Microtubule is a well-known structural protein participating in cell division, motility and vesicle traffic. In this study, we found that β2-tubulin, one of the microtubule components, plays an important role in regulating secondary metabolite deoxynivalenol (DON) biosynthesis in Fusarium graminearum by interacting with isocitrate dehydrogenase subunit 3 (IDH3). We found IDH3 negatively regulate DON biosynthesis by reducing acetyl-CoA accumulation in F. graminearum and DON biosynthesis was stimulated by exogenous acetyl-CoA. In addition, the expression of IDH3 significantly decreased in the carbendazim-resistant mutant nt167 (Fgβ2F167Y). Furthermore, we found that carbendazim-resistance associated β2-tubulin substitutions reducing the interaction intensity between β2-tubulin and IDH3. Interestingly, we demonstrated that β2-tubulin inhibitor carbendazim can disrupt the interaction between β2-tubulin and IDH3. The decreased interaction intensity between β2-tubulin and IDH3 resulted in the decreased expression of IDH3, which can cause the accumulation of acetyl-CoA, precursor of DON biosynthesis in F. graminearum. Thus, we revealed that carbendazim-resistance associated β2-tubulin substitutions or carbendazim treatment increases DON biosynthesis by reducing the interaction between β2-tubulin and IDH3 in F. graminearum. Taken together, the novel findings give the new perspectives of β2-tubulin in regulating secondary metabolism in phytopathogenic fungi.  相似文献

Summary We have examined transport and membrane binding of 6-diazo-5-oxo-l-norleucine (DON, a photoactive diazo-analogue of glutamine) and their relationships to glutamine transport in Xenopus laevis oocytes. DON uptake was stereospecific and saturable (V max of 0.44 pmol/oocyte · min and a K m of 0.065 mm). DON uptake was largely Nau+ dependent (80% at 50 m DON) and inhibited (>75%) by glutamine and arginine (substrates of the System B0,+ transporter) at 1 mm. Glutamine and DON show mutual competitive inhibition of Na+-dependent transport. Preincubation of oocytes in medium containing 0.1 mm DON for 24 or 48 hr depressed the V max for System B0,+ transport (as measured by Na+-dependent glutamine uptake), this effect was highly specific (neither d-DON nor the System B0,+ substrates glutamine and d-alanine showed any independent effect) and required Na+ ions. Glutamine (1 mm in preincubation medium) protected transport from inhibition by DON. The possibility that specific inactivation of System B0,+ by DON reflects attachment of DON to the transporter was tested by examining the binding of [14C]DON to Xenopus oocyte membranes. Oocytes incubated in 100 mm NaCl in the presence of [14C]DON for up to 48 hr showed 2.4-fold higher 14C-binding to membranes than oocytes incubated in choline chloride. Na+-dependent DON binding (31 ± 11 fmol/g membrane protein) was suppressed by external glutamine, arginine or alanine and was largely confined to a membrane protein fraction of 48–65 kDa (as assessed by SDS-polyacrylamide gel electrophoresis). The present studies indicate that DON and glutamine uptake in oocytes are both mediated by System B0,+ and demonstrate that DON binding to a particular membrane protein fraction is associated with inactivation of the transporter, offering the prospect of using [14C]DON as a covalent label for the transport protein in order to facilitate its isolation and subsequent biochemical characterization.This work was supported by The Wellcome Trust, Action Research for the Crippled Child, Ajinomoto GmbH, Pfrimmer GmbH, the Rank Prize Funds, the Medical Research Council and the University of Dundee. High ratios of dissolved organic nitrogen (DON) to dissolved inorganic nitrogen (DIN) have been suggested to favor the growth of the brown tide alga Aureococcus anophagefferens. DON could provide a particular advantage in low light levels, as occur when blooms induce self-shading. We examined the effects of varying DON:DIN ratios on the photosynthetic abilities of cultured Aureococcus at two light intensities, 93 and 17 μmol photons m−2 s−1. Glutamic acid and urea were used as DON sources, and the remainder of the nitrogen was added as nitrate.In experiments examining Aureococcus growth with varying ratios of DONGlu:DINNitrate at two light intensities in batch culture, higher growth rates and biomass were observed in treatments containing DIN than in those with DON only, which contrasts with the results of previous studies. In semi-continuous growth experiments with varying DONUrea:DINNitrate ratios, low light cultures with urea had higher growth rates than those without urea. Also, the effective target area for light absorption per cell and photosystem II efficiency were greater for the low light cultures of each nutrient treatment, particularly when DON:DIN mixtures (33 and 67% NUrea) were used. The same pattern was seen in the maximum photosynthetic rates per cell in the light-saturated (Pmcell) and in the initial slope (αcell) of the PE (photosynthesis versus irradiance) curve, and in PON, POC and chlorophyll a cell−1. This indicates that the ability of Aureococcus to acclimate to low light conditions may be enhanced by the presence of both organic and inorganic nitrogen sources. These results suggest that Aureococcus physiology and photosynthesis are different during growth on a mixture of urea-N and nitrate than when either nitrogen source is present alone. Results of this study suggest that Aureococcus may not respond to all DON substrates in the same way, and that mixtures of DON and DIN may provide for higher photosynthetic rates, especially at low light. Our results did not, however, support earlier suggestions that growth on DON alone provides the brown tide alga with a large advantage at low light levels.  相似文献

