The effect of low doses of UV-A (320–400 nm) and UV-B (280–320 nm) radiation on photosynthetic activities inPhaseolus mungo L. was investigated under field condition. Supplementation of UV-A enhanced the synthesis of chlorophyll and carotenoids than the UV-B supplemented plants. Significant increase was seen in the concentration of UV-B absorbing compounds of UV-B treated plants. Increase of PS 2 activity in UV-A treated plants was seen. Changes in photosynthetic activity were measured in terms of PS 2 mediated O2 evolution and Chl a fluorescence.  相似文献

Sterilized seeds of Isatis indigotica (Brassicacae) were divided into four groups based on irradiation pretreatments. These control groups (C) were non irradiated, He–Ne laser treated seeds (L), UV-B treated seeds (B) and He–Ne laser followed by UV-B radiation treated seeds (LB). Laser radiation was provided by He–Ne laser, UV-B radiation was provided by filtered Qin brand 30 W fluorescent sun lamps. Malondialdehyde (MDA), proline, UV-B absorbing compounds and ascorbic acid (AsA) concentrations, as well as, the activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) were measured in the cotyledons of seedlings from all the four irradiation treatments. The result indicate that UV-B radiation enhanced the concentration of MDA while decreasing the activities of SOD, CAT, POD and the concentration of AsA in the seedlings compared with the controls. The concentration of MDA decreased, while the activities of SOD, CAT, POD and the concentration of AsA increased in seedling treated with He–Ne laser and UV-B compared to UV-B alone. The concentration of proline and UV absorbing compounds increased progressively with treatments i.e. UV-B irradiation, He–Ne laser irradiation, and He–Ne laser irradiation followed by UV-B irradiation compared to the controls. The present data suggest that Isatis indigotica seedlings derived from laser stimulated seeds showed improved resistance to elevated UV-B.  相似文献

Five different doses of ultraviolet-B (UV-B) radiation were supplied to tomato (Lycopersicon esculeutum. Mill) with the doubled CO2 concentration (700 μmol · mol−1) in the winter plastic greenhouse. The influences on the seedling growth, fruit quality and yield of tomato were investigated. Results showed that the seedling growth, and the contents of UV absorbing compounds, soluble sugar, organic acid, vitamin C and lycopene of tomato fruits, and yield of tomato increased under doubled CO2 concentration. Under the doubled CO2 concentration the effects of lost doses of UV-B radiation could further promote the effects of doubled CO2 concentration. However, there is no significant increase in yield of tomato. The best dose of UV-B radiation is about 1.163 kJ·m−2. We used solvent extractions, SiO2 column chromatographies, and HPLC to isolate from the bark of Thanakha (Hesperethusa crenulata L) an active crystalline compound for absorbing UV-A radiation (320 to 380 nm). Analyses of low-and high-resolution FAB-MS revealed a compound, named marmesin, with a formula of C14H14O4 and a molecular mass of 246. To determine its chemical structure, we conducted 300 MMz NMR analyses using various probes,1H,13C, and DEPT13C. Our NMR data showed a structure of 2,3-dihydro-2(1 -hydroxy-1 -methylethyl)-furanocoumarin. This active compound contains UV-absorbing chromophores, an aromatic ring, a double bond at C3-C4, and a carbonyl at C2. Its λmax is 335 nm, indicating that marmesin could be commercially useful as a natural UV-A-filtering product  相似文献

