Eighteen different superoxide dismutases from procaryote, plant, fish, bird and mammalian species have been tested for anti-inflammatory activity in the rat paw pad carrageenan-induced inflammation model Very large differences in activity are observed. Homologous rat Cu-SOD is not active and indeed shows slight pro-inflammatory activity. The different SODs have different iso-electric values, different metal; (Cu, Mn or Fe) at the active centre, different molecular weights and different circulation lifetimes Biological activity is a function of amino acid sequence rather than of such secondary parameters.  相似文献

Various superoxide dismutases from different sources, containing Cu, Mn or Fe at the active centre, have been examined with respect to anti-inflammatory activity in a model using adriamycin-induced edema in rats. Very large differences in efficiency are observed, the most active being E. coli Mn-SOD and bovine Cu-SOD. TheFe-SOD from E coli is active whereas P. leiognathiFe-SODisnot. Human Mn-SOD shows no significant activity and homologous rat Cu-SOD is totally inactive. Yeast Cu-SOD shows proinflammatory properties. Anti-inflammatory activity is not a function of molecular weight or circulation life-time.  相似文献

The increase of ubiquinone content in myocardial mitochondria and succinateubiquinone reductase activity in rat blood leucocytes under hypoxic hypoxia and acute microfocal myocardial damage [table: see text] have been shown. At the same time the succinateubiquinone reductase activity of rat myocardial mitochondria do not change substantially. We simultaneously observe the dramatic drop in NADH-ubiquinone reductase activity under experimental myocarditis. This fact demonstrates higher stability of succinateubiquinone reductase system and differences in metabolical processes under hypoxic conditions of different origin. All vitamin E derivatives studied demonstrate substantial antihypoxic activity, which is connected with increased animals' survivability, ubiquinone content in myocardial mitochondria and stabilization and leveling of succinateubiquinone reducatse activity in rat blood leucocytes. alpha-Tocopherolacetate with shortened to six carbon atoms side chain is the most effective in this respect. We discuss possible mechanisms for the realization of vitamin E and its active derivative's antihypoxic effect.  相似文献

Microelectrode studies of pacemaker and non pacemaker cells in pregnant rat uterus have shown the pacemaker areas to have a constant value of RMP throughout pregnancy which was always significantly smaller than that of non pacemaker cells. The development of new pacemaker areas was associated with membrane depolarization. A number of agents and conditions which caused membrane depolarization also induced pacemaker activity in previously non pacemaker areas but did so at different levels of membrane depolarization. Potassium depolarization failed to induce pacemaker activity. It is concluded that a low level of RMP is an important factor but not sufficient alone to explain the occurence of pacemaker activity.The resting membrane potentials (RMP) of spontaneously active smooth muscles are appreciably smaller than those of nonspontaneously active muscles (3) and comparable in magnitude to those of other tissues showing spontaneous activity such as the frog sinus venosus(7), rabbit sino-auricular node (10) and embryonic heart muscle (6). In intestinal smooth muscle, where all cells appear to be spontaneously active, a clear relationship can be demonstrated between fluctuating levels of RMP and the incidence of action potential activity, and the ionic and metabolic basis of slow wave activity has been extensively investigated (5, 8). In other smooth muscles, such as the ureter and uterus, where electrical activity arises from pacemaker areas (11, 12), the underlying causes of spontaneous activity are less well understood and the relationships between pacemaker activity, RMP and excitability have not been clearly defined. As an initial approach to studying this problem we have chosen to investigate the relationship between RMP and pacemaker activity in the uterus of the pregnant rat.  相似文献

The atrial natriuretic activity of rat heart has been found to exist in multiple forms. One of these factors has been purified to apparent homogeneity by a combination of gel filtration and high pressure liquid chromatography in two different systems and its amino acid composition determined. The purified active peptide is shown to have a molecular weight of approximately 3800. In addition, the vasorelaxant activity of rat atrium has been purified and found to cochromatograph with the natriuretic activity in all chromatographic systems employed. Thus, the vasorelaxant activity resides in the natriuretic factor. The existence of this new multifunctional peptide implies a higher level of complexity for cardiovascular control of blood volume and pressure.  相似文献

