为了探讨水稻白叶枯病成株抗性是否与防卫基因相关 ,通过RT PCR研究了几种防卫基因PAL、LOX、PBZ1、Cht 1和PR1a的表达 ,同时也分析了PAL和LOX的酶活性变化 .结果表明 ,苗期PAL受病原菌诱导上调表达 ,而成株期病原菌和伤害均可诱导PAL上调表达 ,且成株期诱导表达强于苗期 .LOX在苗期和成株期都可受病原菌诱导表达 ,但成株期表达强于苗期且更持久 .PBZ1在苗期和成株期均受病原菌和伤害诱导表达 ,但成株期表达较苗期早且更强 .PR1a和Cht1没检测到期望的扩增片段 .PAL和LOX的酶活性分析结果与基因表达分析结果一致 .因此 ,PAL、LOX和PBZ1在苗期和成株期的表达差异可能与水稻白叶枯病成株抗性相关 .  相似文献

Adult stem cells maintain tissue homeostasis by their ability to both self-renew and differentiate to distinct cell types. Multiple signaling pathways have been shown to play essential roles as extrinsic cues in maintaining adult stem cell identity and activity. Recent studies also show dynamic regulation by epigenetic mechanisms as intrinsic factors in multiple adult stem cell lineages. Emerging evidence demonstrates intimate crosstalk between these two mechanisms. Misregulation of adult stem cell activity could lead to tumorigenesis, and it has been proposed that cancer stem cells may be responsible for tumor growth and metastasis. However, it is unclear whether cancer stem cells share commonalities with normal adult stem cells. In this review, we will focus on recent discoveries of epigenetic regulation in multiple adult stem cell lineages. We will also discuss how epigenetic mechanisms regulate cancer stem cell activity and probe the common and different features between cancer stem cells and normal adult stem cells.  相似文献

六星黑点豹蠹蛾Zeuzera leuconotum Butler (Lepidoptera: Cossidae)是一种重要的园林害虫。在光周期L:D=14:10、温度19-32 ℃、相对湿度75%-85%条件下,对六星黑点豹蠹蛾的羽化、性比、交配行为及能力进行了观察,利用风洞技术和林间诱蛾试验验证了其性趋向。结果表明:六星黑点豹蠹蛾羽化期60 d左右,6月下旬为羽化高峰期,日羽化高峰出现在傍晚17:00-18:00,占全天羽化量的55.4%;雌雄性比为1:1.03;交配高峰期发生在进入暗期5-7 h,成虫1日龄后性成熟,一生交配1次;随雌蛾日龄增加,其交配率逐渐下降,交配时间提前,交配持续时间逐渐加长;雌蛾一生平均产卵378个,孵化率80%,未交配雌蛾平均产卵426个,孵化率0%;未交配雌蛾和雄蛾的平均寿命分别为5.56 d和3.83 d,交配过的雌蛾和雄蛾平均寿命分别为5.66 d和2.41 d,交配缩短雄蛾寿命;六星黑点豹蠹蛾种内个体间引诱作用主要为雌蛾引诱雄蛾,同性间不存在聚集行为;2-3日龄未交配雌蛾林间诱蛾量最高,显著高于1日龄和4-6日龄(P<0.01)。依据研究结果,分析讨论了应用性信息素防治六星黑点豹蠹蛾的可行性,说明六星黑点豹蠹蛾是可利用性信息素进行防控的理想对象。  相似文献

We have analyzed methylation of the rat albumin and alpha-fetoprotein (AFP) genes by hydridizing labeled cDNA clones to HpaII and MspI digests of DNA from different stages of development. These CCGG-cutting enzymes distinguish 5-methylcystosine in mCCGG (sensitive to HpaII) and CmCGG (sensitive to MspI). In the liver, the albumin gene is heavily methylated at 18 days gestation and uniformly demethylated in the adult. The AFP gene is also heavily methylated at 18 days gestation, and develops demethylated regions at the 3' half of the gene in the adult. These methylation changes are not observed in other embryonic or adult tissues. We also evaluated expression of these genes by measuring their corresponding mRNAs. The albumin gene is actively transcribed in 18-day fetal liver, when it is heavily methylated, as well as in adult liver, when it is unmethylated. In contrast, the AFP gene is transcribed only in fetal liver, even though it is less methylated in adult liver. These findings suggest that specific methylation changes are associated with changes in gene expression, but that this association is not adequately described by the simple hypothesis that methylation turns genes off.  相似文献

