Maize (Zea mays L.) seedlings were grown in nutrient solution culture containing 0, 5, and 20 μM cadmium (Cd) and the effects on various aspects of photosynthesis were investigated after 24, 48, 96 and 168 h of Cd treatments. Photosynthetic rate (P N) decreased after 48 h of 20 μM Cd and 96 h of 5μM Cd addition, respectively. Chl a and total Chl content in leaves declined under 48 h of Cd exposure. Chl b content decreased on extending the period of Cd exposure to 96 h. The maximum quantum efficiency and potential photosynthetic capacity of PSII, indicated by Fv/Fm and Fv/Fo, respectively, were depressed after 96 h onset of Cd exposure. After 48 h of 5μM Cd and 24 h of 20 μM Cd treatments, the activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.39) and phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31) in the leaves started to decrease, respectively. We found that the limitation of photosynthetic capacity in Cd stressed maize leaves was associated with Cd toxicity on the light and the dark stages. However, Cd stress initially reduced the activities of Rubisco and PEPC and subsequently affected the PSII electron transfer, suggesting that the Calvin cycle reactions in maize plants are the primary target of the Cd toxic effect rather than PSII.  相似文献

In mature and young leaves of sunflower (Helianthus annuus L. cv. Catissol-01) plants grown in the greenhouse, photosynthetic rate, stomatal conductance, and transpiration rate declined during water stress independently of leaf age and recovered after 24-h rehydration. The intercellular CO2 concentration, chlorophyll (Chl) content, and photochemical activity were not affected by water stress. However, non-photochemical quenching increased in mature stressed leaves. Rehydration recovered the levels of non-photochemical quenching and increased the Fv/Fm in young leaves. Drought did not alter the total Chl content. However, the accumulation of proline under drought was dependent on leaf age: higher content of proline was found in young leaves. After 24 h of rehydration the content of proline returned to the same contents as in control plants.  相似文献

The amounts of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco), total chlorophyll (Chl), and total leaf nitrogen were measured in fully expanded, young leaves of wheat (Triticum aestivum L.), rice (Oryza sativa L.), spinach (Spinacia oleracea L.), bean (Phaseolus vulgaris L.), and pea (Pisum sativum L.). In addition, the activities of whole-chain electron transport and carbonic anhydrase were measured. All plants were grown hydroponically at different nitrogen concentrations. Although a greater than proportional increase in Rubisco content relative to leaf nitrogen content and Chl was found with increasing nitrogen supply for rice, spinach, bean, and pea, the ratio of Rubisco to total leaf nitrogen or Chl in wheat was essentially independent of nitrogen treatment. In addition, the ratio of Rubisco to electron transport activities remained constant only in wheat. Nevertheless, gas-exchange analysis showed that the in vivo balance between the capacities of Rubisco and electron transport in wheat, rice, and spinach remained almost constant, irrespective of nitrogen treatment. The in vitro carbonic anhydrase activity in wheat was very low and strongly responsive to increasing nitrogen content. Such a response was not found for the other C3 plants examined, which had 10- to 30-fold higher carbonic anhydrase activity than wheat at any leaf-nitrogen content. These distinctive responses of carbonic anhydrase activity in wheat were discussed in relation to CO2-transfer resistance and the in vivo balance between the capacities of Rubisco and electron transport.  相似文献