Anti-arrhythmic activity of some piperazine derivatives has been investigated upon cardiac arrythmia induced by aconitine in albinos rat. Among mono-substituted derivatives trimetazidine is active. Among di-substituted derivatives cinepazide and hydroxyzine are active but medibazine have a doubtful activity and piribedil and cinnarizine are devoid of activity.  相似文献

The steroidogenic activities of ACTH, alpha-MSH, beta-MSH as well as analogs of the hormones have been compared in rat and rabbit adrenocortical cells. ACTH is equally active in both species and the melanotropins have very low steroidogenic potency in either species. The steroidogenic potencies of the peptide analogs are strikingly similar in the two species, suggesting that the structural requirements for eliciting steroidogenesis are the same in rat and rabbit adrenocortical cells. The analog NPS-ACTH has low, comparable steroidogenic activity in both species. NPS-ACTH is a potent antagonist of ACTH-induced cAMP production in rat adrenocortical cells but acts as a weak partial agonist in rabbit adrenocortical cells. These results suggest that steroidogenesis may be mediated by receptors different from those involved in the cAMP response observed at supraphysiological concentrations of ACTH.  相似文献

Di- and tetravalent lectins purified from lima beans have mitogenic activity towards human, bovine, rabbit, rat and probably mouse lymphocytes; the effect of the mitogen varies for the different species. The mitogenic activity of the 2 lima bean lectins is related to their valency: LIM 124, the component with molecular weight 124 000 and 2 saccharide binding sites, is a weak mitogen; LIM 247, the component with molecular weight 247 000 and four saccharide binding sites, is several times more active. There are indications that the tetravalent LIM 247 exhibits B cell stimulatory activity.  相似文献

Enzyme activities of the transaminative pathway of cysteine metabolism in various rat tissues were examined. Liver was found the most active tissue, followed by kidney and heart. Liver and kidney were more pronounced in mercaptopyruvate sulfurtransferase activity than in cysteine transaminase activity; heart was more active in the latter. Red blood cells, which have pronounced sulfurtransferase activity, exhibited no transaminase activity, indicating the pathway is negligible in this tissue.  相似文献

The glycosaminoglycan heparin inhibits the growth of several cell types in vitro including smooth muscle cells and rat cervical epithelial cells. The commercially available heparin which has antiproliferative activity is a structurally heterogeneous polymer that undergoes extensive modifications during maturation. In this report we have performed structure-function studies on heparin's antiproliferative activity using three different cell types: both rat and calf vascular aortic smooth muscle cells and rat cervical epithelial cells. The minimal oligosaccharide size requirements for antiproliferative activity were determined for the three cell types by using oligosaccharide fragments of defined length prepared by nitrous acid cleavage and gel filtration and a synthetic pentasaccharide. The size requirements are similar but not identical for the different cell types. Hexasaccharide fragments are antiproliferative for all three cell types but the synthetic pentasaccharide inhibits the growth of only the rat and calf vascular aortic smooth muscle cells. The interdependence between size and charge for antiproliferative activity was investigated using chemically modified oligosaccharides as well as oligosaccharides prepared from heparin and separated into fractions of differing charge by ion-exchange chromatography. There is a strong interdependence between size and charge for antiproliferative activity. For example, increasing the charge of inactive tetrasaccharide fragments by O-oversulfation makes them antiproliferative whereas reducing the charge of active larger fragments causes them to loose their antiproliferative activity. Finally the importance of 2-O-sulfate glucuronic acid moieties for antiproliferative activity was investigated using heparin preparations that lack 2-O-sulfate glucuronic acid. These compounds possess antiproliferative activity indicating that 2-O-sulfate glucuronic acid is not required for antiproliferative activity.  相似文献

