The use of the relative ureide content of xylem sap [(ureide-N/total N) × 100] as an indicator of N2 fixation in soybeans (Merr.) was examined under greenhouse conditions. Acetylene treatments to inhibit N2 fixation were imposed upon the root systems of plants totally dependent upon N2 fixation as their source of N and of plants dependent upon both N2 fixation and uptake of exogenous nitrate. Significant decreases in the total N concentration of xylem sap from plants of the former type were observed, but no significant decrease was observed in the total N concentration of sap from the latter type of plants. In both types of plants, acetylene treatment caused significant decreases in the relative ureide content of xylem sap. The results provided further support for a link between the presence of ureides in the xylem and the occurrence of N2 fixation in soybeans. The relative ureide content of xylem sap from plants totally dependent upon N2 fixation was shown to be insensitive to changes in the exudation rate and total N concentration of xylem sap brought about by diurnal changes in environmental factors. There was little evidence of soybean cultivars or nodulating strains affecting the relative ureide content of xylem sap. `Ransom' soybeans nodulated with Rhizobium japonicum strain USDA 110 were grown under conditions to obtain plants exhibiting a wide range of dependency upon N2 fixation. The relative ureide content of xylem sap was shown to indicate reliably the N2 fixation of these plants during vegetative growth using a 15N method to measure N2 fixation activity. The use of the relative ureide content of xylem sap for quantification of N2 fixation in soybeans should be evaluated further.  相似文献

The yield of N in maize (Zea mays L.) and ricebean (Vigna umbellata [Thumb.] Ohwi and Ohashi) were compared on a Tropoqualf soil in North Thailand in 1984 and 1985. Both species were grown in field plots in monoculture or as intercrops at a constant planting density equivalent to 8 maize or 16 ricebean plants per m2. The contribution of symbiotic N2 fixation to ricebean growth was estimated from measurements of the natural abundance of15N (δ15N) in shoot nitrogen and from analysis of ureides in xylem sap vacuumextracted from detached stems. The natural abundance of15N in the intercropped ricebean was found to be considerably less than that in monoculture in both growing seasons. Using maize and a weed (Ageratum conyzoides L.) as non-fixing15N reference plants the proportions (P 15N) of ricebean shoot N derived from N2 fixation ranged from 0.27 to 0.36 in monoculture ricebean up to 0.86 when grown in a 75% maize: 25% ricebean intercrop. When glasshouse-derived calibration curves were used to calculate plant proportional N2 fixation (Pur) from the relative ureide contents of field collected xylem exudates, the contribution of N2 fixation to ricebean N yields throughout the 1985 growing season were greater in intercrop than in monocrop even at the lowest maize:legume ratio (25∶75). Seasonal patterns of sap ureide abundance indicated that N2 fixation was greatest at the time of ricebean podset. The averagePur andP 15N in ricebean during the first 90 days of growth showed identical rankings of monocrop and intercrop treatments in terms of N2 fixation, although the two sets ofP values were different. Nonetheless, seasonal estimates of N2 fixation during the entire 147 days of legume growth determined from ureide analyses indicated that equivalent amounts of N could be fixed by ricebean in a 75∶25 intercrop and in monoculture despite the former being planted at one-quarter the density.  相似文献

Utilising the 15N dilution technique the relationship between the proportion of N derived from N2 fixation and relative abundance of ureides in xylem sap was evaluated for Phaseolus vulgaris L. cv. Mokcham during vegetative and reproductive development. In order to establish calibration curves for time integrated estimates of N2 fixation, plants were raised in sand culture during the dry season in northern Thailand and continuously supplied with a N-free nutrient solution or the same solution amended with 0, 3, 6 or 9 mol m–3 nitrate. Large changes in plant dependence on N2 fixation were concomitantly reflected by corresponding alterations in N solutes in xylem sap. Regression analyses of the data suggested high correlations between relative ureide content and N2 fixation, but different slopes and line intercepts indicated the requirement for the use of calibration curves established for different phases of the development of the plant. Largest age related differences were noted between vegetative and reproductive development. Judging from 95% confidence limits, utilisation of appropriate calibrations can reduce errors of the technique to close to ±5%.A second experiment, involving similarly cultivated plants exposed to different sources of mineral N, indicated an effect of ammonium on xylem sap composition. This implies that calibrations, in which N2 fixation is regulated only by applications of various concentrations of nitrate, may lead to errors in situations where a major proportion of the plant available soil nitrogen fraction is present in the form of ammonium.  相似文献

