Smelts of the genus Osmerus, O. eperlanus and O. mordax dentes, inhabiting the basins of the Atlantic, Arctic, and Pacific oceans were investigated using RFLP analysis of the mitochondrial DNA segments A8/A6/COIII/ND3 and ND3/ND4, and sequencing of the cytb and COI genes (mtDNA), and intron 1 of the rpS7 gene (nDNA). A total of 14 samples from the populations from most part of the Russian range were examined. The mean values of haplotype and nucleotide diversity constituted 0.5997 ± 0.11264 and 0.003201 for O. m. dentex, and 0.3086 ± 0.09892 and 0.000431 for O. eperlanus, respectively. The high level of interspecific diversity (12.94%) along with the low level of intraspercific diversity (0.049% for O. m. dentex, and 0.001% for O. eperlanus was observed. The dendrograms (UPGMA, NJ, MP, and BA) constructed using the data of RFLP analysis of mtDNA, along with the sequencing data of mitochondrial and nuclear genes were congruent. The representatives of O. eperlanus and O. m. dentex formed steady clusters in accordance with their species affiliation, albeit without subdivision into local populations depending on their geographic locality.  相似文献

Two smelt species that inhabit the northwest of the Sea of Japan, Hypomesus japonicus and H. nipponensis (family Osmeridae), which are both currently referred to as Japanese smelt, have been studied in order to clarify the taxonomic value of morphometric characters. Variations between individuals of these species have been revealed using a multivariate analysis of their morphometric characters and indices. The results of the analysis show that H. japonicus is distinguished from H. nipponensis both by the previously known characters (the eye diameter is smaller and the vertical through the beginning of the dorsal fin is closer to the head than that through the beginning of the pelvic fins) and by a greater height of the dorsal and adipose fins in the former species as defined in the paper. Within the species H. japonicus, the individuals sampled from Olga Bay are most similar morphologically to those from the coastal waters off the village of Terney. In contrast, the individuals of this species from the coastal waters off Russky Island and from Olga Bay manifest the greatest differences.  相似文献

A critical analysis of literature data on the distribution, morphology, and phylogeography of the Arctic lamprey (Lethenteron camtschaticum) and five species of marine and anadromous fish such as navaga (Eleginus navaga), pollock (Theragra chalcogramma), rainbow smelt (Osmerus mordax dentex), Pacific herring (Clupea pallasii), and pond smelt (Hypomesus olidus) has been performed. The results show that all these species have colonized Northern European seas, distributing along the Arctic coastline of Eurasia after the glacier retreat. The reasons that the dispersal of these species in the Atlantic Ocean may be impeded (preference for a cold environment, competition, and decrease of the evolutionary potential) are discussed.  相似文献

Putative phylogenetic relationships between all smelt species inhabiting Russian waters were studied using RFLP and the data on divergence between nucleotide sequences of the mitochondrial gene encoding cytochrome b. All types of phylogenetic trees (NJ, MP, and Bayesian) displayed stable clustering into isolated groups corresponding to the division of the subfamily Osmerinae into three genera, Mallotus, Osmerus, and Hypomesus. It was demonstrated that the Mallotus phylum was the first to diverge from a hypothetical common ancestor of the smelts. Later, it divided into two clusters corresponding to the modern subspecies Mallotus villosus villosus and M. v. catervarius. The phylum of the genus Osmerus diverged later than the genus Mallotus and also divided into two clusters, one including O. mordax of the Pacific (Sea of Japan) and Arctic (Kara and Barents Seas) basins and the other, anadromous and resident ecotypes of the O. eperlanus from the Baltic Sea basin. The smelts of the genus Hypomesus is the youngest phylogenetic group among the taxa studied; it forms three individual clusters matching the species H. olidus, H. nipponensis, and H. japonicus. The proposed phylogenetic hypothesis is discussed from the standpoint of its compliance with the phylogenetic constructions based on the external morphological and osteological traits.  相似文献

