Intact and fully O[emsp4 ]-sulfonated glycosaminoglycans (GAGs) including chondroitin sulfate, dermatan sulfate, hyaluronan, heparan sulfate and heparin were chemically de-O-sulfonated on their hexosamine C-6 position (6-O[emsp4 ]-desulfonation) using N,O[emsp4 ]-bis(trimethylsilyl) acetamide. 1H NMR spectroscopy and chemical compositional analysis showed that the chemical de-O[emsp4 ]-sulfonation at C-6 position of hexosamine residues in both intact and fully O[emsp4 ]-sulfonated GAGs was completely achieved. Since GAGs and their derivatives are often used as anticoagulant agents, their anti-amidolytic activities were determined. While most of anticoagulant activity of fully O[emsp4 ]-sulfonated GAGs (FGAGs) and heparin disappeared following chemical 6-O[emsp4 ]-desulfonation, the activity of 6-O-desulfonated fully O[emsp4 ]-sulfonated dermatan sulfate (De6FDS) remained. This observation suggests the importance of the position of O-sulfonate groups for anti-coagulant activity.  相似文献

The whole tissue of the earthworm (Eisenia andrei) was lyophilized and extracted to purify glycosaminoglycans. Fractions, eluting from an anion-exchange column at 1.0 M and 2.0 M NaCl, showed the presence of acidic polysaccharides on agarose gel electrophoresis. Monosaccharide compositional analysis showed that galactose and glucose were most abundant monosaccharides in both fractions. Depolymerization of the polysaccharide mixture with glycosaminoglycan-degrading enzymes confirmed the presence of chondroitin sulfate/dermatan sulfate and heparan sulfate in the 2.0 M NaCl fraction. The content of GAGs (uronic acid containing polysaccharide) in the 2.0 M NaCl fraction determined by carbazole assay was 2%. Disaccharide compositional analysis using liquid chromatography–electrospray ionization mass spectrometry (LC–ESI–MS) analysis after chondroitinase digestion (ABC and ACII), showed that the chondroitin sulfate/dermatan sulfate contained a 4-O-sulfo (76%), 2,4-di-O-sulfo (15%), 6-O-sulfo (6%), and unsulfated (4%) uronic acid linked N-acetylgalactosamine residues. LC–ESI–MS analysis of heparin lyase I/II/III digests demonstrated the presence of N-sulfo (69%), N-sulfo-6-O-sulfo (25%) and 2-O-sulfo-N-sulfo-6-O-sulfo (5%) uronic acid linked N-acetylglucosamine residues.  相似文献

