To assess the generality of our previous finding (Gao and Macagno, 1987) that segmental homologues play a role in the establishment of the pattern of axonal projections of the heart accessory HA neurons, we have extended our studies to two other identified leech neurons: the anterior pagoda (AP) neurons and the annulus erector (AE) motor neurons. Bilateral pairs of AP neurons are found in the first through the twentieth segmental ganglia (SG1 through SG20) of the leech ventral nerve cord. All AP neurons initially extend axonal projections to the contralateral periphery as well as longitudinal projections along the contralateral interganglionic connective nerves toward anterior and posterior neighboring ganglia. Although the peripheral projections are maintained by all AP neurons throughout the life of the animal, the longitudinal projections disappear in all but two segments: the AP neurons in SG1 maintain their anterior projections and extend them into the head ganglion, and those in SG20 maintain their posterior projections and extend them into SG21 and the tail ganglion. When single AP neurons are deleted anywhere along the nerve cord before processes begin to atrophy, however, the longitudinal projections are retained by their ipsilateral homologues in adjacent ganglia. The rescued processes appear to take over the projections of the deleted neurons. In cases where two or more AP neurons on the same side of the nerve cord are deleted from adjacent ganglia, a contralateral homologue sometimes extends projections to the periphery ipsilaterally or on both sides. We obtained similar results when we deleted single AE neurons from midbody ganglia. Thus, our experiments with three different identified neurons consistently show that the initial pattern of projections is the same in all ganglia, but that the existence of homologues in adjacent ganglia leads to the pruning of some of the initial projections. A consequence of this homologue-dependent process retraction is that neurons normally lacking neighboring homologues will have patterns of projections different from those neurons that do have such neighbors. Process loss by the HA, AP, and AE neurons may be the result either of competition for targets, inputs, or growth factors or of direct interactions among homologous cells.  相似文献

Transplantation of embryonic or stem cell derived neurons has been proposed as a potential therapy for several neurological diseases. Previous studies reported that transplanted embryonic neurons extended long-distance projections through the adult brain exclusively to appropriate targets. We transplanted E14 lateral ganglionic eminence (LGE) and E15 cortical precursors from embryonic mice into the intact adult brain and analyzed the projections formed by transplanted neurons. In contrast to previous studies, we found that transplanted embryonic neurons formed distinct long-distance projections to both appropriate and ectopic targets. LGE neurons transplanted into the adult striatum formed projections not only to the substantia nigra, a normal target, but also to the claustrum and through all layers of fronto-orbital cortex, regions that do not normally receive striatal input. In some cases, inappropriate projections outnumbered appropriate projections. To examine the relationship between the donor cells and host brain in establishing the pattern of projections, we transplanted cortical precursors into the adult striatum. Despite their heterotopic location, cortical precursors not only predominantly formed projections appropriate for cortical neurons, but they also formed projections to inappropriate targets. Transplantation of GFP-expressing cells into beta-galactosidase-expressing mice confirmed that the axonal projections were not created by the fusion of donor and host cells. These results suggest that repairing the brain using transplantation may be more complicated than previously expected, because exuberant ectopic projections could result in brain dysfunction. Understanding the signals regulating axonal extension in the adult brain will be necessary to harness stem cells or embryonic neurons for effective neuronal-replacement therapies.  相似文献

The role of homologues in the establishment of the pattern of axonal projections of identified segmentally homologous neurons was investigated by means of selective cell ablation and dye injection. The cells studied were the bilateral pairs of heart accessory (HA) neurons found in the fifth and sixth segmental ganglia of the leech ventral nerve cord. Homologues start their morphological differentiation with identical axonal projections, and segmental differences are manifested later, when specific branches stop growing and disappear. The deletion of single HA cells at early stages, however, permits these branches to survive in their ipsilateral homologues and to grow and take over the projections of the deleted neurons. In addition, if both HA homologues on the same side of the nerve cord, or three of the four HA cells, are deleted in an animal, the remaining HA cells often extend novel projections. These observations suggest that either competition for targets, inputs or growth factors, or direct interactions among homologous cells may play a role in the differentiation of segment specific patterns of axonal projections.  相似文献