The present study was performed to investigate the adjustment of the rate parameters of the light and dark reactions of photosynthesis to the natural growth light in leaves of an overstorey species, Betula pendula Roth, a subcanopy species, Tilia cordata P. Mill., and a herb, Solidago virgaurea L., growing in a natural plant community in Järvselja, Estonia. Shoots were collected from the site and individual leaves were measured in a laboratory applying a standardized routine of kinetic gas exchange, Chl fluorescence and 820 nm transmittance measurements. These measurements enabled the calculations of the quantum yield of photosynthesis and rate constants of excitation capture by photochemical and non-photochemical quenchers, rate constant for P700+ reduction via the cytochrome b6f complex with and without photosynthetic control, actual maximum and potential (uncoupled) electron transport rate, stomatal and mesophyll resistances for CO2 transport, Km(CO2) and Vm of ribulose-bisphosphate carboxylase-oxygenase (Rubisco) in vivo. In parallel, N, Chl and Rubisco contents were measured from the same leaves. No adjustment toward higher quantum yield in shade compared with sun leaves was observed, although relatively more N was partitioned to the light-harvesting machinery in shade leaves ( H. Eichelmann et al., 2004 ). The electron transport rate through the Cyt b6f complex was strongly down-regulated under saturating light compared with darkness, and this was observed under atmospheric, as well as saturating CO2 concentration. In vivo Vm measurements of Rubisco were lower than corresponding reported measurements in vitro, and the kcat per reaction site varied widely between leaves and growth sites. The correlation between Rubisco Vm and the photosystem I density was stronger than between Vm and the density of Rubisco active sites. The results showed that the capacity of the photosynthetic machinery decreases in shade-adjusted leaves, but it still remains in excess of the actual photosynthetic rate. The photosynthetic control systems that are targeted to adjust the photosynthetic rate to meet the plant's needs and to balance the partial reactions of photosynthesis, down-regulate partial processes of photosynthesis: excess harvested light is quenched non-photochemically; excess electron transport capacity of Cyt b6f is down-regulated by ΔpH-dependent photosynthetic control; Rubisco is synthesized in excess, and the number of activated Rubisco molecules is controlled by photosystem I-related processes. Consequently, the nitrogen contained in the components of the photosynthetic machinery is not used at full efficiency. The strong correlation between leaf nitrogen and photosynthetic performance is not due to the nitrogen requirements of the photosynthetic apparatus, but because a certain amount of energy must be captured through photosynthesis to maintain this nitrogen within a leaf.  相似文献

The effects of 24-epibrassinolide (EBR) spray application on gas-exchange, chlorophyll fluorescence characteristics, Rubisco activity, and carbohydrate metabolism were investigated in cucumber (Cucumis sativus L. cv. Jinchun No. 3) plants grown in a greenhouse. EBR significantly increased the light-saturated net CO(2) assimilation rate (A(sat)) from 3 h to 7d after spraying, with 0.1 mg l(-1) EBR proving most effective. Increased A(sat) in EBR-treated leaves was accompanied by increases in the maximum carboxylation rate of Rubisco (V(c,max)) and in the maximum rate of RuBP regeneration (J(max)). EBR-treated leaves also had a higher quantum yield of PSII electron transport (phi(PSII)) than the controls, which was mainly due to a significant increase in the photochemical quenching (q(P)), with no change in the efficiency of energy capture by open PSII reaction centres (F'(v)/F'(m)). EBR did not influence photorespiration. In addition, significant increases in the initial activity of Rubisco and in the sucrose, soluble sugars, and starch contents were observed followed by substantial increases in sucrose phosphate synthase (SPS), sucrose synthase (SS), and acid invertase (AI) activities after EBR treatment. It was concluded that EBR increases the capacity of CO(2) assimilation in the Calvin cycle, which was mainly attributed to an increase in the initial activity of Rubisco.  相似文献

Photosystem II (PSII) photochemistry was examined by chlorophyll (Chl) a fluorescence analysis in high-yield rice LYPJ flag leaves during senescence. Parameters deduced from the JIP-test showed that inhibition of the donor side of PSII was greater than that of the acceptor side in hybrid rice LYPJ. The natural senescence process was accompanied by the increased inactivation of oxygen-evolving complex (OEC) and a lower total number of active reaction centers per absorption. It indicated that the inhibition of electron transport caused by natural senescence might be caused partly by uncoupling of the OEC and/or inactivation of PSII reaction centers. Chl fluorescence parameters analyzed in this study suggested that energy dissipation was enhanced in order to protect senescent leaves from photodamage. Nevertheless, considerably reduced PSI electron transport activity was observed at the later senescence. Thus, natural senescence inhibited OEC-PSII electron transport, but also significantly limited the PSII-PSI electron flow.  相似文献

