Native populations of Origanum vulgare L. from the Liguria andEmilia regions of northern Italy were analysed for essentialoil content and composition. Morphological characters suggestedthat the Ligurian samples belonged to the ssp. viride. The essentialoil content of inflorescences ranged from less than 5 mg g-1inthe samples from Emilia, to more than 50 mg g-1in some Liguriansamples, values similar to those reported for ssp. hirtum fromsouthern Italy. Sixty-four compounds were identified in theessential oil. The samples were allotted to three main groupson the basis of oil composition: the first group had a highcontent of components belonging to the carvacrol/thymol biosyntheticpathway; the second was characterized by a different sesquiterpenecomposition and a high linalool content; and the third, includingthe two samples of ssp. vulgare from Emilia, was characterizedby the presence of abundant sesquiterpenes. The high essentialoil content of the samples studied, and the presence of someparticular morphological characters suggest that the taxonomyof O. vulgare, especially in its western distribution area,needs further investigation. A new method is reported for the histochemical localizationof monoterpene phenols in essential oil secretory structures.The method was adapted from a spot test originally devised forin vitro detection of phenolic compounds in organic analyses.Plant subjects were the Lamiaceae species Thymus vulgaris L.,Oreganum vulgare L. and Mentha x piperita L., which accumulateessential oil in glandular trichomes. A reagent consisting of4-nitrosophenol in conc. H2SO4was applied to sample leaves ofeach species. A positive test for phenol was indicated by theproduction of coloured indophenols. Using this method, monoterpenephenols were identified in the trichomes of T. vulgaris(thymol)and O. vulgare(carvacrol), indicated by colour changes to redand green respectively. No phenol was detected in trichomesof M. x piperita. Results were confirmed by GC-MS analysis ofleaf volatile extracts from each species, and in vitro testswith thymol and carvacrol. The method could be used in fieldsurveys for rapid identification of potential medicinal plantsand bioactive compounds. The types of glandular hairs and their pattern of distributionon leaves of Ocimum basilicum L. at different stages, were investigated.Their essential oil at the various stages was chemically analysed.The density of the glandular hairs appears to be very high onyoung meristematic leaves and on meristematic regions of olderleaves. When the leaf regions start to expand no new glandularhairs are produced and their density decreases. Sideritis italica (Mill.) Greuter et Burdet belongs to the Lamiaceae family and is endemic to Italy. The glandular trichomes (morphology, distribution, histochemistry, and ultrastructure) of the plant were studied for the first time, along with the chemical composition of the essential oils. Abundant non-glandular hairs and peltate (type A) and capitate (types B, C(1), and C(x)) glandular trichomes were observed both on the vegetative and reproductive organs. The histochemical procedures and the ultrastructural investigation enabled specific location of the main site of essential oil production mainly in type-A peltate hairs. Particular emphasis is given to the release mechanism of the secreted material in all of the types of glands, and the potential taxonomic value of the indumentum in the Lamiaceae family is briefly discussed. Essential oils were hydrodistilled from flowering aerial parts of S. italica, and 136 compounds (112 in flowerheads, 79 in vegetative parts) were identified. The quantitative prevalence of diterpenoids (43.4% in flowerheads and 22.3% in vegetative parts) was the most significant characteristic of the essential oil of S. italica that could be classified as a diterpene-rich essential oil according to the classification of Kirimer.  相似文献

The terpenoid composition of the essential oils of 17 different populations of Origanum vulgare L., collected from wild populations and subsequently grown under similar conditions in the sub‐temperate region of the Western Himalaya, was studied. Analysis by GC (RI) and GC/MS allowed the identification of 51 components, representing 90.15 to 99.94% of the total oil. The two classes of the phenolic compounds and the monoterpenoids were predominant in all the essential oils. On the basis of the major constituents, i.e., marker compounds, and by comparison of the results with previous reports, new chemotypes could be identified. Principal component analysis was performed to determine the chemical variability within the different populations of O. vulgare collected and grown under similar conditions. Based on the marker compounds, six chemotypes with significant variations in their terpenoid profile were noticed within the 17 populations.  相似文献