采用人工模拟低气压低氧的方法比较研究了不同程度(模拟海拔5 km和7 km)和不同时间(24d和5d)低氧暴露,对大鼠和高原鼠兔(Ochotona curzoniae)下丘脑-垂体-肾上腺皮质 (hypothalamo-pituitary-adrenalcortex,HPA)轴活动的影响。结果如下:7 km低氧暴露24 h,大鼠下丘脑的促肾上腺皮质激素释放激素(corticotropin-releasing actor,CRF)和肾上腺皮质激素皮质酮分泌显著增加,大鼠HPA低氧暴露对大鼠HPA 轴活动无显著差异。低氧暴露5天后,大鼠7 km、5 km组的HPA轴活动与对照相比无明显差异。低氧暴露对高原鼠兔的HPA轴无明显影响。上述结果表明:低氧暴露的时间和程度与大鼠HPA的活动密切相关;从HPA的活动来看,高原鼠兔表现出较强的低氧耐受性。  相似文献

The first crystallographic structure of human type 3 3alpha-hydroxysteroid dehydrogenase (3alpha-HSD3, AKR1C2), an enzyme playing a critical role in steroid hormone metabolism, has been determined in complex with testosterone and NADP at 1.25-A resolution. The enzyme's 17beta-HSD activity was studied in comparison with its 3alpha-HSD activity. The enzyme catalyzes the inactivation of dihydrotestosterone into 5alpha-androstane-3alpha,17beta-diol (3alpha-diol) as well as the transformation of androstenedione into testosterone. Using our homogeneous and highly active enzyme preparation, we have obtained 150-fold higher 3alpha-HSD specificity as compared with the former reports in the literature. Although the rat and the human 3alpha-HSDs share 81% sequence homology, our structure reveals significantly different geometries of the active sites. Substitution of the Ser(222) by a histidine in the human enzyme may compel the steroid to adopt a different binding to that previously described for the rat (Bennett, M. J., Albert, R. H., Jez, J. M., Ma, H., Penning, T. M., and Lewis, M. (1997) Structure 5, 799-T812). Furthermore, we showed that the affinity for the cofactor is higher in the human 3alpha-HSD3 than the rat enzyme due to the presence of additional hydrogen bonds on the adenine moiety and that the cofactor is present under its reduced form in the active site in our preparation.  相似文献

We are interested in developing methods to rigorously characterize the intrinsic enzymatic activity of the insulin receptor. We have previously shown that the intact, kinase active form of the receptor can be separated from inactive forms isolated from human placenta. Therefore, the determination of kinase activity, when normalized to the number of receptors based on binding, is not complicated by the presence of insulin receptor forms which bind insulin normally, but are kinase inactive. We now have extended this separation technique to insulin receptor preparations from rat liver. Thus, the determination and comparison of the intrinsic kinase activity of insulin receptor from human placenta and rat liver was performed. When normalized to the same number of insulin receptors which are autophosphorylated to the same degree, the rat liver insulin receptor catalyzes the transfer of phosphate from ATP to three different substrates, on average, 2.8-fold quicker than receptor from human placenta. This probably represents an inherent difference in the intrinsic kinase activity (Vmax), since the values for KM of the substrates are essentially identical, for insulin receptors from both sources. Intrinsic kinase differences may reflect different biological roles and/or differential regulation by exogenous factors. We are now examining this hypothesis in light of reports that demonstrate regulation of intrinsic kinase activity of the insulin receptor in certain physiological and pathological states.  相似文献

To determine whether the ability to induce neurite outgrowth in rat pheochromocytoma cell line PC12 is characteristic of phospholipases of different types, we have studied the influence of phospholipase A(2) (PLA2) from cobra Naja kaouthia venom and two PLA2s from viper Vipera nikolskii venom on PC12 cells. Phospholipases from the viper venom are heterodimers in which only one of the subunits is enzymatically active, while PLA2 from the cobra venom is a monomer. It was found that all three PLA2s induce neurite outgrowth in PC12. The PLA2 from cobra venom exhibits this effect at higher concentrations as compared to the viper enzymes. We have not observed such an activity for isolated subunits of viper PLA2s, since the enzymatically active subunits have very high cytotoxicity, while the other subunits are not active at all. However, co-incubation of active and inactive subunits before addition to the cells leads to a marked decrease in cytotoxicity and to restoration of the neurite-inducing activity. It has also been shown that all enzymatically active PLA2s are cytotoxic, the PLA2 from cobra venom being the least active. Thus, for the first time we have shown that PLA2s from snake venoms can induce neurite outgrowth in PC12 cells.  相似文献

