MyoD基因对肉牛胴体性状影响的分析   总被引:11,自引:1,他引:10

用PCR技术克隆到MyoD基因的第二内含子, 采用PCR-SSCP方法研究了3个黄牛品种(鲁西牛、晋喃牛、秦川牛)及4个杂交肉牛(夏洛莱×鲁西牛、安格斯×鲁西牛、利木赞×鲁西牛、西门塔尔×鲁西牛)群体MyoD基因的多态性, 并分析了基因位点多态性与肉牛肉质性状的相关性。实验结果,在国内首次扩增出肉牛MyoD基因的第二内含子的全部序列, 共261 bp。用SSCP方法检测到MyoD基因内含子2有A和B两个等位基因。测序结果表明该座位的多态性是由于内含子二39 bp处C-T的突变和112 bp处C→G的突变造成的。等位基因B在中国地方品种的分布频率高于引进品种的杂交牛群体。c2检验的结果表明, 在该位点的除夏洛莱和安格斯杂交牛外, 其余五个群体(晋南、鲁西、秦川、西门塔尔杂交牛和利木赞杂交牛)均处于Hardy-Weinberg不平衡状态(P>0.05)。实验群体不同基因型与肉牛的宰前活重、胴体重、净肉重、高档肉重、眼肌面积等性状的影响差异极显著或显著(P<0.01或P<0.05), 并且AA型个体均高于AB型个体。  相似文献

本试验旨在研究贵州白山羊GFI1B基因第八外显子的多态性,及其与生长性状的相关性.通过PCR-RFLP技术对贵州白山羊外显子SNPs位点进行检测,利用一般线形模型分析其与生长性状的关联性.结果显示,供试群体中在外显子上检测到2个SNPs位点,即第八外显子263(G/T)和340(G/A).最小二乘法分析表明,G340A位点,CC型和DD型的体重、体长、胸深和胸宽对CD型达到差异及显著水平(p约0.01).本研究检测到的GFI1B基因第八外显子340(G/A)多态性位点可作为贵州白山羊生长性状的候选分子标记.  相似文献

文章采用单链构象多态性(PCR-SSCP)和DNA测序的方法检测了北极狐生长激素受体(Growth hormne receptor, GHR)基因的单核苷酸多态性(SNPs), 并针对该群体的特点建立合适的统计分析模型, 对GHR基因多态性与生长性状的相关性进行了分析。结果表明, 在北极狐GHR基因的外显子1和外显子5上发现了4个多态位点, 分别为5′UTR上的G3A和外显子1上的C99T突变, 外显子5上的T59C和G65A突变; GHR基因G3A和C99T多态性与母狐的体重性状显著相关(P<0.05), T59C和G65A多态性与公狐的体重性状显著相关(P<0.05), 与母狐的皮张长度性状极显著相关(P<0.01)。因此, 可以利用以上点突变对北极狐的体重及皮张长度性状进行标记辅助选择研究, 以达到快速选育出快大、优质的北极狐的目的。  相似文献

为分析山羊ADIPOQ基因的多态性,筛选出对山羊繁殖性状有显著影响的SNPs位点,本研究以黔北麻羊和贵州黑山羊为试验对象,构建池DNA,采用PCR产物直接测序法对2个品种山羊该基因的外显子1和3进行单核苷酸多态性检测,估算各SNPs等位基因频率,并利用在线软件预测不同基因型的m RNA二级结构。结果显示,4对引物扩增片段均存在多态性,共发现5个单碱基突变,分别位于内含子1中的C109G,外显子3中的A730G、G1055A、A1691T和A2244G。利用生物信息学软件对外显子3中的A1691T、A2244G进行分析,结果表明2个SNPs位点均导致编码的m RNA二级结构发生改变。表明ADIPOQ基因在黔北麻羊和贵州黑山羊群体中存在较高的遗传多样性,ADIPOQ位点有望丰富两个山羊品种繁殖性状的研究内容。  相似文献