The structure, site and histochemistry of glandular hairs onthe vegetative and reproductive parts ofSalvia officinalis wereinvestigated by UV and conventional light microscopy and byscanning electron microscopy. Five distinct types of glandularhair (one peltate and four capitate) with different sites, secretorymodes and secretions, were identified, and a functional rolepostulated for each type. All the hair types show mixed secretions,i.e., hydrophilic and lipophilic, except type I capitate hairs,which have hydrophilic secretions only. In peltate hairs andin type II capitate hairs hydrophilic secretion prevails; inthe remaining types, lipophilic secretion dominates. Origanum vulgare L. subsp. hirtum (Link) Ietsw. is a polymorphic taxon with respect to essential oil production and glandular trichome density. Here it is examined whether the natural populations that are indigenous in continental Greece may be considered as different ecotypes (i.e. populations with different genetic variation) and whether evidence regarding a fitness cost from essential oil production could be obtained. Samples from 30 different natural populations, differing in climatic conditions, were collected across continental Greece in 1998–2000 and were studied for essential oil production and glandular trichome density. Additionally, individual plants, derived from propagated rhizomes, originated from the natural populations, were transplanted in pots in two gardens that varied in terms of mean temperature and aridity. In these experimental populations, the above-ground phytomass and ratio of leaves and inflorescences to above-ground phytomass were additionally measured. The main findings of the study were (a) a strong positive correlation between essential oil concentration and glandular trichome density, (b) a high positive correlation between natural and experimental populations in essential oil concentration (or glandular trichome density), (c) a negative correlation between essential oil concentration and phytomass and (d) a positive relationship between the essential oil concentration and the factors thermal status and summer drought of the location of origin. Results suggest that the examined populations of O. vulgare subsp. hirtum are different ecotypes, the production of essential oils has a cost to plant growth and that the prime target of selection, by producing these oils, is to increase the resistance to drought and heat stresses.  相似文献

Anatomical and histological investigations of the secretoryhairs ofSalvia aurea leaves, and identification of the maincomponents of the essential oil were carried out. Two typesof glandular trichome were found: peltate glands, characterizedby a short stalk and a large six to eight-celled head, and capitatetrichomes which were further subdivided into two kinds, thefirst with a short monocellular stalk and two-cellular head(type I), and the second with a multicellular stalk, a neckcell and a small globose unicellular head (type II). Whereaspeltate glands and type I capitate trichomes were always present,type II capitate glands were not found in all leaf samples.The histochemical study suggested an ‘endodermal’role for the stalk cell (peltate and capitate type I) as wellas for the neck cell (capitate type II), preventing the lossof essential oil. Histological reactions also revealed the complexnature of the material secreted by all types ofS. aurea trichome,including polysaccharides, polyphenols and proteins, in additionto the essential oil. Qualitative and quantitative GC-MS analysisof the essential oil revealed camphor to be the main constituent.The findings are discussed in relation to studies of trichomesfrom other members of the Lamiaceae. The types of glandular trichomes and their distribution on leavesand flowers of Plectranthus ornatus were investigated at differentstages of their development. Five morphological types of glandulartrichomes are described. Peltate trichomes, confined to theleaf abaxial surface, have, in vivo, an uncommon but characteristicorange to brownish colour. Capitate trichomes, uniformly distributedon both leaf surfaces, are divided into two types accordingto their structure and secretory processes. In long-stalkedcapitate trichomes, a heterogeneous secretion (a gumresin) isstored temporarily in a large subcuticular space, being releasedby cuticle rupture, whereas, in the short-stalked capitate trichomes,the secretion, mainly polysaccharidic, is probably exuded viamicropores. On the leaves, digitiform trichomes, which do notshow a clear distinction between the apical glandular cell andthe subsidiary cells, occur with a similar distribution to thecapitate trichomes. They do not develop a subcuticular space,and secrete small amounts of essential oils in association withpolysaccharides. The reproductive organs, particularly the calyxand corolla, exhibit, in addition to the reported trichomes,unusual conoidal trichomes with long unicellular conical heads.A large apical pore, formed by tip disruption, releases thesecretion (a gumresin) stored in a rostrum-like projection.On the stamens and carpels, digitiform, capitate and conoidaltrichomes are absent, but peltate trichomes are numerous. Theyoccur between the two anther lobes, on the basal portion ofthe style, and between the four lobes of the ovary. The resultspresented are compared with those of other studies on Lamiaceaeglandular trichomes. Satureja montana and S. subspicata are used as spice, pepper substitute, for preparing tea, juice, and as a medicine. Fourteen populations (seven per species) of Satureja montana L. and S. subspicata Vis . growing in Croatia were examined to determine the chemical composition of the essential oil (analyzed by GC‐FID and GC/MS), the content of macroelements (Na, K, Ca, Mg) and trace elements (B, Fe, Cu, Mn, Zn, Al, Pb, Cr, Cd, Ni, Hg, As) analyzed by ICP‐AES, antioxidant compounds (analyzed by UV/VIS spectrophotometer), and the types and distribution of trichomes (analyzed by scanning electron microscopy). The main constituents of the essential oil were carvacrol and thymol in S. montana (all populations belong to one phenol chemotype), while α‐eudesmol, β‐eudesmol, and spathulenol dominated in S. subspicata (three chemotypes could be distinguished). Both species possess considerably higher quantities of Ca and Mg, and moderate concentrations of K and Na, while Hg and As levels were below the limit of quantification. The chemical composition of the essential oils of Lippia chevalieri and Lippia multiflora obtained from the air-dried leaves by hydrodistillation were analysed using GC and GC-MS. L. chevalieri and L. multiflora belonged to thymol/p-cymene/2-phenyl ethyl propionate and thymol/p-cymene/thymyl acetate chemotypes, respectively. The essential oils were also tested against 09 strains using a broth microdilution method. The Gram-negative bacteria were the most sensitive. The essential oil of L. multiflora was the most active.  相似文献

