Synchronous, demonstrative, easily reproducible fertilization with the following embryonic development makes the process in the sea urchin extremely attractive for studying many biological enigmas. In particular, germ and embryonic cells of the sea urchin present a wide opportunity for investigating different associated phenomena launched by an increase in concentration of Ca2+ in cells ([Ca2+]i).Ca2+ ions participate in the activation of diverse processes of respiration and sperm motility (Shapiro et al., 1990; Brokaw, 1991), chemotaxis of spermatozoa to components of the egg jelly (Ward et al., 1985), acrosomal reaction (Trimmer et al., 1986; Shapiro et al., 1990), cortical reaction, formation of the fertilization membrane (Sasaki, 1984; Sardet and Chang, 1987), cellular division in the embryo (Poenie et al., 1985; Silver, 1986; Whitaker and Patel, 1990), their adhesion (McClay and Matranga, 1986), differentiation and formation of spicules (Mitsunaga et al., 1988) and metamorphosis (Carpenter et al., 1984).The present review combines information on the function of calcium-binding proteins and their targets, calmodulin regulation of NAD-kinase, exocytosis of cortical granules, Ca2+- and calmodulin-dependent protein phosphatase, Ca2+-dependent protein phosphorylation, regulation of ion-exchanger in the germ and embryonic cells as well as Ca2+- and calmodulin control of sperm motility in sea urchins.  相似文献

The chromatin proteins of different embryonic stages, ranging from 16 cell to gastrula, of the sea urchin Strongylocentrotus purpuratus were labeled, in vivo, with 14C and were labeled, in vitro, with 3H. The proteins thus labeled were separated by high resolution two-dimensional electrophoresis. The extent of possible cytoplasmic contamination has been examined with reconstruction experiments. Gastrula chromatin contains over 200 separable nonhistone proteins, and about 90% of them are also detected at the 60-cell stage; cleavage stages have over all protein gel patterns displaying numerous differences with the pattern shown by chromatin from later stages. Differences in the proportion of histone to nonhistone proteins that are synthesized are observable at the different embryonic stages, with histones predominating in midcleavage. About half of the nonhistone proteins of the developing embryo that can be labeled with 3H, in vitro, are not labeled with 14C, in vivo, and hence, must originate from a reservoir of nonhistone proteins assembled during oogenesis.  相似文献

Surface membrane glycoproteins have been postulated in many mammalian cells to be involved in external surface membrane functions such as cell adhesion, cell-cell recognition, and cell movement. In developing echinoderm embryos, cell adhesion, recognition, and movement of individual cell types have been attributed to differences in the external surface membranes of these cells. Results reported here suggest that the three cell types of 16-cell sea urchin embryos have a mechanism that could establish differences in the carbohydrate portion of glycoproteins located in the external surface membrane. The results demonstrate 1) that glycoproteins are synthesized during early sea urchin development and 2) that slightly different rates of glycoprotein synthesis exist for the three types of blastomeres from 16-cell sea urchin embryos.  相似文献

Extracellular matrix of sea urchin and other marine invertebrate embryos   总被引:3,自引:0,他引:3  
The extracellular matrix surrounding the sea urchin embryo (outer ECM) contains fibers and granules of various sizes which are organized in recognizable patterns as shown by ultrastructural studies, particularly stereoimaging techniques. The use of the ruthenium red method for retaining and staining the ECM, with modifications of the Luft (Anatomical Record 171:347-368, 1971) method for invertebrate embryos, allows for the clarification of certain structures, particularly fiber compaction in the interzonal region, and microvillus-associated bodies. The inner ECM in the sea urchin embryo includes the basal lamina and blastocoel matrix. Stereoimages show that the fibers which are loosely distributed in the blastocoel matrix become compacted around the periphery of the blastocoel to form the basal lamina. The ruthenium red method was also used on a number of marine invertebrate embryos and larvae, representing different phyla, to facilitate comparisons between their surface coats. The similarities observed in the specimens shown suggest that ECMs are widely found on marine invertebrate eggs, embryos, and larvae, and that they resemble vertebrate ECMs and may, therefore, have similar functions.  相似文献