The marine algal species in the Baltic Sea are few due to the low sea water salinity. One of the few species that can be found is Fucus vesiculosus. Even this species is affected by the low salinity and becomes smaller in size in the Baltic. In present work the photosynthesis of F. vesiculosus in the northern Baltic (Bothnian Sea) was compared to the photosynthesis of F. vesiculosus in the Atlantic. Oxygen evolution was measured before and after exposure to 2.3 W of UV-B (280–320 nm) radiation for 5 h, as well as after 48 h recovery in low light. The plants were kept in their own sea water salinity as well as in a changed salinity, this to examine possible correlations between salinity and photosynthesis. The results show a significant higher initial maximal photosynthesis (P max) for Atlantic plants (10.3 nmol O2 g−1 FW s−1) compared to Baltic plants (4.0 nmol O2 g−1 FW s−1). The Baltic plants were found more sensitive to UV-B with a 40–50% decrease of P max as well as a lower degree of recovery (60–70% compared to 75–95% for the Atlantic plants). The higher salinity (35 psu) had a positive effect on the Baltic F. vesiculosus with increased P max as well as increased tolerance to UV-B. The lower salinity (5 psu) had a negative effect on the Atlantic plants with a decreased P max as well as a lower tolerance to UV-B. Pigment content was found higher in Atlantic F. vesiculosus. The pigment content decreased then the Atlantic plants were transferred to 5 psu. The concentration of Chl a as well as the total content of violaxanthin, diadinoxanthin and zeaxanthin in Baltic plants increased when transferred to 35 psu. The Atlantic F. vesiculosus can not survive the low salinity in the northern Baltic (died within 8 weeks). It is likely that a long time acclimation or adaptation to low salinity has taken place for F. vesiculosus in northern Baltic. If this is an ecotypic or genotypic development it is too early to say.  相似文献

The genome sequence of Streptomyces coelicolor A3(2) contains 51 putative lipase and esterase genes mostly of unknown function. The gene estB (locus SCO 6966) was expressed as a His-tagged protein in E. coli. Esterase B was active at low temperatures exerting its maximum activity at 30°C and retaining more than 25% of its activity at 4°C. The optimum pH was 8–8.5. The enzyme was active against short synthetic p-nitrophenylesters (C2–C10) with maximum activity towards the acetate ester (C2). The esterase was tested on 13 series of racemic esters of potential interest for the synthesis of chiral pharmaceutical compounds. 4 of the series were substrates and a modest degree of enantioselectivity was observed (enantiomeric ratios of 1.1–1.9).  相似文献

Nitrogenous excretion in two snails, Littorina saxatilis (high intertidal) and L. obtusata (low intertidal) was studied in relation to temperature acclimation (at 4° and 21°C), including total N excretion rates, the fraction of urea in N excretion, corresponding O:N ratios and the partitioning of deaminated protein between catabolic and anabolic processes at 4°, 11° and 21°C. Aggregate N excretion rates in both species showed no significant compensatory adjustments following acclimation. Total weight specific N excretion rates at 21°C were higher in standard 3 mg L. saxatilis (739 ng N mg−1 h−1) than standard 5 mg L. obtusata (257 ng N mg−1 h−1) for snails acclimated to 21°C. Comparisons of Q10 values of total weight specific N excretion to Q10 values for weight specific oxygen consumption ({xxV}O2) between 4° to 11 °C and 11° to 21°C indicated that, while total rates of catabolic metabolism ({xxV}O2) and protein deamination in L. obtusata were essentially parallel, the relationship between N excretion and {xxV}O2 in L. saxatilis revealed the partitioning of a larger share of deaminated protein carbon into anabolism at 4° and 21°C than at 11°C. Urea N accounted for a larger share of aggregate N excreted in L. saxatilis than in L. obtusata, but in both species urea N is a greater proportion of total N excreted when acclimated at 4°C (urea N: ammonia N ratio range: 1 to 2.15) than in snails acclimated to 21°C (urea N: ammonia N ratio range: 0.46 to 1.39). Molar O:N ratios indicate that the proportion of metabolism supported by protein catabolism is greater in L. saxatilis (O:N range: 2.5–8.4) than in L. obtusata (O:N range: 7.3–13.0). In both species, regardless of acclimation temperature, the O:N ratios are generally lowest (high protein catabolism) at 4°C and highest at 21°C.  相似文献