Waterlogging of soybean plants (Glycine max L.) led to impaired symbiotic N2 fixation and a marked decline in glutamine (Gln) concentration in xylem bleeding sap. Xylem Gln concentration increased during the growth cycle of the plant and was correlated with nodule formation. Treatments known to impair N2 fixation, such as exposing the root system to pure N2 gas or a mixture of Ar and O2 (80:20; v/v), led to specific declines in xylem sap Gln. The decrease in Gln observed during waterlogging was also seen on transfer of nodulated plants to aerated hydroponics, where the decline was highly correlated with ureide content in the xylem sap. Upon flooding the nodulated root system, the specific decline in xylem sap Gln could be detected within 10 min and reached a minimum within 60 min, indicating that waterlogging has an immediate effect on N2 fixation. It is concluded that xylem Gln arises directly from N2-fixation and is a useful indicator of N2 fixation activity of symbiotic soybean plants.  相似文献

Xylem sap composition was examined in nodulated and nonnodulated cowpea (Vigna unguiculata [L.] Walp.) plants receiving a range of levels of NO3 and in eight other ureide-forming legumes utilizing NO3 or N2 as sole source of nitrogen. A 15N dilution technique determined the proportions of plant nitrogen derived from N2 in the nodulated cowpeas fed NO3. Xylem sap composition of NO3-fed, nodulated cowpea varied predictably with the relative extents to which N2 and NO3 were being utilized. The ratios of asparagine to glutamine (N/N) and of NO3 to ureide (N/N) in xylem sap increased with increasing dependence on NO3 whereas per cent of xylem nitrogen as ureide and the ratio of ureide plus glutamine to asparagine plus NO3 (N/N) in xylem sap increased with increasing dependence on N2 fixation. The amounts of NO3 and ureides stored in leaflets, stems plus petioles, and roots of cowpea varied in a complex manner with level of NO3 and the presence or absence of N2 fixation. All species showed higher proportions of organic nitrogen as ureide and several-fold lower ratios of asparagine to glutamine in their xylem sap when relying on N2 than when utilizing NO3. In nodulated (minus nitrate) cowpea and mung bean (Vigna radiata [L.] Wilczek) the percentage of xylem nitrogen as ureide remained constant during growth but the ratio of asparagine to glutamine varied considerably. The biochemical significance of the above differences in xylem sap composition was discussed.  相似文献

Experiments were conducted to characterize the distribution of N compounds in the xylem sap of nodulated and nonnodulated soybean plants through development and to determine the effects of exogenous N on the distribution of N compounds in the xylem. Xylem sap was collected from nodulated and nonnodulated greenhouse-grown soybean plants (Glycine max [L.] Merr. “Ransom”) from the vegetative phase to the pod-filling phase. The sum of the nitrogen in the amino acid, nitrate, ureide (allantoic acid and allantoin), and ammonium fractions of the sap from both types of plants agreed closely with total N as assayed by a Kjeldahl technique. Sap from nodulated plants supplied with N-free nutrient solution contained seasonal averages of 78 and 20% of the total N as ureide-N and amino acid-N, respectively. Sap from nonnodulated plants supplied with a 20 millimolar KNO3 nutrient solution contained seasonal averages of 6, 36, and 58% of total N as ureide-N, amino acid-N, and nitrate-N, respectively. Allantoic acid was the predominant ureide in the xylem sap and asparagine was the predominant amino acid. When well nodulated plants were supplied with 20 millimolar KNO3, beginning at 65 days, C2H2 reduction (N2 fixation) decreased relative to nontreated plants and there was a concomitant decrease in the ureide content of the sap. A positive correlation (r = 0.89) was found between the ureide levels in xylem sap and nodule dry weights when either exogenous nitrate-N or urea-N was supplied at 10 and 20 millimolar concentrations to inoculated plants. The results demonstrate that ureides play a dominant role in N transport in nodulated soybeans and that the synthesis of ureides is largely dependent upon nodulation and N2 fixation.  相似文献