Biological features of the four common fish species, giant grenadier Albatrossia pectoralis (Macrouridae), Pacific herring Clupea pallasii (Clupeidae), Pacific rainbow smelt Osmerus mordax dentex, and Pacific capelin Mallotus villosus catervarius (Osmeridae), were studied under the 20-year dataset (1995?2015). These species inhabit the northwestern Bering Sea in the summer–autumn period and form the schoolings in the Olyutorsky-Navarin region. The size–age parameters of the fish caught by different sampling gear, as well as the peculiarities of the body length and body weight dynamics, spawning periods, spawning range, and conditions, were analyzed. The largest specimens of giant grenadier, Pacific herring, and Pacific rainbow smelt were observed in the catches performed by the bottom setline and the gill nets; the smallest fish were found in the trawl catches. The body length and body weight of Pacific herring were larger in the pelagic trawls compared to the bottom trawls; an opposite pattern was observed for the Pacific capelin. The abundant year-class in the species with short life cycle (capelin and herring) is well tracked on the longterm plots of the fish body size; this is accompanied by the decrease of their biological parameters. Herring stock covers large growing grounds; smaller body size was observed for the herring grazing in the coastal waters; young specimens dominate here.  相似文献

Vicariant geographic isolation and resource partitioning have long been independently identified as processes contributing to the morphological divergence of closely-related species. However, little is known about the extent to which vicariant history influences the adaptive ecological divergence associated with resource partitioning and trophic specialization within species. The present study thus quantified the contribution of vicariant historical genetic divergence to the adaptive contemporary morphological divergence of intraspecific feeding specialists in the Rainbow smelt (Pisces: Osmerus mordax ). This species is characterized by the polyphyletic origin of two lacustrine feeding specialists originating in two intraspecific lineages associated with independent glacial refuges. The historical genetic segregation was initiated approximately 350 000 years ago, whereas the lacustrine trophic segregation arose within the past 10 000 years. Wild caught lacustrine smelt populations were grouped a priori based on known historical genetic identities (Acadian and Atlantic mitochondrial DNA clades) and contemporary feeding specializations (microphageous and macrophageous morphotypes). The present study demonstrated that independent suites of correlated morphological traits are associated with either vicariant history or contemporary feeding specializations. Second, functionally-similar feeding specialists exhibit distinct morphologies resulting largely from vicariant historical processes. Although, the evolutionary processes producing historical phenotypes remains unknown, the results obtained demonstrate how adaptive radiation associated with ecological resource partitioning and feeding specializations can be strongly influenced by intraspecific phenotypic diversification resulting from relatively recent vicariant histories.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 97 , 140–151.  相似文献

The roles of ecological speciation and reinforcement in the formation of contemporary diversity remain contentious. In the present study, we contrast phenotypic and molecular divergence within morphologically diverged bimodal sympatric and allopatric pairs of rainbow smelt, Osmerus mordax. We hypothesize that, in sympatry, evidence of selection associated with resource partitioning will be visible through strong divergence, reinforcement, and greater character displacement. Parallel morphological divergence was observed between the two trophic forms (macrophagous and microphagous), with several examples of greater trait divergence in sympatry than allopatry. Mitochondrial DNA sequence analysis indicated no association between historical clades and morphology; however, Bayesian clustering using microsatellites supported the isolation of these morphs under both allopatry and sympatry. Estimates of genetic isolation were one order of magnitude lower than measures of morphological divergence, consistent with a hypothesis of strong contemporary selection. Using experimental crosses, we obtained similar rates of fertilization success among the allopatric hybrid and pure crosses; whereas, in the sympatric hybrid crosses, fertilization rates dropped by 30–50%, suggesting a clear role for reinforcement through prezygotic incompatibilities. Pond smelt Hypomesus olidus (Pallas, 1814), one of the five species of the genus Hypomesus, family Osmeridae, was examined for intraspecific variation of the mitochondrial DNA cytb (1062 bp) and COI (567 bp) genes. Among the ten single substitutions discovered, only one, leading to the substitution of isoleucine by valine, was nonsynonymous, while the remaining substitutions were synonymous. The degree of genetic divergence among pooled nucleotide sequences in H. olidus populations examined constituted 0.4% on average, ranging from 0.2 to 0.6%. These values were not higher than the levels of divergence between the individuals within the populations. Phylogenetic analysis of the populations examined did not reveal their subdivision depending of their geographic location, and pointed to the absence of intraspecific differentiation of the species.  相似文献

