The photosynthetic and respiratory responses to irradiance, salinity and temperature of the red alga, Gracilaria vermiculophylla, collected from Kumamoto, Shizuoka and Iwate in Japan were studied using an electronic Dissolved Oxygen sensor. The parameters derived from the photosynthesis versus irradiance relationship indicated the potential to acclimate to broad irradiance variations in all of the populations of G. vermiculophylla collected from these three sites. In addition, the light-saturated photosynthesis rate (P max) and the dark respiration rate of all populations increased with increasing temperature up to 20–30°C, while the P max decreased at 35°C. All populations also showed a broad variation of photosynthetic responses to salinity changes in the range from 10 to 30 psu. On the other hand, the population from Iwate showed high photosynthetic efficiency, especially in the temperature range of 5–10°C, and showed low values of saturation irradiance compared to the populations from Shizuoka and Kumamoto. These results suggest that there is greater potential to acclimate to low irradiance and low temperature in the population from Iwate compared to those from the Shizuoka and Kumamoto populations. However, the P max of the populations from Iwate and Shizuoka was reached at 20°C and 25°C, respectively, while the Kumamoto population reached P max at 30°C. This implies that the latter population has greater potential to tolerate higher temperatures than the former. Such characteristics in photosynthesis and respiration of G. vermiculophylla collected from the three locations probably indicate an acclimation to prevailing environmental conditions in their respective habitats.  相似文献

Selecting entomopathogenic fungal isolates for use as biocontrol agents requires an assessment of their growth and virulence characteristics as affected by environmental conditions. Here we demonstrate a wide temperature and moisture range for colony growth, effective conidial germination and virulence against Pissodes strobi Peck (white pine weevil) of several isolates of Lecanicillium Gams and Zare, an entomopathogenic fungus distributed worldwide and indigenous to forests on Vancouver Island, British Columbia, Canada. In order to examine the potential Lecanicillium as a biological control agent, the pathogenicity of isolates collected from different geographical locations on P. strobi cadavers was assessed, and colony growth at different temperatures was evaluated. Colony growth was evident between 5 and 30°C, with optimal growth occurring at 25°C. Various combinations of water activity (0.55, 0.76, 0.85 and 0.99 a w) and temperature (10, 15, 20, and 25°C) were also used to evaluate environmental impacts on conidial germination and cumulative mycosis of adult P. strobi. Certain Lecanicillium isolates displayed xerophilic (0.85 a w) or psychrophilic (10°C) growth optima. Ultimately, identifying the abiotic limits of this entomopathogenic fungus will be used to determine which isolates have potential for future in situ biocontrol trials.  相似文献

The effect of five constant temperatures (16, 22, 26, 31 and 36°C) on biological (survival and duration of developmental stages, fecundity and longevity of females, sex-ratio) and demographic parameters (R o , G, r m and λ) of the two main phytophagous mites that attack cassava in Africa,Mononychellus progresivus Doreste andOligonychus gossypii (Zacher), was studied in the laboratory. Experiments were performed simultaneously on the two mite species reared on the same cassava variety (1M20) under controlled conditions: 70±10% r.h. and 12L∶12D. The lower thermal threshold was 13°C forM. progresivus and 11°C forO. gossypii. Both species developed in the range from 22 to 36°C. The shortest development time was obtained at 31°C for both species; it was 7.2 days forM. progresivus and 8.2 days forO. gossypii. Maximum fecundity of both species was recorded at 26°C with 42.1 eggs forM. progresivus and 36.3 eggs forO. gossypii The highest intrinsic rate of increase (r m ) was obtained at 31°C for both species with 0.289 and 0.214 forM. progresivus andO. gossypii, respectively.  相似文献

