After ovulation, non-pregnant female giant pandas experience pseudopregnancy. During pseudopregnancy, non-pregnant females exhibit physiological and behavioral changes similar to pregnancy. Monitoring hormonal patterns that are usually different in pregnant mammals are not effective at determining pregnancy status in many animals that undergo pseudopregnancy, including the giant panda. Therefore, a physiological test to distinguish between pregnancy and pseudopregnancy in pandas has eluded scientists for decades. We examined other potential markers of pregnancy and found that activity of the acute phase protein ceruloplasmin increases in urine of giant pandas in response to pregnancy. Results indicate that in term pregnancies, levels of active urinary ceruloplasmin were elevated the first week of pregnancy and remain elevated until 20–24 days prior to parturition, while no increase was observed during the luteal phase in known pseudopregnancies. Active ceruloplasmin also increased during ultrasound-confirmed lost pregnancies; however, the pattern was different compared to term pregnancies, particularly during the late luteal phase. In four out of the five additional reproductive cycles included in the current study where females were bred but no birth occurred, active ceruloplasmin in urine increased during the luteal phase. Similar to the known lost pregnancies, the temporal pattern of change in urinary ceruloplasmin during the luteal phase deviated from the term pregnancies suggesting that these cycles may have also been lost pregnancies. Among giant pandas in captivity, it has been presumed that there is a high rate of pregnancy loss and our results are the first to provide evidence supporting this notion.  相似文献

对5只大熊猫发情配种期间阴道粘液电阻值进行了检测,结果发现4只发情表现正常的大熊猫发情配种期阴道粘液电阻值呈现相似的V字形变化曲线,V形曲线的谷底所代表时期恰好与尿雌酮峰值期吻合,其中3只大熊猫于秋后产仔,结果提示测定大熊猫发情配种期间阴道粘液电阻值可有效用于监测排卵,具有良好的应用前景。  相似文献

Artificial insemination (AI) is an important component of captive breeding programs for endangered species, such as the giant panda. The panda has been the subject of increasingly successful captive breeding programs involving a compilation of assisted breeding techniques, including AI using cryopreserved spermatozoa. AI implementation is currently hampered by a lack of understanding of the factors that may cause failure. We investigated factors influencing the probability of success of AI for 14 giant panda females housed at the China Center for Research and Conservation of the Giant Panda (CCRCGP) inseminated in a total of 20 instances using cryopreserved spermatozoa from 11 males currently residing in 6 different captive breeding institutions. One of the pandas was the oldest giant panda female to ever successfully conceive from AI (20.5 years old). The success of AI was significantly affected by the timing of AI in relationship to both timing of peak urinary estrogen of the female and percent decline in urinary estrogen between the peak level and the first AI attempt. Our results suggest that the window for successful AI in giant pandas may be narrower than previously suspected, although individual differences in rates of decline in urinary estrogen may reflect some degree of variation in this crucial window across females. Our results are consistent with recent research on pandas and other species that demonstrates the efficacy of cryopreserved spermatozoa for AI and highlights the need for more in‐depth analysis of factors related to female physiology that may influence its success. 行为谱是动物行为学研究的基础。大熊猫(Ailuropoda melanoleuca)是生物多样性保护中的旗舰物种,其行为生态的研究备受关注,然而由于大熊猫数量稀少且警惕性强,很难在野外进行观察,有关其行为谱等基础行为研究结果主要是基于圈养大熊猫的观察研究。为了促进野生大熊猫行为学的深入研究,2015年1月—2016年6月,2018年8月— 2019年4月我们在卧龙自然保护区基于红外相机图片和视频数据,利用PAE编码系统建立了野生大熊猫行为谱和PAE编码系统。研究共计在14种环境背景中记录到大熊猫12种姿势、52种动作、56种行为。一方面,本研究结果基本涵盖了大熊猫的主要行为,并发现了一些未在圈养大熊猫行为谱中记录到的行为,如野生大熊猫利用植物的茎进行自我除雪行为;另一方面丰富了观察大熊猫行为的环境因素。以上结果为深入研究野生大熊猫行为生态学提供了基础信息。    相似文献

