The smear method of plant cytology as described by Taylor, presented difficulties in obtaining an adequate stain with the iron-alum haematoxylin combination. The writer has largely avoided those difficulties by curtailing the times of mordanting and of staining. The smear method has been applied by the writer in the study of microsporogenesis in the monocotyledonous genera Tradescantia and Rhoeo and in the dicotyledonous genus Podophyllum. Due to the rapidity with which permanent preparations can be completed by the smear method, there is presented a valuable means for securing a more critical evaluation of the available preservatives.  相似文献

This report investigates the reasons for false negative cervical cytology in 94 out of 630 patients (15%) in whom cervical intraepithelial neoplasia (CIN) was diagnosed on colposcopically directed biopsy. Cervical smears were taken immediately before biopsy and the cases with false negative cytology were compared with those whose cytology was abnormal. Patients with false negative cytology were more likely to have been younger (P < 0.01), to have had fewer pregnancies (P < 0.001), to have had a less severe grade of dyskaryosis on their referral smear (P < 0.001), to have had no endocervical cells on the smear (P < 0.05), to have had a less severe grade of CIN on biopsy (P < 0.001), to have had no punctation visible at colposcopy (P < 0.01), and to have had no mosaic pattern seen at colposcopy (P < 0.05). We found no effect attributable to the patient's menstrual history, the interval between referral smear and colposcopy clinic visit, the smear taker or the type of spatula used to take the smear.  相似文献

Granulocytic Ehrlichia infection in sheep is common in Norway in areas with Ixodes ricinus. In this study, 2 sheep flocks that had been grazing on I. ricinus infested pastures the previous season, were blood sampled after being housed indoors for nearly 6 months during wintertime. Thirty animals from each flock were examined for granulocytic Ehrlichia infection in the peripheral blood by blood inoculation studies, stained blood smear evaluation, polymerase chain reaction (PCR) analysis and serology (IFAantibodies). The animals were sampled twice within a three-week period, the first time before and the second time after lambing. Two sheep in one flock were found Ehrlichia positive by both blood smear evaluation and PCR before lambing, and 3 sheep were found positive after lambing; 2 by blood smear examination and 3 by PCR. In the other flock, no sheep was found infected before lambing, but 2 ewes were found positive after lambing by both blood smear evaluation and PCR. In the first flock, 87% of the animals were found seropositive before lambing, and the mean antibody titre (log 10 ± SD) to E. equi was 2.45 ± 0.401. In the second flock, 40% were found seropositive before lambing, and the mean antibody titre was 1.93 ± 0.260. Seroprevalence and mean antibody titre in these 2 flocks were significantly different (p < 0.001). The present study indicates that sheep may be a reservoir host for granulocytic Ehrlichia infection from one grazing season to the next under natural conditions in Norway.  相似文献

Surface contamination of smear cheese by Listeria spp. is of major concern for the industry. Complex smear ecosystems have been shown to harbor antilisterial potential but the microorganisms and mechanisms involved in the inhibition mostly remain unclear, and are likely related to complex interactions than to production of single antimicrobial compounds. Fungus were found to take important role in ear infections of the 344 patients (CSOM 286, Otomycosis 44, Otitis externa 14), significant fungal infections (with positive smear and culture) were detected on 49%, 79.5%, 66.6% patients respectively. 84.8% patients were detected both by smear and culture, 14.1% patients by culture and 0.1% patients in smear preparation only. In CSOM patients, age predominated in 20–27 yrs group, sex in male below 30 yrs, and Aspergillus flavus, A. niger, Penicillium, A. fumigatus in mycelial fungus, Candida albicans, C. parapsillosis in yeast. But in 18 post antibiotic fungus infected patients Penicillium and A. niger were the important isolates. In otomycosis and otitis externa patients A. niger took the main role.  相似文献

Chromosome counts are reported for 372 individuals from 202 populations in 26 taxa of Acmella (Asteraceae: Heliantheae). Chromosome numbers for 15 taxa are first reports. A review of previous counts and the new reports supports a basic chromosome number of 13 for the genus. The results show that polyploidy, sometimes accompanied by hybridization and asexual reproduction, is widespread in Acmella and has contributed to the taxonomic difficulties in the genus. These factors have produced a variable polyploid pillar complex in sect. Acmella. In one taxon in this complex, A. oppositifolia var. oppositifolia, intrataxon and even intrapopulational chromosomal variation has been detected. Morphological studies in conjunction with observations of meiotic pairing suggest that most polyploids are alloploid in origin. The occurrence of polyploidy in 16 of the 27 taxa known chromosomally emphasizes the important role this process has had in speciation within Acmella. Although intrataxon chromosomal variation has limited the taxonomic utility of chromosome numbers, a few examples are presented in which these data have been valuable for separating some pairs of closely related taxa (A. decumbens var. affinis from var. decumbens and A. poliolepidica from A. oppositifolia).  相似文献