The relationship between ureide N and N2 fixation was evaluated in greenhouse-grown soybean (Glycine max L. Merr.) and lima bean (Phaseolus lunatus L.) and in field studies with soybean. In the greenhouse, plant N accumulation from N2 fixation in soybean and lima bean correlated with ureide N. In soybean, N2 fixation, ureide N, acetylene reduction, and nodule mass were correlated when N2 fixation was inhibited by applying KNO3 solutions to the plants. The ureide-N concentrations of different plant tissues and of total plant ureide N varied according to the effectiveness of the strain of Bradyrhizobium japonicum used to inoculate plants. The ureide-N concentrations in the different plant tissues correlated with N2 fixation. Ureide N determinations in field studies with soybean correlated with N2 fixation, aboveground N accumulation, nodule weight, and acetylene reduction. N2 fixation was estimated by 15N isotope dilution with nine and ten soybean genotypes in 1979 and 1980, respectively, at the V9, R2, and R5 growth stages. In 1981, we investigated the relationship between ureide N, aboveground N accumulation, acetylene reduction, and nodule mass using four soybean genotypes harvested at the V4, V6, R2, R4, R5, and R6 growth stages. Ureide N concentrations of young stem tissues or plants or aboveground ureide N content of the four soybean genotypes varied throughout growth correlating with acetylene reduction, nodule mass, and aboveground N accumulation. The ureide-N concentrations of young stem tissues or plants or aboveground ureide-N content in three soybean genotypes varied across inoculation treatments of 14 and 13 strains of Bradyrhizobium japonicum in 1981 and 1982, respectively, and correlated with nodule mass and acetylene reduction. In the greenhouse, results correlating nodule mass with N2 fixation and ureide N across strains were variable. Acetylene reduction in soybean across host-strain combinations did not correlate with N2 fixation and ureide N. N2 fixation, ureide N, acetylene reduction, and nodule mass correlated across inoculation treatments with strains of Bradyrhizobium spp. varying in effectiveness on lima beans. Our data indicate that ureide-N determinations may be used as an additional method to acetylene reduction in studies of the physiology of N2 fixation in soybean. Ureide-N measurements also may be useful to rank strains of B. japonicum for effectiveness of N2 fixation.  相似文献

Nitrogen (N) is often the most limiting nutrient in organic cropping systems. N2 fixing crops present an important option to improve N supply and to maintain soil fertility. In a field experiment, we investigated whether the lower N fertilization level and higher soil microbial activity in organic than conventional systems affected symbiotic N2 fixation by soybean (Glycine max, var. Maple Arrow) growing in 2004 in plots that were since 1978 under the following systems: bio-dynamic (DYN); bio-organic (ORG); conventional with organic and mineral fertilizers (CON); CON with exclusively mineral fertilizers (MIN); non-fertilized control (NON). We estimated the percentage of legume N derived from the atmosphere (%Ndfa) by the natural abundance (NA) method. For ORG and MIN we additionally applied the enriched 15N isotope dilution method (ID) based on residual mineral and organic 15N labeled fertilizers that were applied in 2003 in microplots installed in ORG and MIN plots. These different enrichment treatments resulted in equal %Ndfa values. The %Ndfa obtained by NA for ORG and MIN was confirmed by the ID method, with similar variation. However, as plant growth was restricted by the microplot frames the NA technique provided more accurate estimates of the quantities of symbiotically fixed N2 (Nfix). At maturity of soybean the %Ndfa ranged from 24 to 54%. It decreased in the order ORG > CON > DYN > NON > MIN, with significantly lowest value for MIN. Corresponding Nfix in above ground plant material ranged from 15 to 26 g N m-2, with a decreasing trend in the order DYN = ORG > CON > MIN > NON. For all treatments, the N withdrawal by harvested grains was greater than Nfix. This shows that at the low to medium %Ndfa, soybeans did not improve the N supply to any system but removed significant amounts of soil N. High-soil N mineralization and/or low-soil P availability may have limited symbiotic N2 fixation.  相似文献

Sustainable management for existing Amazonian forests requires an extensive knowledge about the limits of ecosystem nutrient cycles. Therefore, symbiotic nitrogen (N2) fixation of legumes was investigated in a periodically flooded forest of the central Amazon floodplain (Várzea) over two hydrological cycles (20 months) using the 15N natural abundance method. No seasonal variation in 15N abundance (δ 15N values) in trees which would suggest differences in N2 fixation rates between the terrestrial and the aquatic phase was found. Estimations of the percentage of N derived from atmosphere (%Ndfa) for the nodulated legumes with Neptunia oleracea on the one side and Teramnus volubilis on the other resulted in mean %Ndfa values between 9 and 66%, respectively. More than half of the nodulated legume species had %Ndfa values above 45%. These relatively high N gains are important for the nodulated legumes during the whole hydrological cycle. With a %Ndfa of 4–5% for the entire Várzea forest, N2 fixation is important for the ecosystem and therefore, has to be taken into consideration for new sustainable land-use strategies in this area.  相似文献