Fast atom bombardment tandem mass spectrometry has been used in the characterization of non-, mono-, di- and trisulfated disaccharides from chondriotin sulfate, dermatan sulfate and hyaluronan. The positional isomers of the sulfate group of mono- and disulfated disaccharides were distinguished from each other by both positive- and negative-ion fast atom bombardment tandem mass spectra, which gave sufficient information characteristic of the isomers. The anomeric isomers of nonsulfated disaccharides were characterized by the technique in the positive-ion mode. Studies using lysosomal membrane vesicles have suggested that efflux of the sulfate that results from lysosomal glycosaminoglycan degradation is carrier-mediated. In this study, glycosaminoglycan degradation and sulfate efflux were examined using cultured skin fibroblasts and lysosomes deficient in the lysosomal enzymeN-acetylgalactosamine-4-sulfatase. Such fibroblasts store dermatan sulfate lysosomally, which could be labelled biosynthetically with Na 2 35 SO4. The addition of recombinantN-acetylgalactosamine-4-sulfatase to the media of35S labelled fibroblasts degraded up to 82% of the stored dermatan [35S] sulfate over a subsequent 96 h chase and released inorganic [35S] sulfate into the medium. In the presence of 4-acetamido-4-isothiocyanatostilbene-2,2-disulfonic acid (SITS), sulfate was reused to a minor extent in newly synthesized proteoglycan. Isolated granules from recombinant enzyme supplemented fibroblasts degraded stored dermatan [35S]sulfate to sulfate which was rapidly released into the medium at a rate that was reduced by the extra-lysosomal presence of the lysosomal sulfate transport inhibitors SITS, Na2SO4 and Na2MoO4. SITS also inhibited dermatan sulfate turnover, although it had no effect on the action of purified recombinant enzymein vitro. To generate xyloside-primed dermatan sulfate suitable for sequence analysis, skin fibroblasts were incubated withp-hydroxyphenyl--d-xylopyranoside and [3H]galactose, and free [3H]glycosaminoglycan chains were isolated from the culture medium by ion exchange and gel chromatography. After125I labelling of their reducing-terminal hydroxyphenyl groups, chains were subjected to various chemical and enzymatic degradations, both partial and complete, followed by gradient polyacrylamide gel electrophoresis and autoradiographic identification of fragments extending from the labelled reducing-end to the point of cleavage. Results of periodate oxidation-alkaline scission indicated that the xylose moiety remained unsubstituted at C-2/C-3; exhaustive treatment with chondroitin AC-I lyase afforded the fragment HexA-Gal-Gal-Xyl-R (R = radio-iodinated hydroxyphenyl group), and complete degradations with chondroitin ABC lyase as well as testicular hyaluronidase yielded the fragments HexA/HexA-GalNAc-GlcA-Gal-Gal-Xyl-R with or without sulfate on theN-acetylgalactosamine. Partial digestions with testicular hyaluronidase or chondroitin B lyase indicated that glucuronic acid was common in the first three repeats after the linkage region and that iduronic acid could occupy any position thereafter. The pentasaccharide chain of the Forssman antigen, O-(2-acetamido-2-deoxy-α-d-galactopyranosyl)-(1→3)-O-(2-acetamido-2-deoxy-β-d-galactopyranosyl)-(1→3)-O-α-d- galactopyranosyl-(1→4)-O-β-d-galactopyranosyl-(1→4)-d-glucopyranose (46) was synthesized by a block synthesis in which an α-d-glycoside linkage between two d-galactose residues was formed. The trisaccharide O-(6-O-acetyl-2-azido-3,4-di-O-benzoyl-2-deoxy-α-d-galactopyranosyl)- (1→3)-O-(6-O-acetyl-4-O-benzyl-2-deoxy-2-phthalimido-β-d-galactopyranosyl)-(1→3)-6-O-acetyl-2,4-di-O-benzyl- α-d-galactopyranosyl bromide (40) (this was obtained through acetolysis of O-(6-O-acetyl-2-azido-3,4-di-O-benzoyl-2-deoxy-α-d-galactopyranosyl)- (1→3)-O-(6-O-acetyl-4-O-benzyl-2-deoxy-2-phthalimido-β-d-galactopyranosyl)-(1→3)-1,6-anhydro-2,4-di-O-benzyl-β-d- galactopyranose