Strongyle infection is an important issue in horse breeding. It impairs horse health and performance, with young horses being the most sensitive. Strongyle control has long relied on the systematic use of chemical treatments. However, expanding anthelmintic resistance among strongyles calls for alternative options. Mixed grazing is assumed to reduce strongyle load on the pasture as the result of a dilution effect. This has been shown in small ruminants grazing with cattle, but the putative benefits of co-grazing between horses and cattle have not yet been evaluated. Here, we conducted field surveys and face-to-face interviews on 44 farms from two contrasted saddle-horse production areas, Normandy and northern Massif Central, to compare equine strongyle management practices between specialized systems and mixed horse-cattle systems. Our goals were (i) to quantify breeders’ awareness of the putative benefits associated with the co-grazing of horses and cattle, (ii) to establish whether mixed farming was associated with different strongyle management strategies and (iii) to test whether strongyle egg excretion was reduced in horses grazed with beef cattle. Every breeder relied on systematic calendar treatments, and only 8 out of the 23 mixed breeders were aware that co-grazing of horses with cattle could be used as part of their strongyle control strategy. Management practices were similar across both systems in Normandy. In Massif Central, mixed breeders formed a distinct cluster from their specialized counterparts: deworming was less frequent and stocking density was higher in mixed farms, while specialized breeders seemed more willing to integrate herd and plot management into control strategies. Faecal egg counts measured in horses from Massif Central were significantly reduced when horses were grazed with cattle. This was the result of an increased reliance on macrocyclic lactones in mixed farms (P < 0.01) and a significant dilution effect (P < 0.01). When considering a subsample of horses treated with macrocyclic lactones only, young horses grazed with cattle had 50% fewer strongyle eggs excreted in their faeces than horses grazed in equine-only pastures (P < 0.01). This is the first evidence of the benefits of mixed grazing with cattle as an alternative to control strongyle infection in horses, although this promising alternative remains largely unknown by horse breeders.  相似文献

Horses are ubiquitously infected by a diversity of gastro-intestinal parasitic helminths. Of particular importance are nematodes of the family Strongylidae, which can significantly impact horse health and performance. However, knowledge about equine strongyles remains limited due to our inability to identify most species non-invasively using traditional morphological techniques. We developed a new internal transcribed spacer 2 (ITS2) DNA metabarcoding ‘nemabiome’ assay to characterise mixed strongyle infections in horses and assessed its performance by applying it to pools of infective larvae from fecal samples from an experimental herd in Kentucky, USA and two feral horse populations from Sable Island and Alberta, Canada. In addition to reporting the detection of 33 different species with high confidence, we illustrate the assay’s repeatability by comparing results generated from aliquots from the same fecal samples and from individual horses sampled repeatedly over multiple days or months. We also validate the quantitative potential of the assay by demonstrating that the proportion of amplicon reads assigned to different species scales linearly with the number of larvae present. This new tool significantly improves equine strongyle diagnostics, presenting opportunities for research on species-specific anthelmintic resistance and the causes and consequences of variation in mixed infections.  相似文献

Ponies reared with minimal or no exposure to anthelmintics were surveyed for intestinal helminths in order to estimate prevalence and intensity of parasite populations unaltered by frequent exposure to anthelmintics. Thirty-seven mixed breed ponies of varying ages were examined. Thirty-four species of nematodes and 2 species of cestodes were found. Twenty-four of the nematode species (including 1 new species) were in the subfamily Cyathostominae (small strongyles). Eighty-seven percent of the total burden of adult small strongyles in the large intestine was composed of 10 species. By comparing the results of the present survey with those of recent surveys of horses from herds which had been subjected to treatments with anthelmintics, the effect of prolonged usage of anthelmintic treatment on the prevalence of individual species possibly can be estimated. The general ranking of the 10 most common cyathostome species was similar to those described in recent surveys of horses, suggesting that anthelmintic pressure does not affect the prevalence of most cyathostome species. The lack of anthelmintic treatment appeared not to affect prevalence rates for Anoplocephala perfoliata and Anoplocephala magna when compared to other studies. Conversely, prevalence rates for Strongylus spp., Triodontophorus spp., Craterostomum acuticaudatum, Oxyuris equi, and Parascaris equorum were higher than those reported for these species in recent studies of horses.  相似文献