In field-grown soybeans (Glycine max L. Merr. cv Harosoy), the percentage of N in the xylem as ureides increased with increasing N2 fixation. During a 9-week collection period, the ureide content varied from 9.0 to 69.2% of the xylary N. Between 9 and 11 weeks (early pod fill), there was a good correlation (r = 0.93) between C2H2 reduction and the per cent N in xylem as ureides. The per cent N as ureides, however, does not always indicate the reliance of the plant on symbiotic N2 fixation. This ureide content also depended on the level of NO3 available to the roots. Non-nodulated soybeans given from 0 to 200 kilogram N per hectare produced xylem sap which averaged from 31.8% to 9.0% N, respectively, in the xylem as ureides over the 9-week period.

Feeding of 15N2, 15NH4, or 15NO3 to greenhouse-grown soybeans indicated substantial differences in the initial distribution of N by the xylem stream, but the ultimate distribution of N between plant parts and grain did not vary with available N or percentage of xylary N as ureides. Amino acids, not ureides, were the major source of N in the phloem. The soybeans maintained a similar composition in phloem irrespective of the xylem sap constituents, with N derived from N2, NH4, or NO3 being equally accessible to the phloem stream.

Background: Nitrogen fixation has been quantified for a range of crop legumes and actinorhizal plants under different agricultural/agroforestry conditions, but much less is known of legume and actinorhizal plant N2 fixation in natural ecosystems.

Aims: To assess the proportion of total plant N derived from the atmosphere via the process of N2 fixation (%Ndfa) by actinorhizal and legume plants in natural ecosystems and their N input into these ecosystems as indicated by their 15N natural abundance.

Methods: A comprehensive collation of published values of %Ndfa for legumes and actinorhizal plants in natural ecosystems and their N input into these ecosystems as estimated by their 15N natural abundance was carried out by searching the ISI Web of Science database using relevant key words.

Results: The %Ndfa was consistently large for actinorhizal plants but very variable for legumes in natural ecosystems, and the average value for %Ndfa was substantially greater for actinorhizal plants. High soil N, in particular, but also low soil P and water content were correlated with low legume N2 fixation. N input into ecosystems from N2 fixation was very variable for actinorhizal and legume plants and greatly dependent on their biomass within the system.

Conclusions: Measurement of 15N natural abundance has given greater understanding of where legume and actinorhizal plant N2 fixation is important in natural ecosystems. Across studies, the average value for %Ndfa was substantially greater for actinorhizal plants than for legumes, and the relative abilities of the two groups of plants to utilise mineral N requires further study.  相似文献

Nodulation, nitrogen (N2) fixation and xylem sap composition were examined in sand cultured plants of Bambara groundnut (Vigna subterranea L.) and Kersting's bean (Macrotyloma geocarpum L.) inoculated with Bradyrhizobium strain CB756 and supplied via the roots for a 4 week period from the third week onwards with different levels of (15N)-nitrate (0–15 mM). The separate contributions of nitrate and N2 to plant nitrogen were measured by isotope dilution. Increasing levels of nitrate inhibited nodule growth (measured as dry matter or nodule N) of both species parallel with decreased dependence on symbiotically-fixed N. Specific nodule activity (N2 fixed g nodule dry−1 d−1 of nodules) was reduced progressively with time in V. subterranea at higher (5 or 15 mM) levels of NO3, but this was not so for M. geocarpum. Root xylem bleeding sap of both species showed ureides (allantoin and allantoic acid) as predominant (>90%) solutes of nitrogen when plants were relying solely on atmospheric N. Levels of ureide and glutamine decreased and those of asparagine and nitrate in xylem increased with increasing level of applied nitrate. Relative levels of xylem ureide-N were positively correlated (R2=0.842 for M. geocarpum and 0.556 for V. subterranea), and the ratio of asparagine to glutamine in xylem exudate negatively correlated (R2=0.955 for M. geocarpum and 0.736 for V. subterranea) with plant reliance on nitrogen fixation. The data indicate that xylem sap analyses might be useful for indirect field assays of nitrogen fixation by the species and that Kersting's bean might offer some potential as a symbiosis in which N2 fixation is relatively tolerant of soil N.  相似文献