Genetic differentiation of two sympatric char species, the Dolly Varden char (Salvelinus malma Walbaum) and the Taranetz char (S. taranetzi Kaganovski), has been studied. Restriction fragment length polymorphism (RFLP) analysis has been used to compare three mitochondrial DNA (mtDNA) fragments (ND1/ND2, ND5/ND6, and Cytb/D-loop) amplified via polymerase chain reaction (PCR). The divergence between S. malma and S. taranetzi inferred from mtDNA nucleotide sequences is 2.8%; between S. leucomaenis and S. taranetzi, 7.1%; and between S. malma and S. leucomaenis, 7.5%. The absence of common haplotypes and the degree of divergence indicate that the Dolly Varden char and the Taranetz char are genetically isolated and confirm that S. taranetzimay be regarded as a separate species.  相似文献

An extensive mortality of young-of-the-year rainbow smelt (Osmerus mordax) occurred in Lake Erie during the fall of 1969. Dead and dying smelt were observed along the north shore from west of Long Point in the central basin to Port Maitland in the eastern basin. No other fish species was involved. Glugea hertwigi, a microsporidan parasite, was observed in 90% of the distressed smelt examined, and is believed to have been a major contributing factor to the mortality.  相似文献

The rainbow smelt, Osmerus mordax (Mitchill), is an osmerid fish that exhibits extensive life-history diversity throughout watersheds of northeastern North America. There are both ???sea-run (anadromous) and lake-resident (lacustrine) populations and the latter have diversified further into “dwarf-” and “normal-sized” life-history types. Anadromous and lacustrine smelt may inhabit the same watershed and there are several instances where dwarf and normal populations reside within the same lake. We assayed variation among smelt for morphological traits linked to feeding performance in fishes to see if trophic ecology might promote life-history diversity in Osmerus. We also examined mitochondrial DNA (mtDNA) restriction site variation among forms to assess their evolutionary interrelationships. Dwarf smelt had significantly more gill rakers, larger eyes, but shorter upper jaws than normal lake and anadromous smelt. The populations clustered into two trophic “morphotypes”; an anadromous/normal lake group of populations and a group consisting only of dwarf smelt. The mtDNAs of 444 smelt from 16 populations were digested with 12 restriction enzymes revealing 93 composite mtDNA genotypes that clustered (UPGMA) into two major phylogenetic groups differing by 0.78% in sequence. Both genetic groups were present in dwarf and normal smelt as well as in anadromous fish. Further, geographic proximity, rather than trophic morphotype, appeared to be the major determinant of genetic affinities among populations. In two lakes, however, dwarf and normal smelt populations had significantly different mtDNA genotype frequency distributions indicating that the forms are reproductively isolated within both lakes. A clustering analysis of population affinities suggested that the divergence of sympatric dwarf and normal populations had occurred independently in the two lakes. We concluded (1) that trophic ecology is an important factor promoting differentiation in smelt life histories; (2) that smelt ecotypes are polyphyletic and there have been multiple, independent divergences of Osmerus life-history types throughout northeastern North America; and (3) that the biological and mtDNA differences between coexisting dwarf and normal lake smelt argue strongly that their genetic isolation may have developed sympatrically.  相似文献