The activities of aminotransferases catalysing the transfer of amino groups from aspartate, alanine and leucine to 2-oxoglutarate in different tissues of the rat, pigeon and trout have been determined. 2. Alanine-2-oxoglutarate aminotransferase was high in the liver of the rat and trout and low in that of the pigeon. 3. Aspartate-2-oxoglutarate aminotransferase was usually the dominant aminotransferase in all tissues and was highest in oxidative tissues where the TCA cycle is active. Its activity in the various livers is not correlated with the function of aspartate in nitrogen excretion. 4. The activity of aspartate-2-oxoglutarate aminotransferase in oxidative tissues argues that aspartate in conjunction with this enzyme serves as a buffer of oxaloacetate to keep the TCA cycle running and/or to mediate the transfer of reducing equivalents across mitochondrial membranes.  相似文献

Angiotensin-converting enzyme (ACE) has been studied in different reproductive organs of the male rat, in somatic cell lines clonally derived from both rat and mouse testes, and in isolated spermatogenic cells of the mouse. Among the various reproductive organs only testis and epididymis show high levels of enzyme activity. The testicular activity is found mainly in the isolated germinal cells and residual bodies, whereas somatic cell lines contain negligible levels of activity even after addition of hormones and growth factors. Testicular homogenates, spermatogenic cells, epididymal spermatozoa, and spermatozoan cytoplasmic droplets, when fractionated by anion exchange chromatography, contain one major and one minor activity peak, whereas epididymal homogenates and epididymal secretions reveal an additional major activity peak together with the minor peak. All forms of ACE have a similar response to different modifiers, and are equally sensitive to the specific inhibitor N-[(S)-1-(ethoxycarbonyl)-3-phenylpropyl]-L-alanyl-L-proline (Enalapril). The testicular enzyme could provide a useful marker for spermatogenic maturation and/or cytoplasmic processes both in testis and epididymis. The separate epididymal peak is a secretory enzyme that may be responsible for the processing of spermatozoan plasma membrane constituents during epididymal transit, or may have a role in attacking some biologically active compounds.  相似文献

Vasostatin-1, the natural N-terminal 1-76 chromogranin A (CGA)-derived fragment in bovine sequence, has been purified from chromaffin secretory granules and identified by sequencing and matrix-assisted laser desorption time-of-flight mass spectrometry. This peptide, which displays antibacterial activity against Gram-positive bacteria at micromolar concentrations, is also able to kill a large variety of filamentous fungi and yeast cells in the 1-10 microM range. We have found that the C-terminal moiety of vasostatin-1 is essential for the antifungal activity, and shorter active peptides have been synthesized. In addition, from the comparison with the activity displayed by related peptides (human recombinant and rat synthetic fragments), we could determine that antibacterial and antifungal activities have different structural requirements. To assess for such activities in vivo, CGA and CGA-derived fragments were identified in secretory material released from human polymorphonuclear neutrophils upon stimulation. Vasostatin-1, which is stored in a large variety of cells (endocrine, neuroendocrine, and neurons) and which is liberated from stimulated chromaffin and immune cells upon stress, may represent a new component active in innate immunity.  相似文献

The anti-inflammatory activity of amylin was studied in different models of inflammation, and compared to that of CGRP. Both peptides were active against mouse ear oedema induced by croton oil and acetic acid-induced peritonitis in the rat. CGRP was more potent than amylin in both models. Pretreatment with CGRP 8–37 fragment blocked the anti-inflammatory activity of both peptides in croton oil ear oedema. No anti-inflammatory activity was evidenced against serotonin-induced rat paw oedema and plasma protein extravasation induced by dextran in rat skin. Our results suggest that amylin exerts anti-inflammatory activity only in inflammatory models characterized by a vascular component. This effect appears to be mediated by the involvement of CGRP receptors.  相似文献