Vestibular nucleus neurons projecting to the cat bulbar lateral reticular nucleus were revealed using horseradish peroxidase axonal transport techniques. Neurons giving rise to such projections — relatively few in number — were confined to homolateral locations and nearly all occurred within Deiter's nucleus. Large as well as small and medium-sized neurons of the vestibular nucleus projected to the lateral reticular nucleus. We have generated mice homozygous for a mutation that disrupts the gene encoding EphA8, a member of the Eph family of tyrosine protein kinase receptors, previously known as Eek. These mice develop to term, are fertile and do not display obvious anatomical or physiological defects. The mouse ephA8/eek gene is expressed primarily in a rostral to caudal gradient in the developing tectum. Axonal tracing experiments have revealed that in these mutant mice, axons from a subpopulation of tectal neurons located in the superficial layers of the superior colliculus do not reach targets located in the contralateral inferior colliculus. Moreover, ephA8/eek null animals display an aberrant ipsilateral axonal tract that projects to the ventral region of the cervical spinal cord. Retrograde labeling revealed that these abnormal projections originate from a small subpopulation of superior colliculus neurons that normally express the ephA8/eek gene. These results suggest that EphA8/Eek receptors play a role in axonal pathfinding during development of the mammalian nervous system.  相似文献

Treatment of cultured vertebrate neurons with nitric oxide leads to growth-cone collapse, axon retraction and the reconfiguration of axonal microtubules. We show that the light chain of microtubule-associated protein (MAP) 1B is a substrate for S-nitrosylation in vivo, in cultured cells and in vitro. S-nitrosylation occurs at Cys 2457 in the COOH terminus. Nitrosylation of MAP1B leads to enhanced interaction with microtubules and correlates with the inhibition of neuroblastoma cell differentiation. We further show, in dorsal root ganglion neurons, that MAP1B is necessary for neuronal nitric oxide synthase control of growth-cone size, growth-cone collapse and axon retraction. These results reveal an S-nitrosylation-dependent signal-transduction pathway that is involved in regulation of the axonal cytoskeleton and identify MAP1B as a major component of this pathway. We propose that MAP1B acts by inhibiting a microtubule- and dynein-based mechanism that normally prevents axon retraction.  相似文献

It was shown by the method of retrograde axonal transport of horseradish peroxidase that the posterolateral thalamic nucleus (NPL) in rats receives considerable ascending projections from the superior colliculus (SC), the dorsal part of the lateral geniculate body (LGB), and the pretectal region (PT) and smaller projections from n. ventralis posterior (VP) and n. ventralis lateralis (VL) of the thalamus, the ventral part of LGB, the zona incerta, and anterior hypothalamus. The most marked descending projections run into NPL from area 18A of the cortex and the dentate fascia of the hippocampus, whereas inputs from cortical areas 18, 20, 7, 29c, 17, and 36 are less marked. In electrophysiological experiments with peripheral stimulation of visual, auditory, and somatosensory systems, polysensory convergence and interaction between signals from these systems were studied during isolated and simultaneous presentation of heterosensory stimuli. Of 229 neurons tested, 134 (58.5%) responded to at least one of the stimuli mentioned. Among monomodal neurons (53 of 134) there were some cells which responded to visual (77.4%) and somatic (22.6%) stimulation; neurons which responded only to acoustic stimulation were not found in the nucleus. As far as polymodal neurons (81 of 134) responding to two or three sensory stimuli are concerned, the most effective inputs of these units were visual and somatosensory. Interaction between stimuli acting on polymodal neurons was expressed as mutual inhibition or facilitation of responses; opposite effects could be observed on the various components of these responses.I. Rhodamine-coated microspheres (RCMs) were injected into the primary somatosensory cortex (SI) of rats ranging in age from postnatal (PN) day 1 to adulthood. Ipsilateral corticocortical and callosal projections within the SI were identified as early as PN day 1. At the end of the first PN week, ipsilaterally projecting neurons located in sublayer VIb were the first to assume an adult-like pattern of connectivity. Injections at subsequent postnatal ages revealed that an adult pattern of lamination of ipsilateral corticocortical projections within the SI is established between PN weeks 2 and 3, comprising projection neurons from layers II/III, layer V, and sublayer VIb. Therefore, local interactions in the rat SI are mediated not only by pyramidal neurons of layers III and V, derived from the cortical plate, but also by a subpopulation of ontogenetically older neurons located in the sublayer VIb, which may correspond to the subplate neurons of other species. Overall, these results suggest the existence of three independent short-range corticocortical systems of projections within the rat SI, which differ in terms of the laminar distribution and ontogenetic origin of their cells.  相似文献