Gastrointestinal nematodes (GIN) infection can impair milk production (MP) in dairy cows. To investigate whether MP would be optimized by spring targeted-selective anthelmintic treatment in grazing cows, we assessed (1) the effect on MP of an anthelmintic treatment applied 1.5 to 2 months after turn-out, and (2) herd and individual indicators associated with the post-treatment MP response. A randomized controlled clinical trial was conducted in 13 dairy farms (578 cows) in western France in spring 2012. In each herd, lactating cows of the treatment group received fenbendazole orally, control cows remained untreated. Daily cow MP was recorded from 2 weeks before until 15 weeks after treatment. Individual serum pepsinogen and anti-Ostertagia antibody levels (expressed as ODR), faecal egg count and bulk tank milk (BTM) Ostertagia ODR were measured at treatment time. Anthelmintic treatment applied during the previous housing period was recorded for each cow. In each herd, information regarding heifers’ grazing and anthelmintic treatment history was collected to assess the Time of Effective Contact (TEC, in months) with GIN infective larvae before the first calving. The effect of treatment on weekly MP averages and its relationships with herd and individual indicators were studied using linear mixed models with two nested random effects (cow within herd). Unexpectedly, spring treatment had a significant detrimental effect on MP (-0.92 kg/cow/day on average). This negative MP response was particularly marked in high producing cows, in cows not treated during the previous housing period or with high pepsinogen levels, and in cows from herds with a high TEC or a high BTM ODR. This post-treatment decrease in MP may be associated with immuno-inflammatory mechanisms. Until further studies can assess whether this unexpected result can be generalized, non-persistent treatment of immunized adult dairy cows against GIN should not be recommended in early grazing season.  相似文献

Species composition and structure of the strongylid community was studies on helminthological material collected from 162 domestic horses from 11 regions of Ukraine by the in vivo method. Animals were treated with anthelmintic drug "Univerm" (0.2% aversectin C, Russia). Faecal samples (200 g each) were collected from every horse at 24, 36, 48 and 60 hours after treatment; all nematodes expelled (90.851 specimens) were collected and identified. Thirty-three strongylid species from 12 genera (8 species of subfamily Strongylinae and 25--of Cyathostominae) were found in domestic horses in Ukraine. Cyathostominae dominated in the strongylid community; they were found in 100 % horses and composed 98.21% of community. "Core" of the strongylid community was composed by 7 cyathostome species. Decreasing of proportion of Strongylinae in the community for last 40 years was registered; strongylines were found in 37.6% of horses and composed 1.25% of community. Maximal prevalence was 20.98% (Strongylus vulgaris). Bray-Curtis cluster analysis revealed high similarity of strongylid communities in horses from various regions of Ukraine. Difference in general structure of strongylid communities of horses from different horse-keeping conditions was established. Horses from farms with stable-paddock keeping conditions had bimodal strongylid community structure; while horses from stable-pasture keeping conditions possessed multimodal community structure.  相似文献