Mononuclear cells from human cord blood (CBMC) are able to mount an antigen-specific IgM plaque-forming cell (PFC) response after primary in vitro stimulation with the T cell-dependent antigen ovalbumin (OA). The antigen dose-response relationship for the induction of PFC in cultures of CBMC is represented by a bell-shaped curve comparable to that found for mononuclear cells from adult peripheral blood (adult PBMC). The dose of OA optimal for the induction of a response in cultures of CBMC consistently, however, is 100-fold lower than the antigen dose optimal for adult PBMC (0.03 microgram OA/ml vs 3.0 micrograms OA/ml). Results obtained from co-culture experiments in which semiallogeneic combinations of parental/neonatal lymphocytes and monocytes were stimulated with a variable dose of OA indicate that the adherent cell (AC) plays a pivotal role in the establishment of the optimum antigen dose. From experiments using antigen-pulsed AC, it was concluded that neonatal and adult AC differ in their antigen handling capacity. In the presence of the prostaglandin synthetase inhibitor indomethacin the antigen dose-response relationship for the induction of PFC in cultures of CBMC shifts to an "adult type" of curve. From pulsing experiments it emerges that indomethacin affects the interaction between antigen and monocytes. Indomethacin causes an enhancement of the expression of HLA-DR at the surface of neonatal as well as adult AC; this can be down regulated by the addition of prostaglandin E2 (PGE2). The addition of PGE2 to cultures of adult PBMC leads to a shift of the optimal antigen dose for induction of PFC toward lower concentrations. Although higher levels of PGE2 were measured in the supernatant of cultured neonatal AC compared with adult AC, it seems unlikely that this observation can explain the distinct antigen dose-response relationship for the induction of a PFC response in cultures of CBMC.  相似文献

Social investigatory behaviour was used as a measure of olfactory recognition in two experiments to assess social memory in adult male rats. In Experiment 1, time spent in social investigation of juvenile males by 3-month-old adults was significantly higher than time spent by 7- and 11-month-old animals. Furthermore, a reexposure to the same juvenile male 30 min after the initial exposure elicited significantly less social investigation in adult males aged 7 and 11 months but not in those aged 3 months. If the reexposure occurs 2 h later, the same juvenile is thoroughly investigated by adult males irrespective of the age. The age-related differences in social recognition are discussed in terms of the internal readiness of adult males. While the social recognition was confirmed in older adult males, it is suggested that an ability to recognize the same juvenile may be masked in young animals by a high sexual arousal. Behavioural phenomenon of the social recognition was used in Experiment 2. An administration of hypothalamic MIF-I or its synthetic derivative Alaptide to adult males 7 or 11 months old immediately after their 1st exposure to a juvenile male resulted in decreasing the time spent in social investigation of the same juvenile during a reexposure performed 120 min later. Both drugs were ineffective if adult males were reexposed to a novel juvenile. The results suggest that both MIF-I and Alaptide improved an animal's capacity to store information received through olfactory cues.  相似文献

Natural selection on juveniles is often invoked as a constraint on adult evolution, but it remains unclear when such restrictions will have their greatest impact. Selection on juveniles could, for example, mainly limit the evolution of adult traits that mostly develop prior to maturity. Alternatively, selection on juveniles might primarily constrain the evolution of adult traits that experience weak or context-dependent selection in the adult stage. Using a comparative study of dragonflies, I tested these hypotheses by examining how a species’ larval habitat was related to the evolution of two adult traits that differ in development and exposure to selection: adult size and male ornamentation. Whereas adult size is fixed at metamorphosis and experiences consistent positive selection in the adult stage, ornaments develop throughout adulthood and provide context-dependent fitness benefits. My results show that species that develop in less stable larval habitats have smaller adult sizes and slower rates of adult size evolution. However, these risky larval habitats do not limit ornament expression or rates of ornament evolution. Selection on juveniles may therefore primarily affect the evolution of adult traits that mostly develop prior to maturity.  相似文献