Two intertidal snails, Littorina saxatilis (Olivi, 1972) (upper eulittoral fringe/maritime zone) and Littorina obtusata (Linnaeus, 1758) (lower eulittoral) were collected from a boulder shore on Nobska Point, Cape Cod, Massachusetts, in July and acclimated for 15–20 days at 4 ° or 21 °C. Oxygen consumption rate (Vo2) was determined for 11–15 subsamples of individuals at 4 °, 11 ° and 21 °C with silver/platinum oxygen electrodes. Multiple factor analysis of variance (MFANOVA) of lo10 transformed values of whole animal Vo2 with log10 dry tissue weight (DTW) as a covariant revealed that increased test temperature induced a significant increase in Vo2 in both species (P<0.00001). In contrast, MFANOVA revealed that temperature acclimation did not affect Vo2 in either L. saxatilis (P= 0.35) or L. obtusata (P= 0.095). Thus, neither species displayed a capacity for the typical metabolic temperature compensation marked by an increase in Vo2 at any one test temperature in individuals acclimated to a lower temperature that is characteristic of most ectothermic animals. Lack of capacity for metabolic temperature acclimation has also been reported in other littorinid snail species, and may be characteristic of the group as a whole. Lack of capacity for respiratory temperature acclimation in these two species and other littorinids may reflect the extensive semi-diurnal temperature variation that they are exposed to in their eulittoral and eulittoral fringe/maritime zone habitats. In these habitats, any metabolic benefits derived from longer-term temperature compensation of metabolic rates are negated by extreme daily temperature fluctuations. Instead, littorinid species appear to have evolved mechanisms for immediate metabolic regulation which, in L. saxatilis and L. obtusata and other littorinids, appear to centre on a unique ability for near instantaneous suppression of metabolic rate and entrance into short-term metabolic diapause at temperatures above 20–35 °C, making typical seasonal respiratory compensation mechanisms characteristic of most ectotherms of little adaptive value to littorinid species.  相似文献

为探索贵州苗药光枝勾儿茶内生真菌类群特征、分布部位及其抑菌活性,该研究采用传统方法对贵州省贵阳市和黔西市光枝勾儿茶内生真菌进行分离,并基于分子生物学及统计学对其分类地位进行鉴定及多样性评价,最后通过微量肉汤倍比稀释法筛选具有抑菌活性的菌株。结果表明:(1)从光枝勾儿茶中分离到191 株内生真菌,隶属于3 个门5 个纲10 个目15 个科19 个属,优势属为叶点霉属(Phyllosticta)、间座壳属(Diaporthe)、葡萄座腔菌属(Botryosphaeria)和刺盘孢属(Colletotrichum)。(2)黔西光枝勾儿茶内生真菌香农-维纳多样性指数(H''Q=2.112)较贵阳(H''G=1.801)高,索伦森相似性指数CsG-Q为0.923,不同组织香农-维纳多样性指数为茎(H''S=2.004)>根(H''R=1.764)>叶(H''L=1.654)>果实(H''F=1.473),茎和叶内生真菌的索伦森相似性最高(CsS-L=0.667)。(3)筛选出的21株内生真菌对供试菌大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)和沙门氏菌(Salmonella enterica)具有抑菌效果,其中Diaporthe sp. QX4G6对大肠杆菌、金黄色葡萄球菌和沙门氏菌的最小抑菌浓度分别为12.5、6.25、12.5 mg·mL-1,最小杀菌浓度分别为12.5、6.25、12.5 mg·mL-1。以上研究结果揭示了光枝勾儿茶蕴藏丰富的内生真菌资源,不同地区及组织内生真菌类群组成有差异,多个分离菌株具有抗菌活性,为光枝勾儿茶内生真菌天然抗菌药物或药源研发奠定了基础。  相似文献