Considerable attention has been given by anthropologists, including ethnohistorians, to the impact of domesticated horse use on aboriginal societies in western North America. The spatial and temporal distribution of horses, as well as variation in herd size, has traditionally been explained within a cultural diffusionist or historical particularist paradigm. It is argued here that not only has such an approach failed to account for the distribution and abundance of horses, but it also has proven to be an unproductive research strategy. An ecologically based explanatory model is presented that accounts for 65% of the observed variability in aboriginal horse herd size(s). An explanation of variability in horse numbers and horse-to-person ratios is essential if anthropologists seek to better understand equestrian adaptations. Wild horses (Equus caballus) are a non‐native species occupying over 2800 km2 of the nationally significant Australian Alps National Parks. We estimated key demographic parameters (fecundity, adult and juvenile survival and annual finite population growth rate) over 3 years and related these to horse body condition and available food for three populations under natural conditions, and found a trend consistent with food limitation. The populations were independent, with different site characteristics and occupied areas, identified by land managers, as areas of concern about possible conservation impacts. Annual fecundity and juvenile survival varied across sites averaging between 0.21 and 0.31 female young per adult female, and 0.83 and 0.90 per annum, respectively, and annual adult survival was consistent across sites averaging 0.91 per annum. One population was increasing (λ = 1.09 year?1; 95% CI 1.04–1.14) and two populations were stable (λ ~ 1.0 year?1). Mean body condition of horses was positively correlated with mean pasture biomass rank. Across the three populations, fecundity, recruitment, body condition and annual finite population growth rate were lowest when mean pasture biomass rank was lowest and conversely highest when pasture rank was highest. We conclude that food limitation appears to be operating across these three sites. We used our results to assess the sensitivity of annual finite rate of increase (λ) to changes in key demographic parameters and found that λ was most sensitive to a change in adult survival, with the second most sensitive parameter being fecundity. Thus, if the aim of management is to reduce the size of the wild horse population then targeting adult survival is most important, followed by fecundity. Finally, we estimated the linear, negative, numerical response for wild horses between annual λ and horses per unit pasture biomass.  相似文献

Background

In a stable of eight horses in Northern Iceland, six horses presented with clinical signs, such as ataxia and reduced appetite, leading to euthanasia of one severely affected horse. Serological investigations revealed no evidence of active equine herpes virus type 1 infection, a common source of central nervous system disease in horses, nor equine arteritis virus and West Nile virus. Another neurotropic virus, Borna disease virus, was therefore included in the differential diagnosis list.

Findings

Serological investigations revealed antibodies against Borna disease virus in four of five horses with neurological signs in the affected stable. One horse without clinical signs was seronegative. Four clinically healthy horses in the stable that arrived and were sampled one year after the outbreak were found seronegative, whereas one of four investigated healthy horses in an unaffected stable was seropositive.

Conclusions

This report contains the first evidence of antibodies to Borna disease virus in Iceland. Whether Borna disease virus was the cause of the neurological signs could however not be confirmed by pathology or molecular detection of the virus. As Iceland has very restricted legislation regarding animal imports, the questions of how this virus has entered the country and to what extent markers of Bornavirus infection can be found in humans and animals in Iceland remain to be answered.

Virtually all horses are infected with helminth parasites. For some decades, the control of parasites of Swedish horses has been based on routine treatments with anthelmintics, often several times per year. Since anthelmintic resistance is becoming an increasing problem it is essential to develop more sustainable control strategies, which are adapted to different types of horse management. Corroborative results from various horse studies led to the conclusion that management factors might influence horse social organisation. In a free-living range-restricted population of 130 horses in the Camargue, a “young herd” whose formation had not previously been recorded, was observed for a period of one year in order to study the effects of still another management system. The uneven distribution of positive and agonistic interactions resulted in a social structure. This structure was stable for 9 months but changed afterwards. The results are compared with those from other horse studies and three variables, i. e. management, population composition and individual differences are developed into a preliminary model from which to predict what factorial combinations lead to what formations.  相似文献

A computer model was developed to simulate Trichostrongylus colubriformis populations, their level of resistance to the common anthelmintics, host mortalities and acquired immunity. Predictions were based on sheep management practices such as lambing, weaning, sheep/paddock rotation, anthelmintic treatment, the use of controlled release devices (capsules) for anthelmintic delivery and daily meteorological records to determine the development and survival of infective larvae (L3) on pasture. Evolution of drug resistance was determined by a simple genetic system which allowed for up to three genes, each with two alleles, to give a maximum of 27 genotypes associated with one drug or three genotypes for each of three drugs. The model was validated against egg counts, L3 counts on pasture and host mortalities observed in a grazing trial, however, aspects of the model such as the development of drug resistance and use of the model in a variety of climatic zones have yet to be tested against field observations. The model was used to examine the impact of grazing management and capsule use on anthelmintic resistance and sheep production over 20 years using historical weather data. Predictions indicated that grazing management can play a dominant role in parasite control and that capsule use will reduce sheep mortalities and production losses, and in some circumstances will not cause a substantial increase in anthelmintic resistance for up to 5 years.  相似文献

