in mice. The results of this study show that ESTR loci provide a new efficient experimental system for monitoring the genetic effects of chemical mutagens, capable of detecting increases in mutation rates at low doses of exposure.  相似文献

We assessed whether exposure to solar ultraviolet-B radiation (UV-B) affects the mass loss of Larrea tridentata (creosotebush) litter in the Sonoran Desert of central Arizona. We placed three types of litter (leaves, twigs, or a natural mixture of leaves, twigs, and seeds) in bags constructed of UV-B-transmitting or UV-B-absorbing filter material that allowed either 85% (near-ambient UV-B treatment) or 15% (reduced UV-B treatment) of the biologically effective solar UV-B to reach litter inside the bags. Bags were placed outdoors for 4–5 months during the winter at two sites: a balcony or on the soil surface of the desert. Mass loss of leaf litter was greater under near-ambient UV-B than reduced UV-B at both sites: 21 (near-ambient) vs. 18% (reduced) on the balcony, and 18 vs. 14% at the desert site. Mass loss of twig litter was also greater under near-ambient UV-B at the desert site. Mass loss of the natural mixture of litter was also greater when exposed to near-ambient UV-B on the balcony, and tended to be greater at the desert site. We estimate that about 14–22% of the total mass loss of leaf litter during our 4–5 month experiments was attributable to solar UV-B exposure. Leaf litter exposed to near-ambient UV-B had lower concentrations of lignin, and fats and lipids, and slightly higher concentrations of holocellulose. The greater mass loss of litter under near-ambient UV-B appeared mainly attributable to loss of lignin, although losses of fats and lipids were also appreciable. A primary reason for greater mass loss of litter under solar UV-B appeared to be photodegradation, particularly of lignin.  相似文献

The primary objective of this study was to examine a possible correlation among the three endpoints of toxicity, namely, stress gene expression (hsp16), feeding, and acetylcholinesterase (AChE) activity in transgenic C. elegans (hsp16‐lacZ) exposed to sublethal concentrations of dichlorvos, an organophosphorus insecticide. Worms exposed to dichlorvos (at 5, 40, and 80 μM) exhibited a concentration‐dependent inhibition in feeding with total cessation in feeding occurring beyond 4 h of exposure. Concomitantly, marked and dose‐dependent inhibition (69%–90%) of AChE was also evident after 4 h of exposure. Induction of heat shock protein (Hsp) was evident after 4 h of exposure (as seen from quantitative analysis), although maximum expression of Hsp was evident only after 24 h of exposure (as evident from qualitative analysis). Interestingly, the Hsp induction was restricted only to the pharyngeal region. Significant correlation was discernible between the three evaluated end points suggesting their possible interrelated role in the physiological dysfunctions evoked by sublethal concentrations of dichlorvos. © 2009 Wiley Periodicals, Inc. House mice (Mus domesticus) in the Victorian mallee region of southeastern Australia show irregular outbreaks. Changes in reproductive output that could potentially drive changes in mouse numbers were assessed from 1982 to 2000. Litter size in females is positively correlated with body size. When standardized to an average size female, litter size changes seasonally from highest in spring to lowest in autumn and winter. Litter size is depressed throughout breeding seasons that begin when the abundance of mice is high, but is similar in breeding seasons over which the abundance of mice increases rapidly or remains low. Breeding begins early and is extended on average by about five weeks during seasons when mouse abundance increases rapidly. The size at which females begin to reproduce is larger during breeding seasons that begin when mouse abundance is high. An extended breeding season that begins early in spring is necessary for the generation of a house mouse plague, but it is not in itself sufficient. Reproductive changes in outbreaks of house mice in Australia are similar but not identical to reproductive changes that accompany rodent population increases in the Northern Hemisphere. We conclude that food quality, particularly protein, is a probable mechanism driving these reproductive changes, but experimental evidence for field populations is conflicting.  相似文献

In order to evaluate the importance of gestational age in possible effects due to exposure to a 20 kHz sawtooth magnetic field, pregnant ICR mice at gestational 2.5-15.5 days post-coitus, which is the most sensitive stage for the induction of major congenital malformations, were exposed in a carrousel irradiator. The mice were exposed to a 20 kHz intermediate frequency (IF) sawtooth magnetic field had a 6.5 microT peak intensity for 8 h/day. The animals were sacrificed on the 18th day of gestation; and the fetuses were examined for mortality, growth retardation, changes in head size, and other morphological abnormalities. From the above conditions, it is concluded that the exposure to a 20 kHz sawtooth magnetic field with 6.5 microT peak intensity does not inflict any adverse effect on fetuses of pregnant mice.  相似文献