The chemical composition of the chromatin, the fractional content of histones and nonhistone chromatin proteins (NHP), and the biosynthesis of these proteins in normal, vegetalized, and animalized embryos of the sea urchin Strongylocentrotus droebachiensis at the blastula, mesenchyme blastula, and gastrula stages have been studied. The amount of the NHP in the chromatin from normal and vegetalized embryos increases during early embryonic development while that in animalized embryos remains without change at the mesenchyme blastula stage and then decreases. During development the histone content in all three cases slightly decreases. Polyacrylamide gel electrophoresis reveals that both fractional composition of histones and their biosynthesis in normal, vegetalized, and animalized embryos display no differences. During development, however, some changes occur, so that the relative amount of histones F1 and F2a2 increases, F2b decreases, while F3 and F2a1 remains constant. Histone F1 at the blastula stage consists of two subfractions while at the gastrula stage it consists of three subfractions. The histone F2a1 consists of one and two, respectively. Histone F3 at all stages is made up of three subfractions; histone F2b is made up of two; and the histone F2a2 is electrophoretically homogeneous. Specific radioactivity of the arginine-rich histones F3 and F2a1 tends to increase during development, while that of moderately lysine-rich histones F2b and F2a2 does not change, and that of the lysine-rich histone F1 decreases. The NHP in normal, vegetalized, and animalized embryos at different developmental stages consist of 17 fractions that can be separated by isoelectrofocusing within the 4.5-8.8 pH range. Quantitative changes have been observed in the fractions focused at pH 4.5-6.1 during development and in normal and modified embryos at the gastrula stage.  相似文献

Degradation of yolk proteins in sea urchin eggs and embryos   总被引:6,自引:0,他引:6  
Yolk granules isolated from unfertilized and fertilized eggs of the sea urchins, Hemicentrotus pulcherrimus and Anthocidaris crassispina, were incubated in acidic media, and the protein components were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. By the incubation, a protein (molecular weight 180,000 in H. pulcherrimus and 178,000 in A. crassispina) most abundant in unfertilized eggs decreased, while proteins (molecular weight 61,000, 72,000, 94,000, 114,000 in H. pulcherrimus and 56,000, 70,000, 92,000, 112,000 in A. crassispina) dominant in developed embryos increased. Neither alkaline nor neutral condition resulted in such changes in the electrophoretic patterns of proteins as observed in acidic media. Experiments with various inhibitors of proteases suggested that thiol protease(s), such as cathepsin B, may be the most important enzyme(s) in the degradation of yolk proteins in embryogenesis of the sea urchin.  相似文献

In Paracentrotus lividus embryos, treatment with zinc ions induces the synthesis of the two major stress proteins with the same molecular weight as those induced by heat shock. The developmental stages responsive to zinc ion treatment are the same as those responsive to heat shock. However, zinc treatment induces a longer lasting synthesis of the stress proteins, and, unlike heat shock, does not induce thermotolerance and does not inhibit synthesis of the bulk proteins.  相似文献

In Arbacia punctulata and Strongylocentrotus purpuratus, two species of sea urchins, collagen synthesis begins during gastrulation and increases many-fold before reaching a plateau in the late pluteus stage. A collagen extraction method involving treatment with 0.1 M NaOH and hot 10% trichloroacetic acid provided the basis for a sensitive assay of collagen synthesis.  相似文献

Newly synthesized DNA-binding proteins were isolated from the nuclei and, separately from, the cytoplasm of sea urchin mofula stage embryos. The presence of 5-bromodeoxyuridine during embryogenesis did not appear to alter the synthesis of either class of DNA-binding proteins. This result tends to argue that cell differentiation in early embryos is not regulated by differential synthesis of DNA-binding proteins. Sea urchin mofulae synthesize a broad range, by molecular weight, or cytoplasmic DNA-binding proteins which dissociate from sea urchin DNA-cellulose at relatively high salt concentrations (0.6-2.0 M NaCl). The most prominant of these apparently high-binding-affinity proteins has an approximate molecular weight of 33,000.  相似文献

Summary The developmental changes in the pattern of chromatin nonhistone proteins have been investigated. The main feature of the observed changes was not the introduction of new or the disappearence of earlier existing species, but the stage-specific alterations in the rate of biosynthesis of the nonhistone protein constituents.  相似文献