物种多样性(SD)与功能多样性(FD)之间存在多种关系,但由于生态系统功能主要由物种的功能属性决定,因而功能多样性对生态系统功能的影响大于物种多样性的影响。但在种间性状差异和物种均匀度这两个构成功能多样性的基本成分中,何者对功能多样性影响更大,并进而决定SD-FD关系尚不明确。通过在高寒矮嵩草(Kobresia humilis)草甸为期6a的刈割(留茬1 cm、3 cm及不刈割)和施肥(尿素7.5 g m~(-2)a~(-1)+磷酸二胺1.8 g m~(-2)a~(-1)、不施肥)控制实验,研究了种间性状差异(33个物种13个性状)和物种均匀度(所有物种)对物种多样性(所有物种)与功能多样性(33个物种13个性状)之间关系的影响。研究结果显示:(1)物种多样性与功能多样性正相关,它们与多性状种间差异负相关,而与物种均匀度正相关。物种均匀度是导致功能多样性变化的主要因素,也是导致SD-FD正相关的原因,这是因为随着物种多样性增加,物种均匀度的增加程度大于多性状种间差异的减少程度,因而功能多样性增加,SD-FD正相关;(2)功能多样性指数(FD_(Rao)和FDis)随物种多样性指数(H')减速递增,表明群落存在一定的功能冗余,且功能冗余随物种多样性的增大而增大,但尚未达到产生SD-FD无相关性的极限H'值;(3)功能多样性对高寒草甸生态系统地上净初级生产力(ANPP)的影响大于物种多样性的影响,二元线性回归显示在同时考虑二者对ANPP的影响时,可排除物种多样性的作用。但由于物种多样性下降或物种丧失引起的物种功能性状丢失或性状空间维度减小将导致功能多样性降低,表明它们之间存在一定互补性,在研究生物多样性与生态系统功能关系时,同时考虑物种多样性和功能多样性的影响仍十分必要。  相似文献

The marcoalga Ulva pertusa was cultured under (20 ± 2)°C, (20 ± 4)°C, (20 ± 6)°C, (20 ± 8)°C and (20 ± 10)°C circadian rhythms of fluctuating temperature conditions, and constant temperature of 20°C was used as the control. The growth rate of macroalga at (20 ± 2)°C, (20 ± 4)°C and (20 ± 6)°C were significantly higher than that at constant temperature of 20°C, while growth rate at (20 ± 8)°C and (20 ± 10)°C were significantly lower than that at constant temperature of 20°C. The growth rate of macroalga was a quadratic function of the thermal amplitude. Such a growth model can be described by G = β 0 + β 1(TA) + β 2(TA)2, where G represents the relative growth rate, TA is thermal amplitude in degree Celsius, β 0 is the intercept on the G axis, and β 1 and β 2 are the regression coefficients. The optimal thermal amplitude for the growth of thallus at mean temperature of 20°C was estimated to be ± 3.69°C. Analysis of biochemical composition at the final stages of thaulls growth revealed that diel fluctuating temperature caused various influences (P < 0.05). The content of chlorophyll, protein and total solute carbohydrate at (20 ± 2)°C and (20 ± 4)°C were slightly higher than those at constant temperature of 20°C, however no statistically significant differences were found among them (P > 0.05). While osmolytes (total solute carbohydrate and free proline) at (20 ± 10)°C were significantly higher than that at 20°C (P < 0.05). Therefore, more chlorophyll and carbohydrate production might account for the enhancement in the growth of macroalga at the diel fluctuating temperatures in the present study. Handling editor: S. M. The brown planthopper (BPH) is one of the most destructive insect pests of rice in Thailand. We performed a cluster analysis that revealed the existence of four groups corresponding to the variation of virulence against BPH resistance genes in 45 BPH populations collected in Thailand. Rice cultivars Rathu Heenati and PTB33, which carry Bph3, showed a broad-spectrum resistance against all BPH populations used in this study. The resistant gene Bph3 has been extensively studied and used in rice breeding programs against BPH; however, the chromosomal location of Bph3 in the rice genome has not yet been determined. In this study, a simple sequence repeat (SSR) analysis was performed to identify and localize the Bph3 gene derived from cvs. Rathu Heenati and PTB33. For mapping of the Bph3 locus, we developed two backcross populations, BC1F2 and BC3F2, from crosses of PTB33 × RD6 and Rathu Heenati × KDML105, respectively, and evaluated these for BPH resistance. Thirty-six polymorphic SSR markers on chromosomes 4, 6 and 10 were used to survey 15 resistant (R) and 15 susceptible (S) individuals from the backcross populations. One SSR marker, RM190, on chromosome 6 was associated with resistance and susceptibility in both backcross populations. Additional SSR markers surrounding the RM190 locus were also examined to define the location of Bph3. Based on the linkage analysis of 208 BC1F2 and 333 BC3F2 individuals, we were able to map the Bph3 locus between two flanking SSR markers, RM589 and RM588, on the short arm of chromosome 6 within 0.9 and 1.4 cM, respectively. This study confirms both the location of Bph3 and the allelic relationship between Bph3 and bph4 on chromosome 6 that have been previously reported. The tightly linked SSR markers will facilitate marker-assisted gene pyramiding and provide the basis for map-based cloning of the resistant gene.  相似文献