During vegetative regrowth of Medicago sativa L., soil N, symbiotically fixed N2 and N reserves meet the nitrogen requirements for shoot regrowth. Experiments with nodulated or non-nodulated plants were carried out to investigate the changes in N flows originating from the different N sources and in xylem transport of amino acids during regrowth. Exogenous N uptake, N2 fixation and endogenous N remobilization were estimated by 15N labelling and amino acids in xylem sap were analysed. Removal of shoots resulted in great declines of exogenous N flows derived either from N2 or from NH4NO3 during the first week of regrowth, thereafter recovery increased linearly. Mineral N uptake as well as N2 fixation occurred mainly between the 10th and 18th day after removal of shoots while exogenous N assimilation in intact plants remained at a steady level. Nitrogen remobilization rates in defoliated plants increased by at least three to five-fold, especially during the first 10 days following shoot removal. Compared to control plants, contents of amino acids in xylem sap, during the first 10 days of regrowth, were reduced by about 72% and 82% in NH4NO3 grown and in N2 fixing plants, respectively. Asparagine was the main amino acid transported in xylem sap of both treated plants. Its relative contents during this period significantly decreased from 75% to 59% and from 67% to 36% respectively in non-nodulated plants and in nodulated ones. This decline was accompanied by compensatory increase in the relative contents of aspartate and glutamine.  相似文献

Nodulated and denodulated roots of adzuki bean (Vigna angularis), soybean (Glycine max), and alfalfa (Medicago sativa) were exposed to 14CO2 to investigate the contribution of nodule CO2 fixation to assimilation and transport of fixed nitrogen. The distribution of radioactivity in xylem sap and partitioning of carbon fixed by nodules to the whole plant were measured. Radioactivity in the xylem sap of nodulated soybean and adzuki bean was located primarily (70 to 87%) in the acid fraction while the basic (amino acid) fraction contained 10 to 22%. In contrast, radioactivity in the xylem sap of nodulated alfalfa was primarily in amino acids with about 20% in organic acids. Total ureide concentration was 8.1, 4.7, and 0.0 micromoles per milliliter xylem sap for soybean, adzuki bean, and alfalfa, respectively. While the major nitrogen transport products in soybeans and adzuki beans are ureides, this class of metabolites contained less than 20% of the total radioactivity. When nodules of plants were removed, radioactivity in xylem sap decreased by 90% or more. Pulse-chase experiments indicated that CO2 fixed by nodules was rapidly transported to shoots and incorporated into acid stable constituents. The data are consistent with a role for nodule CO2 fixation providing carbon for the assimilation and transport of fixed nitrogen in amide-based legumes. In contrast, CO2 fixation by nodules of ureide transporting legumes appears to contribute little to assimilation and transport of fixed nitrogen.  相似文献

The apparently diminished capacity for N2 fixation by the shrub legume Calliandra calothyrsus (Calliandra) relative to other woody perennial legumes was investigated in a field experiment in northern Queensland, Australia. In this trial, (i) the proportion of plant nitrogen (N) derived from symbiotic N2 fixation (%Pfix) and the amounts of N2 fixed were compared in Calliandra, Gliricidia sepium (Gliricidia) and Codariocalyx gyroides (Codariocalyx), (ii) variations in N2 fixation due to season or tree age were determined, (iii) estimates of Pfix derived with the 15N natural abundance technique were compared with values obtained from 15N enrichment or xylem sap ureide procedures to determine whether the previous conclusions about Calliandra's ability to fix N had resulted from specific problems with the natural abundance methodology used in the earlier studies.Inoculated seedlings of each of the three shrub legume species were planted in dense stands (1.5 m rows, 0.5 m between trees) in two randomised blocks. The northern block was used solely for natural abundance measurements, while 15N-enriched KNO3 (10 atom % 15N excess) was applied four times over a 52 week period to plots in the southern block. The non-nodulating tree legume Senna spectabilis (formally Cassia spectabilis) was used as a non-N2-fixing reference for the 15N-based procedures, with Guinea grass (Panicum maximum) included as an additional non-fixing check. Growth by the trees above 75 cm was first cut and removed after 22 weeks and regrowth was subsequently pruned periodically for another 95 weeks. Sampling for dry matter production, N yield and estimates of Pfix were restricted to the central four of the 32 plants which constituted each replicate plot. Information generated during the 117 week study indicated that estimates of Pfix by 15N natural abundance were closely similar to values derived with 15N-enrichment or sap ureides. The data indicated that Calliandra had a reduced reliance upon N2 fixation relative to Gliricidia and Codariocalyx for the first 65 weeks after establishment. This appeared to be due to more prolifc root growth by Calliandra than either of the other N2-fixing species and an ability to extract a greater proportion of its N requirements from soil mineral N. However, after week 65 and for the remainder of the experiment, estimates of Pfix for Calliandra were similar to the other shrub legumes. Over 117 weeks, prunings from Calliandra and Gliricidia had removed 52–58 t dry matter ha-1, and between 1471 and 1678 kg N ha-1, of which 1026–1063 kg N ha-1 was estimated to have been derived from N2 fixation. At the time of final harvest, 65–73% of the fixed N was present in shoot regrowth of the N2 fixing shrubs, 9–18% in the roots, 15% in the trunk, and 2–6% in fallen leaves.  相似文献