研究旨在探讨生长激素释放激素基因(Growth hormone-releasing hormone,GHRH)对斑点叉尾(Ictalurus punctatus)生长性状的影响。采用DNA混池测序法筛选GHRH基因的单核苷酸多态性(Single nucleotide polymorphisms,SNPs)位点,使用SNa Pshot法将筛选到的SNPs多态性位点进行分型,并对这些位点进行连锁不平衡和单倍型分析。结果表明,在GHRH基因内含子区域共检测到4个SNPs位点,并成功地对3个位点进行了分型,3个位点间均不存在强连锁不平衡;3个SNPs位点在176尾斑点叉尾中形成了6种有效单倍型。关联分析表明SNP位点g.6301 GA的AA基因型的体质量显著性地高于AG和GG型(P0.05),比群体的平均体质量高14%。单倍型组合H1/H4和H1/H5个体的体质量和体长极显著性地高于其他单倍型组合(P0.01),体质量比群体平均体质量分别高30%和15%,体长比群体平均体长分别高7%和6%。研究为斑点叉尾生长性状分子标记辅助选育和QTL定位提供了参考依据。  相似文献

为研究中华绒螯蟹(Eriocheir sinensis)肌肉生长抑制素基因(myostatin, MSTN)的多态性及其与生长性状的相关性, 对中华绒螯蟹3个群体(育种群体、大赛群体、野生群体)共321个个体MSTN基因的多态性进行筛选, 发现该基因的第1外显子存在3个多态性SNP位点(S1: C714T; S2:G729A; S3:G753T), 均为处于Hardy-Weinberg平衡(P>0.05)的中、高度多态性位点。利用一般线性模型分析3个位点及其基因型组合与生长性状的相关性, 发现S1位点对中华绒螯蟹的体重和壳长等生长性状有显著影响(P≤0.05), 而其余2个位点与生长性状无显著关联性。结果表明S1位点的TT基因型对中华绒螯蟹的生长最为有利, 可作为分子标记辅助育种的候选标记。  相似文献

为探究GHSR基因多态性对中华鳖(Pelodiscus sinensis)生长相关性状的影响,采用直接测序法在GHSR基因5'侧翼和3'侧翼上筛选SNPs位点,共检测到5个单核苷酸多态性位点:A335T、G397T、A527G、A13482C和T13526A。随机选取同批繁殖的1冬龄200只中华鳖用直接测序法进行SNPs位点的分型,并分析与生长性状的相关性。检测结果显示,所有SNP位点均符合Hardy-Weinberg平衡状态(p>0.05)。方差分析显示,A336T位点的AT、TT基因型的体重、背甲长、背甲宽和裙边宽4项生长数据均显著高于AA基因型。A13482C位点的AC基因型的体重、背甲长、背甲宽和裙边宽4项数据均显著高于AA基因型(p<0.05)。研究表明,本实验在GHSR基因上获得的这些SNP位点可能影响着中华鳖的生长性状或与之紧密连锁,可为中华鳖分子辅助育种提供助力与参考。  相似文献

以五指山猪为试验材料,采用PCR-SSCP和测序相结合技术,对GH基因和IGFBP3基因进行多态性检测,并分析其与生长性状(体质量,体高,体长和胸围)的相关性。结果表明,GH基因第2外显子存在一处G→A转换,属沉默突变,该位点与生长性状关联分析显示,BB基因型的体重显著高于AA基因型和AB基因型;IGFBP3基因第2外显子存在一处G→C颠换,属沉默突变,该位点与生长性状关联分析显示,AA基因型的体高显著高于BB基因型;IGFBP3基因第4内含子存在一处碱基C缺失,该位点与生长性状关联分析显示,AA基因型体重显著高于AB基因型和BB基因型。研究将为五指山猪生长发育规律、系统选育及矮小机制等方面研究提供了遗传学依据。  相似文献

