Glycosaminoglycans (GAGs) involved in the formation of the teeth of Ambystoma mexicanum were located and characterized with the cuprolinic blue (CB) staining method and transmission electron microscopy (TEM). Glycosaminoglycan-cuprolinic blue precipitates (GAGCB) were found in different compartments of the mineralizing tissue. Various populations of elongated GAGCB could be discriminated both according to their size and their preferential distribution in the extracellular matrix (ECM). GAGCB populations that differ in their composition could be attributed not only to the compartments of the ECM but also to different zones and to different tooth types (early-larval and transformed). Larger precipitates were only observed within the dentine matrix of the shaft of the early-larval tooth. The composition of the populations differed significantly between the regions of the transformed tooth: pedicel, shaft and dividing zone. In later stages of tooth formation, small-sized GAGCBs were seen as intracellular deposits in the ameloblasts. It is concluded that the composition of GAGCB populations seems to play a role in the mineralization processes during tooth development in A. mexicanum and influence qualitative characteristics of the mineral in different tooth types and zones, and it is suggested that GAGs might be resorbed by the enamel epithelium during the late phase of enamel formation.  相似文献

Summary Regenerating tails and limbs of axolotl larvae (A. mexicanum) were studied for overall growth and for mitosis after the animals received intraperitoneal injections of all-trans retinoic acid. Both processes were depressed to approximately the same extent (60–70%). Some mitosis always survived, even when the treatment was in effect during the entire history of the regenerate. The treatment duration was a major variable in the severity of the effect, whereas the post-amputation age of the regenerate was not. In limb regenerates the epithelial cap and the mesenchymal blastema were affected to roughly the same degree.Supported by PHS Grant 507RR7031H of the BMRG, Indiana University  相似文献

The crystal-face dependence of the dye-sensitized photocurrents and the adsorption properties of benzothiazole merocyanine (Mc[18,1]) dye molecules were investigated, using atomically flat (1 0 0) and (1 1 0) TiO2 single crystal surfaces. From the estimation of the amount of the transferred charge from the TiO2 surface to CO groups of dye molecules based on NEXAFS data, it was revealed that the interaction of the adsorbed molecules and the (1 1 0) surface was much stronger than that for the (1 0 0) surface. On the other hand, the absorbed photon to current conversion efficiency (APCE) value was almost the same for both surfaces. We suggested a possible explanation as follows: the energy difference between the LUMO of Mc[18,1] and the conduction band of TiO2 was large enough to give a nearly 100% quantum efficiency of electron transfer from photoexcited dye to TiO2, which made the difference in the interaction between dye molecules and TiO2 not apparent. The incident photon to current conversion efficiency (IPCE) for the (1 0 0) surface was much larger than that for the (1 1 0) surface, which was explained by the fact that the amount of the adsorbed dye molecules on the (1 0 0) surface was larger than on the (1 1 0) surface, probably due to the larger surface density of five-coordinated Ti sites in the former surface.  相似文献

The objective of this study was to develop a preservation method for canine sperm using microencapsulation. Pooled ejaculates from three beagles (Canis familiaris) were extended in egg yolk Tris extender and were encapsulated in gel (alginate only) or polycation (poly-l-lysine membrane bound) microcapsules at 0.75% and 1.0% alginate concentration. In Experiment 1, characteristics of microcapsule and microencapsulated sperm were evaluated during chilling storage for 48 h. Gel microcapsules at 0.75% alginate concentration had a teardrop-like structure with fragility, whereas those at 1.0% alginate had a solid spherical structure. In all groups, diameter of the microcapsules increased with duration of storage (P < 0.05). Alginate concentration did not affect the sperm recovery rate from microcapsules. Total average recovery rate of sperm from polycation microcapsules was lower than that of gel microcapsules (P < 0.05). Progressive motility of polycation microencapsulated sperm and unencapsulated sperm (control) was higher than that of the gel microencapsulated sperm, both at 0.75% and 1.0% alginate concentration (P < 0.05), although viability of sperm was similar among the three groups. In Experiment 2, to evaluate the sperm longevity after chilling storage, sperm were microencapsulated in polycation microcapsules at 1.0% alginate concentration, stored at 4 °C for 0, 1, 4, and 7 d, and then cultured at 38.5 °C for 0, 6, and 24 h. Progressive motility and viability of microencapsulated sperm were higher than those of unencapsulated spermatozoa at 0 to 24 h of culture after 4 and 7 d of chilling storage (P < 0.05). In conclusion, polycation microencapsulation at 1.0% alginate concentration can be successfully applied for chilling storage of canine sperm by maintaining motility and viability for up to 7 d.  相似文献