The 15N isotope dilution and A-value methods were used to measure biological nitrogen (N2) fixation in field grown fababean (Vicia faba L.), over a 2-year period. Four N rates, 20, 100, 200 and 400 kg N ha–1 were examined. The two isotope methods gave similar values of % N derived from the atmosphere (%Ndfa). With 20 kg N ha–1, %Ndfa in fababean was about 85% in both years. Increasing the N rate to 100 kg N ha–1 decreased N2 fixation slightly to 75%. Further reductions in N2 fixed to 60 and 43% occurred where 200 and 400 kg N ha–1 were applied, respectively. Thus even higher rates of N than normally applied in farming practice could not completely suppress N2 fixation in fababean.We also devised one equation for both the isotope dilution and A-value approaches, thereby (i) avoiding the need for different calculations for the 15N isotope methods, and (ii) showing once again that the isotope dilution and A-value methods are mathematically and conceptually identical.  相似文献

The effects of dairy cow urine and defoliation severity on biological nitrogen fixation and pasture production of a mixed ryegrass-white clover sward were investigated over 12 months using mowing for defoliation. A single application of urine (equivalent to 746 kg N ha–1), was applied in late spring to plots immediately after light and moderately-severe defoliation (35 mm and 85 mm cutting heights, respectively) treatments were imposed. Estimates of percentage clover N derived from N2 fixation (%Ndfa) were compared by labelling the soil with 15N either by applying a low rate of 15N-labelled ammonium sulphate, immobilising 15N in soil organic matter, adding 15N to applied urine, or by utilising the small differences in natural abundance of 15N in soil. Urine application increased annual grass production by 85%, but had little effect on annual clover production. However, urine caused a marked decline in %Ndfa (using an average of all 15N methods) from 84% to a low of 22% by 108 days, with recovery to control levels taking almost a year. As a result, total N fixed (in above ground clover herbage) was reduced from 232 to 145 kg N ha–1 yr–1. Moderately–severe defoliation had no immediate effect on N2 fixation, but after 108 days the %Ndfa was consistently higher than light defoliation during summer and autumn, and increased by up to 18%, coinciding with an increase in growth of weeds and summer-grass species. Annual N2 fixation was 218 kg N ha–1 yr–1 under moderately-severe defoliation compared to 160 kg N ha–1 yr–1 under light defoliation. Estimates of %Ndfa were generally similar when 15N-labelled or immobilised 15N were used to label soil regardless of urine and defoliation severity. The natural abundance technique gave highly variable estimates of %Ndfa (–56 to 24%) during the first 23 days after urine application but, thereafter, estimates of %Ndfa were similar to those using 15N-labelling methods. In contrast, in urine treated plots the use of 15N-labelled urine gave estimates of %Ndfa that were 20–30% below values calculated using conventional 15N-labelling during the first 161 days. These differences were probably due to differences in the rooting depth between ryegrass and white clover in conjunction with treatment differences in 15N distribution with depth. This study shows that urine has a prolonged effect on reducing N2 fixation in pasture. In addition, defoliation severity is a potential pasture management tool for strategically enhancing N2 fixation.  相似文献

