The inhibitory effect of halothane on the emetic response in the ferret   总被引:1,自引:0,他引:1  
Emesis and nausea are often associated with anaesthesia and continue to be a common clinical problem. Past clinical studies have demonstrated that halothane produces a higher incidence of vomiting compared with other anaesthetics, but some investigators have described an antiemetic effect. The purpose of this study was to investigate the effects of various doses of halothane on the emetic response in the decerebrate ferret. Following a control emetic response, a maximum of six increasing cumulative concentrations of halothane were delivered. At the end of each delivery period, the supradiaphragmatic vagal communicating branch, which has been shown to reproducibly elicit vomiting, was electrically stimulated and the emetic response was monitored. An increase in halothane concentration produced a marked depression of tongue, abdominal muscle, and diaphragm EMG activity as well as a decrease in central venous pressure. Licking, a prodromal response comparable to nausea in the human, appeared to be most sensitive. An increase in latency of the emetic response occurred as the concentration of halothane was increased. All phases of the response were observed at concentrations below 0.6 vol% halothane. At 0.6 vol% halothane, 75% of the animals vomited. At higher concentrations, the emetic response was completely abolished. One hour post-halothane, all latencies had returned to near control values. The methods utilized in this study provided a model that was not complicated by a large number of variables usually present in clinical studies. These data demonstrate that halothane exerts an inhibitory, concentration-dependent, and reversible effect on the emetic response in the ferret and provide further support that halothane alone does not possess emetic properties at clinical properties at clinical concentrations.  相似文献

Beef cows fed 0.37 Kg crude protein/day during the last four months of pregnancy had significantly shortened gestation (274 days vs. 282 days) and decreased weight gain (25 Kg vs. 66 Kg) compared to control cows fed 0.96 Kg crude protein/day. Calf mortality in the low protein group was associated with either dystocia (3 calves) or prematurity (2 calves). There were no deaths among control calves. It is suggested that protein malnutrition in late pregnancy may be a contributing factor to neonatal mortality among beef calves.  相似文献

Summary Although the regulatory activity of steroid hormones on amino acid metabolism has been described, no information is published on the effect of ovariectomy. We studied the influence of ovariectomy in Wistar rats determining the amino acids phenylalanine and tyrosine in liver, kidney, plasma and urine. 32 animals were used in the study, 12 animals were sham operated, 9 animals were ovariectomized and 11 rats were ovariectomized and supplemented with estradiol. No quantitative changes were detected comparing liver and kidney phenylalanine and tyrosine between the groups (sham operated rats liver phenylalanine 2,53nM/mg ± 1,07; liver tyrosine 1.95nM/mg ± 0.92; kidney phenylalanine 2.16nM/mg ± 0.53; kidney tyrosine 1.80nM/mg ± 0.39. Ovariectomized rats showed liver phenylalanine 3.07nM/mg ± 1.14; liver tyrosine 2.63nM/mg ± 1.01; kidney phenylalanine 2.30 nM/mg ± 0.74; kidney tyrosine 1.93nM/mg ± 0.63. Ovariectomized and estradiol supplemented rats presented with liver phenylalanine 2.84nM/mg ± 1.40; liver tyrosine 2.35nM/mg ± 1.28; kidney phenylalanine 1.91nM/mg ± 0.26, kidney tyrosine 1.67nM/mg ± 0.23.). When, however, the phenylalanine/tyrosine ratio in the liver was evaluated, ovariectomized rats showed a significant decrease of the quotient (p = 0.001). The phenylalanine/tyrosine ratio was restored by estradiol replacement. Our findings show that phenylalanine and tyrosine metabolism is under estradiol control. The effect on the metabolic changes could be mediated by enzyme systems as phenylalanine hydroxylase, tyrosine hydroxylase and tyrosine aminotransferase. Our results would be compatible with previous reports on the stimulatory effect of estradiol on these enzymes. The kidney phenylalanine/tyrosine ratio was unaffected by ovariectomy and/or estradiol replacement which can be easily explained by different pools, enzyme activities, filtration/reabsorption effects, etc.The urinary P/T ratio was decreased by ovariectomy and restored by estradiol replacement indicating endocrine control of renal reabsorption and secretion mechanisms.  相似文献