We examined differentiation of three sympatric charr species Dolly Varden charr Salvelinus malmaWalbaum, white-spotted charrS.leucomaenisPallas, and Levanidov charrS. levanidovi Chereshnev, Scopetz, Gudkov. Using restriction fragment length polymorphism analysis (RFLP), three mitochondrial DNA fragments (ND1/ND2, ND5/ND6, and Cytb/D-loop) amplified in PCR were compared. The divergence of mtDNA nucleotide sequences betweenS. levanidovi and S. leucomaenis was about 9.7%, between S. levanidovi and S. malma, 8.7%, and between S. malma andS. laecomaenis, 7.5%. These results indicate genetic isolation of the species examined and support the earlier suggestion on closeness of Levanidov charr to the common ancestor of the genus Salvelinus.  相似文献

Episodes of trans-Arctic faunal exchange and isolation between the north Pacific and Atlantic ocean basins have been implicated as important historic geological events contributing to extant patterns of genetic diversity and structure in Holarctic faunas. We made a further test of the significance of such biogeographic events by examining mitochondrial DNA (mtDNA) restriction fragment length and cytochrome b sequence polymorphism among north Pacific and Arctic, north-western Atlantic (north-eastern North American), and north-eastern Atlantic (European) regional forms of the boreal smelt, genus Osmerus. Our analyses also assessed whether the regional forms within this ‘species complex’: (i) represent a single widely distributed and polytypic species, or is composed of three geographically distinct species, and (ii) resulted from a single split from north Pacific ancestral Osmerus or two independent Pacific-Atlantic divergences. MtDNA sequence divergence estimates among forms ranged from 5.6–8.9% and from 6.1–8.5% based on restriction fragment and 300 base pairs of cytochrome b sequencing, respectively. Divergence within forms averaged less than 0.5% for fragment analysis and no differences were detected from sequence analysis. Provisional dating of lineage separations in Osmerus based on our sequence divergence estimates suggested a mid-Pliocene to early Pleistocene time frame for diversification among the forms. These estimated lineage separation dates support the idea that geological events in ‘Beringia’ and the surrounding trans-Arctic area (e.g. opening of the Bering Seaway, Pleistocene glacial advances), occurring over a similar time frame, have influenced radiation in Osmerus. Phenetic and parsimony analyses of the sequence divergence estimates and of sequence polymorphisms suggested that the north Pacific/Arctic form and the northwestern Atlantic form shared a common ancestor more recently than either has with the north-eastern Atlantic form, thus supporting the hypothesis that the species complex has arisen from two independent Pacific-Atlantic divergences probably beginning during the mid-Pliocene.  相似文献

The interplay between structural connectivity ( i.e. habitat continuity) and functional connectivity ( i.e. dispersal probability) in marine fishes was examined in a coastal fjord (Holyrood Pond, Newfoundland, Canada) that is completely isolated from the North Atlantic Ocean for most of the year. Genetic differentiation was described in three species (rainbow smelt Osmerus mordax , white hake Urophycis tenuis and Atlantic cod Gadus morhua ) with contrasting life histories using seven to 10 microsatellite loci and a protein-coding locus, Pan I ( G. morhua ). Analysis of microsatellite differentiation indicated clear genetic differences between the fjord and coastal regions; however, the magnitude of difference was no more elevated than adjacent bays and was not enhanced by the fjord's isolation. Osmerus mordax was characterized by the highest structure overall with moderate differentiation between the fjord and St Mary's Bay ( F ST c. 0·047). In contrast, U. tenuis and G. morhua displayed weak differentiation ( F ST < 0·01). Nonetheless, these populations did demonstrate high rates (< 75%) of Bayesian self-assignment. Furthermore, elevated differentiation was observed at the Pan I locus in G. morhua between the fjord and other coastal locations. Interestingly, locus-specific genetic differentiation and expected heterozygosity were negatively associated in O. mordax , in contrast to the positive associations observed in U. tenuis and G. morhua . Gene flow in these species is apparently unencumbered by limited structural connectivity, yet the observed differentiation suggests that population structuring exists over small scales despite high dispersal potential.  相似文献

