Background  

For typing of Staphylococcus aureus, DNA sequencing of the repeat region of the protein A (spa) gene is a well established discriminatory method for outbreak investigations. Recently, it was hypothesized that this region also reflects long-term epidemiology. However, no automated and objective algorithm existed to cluster different repeat regions. In this study, the Based Upon Repeat Pattern (BURP) implementation that is a heuristic variant of the newly described EDSI algorithm was investigated to infer the clonal relatedness of different spa types.  相似文献

The BURP‐domain protein family comprises a diverse group of plant‐specific proteins that share a conserved BURP domain at the C terminus. However, there have been only limited studies on the functions and subcellular localization of these proteins. Members of the RD22‐like subfamily are postulated to associate with stress responses due to the stress‐inducible nature of some RD22‐like genes. In this report, we used different transgenic systems (cells and in planta) to show that the expression of a stress‐inducible RD22‐like protein from soybean (GmRD22) can alleviate salinity and osmotic stress. We also performed detailed microscopic studies using both fusion proteins and immuno‐electron microscopic techniques to demonstrate the apoplast localization of GmRD22, for which the BURP domain is a critical determinant of the subcellular localization. The apoplastic GmRD22 interacts with a cell wall peroxidase and the ectopic expression of GmRD22 in both transgenic Arabidopsis thaliana and transgenic rice resulted in increased lignin production when subjected to salinity stress. It is possible that GmRD22 regulates cell wall peroxidases and hence strengthens cell wall integrity under such stress conditions.  相似文献

Background and aims

Localized supply of P plus ammonium improves root-proliferation and nutrient-uptake by maize (Zea mays L.) at seedling stage, but it is largely unknown how localized supply of nutrients at both early and late stages influences maize-growth, nutrient-uptake and grain-yield.

Methods

A 2-year field experimentation with maize was conducted with localized application of P plus ammonium as diammonium phosphate (LDAP) or ammonium sulfate plus P (LASP) at sowing or jointing stage, with broadcast urea and P (BURP) or no nitrogen (F0) as controls.

Results

Localized supply of P plus ammonium significantly increased root-proliferation, shoot dry-weight and nutrient-uptake at seedling stage. The positive effect disappeared at 53 days after sowing. However, plant-growth and nutrient-uptake increased again after the second localized application of P plus ammonium at jointing. The density and average length of the first-order lateral roots in local patches increased by 50 % in LDAP and LASP compared with F0 and BURP. Maize-yield increased by 8–10 % compared with BURP. Agronomic N efficiency and N-use efficiency increased by 41–48 % and 25–57 % compared with the BURP.

Conclusions

It is suggested that enhanced root-proliferation in the nutrient-rich patches with localized supply of ammonium and P at sowing and jointing stages is essential for improving nutrient-uptake and ultimately grain-yield.  相似文献

The exported repetitive protein (erp) gene encodes a secreted 36-kDa protein with a central domain containing several proline-glycine-leucine-threonine-serine (PGLTS) repeats. The KdpD/KdpE two-component system of Escherichia coli regulates expression of the kdpFABC operon encoding the high affinity K+ transport system KdpFABC. The input domain of KdpD comprises a domain that belongs to the family of universal stress proteins (Usp). It has been previously demonstrated that UspC binds to this domain, resulting in KdpD/KdpE scaffolding under salt stress. However the mechanistic significance of this domain for signaling remains unclear. Background  

In Drosophila, the transport regulator Klar displays tissue-specific localization: In photoreceptors, it is abundant on the nuclear envelope; in early embryos, it is absent from nuclei, but instead present on lipid droplets. Differential targeting of Klar appears to be due to isoform variation. Droplet targeting, in particular, has been suggested to occur via a variant C-terminal region, the LD domain. Although the LD domain is necessary and sufficient for droplet targeting in cultured cells, lack of specific reagents had made it previously impossible to analyze its role in vivo.  相似文献