Growth of young sporophytes of the brown algae Laminaria digitata, L. saccharina and L. solidungula from Spitsbergen were measured in the laboratory after being exposed for 21 days to either photosynthetically active radiation (PAR=P) or to full light spectrum (PAR + UV-A + UV-B=PAB) using of cutoff glass filters. The plants were grown at 8±2°C and 16 h light : 8 h dark cycles with 6 h additional ultraviolet radiation (UVR) exposure in the middle of the light period. Growth was measured every 10 min using growth chambers with online video measuring technique. Tissue morphology and absorption spectra were measured in untreated young sporophytes while chlorophyll (Chl) a content and DNA damage were measured in treated thalli at the end of the experiment. In all species, growth rates were significantly higher in sporophytes exposed to P alone compared to sporophytes exposed to PAB. Tissue DNA damage is dependent on thallus thickness and absorption spectra characteristics of pigments and UV-absorbing compounds. In sporophytes exposed to UVR, energy demands for repair of DNA damage and synthesis of UV-absorbing compounds for protection effectively diverts photosynthate at the expense of growth. Photosynthetic pigment was not significantly different between treatments suggesting a capacity for acclimation to moderate UVR fluence. The general growth pattern in sporophytes exposed to P alone showed an increasing growth rate from the onset of light (0500–0900 hours) to a peak at the middle of the light phase (0900–1500 hours), a decline towards the end of the light phase (1500–2100 hours) and a minimum “low” growth in the dark (2100–0500 hours) relative to growth during the entire light phase. Under PAB, different growth patterns were observed such as growth compensation at night in L. digitata, delayed growth recovery in L. saccharina and minimal but continuous growth in L. solidungula. Growth as an integrative parameter of all physiological processes showed that the effect of UVR is correlated to the depth distribution of these species.  相似文献

The toxicity of para-menthane-3,8-diol (PMD), the main arthropod-repellent compound in the oil of the lemon eucalyptus, Corymbia citriodora, was evaluated against nymphs of Ixodes ricinus using five methods (A–E) of a contact toxicity bioassay. Mortality rates were estimated by recording numbers of dead nymphs at 30 min intervals during the first 5 h after the start of exposure and at longer intervals thereafter. The mortality rate increased with increasing concentration of PMD and duration of exposure with a distinct effect after 3.5 h. From the results obtained by methods A, C and E, the LC50 range was 0.035–0.037 mg PMD/cm2 and the LC95 range was 0.095–0.097 mg PMD/cm2 at 4 h of exposure; the LT50 range was 2.1–2.8 h and the LT95 range was 3.9–4.2 h at 0.1 mg PMD/cm2. To determine the duration of toxic activity of PMD, different concentrations (0.002, 0.01, 0.1 mg PMD/cm2) were tested and mortality was recorded at each concentration after 1 h; thereafter new ticks were tested. This test revealed that the lethal activity of PMD remained for 24 h but appeared absent after 48 h. The overall results show that PMD is toxic to nymphs of I. ricinus and may be useful for tick control.  相似文献

We examined, under laboratory conditions, the influence of temperature (2 °C vs. 10 °C) on the physiological responses of two aquatic bryophytes from a mountain stream to artificially enhanced UV-B radiation for 82 d. These organisms may be exposed naturally to relatively low temperatures and high levels of UV-B radiation, and this combination is believed to increase the adverse effects of UV-B radiation. In the moss Fontinalis antipyretica, UV-B-treated samples showed severe physiological damages, including significant decreases in chlorophyll (Chl) and carotenoid (Car) contents, Chl a/b and Chl/phaeopigment ratios, Chl a fluorescence parameters Fv/Fm and PS2, electron transport rate (ETRmax), and growth. In the liverwort Jungermannia cordifolia, UV-B radiation hardly caused any physiological change except for growth reduction. Thus, this liverwort seemed to be more tolerant to UV-B radiation than the moss under the specific experimental conditions used, maybe partly due to the accumulation of UV-B absorbing compounds. The influence of temperature on the effects of UV-B radiation depended on the species: the higher the UV-B tolerance, the lower the influence of temperature. Also, different physiological variables showed varied responses to this influence. Particularly, the lower temperature used in our study enhanced the adverse effects of UV-B radiation on important physiological variables such as Fv/Fm, growth, and Chl/phaeopigment ratios in the UV-B-sensitive F. antipyretica, but not in the more UV-B-tolerant J. cordifolia. Thus, the adverse effects of cold and UV-B radiation were apparently additive in the moss, but this additiveness was lacking in the liverwort. The Principal Components Analyses (PCA) conducted for both species with the physiological data obtained after 36 and 82 d of culture confirmed the above results. Under natural conditions, the relatively high water temperatures in summer might facilitate the acclimation of aquatic bryophytes from mountain streams to high levels of UV-B radiation. This may be relevant to predict the consequences of concomitant global warming and increasing UV-B radiation.  相似文献