to the acetyl derivative, followed by reaction with titanium tetrabromide under anhydrous conditions) was condensed with benzyl-4-O-(6-O-benzoyl-2,3-di-O-benzyl-β-d-galactopyranosyl)-2,3,6- tri-O-benzyl-β-d-glucopyranoside were in the presence of silver carbonate and perchlorate. The plasma membrane-associated proteoglycans of a malignant human breast cell line (MDA-MB-231) were compared with the corresponding proteoglycans from a normal cell line (HBL-100). The labeled proteoglycans were isolated from the plasma membranes of cells grown in the presence of [3H]glucosamine and [35S]Na2SO4 by extraction with guanidine hydrochloride and subsequently purified by DEAE-ion exchange chromatography. Their structural properties were established by treatment with nitrous acid, heparitinase and chondroitinase ABC, and by gel filtration before and after alkaline -elimination. About 18% of the proteoglycans synthesized by these cell lines were associated with the plasma membranes. The HBL plasma membranes contained 80% heparan sulfate and 20% chondroitin sulfate proteoglycans whereas MDA plasma membranes had 50% heparan sulfate and 50% chondroitin sulfate proteoglycans. The MDA plasma membrane contained two heparan sulfate proteoglycans, both having nearly the same molecular size as the two species secreted into the medium by these cells. The HBL plasma membrane also contained two hydrodynamic size heparan sulfate proteoglycans. The larger hydrodynamic size species has a slightly lower molecular size than that secreted into the medium, and the smaller hydrodynamic size species was not detectable in the medium. Investigation of the acetolysis products of the sulphated polysaccharide of the seaweed Aeodes ulvoidea led to the isolation and characterization of the following oligosaccharides: 3-O-α- -galactopyranosyl- -galactose (1), 3-O-(2-O-methyl-α- -galactopyranosyl)- -galactose (2), 4-O-β- -galactopyranosyl-2-O-methyl- -galactose (3), 4-O-β- -galactopyranosyl-2-O-methyl- -galactose (4), O-β- -galactopyranosyl-(1→4)-O-α- -galactopyranosyl-(1→3)- -galactose (5), O-α- -galactopyranosyl-(1→3)-O-β- -galactopyranosyl-(1→4)- -galactose (6), O-α- -galactopyranosyl-(1→3)-O-β- -galactopyranosyl-(1→4)-2-O-methyl- -galactose (7), O-(2-O-methyl-α- -galactopyranosyl)-(1→3)-O-β- -galactopyranosyl-(1→4)-2-O-methyl- -galactose (10), and O-α- -galactopyranosyl-(1→3)-O-β- -galactopyranosyl-(1→4)-O-α- -galactopyranosyl-(1→3)- -galactose. A novel 1,2-cis stereoselective synthesis of protected α-d-Gal-(1→2)-d-Glc fragments was developed. Methyl 2-O-acetyl-3-O-allyl-4,6-O-benzylidene-α-d-galactopyranosyl-(1→2)-3-O-benzoyl-4,6-O-benzylidene-α-d-glucopyranoside (13), methyl 2-O-acetyl-3-O-allyl-4,6-O-benzylidene-α-d-galactopyranosyl-(1→2)-3,4,6-tri-O-benzoyl-α-d-glucopyranoside (15), methyl 2-O-acetyl-3-O-allyl-4,6-O-benzylidene-α-d-galactopyranosyl-(1→2)-3-O-benzoyl-4,6-O-benzylidene-β-d-glucopyranoside (17), and methyl 2-O-acetyl-3-O-allyl-4,6-O-benzylidene-α-d-galactopyranosyl-(1→2)-3,4,6-tri-O-benzoyl-β-d-glucopyranoside (19) were favorably obtained by coupling a new donor, isopropyl 2-O-acetyl-3-O-allyl-4,6-O-benzylidene-1-thio-β-d-galactopyranoside (2), with acceptors, methyl 3-O-benzoyl-4,6-O-benzylidene-α-d-glucopyranoside (4), methyl 3,4,6-tri-O-benzoyl-α-d-glucopyranoside (5), methyl 3-O-benzoyl-4,6-O-benzylidene-β-d-glucopyranoside (8), and methyl 3,4,6-tri-O-benzoyl-β-d-glucopyranoside (12), respectively. By virtue of the concerted 1,2-cis α-directing action induced by the 3-O-allyl and 4,6-O-benzylidene groups in donor 2 with a C-2 acetyl group capable of neighboring-group participation, the couplings were achieved with a high degree of α selectivity. In particular, higher α/β stereoselective galactosylation (5.0:1.0) was noted in the case of the coupling of donor 2 with acceptor 12 having a β-CH3 at C-1 and benzoyl groups at C-4 and C-6.  相似文献