In vitro culture is an important aid for ex situ conservation of rare, endemic or threatened plants. In this work, we establish an efficient method for the seed germination, seedling development, and axillary shoot propagation of Centaurea zeybekii Wagenitz. The seeds, collected from a wild population, were surface sterilised and cultured on various in vitro germination media. The effects of photoperiod and temperature on seed germination were also investigated. Germinations were obtained after 6 weeks in culture and the radicle emergence was evaluated as a main indicator. A high frequency of germination was obtained on distilled water supplemented with vitamines and 1 mg/L GA3. Although the seed germination frequencies were not affected by photoperiod, the highest germination frequency was obtained at 24 ± 2°C. A high frequency of axillary shoot proliferation was produced on MS medium supplemented with 1 mg/L BA. Then, the axillary shoots were separated and transferred to MS medium with or without plant growth regulators for rooting. Rhizogenezis was promoted after 6 weeks only in MS and 1/2 MS media containing 0.5 mg/L IBA. The rooting process was very slow and the percentage of shoot rooting was also very low (15%). The present study not only enables reinforcement of wild plant populations using ex situ growth of individuals, but it also helps to large number of aseptic seedling to use it in clonaly micropropagation studies.  相似文献

Tetranychus evansi Baker and Pritchard and Tetranychus urticae Koch (Acari: Tetranychidae) are important pests of Solanaceae in many countries. Several studies have demonstrated that T. urticae is an acceptable prey to many predatory mites, although the suitability of this prey depends on the host plant. T. evansi, has been shown to be an unfavorable prey to most predatory mites that have been tested against it. The predator Phytoseiulus fragariae Denmark and Schicha (Acari: Phytoseiidae) has been found in association with the two species in Brazil. The objective of this work was to compare biological parameters of P. fragariae on T. evansi and on T. urticae as prey. The study was conducted under laboratory conditions at 10, 15, 20, 25 and 30°C. At all temperatures, survivorship was lower on T. evansi than on T. urticae. No predator reached adulthood at 10°C on the former species; even on the latter species, only about 36% of the predators reached adulthood at 10°C. For both prey, in general, duration of each life stage was shorter, total fecundity was lower and intrinsic rate of population increase (r m ) was higher with increasing temperatures. The slower rate of development of P. fragariae on T. evansi resulted in a slightly higher thermal requirement (103.9 degree-days) on that prey than on T. urticae (97.1 degree-days). The values of net reproduction rate (R 0), intrinsic rate of increase (r m ) and finite rate of increase (λ) were significantly higher on T. urticae, indicating faster population increase of the predator on this prey species. The highest value of r m of the predator was 0.154 and 0.337 female per female per day on T. evansi and on T. urticae, respectively. The results suggested that P. fragariae cannot be considered a good predator of T. evansi.  相似文献

植物化石气孔参数分析是目前恢复古大气二氧化碳浓度较为精准的方法之一,银杏类和松柏类等是恢复古大气CO_2浓度常用的化石类群。本文利用新疆准噶尔盆地下侏罗统三工河组的松柏类掌鳞杉科Brachyphyllum(Hirmeriella?)sp.化石对早侏罗世大气CO_2浓度进行了重建,获得早侏罗世大气CO_2浓度为~1200ppm,丰富了早侏罗世大气CO_2浓度信息,进一步说明掌鳞杉科植物通过气孔比率法在重建侏罗纪大气CO_2浓度方面的可靠性。掌鳞杉科植物的旱生构造和较高的大气CO_2浓度表明早侏罗世Toarcian期大洋缺氧事件在陆地生态系统内可能产生了一定的响应。  相似文献