The ecophysiological responses of the homoiochlorophyllous desiccation-tolerant (HDT) plant Haberlea rhodopensis showed that this plant could tolerate water deficit and both leaves and roots had high ability to survive severe desiccation. The changes and correlation between CO2 assimilation, stomatal conductance, contents of photosynthetic pigments, root respiration and specific leaf area during dehydration–rehydration cycle were investigated. The physiological activity of leaves and roots were examined in fully hydrated (control) plants and during 72 h of dehydration, as well as following 96 h of rehydration every 6 and 24 h. After 6 h of dehydration, the stomatal conductance declined and the intercellular CO2 concentration increased. The reduction in CO2 assimilation rate was observed after 54 h of dehydration. There was a good correlation between the root respiration and water content. Our results showed that the plasticity of adaptation in leaves and roots were different during extreme water conditions. Roots were more sensitive and reacted faster to water stress than leaves, but their activity rapidly recovered due to immediate and efficient utilization of periodic water supply.  相似文献

Young poplar trees (Populus tremula Michx. x Populus alba L. clone INRA 717-1B4) were subjected to 120 ppb of ozone for 35 days in phytotronic chambers. Treated trees displayed precocious leaf senescence and visible symptoms of injury (dark brown/black upper surface stippling) exclusively observed on fully expanded leaves. In these leaves, ozone reduced parameters related to photochemistry (Chl content and maximum rate of photosynthetic electron transport) and photosynthetic CO(2) fixation [net CO(2) assimilation, Rubisco (ribulose-1,5-bisphosphate carboxylase oxygenase) activity and maximum velocity of Rubisco for carboxylation]. In fully expanded leaves, the rate of photorespiration as estimated from Chl fluorescence was markedly impaired by the ozone treatment together with the activity of photorespiratory enzymes (Rubisco and glycolate oxidase). Immunoblot analysis revealed a decrease in the content of serine hydroxymethyltransferase in treated mature leaves, while the content of the H subunit of the glycine decarboxylase complex was not modified. Leaves in the early period of expansion were exempt from visible symptoms of injury and remained unaffected as regards all measured parameters. Leaves reaching full expansion under ozone exposure showed potential responses of protection (stimulation of mitochondrial respiration and transitory stomatal closure). Our data underline the major role of leaf phenology in ozone sensitivity of photosynthetic processes and reveal a marked ozone-induced inhibition of photorespiration.  相似文献

Changes in photochemical efficiency, non-radiative energy dissipation (NRD), de-epoxidation state of xanthophyll cycle components (DPS) and contents of the antioxidants ascorbic acid and glutathione were studied in leaves of the poikilohydric Ramonda serbica Panc. (Gesneriaceae) during cycles of dehydration and subsequent rehydration. In drying leaves, the intrinsic efficiency of PS II photochemistry and the photon yield of PS II electron transport showed strong progressive decreases. Simultaneously, the fraction of excitation energy dissipated as heat in the PS II antenna increased markedly. The energy-dependent component of non-photochemical quenching (NPQ) showed an increase in dehydrating leaves down to relative water contents (RWC) values near 30%. Further decreases in RWC below these values caused a decrease in NPQ. Accordingly, DPS showed a similar behaviour, with a sharp increase and a subsequent decrease at very low RWC, although the maximum DPS was reached at slightly lower RWC than that for the maximum NPQ. The pools of reduced ascorbate and glutathione increased strongly when the RWC values fell below 40% and remained high in fully dehydrated leaves. When plants were re-watered photosynthetic efficiency, NRD, DPS and antioxidant contents recovered their initial control values. However, during rehydration, the zeaxanthin content showed a transient increase, as did NPQ, indicating an increasing demand for non-radiative dissipation. On the other hand, the contents of reduced ascorbate and reduced glutathione decreased but were still relatively high in the initial phase of rehydration, when the rate of photosynthetic electron transport, proton pumping and NRD were still relatively low. These results indicate that several photoprotective mechanisms are operating in R. serbica. Protection from photo-oxidation and photoinhibition appears to be achieved by co-ordinated contributions by ascorbate, glutathione and zeaxanthin-mediated NPQ. A novel A-Ci curve (net CO2 assimilation rate of a leaf -An- as a function of its intercellular CO2 concentration -Ci) analysis method (Plant, Cell & Environment 27, 137-153, 2004) was used to estimate the CO2 transfer conductance (gi) and the maximal carboxylation (Vcmax) and electron transport (Jmax) potentials of ageing, non-senescing Pseudotsuga menziesii leaves in relation to their nitrogen (N) content and protein and pigment composition. Both gi and the stomatal conductance (gsc) of leaves were closely coupled to Vcmax, Jmax and An with all variables decreasing with increasing leaf age. Consequently, both Ci and Cc (chloroplastic CO2 concentration) remained largely conserved through successive growing seasons. The N content of leaves, as well as the amount of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and other sodium dodecyl sulfate-soluble proteins, increased during the first three growing seasons, then stabilized or decreased only slightly afterwards. Thus, the age-related photosynthetic nitrogen use efficiency (PNUE) decline of leaves was not a consequence of decreased allocation of N towards Rubisco and other proteins involved in bioenergetics and light harvesting. Rather, loss of photosynthetic capacity was the result of the decreased activation state of Rubisco and proportional down-regulation of electron transport towards the photosynthetic carbon reduction (PCR) and photorespiratory (PCO) cycles in response to a reduction of CO2 supply to the chloroplasts' stroma. This study emphasizes the regulatory potential and homeostaticity of Cc- rather than photosynthetic metabolites or Ci- in relation to the commonly observed correlation between photosynthesis and gsc.  相似文献