This study was performed to determine the effects of levamisole and levamisole + vitamin C against Syphacia muris naturally infection in rats and to detect its effect on the oxidative parameters in blood and tissues of host. For this purpose, natural infection was diagnosed using the cellophane tape method on the perianal region of rats. Infected rats (total 18) were divided into three groups. On the other hand six without helminth rats were used in this study as negative control group. Group 2 was given an orally levamisole HCl treatment with gastric gavage at a dose level of 20 mg/kg body weight in distilled water, every alternate day. Group 3 was given levamisole HCl via gastric gavage at a dose level of 20 mg/kg and vitamin C was given 1 g/L added to the drinking water. All the treatments continued for a period of 7 days. As a result; levamisole administered to rats at dose of 20 mg/kg orally 98.34% was found to be effective against adult S. muris in the rats. In addition to levamisole + vitamin C is effective to alleviate the oxidative damage in rats infected with S. muris.  相似文献

PsbV2 is a c-type cytochrome present in a very low abundance in the thermophilic cyanobacterium Thermosynechococcus elongatus. We purified this cytochrome and solved its crystal structure at a resolution of 1.5 Å. The protein existed as a dimer in the crystal, and has an overall structure similar to other c-type cytochromes like Cytc6 and Cytc550, for example. However, the 5th and 6th heme iron axial ligands were found to be His51 and Cys101, respectively, in contrast to the more common bis-His or His/Met ligands found in most cytochromes. Although a few other c-type cytochromes were suggested to have this axial coordination, this is the first crystal structure reported for a c-type heme with this unusual His/Cys axial coordination. Previous spectroscopic characterizations of PsbV2 are discussed in relation to its structural properties.  相似文献

Of the several proteins located within sarcomeric A-bands, C-protein, a myosin binding protein (MyBP) is thought to regulate and stabilize thick filaments during assembly. This paper reports the characterization of C-protein isoforms in juvenile and adult axolotls, Ambystoma mexicanum, by means of immunofluorescent microscopy and Western blot analyses. C-protein and myosin are found specifically within the A-bands, whereas tropomyosin and -actin are detected in the I-bands of axolotl myofibrils. The MF1 antibody prepared against the fast skeletal muscle isoform of chicken C-protein specifically recognizes a cardiac isoform (Axcard1) in juvenile and adult axolotls but does not label axolotl skeletal muscle. The ALD66 antibody, which reacts with the C-protein slow isoform in chicken, localizes only in skeletal muscle of the axolotl. This slow axolotl isoform (Axslow) displays a heterogeneous distribution in fibers of dorsalis trunci skeletal muscle. The C315 antibody against the chicken C-protein cardiac isoform identifies a second axolotl cardiac isoform (Axcard2), which is present also in axolotl skeletal muscle. The effect of ZnCl2 on the degradation of cellulose was studied to develop conditions to produce useful feedstock chemicals directly from cellulosic biomass. Cellulose containing 0.5 mol of ZnCl2/mol of glucose unit of cellulose was found to degrade at 200 °C when heated for more than 60 s in air. The major non-gaseous products of the degradation were identified as furfural, 5-hydroxymethylfurfural and levulinic acid. The maximum yields for furfural and 5-hydroxymethylfurfural are 8% and 9%, respectively, based on glucose unit of cellulose. These yields are reached after 150 s of heating at 200 °C. A cellulose sample containing 0.5 mol of ZnCl2/mol of glucose unit of cellulose and 5.6 equivalents of water when heated for 150 s at 200 °C produced levulinic acid as the only product in 6% yield. The ZnCl2 mediated controlled degradation of cellulose at 200 °C is shown to produce useful feedstock chemicals in low yield.  相似文献