We constructed a fine physical map for a 260-kb rice BAC contig surrounding the waxy locus. In order to identify variable regions within this 260-kb as to the restriction fragments length polymorphisms and copy numbers, sixty overlapping fragments derived from the 260-kb contig were used as probes to compare their corresponding structures among the Oryza species with AA-genome. According to the hybridization patterns, each fragment was classified into four types; true single copy (class 1), single copy with a smear background (class 2), multiple copy without a smear background (class 3), and only a smear background (class 4). Out of 16 single copy (class 1 and class 2) regions obtained in this map, the one site corresponding to wx gave rise to remarkable polymorphisms among AA-genome species in Oryza. In most of the fragments observed as repetitive segments (class 4), we could not find obvious differences in the hybridization pattern. However, interestingly, one site sorted into class-3 showed copy numbers varying among the lines. The lines belonging to O. sativa O. rufipogon, O. meridionalis,and O. longistaminata possessed high-copy numbers of this fragment, whereas only a few bands were detected in the lines from O. glaberrima, O. barthii, and O. glumaepatula. Albert's method, of staining diphtheria cultures consists of staining a fixed smear for one minute (some laboratories stain for five minutes) with a solution containing toluidine blue and malachite (or methyl) green, washing with water, and applying Albert's iodine for one minute. This procedure is discussed and criticized, and in addition the mechanism of the stain is elucidated. Also, the procedure which involves staining a fixed smear for one minute with Loeffler's alkaline methylene blue solution is discussed and criticized.

To overcome the objections to the above staining methods, a different method is proposed. This consists of staining a fixed smear with an acid solution of toluidine blue, washing with water, applying Albert's iodine for one minute, washing with water, and finally applying a safranin solution for 15-20 seconds. The rate of recombination between linked inversions of the third chromosome of Drosophila ananassae has been investigated by means of the salivary gland smear technique. Different homozygous strains of the species were utilised during the present investigation. The data show that the crossover rate between heterozygous inversions of the third chromosome of D. ananassae varies among different strains. This suggests that recombination between heterozygous inversions is influenced by genic (strain) factors.  相似文献

Summary

In order to determine how long the primary root of Asparagus officinalis L. keeps living, the writer has periodically observed plants of this species grown on two soils differing in texture an pH; so he could ascertain that in the coarse-textured and less acid soil the primary root disappears in some individual already 45 days after germination, being quite lost in all of them 75 days after germination; while in the fine-textured and more acid soil such terms are respectively antedated at the 40th and 70th day after germination. The writer also relates on early development of the first order and second order lateral rootlets, and of the first and second secondary asparagus roots.  相似文献

The determination of trigger features of single neurons in afferent pathways has been one of the central problems in sensory physiology. A novel method, called Alopex, has been developed, in which response feedback is used to construct visual patterns that optimize the responses. Data are presented which show the emergence of trigger features of cells monitored in frog visual tectum. The method is checked against results obtained by scanning the visual field with a small spot. Correlations between Alopex patterns and scan patterns are generally between 0.3 and 0.5 but may be as high as 0.9 when smoothing and/or averaging procedures are applied to the Alopex patterns. The dynamics of the Alopex process are discussed and details of the algorithms are presented. The series of experiments presented here has established the validity of the method and suggests that this approach should find wide application in receptive field studies. For that purpose data on the instrumentation and software are also presented.This research has been supported by the National Institutes of Health, under grant EY 01215  相似文献

ObjectivesTo describe women''s understanding of a negative smear test result when presented using the term “normal smear result,” as required by the NHS cervical screening programme, and to evaluate the impact on understanding of different ways of presenting the residual risk inherent in such a result.DesignExperimental questionnaire based study.InterventionsParticipants were asked to imagine that they had received a normal smear result. The meaning of this result was then presented using different combinations of three different expressions of residual risk of having or developing cervical cancer over the next five years: a verbal probability of absolute risk (low risk), a numerical probability of absolute risk (1 in 5000), or a numerical probability of risk relative to an unscreened woman (five times lower).Participants1027 women aged 20 to 64.ResultsWhen informed only that their smear result was normal, 52% (80 of 153 women) of participants correctly understood that this entailed a residual risk of cervical cancer, compared with 70% (107 of 152) given the additional sentence explaining the meaning of a normal smear result using a verbal probability of absolute risk (difference 18%; 95% confidence interval 7% to 29%). Additionally, explaining the results using a numerical probability of absolute or relative risk did not increase the proportion who correctly understood that there was a residual risk of cervical cancer.ConclusionsNHS policy for reporting normal smears needs to change to make it a definite requirement that the reporting of a “normal smear result” is accompanied by a sentence stating that this means a low risk for having or developing cervical cancer in the next five years.  相似文献