Photosynthetic characteristics in rice (Oryza sativa L.) leaves were examined after treatment with low temperature (15 degrees C) and high irradiance (1,500 micromol quanta m(-2) s(-1)). Decreases in quantum efficiencies in PSII (PhiPSII) and PSI (PhiPSI) and in the rate of CO2 assimilation were observed with a decrease in the maximal quantum efficiency of PSII (F(v)/F(m)) by simultaneous measurements of Chl fluorescence, P700+ absorbance and gas exchange. The decreases in PhiPSII were most highly correlated with those in CO2 assimilation. Although the initial (the activity immediately measured upon extraction) and total (the activity following pre-incubation with CO2 and Mg2+) activities of ribulose-1,5-bisphosphate (RuBP) carboxylase/oxygenase (Rubisco) decreased slightly, the maximal activity (the activity following treatment with SO4(2-)) of Rubisco remained almost constant. These results indicate that the decrease in CO2 assimilation rate with the decreasing F(v)/F(m) was not caused by a decrease in Rubisco activity but rather by a decrease in RuBP regeneration capacity which resulted from the decrease in the rate of the linear electron transport. On the other hand, the decrease in PhiPSI was very small and the ratio of PhiPSI to PhiPSII increased. The de-epoxidation state of xanthophyll cycle pigments also increased. Thus, the cyclic electron transport around PSI occurred in photoinhibited leaves.  相似文献

Protease activities and its relation to the contents of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and Rubisco activase were investigated in detached leaves of rice (Oryza sativa L.) floated on the solutions containing abscisic acid (ABA) or benzyladenine (BA). Rubisco and Rubisco activase contents were decreased during the time course and the decreases were enhanced by ABA and suppressed by BA. The decrease in Rubisco activase was faster than that in Rubisco. SDS-dependent protease activities at 50–70 kDa (rice SDS-dependent protease: RSP) analyzed by the gelatin containing PAGE were significantly enhanced by ABA. RSPs were also increased in attached leaves during senescence. RSPs had the pH optimum of 5.5, suggesting that RSPs are vacuolar protease. Both decrease in Rubisco and Rubisco activase contents and increase in RSPs activities were suppressed by cycloheximide. These findings indicate that the activities of RSPs are well correlated with the decrease in these protein contents. Immunoblotting analysis showed that Rubisco in the leaf extracts was completely degraded by 5 h at pH 5.5 with SDS where it was optimal condition for RSPs. However, the degradation of Rubisco did not proceed at pH 7.5 without SDS where it is near physiological condition for stromal proteins. Rubisco activase was degraded at similar rate under both conditions. These results suggest that RSPs can functions in a senescence related degradation system of chloroplast protein in rice leaves. Rubisco activase would be more susceptible to proteolysis than Rubisco under physiological condition and this could affect the contents of these proteins in leaves.  相似文献

