Reactive oxygen species (ROS) are highly reactive oxygen‐containing molecules associated with aging and a broad spectrum of pathologies. We have previously shown that transgenic expression of the antioxidant enzyme catalase targeted to the mitochondria (mCAT) in mice reduces ROS, attenuates age‐related disease, and increases lifespan. However, it has been increasingly recognized that ROS also has beneficial roles in signaling, hormesis, stress response, and immunity. We therefore hypothesized that mCAT might be beneficial only when ROS approaches pathological levels in older age and might not be advantageous at a younger age when basal ROS is low. We analyzed abundance and turnover of the global proteome in hearts and livers of young (4 month) and old (20 month) mCAT and wild‐type (WT) mice. In old hearts and livers of WT mice, protein half‐lives were reduced compared to young, while in mCAT mice the reverse was observed; the longest half‐lives were seen in old mCAT mice and the shortest in young mCAT. Protein abundance of old mCAT hearts recapitulated a more youthful proteomic expression profile (P‐value < 0.01). However, young mCAT mice partially phenocopied the older wild‐type proteome (P‐value < 0.01). Age strongly interacts with mCAT, consistent with antagonistic pleiotropy in the reverse of the typical direction. These findings underscore the contrasting roles of ROS in young vs. old mice and indicate the need for better understanding of the interaction between dose and age in assessing the efficacy of therapeutic interventions in aging, including mitochondrial antioxidants.  相似文献

The Society for Ecological Restoration's 2016 (SER) “International Standards for the Practice of Ecological Restoration” is a living document intended to guide restoration projects “anywhere in the world.” Given its intended global scope and in hopes of informing future editions, this document is critically assessed in light of the role people have played in ecosystems around the world. We argue that the Standards has an underlying nature–culture dichotomization that limits its applicability; in qualifying what it calls “cultural ecosystems” for rehabilitation, rather than restoration, the Standards privileges colonial visions of ecological restoration. We also discuss the Standards' representation of the ecological impacts and practices of indigenous groups. Whereas the Standards claims that preindustrial cultural ecosystems exist in states similar to unmodified areas, many historians, anthropologists, and paleoecologists would point out that preindustrial people sometimes had massive environmental impacts through agriculture, hydrological engineering, over‐hunting, living in dense urban environments, transporting species, burning on a scale capable of changing the climate, and other practices. Furthermore, the Standards does not discuss how the cultural goals of indigenous groups fit into the overall picture of ecological restoration. Future drafts of the Standards should more accurately frame the diverse roles people play in nature, and create global standards that account for the validity of cultural goals for ecological restoration.  相似文献

The temperature dependence for Na(+)-K(+)-ATPase has been examined in the proximal-distal axis of the intestine of gilthead seabream (Sparus aurata), i.e. pyloric caeca (PC), anterior intestine (AI) and posterior intestine (PI). Data derived from the Arrhenius plots showed differences in terms of temperature discontinuity points ( Td) (13.29 degrees C, 16.39 degrees C and 17.48 degrees C for PC, AI and PI, respectively) and activation energy ratios (Ea(2)/Ea(1)) obtained at both sides of Td (2.38, 1.98 and 1.78, for PC, AI and PI, respectively). The analyses of polar lipids showed differences in the levels of certain fatty acids among intestinal regions. The content of each fatty acid and different fatty acid ratios were correlated with the corresponding Td and Ea(2)/Ea(1) values. Regression analyses revealed the existence of strong negative correlations between docosahexaenoic acid (22:6n-3, DHA) or the DHA/monoenes ratio and Td. No obvious relationships were observed for other polyunsaturated fatty acids (PUFA) nor saturated fatty acids. The results obtained in the present study indicate that the heterogeneous values of Td displayed by the Na(+)-K(+)-ATPase along the intestinal tract could be related to a modulatory role of certain fatty acid within the lipid microenvironment of the enzyme.  相似文献

A real-time polymerase chain reaction (PCR) was developed for the rapid detection and identification of Phaeoacremonium species, the fungi associated with severe diseases in grapevines. A degenerate primer pair (F2bt–R1bt) with homology to the β-tubulin gene was designed to be used in the amplification of 11 species of Phaeoacremonium. Four species-specific probes labelled with three different fluorescent dyes were designed to be used with the degenerate primers in a real-time PCR for the identification of Phaeoacremonium aleophilum, P. parasiticum, P. viticola and P. mortoniae. Combinations of two probes in a duplex real-time PCR allowed to detect and identify a mixture of Phaeoacremonium species and cross-amplifications were not detected. This method was applied to detect Phaeoacremonium species in eight wood fragments from grapevine plants naturally infected, and results were compared with those obtained with nested PCR and culturing on growth media. Real-time PCR detected Phaeoacremonium in 100% of the analysed fragments, whereas nested PCR did only in the 62% of them and requiring subsequent restriction fragment-length polymorphism analysis to identify the species. This method is a sensitive tool to detect and identify Phaeoacremonium species in infected grapevine wood. Real-time PCR assay defined here can be used in a plant nursery program to identify pathogen-free plants in order to manage Petri disease of grapevines.  相似文献