A facultatively anaerobic, thermophilic, xylanolytic bacterium was isolated from a sample collected from the Diyadin Hot Springs, Turkey. According to morphological, biochemical and molecular identification, this new strain was suggested to be representative of the Anoxybacillus pushchinoensis and it was designated as Anoxybacillus pushchinoensis strain A8. It exhibited 97% similarity to 16S rRNA gene sequence of A. pushchinoensis and 77% DNA homology by DNA-DNA hybridization studies. Q-sepharose and CM-sepharose chromatography was used to purify an extracellular xylanase to >90% purity from this species. The enzyme had a molecular mass of approximately 83 kDa. The enzyme showed optimum activity at pH 6.5 and it was 96% stable over a broad pH range of 6.5–11 for 24 hours. The enzyme had optimum activity at 55°C and it was 100% stable at temperature between 50–60°C up to 24 hours. Kinetic characterization of the enzyme was performed at temperature optima (55°C) and Vmax and K m were found to be 59.88 U/mg protein and 0.909 mg/mL, respectively. Oat spelt xylan but not xylooligosaccharides was degraded by the enzyme and xylose was the only product detected from oat xylan degradation. This suggested that the enzyme was an exo-acting xylanase.  相似文献

Milk-clotting enzyme from Bacillus licheniformis 5A1 was immobilized on Amberlite IR-120 by ionic binding. Almost all the enzyme activity was retained on the support. The immobilized milk-clotting enzyme was repeatedly used to produce cheese in a batch reactor. The production of cheese was repeated 5 times with no loss of activity. The specific activity calculated on a bound-protein basis was slightly higher than that of free enzyme. The free and immobilized enzyme were highly tolerant to repeated freezing and thawing. The optimum temperature for milk-clotting activity was 70 °C with the free enzyme whereas, it was ranged from 70 to 80 °C with the immobilized milk-clotting enzyme. The activation energy (E A) of the immobilized milk-clotting enzyme was lower than the free enzyme (E A = 1.59 and 1.99 Kcal mol−1 respectively). The immobilized milk-clotting enzyme exhibited great thermal stability. The milk-clotting optimum pH was 7.0 for both free and immobilized enzyme. The Michaelis constant K m of the immobilized milk-clotting enzyme was slightly lower than the free enzyme.  相似文献

The influence of leaves of Quercus suber L. and Eucalyptus globulus Labill. and needles of Pinus pinaster Ait. on a sandstone substrate was assessed through lysimetric studies during a ten-year period at a site in Central Portugal. The decomposition rate of Q. suber leaf litter was similar to that of E. globulus and higher than that of P. pinaster needle litter. The proportion of nitrogen released from the Q. suber leaf litter was higher than that lost from the other organic species. Such a release was proportional to the initial nitrogen content in the substrates. The concentrations of both NH4-N and NO3-N were much higher in leachates collected under Q. suber leaf litter than in those collected under the other organic substrates. A similar trend was found in the leachates collected under the mineral substrate influenced by the studied organic substrates. The leachate concentrations of mineral N (especially NO3-N) were higher from the mineral substrate under Q. suber leaf litter than from this organic substrate itself. The mineral substrate under leaf litter of E. globulus or needle litter of P. pinaster showed an increase in exchangeable base cations and pH values, and a decrease in extractable Al. Conversely, in the substrate with Q. suber leaf litter there was only a slight increase in exchangeable base cations and pH values, and a decrease in extractable Al. These results combined with those obtained in soils under E. globulus plantations indicate that changes found in these soils are due to soil and forest management practices rather than to the decomposition process of the respective of leaf litter.  相似文献

A fermentation medium for avilamycin production by Streptomyces viridochromogenes Tü57-1 has been optimized. Important components and their concentrations were investigated using fractional factorial design and Box–Behnken Design. The results showed that soybean flour, soluble starch, MgSO4·7H2O and CaCl2·2H2O are important for avilamycin production. A polynomial model related to medium components and avilamycin yield had been established. A high coefficient of determination (R 2 = 0.92) was obtained that indicated good agreement between the experimental and predicted values of avilamycin yield. Student’s T-test of each coefficient showed that all the linear and quadratic terms had significant effect (P > |T| < 0.05) on avilamycin yield. The significance of tested components was related to MgSO4·7H2O (0.37 g/L), CaCl2·2H2O (0.39 g/L), soybean flour (21.97 g/L) and soluble starch (37.22 g/L). The yield of avilamycin reached 88.33 ± 0.94 mg/L (p < 0.05) that was 2.8-fold the initial yield.  相似文献