Somatic nuclear transfer has been performed with frogs since the early 1960s, yet it has proved impossible to generate an adult frog using an adult cell as nuclear donor. After some initial skepticism, the birth of sheep, cows, goats, and mice using this technique with fetal or adult cell donors is now established fact. The success with adult mammalian cell donors extends the historic work in frogs by attesting to the totipotency of nuclei in at least some adult, differentiated cell types. Because the technique offers a developmental read out of the totality of genetic and molecular lifetime changes accumulated by the nucleus of a single somatic cell, basic research applications are seen in the fields of ageing, cancer, X chromosome inactivation, and imprinting. The prospect of a method for gene targeting in livestock holds particular promise for commercial applications; whilst for humans, the use of nuclear transfer to provide diverse populations of customized stem cells for therapeutic purposes presents a tantalizing future goal.  相似文献

Laouris et al. (1990) reported that the development of air-righting in growing new-born rats was clearly suppressed when their neck and/or back rotations were prevented. Thus, it is suggested that the neck and back muscles play an important role for a quick righting in response to drop from supine position. Further, we reported that an inhibiton of posture adjustment in response to head-down drop from a height of approximately 30 cm following chronic hindlimb suspension of adult rats was accompanied by a decrease of electromyogram (EMG) activity in both neck and back muscles (Kawano et al., 2002). However, it is still unclear how the air-righting performance is influenced by hindlimb suspension in adult rats. Thus, the effects of chronic hindlimb suspension of adult rats on the patterns and time of air-righting and role of neck and back muscles in the air-righting were investigated in the current study.  相似文献

The quality of food eaten by larval insects will affect traits such as gamete production, fat reserves, muscle bulk and body size in the adult. Moreover, larvae also depend on high moisture content in the diet for survival. The almond moth (Ephestia cautella) (W.) (Lepidoptera; Pyralidae) does not feed as an adult although it continues to drink water. We tested the idea that an almond moth could compensate for a low-water diet as a larva by increasing its water intake as an adult. We reared larvae on two different food sources with different moisture regimes; standard laboratory diet with glycerol (relatively wet) and standard diet without glycerol (relatively dry). Half the adult moths from each treatment were given water to drink before their first and only mating. Our results show that wet larval diets (i.e. containing glycerol) significantly decreased fecundity (total number of eggs laid and the proportion of hatched larvae), whilst it significantly increased male and female longevity. The interaction effect of water access for adult males and females was significant, independent of the glycerol in the larval diet. Longevity in females that were not presented with water as adults was slightly higher if mated with a male that had had access to water, suggesting a mating donation of water. However, females that received water as adults showed a decreased longevity if mated with a male who had also had access to water as an adult, indicating a negative effect of water if received by both males and females. In addition, when the larval diet included glycerol, increased number of eggs laid decreased female longevity, whilst an absence of glycerol in the larval diet resulted in low female longevity that was unlinked with fecundity. Glycerol is used in many artificial insect diets and the fact that it shows a strong effect on key life-history traits (reproductive output and longevity in this species), merits careful re-examination of its effects on these important traits in other laboratory models. We also discuss the possibility that larval diet can affect female reproductive decisions.  相似文献

Measurements of whole-body dry matter, total nitrogen, water-soluble protein, amino acids, and uric acid were determined at successive stages during metamorphosis in Neodiprion sertifer. The major change was in the uric acid fraction: in females, it increased up to the non-pharate pupa and then decreased during the subsequent stages of adult development and egg production; in males, it continued to increase during adult development. The decline of uric acid could not be explained by the accumulation of allantoin, allantoic acid, urea, or uric acid riboside. Examination of amino acid levels in the gut revealed an accumulation in the pupa followed by a depletion at the onset of adult development. This was followed by an excretory phase marked by the progressive accumulation of large quantities of uric acid and small quantities of urea, ammonia, and amino acids during the formation of the meconium. Amino acid analysis of the meconium revealed the presence of large proportions of proline, hydroxyproline, and histidine in comparison with the other amino acids.  相似文献