1. This study was part of the Lotic Intersite Nitrogen eXperiment (LINX); a series of identical 15NH4 tracer additions to streams throughout North America. 15NH4Cl was added at tracer levels to a Puerto Rican stream for 42 days. Throughout the addition, and for several weeks afterwards, samples were collected to determine the uptake, retention and transformation pathways of nitrogen in the stream. 2. Ammonium uptake was very rapid. Nitrification was immediate, and was a very significant transformation pathway, accounting for over 50% of total NH4 uptake. The large fraction of NH4 uptake accounted for by nitrification (a process that provides energy to the microbes involved) suggests that energy limitation of net primary production, rather than N limitation, drives N dynamics in this stream. 3. There was a slightly increased 15N label in dissolved organic nitrogen (DON) the day after the 15NH4 addition was stopped. This DO15N was < 0.02% of DON concentration in the stream water at the time, suggesting that nearly all of the DON found in‐stream is allochthonous, or that in‐stream DON production is very slow. 4. Leptophlebiidae and Atya appear to be selectively feeding or selectively assimilating a very highly labelled fraction of the epilithon, as the label found in the consumers became much higher than the label found in the food source. 5. A large spate (>20‐fold increase in discharge) surprisingly removed only 37% of in‐stream fine benthic organic matter (FBOM), leaves and epilithon. The fraction that was washed out travelled downstream a long distance (>220 m) or was washed onto the stream banks. 6. While uptake of 15NH4 was very rapid, retention was low. Quebrada Bisley retained only 17.9% of the added 15N after 42 days of 15N addition. Most of this was in FBOM and epilithon. Turnover rates for these pools were about 3 weeks. The short turnover times of the primary retention pools suggest that long‐term retention (>1 month) is minimal, and is probably the result of N incorporation into shrimp biomass, which accounted for < 1% of the added 15N.  相似文献

Ice cores were collected between 10.03.93 and 15.03.93 along a 200 m profile on a large ice floe in Fram Strait. The ice was typical of Arctic multi-year ice, having a mean thickness along the profile of 2.56 ±0.53 m. It consisted mostly of columnar ice (83%) grown through congelation of seawater at the ice bottom, and the salinity profiles were characterized by a linear increase from 0 psu at the top to values ranging between 3 and 5 psu at depth. Distributions of dissolved organic carbon (DOC) and nitrogen (DON) and major nutrients were compared with ice texture, salinity and chlorophyll a. DOC, DON, dissolved inorganic nitrogen (DIN), NH4 + and NO2 were present in concentrations in excess of that predicted by dilution curves derived from Arctic surface water values. Only NO3 was depleted, although not exhausted. High DOC and DON values in conjunction with high NH4 + levels indicated that a significant proportion of the dissolved organic matter (DOM) was a result of decomposition/grazing of ice algae and/or detritus. The combination of high NH4 + and NO2 points to regeneration of nitrogen compounds. There was no significant correlation between DOC and Chl a in contrast to DON, which had a positively significant correlation with both salinity and Chl a, and the distribution of DOM in the cores might best be described as a combination of both physical and biological processes. There was no correlation between DOC and DON suggesting an uncoupling of DOC and DON dynamics in multi year ice.  相似文献

