Summary 1. Catecholaminergic structures in the male Brachionus plicatilis were investigated, using aqueous dansylpropranolol as fluorescent label of neurotransmitter receptors. 2. All major organs of the male are innervated by catecholaminergic systems, that may also be involved in the regulation of copulatory behavior. 3. Cryostat-sectioned preparations of the female B. plicatilis were also investigated. They provided additional information to findings on whole animals reported in our previous paper (Keshmirian and Nogrady 1987a).  相似文献

1. Catecholaminergic neuronal structures were investigated in the rotifers Brachionus plicatilis, Asplanchna priodonta and Asplanchna herricki, using three different aqueous histofluorescent methods. 2. The adrenergic receptors were labelled using the dansyl analog of propranolol, a beta-adrenergic blocker. Catecholamine neurotransmitters were visualized by derivatizing with glyoxylic acid and formaldehyde respectively. 3. Although all three methods lead to similar results, dansyl-propranolol gave the most rapid and strong fluorescence. 4. The results reveal a complex and highly developed catecholaminergic neuronal system in all adult organs and sensory structures. While developed embryos in the egg show strong fluorescence, immature eggs do not.  相似文献

Brachionus quadridentatus and Brachionus plicatilis were examined in laboratory cultures to investigate their (1) food requirements and (2) ability to select particles on the basis of size. Growth experiments showed that the Chlorophyceans Nannochloris sp. and Chlorella vulgaris possess good nutritional value for these rotifers. However, B. quadridentatus declined in cultures with Microcystis firma, detritus of Enteromorpha sp. or Pseudomonas sp. (bacteria) as a sole source of food, thus indicating that these foods were inadequate. Utilization of latex microspheres by these two species indicate that they respond to particle size in different ways: B. quadridentatus had a higher selectivity index for 3 and 5 µm beads, while B. plicatilis had a higher selectivity index for 2 µm beads.  相似文献

gamma-Aminobutyric acid (GABA) and 5-hydroxytryptamine (5-HT) have been shown to increase the reproduction of the Brachionus plicatilis (NH3L strain). In the present study, the endogenous presence of GABA and 5-HT in the rotifers B. plicatilis (NH3L and Kamiura strains) and Brachionus rotundiformis (Langkawi strain) were confirmed by dot blot immunoassay and high-performance liquid chromatography (HPLC). HPLC showed that GABA and 5-HT concentrations in the three rotifer strains range from 71 to 188 pmol/mg and from 12 to 64 pmol/mg, respectively. A total of 33 amino acids were also detected in B. plicatilis and B. rotundiformis, with glutamic acid, serine, glycine, taurine, threonine, alanine, arginine, proline, valine and isoleucine in high concentrations relative to other amino acids.  相似文献

Fecundity patterns of S and L type rotifers Brachionus plicatilis, which were previously found to have distinct growth ability, were analyzed using data of age-specific fecundity. The data were obtained by individual cultures of S and L type strains at 17, 24 and 34 °C. The pattern was analyzed by using normal probability functions. When the age was transformed into logarithmic value, the S and L types had an identical pattern of fecundity at every temperature. This fact indicates that the difference of the growth response to the temperature between S and L type strains stemmed only from the differences in net reproduction rate, not from the pattern.  相似文献

Examination by scanning electron microscopy of sectioned rotifers provides views on their internal structures which complement the results of other techniques. Thus, additional information has been obtained for example, on the digestive tract, on nerve connections and on the morphology of the mastax. Our observations confirm that nearly all organs are connected in some way to the integument, suggesting that integument structures may be responsible for the holding together of the whole rotifer body.  相似文献

Rotifers (Brachionus plicatilis), maintained on baker's yeast, were fed for 24h upon two algal diets, Isochrysis galbana (diet A) and Isochrysis galbana + Nannochloropsis gaditana (diet B). (These algal diets were selected for their potential use as essential fatty acid (EFA) boosters, taking into account the requirements of fish larvae). The effect of these algal diets on total lipid content, lipid classes and fatty acid composition was studied. The total lipid content increased after feeding upon both diets but no significant differences were found between the two types. Neutral lipid and polar lipid contents increased and a positive correlation was observed between the neutral lipids content of rotifers and that of the food supplied. However, the content of polar lipids in rotifers did not depend upon that of the diet. The increase in neutral lipid content was found to be higher in rotifers fed upon diet B, compared to diet A which increased the phospholipid content. Non-enriched rotifers contained only small amounts of polyenoic fatty acids, i.e. 18:3n-6, 18:3n-3, 20:4n-6, 20:5n-3 and 22:6n-3, the contents of which increased significantly by feeding both diets. The EFA composition (20:4n-6, 20:5n-3 and 22:6n-3) of neutral lipids and phopholipids in rotifers reflected the EFA composition of each diet. Diet B-fed rotifers had the highest content in 20:4n-6 and 20:5n-3, whereas rotifers fed diet A and the highest 22:6n-3 content. The mixed diet I. galbana + N. gaditana enhanced substantially the composition of lipid classes i.e. neutral lipids and of n-3 PUFA of rotifers in comparison with Isochrysis or yeast diets.  相似文献

