Old yellow enzymes (OYEs, EC 1.6.99.1) are flavin-dependent oxidoreductases that catalyze the stereoselective trans-hydrogenation of the double bond, representing a promising alternative to metal-based catalysis. Bioconversion of ketoisophorone (KIP) by 28 non-conventional yeasts belonging to 16 different species was investigated. Growing cells of most of the strains reduced KIP via OYE and showed high stereoselectivity, producing R-levodione as major product. Competition by carbonyl reductase (CR) activity was observed in several strains. The best performing yeasts belong to Candida castellii, Kazachstania spencerorum and Kluyveromyces marxianus exhibited yields of levodione ≥77% up to 95% e.e., and. Candida freyschussii, the sole strain lacking the OYE gene, reduced KIP only to unsaturated alcohols via CR. Nine unedited OYE genes were cloned, sequenced, and heterologously expressed in Saccharomyces cerevisiae BY4741ΔOye2, a mutant that showed negligible OYE and CR activities. Compared with the corresponding wild-type yeasts, growing cells of the recombinant strains bioconverted KIP with improved yields of OYE products, minor competition by CR activity, and lower enantioselectivity. In particular, resting cells of recombinant S. cerevisae presented the best performance in KIP bioconversion. Based on the results herein reported, selected strains of non-conventional yeasts and novel OYE genes can be profitably used as innovative biocatalysts in asymmetric reductions. 相似文献

Kittiwake growth rates and breeding success are examined in relation to survival between fledging and breeding and to adult survival rates. High chick growth rates lead to increased survival after fledging. Broods of three (the maximum brood size) did not suffer lower fledging success than broods of two and clutches of three fledged appreciably more chicks per pair than did clutches of two or one. On average, the a- and b -chicks in broods of three grew at a faster rate and had a higher survival before breeding than those from smaller broods. Chicks from broods of two with experienced female parents grew at a faster rate than those of inexperienced female parents. Female parents which laid three egg clutches had a higher survival rate than those which laid clutches of two or one. We contend that three egg clutches were laid by higher quality individuals. We believe that clutch size indicates the condition of the Kittiwakes forming the pair. This condition probably has a genetical component, but is modified by environmental factors. 相似文献

The analysis of a cerebro-vascular insult hospitalized cases in the Clinical Hospital Mostar as a retrospective epidemiological study was done in the Clinical Hospital Mostar for the period from 1999 to 2003. The major source of data was medical documentation of this hospital (an institutional register), the only hospital for the treatments of 457,491 inhabitants who gravitate by a health insurance for the treatment in this hospital. The study included a total of 1,555 cerebro-vascular insult cases treated in the Clinical Hospital Mostar Among them 727 (46.8%) were male patients, while 828 (53.2%) cases were female. The majority of the cases were above 50 years of life. Majority of treated female patients were older than 61 (45.6% of all cases), as well as among male patients (31.3%). The least number of cases was under 41 years in both groups (1.2%). Prevalence of risk factors was 2,035 cases (74%). During the same period risk factors research for entire Federation of Bosnia and Herzegovina (FBiH) was performed on the sample of 2,750 national insurance holders, out of which 852 gravitate for treatment in CB Mostar. Background
Patients with chronic fatigue syndrome (CFS) often complain of persistent fatigue even after conventional therapies such as pharmacotherapy, cognitive behavioral therapy, or graded exercise therapy. The aim of this study was to investigate in a randomized, controlled trial the feasibility and efficacy of isometric yoga in patients with CFS who are resistant to conventional treatments.Methods
This trial enrolled 30 patients with CFS who did not have satisfactory improvement after receiving conventional therapy for at least six months. They were randomly divided into two groups and were treated with either conventional pharmacotherapy (control group, n?=?15) or conventional therapy together with isometric yoga practice that consisted of biweekly, 20-minute sessions with a yoga instructor and daily in-home sessions (yoga group, n?=?15) for approximately two months. The short-term effect of isometric yoga on fatigue was assessed by administration of the Profile of Mood Status (POMS) questionnaire immediately before and after the final 20-minute session with the instructor. The long-term effect of isometric yoga on fatigue was assessed by administration of the Chalder’s Fatigue Scale (FS) questionnaire to both groups before and after the intervention. Adverse events and changes in subjective symptoms were recorded for subjects in the yoga group.Results
All subjects completed the intervention. The mean POMS fatigue score decreased significantly (from 21.9?±?7.7 to 13.8?±?6.7, P?<?0.001) after a yoga session. The Chalder’s FS score decreased significantly (from 25.9?±?6.1 to 19.2?±?7.5, P?=?0.002) in the yoga group, but not in the control group. In addition to the improvement of fatigue, two patients with CFS and fibromyalgia syndrome in the yoga group also reported pain relief. Furthermore, many subjects reported that their bodies became warmer and lighter after practicing isometric yoga. Although there were no serious adverse events in the yoga group, two patients complained of tiredness and one of dizziness after the first yoga session with the instructor.Conclusions
Isometric yoga as an add-on therapy is both feasible and successful at relieving the fatigue and pain of a subset of therapy-resistant patients with CFS.Trial registration