Mature maize (Zea mays) embryos were exposed to 5, 10 and 25 µg ml–1 of deoxynivalenol (DON), zearalenone (ZEA), ochratoxin A (OA) and a mixture of zearalenone and deoxynivalenol (ZEA/DON) for 9 days. DON and the ZEA/DON combination were consistently more inhibitory of the measured parameters than either ZEA or OA. Based on the predicted additive values, it would appear that, in combination, ZEA and DON act synergistically to inhibit root and shoot growth. For ZEA alone, a concentration of 5 µg ml–1 ZEA was generally inhibitory of root and shoot elongation and fresh mass accumulation, while at 10 and 25 µg ml–1, this toxin had a stimulatory effect on these parameters. For OA, the measured effects on root and shoot growth at 5 and 25 µg ml–1 were stimulatory, while at 10 µg ml–1 OA, an inhibitory effect was observed. The variability of the percentage of extracellular dissolved organic nitrogen (DON) release (PER), along with the relationship between DON release and bacterioplankton activity, was examined during five oceanographic cruises, carried out in the upwelling region of the NW Iberian Peninsula, the SW Bay of Biscay, and a latitudinal transect in the Atlantic Ocean (50°N–35°S). Rates of nitrogen uptake, DON release, and bacterial production were measured at 66 stations and sampled between August 1998 and October 2000. The percentage of DON release relative to the gross uptake of ammonium (PERNH4+) ranged from 3 to 46%, whereas that relative to total nitrogen (NH4 + + NO3 + urea) gross uptake (PERtotal) varied between 21 and 82%. The highest values for both PERNH4+ and PERtotal were found in oligotrophic oceanic waters (<0.25 mg chlorophyll a m−3). In oceanic environments, a positive relationship was found between nitrogen uptake and DON release rates, with a log–log linear regression slope significantly lower than 1, suggesting an inverse relationship between PER and gross nitrogen uptake rates. In contrast, in areas with higher productivity levels (>6 mg chlorophyll a m−3), such as the continental shelf off the NW Iberian Peninsula, PER held constant as nitrogen uptake increased. These results suggest the dominance of different processes controlling DON release in oceanic and neritic zones. DON release rates accounted for less than 15% of the variability observed in bacterial production rates, suggesting a weak response of bacterioplankton to phytoplankton on short time scales (hours). Furthermore, nitrogen budgets showed an excess of DON release in relation to bacterial requirements.  相似文献

The aim of this study was to investigate the potential of different isolates of ectomycorrhizal (EM) fungi to enhance the growth of Pinus sylvestris seedlings in five natural peat substrates with different nitrogen concentrations, and the effect of the Scots pine seedlings and fungal inoculum on the formation of dissolved inorganic and organic nitrogen in peat. Utilization of different organic nitrogen compounds by microbial community in the peat was also investigated using Biolog MT MicroPlates. Inoculation of the seedlings with EM fungi enhanced seedling growth. Piloderma croceum increased root growth especially, whereas Lactarius rufus increased needle growth and Suillus variegatus I, II and III improved both root and needle growth. All the EM fungi also significantly affected stem growth. Nitrogen concentration of the peat did not affect seedling growth as much as the EM fungi. At the lowest peat N concentration (1.17%) NH 4 + mineralisation was lower and DON (dissolved organic nitrogen) accumulation higher than at higher peat N concentrations. The EM fungal isolates had different effects on NH 4 + and DON accumulation/degradation in peat. The EM fungal isolates significantly increased NH 4 + formation in the peat, whereas L. rufus and P. croceum had an opposite effect on DON accumulation. S. variegatus I significantly decreased the DON concentrations during peat incubation. The N concentration of the peat slightly affected the utilization of amino acids and polyamines by the microbial community, whereas inoculation with S. variegatus I, II or III had no effect.  相似文献