Efferent projections of the lateral septal nucleus (LS) to the preoptic area and the hypothalamus were identified in 20 female guinea pigs after iontophoretic injection of the anterograde axonal tracer Fluoro-Ruby. Tubero-infundibular (TI) neurons of the preoptic area and the hypothalamus were retrogradely labeled after intracardiac injection of Granular Blue or Fluoro-Gold. Magnocellular neurons of the supraoptic and paraventricular nuclei were also labeled. The double labeling procedure allowed an estimation of the extent of the direct relationship between LS efferents and TI neurons. Contacts between lateral septal fibers and TI cell bodies were mainly observed at the light-microscopical level in the preoptic area. A group of labeled fibers coursing along the third ventricle established sparse connections with hypothalamic periventricular TI neurons. A few appositions was observed in the infundibular (arcuate) nucleus, suggestive of a monosynaptic regulation of TI neurons by a septo-arcuate tract. Close association with labeled magnocellular neurons was also noted at the edge of the supraoptic and paraventricular nuclei. The sparse but direct connections between LS and TI neurons may be involved in the neuroendocrine functions of the LS.  相似文献

The formation of topographic neural maps relies on the coordinate assignment of neuronal cell body position and axonal trajectory. The projection of motor neurons of the lateral motor column (LMC) along the dorsoventral axis of the limb mesenchyme constitutes a simple topographic map that is organized in a binary manner. We show that LIM homeodomain proteins establish motor neuron topography by coordinating the mediolateral settling position of motor neurons within the LMC with the dorsoventral selection of axon pathways in the limb. These topographic projections are established, in part, through LIM homeodomain protein control of EphA receptors and ephrin-A ligands in motor neurons and limb mesenchymal cells.  相似文献

In female rats, sexual behavior requires the convergence of ovarian hormone signals, namely estradiol and progesterone, and sensory cues from the male on a motor output pathway. Estrogen and progestin receptors (ER and PR) are found in neurons in the hypothalamic ventromedial nucleus (VMH), a brain region necessary for lordosis, the stereotypic female copulatory posture. A subset of VMH neurons sends axonal projections to the periaqueductal gray (PAG) to initiate a motor output relay, and some of these projection neurons express PR. Previous studies showed that VMH neurons are activated during mating, based on the expression of the immediate early gene Fos. Many of the activated neurons expressed ER; however, it is not known if such activated neurons co-express PR. Fluorogold, a retrograde tracer, was injected into the PAG of ovariectomized rats to label neurons projecting from the VMH. Hormone-treated animals then were mated, and their brains were immunohistochemically stained for PR and Fos. Of the Fos-positive neurons, 33% were double-labeled for PR, 19% were double-labeled with Fluorogold, and 5% were triple-labeled for Fos, PR, and the retrograde tracer. The majority of triple-labeled neurons were found in the rostral, rather than caudal, portion of the VMH. These results show that PR-containing neurons are engaged during sexual behavior, which suggests that these neurons are the loci of hormonal-sensory convergence and hormonal-motor integration.  相似文献

Axons of olfactory sensory neurons expressing a given odorant receptor converge to a few glomeruli in the olfactory bulb. We have generated mice with unresponsive olfactory sensory neurons by targeted mutagenesis of a cyclic nucleotide-gated channel subunit gene, OCNC1. When these anosmic mice were crossed with mice in which neurons expressing a given odorant receptor can be visualized by coexpression of an axonal marker, the pattern of convergence was affected for one but not another receptor. In a novel paradigm, termed monoallelic deprivation, axons from channel positive or negative neurons that express the same odorant receptor segregate into distinct glomeruli within the same bulb. Thus, the peripheral olfactory projections are in part influenced by mechanisms that depend on neuronal activity.  相似文献