Populations of obliquebanded leafroller, Choristoneura rosaceana Harris, and three-lined leafroller, Pandemis limitata Robinson, were obtained from seven sites in the Okanagan and Similkameen Valleys of British Columbia and assayed for their responses to three insecticides using a leaf disk bioassay. Lethal concentration ratios (LCRs) were calculated for all populations compared with a susceptible laboratory colony of C. rosaceana; significant variation was detected in response to all three insecticides. LCRs were 0.86-15.52 for azinphosmethyl, 0.38-2.37 for spinosad (Success), and 0.58-4.89 for Bacillus thuringiensis (Foray). Correlation analysis indicated no cross-resistance among the three insecticides. Leafroller populations obtained from apple orchards managed with organic production practices were more susceptible to azinphosmethyl than leafrollers obtained from conventionally managed sites. Conversely, the highest levels of tolerance to B. thuringiensis were observed in the populations from organic sites, possibly reflecting usage patterns; B. thuringiensis is one of the few insecticides allowed under organic production guidelines. All populations were highly susceptible to spinosad, which may be a useful tool for resistance management programs if used judiciously. 相似文献

German linking elements are sometimes classified as inflectional affixes, sometimes as derivational affixes, and in any case
as morphological units with at least seven realisations (e.g. -s-, -es-, -(e)n-, -e-). This article seeks to show that linking elements are hybrid elements situated between morphology and phonology. On the
one hand, they have a clear morphological status since they occur only within compounds (and before a very small set of suffixes)
and support the listener in decoding them. On the other hand, they also have to be analysed on the phonological level, as
will be shown in this article. Thus, they are marginal morphological units on the pathway to phonology (including prosodics).
Although some alloforms can sometimes be considered former inflectional endings and in some cases even continue to demonstrate
some inflectional behaviour (such as relatedness to gender and inflection class), they are on their way to becoming markers
of ill-formed phonological words. In fact, linking elements, above all the linking -s-, which is extremely productive, help the listener decode compounds containing a bad phonological word as their first constituent,
such as Geburt+s+tag ‘birthday’ or Religion+s+unterricht ‘religious education’. By marking the end of a first constituent that differs from an unmarked monopedal phonological word,
the linking element aids the listener in correctly decoding and analysing the compound. German compounds are known for their
length and complexity, both of which have increased over time—along with the occurrence of linking elements, especially -s-. Thus, a profound instance of language change can be observed in contemporary German, one indicating its typological shift

In the haloarchaea Haloferax volcanii, ribosomes are found in the cytoplasm and membrane-bound at similar levels. Transformation of H. volcanii to express chimeras of the translocon components SecY and SecE fused to a cellulose-binding domain substantially decreased ribosomal membrane binding, relative to non-transformed cells, likely due to steric hindrance by the cellulose-binding domain. Treatment of cells with the polypeptide synthesis terminator puromycin, with or without low salt washes previously shown to prevent in vitro ribosomal membrane binding in halophilic archaea, did not lead to release of translocon-bound ribosomes, indicating that ribosome release is not directly related to the translation status of a given ribosome. Release was, however, achieved during cell starvation or stationary growth, pointing at a regulated manner of ribosomal release in H. volcanii. Decreased ribosomal binding selectively affected membrane protein levels, suggesting that membrane insertion occurs co-translationally in Archaea. In the presence of chimera-incorporating sterically hindered translocons, the reduced ability of ribosomes to bind in the transformed cells modulated protein synthesis rates over time, suggesting that these cells manage to compensate for the reduction in ribosome binding. Possible strategies for this compensation, such as a shift to a post-translational mode of membrane protein insertion or maintained ribosomal membrane-binding, are discussed. 相似文献