The contribution of N2 fixation to overall soybean N uptake has most commonly been quantified by N isotope‐based methods, which rely on isotopic differences in plant N between legumes and non‐fixing reference plants. The choice of non‐fixing reference plants is critical for the accuracy of isotope‐based methods, and mismatched reference plants remain a potential source of error. Accurate estimates of soybean N2 fixation also require information on N isotopic fractionation within soybean. On the basis of a previous observation of a close correlation between an expression of N fractionation within soybean and the proportion of plant N derived from atmosphere (%Ndfa) determined by 15N natural abundance, this field study aimed at assessing the relationship between various expressions describing intraplant 15N or N partitioning and %Ndfa during soybean development. Starting from a late vegetative stage until beginning senescence, the N content and N isotopic composition of shoots, roots and nodules of nodulated and non‐nodulated soybeans was determined at eight different developmental stages. Regression analysis showed that %Ndfa most closely correlated with the difference in the N isotopic composition of shoot N minus that of root including nodule N, and that this relationship was similar to that obtained in a previous multi‐site field study. We therefore consider this expression to hold promise as a means of quantifying %Ndfa independent of a reference plant, which would avoid some of the external sources of error introduced by the use of reference plants in determining %Ndfa.  相似文献

Improvement of dinitrogen fixation in beans (Phaseolus vulgaris L.) will depend on the selection of superior plant genotypes and the presence of efficient rhizobial strains. This study was conducted to evaluate diverse bean lines for N2 fixation potential using the15N-depleted dilution technique under field conditions in Wisconsin, USA. Plants of 21 bean lines and three non-nodulating isolines of soybean received appliin Wisconsin, USA. Plants of 21 bean lines and three non-nodulating isolines of soybean received applications of15N-depleted ammonium sulphate. Shoots harvested at the V6, R3 and R7 stages and dry seeds were analyzed for total N using the Kjeldahl procedure, and the ratio of15N to14N was determind on a MAT 250 mass spectrometer. Nodule occupancy of the applied strain ofR. leguminosarum biovarphaseoli, CIAT 899, was determined in five of the bean lines. Total shoot N content showed a pattern of accumulation similar to shoot dry weight and fixed N2 in the shoot. Based on shoot total N, N2 fixed in the shoot and shoot dry weight Riz 30 and Preto Cariri were identified as being as good fixers as Puebla 152 and Cargamanto appear to begin N2 fixation early. Furthermore, some bean lines that fixed considerable N2 did not translocate a large amount of N to the grains. Preto Cariri accumulated 21.2 kg N ha−1 in the seeds compared to Puebla 152 which accumulated 43.8 kg N ha−1 of the fixed N2 into the grains. At the early sampling, Puebla 152 and 22–27 had a considerable higher percentage of their crown nodules formed by the inoculant strain CIAT 899, than did Rio Tibagi which has been considered a poor N2 fixer.  相似文献

A pot experiment was conducted in a greenhouse using the 15N isotope dilution method and two reference plants, Parkia biglobosa and Tamarindus indica to estimate nitrogen fixed in four Acacia species: A raddiana, A. senegal, A. seyal and Faidherbia albida (synonym Acacia albida). For the reference plants, the 15N enrichments in leaves, stems and roots were similar. With the fixing plants, leaves and stems had similar 15N enrichments; they were higher than the 15N enrichment of roots. The amounts of nitrogen fixed at 5 months after planting were similar using either reference plant. Estimates of the percentage of N derived from fixation (%Ndfa) for the above ground parts, in contrast to %Ndfa in roots, were similar to those for the whole plant. However, none of the individual plant parts estimated accurately total N fixed in the whole plant, and excluding the roots resulted in at least 30% underestimation of the amounts of N fixed. Between species, differences in N2 fixation were observed, both for %Ndfa and total N fixed. For %Ndfa, the best were A. seyal (average, 63%) and A. raddiana (average, 62%), being at least twice the %Ndfa in A. senegal and F. albida. Because of its very high N content, A. seyal was clearly the best in total N fixed, fixing 1.62 g N plant–1 compared to an average of 0.48 g N plant–1 for the other Acacia species. Our results show the wide variability existing between Acacia species in terms of both %Ndfa and total N fixed: A. seyal was classified as having a high N2 fixing potential (NFP) while the other Acacia species had a low NFP.  相似文献