Background/ObjectiveThe giant panda belongs to the family Ursidae and, as a species of bear, still retains the simple digestive system of a Carnivoran. However, under the pressure of a specific habitat they had to adapt to a plant mono-diet consisting of bamboo with different species and growth stages around the year. A plant-based diet has relatively low iodine content with risk of iodine deficiency. Furthermore, bamboo contains cyanogenic glycosides releasing cyanide whose detoxification metabolite the thiocyanate acts as antagonist against iodine uptake and storage in the thyroid. To date very little is known about the iodine nutritional status of the giant panda, thus this study was conducted to receive the first information about the iodine nutrition of captive giant panda.Subjects/MethodsHere we investigated the iodine content of bamboo with different plant parts/vegetation stage and species and further compounds of the captive giant panda diet. Next, the urinary iodine (UI) and urinary thiocyanate (UT) levels of infant, sub-adult, adult and geriatric captive giant pandas was measured during the periods when the pandas consume both bamboo leaves- and culm (bamboo leaf-culm stage). Afterwards, the UI of 19 adult giant pandas was measured again for the different iodine intake during bamboo shoot stage. Finally, in this study part also the fecal iodine concentration was analyzed for calculation of total iodine excretion in relation to the iodine intake.ResultsBamboo leaves had the highest iodine content (453 μg/kg dry matter (DM)), followed by the shoots (84 μg/kg DM, p < 0.05), while bamboo culm had the lowest value (12 μg/kg DM, p < 0.05). During bamboo leaf-culm stage, giant pandas of different age groups had different UI and UT levels (p < 0.05). Furthermore, UI and UT were positively correlated among sub-adult, adult and geriatric giant pandas (p < 0.05). In adult giant pandas during bamboo shoot stage, the iodine excretion in feces was not different from that in urine while their total iodine excretion was less than their iodine intake (p < 0.05). Moreover, during bamboo shoot stage, the UI level of adult giant pandas was much lower than noted during bamboo leaf-culm stage (p < 0.05).ConclusionsOur results indicate that UI of captive giant pandas was related to their age as well as to the vegetation stage/part of bamboo they consumed reflecting a different periodic iodine supply. Thiocyanate and fecal excretion should be emphasized when considering the iodine nutrition of giant pandas.  相似文献

The giant panda (Ailuropoda melanoleuca) is classified as a carnivore, yet subsists on a diet comprised almost exclusively of bamboo. Wild and captive giant pandas use highly selective foraging behaviors for processing and consuming bamboo. These behaviors are for the first time quantified in captive giant pandas over a 5‐year period of time showing highly specific seasonal trends. Giant panda feeding behavior was recorded using live video observations of two giant pandas housed at the Memphis Zoo from November 2003 to June 2008. Leaf was the primary plant part consumed from June to December, whereas culm was consumed primarily from February to May, with both bears displaying similar seasonal shifts in plant part consumption. From May to June, leaf consumption increased significantly (P‐values<0.001); from June to August, leaf consumption remained high and stable. From December to March, leaf consumption decreased significantly (P‐values<0.001). Specific behaviors for bamboo leaf and culm consumption were also observed. Both bears formed wads of leaves before ingestion while feeding on leaf, but the male employed this feeding behavior more often than the female (54 and 33%, respectively). Both bears used similar culm‐stripping behavior (26 and 25%), used to remove the outer layer and isolate the pith for consumption. This study indicates that unique seasonal foraging behaviors observed in wild pandas are also apparent in captive animals in relation to plant part selectivity and feeding behaviors. Ursids (bears) in general, and giant pandas in particular, are highly altricial at birth. The components of bear milks and their changes with time may be uniquely adapted to nourish relatively immature neonates, protect them from pathogens, and support the maturation of neonatal digestive physiology. Serial milk samples collected from three giant pandas in early lactation were subjected to untargeted metabolite profiling and multivariate analysis. Changes in milk metabolites with time after birth were analysed by Principal Component Analysis, Hierarchical Cluster Analysis and further supported by Orthogonal Partial Least Square-Discriminant Analysis, revealing three phases of milk maturation: days 1–6 (Phase 1), days 7–20 (Phase 2), and beyond day 20 (Phase 3). While the compositions of Phase 1 milks were essentially indistinguishable among individuals, divergences emerged during the second week of lactation. OPLS regression analysis positioned against the growth rate of one cub tentatively inferred a correlation with changes in the abundance of a trisaccharide, isoglobotriose, previously observed to be a major oligosaccharide in ursid milks. Three artificial milk formulae used to feed giant panda cubs were also analysed, and were found to differ markedly in component content from natural panda milk. These findings have implications for the dependence of the ontogeny of all species of bears, and potentially other members of the Carnivora and beyond, on the complexity and sequential changes in maternal provision of micrometabolites in the immediate period after birth.  相似文献