In adult mammals, the severing of the optic nerve near the eye is followed by a loss of retinal ganglion cells (RGCs) and a failure of axons to regrow into the brain. Experimental manipulations of the non-neuronal environment of injured RGCs enhance neuronal survival and make possible a lengthy axonal regeneration that restores functional connections with the superior colliculus. These effects suggest that injured nerve cells in the mature central nervous system (CNS) are strongly influenced by interactions with components of their immediate environment as well as their targets. Under these conditions, injured CNS neurons can express capacities for growth and differentiation that resemble those of normally developing neurons. An understanding of this regeneration in the context of the cellular and molecular events that influence the interactions of axonal growth cones with their non-neuronal substrates and neuronal targets should help in the further elucidation of the capacities of neuronal systems to recover from injury.  相似文献

What causes motor neurons to project into the periphery is not well understood. We here show that forced expression of the homeodomain protein Phox2b, shown previously to be necessary and sufficient for branchio-visceromotor neuron development, and of its paralogue Phox2a imposes a branchiomotor-like axonal phenotype in the spinal cord. Many Phox2-transfected neurons, whose axons would normally stay within the confines of the neural tube, now project into the periphery. Once outside the neural tube, a fraction of the ectopic axons join the spinal accessory nerve, a branchiomotor nerve which, as shown here, does not develop in the absence of Phox2b. Explant studies show that the axons of Phox2-transfected neurons need attractive cues to leave the neural tube and that their outgrowth is promoted by tissues, to which branchio-visceromotor fibers normally grow. Hence, Phox2 expression is a key step in determining the peripheral axonal phenotype and thus the decision to stay within the neural tube or to project out of it.  相似文献

We used confocal microscopy in conjunction with specific antibodies and enhancer trap strains to investigate the development of specific neuronal connections in a simple model system, the larval visual system of Drosophila. We find that the establishment of axonal projections from the larval photoreceptor neurons to their central nervous system targets involves a series of discrete steps. During embryogenesis, the larval optic nerve contacts several different cell types, including optic lobe pioneer (OLP) neurons and a number of glial cells. We demonstrate that OLP neurons are present and project normally in glass (gl) mutant embryos in which the larval optic nerve fails to develop, suggesting that they do not depend on interactions with the larval optic nerve for differentiation and proper axonal projection. The OLPs fail to differentiate properly in disconnected (disco) mutant embryos, where appropriate connections between the larval optic nerve and its targets in the brain are not formed. The disco gene is expressed in the OLPs and may therefore act autonomously to direct the differentiation of these cells. Taken together, our results suggest that the OLPs act as an intermediate target required for the establishment of normal optic nerve projection and connectivity. © 1995 John Wiley & Sons, Inc.  相似文献

Previous studies have indicated that the formation of stereotyped segmental nerves in leech embryos depends on the interactions between CNS projections and ingrowing afferents from peripheral neurons. Especially, CNS-ablation experiments have suggested that CNS-derived guidance cues are required for the correct navigation of several groups of peripheral sensory neurons. In order to directly test this hypothesis we have performed transplantations of CNS ganglia into ectopic sites in segments from which the resident ganglia have been removed. We find that the transplanted ganglia extend numerous axons distributed roughly equally in all directions. When these CNS projections reach and make contact with peripheral sensory axons they are used as guides for peripheral neurons to grow toward and into the ectopic ganglia even when this means following novel pathways that cross the midline and/or segmental boundaries. The peripheral sensory axons turn and grow toward the ectopic ganglia only when in physical contact with CNS axons, suggesting that diffusible chemoattractants are not a factor. These results demonstrate that the guidance cues provided by ectopic CNS projections are both necessary and sufficient to steer peripheral sensory neuron axons into the CNS.  相似文献

The location of sources of direct projections to the hypothalamic locomotor region, electrical stimulation of which in the lightly anesthetized animal induced stepping along a moving treadmill, was studied by the retrograde axonal transport of horseradish peroxidase method in the cat brain stem. Different formations in the brain stem were shown to have direct connections with hypothalamic locomotor regions on both sides. Most sources of these afferent projections were located at sites of catecholamine- (nucl. reticularis lateralis, locus coeruleus, nucl. tractus solitarii) and serotonin-containing (nucl. raphe and substantia grisea centralis) neurons, parabrachial nuclei, and various sensory nuclei.