In this study the glandular hair morphology, chemical composition and antimicrobial activity of the essential oils from three Ocimum species have been investigated (Ocimum basilicum L. var. Genovese, O. gratissimum and O. tenuiflorum). The indumentum shows little variation among the investigated species with both glandular and non-glandular hairs presents. Glandular hairs on the three species are peltate and capitate (with various cell numbers in the stalk and gland). The samples of essential oils obtained from the plant aerial organs by hydrodistillation have been analyzed by GC-MS. Linalool (65.38%, 74.22%, 38.60%), eugenol (5.26%, 3.47%, 10.20%) and tau-cadinol (8.18%, 3.47%, 10.20%) appear as the main components in Ocimum basilicum L. var. Genovese, O. gratissimum and O. tenuiflorum. The oils also contain lower levels of α-bergamotene, 1,8-cineole, germacrene D, β-ocimene, α-caryophyllene, camphor, and α-guaiene. All essential oils showed antibacterial activity against Staphylococcus aureus and Escherichia coli depending on their concentration. Ocimum basilicum L. var. Genovese oil produced the strongest antibacterial effect on S. aureus and E. coli.  相似文献

Turkish oregano (Origanum onites L.) is a major aromatic plant that belongs to Labiatae family. In this work fourteen Turkish oregano clones have been characterized chemically and genetically. Essential oil obtained by hydro-distillation was characterized by gas chromatography. Genetic variation was determined by use of random amplified polymorphic DNA (RAPD) markers. Variation of essential oils in the selected clones was subjected to cluster analysis, and two chemotypes, carvacrol and thymol, were identified. Carvacrol was found to be the main component in all clones except clone-661, the main component of which was thymol. The oregano clones were divided into three main groups by clustering on the basis of RAPD markers. Genetic similarity values among the oregano clones ranged between 0.49 and 0.73 which was indicative of a low level of genetic variation. Clones originating from locations close to each other had similar RAPD markers. Correlation analysis of the genetic distance matrix and the Euclidian distance matrix revealed no significant correlation between them. The results also indicated that there is no relationship between genetic structure of the selected clones and essential oil composition.  相似文献

Plants of Origanum vulgare L. ssp. vulgare from the Veneto region (NE Italy) were selected to study the variability of the essential‐oil composition from leaves and inflorescences throughout an elevation gradient. We investigated also the morphology of non‐glandular and glandular trichomes, their distribution on the vegetative and reproductive organs, as well as the histochemistry of the secreted products, with special focus on the terpenoidic fraction. Since glandular trichomes are prerequisite for the essential‐oil synthesis, the second objective was to establish whether its production is related to glandular hair number, and density. Essential‐oil contents decline with increasing altitude, and the micromorphological observations revealed a decrease in trichome density along the same direction. Moreover, GC/MS analysis together with principal component analysis (PCA) showed that the three investigated populations were significantly different in chemical composition. Therefore, an important interpopulation variability for low‐, mid‐, and high‐altitude sites was established, suggesting the likely occurrence of different biotypes associated with altitudinal levels. Hence, the involvement of abiotic factors such as temperature and drought in the chemical polymorphism of O. vulgare associated with elevation is briefly discussed.  相似文献