Evidence suggests that phytoplankton can release significant amounts of dissolved organic carbon (DOC), particularly when they are nitrogen (N) deficient. To investigate this phenomenon from the perspective of dissolved organic N (DON), 15N tracer techniques were used to trace the flow of N through two clones of Synechococcus, WH7803 and WH8018, which were grown in batch culture under N-sufficient and N-deficient conditions. At various stages during the experiment, a subsample of the culture was removed and the response to a pulse of 15N-labeled NH4+ was measured with respect to rates of NH4+ uptake, intracellular NH4+ transformation, and release of total and low-molecular-weight (LMW; <10 000 Da) DON. In contrast to what is commonly observed for DOC, high rates of DON release were not observed during N starvation in either clone. The highest rates of DON release, both absolute and as a percentage of gross N uptake, occurred during N-sufficient growth, and release rates decreased by a factor of 4-7 when NH4+ was depleted in the medium.   相似文献

Intense efforts are underway to identify inhibitors of the enzyme gamma‐glutamyl transpeptidase 1 (GGT1) which cleaves extracellular gamma‐glutamyl compounds and contributes to the pathology of asthma, reperfusion injury and cancer. The glutamate analog, 6‐diazo‐5‐oxo‐norleucine (DON), inhibits GGT1. DON also inhibits many essential glutamine metabolizing enzymes rendering it too toxic for use in the clinic as a GGT1 inhibitor. We investigated the molecular mechanism of human GGT1 (hGGT1) inhibition by DON to determine possible strategies for increasing its specificity for hGGT1. DON is an irreversible inhibitor of hGGT1. The second order rate constant of inactivation was 0.052 mM ?1 min?1 and the K i was 2.7 ± 0.7 mM . The crystal structure of DON‐inactivated hGGT1 contained a molecule of DON without the diazo‐nitrogen atoms in the active site. The overall structure of the hGGT1‐DON complex resembled the structure of the apo‐enzyme; however, shifts were detected in the loop forming the oxyanion hole and elements of the main chain that form the entrance to the active site. The structure of hGGT1‐DON complex revealed two covalent bonds between the enzyme and inhibitor which were part of a six membered ring. The ring included the OG atom of Thr381, the reactive nucleophile of hGGT1 and the α‐amine of Thr381. The structure of DON‐bound hGGT1 has led to the discovery of a new mechanism of inactivation by DON that differs from its inactivation of other glutamine metabolizing enzymes, and insight into the activation of the catalytic nucleophile that initiates the hGGT1 reaction.  相似文献

Here we report measurements of organic and inorganic nitrogen (N) fluxes from the high-elevation Green Lakes Valley catchment in the Colorado Front Range for two snowmelt seasons (1998 and 1999). Surface water and soil samples were collected along an elevational gradient extending from the lightly vegetated alpine to the forested subalpine to assess how changes in land cover and basin area affect yields and concentrations of ammonium-N (NH4-N), nitrate-N (NO3-N), dissolved organic N (DON), and particulate organic N (PON). Streamwater yields of NO3-N decreased downstream from 4.3 kg ha−1 in the alpine to 0.75 kg ha−1 at treeline, while yields of DON were much less variable (0.40–0.34 kg ha−1). Yields of NH4-N and PON were low and showed little variation with basin area. NO3-N accounted for 40%–90% of total N along the sample transect and was the dominant form of N at all but the lowest elevation site. Concentrations of DON ranged from approximately 10% of total N in the alpine to 45% in the subalpine. For all sites, volume-weighted mean concentrations of total dissolved nitrogen (TDN) were significantly related to the DIN:DON ratio (R 2 = 0.81, P < 0.001) Concentrations of NO3-N were significantly higher at forested sites that received streamflow from the lightly vegetated alpine reaches of the catchment than in a control catchment that was entirely subalpine forest, suggesting that the alpine may subsidize downstream forested systems with inorganic N. KCl-extractable inorganic N and microbial biomass N showed no relationship to changes in soil properties and vegetative cover moving downstream in catchment. In contrast, soil carbon–nitrogen (C:N) ratios increased with increasing vegetative cover in catchment and were significantly higher in the subalpine compared to the alpine (P < 0.0001) Soil C:N ratios along the sample transect explained 78% of the variation in dissolved organic carbon (DOC) concentrations and 70% of the variation in DON concentrations. These findings suggest that DON is an important vector for N loss in high-elevation ecosystems and that streamwater losses of DON are at least partially dependent on catchment soil organic matter stoichiometry. The mycotoxin deoxynivalenol (DON) causes serious problems worldwide in the production of crops such as wheat and barley because of its toxicity toward humans and livestock. A bacterial culture capable of degrading DON was obtained from soil samples collected in wheat fields using an enrichment culture procedure. The isolated bacterium, designated strain WSN05-2, completely removed 1,000???g/mL of DON from the culture medium after incubation for 10?days. On the basis of phylogenetic studies, WSN05-2 was classified as a bacterium belonging to the genus Nocardioides. WSN05-2 showed significant growth in culture medium with DON as the sole carbon source. High-performance liquid chromatography analysis indicated the presence of a major initial metabolite of DON in the culture supernatant. The metabolite was identified as 3-epi-deoxynivalenol (3-epi-DON) by mass spectrometry and 1H and 13C nuclear magnetic resonance analysis. The amount of DON on wheat grain was reduced by about 90% at 7?days after inoculation with WSN05-2. This is the first report of a Nocardioides sp. strain able to degrade DON and of the yet unknown 3-epi-DON as an intermediate in the degradation of DON by a microorganism.  相似文献