截至2003年底,我国野生大熊猫种群数量达1596只,分布在陕西、四川和甘肃3省的45个县境内,总栖息地面积达2304991hm^2。与第2次大熊猫调查相比,野生大熊猫生存状况已得到改善,分布范围扩大、栖息地面积增加、种群数量进一步增长。本文在第3次大熊猫调查的基础上,就野生大熊猫种群及栖息地现状进行了分析,指出未来保护大熊猫所面临的3个方面的挑战,即来自物种自身生物学特性的挑战、栖息地破碎化及隔离小种群未来命运的挑战以及大熊猫保护与社区经济发展需求相冲突的挑战。作者还就我国大熊猫保护前景进行了展望,即自然保护区数量将进一步增加,栖息地状况将进一步改善;种群数量在总体保持稳定的基础上将逐步增长,但局部小种群灭绝风险将加剧;圈养种群将形成能自我维持的种群,圈养个体通过培训将逐步放归到隔离野生小种群中以改变其命运。  相似文献

有大量的研究提出,大熊猫具有发达的嗅觉和听觉系统但是视力很差。 虽然有些关于大熊猫视力和色觉方面的研究,但是至今我们对大熊猫的具体视力值方面的内容知之甚少。为了定量化大熊猫的视力值,我们观察并记录了8只圈养大熊猫(雌:雄=4:4)对于移动不同水平黑白相间条纹宽度的纸片的行为反应。我们发现,在测量距离为50 cm的情况下,大熊猫能够区分大约为0.46mm宽的黑白相间条纹。这个研究结果为大熊猫视网膜结构的解剖学研究提供了行为学支持,也为今后研究大熊猫的通讯和认知提供一定的参考。  相似文献

为了研究大熊猫对食物的化学性消化特点和机制,测定了9只大熊猫唾液和3只大熊猫胃肠道中主要消化酶的活力,并与其他动物进行了比较.结果显示,大熊猫唾液呈碱性,蛋白酶和淀粉酶等消化酶活力低;肠道中淀粉酶活力高,而脂肪酶活力明显低于棕熊.大熊猫小肠粘膜中存在显著量的蔗糖酶、乳糖酶和麦芽糖酶活力.另外,在1只大熊猫胃和直肠液中检测到了少量纤维素酶活力.研究结果提示,大熊猫唾液直接参与食物消化的作用可能很弱;大熊猫对淀粉类食物有很好的消化能力,但对脂肪类食物消化能力相对不高.大熊猫胃肠道消化酶的活力特点适应其消化天然食物中的营养物质.  相似文献

 大熊猫是生物多样性保护的旗舰种, 保护大熊猫及其栖息地是保护生物多样性和生态系统功能完整性与稳定性的重要保障体现。汶川地震灾区位于大熊猫重点分布区岷山-邛崃山, 地震及其次生灾害导致该区27个大熊猫自然保护区不同程度受损, 8.3%的大熊猫栖息地因地震而被破坏。地震及其次生灾害对大熊猫栖息地的影响主要表现在: 1) 地震埋没和砸毁大熊猫赖以生存的主食竹, 地震可能诱发主食竹开花, 威胁到大熊猫的健康和食物安全; 2) 地震及其诱发的土壤和山石运动显著影响森林的动态特征, 森林大面积丧失或质量下降; 3) 地震改变大熊猫栖息地生境特征, 大熊猫个体交流的廊道阻断, 形成“生殖孤岛”, 遗传多样性降低, 栖息地破碎化进程加快。应对震后大熊猫栖息地恢复的主要对策有: 1) 重新评估震后大熊猫栖息地质量, 并重新规划现有大熊猫保护区群的布局; 2) 应用地理信息系统、遥感及数学模型等手段与野外实地实证研究相结合的方法, 全面查清震后大熊猫栖息地主食竹资源状况及分布规律并及时监测其动态, 复壮更新大熊猫主食竹; 3) 利用天然植被自然恢复和人工重建等措施恢复因地震而退化或丧失的大熊猫栖息地。  相似文献