The human immune response to extracellular substances (ES) from Actinobacillus actinomycetemcomitans Y4 were analyzed. Twenty-nine periodontal patients with generalized severe alveolar bone loss and 13 healthy volunteers were examined for serum IgG antibody titers against ES. Among the patients, 17 had higher antibody titers than healthy individuals (high-titer patients) but the remainder (low-titer patients) did not. Two-dimensional electrophoresis (2DE) and Western blots demonstrated that two proteins (40 and 37 kDa) and three smeared substances reacted with IgGs from high-titer patients, but not with IgGs from healthy volunteers. The low-molecular-mass smear at the acid side reacted with over 90% of all patients. This smear reacted with anti-A. actinomycetemcomitans lipopolysaccharide (LPS) monoclonal antibody which recognized LPS from each A. actinomycetemcomitans serotype. The high molecular mass smear at the acid side might be serotype-specific O-antigens of LPS. Another high molecular mass smear which located from the alkaline to the neutral side reacted with anti-serotype-b-specific capsular polysaccharide monoclonal antibody. Moreover, the 40- and 37-kDa proteins reacted with this anti-capsular polysaccharide antibody. This results suggested that 40-and 37-kDa proteins which reacted with 100% or 88% of high-titer patients might be glycoproteins linked with capsular polysaccharide.  相似文献

A method for single-chromosome microdissection and microcloning was established in forest plants using poplar (Populus tremula) as a model. By use of meristematic cell division in root tip and the wall degradation hypotonic method, well-spread poplar metaphase chromosome spreads showing low contamination were quickly prepared and fitted for chromosome microdissection. An individual chromosome 1 was microdissected from the metaphase spreads of poplar root-tip cells with a fine glass needle controlled by a micromanipulator. The dissected chromosome was amplified in vitro by theSau3A linker adaptor-mediated PCR technique, by which 200- to 3000-bp smear DNA fragments were obtained. Southern hybridization results showed that the PCR products from the single poplar chromosome were homogeneous with poplar genomic DNA, indicating that DNA from the single chromosome has been successfully amplified. Next, the second-round PCR products from the single chromosome 1 were cloned into T-easy vectors to generate a DNA library of the chromosome 1. About 3×105 recombinant clones were obtained. Evaluation based on 160 randomly selected clones showed that the sizes of the cloned inserts varied from 230–2200 bp, with an average of 800 bp. Therefore, this research suggests that microdissection and microcloning of single small chromosomes in forest plants is feasible.  相似文献

A new isolation method for labyrinthulids, marine microbes with spindle-shaped vegetative cells and gliding movement, is presented. The method for isolating labyrinthulids has been found to be more difficult and less reproducible than that for thraustochytrids, classified in the same order. So far serum seawater agar fortified with antibiotics has been proposed to be the best for isolation of labyrinthulids. The method presented here involves placing plant samples on an agar medium on which a marine bacterium, Psychrobacter phenylpyruvicus, has been grown. The new method, which utilizes fallen mangrove leaves as source material, was more than twice as effective as isolation agar medium without the bacterium. The increased effectiveness appears to derive partly from the bacterial colonies' delaying extension of fungal mycelium. The bacterium was more effective for the isolation of labyrinthulids than either the bacterium Shewanella sp. or the yeast Rhodotorula rubra.  相似文献

Since the turn of this century it has been suggested between whiles that brachiopterygians (i.e. the species of Polypterus and Calamoichthys) are actinopterygians. One of the arguments used recently by advocates of this widespread contention is the dermohyal, an exoskeletal branchial-arch element which they assert occurs in both groups. However, this argument has to be rejected: the dermohyal does not exist in brachiopterygian fishes. The writer proposes to maintain the view that brachiopterygians are not actinopterygians until better arguments to the contrary are forthcoming.  相似文献

A method has been described for the detection of maltase activity in starch gels. This method has been used to resolve the difficulties encountered when analysing animals with weak AmIB activity for Ami genotype. It was found that, with the herd of the Hannah Research Institute, the heterozygote excess observed could be explained largely on the basis of these weak AmIB bands.  相似文献

Earlier determinations of density gradient proportionality constants β0, density distributions ρ(r), and the effect of pressure on density gradients in the analytical ultracentrifuge have been of limited precision and usefulness in the study of proteins and polypeptides. Reasons for these difficulties are that numerous intermediate relationships were required in the calculations, and the density ranges studied were generally above 1.2 g/ml. Relations are derived in the present paper to directly compute β0(ρ) values and β0′(ρ) values from the original data without any intermediate expansions or approximations. Data are presented for CsCl, CsBr, and Cs2SO4 and compared with literature values. Density distributions are computed for all three salts under a wide variety of experimental conditions of density, column length, and angular velocity. These values of ρ(r) and re are obtained by a numerical iterative technique. Values obtained by this new method are compared with values obtained using closed-form expressions. The effects of pressure on the composition density gradient for the three salts given above are calculated and found to be significant for Cs2SO4 solutions.  相似文献