The effect of the genetic and biological factors on resistance are generally beyond the control of the operator. However, an understanding of these factors can enable the “resistance risk” (Georghiou, 1983), to be assessed when designing strategies to aid in the control of resistance. The important operator controlled factors relate to the frequency of anthelmintic usage, its efficiency and relation to previously used compounds and integration with stock management.The small post-treatment parasitic sub-population which remains after anthelmintic treatment is likely to have a higher resistance gene frequency compared to the larger pre-treatment parasitic and the freeliving sub-population on pasture. Where treatment frequencies approach the pre-patent period for nematodes, only resistant worms will persist. Likewise if stock are moved after treatment to a pasture free of nematode larvae, subsequent generations will result from the resistant worms carried over, which once again will lead to a resistant population. However, in either of these situations there would initially be a substantial reduction in the size of the parasite population. The subsequent aim is to prevent the increase in the size of the resistant population to the level where production losses occur.The complexities of the association between resistance, anthelmintic usage and stock management indicate the need to understand not just the general factors that effect resistance but also the ecological factors pertinent to a particular region and grazing enterprise. Difficulties will be faced by extension workers who will need to acquire the relevant knowledge to integrate anthelmintic treatment with management strategies that are acceptable to farmers. Some encouragement for the success of this comes from the rapid acceptance of the “Wormkill” drenching program introduced in New South Wales (Dash et al., 1985).  相似文献

Strongylus vulgaris is an important strongyle nematode with high pathogenic potential infecting horses world-wide. Several decades of intensive anthelmintic use has virtually eliminated clinical disease caused by S. vulgaris, but has also caused high levels of anthelmintic resistance in equine small strongyle (cyathostomin) nematodes. Recommendations aimed at limiting the development of anthelmintic resistance by reducing treatment intensity raises a simultaneous demand for reliable and accurate diagnostic tools for detecting important parasitic pathogens. Presently, the only means available to differentiate among strongyle species in a faecal sample is by identifying individual L3 larvae following a two week coproculture procedure. The aim of the present study is to overcome this diagnostic obstacle by developing a fluorescence-based quantitative PCR assay capable of identifying S. vulgaris eggs in faecal samples from horses. Species-specific primers and a TaqMan probe were designed by alignment of published ribosomal DNA sequences of the second internal transcribed spacer of cyathostomin and Strongylus spp. nematodes. The assay was tested for specificity and optimized using genomic DNA extracted from identified male worms of Strongylus and cyathostomin species. In addition, eggs were collected from adult female worms and used to evaluate the quantitative potential of the assay. Statistically significant linear relationships were found between egg numbers and cycle of threshold (Ct) values. PCR results were unaffected by the presence of cyathostomin DNA in the sample and there was no indication of PCR inhibition by faecal sources. A field evaluation on faecal samples obtained from four Danish horse farms revealed a good agreement with the traditional larval culture (kappa-value=0.78), but with a significantly higher performance of the PCR assay. An association between Ct values and S. vulgaris larval counts was statistically significant. The present assay can reliably and semi-quantitatively detect minute quantities of S. vulgaris eggs in faecal samples.  相似文献

Summary Spores of V-A mycorrhizal endophytes were extracted and counted from three different soils under three different rotations; continuous wheat, annual pasture and an alternating crop: pasture rotation.High V-A spore abundance in the autumn fell at the onset of plant growth. Spore abundance remained low under wheat but recovered under pasture regimes in the spring, coincidently with host maturation in all except one of the experiments sampled.Weak positive correlations were found between V-A spore abundance and components of pasture biomass while wheat biomass was negatively correlated with V-A spore numbers.  相似文献