1. The enzyme glutathione S-transferase (GST), a critical element in xenobiotic metabolism, was isolated from the marine rotifer Brachionus plicatilis and its freshwater congener B. calyciflorus. 2. In B. plicatilis, GST comprised 4.2% of cytosolic protein and was present as three separate isozymes with mol. wts 30,000, 31,400 and 33,700. Specific activity of crude homogenates was 56 nmol min-1 mg-1 protein, while that of affinity chromatography purified GST was 1850. 3. In B. calyciflorus, GST was present as two isozymes with mol. wts of 26,300 and 28,500, representing 1.0% of cytosolic protein. Crude GST specific activity was 1750 nmol min-1 mg-1 protein and purified was 72,400. 4. Rotifer GSTs are unusual because they are monomers whereas all other animals thus far investigated posses dimeric GSTs.  相似文献

The cuticular oesophagus is a simple expansion of the dorsal pharyngeal wall of the mastax. The ciliary oesophagus is the cellular anterior wall of the stomach lumen, but seems to have the same embryological origin as the pharynx.In Brachionus calyciflorus, its cilia are surrounded by cuticular velums which have the same myelin-like structure and the same function as the buccal velum of Philodina roseola. In all cases, the oesophagus prevents the return of food particles from the stomach to the mastax lumen.  相似文献

This study was initiated to search for species-specific and strain-specific satellite DNA sequences for which oligonucleotide primers could be designed to differentiate between various commercially important strains of the marine monogonont rotifers Brachionus rotundiformis and Brachionus plicatilis. Two unrelated, highly reiterated satellite sequences were cloned and characterized. The eight sequenced monomers from B. rotundiformis and six from B. plicatilis had low intrarepeat variability and were similar in their overall lengths, A + T compositions, and high degrees of repeated motif substructure. However, hybridizations to 19 representative strains, sequence characterizations, and GenBank searches indicated that these two satellites are morphotype-specific and population-specific, respectively, and share little homology to each other or to other characterized sequences in the database. Primer pairs designed for the B. rotundiformis satellite confirmed hybridization specificities on polymerase chain reaction and could serve as a useful molecular diagnostic tool to identify strains belonging to the SS morphotype, which are gaining widespread usage as first feeds for marine fish in commercial production. A number of compounds showing general anesthetic action in the rotifer Brachionus calyciflorus were investigated in the presence of acetylcholine. Non-ionizing anesthetics, including tricaine, showed no interaction with acetylcholine. However, highly ionized compounds like the local anesthetics procaine and lidocaine, the muscarinic blocker and local anesthetic atropine, and the beta-adrenergic blocker propranolol showed a synergistic effect with acetylcholine. ACh increased the general anesthetic effect of these compounds in a statistically highly significant dose-dependent fashion. To account for the mechanism of this unusual and novel effect it is proposed that these compounds interact with the anesthetic binding site of the rotifer cholinoceptor ionophore in the open state. It is also proposed that non-ionizing compounds have a general membrane effect only. In addition to anesthesia, atropine and propranolol cause foot paralysis in B. calyciflorus. This other novel effect is also enhanced by acetylcholine as well as decamethonium, a neuromuscular blocker.  相似文献

Involvement of nine classical neurotransmitters in the abnormal sessile behavior of the rotiferBrachionus plicatilis was investigated. Only norepinephrine (NE), dopamine (DA) and octopamine (OA) induced sessility. NE and DA behaved as a full, OA as a partial agonist; NE was more potent than DA. Summary

The decline with age of mictic female susceptibility to fertilization and male capacity for fertilization is characterized for the rotifer Brachionus plicatilis. All mictic females were susceptible to fertilization until age 4 hr. Susceptibility then declined non-linearly according to the quadratic equation Y = 140.6—14.3X + 0.36X 2. By age 24 hr, sexual females no longer could be fertilized. Only 83% of newborn males were capable of fertilization. This level of fertility held until age 8 hr, then declined linearly. The age when 50% of individuals were no longer fertile was termed the length of fertilizability 50(LF50) and is 7.9 hr (16.7% of lifespan) and 18.8 hr (26.1% of lifespan) for females and males, respectively. Newborn males had an average of 30.1 ± 1.40 motile sperm. Males transferred a mean of 2.3 motile sperm into the pseudocoelom of females on each insemination. Sperm inseminated per copulation closely corresponds to the mean number of resting cysts produced by fertilized females. It is not likely that resting cyst production is limited by sperm availability.  相似文献