In the southeast of the Qinghai-Tibetan Plateau of China, Mono Maple is a common species in reforestation processes. The paper mainly investigated the changes in morphological, photosynthetic and physiological responses of Mono Maple seedlings to UV-B radiation, nitrogen supply and their combination. The experimental design included two levels of UV-B treatments (ambient UV-B, 11.02 KJ m−2 day−1; enhanced UV-B, 14.33 KJ m−2 day−1) and two nitrogen levels (0; 20 g N m−2 a−1)—to determine whether the adverse effects of UV-B on plants are eased by nitrogen supply. Enhanced UV-B caused a marked decline in growth parameters, net photosynthetic rate, and photosynthetic pigments, whereas it induced an increase in reaction oxygen species (hydrogen peroxide accumulation and the rate of superoxide radical production) and malondialdehyde content. Enhance UV-B also induced an increase in antioxidant compounds of Mono Maple, such as UV-B absorbing compounds, proline content, and activities of antioxidant enzymes (peroxidase, superoxide dimutase and catalase). On the other hand, nitrogen supply caused an increase in some growth parameters, net photosynthetic rate, photosynthetic pigments and antioxidant compounds (peroxidase, proline content and UV-B absorbing compounds), and reduced the content of reaction oxygen species (H2O2 accumulation, the rate of O2production) and malondialdehyde content under ambient UV-B. However, under enhanced UV-B, nitrogen supply inhibited some growth parameters, and increased H2O2 accumulation, the rate of O2production and MDA content, though proline content, UV-B absorbing compounds and activities of POD and SOD increased. These results implied that enhanced UV-B brought harmful effects on Mono Maple seedlings and nitrogen supply made plants more sensitive to enhanced UV-B, though increased some antioxidant activity.  相似文献

n-Alkanes pattern in response to NaCl stress has been studied in the cyanobacterium Anabaena cylindrica. Saturated hydrocarbons were separated and identified by gas chromatography-mass spectrometry (GC-MS) using serially coupled capillary column. Light chain n-alkanes in the range of C9–C17 (43%) and heavy chain n-alkanes in range of C17–C23 (34%) and C23–C31 (23%) were identified as the major components of total hydrocarbons in the NaCl adapted cells of A. cylindrica. In contrast, NaCl-untreated cells of A. cylindrica had dominance of only long chain n-alkanes in the range of C23–C31 comprising about 94% of its total n-alkanes. The persistence of high level (43%) of short chain n-alkanes (C9–C17) in NaCl adapted cells of A. cylindrica as compared to its negligible level (0.2%) in NaCl untreated counterpart clearly indicates that NaCl stress causes the A. cylindrica to shift towards the synthesis of short chain n-alkanes.  相似文献

The effect of pre-treatment with 200 mM NaCl on the response of four barley cultivars (Hordeum vulgare L. cv. Bülbül-89, Kalaycı-97, Tarm-92 and Tokak-157/37) to UV-B radiation was investigated. Salt stress as well as UV-B irradiation led to a decrease of the total chlorophyll (chl) content in all cultivars, except in Kalaycı-97. While carotenoids are almost not affected by NaCl treatment, UV-B irradiation caused an increase by 5–20% of carotenoid content of all cultivars. UV-B induced damages of photosynthetic apparatus were estimated by the rate of photosynthetic electron transport measured by chl fluorescence and the rate of oxygen evolution, the latter being more affected. Pre-treatment with NaCl alleviated harmful effect of UV-B irradiation on F v/F m and ETR, but not on oxygen evolution. UV-B-induced and UV-B-absorbing compounds with absorption at 300 and 438 nm increased as a result of UV-B treatment. The level of stress marker proline increased considerably as a result of NaCl treatment, while UV-B irradiation resulted in a pronounced increase of the level of H2O2. MDA enhanced in the seedlings subjected to salt and UV-B stress. Established cross-acclimation to UV-B as a result of salt treatment could be due to the increased free proline and the level of UV-B absorbing compounds in barley seedlings subjected to NaCl.  相似文献