Cobalt(III) and rhodium(III) complexes of the series of [MIIICl3 − n(P)3 + n]n+ (M = Co or Rh; n = 0, 1, 2 or 3) have been prepared with the use of 1,1,1-tris(dimethylphosphinomethyl)ethane (tdmme) and mono- or didentate phosphines. The single-crystal X-ray analyses of both series of complexes revealed that the M-P and M-Cl bond lengths were dependent primarily on the strong trans influence of the phosphines, and secondarily on the steric congestion around the metal center resulting from the coordination of several phosphine groups. In fact, the M-P(tdmme) bonds became longer in the order of [MCl3(tdmme)] < [MCl2(tdmme)(PMe3)]+ < [MCl(tdmme)(dmpe)]2+ (dmpe = 1,2-bis(dimethylphosphino)ethane) < [M(tdmme)2]3+ for both CoIII and RhIII series of complexes, while the M-Cl bond lengths were shortened in this order (except for [M(tdmme)2]3+). Such a steric congestion around the metal center can also account for the structural and spectroscopic characteristics of the series of complexes, [MCl(tdmme)(dmpm, dmpe or dmpp)]2+ (dmpm = bis(dimethylphosphino)methane, dmpp = 1,3-bis(dimethylphosphino)propane). The X-ray analysis for [CoCl(tdmme)(dmpm or dmpe)](BF4)2 showed that all Co-P bonds in the dmpm complex were shorter by 0.03-0.04 Å than those in the dmpe complex. Furthermore, the first d-d transition energy of the CoIII complexes and the 1JRh-P(tdmme) coupling constants observed for the RhIII complexes indicated an unusual order in the coordination bond strengths of the didentate diphosphines, i.e., dmpm > dmpe > dmpp.  相似文献

The crystal structures of CsGST in two different space groups revealed that Asp26 and His79 coordinate a zinc ion. In one space group, His46 of an adjacent molecule participates in the coordination within 2.0 Å. In the other space group, Asp26, His79 and a water molecule coordinate a zinc ion. The CsGST–D26H structure showed that four histidine residues – His26 and His79 from one molecule and the same residues from a symmetry-related neighboring molecule – coordinate a zinc ion. The coordinated zinc ions are located between two molecules and mediate molecular contacts within the crystal.  相似文献

Five novel complexes, Co(OBt)2 · 7H2O (1) (OBt = 1-hydroxybenzotriazole ion), Ni3(OBt)6 · 6H2O (2), [Ag(OBt)(HOBt)]n (3), [Zn(OBt)2]n (4) and [Cu2(OBt)4 · 3H2O]n (5) were synthesized by hydrothermal method and characterized by elemental analysis, IR spectroscopy, TGA, XRPD, and single-crystal X-ray diffraction. The results from single-crystal X-ray diffraction indicate that 1-5 are zero-dimensional (0D), zero-dimensional, one-dimensional (1D), and three-dimensional (3D) frameworks, respectively. In particular, 3 is twin crystal; 4 possesses of double-stranded chains; 5 crystallizes in orthorhombic space group P212121 with a helical chain in its structure. The luminescence properties and the magnetic properties of the five complexes were investigated.  相似文献

Alicyclobacillus acidocaldarius endoglucanase Cel9A (AaCel9A) is an inverting glycoside hydrolase with β-1,4-glucanase activity on soluble polymeric substrates. Here, we report three X-ray structures of AaCel9A: a ligand-free structure at 1.8 Å resolution and two complexes at 2.66 and 2.1 Å resolution, respectively, with cellobiose obtained by co-crystallization and with cellotetraose obtained by the soaking method. AaCel9A forms an (α/α)6-barrel like other glycoside hydrolase family 9 enzymes. When cellobiose is used as a ligand, three glucosyl binding subsites are occupied, including two on the glycone side, while with cellotetraose as a ligand, five subsites, including four on the glycone side, are occupied. A structural comparison showed no conformational rearrangement of AaCel9A upon ligand binding. The structural analysis demonstrates that of the four minus subsites identified, subsites − 1 and − 2 show the strongest interaction with bound glucose. In conjunction with the open active-site cleft of AaCel9A, this is able to reconcile the previously observed cleavage of short-chain oligosaccharides with cellobiose as main product with the endo mode of action on larger polysaccharides.  相似文献