Hexokinase, an enzyme that is capable of regulating the entry of glucose into metabolic sequences, is known to exist in four isozyme forms in a variety of mammalian tissues. Each of these isozymes possesses different kinetic properties, suggesting that they may serve in different regulatory capacities. In addition, the proportions of the four isozymes are characteristically different for various differentiated tissues, presumably reflecting different metabolic capabilities of the tissues.Extracts of rabbit and human placentas from early gestational age and term pregnancies were chromatographed and assayed for hexokinase activity. Four peaks of activity were observed. Elution positions of the four placental hexokinase isozymes were comparable to those from liver; however, the relative proportions were considerably different. In both rabbit and human placentas, the proportion of Type I hexokinase increased during gestation and Type III decreased, while Types II and IV remained essentially unchanged. Implications of the gestational changes for placental carbohydrate metabolism are discussed.  相似文献

The contractile response to acidosis is the final product of a number of different changes in the excitation-contraction coupling pathway: (i) Cai increases and subsequently decreases during acidosis; (ii) the action potential becomes longer; (iii) the sensitivity of the contractile proteins to Ca2+ decreases. The increase of Cai and the lengthening of the action potential may help to maintain contractile function, although this advantage may be offset if spontaneous Ca2– release from the s.r. occurs, secondary to the increase of Cai. The recovery of force shown in figure 1 occurs at a time when the calcium transient is decreasing, and therefore represents an increasing sensitivity of the contractile proteins to Cai, probably due to a recovery of intracellular pH(6), although it is also possible that a disappearance of spontaneous Ca2+ releases from the s.r. may be contributing [2].  相似文献

The vomeronasal organ (VNO) is known to play a major role in sexual behavior in many mammals. This study is the first report that the adult male ferret has a VNO, which is considerably smaller and morphologically different from the usually crescent-shaped epithelium in several mammalian species, particularly rodents. There were no differences in the size or structure of the ferret VNO between the mating season in spring and the sexually quiescent season in autumn, although plasma testosterone, testis size and brain size are dramatically increased in spring and behavior changes significantly. The histological data suggest that the VNO might be not as important a structure in male ferret sexual behavior as in rodents.  相似文献

The relationship between calcitonin (CT) and prolactin (PRL) was studied by means of the injection of salmon calcitonin (SCT) i.p. on day 1 of gestation. An estrogen inhibitor - clomiphene - was also administered to certain groups of animals on day 4 and 5 of gestation. SCT did not affect PRL levels on day 1 of gestation nor on days 5 or 7, but it prevented the rise of PRL levels observed in animals submitted to injection stress on days 4 and 5. In animals treated with clomiphene, the inhibition by SCT on PRL increase after injection stress was partially abolished. SCT while not affecting basal PRL level prevented the rise observed after stress and this effect occurred some days later. Thus SCT could exercise a delayed neuroendocrine control. This action of SCT seemed to be partially dependent upon the presence of estrogens.  相似文献

The effects of terminating pregnancy by means of ovariectomy (O), hysterectomy (H), or ovariectomy plus hysterectomy (OH) at different stages of gestation upon the latency to initiation of maternal behavior were examined in primigravid rats. O, H, or OH on either Day 13 or 17 of pregnancy resulted in an accelerated onset of maternal behaviors (median range = 1.0–2.0 day latency for O, H, or OH animals vs 4.0–5.0 day latency for sham-operated intacts). However, O and H on Day 8 of pregnancy were ineffective in inducing a rapid onset of maternal behavior, while OH on Day 8 of pregnancy only slightly facilitated the onset of maternal behavior when compared to animals sham-operated on Day 8 of pregnancy. O, H, and OH on either Day 13 or 17 of gestation were also significantly more effective in rapidly inducing maternal behaviors than the corresponding surgeries performed on Day 8 of pregnancy. These data suggest that the onset of maternal behavior in the pregnant rat can be rapidly induced after mid-pregnancy by surgical procedures that presumably result in a rapid decline in serum steroids of ovarian origin.  相似文献