Cocoa and coffee are among the most valuable tropical crops, with much of their production in areas of high biodiversity. Although this could suggest a conflict between agricultural expansion and biodiversity conservation, these crops are normally grown in shade plantations—a more biodiversity‐friendly agroforestry system. Using São Tomé Island as a case study, we examined if shade plantation can benefit biodiversity by protecting extinction‐prone island endemic species. We found that shade plantations held rich assemblages, even in comparison with forest: we estimated 30 bird and 74 tree species occurring in plantations compared to 24 bird and 108 tree species in forests. However, the structure of the assemblages was significantly different between ecosystems, with an average dissimilarity of 33.8 percent and 87.9 percent for birds and trees, respectively. Shade plantations had consistently less endemic species than the forest; we estimated a drop from 17 to 13 in birds and from 17 to 3 in trees. We also found that despite marked differences in the response of bird species to the vegetation characteristics of shade plantations, there was a strong positive link between aboveground tree biomass and endemic species richness and abundance. These results show that shade plantations hold assemblages impoverished in endemic species, but that management can improve their value for the conservation of these species. Finally, we suggest that biodiversity‐friendly certification and carbon markets are used to implement and guarantee the long‐term economical sustainability of practices that favor the endemic species in São Tomé's shade plantations.  相似文献

The content and organization of mental health care have been heavily influenced by the view that mental difficulties come as diagnosable disorders that can be treated by specialist practitioners who apply evidence‐based practice (EBP) guidelines of symptom reduction at the group level. However, the EBP symptom‐reduction model is under pressure, as it may be disconnected from what patients need, ignores evidence of the trans‐syndromal nature of mental difficulties, overestimates the contribution of the technical aspects of treatment compared to the relational and ritual components of care, and underestimates the lack of EBP group‐to‐individual generalizability. A growing body of knowledge indicates that mental illnesses are seldom “cured” and are better framed as vulnerabilities. Important gains in well‐being can be achieved when individuals learn to live with mental vulnerabilities through a slow process of strengthening resilience in the social and existential domains. In this paper, we examine what a mental health service would look like if the above factors were taken into account. The mental health service of the 21st century may be best conceived of as a small‐scale healing community fostering connectedness and strengthening resilience in learning to live with mental vulnerability, complemented by a limited number of regional facilities. Peer support, organized at the level of a recovery college, may form the backbone of the community. Treatments should be aimed at trans‐syndromal symptom reduction, tailored to serve the higher‐order process of existential recovery and social participation, and applied by professionals who have been trained to collaborate, embrace idiography and maximize effects mediated by therapeutic relationship and the healing effects of ritualized care interactions. Finally, integration with a public mental health system of e‐communities providing information, peer and citizen support and a range of user‐rated self‐management tools may help bridge the gap between the high prevalence of common mental disorder and the relatively low capacity of any mental health service.  相似文献

Through simulation, Whitlock showed that when all the alternatives have the same effect size, the weighted z-test is superior to both unweighted z-test and Fisher's method when combining P-values from independent studies. Trichocorixa verticalis is a North American water bug (Heteroptera: Corixidae) that occurs in brackish and saline aquatic systems. Recently, it has been found invading three continents including Europe. Its invasive success has been attributed to the capacity of tolerating hypersalinity. We compared both the realized and standardized salinity niche of invasive T. verticalis and native Corixidae to verify if T. verticalis may fill in an unoccupied niche. We first established the field distribution of T. verticalis and native Corixidae along a salinity gradient. Second, we experimentally tested the salinity tolerance of T. verticalis and three common native Corixidae species. Of the seven Corixidae species found in the field study, three were positively related to the salinity gradient: S. selecta, S. stagnalis, and T. verticalis. T. verticalis showed the highest salinity optimum, however, after correcting for environmental background variation, salinity optima differed little among the three halophilic species. In the salinity tolerance experiment, S. selecta outperformed T. verticalis, which performed as well as S. stagnalis. Based on our experimental results, we cannot support the hypothesis that T. verticalis’ invasion is mediated by a high salinity tolerance that allows this species to colonize habitats unoccupied by native Corixidae. Although these findings contrast with the field patterns in which T. verticalis showed the highest niche optimum and tolerance, these patterns may have been partly due to other environmental factors, particularly anthropogenic disturbance. Our comparative results are for adults only, and it remains possible that relative salinity tolerance patterns for juveniles differ from that for adults, which may add to the observed field pattern.  相似文献