Background
Patients with pituitary stalk interruption syndrome (PSIS) are initially referred for hypoglycemia during the neonatal period or growth retardation during childhood. PSIS is either isolated (nonsyndromic) or associated with extra-pituitary malformations (syndromic).Objective
To compare baseline characteristics and long-term evolution in patients with PSIS according to the initial presentation.Study Design
Sixty-seven patients with PSIS were included. Data from subgroups were compared: neonates (n = 10) versus growth retardation patients (n = 47), and syndromic (n = 32) versus nonsyndromic patients (n = 35).Results
Neonates displayed a more severe hormonal and radiological phenotype than children referred for growth retardation, with a higher incidence of multiple hormonal deficiencies (100% versus 34%; P = 0.0005) and a nonvisible anterior pituitary lobe (33% versus 2%; P = 0.0017). Regular follow-up of growth might have allowed earlier diagnosis in the children with growth retardation, as decreased growth velocity and growth retardation were present respectively 3 and 2 years before referral. We documented a progressive worsening of endocrine impairment throughout childhood in these patients. Presence of extra-pituitary malformations (found in 48%) was not associated with more severe hormonal and radiological characteristics. Growth under GH treatment was similar in the patient groups and did not vary according to the pituitary MRI findings.Conclusions
PSIS diagnosed in the neonatal period has a particularly severe hormonal and radiological phenotype. The progressive worsening of endocrine impairment throughout childhood justifies periodic follow-up to check for additional hormonal deficiencies. 相似文献14.

It has been previously reported that presentation of square-wave gratings to either side of the horizontal meridian of the visual field gives rise to different Simple Reaction Times (RTs), depending upon the spatial frequency of the stimuli. Specifically, for 1 c/deg stimulus RT is faster in the lower hemefield, whereas the reverse is true for 3 c/deg pattern, RT being faster in the upper visual field. In the reported experiment, RT to simultaneous presentation of either the same (alternatively 1 c/deg or 3 c/deg) or different (i.e. 1 c/deg and 3 c/deg combined) spatial frequencies to both hemifields was analyzed. The data show that whenever the two half components correspond to different RTs, the resulting RT equates that of the faster component. Conversely, when the two components give rise to identical RTs, the resulting RT does not differ from the value obtained with each half stimulus. Implications of this result for the functional organization of the visual system are discussed. 相似文献

Bla g allergens are major targets of IgE responses associated with cockroach allergies. However, little is known about corresponding T cell responses, despite their potential involvement in immunopathology and the clinical efficacy of specific immunotherapy. Bioinformatic predictions of the capacity of Bla g 1, 2, 4, 5, 6, and 7 peptides to bind HLA-DR, -DP, and -DQ molecules, and PBMC responses from 30 allergic donors, identified 25 T cell epitopes. Five immunodominant epitopes accounted for more than half of the response. Bla g 5, the most dominant allergen, accounted for 65% of the response, and Bla g 6 accounted for 20%. Bla g 5 induced both IL-5 and IFN-γ responses, whereas Bla g 6 induced mostly IL-5, and, conversely, Bla g 2 induced only IFN-γ. Thus, responses to allergens within a source are independently regulated, suggesting a critical role for the allergen itself, and not extraneous stimulation from other allergens or copresented immunomodulators. In comparing Ab with T cell responses for several donor/allergen combinations, we detected IgE titers in the absence of detectable T cell responses, suggesting that unlinked T cell-B cell help might support development of IgE responses. Finally, specific immunotherapy resulted in IL-5 down modulation, which was not associated with development of IFN-γ or IL-10 responses to any of the Bla g-derived peptides. In summary, the characteristics of T cell responses to Bla g allergens appear uncorrelated with IgE responses. Monitoring these responses may therefore yield important information relevant to understanding cockroach allergies and their treatment. 相似文献