Ageing is characterized by a decline in organ maintenance and repair. Adult stem cells contribute to tissue repair and organ maintenance. Thus it is conceivable that ageing is partly due to a decline of stem cell function. At molecular level, ageing is associated with an accumulation of damage affecting DNA, proteins, membranes, and organelles, as well as the formation of insoluble protein aggregates. Telomere shortening represents a cell intrinsic mechanism, which contributes to the accumulation of DNA damage during cellular ageing. Telomere dysfunction in response to critical telomere shortening induces DNA damage checkpoints that lead to cell cycle arrest and/or cell death. Checkpoint responses induced by telomere dysfunction have mostly been studied in somatic cells but there are emerging data on cell intrinsic checkpoints that impair the maintenance and function of adult stem cell in response to telomere dysfunction. Moreover, telomere dysfunction induces alterations in the stem cell environment that limit the function of adult stem cells. In this review we summarize our current knowledge on the role of telomere dysfunction in adult stem cell ageing.  相似文献

Larval to adult microvascular anatomy of the esophagus was studied in the South African Clawed Toad, Xenopus laevis (Daudin) by scanning electron microscopy of vascular corrosion casts and correlative light microscopy of paraplast embedded stained tissue sections. Analyses of vascular corrosion casts of tadpole esophaguses at premetamorphosis revealed a wide‐meshed, but mature looking capillary bed which during following prometamorphosis increased in density and gained the adult‐like pattern during late metamorphic climax by sprouting and nonsprouting angiogenesis. In adult Xenopus, the esophageal mucosa possessed a dense subepithelial capillary bed fed by one or two esophageal arteries that originated from right and/or left thoracic aorta just distal to the origin of the subclavian arteries. In the adult undistended esophagus, esophageal arteries revealed an undulating course, a pattern that guarantees a continuous blood supply when the esophagus is extremely wide expanded as it is the case when adult Xenopus swallows large prey.  相似文献

为了研究水稻白叶枯病成株抗性是否与逆转子激活有关,运用SSAP (sequence-specific amplification polymorphism) 对成株抗性品种苗期和成株期接种白叶枯病原菌、清水接种及健康植株的基因组进行了逆转座子扫描。在筛选的约2000个逆转座子基因片段中,9个受苗期生长发育诱导激活,两个受成株期生长发育诱导激活,苗期和成株期各有3个受病原菌诱导激活。苗期生长发育诱导激活产生的逆转座子数目高于成株期,而病原菌诱导产生的逆转座子数目与成株期相当,表明水稻白叶枯病成株抗性可能与生长发育诱导的逆转座子激活相关。  相似文献

The role of tactile stimuli and pheromonal stimuli in the induction of oestrous cycle irregularities in mice was evaluated. In contrast to an adult regularly cycling female (the test female) housed in contact with 6 adult females, a test female housed in contact with 6 impuberal females failed to show disruption of oestrous cycle. Likewise, a test female housed in contact with bedding soiled by 6 group-housed adult females failed to exhibit disruption of oestrous cycle. By contrast, a test female housed in contact with 6 impuberal females on bedding soiled by 6 adult group-housed females exhibited a significant increase in the incidence of prolonged cycles. However, a test female housed with 6 impuberal females without direct contact with the bedding soiled by adult group-housed females exhibited a significant decrease in the incidence of prolonged cycles. The results suggest that tactile stimuli and pheromonal stimuli (probably present in the excrete of adult females) act synergistically in inducing cycle disruption. The findings further indicate that the female-originating pheromone involved in oestrous cycle disruption is non-volatile (not air-borne) and that it acts through contact.  相似文献

Insect neurogenesis has been subjected to extensive study and as a result is regarded as being well understood. It is, therefore, all the more surprising when a fundamentally novel aspects of the process is uncovered. Until recently it was thought that the production of central neurons ceased before the emergence of the adult. Recently, however, Cayre et al. have shown that neurogenesis also occurs in the adult brain. Their studies also show that the rate at which adult neuroblasts divide is controlled by hormones, suggesting that hormones may play a more important role in regulating neurogenesis than previously suspected.  相似文献