So far, it has not been possible to identify a general sequence motif for O-glycosylation in bacteria. In this issue, Charbonneau et al. (2012) demonstrate why O-glycosylation is mediated by a 13-residue strand-loop-strand motif which is part of a 19-residue imperfect repeat in the passenger domain of bacterial autotransporters. This motif provides a convenient 'glycosylation cassette' and raises intriguing questions about the structural regulation of the glycosylation pathway.  相似文献

Nested clade phylogeographic analysis (NCPA) has become a popular method for reconstructing the history of populations across species ranges. Ever since its invention in 1995, criticisms have been formulated, but the method, which has been regularly updated, continues to attract investigators. Molecular Ecology has published a large fraction of the literature on the topic — both pro and con. A recent study by Panchal and Beaumont (2007) finally allows a precise evaluation of the method by developing software that automates the somewhat complicated NCPA procedure. Using simulations of random-mating populations, Panchal and Beaumont find a high frequency of false-positives with their automated NCPA procedure (over 75%). These findings, which echo and amplify earlier warnings, appear serious enough to suggest to researchers to await further evaluation of the method. Although no other all-encompassing method such as the NCAP currently exists to evaluate phylogeographic data sets, researchers have many alternative methods to test ever more refined hypotheses.  相似文献

Sampling for rare events, such as a new weed incursion, is not easy. At most of the sample points the species of interest is absent and only occasionally the species is recorded. Very often surveillance and monitoring for rare events (e.g. new weed incursions) is done using local knowledge and a statistical sampling design is not used. The stated reason for this approach is usually because the biodiversity managers knew where to look and didn’t need statistics. Adaptive, unequal probability survey designs can be used in these situations, ensuring both sample effort is focused on locations where there is a high likelihood of a weed being present. Time in the field is spent within locations where weeds are present and minimal time spent where weeds are absent. Any relevant information on where weeds are likely to be found (e.g. local knowledge and expertise) can be used to target survey effort in unequal probability survey designs. The advantage of an adaptive, unequal probability survey design is that not only can field effort be focused on areas where the weeds are thought to be. In addition, important weed parameters can be estimated and reported along with estimates of uncertainty. Weed parameters include the proportion of the total area that weeds are present, the diversity of weed species, the total abundance of weeds, and the total area covered by weeds. With reliable and consistent estimates of these weed parameters (e.g. weed cover or abundance) the efficacy of weed management can be tracked. Over time, with regular reporting of weed cover or abundance, the success (or otherwise) of weed management strategies can be measured.  相似文献

Computational prediction of stabilizing mutations into monomeric proteins has become an almost ordinary task. Yet, computational stabilization of protein–protein complexes remains a challenge. Design of protein–protein interactions (PPIs) is impeded by the absence of an energy function that could reliably reproduce all favorable interactions between the binding partners. In this work, we present three energy functions: one function that was trained on monomeric proteins, while the other two were optimized by different techniques to predict side-chain conformations in a dataset of PPIs. The performances of these energy functions are evaluated in three different tasks related to design of PPIs: predicting side-chain conformations in PPIs, recovering native binding-interface sequences, and predicting changes in free energy of binding due to mutations. Our findings show that both functions optimized on side-chain repacking in PPIs are more suitable for PPI design compared to the function trained on monomeric proteins. Yet, no function performs best at all three tasks. Comparison of the three energy functions and their performances revealed that (1) burial of polar atoms should not be penalized significantly in PPI design as in single-protein design and (2) contribution of electrostatic interactions should be increased several-fold when switching from single-protein to PPI design. In addition, the use of a softer van der Waals potential is beneficial in cases when backbone flexibility is important. All things considered, we define an energy function that captures most of the nuances of the binding energetics and hence, should be used in future for design of PPIs.  相似文献

The β-clamp protein and the γ clamp loader complex are essential components of bacterial DNA replication machinery. The β-clamp is a ring-shaped homodimer that encircles DNA and increases the efficiency of replication by providing a binding platform for DNA polymerases and other replication-related proteins. The β-clamp is loaded onto DNA by the five-subunit γ clamp loader complex in a multi-step ATP-dependent process. The initial steps of this process involve the cooperative binding of the β-clamp by the five subunits of ATP-bound clamp loader, which induces or traps an open conformation of the clamp. Remarkably, the δ subunit of the E. coli clamp loader, or even its 140 residue N-terminal domain (called mini-δ), alone can shift conformational equilibrium of the β-clamp towards the open state. Here we report nearly complete backbone and side-chain 1H, 13C and 15N NMR resonance assignments of mini-δ that will facilitate NMR studies of the mechanisms of β-clamp opening and its loading on DNA by the clamp loader.  相似文献