The genetic structure of Salmo dentex and its phylogenetic relations to sympatric salmonids in the Neretva and Skadar River basins were evaluated using mitochondrial DNA (mtDNA) control region, eight microsatellites, and somatolactin (SL) gene. In the Neretva River basin of Bosnia–Herzegovina, the results based on mtDNA analysis showed extensive haplotype sharing between S. marmoratus, S. dentex, and S. trutta, and were therefore not conclusive; however, F-statistics and assignment testing based on nuclear DNA markers indicated that S. dentex of the Neretva basin were grouped in a genetically unified cluster with S. marmoratus in the Neretva basin. Using the same analytical approach, S. dentex from the Skadar basin in Montenegro appeared to be genetically distinct from S. marmoratus in the same basin and indistinct from local S. trutta. Molecular data also indicated that S. dentex of the Neretva basin in Bosnia–Herzegovina are not closely related to S. dentex of the Skadar basin in Montenegro. Based on these results, we hypothesize S. dentex to be a particular life history form of S. marmoratus in the Neretva basin and of S. trutta in the Skadar basin. These results clearly demonstrate that S. dentex does not represent a monophyletic lineage and should not be considered a distinct species.  相似文献

Low rates of evolution in cnidarian mitochondrial genes such as COI and 16S rDNA have hindered molecular systematic studies in this important invertebrate group. We sequenced fragments of 3 mitochondrial protein-coding genes (NADH dehydrogenase subunits ND2, ND3 and ND6) as well as the COI-COII intergenic spacer, the longest noncoding region found in the octocoral mitochondrial genome, to determine if any of these regions contain levels of variation sufficient for reconstruction of phylogenetic relationships among genera of the anthozoan subclass Octocorallia. Within and between the soft coral families Alcyoniidae and Xeniidae, sequence divergence in the genes ND2 (539 bp), ND3 (102 bp), and ND6 (444 bp) ranged from 0.5% to 12%, with the greatest pairwise distances between the 2 families. The COI-COII intergenic spacer varied in length from 106 to 122 bp, and pairwise sequence divergence values ranged from 0% to 20.4%. Phylogenetic trees constructed using each region separately were poorly resolved. Better phylogenetic resolution was obtained in a combined analysis using all 3 protein-coding regions (1085 bp total). Although relationships among some pairs of species and genera were well supported in the combined analysis, the base of the alcyoniid family tree remained an unresolved polytomy. We conclude that variation in the NADH subunit coding regions is adequate to resolve phylogenetic relationships among families and some genera of Octocorallia, but insufficient for most species - or population-level studies. Although the COI-COII intergenic spacer exhibits greater variability than the protein-coding regions and may contain useful species-specific markers, its short length limits its phylogenetic utility.  相似文献

Multiple, sympatric morphotypes of rainbow smelt (Osmerus mordax) are known in Lake Utopia, New Brunswick. The largest, ‘giant’ form is predominantly a piscivore, the smaller, ‘dwarf’ form are predominantly planktivores, and there is an intermediate body-sized form. The forms exhibit some genetic variability, but it is body size that best defines morphotypes, trophic status, and spawning behaviour. We compared egg size, spawning date, incubation time, size at hatching, and daily and annual growth to determine when divergence in body size occurs among morphotypes. Giant form larvae hatched earlier and grew faster during their first year. Dwarf and intermediate form larvae displayed inter-annual variability in degree of overlap and divergence in growth which occurred in their first growing season or at age 1+ or 2+. We conclude that earlier hatching, early growth trajectories, and later niche shifting are linked to the persistence of morphotypes, i.e., the process is controlled by the environment and sustained to some degree by spawning segregation.  相似文献