The physiological photosynthetic characteristics and antioxidant enzyme system of the high-chlorophyll rice (Oryza sativa L.) mutant (Gc) and its wild type (Zhenshan 97B) were compared and analyzed. Resulting data showed that the total chlorophyll (Chl) and Chl b contents in the Gc mutant were significantly increased by 19.0 and 81.7%, respectively, while the increase in Chl a and thylakoid membrane protein contents was insignificant. The net photosynthetic rate (P N) was significantly higher in the mutant; stomatal conductance, intercellular CO2 concentration, and transpiration rate decreased significantly, and water-use efficiency increased significantly, indicating the higher photochemical efficiency of the mutant. The chlorophyll fluorescence parameters: electron transport rate and effective quantum yield of PSII photochemistry of the mutant were significantly higher than those of Zhenshan 97B. The nonphotochemical quenching of the mutant under light adaptation increased by 52.3%. The enzymatic activity of superoxide dismutase, peroxidas, and catalase in the mutant roots and leaves were all higher than those for the wild-type plants. It is believed that the higher activity of antioxidant enzymes in the mutant may be an important factor making difficult the photo-inactivation of Chl, and thus, increasing the content of Chl, especially Chl b.  相似文献

We studied the development of chloroplasts from etioplasts in the cotyledonary leaves of 4-d-old dark-grown cucumber (Cucumis sativus) seedlings after irradiation (20 μmol m-2 s-1). Upon irradiation, the triggering of chlorophyll (Chl) synthesis and accumulation showed a relatively short lag phase. The irradiation of etiolated seedlings initiated the synthesis of apoproteins of pigment-protein complexes. While Chl-protein 2 (CP2) was detected at 6 h after irradiation, CP1 only after 29 h. The appearance and accumulation of some of the apoproteins were monitored by Western-blotting. LHC2 apoprotein was detected after a 6 h-irradiation. The amounts of D1 protein of photosystem (PS) 2 and PsaA/B protein of PS1 were quantitated by ELISA. Further, the thylakoid membrane function during this time period in terms of PS1- and PS2-mediated electron transfer activity and intersystem electron pool size were analyzed. While PS1 activity was detected after 4 h, PS2-mediated O2 evolution was detected only after a 17 h-irradiation. Fv/Fm value of Chl a fluorescence measurements indicated that the photochemical efficiency of these leaves reached its maximum after 29 h of irradiation. The intersystem pool size of cotyledonary leaves was equivalent to that of the control cotyledonary leaves grown for 25 h under continuous irradiation. Thus etioplasts develop into fully functional chloroplasts after approximately 25 h when 4 d-dark grown cucumber seedlings are continuously moderately irradiated. The development of photosynthetic electron transport chain seems to be limited in time at the level of PS2, possibly at the donor side. The objectives of this work using Phaseolus vulgaris were to examine whether the light spectrum incident on mature primary leaves (PLs) is related to leaf‐to‐leaf systemic regulation of developing trifoliate leaves (TLs) in photosynthetic characteristics, and to investigate the relative importance of spectrum and photosynthetic photon flux density (PPFD) in light‐induced systemic regulation. Systemic regulation was induced by altering PPFD and the spectrum of light incident on PLs using a shading treatment and lighting treatments including either white, blue, green or red light‐emitting diodes (LEDs). Photosynthetic characteristics were evaluated by measuring the light‐limited and light‐saturated net photosynthetic rates and the amounts of nitrogen (N), chlorophyll (Chl) and ribulose‐1,5‐bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39). Shading treatment on PLs decreased the amounts of N, Chl and Rubisco of TLs and tended to decrease the photosynthetic rates. However, we observed no systemic effects induced by the light spectrum on PLs in this study, except that a higher amount of Rubisco of TLs was observed when the PLs were irradiated with blue LEDs. Our results imply that photoreceptors in mature leaves have little influence on photosynthetic rates and amounts of N and Chl of developing leaves through systemic regulation, although the possibility of the action of blue light irradiation on the amount of Rubisco cannot be ruled out. Based on these results, we concluded that the light spectrum incident on mature leaves has little systemic effect on developing leaves in terms of photosynthetic characteristics and that the light‐induced systemic regulation was largely accounted for by PPFD.  相似文献