In our continuing efforts to document genetic diversity in Przewalski's horses and relatedness with domestic horses, we report genetic variation at 22 loci of blood group and protein polymorphisms and 29 loci of DNA (microsatellite) polymorphisms. The loci have been assigned by linkage or synteny mapping to 20 autosomes and the X chromosome of the domestic horse (plus four loci unassigned to a chromosome). With cumulative data from tests of 568 Przewalski's horses using blood, hair or tooth samples, no species-defining markers were identified, however a few markers were present in the wild species but not in domestic horses. Inheritance patterns and linkage relationships reported in domestic horses appeared to be conserved in Przewalski's horses. A derived type for the last wild caught mare 231 Orlitza III provided evidence for markers apparently not found in (or not currently available by descent from) the other species founders that were captured at the end of the nineteenth century. This information has been critical to the development of parentage analyses in the studbook population of Przewalski's horses at Askania Nova, at one time the largest herd of captive animals and the source of stock for reintroduction efforts. Some horses in the study showed genetic incompatibilities with their sire or dam, contradicting published studbook information. In many cases alternative parentage could be assigned from living animals. To assist in identification of correct parentage, DNA marker types for deceased horses were established from archived materials (teeth) or derived from offspring. Genetic markers were present in pedigreed animals whose origin could not be accounted for from founders. Genetic distance analysis of erythrocyte protein, electrophoretic and microsatellite markers in Przewlaski's horses and ten breeds of domestic horse place the Przewalski's horse as an outgroup to domestic horses, introgression events from domestic horses not withstanding.  相似文献

This study investigated the occurrence of wounds in Nigerian horses. The study population was 1,621 horses sold at the Obollo Afor horse lairage in Enugu State, Nigeria, during a 6-month period: 3 months of dry season and 3 months of rainy season (February–April and June–August 2012). A total of 207 horses were systematically sampled and subjected to a comprehensive physical examination. Those with wounds were marked, recorded, and clinically examined. Of the 207 horses sampled, 21 (10.1%) had wounds. The body distribution of the wounds was 9.5% head, 9.5% forelimbs, 19.1% hind limbs, 4.8% tail, 14.3% flank, 9.5% loin, 19.1% hip, 9.5% barrel, and 4.8% croup. The occurrence of the wounds was not significantly associated with sex or season, but the occurrence in adults was significantly (p < .05) higher than that in the young and aged horses. It was concluded that the occurrence of wounds is relatively high (10.1%), and mainly the hind limbs, hip, and flank of adult horses are affected. It was recommended that horse guardians and handlers should be properly educated on the care of horses.  相似文献

Recurrent airway obstruction (RAO) is characterized by neutrophilic airway inflammation and obstruction, and stabling of susceptible horses triggers acute disease exacerbations. Stable dust is rich in endotoxin, which is recognized by Toll-like receptor (TLR) 4. In human bronchial epithelium, TLR4 stimulation leads to elevation of interleukin (IL)-8 mRNA expression. The zinc finger protein A20 negatively regulates this pathway. We hypothesized that TLR4 and IL-8 mRNA and neutrophil numbers are elevated and that A20 mRNA is not increased in RAOs during stabling compared with controls and with RAOs on pasture. We measured the maximal change in pleural pressure (DeltaPpl(max)), determined inflammatory cell counts in bronchoalveolar lavage fluid (BAL), and quantified TLR4, IL-8, and A20 mRNA in bronchial epithelium by quantitative RT-PCR. We studied six horse pairs, each pair consisting of one RAO and one control horse. Each pair was studied when the RAO-affected horse had airway obstruction induced by stabling and after 7, 14, and 28 days on pasture. Stabling increased BAL neutrophils, DeltaPpl(max), and TLR4 (4.14-fold change) significantly in RAOs compared with controls and with RAOs on pasture. TLR4 correlated with IL-8 (R2 = 0.75). Whereas stabling increased IL-8 in all horses, A20 was unaffected. IL-8 was positively correlated with BAL neutrophils (R2 = 0.43) and negatively with A20 (R2 = 0.44) only in RAO-affected horses. Elevated TLR4 expression and lack of A20 upregulation in bronchial epithelial cells from RAO-affected horses may contribute to elevated IL-8 production, leading to exaggerated neutrophilic airway inflammation in response to inhalation of stable dust.  相似文献