目的:探讨陕西汉族人群中LKB1基因位点rs741765(380CT)及rs6510599(459GA)单核苷酸多态性(SNPs)与2型糖尿病遗传易感性及相关临床代谢指标的关系。方法:采用等位基因特异性引物PCR(SASP-PCR)对2型糖尿病患者130例及健康对照组100例进行LKB1基因内含子6 rs741765(380CT)及内含子1 rs6510599(459GA)两个位点进行基因多态性筛查,并测序鉴定,分析其基因多态性位点与2型糖尿病临床代谢指标关系。结果:rs741765(380CT)基因突变情况:2型糖尿病患者TT基因型频率显著高于健康对照组(P=0.023);TT基因2型糖尿病组中糖化血红蛋白水平及低密度脂蛋白胆固醇水平在型中明显升高(P=0.030;P=0.002);健康对照组中,空腹血糖水平在TT基因型中明显升高(P=0.011)。rs6510599(459GA)基因突变情况:AA基因型频率在2型糖尿病组及健康对照组间无显著性差异(P0.05);该基因位点与临床指标亦无相关性(P0.05)。结论:陕西汉族人群中LKB1基因内含子6 rs741765(380CT)及内含子1 rs6510599(459GA)存在基因多态性。LKB1基因内含子6 rs741765(380CT)基因多态性与2型糖尿病的发病有相关性。LKB1基因内含子1 rs6510599(459GA)基因多态性与2型糖尿病的发病无相关性。  相似文献

Estrogen receptor (ERalpha) modifies the expression of genes involved in cell growth, proliferation and differentiation through binding to estrogen response elements (EREs) located in a number of gene promoters, so the ERalpha gene is considered as an important factor affecting reproductive endocrinology in Japanese flounder (Paralichthys olivaceus). In this study, twelve single nucleotide polymorphisms (SNPs) within eight CDS exons and 1 kb of 3'-UTR of the ERalpha gene were tested to association with four reproductive traits in a population of 119 Japanese flounder individuals with polymerase chain reaction-single stranded conformational polymorphism (PCR-SSCP). The association analysis of SNPs within Japanese flounder ERalpha gene with the reproductive traits was carried out using General Linear Model (GLM) estimation. Results indicated that two SNPs in the exon4 of ERalpha gene, P1 (A803G and C864T), were significantly associated with hepatosomatic index (HSI) (P<0.05) in female Japanese flounder. Other ten SNPs in 3'-UTR associated to serum 17beta-estradiol (E(2)) and HSI showed that P2 (A1982T) was significantly associated with E(2) (P<0.01) and P3 (A2149G, 2181TTACAAG2182 insertion or deletion, T2324G, A2359G and G2391A) was significantly associated with HSI (P<0.05) in female Japanese flounder. However, P2 (A1982T) and P4 (G2256T, T2294C, T2309G and A2333T) had significant effects on E(2) (P<0.05 and P<0.01, respectively) in male Japanese flounder. In addition, there were significant associations between diplotype D1 based on fourteen SNPs and reproductive traits. The genetic effects for HSI (female) or E(2) (male) of diplotype D1 were significantly higher than those of other eight diplotypes (P<0.05), respectively. Our findings implied that P1 of ERalpha gene affecting the reproductive traits could be a potential QTN (quantitative trait nucleotide) which would be useful genetic marker in the selection of some reproductive traits for its in Japanese flounder.  相似文献

肌肉生长抑制素基因(Myostatin,MSTN)属于转化生长因子β家族,其主要功能是负向调控肌肉的生长发育。采用PCR-SSCP技术对黄颡鱼MSTN基因进行单核苷酸多态性检测和分型,并与其生长性状进行关联分析。结果表明,在第一内含子部分检测到1个缺失位点和2个突变位点(T1003del、G1022A和T1063G),基因型分别为:AA、AB、CC、CD和DD;在第三外显子部分检测到1个突变位点(T132C),基因型分别为:EE和EF。关联性分析表明,AA基因型个体的全长、体长、体高、体厚、头长和体重显著大于CD和DD基因型(P<0.05),AA基因型雌性个体的全长、体长、体高、体厚、头长、尾柄高、尾柄厚和体重也显著大于DD基因型(P<0.05)。由此推断,AA基因型是影响雌性黄颡鱼生长性状的有利基因型,DD基因型是影响雌性黄颡鱼生长性状的不利基因型,可以尝试利用这两个位点对雌性黄颡鱼进行标记辅助选育。  相似文献