We prepared chitosan sulfated derivatives to address the common structural requirement of the sulfate pattern to block P-selectin-mediated tumor cell adhesion. Our results indicate that 6-O-sulfation of chitosan is indispensable for inhibition of P-selectin binding to human melanoma A375 cells. Furthermore, additional N-sulfation or 3-O-sulfation dramatically enhanced the inhibitory activity of 6-O-sulfated chitosan, suggesting that efficient anti-P-selectin adhesion activity of sulfated saccharides requires the synergy of 6-O-sulation and N- or 3-O-sulfation in glucosamine units.  相似文献

The Halide ion-catalysed reaction of benzyl exo-2,3-O-benzylidene-α- -rhamnopyranoside with tetra-O-benzyl-α- -galactopyranosyl bromide and hydrogenolysis of the exo-benzylidene group of the product 2 gave benzyl 3-O-benzyl-4-O-(2,3,4,6-tetra-O-benzyl-α- -galactopyranosyl)-α- -rhamnopyranoside (6). Compound 2 was converted into 4-O-α- -galactopyranosyl- -rhamnose. The reaction of 6 with tetra-O-acetyl-α- -glucopyranosyl bromide and removal of the protecting groups from the product gave 4-O-α- -galactopyranosyl-2-O-β- -glucopyranosyl- -rhamnose.  相似文献

The nondialyzable sialoglycopeptides dcrived from defatted, protease digested whole boyine heifer brain tissue were isolated and characterized. The partially purified bovine heifer brain tissue were isolated and characterized. The partially purified sialoglycopeptide mixtures contained 7-8% ester sulfate. The amount of nonsulfated material present was of the order of 15%. The total mixture was fractionated by anion exchange chromatography on AG1-X2(CI-) using a sodium chloride gradient to yield twelve fractions all of which contained ester sulfate to varying degrees (5.6-21.9%). The molecular size of the fractions ranged from molecular weights of 10,400-12,500. The molar sulfate/galactose plus glucosamine ratios ranged from 0.3 to 1.1. Carbohydrate peptide linkage studies of the fractions by treatment with 0.2 M-NaOH-O.3 M-NaBH4 led to the destruction of a portion of the threonine, serine and galactosamine and the appearance in acid hydrolysates of α-amino-n-butyric acid and galactosaminitol with an increase in alanine. Partial acid hydrolysis of the most abundant sialoglycopeptide liberated detectable amounts of 2-acetarnido-1-(L-4-aspartamido-)-1,2-dideoxy-β-D- glucose. These results indicated that the GalNac at the reducing end of the alkali labile carbohydrate prosthetic groups were linked O-glycosidically to thc hydroxyl groups of threonine and serine and the alkali-stable region of the principal sialoglycopeptide involves N-acetylglycosaminyl-asparagine linkages. Mild acid hydrolysis of the alkali-stable portion of the sulfated fractions indicated that the ester sulfate was confined to the alkali-stable region and exists as galactose 6-O-sulfate and N-acetylglucosa- mine 6-O-sulfate, structural fcatures similar to those present in rat brain glycopcptides (Markgolis & Margolis , 1970; Margolis et al.; 1972). Heparinase and heparitinase were separated from an extract of Flavobacterium heparinum, induced with heparin by using column chromatography on hydroxylapatite. As the heparinase preparation contained chondroitinases B and C, chondroitinase B was removed by rechromatography on a hydroxylapatite column. Chondroitinase C was then eliminated by column chromatography on O-phosphono(“phospho”)-cellulose. The heparinase preparation thus obtained was free from sulfoamidase for 2-deoxy-2-sulfoamino-D-glucose (GlcN-2S), sulfatase for 2-amino-2-deoxy-6-O-sulfo D-glucose (GlcN-6S), as well as (δ4,5glycosiduronase for the unsaturated disaccharides obtained from heparin. The remaining sulfatase for 4-deoxy-α-L-thero-hex-4-enopyranosyluronic acid 2-sulfate (δUA-2S) in the heparinase preparation was removed by affinity chromatography with dermatan sulfate-bound AH-Sepharose 4B coated with dermatan sulfate. The heparitinase preparation separated by column chromatography on hydroxyla patite was purified by affinity chromatography with heparin-bound AH-Sepharose 4B coated with heparin. Sulfatase for 2-amino-2-deoxy-6-O-sulfo-D-glucose (GlcN-6S) and δ4,5glycosiduronase for the unsaturated disaccharides obtained from heparin were removed by this chromatography. Sulfatase for 4-deoxy-α-L-threo-hex-4-enopyranosyluronic acid 2-sulfate (δUA-2S) remaining in the heparitinase preparation was finally removed by column chromatography on hydroxylapatite. The recoveries of the purified preparations of heparinase and heparitinase were estimated to be 39 and 50%, respectively, from the crude enzyme fractions obtained by the first column chromatography on hydroxyl- patite. The purified heparinase and heparitinase were free from all enzymes that could degrade the sulfated unsaturated disaccharides produced from heparin with heparinase.  相似文献