Summary. Branched-chain amino acid aminotransferase was purified by several column chromatographies from Helicobacter pylori NCTC 11637, and the N-terminal amino acid sequence was analyzed. The enzyme gene was sequenced based on a putative branched-chain amino acid aminotransferase gene, ilvE of H. pylori 26695, and the whole amino acid sequence was deduced from the nucleotide sequence. The enzyme existed in a homodimer with a calculated subunit molecular weight (MW) of 37,539 and an isoelectric point (pI) of 6.47. The enzyme showed high affinity to 2-oxoglutarate (K m = 0.085 mM) and L-isoleucine (K m = 0.34 mM), and V max was 27.3 μmol/min/mg. The best substrate was found to be L-isoleucine followed by L-leucine and L-valine. No activity was shown toward the D-enantiomers of these amino acids. The optimal pH and temperature were pH 8.0 and 37 °C, respectively.  相似文献

A life-table was constructed for a little known aphidiid waspLysiphlebia mirzai, a parasitoid of cereal aphid,Rhopalosiphum maidis. The female parasitoid survived 6.4 ± 1.17 (SD) days and oviposited intensively 4.0 ± 0.47 days. The total fecundity rate, Rt, was 169.2 ± 6.94 mummies/female and net reproductive rate, Ro, was 92.70 female offspring/female. The intrinsic total fecundity rate, rt, and intrinsic rate of natural increase, rm, the finite rate of total fecundity, λt, and finite rate of increase, λm, was 0.27048, 0.24155, 1.31059 and 1.27322 respectively. The mean generation time (18.75 days) and doubling time of the population (2.87 days) was slightly higher than other aphidiids studied so far. The proportion of female progenies decreased significantly on the successive oviposition days.   相似文献

The photosynthetic and respiratory responses of Gracilaria salicornia in the subtropical waters of Japan (in Okinawa) and in the tropical waters of Thailand (in Rayong and Phuket Provinces), were studied under various conditions of irradiance, salinity and temperature. This alga showed adaptability in its photosynthetic and respiratory responses to oceanic salinity as well as to subtropical to tropical temperature. Significant differences in the photosynthetic and respiratory rates among the G. salicornia populations collected from the above sites were observed. The Phuket population showed adaptability to high irradiance and temperature, characteristic of its natural sun-exposed environment, as it exhibited the highest I k and I c, and lowest α and higher P max at 30–35°C. On the other hand, the Okinawa population demonstrated adaptability to low submarine irradiance as it had a lower I k and I c than either population from Thailand. Its P max at 20–25°C was also higher than that of the Phuket population. The Rayong population, however, showed the highest P max, Rd, α, I k and I c, suggesting its adaptability to both sun and shade light conditions in its natural environment. An arginine specific protease, Sp-protease, was purified by column chromatography from freeze-dried Spirulina platensis using a five-step process. Purified Sp-protease has a molecular weight of 80 kDa. It hydrolyzed the synthetic substrates containing arginine residue in the P1 position but did not hydrolyze synthetic substrates containing other amino acid residues, including lysine residue in the P1 position. Among the synthetic substrates tested, a substrate of plasminogen activator (Pyr-Gly-Arg-MCA) was hydrolyzed most effectively with the enzyme (Km = 5.5 × 10−6 M), and fibrin gel was solubilized via activation of intrinsic plasminogen to plasmin with the enzyme. Activity was inhibited completely with camostat mesilate (Ki = 1.1 × 10−8 M) and leupeptin (Ki = 3.9 × 10−8 M) but was not inhibited with Nα-tosyl-L-lysine chloromethyl ketone (TLCK). The optimum pH of the enzyme has a range of pH 9.0 to pH 11.0. The optimum temperature was 50°C; the enzyme was stable at 0–50°C.  相似文献

The genome sequence of Streptomyces coelicolor A3(2) contains 51 putative lipase and esterase genes mostly of unknown function. The gene estB (locus SCO 6966) was expressed as a His-tagged protein in E. coli. Esterase B was active at low temperatures exerting its maximum activity at 30°C and retaining more than 25% of its activity at 4°C. The optimum pH was 8–8.5. The enzyme was active against short synthetic p-nitrophenylesters (C2–C10) with maximum activity towards the acetate ester (C2).