Patterns of genetic variation among Brachionus plicatilis clones collected at different times from Soda Lake, Nevada (U.S.A.) were determined using polyacrylamide gel electrophoresis of isozymes. Six of the eleven enzymes assayed were variable. Three of these — malic dehydrogenase (MDH), cholinesterase (CHO), and phosphoglucomutase (PMG) - were selected to screen the 138 clones established from the field collections. Each enzyme revealed variation both within and between field samples. Thirty-eight of the 100 potential composite electromorphs formed by combining isozyme variants were found in the 138 clones. A substantial amount of genetic variation is therefore present. Patterns of electromorph frequency change resembled those reported for cladocerans. No evidence was found for a seasonal succession of distinct electromorphs. We suggest that the B. plicatilis in Soda Lake constitute a single population.  相似文献

An attempt is made to identify sibling species within the Brachionus plicatilis complex (Rotifera). Allozyme and morphological data for nine strains from all over the world are provided. Although the analysis of morphological data classified individuals from nine strains into two groups, cluster analysis using genetic distance data obtained from allozyme data revealed at least three groups. A male choice design is described for the first time in rotifers and was used to test for male mating preferences among sympatric strains belonging to three distinct species. The results suggest that the B. plicatilis complex is actually composed of more than three sibling species.  相似文献

Brachionus calyciflorus contained no detectable tocopherol (vitamin E) when cultured for long periods on the yeast Rhodotorula glutinis. Using a spectrofluorometric assay and the Asplanchna body-wall-outgrowth response bioassay the limits of detection were 6.4 ng and 3.78 pg alpha-tocopherol per g dry weight Brachionus, respectively. These tocopherol-free Brachionus reproduced normally both parthenogenetically and sexually, producing active and potent males and fertilized resting eggs which developed into viable young rotifers. The apparent lack of a tocopherol requirement for male fertility in B. calyciflorus probably also applies to Asplanchna sieboldi and A. brightwell -- rotifers whose male-producing (mictic) females are induced by dietary tocopherol and whose males therefore are never tocopherol-deficient. The adaptive significance of the tocopherol requirement for the initiation of sexual reproduction in Asplanchna sieboldi--formerly hypothesized to be the result of a tocopherol requirement for male fertility--is now thought to be related to the role of tocopherol in controlling female polymorphism in this species.  相似文献

1. Exposure of the rotifer Brachionus plicatilis to elevated temperature resulted in the synthesis of a number of proteins, including a prominent one of 58,000 Da (SP58). 2. This protein is immunologically crossreactive with the 65,000 Da heat shock protein of the moth Heliothis virescens, which is a member of a highly conserved family of mitochondrial proteins. 3. Exposure of rotifers to sublethal doses of CuSO4 leads to a 4-5-fold increase in abundance of SP58, with maximum increase occurring at a dose that is approximately 5% of the LC50 for that compound. 4. A similar response was seen with tributyl tin (TBT). Kinetics of induction were sigmoidal, with induction occurring in the range of 20-30 micrograms/l. 5. No response was observed when rotifers were exposed to aluminum chloride, mercury chloride, pentachlorophenol, sodium arsenite, sodium azide, sodium dodecyl sulfate, or zinc chloride. 6. These results indicate that changes in stress protein abundance may prove useful as a biomarker of exposure to particular toxicants.  相似文献

Three species of rotifers were fixed with glutaraldehyde and postfixed in a OsO4-ferricyanide solution. This procedure results in staining of the sarcoplasmic reticulum (SR) and the surface coat of muscle cells as well as in staining of the surface coat of glands and nerve cells. Section-staining with leadcitrate increased the electron opacity of the precipitates, which are interpreted as polysaccharides. The partial decrease of staining of polysaccharides, which sometimes is observed, may be related to processes of function. The precipitates within the SR are considered as glycogen-beta-particles.  相似文献

Continental lake-dwelling zooplanktonic organisms have long been considered cosmopolitan species with little geographic variation in spite of the isolation of their habitats. Evidence of morphological cohesiveness and high dispersal capabilities support this interpretation. However, this view has been challenged recently as many such species have been shown either to comprise cryptic species complexes or to exhibit marked population genetic differentiation and strong phylogeographic structuring at a regional scale. Here we investigate the molecular phylogeny of the cosmopolitan passively dispersing rotifer Brachionus plicatilis (Rotifera: Monogononta) species complex using nucleotide sequence variation from both nuclear (ribosomal internal transcribed spacer 1, ITS1) and mitochondrial (cytochrome c oxidase subunit I, COI) genes. Analysis of rotifer resting eggs from 27 salt lakes in the Iberian Peninsula plus lakes from four continents revealed nine genetically divergent lineages. The high level of sequence divergence, absence of hybridization, and extensive sympatry observed support the specific status of these lineages. Sequence divergence estimates indicate that the B. plicatilis complex began diversifying many millions of years ago, yet has showed relatively high levels of morphological stasis. We discuss these results in relation to the ecology and genetics of aquatic invertebrates possessing dispersive resting propagules and address the apparent contradiction between zooplanktonic population structure and their morphological stasis.  相似文献