The types of glandular trichomes, their ontogeny and patternof distribution on the vegetative and reproductive organs ofLeonotis leonurus at different stages of development, are studiedby light and scanning electron microscopy. Two morphologicallydistinct types of glandular trichomes (peltate and capitate)are described. Peltate trichomes, at the time of secretion,are characterized by a short stalk, which is connected witha large spherical head composed of eight cells in a single layer.Capitate trichomes can be divided into various types. Generally,they consist of a four-celled head supported by one or threestalk cells. The two kinds of trichomes differ in the secretionprocess. In the peltate trichomes, the secretory product seemsto remain accumulated in a subcuticular space, unless an externalfactor damages it. In the capitate trichomes, this product probablybecomes released through micropores. Sideritis syriaca ssp. syriaca is a taxon with a low essential oil content. Its leaves bear glandular trichomes of two types: long hairs with a 4-celled head, a 4-celled stalk and a 4-celled foot (reported for the first time in Lamiaceae) and short hairs with a 4–celled head, a unicellular stalk and a unicellular foot. The second type is considered intermediate between the capitate and peltate hairs, common in Lamiaceae, but found in S. syriaca ssp. syriaca. The ontogeny of the trichome types is described. The possible significance of the glandular trichome structure to Lamiaceae systematics is further discussed.  相似文献

Thymus pulegioides plants were collected from various natural habitats of Lithuania and transferred into a new uniform environment. The plants were cloned annually at controlled conditions and their essential oil composition was monitored by capillary GC and GC/MS. The geraniol/geranial/neral (G/G/N), thymol (T), linalool (L), carvacrol/γ-terpinene/p-cymene (C/γT/pC) and thymol/carvacrol/γ-terpinene/p-cymene (T/C/γT/pC) chemotypes of T. pulegioides were studied. It was found that according to the stability of essential oil composition on a sudden change of environmental conditions two types of T. pulegioides can be distinguished: (1) plants which preserve their chemical composition of the essential oils; (2) plants, which considerably change their chemical composition of the essential oils.  相似文献

The aim of the present study was to determine the bioactive compounds in four essential oils (EO’s) from Origanum heracleoticum, Origanum vulgare, Thymus vulgaris and Thymus serpyllum and to assess their antimicrobial and anti-biofilm activity against Salmonella Enteritidis. Strains were previously characterized depending on the expression of the extracellular matrix components cellulose and curli fimbriae as rdar (red, dry and rough) and bdar morphotype (brown, dry and rough). This study revealed that the EO’s and EOC’s (carvacrol and thymol) investigated showed inhibition of biofilm formation at sub-minimum inhibitory concentration. Comparing the efficacy of EO’s and EOC’s in the inhibition of biofilm formation between the strains with different morphotype (rdar and bdar) did not show a statistically significant difference. Results related to the effectiveness of EO’s and EOC’s (the essential oil components, carvacrol and thymol) on eradication of preformed 48?h old biofilms indicated that biofilm reduction occurred in a dose-dependent manner over time.  相似文献

The chemical composition of the essential oils of Origanum vulgare ssp. hirtum, growing wild in three different localities in the Southern Apennines, was studied by GC‐FID and GC/MS analyses. In total, 103 compounds were identified. The oils were mainly composed of phenolic compounds and all oils belonged to the chemotype carvacrol/thymol. The three essential oils were evaluated for their in vitro phytotoxic activity by determining their influence on the germination and initial radicle elongation of Sinapis arvensis L., Phalaris canariensis L., Lepidium sativum L., and Raphanus sativus L. The seed germination and radicle growth were affected in various degrees. Moreover, the antifungal activity of the three essential oils was assayed against three species causing pre‐ and postharvest fruit decay (Monilinia laxa, M. fructigena, and M. fructicola). At 1000 ppm, the three oils completely inhibited fungal growth. The hemolytic activity of the oils was assayed and showed no effect on the cell membranes of bovine erythrocytes.  相似文献

对羽叶薰衣草(LavandulapinnataL.)茎和叶上两种表皮毛(腺毛和非腺毛)发育的解剖学观察表明,两者的发生都源于茎或叶的原表皮细胞,但外部形态、发育过程及功能明显不同。腺毛有头状腺毛和盾状腺毛两种类型,均由1个基细胞、1个柄细胞和头部细胞构成。头状腺毛的头部只有1个或2个分泌细胞,盾状腺毛由8个分泌细胞构成头部。非腺毛由3-20个细胞组成,可分为三种类型:单列不分枝、二叉分枝和三叉及三叉以上多分枝的树状分枝。非腺毛的顶部细胞由基部到顶部逐渐变细,先端成尖形。腺毛发育由原表皮细胞经两次平周分裂形成,由于柄细胞和头部细胞所处的分化状态不同而发育成两类腺毛。非腺毛由非腺毛原始细胞经二次或多次平周分裂和不均等分裂,再发育成数个至二十多个子细胞。  相似文献