The ability of leaves to acclimate photosynthetically to low temperature was examined during leaf development in winter rye plants ( Secale cereale L. cv. Puma) grown at 20°C or at 6°C. All leaves grown at 6°C exhibit increased chlorophyll (Chl) levels per leaf area, higher rates of uncoupled, light-saturated photosystem I (PSI) electron transport, and slower increases in photosystem II (PSII) electron transport capacity, when compared with 20°C leaves. The stoiehiometry of PSI and PSII was estimated for each leaf age class by quantifying Chl in elcctrophorctic separations of Chl-protein complexes. The ratio of PSII/PSI electron transport in 20°C leaves is highly correlated with the ratio of core Chl a -proteins associated with PSII (CPa) to those associated with PSI (CP1). In contrast, PSII/PSI electron transport in 6°C leaves is not as well correlated with CPa/CP1 and is related, in part, to the amount and organization of light-harvesting Chl a/b -proteins associated with PSII. CPa/CP1 increases slowly in 6°C leaves, although the ratio of CPa/CP1 in mature 20°C and 6°C leaves is not different. The results suggest that increased PSI activity at low temperature is not related to an increase in the relative proportion of PSI and may reflect, instead, a regulatory change. Photosynthetic acclimation to low environmental temperature involves increased PSI activity in mature leaves shifted to 6°C. In leaves grown entirely at 6°C, however, acclimation includes both increased PSI activity and modifications in the rate of accumlation of PSII and in the organization of LHCII.  相似文献

Caragana korshinskii Kom. is a perennial xerophytic shrub, well known for its ability to resist drought. In order to study ecophysiological responses of C. korshinskii under extreme drought stress and subsequent rehydration, diurnal patterns of gas exchange and chlorophyll (Chl) fluorescence parameters of photosystem II as well as Chl content were analyzed. Plant responses to extreme drought included (1) leaf abscission and using stem for photosynthesis, (2) improved instantaneous water-use efficiency, (3) decreased photosynthetic rate and partly closed stomata owing to leaf abscission and low water status, (4) decreased maximum photochemical efficiency of photosystem II (PSII) (variable to maximum fluorescence ratio, Fv/Fm), quantum efficiency of noncyclic electron transport of PSII, and Chl a and Chl b. Four days after rehydration, new leaves budded from stems. In the rewatered plants, the chloroplast function was restored, the gas exchange and Chl fluorescence returned to a similar level as control plant. The above result indicated that maintaining an active stem system after leaf abscission during extreme drought stress may be the foundation which engenders these mechanisms rapid regrowth for C. korshinskii in arid environment.  相似文献

In canopy shade leaves of grapevine (Vitis vinifera L. cv. Moscato giallo) grown in the field the contents of chlorophyll (Chl), carotenoids (Car), and soluble protein per fresh mass were lower than in sun leaves. RuBPC activity, in vivo nitrate reductase activity (indicator of nitrate utilisation), apparent electron transport rate, and photochemical fluorescence quenching were also significantly reduced in canopy shade leaves. When various photosynthetic activities were followed in isolated thylakoids, canopy shade leaves exerted a marked inhibition of whole chain and photosystem (PS) 2 activity. Smaller inhibition of PS1 activity was observed even in high-level canopy shade (HS) leaves. The artificial exogenous electron donors, DPC and NH2OH, significantly restored the loss of PS2 activity in HS leaves. Similar results were obtained when Fv/Fm was evaluated by Chl fluorescence measurements. The marked loss of PS2 activity in canopy shade leaves was due to the loss of 47, 43, 33, 28–25, 23, 17, and 10 kDa polypeptides.  相似文献