Feral horses on Assateague Island, Maryland, were observed in June and August 2000 to determine what behavioral and ecological factors affect the intensity of biting fly harassment and whether habitat use by horses was influenced by biting flies. Fly counts and frequencies of comfort movements (i.e., movements designed to dislodge insects) were recorded during focal animal samples, as well as data on sex, group size, habitat type, temperature, humidity, wind speed, and behavior. Seasonal habitat use patterns were assessed using 7 years of monthly census data on the horses. The number of biting flies on the horse was affected by horse sex, habitat, temperature, and group size. The number of comfort movements a horse showed was affected by habitat, temperature, wind speed, group size, and number of horses within one body length of the focal. The number of comfort movements made by a horse was found to be highly correlated with fly numbers. Though marshes were used most throughout the year, the pattern of use of dune, scrub, and human-altered habitats reflects a pattern of biting fly avoidance and refuge-seeking by the horses.  相似文献

Recent studies have found that copy number variations (CNVs) are widespread in human and animal genomes. CNVs are a significant source of genetic variation, and have been shown to be associated with phenotypic diversity. However, the effect of CNVs on genetic variation in horses is not well understood. In the present study, CNVs in 6 different breeds of mare horses, Mongolia horse, Abaga horse, Hequ horse and Kazakh horse (all plateau breeds) and Debao pony and Thoroughbred, were determined using aCGH. In total, seven hundred CNVs were identified ranging in size from 6.1 Kb to 0.57 Mb across all autosomes, with an average size of 43.08 Kb and a median size of 15.11 Kb. By merging overlapping CNVs, we found a total of three hundred and fifty-three CNV regions (CNVRs). The length of the CNVRs ranged from 6.1 Kb to 1.45 Mb with average and median sizes of 38.49 Kb and 13.1 Kb. Collectively, 13.59 Mb of copy number variation was identified among the horses investigated and accounted for approximately 0.61% of the horse genome sequence. Five hundred and eighteen annotated genes were affected by CNVs, which corresponded to about 2.26% of all horse genes. Through the gene ontology (GO), genetic pathway analysis and comparison of CNV genes among different breeds, we found evidence that CNVs involving 7 genes may be related to the adaptation to severe environment of these plateau horses. This study is the first report of copy number variations in Chinese horses, which indicates that CNVs are ubiquitous in the horse genome and influence many biological processes of the horse. These results will be helpful not only in mapping the horse whole-genome CNVs, but also to further research for the adaption to the high altitude severe environment for plateau horses.  相似文献

Protected areas can be impacted by the presence and proliferation of feral species. Effective management of feral species requires reliable tools to monitor their population size and ecological impacts. Here, we used drone-based image analysis to assess evidence of feral horses and horse-specific ecological impacts on alpine riparian habitat. Valleys with low (0), medium (1–16) and high (>16) horse abundances were chosen for drone imagery analysis based on independent aerial counts of horses. Data collection trips were carried out pre- and post-2019/2020 wildfires, which unexpectedly burnt valleys with low horse presence. Drone-based RGB orthomosaic imagery was sufficient to identify seven indicators of horse presence and determine the severity of feral horse impacts. Despite the impact of fire, drone-derived classifications were able to accurately detect a gradient of horse impacts, showing a significant difference in indicators from low presence valleys compared with medium and high presence valleys, which did not differ significantly from each other. The significance of differences between valleys reveals that regions routinely inhabited by feral horses will display significant environmental impacts. Our results clearly indicated significant differences between valleys with low horse presence compared with either medium or high horse presence regions (0.01 for differences between both low and medium and low and high horse presence). This was evident both before and after the 2019/2020 fires, suggesting that wildfires did not significantly impact horse populations or distribution in the sampled region. Overall, it was evident that feral horses have a clear and definable impact on alpine riparian vegetation, and drone surveying can be used to routinely monitor potential spread and the outcome of management actions.  相似文献