对圈养大熊猫 (Ailuropoda melanoleuca) 开展长期行为监测能及时了解其所处生理周期和健康状况,有助于繁殖饲养机构迅速采取相应繁育保护措施提高饲养管理水平,但目前无法对大熊猫进行24 h监控并及时地获得相应的行为信息。准确的动物姿态估计是动物行为研究的关键,也是诸多下游应用的基础。了解大熊猫的姿态可以促进大熊猫行为研究并提升保护管理水平。为了提高复杂环境下大熊猫姿态估计的准确率,本文以高分辨率网络 (High resolution net, HRNet) 为基础网络架构提出了一种大熊猫姿态估计方法:针对大熊猫不同部位尺度差异较大的问题,在HRNet-32中引入了空洞空间金字塔池化 (Atrous spatial pyramid pooling, ASPP) 模块,在提升特征感受野的同时捕获多尺度信息;同时对大熊猫身体关键点进行分组,引入基于部位的多分支结构来学习特定于每个部位组的表征。多次对比实验结果表明本文所用模型具有较高的检测精度:在PCK@0.05中所用模型精度达到了81.51%。本文提出的方法可为大熊猫的行为分析和健康评估提供技术支撑。  相似文献

Aim To examine the effects of forest fragmentation on the distribution of the entire wild giant panda (Ailuropoda melanoleuca) population, and to propose a modelling approach for monitoring the spatial distribution and habitat of pandas at the landscape scale using Moderate Resolution Imaging Spectro‐radiometer (MODIS) enhanced vegetation index (EVI) time‐series data. Location Five mountain ranges in south‐western China (Qinling, Minshan, Qionglai, Xiangling and Liangshan). Methods Giant panda pseudo‐absence data were generated from data on panda occurrences obtained from the third national giant panda survey. To quantify the fragmentation of forests, 26 fragmentation metrics were derived from 16‐day composite MODIS 250‐m EVI multi‐temporal data and eight of these metrics were selected following factor analysis. The differences between panda presence and panda absence were examined by applying significance testing. A forward stepwise logistic regression was then applied to explore the relationship between panda distribution and forest fragmentation. Results Forest patch size, edge density and patch aggregation were found to have significant roles in determining the distribution of pandas. Patches of dense forest occupied by giant pandas were significantly larger, closer together and more contiguous than patches where giant pandas were not recorded. Forest fragmentation is least in the Qinling Mountains, while the Xiangling and Liangshan regions have most fragmentation. Using the selected landscape metrics, the logistic regression model predicted the distribution of giant pandas with an overall accuracy of 72.5% (κ = 0.45). However, when a knowledge‐based control for elevation and slope was applied to the regression, the overall accuracy of the model improved to 77.6% (κ = 0.55). Main conclusions Giant pandas appear sensitive to patch size and isolation effects associated with fragmentation of dense forest, implying that the design of effective conservation areas for wild giant pandas must include large and dense forest patches that are adjacent to other similar patches. The approach developed here is applicable for analysing the spatial distribution of the giant panda from multi‐temporal MODIS 250‐m EVI data and landscape metrics at the landscape scale.  相似文献

In order to improve breeding of in situ populations of bears, a comprehensive study of reproductive physiology in Brown (Ursus arctos), Spectacled (Tremarctos ornatus) and Giant panda bears (Ailuropoda melanoleuca) was performed. The objective was to perform non-invasive analyses of urinary and fecal steroid metabolites. In addition, we investigated the presence of reproduction-related urinary volatile substances of these bears that might trigger the reproductive behavior. Urinary estrogen concentrations, routinely used to monitor follicular activity in Giant panda, were inappropriate for monitoring follicular activity in Spectacled bear. In addition, no estrogen peak related to mating activity was observed in Brown bear. Further contrasting Giant panda, although urinary pregnanediol analyses failed to indicate luteal activity in either Spectacled or Brown bears, urinary (Spectacled bear) and fecal (Brown bear) concentrations of progesterone were an appropriate indicator of luteal activity. The Giant panda had volatile components (medium-chain fatty acids) in their urine that increased simultaneously with the seasonal increase of estrogens. These fatty acids were also detected in the Brown during estrus and Spectacled bear. Further studies on the behavioral relevance of these fatty acids are required to determine if they are pheromones.  相似文献

The giant panda is seasonally monoestrus, experiencing a single estrous with spontaneous ovulation in the spring. Therefore, accurate monitoring of the estrous cycle to pinpoint the time of ovulation is critical for the success of timed mating or artificial insemination. Analysis of exfoliated vaginal epithelial cells is a simple technique that rapidly yields information about the estrous status of a panda. Vaginal swabs were obtained during five estrous cycles of two nulliparous females. Cells were stained with the trichrome Papanicolaou and classified as basophils, intermediates or superficials. The color of stained cells, basophilic, acidophilic or keratinized, was recorded as a characteristic independent of the three standard cell types. The day urinary conjugates of estrogen fell from peak levels was considered the day of ovulation. A chromic shift occurred 8-9 days before ovulation when the majority of exfoliated vaginal cells changed from basophilic (blue) to acidophilic (pink) without accompanying nuclear or cytoplasmic changes. A second chromic shift was consistently observed 2 days prior to ovulation when keratinized (orange) cells replaced acidophils as the majority of vaginal cells. Monochrome staining of vaginal cells is sufficient to quantify superficial cells, which is a useful adjunct to behavioral and endocrinological data in determining estrous in the giant panda. However, the timing and duration of superficial cell elevations are substantially different between and within individual females, which limits the accuracy of timing ovulation for artificial insemination. The predictive value of vaginal cytology was greatly enhanced with the trichrome stain and evaluation of cell color.  相似文献