Germinating seed ofDalbergia dolichopetala converted both [2H5]l-tryptophan and [2H5]indole-3-ethanol to [2H5]indole-3-acetic acid (IAA). Metabolism of [2-14C]IAA resulted in the production of indole-3-acetylaspartic acid (IAAsp), as well as several unidentified components, referred to as metabolites I, II, IV and V. Re-application of [14C]IAAsp to the germinating seed led to the accumulation of the polar, water-soluble compound, metabolite V, as the major metabolite, together with a small amount of IAA. Metabolites I, II and IV were not detected, nor were these compounds associated with the metabolism of [2-14C]IAA by shoots and excised cotyledons and roots from 26-d-oldD. dolichopetala seedlings. Both shoots and cotyledons converted IAA to IAAsp and metabolite V, while IAAsp was the only metabolite detected in extracts from excised roots. The relationship between deoxynivalenol (DON) intake and first morning urinary DON was examined in UK adults to validate the latter as a biomarker of human exposure. DON was assessed in first morning samples collected during a period of normal diet, a wheat-restriction intervention diet, and partial wheat-restriction intervention in which bread was allowed. During the partial intervention duplicate bread portions were collected for DON analysis. During the normal diet, partial intervention and full intervention, urinary DON was detected in 198/210 (geometric mean 10.1?ng DON mg?1 creatinine, 95% confidence interval (CI) 8.6–11.6?ng mg?1; range nd–70.7?ng mg?1), in 94/98 (5.9?ng mg?1, 95% CI 4.8–7.0?ng mg?1; range nd–28.4?ng mg?1), and 17/40 (0.5?ng mg?1, 95% CI 0.3–0.7?ng mg?1; range nd–3.3?ng mg?1) volunteers, respectively. A strong correlation between DON intake and the urinary biomarker was observed (p <0.001, adjusted r2?=?0.83) in models adjusting for age, sex and body mass index. These data demonstrate a quantitative correlation between DON exposure and urinary DON, and serve to validate the use of urinary DON as an exposure biomarker.  相似文献