Targeting of MHCII-invariant chain complexes from the trans-Golgi network to endosomes is mediated by two di-leucine-based signals present in the cytosolic domain of invariant chain. Generation of this endosomal targeting signal is also dependent on multimerization of the invariant chain cytosolic domain sequences, mediated through assembly of invariant chain into homotrimers. A small subset of invariant chain is modified by the addition of chondroitin sulfate and is expressed on the cell surface in association with MHCII. In the present study, we have followed the biosynthetic pathway and route of intracellular transport of this proteoglycan form of invariant chain. We found that the efficiency of chondroitin sulfate modification can be increased by altering the invariant chain amino acid sequence around Ser-201 to the xylosylation consensus sequence. Our results also indicate that, following sulfation, the proteoglycan form is transported rapidly from the trans-Golgi network to the cell surface and is degraded following internalization into an endocytic compartment. Invariant chain-chondroitin sulfate is present in invariant chain trimers that also include conventional non-proteoglycan forms of invariant chain. These data indicate that invariant chain-chondroitin sulfate-containing complexes are transported rapidly from the trans-Golgi network to the cell surface in spite of the presence of an intact endosomal localization signal. Furthermore, these results suggest that invariant chain-chondroitin sulfate may play an important role in the generation of cell-surface pools of invariant chain that can serve as receptors for CD44 and macrophage migration inhibitory factor. 相似文献

The primary focus of this investigation was to evaluate the relevance of the first attempt to conceptualize the Ichkeul aquatic ecosystem. This ‘draft model’, published in 1994, was the conceptual working basis of a multidisciplinary research programme undertaken at Ichkeul Lagoon between 1993 and 1995 by a French–Tunisian–German team. The research programme was intended as the basis for devising conservation measures for the lagoon, and the data collected by the research team are used here to assess the validity of the draft model. A related goal is to make the unpublished data from the multidisciplinary study available to other researchers to improve knowledge of Mediterranean wetlands. We have compiled the biomasses of all functional groups recorded during the multidisciplinary study and compared them with the assumptions about biomass relationships in the draft model. Previously collated biomass data for seagrasses and benthic invertebrates collected from July 1993 to April 1994 are complemented by biomass estimates of phytoplankton, zooplankton, fishes and waterfowl from the same period. Thus, for the first time, biomasses of all major components of the Ichkeul aquatic community, from phytoplankton to birds, are presented together. Comparisons of these data with the qualitative biomass estimates in the draft model reveal important shortcomings in the earlier work. Biomasses of zooplankton and benthic invertebrates were underestimated in the draft model, while the biomass of migratory waterfowl was overestimated. This study is an important step towards providing the baseline information required for sound management and conservation decisions regarding the lagoon. 相似文献

Methods to research the origin of microbial life are limited. However, microorganisms were the first organisms on the Earth capable of cell growth and division, and interactions with their environment, other microbial cells, and eventually with diverse eukaryotic organisms. The origin of microbial life and the supporting scientific evidence are both an enigma and a scientific priority. Numerous hypotheses have been proposed, scenarios imagined, speculations presented in papers, insights shared, and assumptions made without supporting experimentation, which have led to limited progress in understanding the origin of microbial life. The use of the human imagination to envision the origin of life events, without supporting experimentation, observation and independently replicated experiments required for science, is a significant constraint. The challenge remains how to better understand the origin of microbial life using observations and experimental methods as opposed to speculation, assumptions, scenarios, envisioning events and un-testable hypotheses. This is not an easy challenge as experimental design and plausible hypothesis testing are difficult. Since past approaches have been inconclusive in providing evidence for the origin of microbial life mechanisms and the manner in which genetic instructions was encoded into DNA/RNA, it is reasonable and logical to propose that progress will be made when testable, plausible hypotheses and methods are used in the origin of microbial life research, and the experimental observations are, or are not reproduced in independent laboratories. These perspectives will be discussed in this article as well as the possibility that a pre-biotic film preceded a microbial biofilm as a possible micro-location for the origin of microbial cells capable of growth and division. 相似文献