In this study, the polymorphisms of growth hormone (GH) gene 5' promoter region and intron 8, exons 4 and 10 of growth hormone receptor (GHR) gene were analyzed in Xinong Saanen goats (SG) and Boer goats (BG). Two alleles (A and B) and three genotypes (AA, AB and BB) were detected at P1 locus of GH gene, and two alleles (G and T) and two genotypes (GG and GT) were detected at P4 locus of GHR gene by PCR-SSCP analysis. In addition, two single nucleotide polymorphisms (SNPs)-A73C (P1 locus) and G114T (P4 locus), were identified by DNA sequencing. The frequencies of alleles A and B in the two goat breeds were 0.61-0.62, and 0.39-0.38, respectively, and the frequencies of alleles G and T in the two goat breeds were 0.82-0.86, and 0.18-0.14, respectively. The SNP loci were in Hardy-Weinberg disequilibrium in both goat breeds (P<0.05). Polymorphisms of GH and GHR genes were shown to be associated with growth traits in BG breed. AA and GG genotypes were associated with superior growth traits in 1-, 2- and 3-month old individuals. Hence, AA and GG genotypes are suggested to be a molecular marker for superior growth traits in BG breed.  相似文献

PCR-SSCP and DNA sequencing methods were applied to reveal three novel single nucleotide polymorphisms (SNPs) in exon 2 of the POU1F1 gene in 963 Chinese cattle belonging to eight breeds. Among them, a silent SNP (NM_174579:c.545G > A) detected by TaqI endonuclease is described. Frequencies of the POU1F1-G allele varied from 0.685 to 1.000. The association of TaqI polymorphism with growth traits was analyzed in 251 Nanyang cattle. No significant associations of the TaqI polymorphism with body weight and average daily gain for different growth periods (6, 12, 18, and 24 months old) were observed (P > 0.05), as well as for body sizes (P > 0.05).  相似文献

In this study, polymorphism in the bovine LEPR gene exon 4 was detected by PCR-SSCP and DNA sequencing methods in 653 individuals from five Chinese cattle breeds. Two haplotypes (M and N), three observed genotypes (MM, MN, and NN), and five single nucleotide polymorphisms (SNPs) (NC_007301:g.26767T>C, NC_007301:g.26805C>T, NC_007301:g.27050A>G, NC_007301:g.27063G>A, NC_007301:g.27079G>A) were detected. The frequencies of haplotypes M and N in the five breeds were 0.661-0.747 and 0.253-0.339, respectively. The SNP locus was in Hardy-Weinberg equilibrium in Nanyang, Jiaxian red, Angus, and Jinnan cattle (P > 0.05) and was in Hardy-Weinberg disequilibrium in Qinchuan cattle (P < 0.05). Polymorphism of the LEPR gene was shown to be associated with growth traits in the Nanyang breed. The SNP in the bovine LEPR gene had significant effects on body height, body length, body weight, heart girth, and average daily gain at 6 and 12 months old (P < 0.01 or P < 0.05). Therefore, these results suggest that the LEPR gene is a strong candidate gene that affects growth traits in cattle.  相似文献

Myostatin (MSTN), or growth and differentiation factor 8 (GDF8), is a member of the transforming growth factor (TGF)-β superfamily. This family functions as a negative regulator of skeletal muscle development and growth in mammals. Single-nucleotide polymorphisms in exon 1 of the Bian chicken myostatin gene were detected by polymerase chain reaction-restriction fragment length polymorphism. A mutation (c.234G>A) in exon 1 was found. Female Bian chickens of genotypes AA and GA had significantly higher body weights than those of genotype GG (P < 0.05 or P < 0.01) from 6 to 18 weeks of age. These results suggested that the mutation c.234G>A in exon 1 could be used as a genetic marker for Bian chicken growth traits.  相似文献