The objective of the present study was to demonstrate the efficiency of the decontamination process applied to deoxynivalenol (DON)-contaminated maize by sodium sulphite (Na2SO3) treatment in vivo. Additionally, in vitro characterisation of the toxicity of the DON sulphonates (DONS 1, 2 and 3 denote structurally different forms), the resulting DON metabolites, on peripheral blood mononuclear cells (PBMC) should substantiate the inactivation of DON. In a piglet experiment, both DON-contaminated maize and -uncontaminated control maize either untreated (DON?, CON?) or Na2SO3-treated (DON+, CON+) were mixed into feed and fed for 42 d starting from weaning. The results showed that feed intake and daily weight gain of animals fed DON? were significantly lower compared to animals fed CON? and CON+, whereas group DON+ reached the control level or even exceeded it. The feed-to-gain ratio was unaffected (p = 0.45). Furthermore, DON concentrations in plasma markedly reflected the diets’ DON concentrations. These were < 0.1, < 0.1, 5.4 and 0.8 mg/kg feed for CON?, CON+, DON? and DON+, and amounted to 0.3, 0.4, 33.0 and 9.3 ng/ml in plasma, respectively. Whereas DONS 2 and 3 were detected in the DON+ diet, only DONS 2 was recovered in plasma. Regarding the toxicity of DONS, no or much lower toxicity was found compared to DON. DONS 1 and Na2SO3 did not affect the viability of PBMC. At 32.71μM DONS2 the viability was reduced by 50% and thus this compound was less toxic than DON by a factor of 73. Consequently, wet preservation of maize with Na2SO3 was an effective tool to avoid the adverse effects of DON on performance of piglets.  相似文献

The effects of nutrient enrichment on the release of dissolved organic carbon and nitrogen (DOC and DON, respectively) from the coral Montipora digitata were investigated in the laboratory. Nitrate (NO3 ) and phosphate (PO4 3−) were supplied to the aquarium to get the final concentrations of 10 and 0.5 μmol l−1, respectively, and the corals were incubated for 8 days. The release rate of DON per unit coral surface area significantly decreased after the nutrient enrichment, while the release rate of DOC was constant. Because the chlorophyll a (chl a) content of zooxanthellae per unit surface area increased, the release rate of DOC significantly decreased when normalized to unit chl a. These results suggested that the incorporation of NO3 and PO4 3− stimulated the synthesis of new cellular components in the coral colonies and consequently, reduced extracellular release of DOC and DON. Actually, significant increase in N and P contents relative to C content was observed in the coral’s tissue after the nutrient enrichment. The present study has concluded that inorganic nutrient enrichment not only affects coral-algal metabolism inside the colony but also affects a microbial community around the coral because the organic matter released from corals functions as energy carrier in the coral reef ecosystem.  相似文献

Dissolved organic nitrogen (DON) potentially plays a major role in sustaining the high productivity and biological diversity of coral reefs. However, data are scarce regarding sources and sinks of DON. This study, for the first time, determined the 15N isotopic composition of total dissolved nitrogen (δ15NTDN), reflecting the isotopic signature of DON, in the water column over a coral reef. The uniformity in δ15NTDN during high tide (3.2 ± 0.3 ‰) indicated that the DON was mainly derived from offshore waters. In contrast, higher spatial heterogeneity of δ15NTDN (3.1 ± 0.9 ‰) and DON concentrations during low tide indicated the existence of local DON sources patchily distributed over the reef. Low δ15NTDN values located mid-reef were indicative of DON release from organisms that obtained their N via N2 fixation, whereas high δ15NTDN appeared to reflect localized release of DON by organisms exposed to dissolved inorganic nitrogen with elevated 15N, such as from terrestrial and offshore inputs. Collectively, the results highlight the importance of spatial patterns in DON release from reef communities in the N cycling of coral reefs.  相似文献

Mycotoxins are fungal secondary metabolites that elicit a wide spectrum of toxicological effects, including the alteration of normal immune function. In the present study we investigated the independent effect of four mycotoxins, aflatoxin B1 (AFB1), fumonisin B1 (FB1), deoxynivalenol (DON) and nivalenol (NIV), on lymphocyte proliferation using human and porcine lymphocytes. Human and porcine peripheral blood mononuclear cells and porcine splenocytes were cultured with increasing concentrations of mycotoxins for 72 hours and labelled in the last 24 hours with [methyl-3H]-thymidine. The results showed that increased concentrations of AFB1, DON and NIV affected the [methyl-3H]-thymidine cellular proliferation following mitogen stimulation in both species and cell types. Lower concentrations of mycotoxins enhanced cellular proliferation, which was more pronounced in human than in porcine cells, while higher concentrations caused a dose-dependent decrease. DON and NIV were the most potent mycotoxin in both species and both cell types. Based on the results of this in vitro study, high correlations were found between proliferation of human and porcine lymphocytes after mycotoxin exposure, especially for DON and NIV.  相似文献