Two apiose-containing kaempferol triosides, together with nine known flavonoids were isolated from the leaves of Silphium perfoliatum L. Their structures were elucidated by acid hydrolysis and spectroscopic methods including UV, LSI MS, FAB MS, CI MS, 1H, 13C and 2D-NMR, DEPT, HMQC and HMBC experiments. The two new compounds were identified as kaempferol 3-O-β- -apiofuranoside 7-O-α- -rhamnosyl-(1′→6)-O-β- -galactopyranoside and kaempferol 3-O-β- -apiofuranoside 7-O-α- -rhamnosyl-(1→ 6)-O-β- (2-O-E-caffeoylgalactopyranoside).  相似文献

Condensation of methyl 4-O-acetyl-3-O-(2,3,4-tri-O-acetyl-α- -rhamnopyranosyl)-α- -rhamnopyranoside with 2,3,4,6-tetra-O-benzyl-α- -glucopyranosyl chloride gave a mixture of methyl O-[2,3,4,6-tetra-O-benzyl-α- (4) and -β- -glucopyranosyl]-(1→2)-O-[(2,3,4-tri-O-acetyl-α- -rhamnopyranosyl)-(1→3)]-4-O-acetyl-α- -rhamnopyranoside (9) in 43:7 proportion in 63% yield. After chromatographic separation, removal of the benzyl and acetyl groups gave methyl O-α- -glucopyranosyl-(1→2)-[O-α- -rhamnopyranosyl-(1→3)]-α- -rhamnopyranoside and the β anomer. Removal of benzyl groups of 4 was followed by tritylation, acetylation, and detritylation of the α- -glucopyranosyl group, and finally condensation with benzyl (2,3,4-tri-O-benzyl- -glucopyranosyl chloride)uronate gave a mixture of two tetrasaccharides (15 and 16), containing the α- and β- -glucopyranosyluronic acid groups in the ratio 81:19, and an overall yield of 71%. After chromatographic separation, alkaline hydrolysis and hydrogenation of 15 gave methyl O-α- -glucopyranosyluronic acid-(1→6)-O-α- -glucopyranosyl-(1→2)-[O-α- -rhamnopyranosyl-(1→3)]-α- -rhamnopyranoside. The β- anomer was obtained by similar treatment of 16. 6-O-α- -glucopyranosyluronic acid-α,β- -glucopyranose was synthesized as a model compound.  相似文献