Summary As steroid hormones are known to influence amino acid metabolism we tested the hypothesis that ovariectomy should lead to significant changes in this system.We found that after ovariectomy serum alanine was significantly decreased (p = 0.0006) in contrast to serum glycine and branched chain amino acids (BCAA). The ratio of glycine/BCAA, a parameter for anabolism or catabolism was not changed after ovariectomy. If, however, the amino acid alanine as the link to carbohydrate and lipid metabolism was introduced the alanine/BCAA ratio was significantly altered (p = 0.01).Although serum cholesterol was altered as well (increased,p = 0.03), no significant correlation with alanine was found. We can therefore assume that there are two independent mechanisms for lipid and amino acid changes after ovariectomy.The most prominent finding was that estradiol replacement after ovariectomy restored increased cholesterol levels but did not restore alanine levels. Other ovarial hormones must be incriminated for the regulation of alanine metabolism. The anabolic effects of estradiol as decreasing glycine and BCAA were noticed which rules out insufficient estradiol replacement.  相似文献

1. The preparation of cell suspensions by treatment of chick embryo hearts with collagenase at various stages of development is described. 2. Measurements of oxygen consumption, incorporation of labelled leucine into protein and accumulation of labelled alpha-aminoisobutyric acid against a concentration gradient indicated a long-lasting viability of the isolated heart cells in vitro; a satisfactory preservation of subcellular structures, including plasma membrane, was assessed by electron-microscopic examination. 3. The rate of alpha-aminoisobutyric acid accumulation by cardiac cells isolated from hearts at different stages of embryological development decreased with aging; insulin stimulated the intracellular accumulation of this amino acid analogue. 4. Insulin increased the uptake by isolated heart cells of several (14)C-labelled naturally occurring amino acids; however, the fraction of amino acid taken up by the cells that was recovered free intracellularly, and therefore the concentration ratio (between intracellular water and medium), was enhanced by the hormone only with glycine, proline, serine, threonine, histidine and methionine. When isolated heart cells were incubated in the presence of a mixture of labelled amino acids, the addition of insulin increased the disappearance of radioactivity from the medium. 5. The general pattern of amino acid transport (in the absence and in the presence of insulin) in isolated cardiac cells was similar to that found in intact hearts, suggesting that the biological preparation described in this paper might be useful for studies of cell permeability and insulin action.  相似文献

The effect of ovariectomy on the quality and quantity of cancellous bone using the young cynomolgus monkey was evaluated after a 2-year period. The bodies of the second lumbar vertebrae were analyzed for changes in bone mineral quality using density fractionation, chemical analysis, and X-ray diffraction techniques. Changes in bone tissue quality and quantity were evaluated using bone histomorphometry and image analysis. The experimental group (n=14) was made surgically menopausal (bilaterally ovariectomized), compared with intact controls (n=16), and then sacrificed after a 2-year period. There was a non-significant shift in the mineralization profile towards less dense bone in the ovariectomized (OVX) vertebrae compared with controls. Physical characteristics of the bone mineral in terms of crystal size or strain were unaffected by OVX. There was a parallel increase in mineral content with fractions of increasing density, however there was no difference in mineral content or the Ca/P ratio in each fraction between treatment groups. Histomorphometric analysis for structural parameters demonstrated no difference in bone volume between control and OVX groups. There was no significant change in trabecular width in the OVX vertebrae compared with controls. There was a significant increase in both osteoid volume and osteoid surface in the OVX vertebrae (P<0.001). Trabecular architecture as measured by image analysis was unchanged. There was a significant increase in eroded surface in the OVX vertebrae (P<0.03) compared with the controls. In conclusion, young, ovariectomized female cynomolgus macaques do not appear to be a useful animal model for the study of postmenopausal bone loss, however they may be a useful model to evaluate skeletal pathology which might be observed after surgical ovariectomy in young human females.  相似文献

The incidence of infections was studied in 137 iron-deficient Somali nomads, 67 of whom were treated with placebo and 71 with iron. Seven episodes of infection occurred in the placebo group and 36 in the group treated with iron; these 36 episodes included activation of pre-existing malaria, brucellosis, and tuberculosis. This difference suggested that host defence against these infections was better during iron deficiency than during iron repletion. Iron deficiency among Somali nomads may be part of an ecological compromise, permitting optimum co-survival of host and infecting agent.  相似文献