The giant panda (Ailuropoda melanoleuca) is one of the world’s most beloved endangered mammals. Although the draft genome of this species had been assembled, little was known about the composition of its microRNAs (miRNAs) or their functional profiles. Recent studies demonstrated that changes in the expression of miRNAs are associated with immunity. In this study, miRNAs were extracted from the blood of four healthy giant pandas and sequenced by Illumina next generation sequencing technology. As determined by miRNA screening, a total of 276 conserved miRNAs and 51 novel putative miRNAs candidates were detected. After differential expression analysis, we noticed that the expressions of 7 miRNAs were significantly up-regulated in young giant pandas compared with that of adults. Moreover, 2 miRNAs were up-regulated in female giant pandas and 1 in the male individuals. Target gene prediction suggested that the miRNAs of giant panda might be relevant to the expressions of 4,602 downstream genes. Subseuqently, the predicted target genes were conducted to KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway analysis and we found that these genes were mainly involved in host immunity, including the Ras signaling pathway, the PI3K-Akt signaling pathway, and the MAPK signaling pathway. In conclusion, our results provide the first miRNA profiles of giant panda blood, and the predicted functional analyses may open an avenue for further study of giant panda immunity.  相似文献

大熊猫(Ailuropoda melanoleuca)是中国的特有物种,其成功放归可能受放归地气候、栖息地、同域动物等多种因素的影响。本研究在野外调查的基础上,基于生态位理论等方法,探讨了栗子坪国家级自然保护区大熊猫与其大中型同域动物的空间关系。调查结果表明,该保护区内大熊猫同域动物共20种,分别属于2纲5目,其中东洋型分布物种占优势(30%)。大熊猫同域动物分布海拔显著低于大熊猫的分布海拔(P<0.05),大熊猫分别与灵长目等5个目的动物在海拔分布上均存在显著的生态位分化(P<0.001)。灵长目动物在空间分布上与大熊猫分布相异,而其他目的动物与大熊猫空间分布类似。偶蹄目动物痕迹数量占比最高(54.14 %),而食肉目的小熊猫与大熊猫样线共同遇见率达到了43.75%。本研究结果表明,该地大熊猫同域动物较为丰富,在海拔分布上与大熊猫存在生态位分化。该研究可为圈养大熊猫的放归提供参考依据。  相似文献

Livestock production is one of the greatest threats to biodiversity worldwide. However, impacts of livestock on endangered species have been understudied, particularly across the livestock–wildlife interface in forested protected areas. We investigated the impact of an emerging livestock sector in China's renowned Wolong Nature Reserve for giant pandas. We integrated empirical data from field surveys, remotely sensed imagery, and GPS collar tracking to analyze (1) the spatial distribution of horses in giant panda habitat, (2) space use and habitat selection patterns of horses and pandas, and (3) the impact of horses on pandas and bamboo (panda's main food source). We discovered that the horse distribution overlapped with suitable giant panda habitat. Horses had smaller home ranges than pandas but both species showed similarities in habitat selection. Horses consumed considerable amounts of bamboo, and may have resulted in a decline in panda habitat use. Our study highlights the need to formulate policies to address this emerging threat to the endangered giant panda. It also has implications for understanding livestock impacts in other protected areas across the globe.  相似文献

Giant panda hair samples obtained by noninvasive methods served as a source of DNA for amplification of seven giant panda microsatellite loci utilizing the polymerase chain reaction. Thirteen giant pandas held in Chinese zoos were tested for identification of paternity. Some males listed as sires have been excluded as the biological father of captive-born giant pandas. Because of the death of some potential sires, paternity is still not assigned for some giant pandas, although there is a high likelihood that paternity assignment could be made if postmortem samples are available for genetic analysis. The DNA microsatellite variation assayed by the test we have developed provides a rapid, highly informative, and noninvasive method for paternity identification in giant pandas. © 1994 Wiley-Liss, Inc.  相似文献