Three phenolic glycosides 5-O-{[5′′-O-E-(4′′′-O-threo-guaiacylglycerol)-feruloyl]-β-apiofuranosyl-(1→2)-β-xylopyranosyl} gentisic acid, 5-O-[(5′′-O-vanilloyl)-β-apiofuranosyl-(1→2)-β-xylopyranosyl] gentisic acid and 1-O-[E-(4′′′-O-threo-guaiacylglycerol)-feruloyl]-3-O-β-galacturonopyranosyl glycerol were isolated and identified from the roots of Medicago truncatula together with four known 5-O-β-xylopyranosyl gentisic acid, vicenin-2, hovetrichoside C and pterosupin identified for the first time in this species. Benzoylation of benzyl 2-acetamido-2-deoxy-4,6-O-isopropylidene-α-d-glucopyranoside, benzyl 2-deoxy-2-(dl-3-hydroxytetradecanoylamino)-4,6-O-isopropylidene-α-d-glucopyranoside, and benzyl 2-deoxy-4,6-O-isopropylidene-2-octadecanoylamino-β-d-glucopyranoside, with subsequent hydrolysis of the 4,6-O-isopropylidene group, gave the corresponding 3-O-benzoyl derivatives (4, 5, and 7). Hydrogenation of benzyl 2-acetamido-4,6-di-O-acetyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-α-d-glucopyranoside, followed by chlorination, gave a product that was treated with mercuric actate to yield 2-acetamido-1,4,6-tri-O-acetyl-2-deoxy-3-O-[d-1-(methoxycarbonyl)ethyl]-β-d-glucopyranose (11). Treatment of 11 with ferric chloride afforded the oxazoline derivative, which was condensed with 4, 5, and 7 to give the (1→6)-β-linked disaccharide derivatives 13, 15, and 17. Hydrolysis of the methyl ester group in the compounds derived from 13, 15, and 17 by 4-O-acetylation gave the corresponding free acids, which were coupled with l-alanyl-d-isoglutamine benzyl ester, to yield the dipeptide derivatives 19–21 in excellent yields. Hydrolysis of 19–21, followed by hydrogenation, gave the respective O-(N-acetyl-β-muramoyl-l-alanyl-d-isoglutamine)-(1→6)-2-acylamino-2-deoxy-d-glucoses in good yields. The immunoadjuvant activity of these compounds was examined in guinea-pigs.  相似文献

Background

Sulf1 is a cell-surface sulfatase removing internal 6-O-sulfate groups from heparan sulfate (HS) chains. Thereby it modulates the activity of HS-dependent growth factors. For HS interaction Sulf1 employs a unique hydrophilic domain (HD).

Methods

Affinity-chromatography, AFM-single-molecule force spectroscopy (SMFS) and immunofluorescence on living cells were used to analyze specificity, kinetics and structural basis of this interaction.

Results

Full-length Sulf1 interacts broadly with sulfated glycosaminoglycans (GAGs) showing, however, higher affinity toward HS and heparin than toward chondroitin sulfate or dermatan sulfate. Strong interaction depends on the presence of Sulf1-substrate groups, as Sulf1 bound significantly weaker to HS after enzymatic 6-O-desulfation by Sulf1 pretreatment, hence suggesting autoregulation of Sulf1/substrate association. In contrast, HD alone exhibited outstanding specificity toward HS and did not interact with chondroitin sulfate, dermatan sulfate or 6-O-desulfated HS. Dynamic SMFS revealed an off-rate of 0.04/s, i.e., ~ 500-fold higher than determined by surface plasmon resonance. SMFS allowed resolving the dynamics of single dissociation events in each force–distance curve. HD subdomain constructs revealed heparin interaction sites in the inner and C-terminal regions of HD.

Conclusions

Specific substrate binding of Sulf1 is mediated by HD and involves at least two separate HS-binding sites. Surface plasmon resonance KD-values reflect a high avidity resulting from multivalent HD/heparin interaction. While this ensures stable cell–surface HS association, the dynamic cooperation of binding sites at HD and also the catalytic domain enables processive action of Sulf1 along or across HS chains.

General significance

HD confers a novel and highly dynamic mode of protein interaction with HS.  相似文献