Ectothermy in reptiles involves strong environmental temperature dependence to maintain body temperature. High mountains are extreme habitats which, because of their marked temperature decrease with altitude, impose difficulties for reptile thermoregulation, as the number of optimum-temperature days diminish in altitude. We studied the activity patterns of the lizard Psammodromus algirus, which has a wide altitudinal distribution, at six sampling stations that spanned an altitudinal range from 300 to 2500 m asl in Sierra Nevada (SE Iberian Peninsula). We performed censuses of the species during the seasonal and daily activity period, and measured lizard body temperature and environmental temperature in the six sampling plots along the altitude gradient. The body temperature of the lizards remained closely the same even though environmental temperature decreased around 9°C along that gradient. Seasonal activity peaked at the beginning of the activity period in highlands, but in the middle of the season in middle and lowland regions. However, newborns appeared in the field almost at the same time along the altitude gradient. We suggest that the thermoregulatory strategies of the lizards must vary along this altitudinal gradient, permitting lizards to keep their body temperature close to a very similar optimum and thus, develop very similar circa-annual activity patterns.  相似文献

The effects of a rapid transfer from a low (3 °C) to a warm (23 °C) temperature on oxidative stress markers and antioxidant defenses were studied in the brain, liver and kidney of the goldfish, Carassius auratus. Cold-acclimated fish were acutely moved to 23 °C and sampled after 1, 6, 12, 24, 48 or 120 h of warm temperature exposure. Lipid peroxide levels increased quickly during the first few hours at 23 °C, but thiobarbituric acid-reactive substances changed little. Protein carbonyl content was reduced by 20–40% in the liver over the entire experimental course, but increased transiently in the kidney. The content of high-molecular mass thiols decreased by two-thirds in the brain and was affected slightly in other organs. By contrast, total low-molecular mass thiols (e.g. glutathione and others) increased transiently. Activities of the primary antioxidant enzymes—superoxide dismutase and catalase—were generally unaffected in goldfish organs, whereas glutathione-dependent enzymes were elevated in the brain and kidney after 24–48 h at 23 °C. Glutathione peroxidase increased by 1.5–2.3-fold and glutathione-S-transferase by 1.7-fold. Hence, a short-term exposure to warm temperature disturbed several oxidative stress markers, but only slightly affected the activities of antioxidant enzymes. However, comparison of the current data for cold-acclimated winter fish with the same parameters in summer fish suggests that longer exposure to high ambient temperature requires the enhancement of activities of glutathione-dependent enzymes for maintaining the steady-state levels lipid peroxidation and protein oxidation in goldfish tissues.  相似文献

Oxygen transport in Myriapoda is maintained by a unique 6 × 6mer hemocyanin, that is, 36 subunits arranged as six hexamers (1 × 6mers). In the sluggish diplopod Spirostreptus, the 1 × 6mers seem to operate as almost or fully independent allosteric units (h ∼ 1.3; P50 ∼ 5 torr), whereas in the swift centipede Scutigera, they intensively cooperate allosterically (h ∼ 10; P50 ∼ 50 torr). Here, we show the chemomechanical basis of this differential behavior as deduced from hybrid 6 × 6mer structures, obtained by single-particle cryo-electron microscopy of the Scutigera 6 × 6mer (10.0 Å resolution according to the 0.5 criterion) and docking of homology-modeled subunits from Scutigera and two diplopods, Spirostreptus and Polydesmus. The Scutigera 6 × 6mer hemocyanin is a trigonal antiprism assembled from six smaller trigonal antiprisms (1 × 6mers), thereby exhibiting D3 point group symmetry. It can be described as two staggered 3 × 6mers or three oblique 2 × 6mers. Topologically, the 6 × 6mer is subdivided into six subunit zones, thereby exhibiting a mantle (24 subunits) and a core (12 subunits). The six hexamers are linked by 21 bridges, subdivided into five types: two within each 3 × 6mer and three between both 3 × 6mers. The molecular models of the 6 × 6mer reveal intriguing amino acid appositions at these inter-hexamer interfaces. Besides opportunities for salt bridges, we found pairs of carboxylate residues for possible bridging via a Ca2+ or Mg2+ ion. Moreover, we detected histidine clusters, notably in Scutigera, allowing us to advance hypotheses as to how the hexamers are allosterically coupled in centipede hemocyanin and why they act more independently in diplopod hemocyanin.  相似文献