CNSgp 130 is a CNS-specific membrane glycoprotein present in large amounts in the adult mammalian CNS. Using immunohistological techniques, we demonstrated that CNSgp130 is not detectable in the rat cerebellum at birth, and does not appear in the cerebellum until the tenth day of postnatal life. It is expressed first in the white matter of the cerebellar folia, and subsequently (by day 14) it is expressed also in the molecular layer. Expression in the granular layer is not seen until the 18th day of postnatal life, by which time the adult pattern of expression is established. CNSgp130 is also not detectable in the cerebrum at birth. However, it is expressed weakly but diffusely in the cerebrum by the fourth day of life. By the 10th day, there is strong expression in the cerebrum, in marked contrast to its virtual absence from the cerebellum at this stage. By quantitative absorption analysis, CNSgp 130 was undetectable on the day of birth, and increased steadily to 80% of adult values by the 22nd day of postnatal life. Binding studies with pure CNSgp130 demonstrated a Pronase-sensitive ligand in adult chicken brain. This ligand was absent from neonatal rat brain and non-CNS tissues.  相似文献

The myosin light chains of cultured muscle cells and embryonic muscle tissue have been examined by two-dimensional gel electrophoresis. Myosin purified from primary cultures of rat muscle cells or the myogenic cell line L6 contain not only the light chains corresponding to those of fast twitch muscle but also another protein, differing slightly in molecular weight and isoelectric point from the adult LC1 protein. By a number of criteria this additional protein is shown to be a myosin light chain: (1) it is found in highly purified myosin preparations; (2) in L6 myosin it replaces the other LC1-type light chains in stoichiometric amounts; (3) it is part of the subfragment-1 complex of myosin produced by chymotrypsin. as expected for an LC1-type light chain. Total extracts of fused cultured muscle cells, when analyzed by two-dimensional electrophoresis, contain substantial amounts of this additional LC1-type protein, strongly suggesting that it is not a proteolytic fragment produced during myosin isolation. Unfused cultures do not synthesize detectable amounts of the adult light chains or the additional LC1-type light chain. This additional LC1 protein can be detected in embryonic or newborn muscle tissue but it is not present in adult myosin or myofibrils. These results indicate that a novel form of myosin light chain, referred to as an embryonic LC1 or LC1emb, is characteristic of the early stages of muscle development.  相似文献

Non-maternal infant care in many of the small-bodied New World primate species has been hypothesized by some researchers to be related to the high infant/adult weight ratio found in these species. The spectral tarsier, Tarsius spectrum, an Old World primate, has one of the highest infant/adult weight ratios of any primate, with infants weighing between 20-33% of adult weight at birth. On the basis of the hypothesized relationship between allocare and the infant/adult weight ratio, it is predicted that the spectral tarsier will also exhibit extensive allocaretaking behaviour. The results of this study indicate that although spectral tarsiers show care by male and female subadults as well as adult males, it is extremely limited compared to the extensive allocaretaking behavior observed in New World primate species such as Aotus and Callicebus. Spectral tarsier subadult females provide substantially more allocare to infants than do subadult males or adult males. Female subadults were observed sharing food, transporting, grooming, playing, alarm calling, baby-sitting and maintaining physical contact with infants more than other age/sex classes. Although the amount of allocare exhibited by adult males and subadult males was much less than that exhibited by female subadults, the data suggest that adult and subadult male spectral tarsiers do play a small part in the care and socialization of the infant. Adult males and subadult males were both observed occasionally engaging in allocaretaking behaviors such as grooming and playing, as well as frequently patrolling and defending the territory's boundaries. The results from this study suggest that although a high infant/adult weight ratio may be a prerequisite for selection to favor extensive allocare, it is not a causal factor. Additional research is needed in order to understand better the selective pressures involved and the costs and benefits of providing allocare to subadults and adult male spectral tarsiers.  相似文献