Alterations in photosynthetic capacity of primary leaves of wheat seedlings in response to ultraviolet-B (UV-B; 280–320 nm; 60 μmol m−2 s−1) exposure alone and in combination with photosynthetically active radiation (PAR; 400–800 nm; 200 μmol m−2 s−1) during different phases of leaf growth and development were assessed. UV-B exposure resulted in a phase-dependent differential loss in photosynthetic pigments, photochemical potential, photosystem 2 (PS2) quantum yield, and in vivo O2 evolution. UV-B exposure induced maximum damage to the photosynthetic apparatus during senescence phase of development. The damages were partially alleviated when UV-B exposure was accompanied by PAR. UV-B induced an enhancement in accumulation of flavonoids during all phases of development while it caused a decline in anthocyanin content during senescence. The differential changes in these parameters demonstrated the adaptation ability of leaves to UV-B stress during all phases of development and the ability was modified in UV-B+ PAR exposed samples.  相似文献

Brassica rapa L. is an important vegetable crop in eastern Asia. The objective of this study was to investigate the genetic variation in leaf Zn, Fe and Mn accumulation, Zn toxicity tolerance and Zn efficiency in B. rapa. In total 188 accessions were screened for their Zn-related characteristics in hydroponic culture. In experiment 1, mineral assays on 111 accessions grown under sufficient Zn supply (2 μM ZnSO4) revealed a variation range of 23.2–155.9 μg g−1 dry weight (d. wt.) for Zn, 60.3–350.1 μg g−1 d. wt. for Fe and 20.9–53.3 μg g−1 d. wt. for the Mn concentration in shoot. The investigation of tolerance to excessive Zn (800 μM ZnSO4) on 158 accessions, by using visual toxicity symptom parameters (TSPs), identified different levels of tolerance in B. rapa. In experiment 2, a selected sub-set of accessions from experiment 1 was characterized in more detail for their mineral accumulation and tolerance to excessive Zn supply (100 μM and 300 μM ZnSO4). In this experiment Zn tolerance (ZT) determined by relative root or shoot dry biomass varied about 2-fold. The same six accessions were also examined for Zn efficiency, determined as relative growth under 0 μM ZnSO4 compared to 2 μM ZnSO4. Zn efficiency varied 1.8-fold based on shoot dry biomass and 2.6-fold variation based on root dry biomass. Zn accumulation was strongly correlated with Mn and Fe accumulation both under sufficient and deficient Zn supply. In conclusion, there is substantial variation for Zn accumulation, Zn toxicity tolerance and Zn efficiency in Brassica rapa L., which would allow selective breeding for these traits.  相似文献

We developed 11 polymorphic microsatellite markers for an endangered shrub Ammopiptanthus mongolicus using the Fast Isolation by AFLP of Sequences Containing repeats protocol. Polymorphism of each locus was assessed in 24 individuals from five natural populations of A. mongolicus. The average allele number of these markers was 4.2, ranging from 2 to 7. The observed (H O) and expected (H E) heterozygosities were 0.044–0.956 and 0.044–0.827, respectively. Further assessment in the congeneric species A. nanus revealed that nine of eleven loci were successfully amplified, but only two loci showed polymorphism. These markers provide powerful tools for the conservation genetics studies of A. mongolicus.  相似文献

Industrial wastewaters contain various heavy metal components and therefore threaten aquatic bodies. Heavy metals can be adsorbed by living or non‐living biomass. Submerged aquatic plants can be used for the removal of heavy metals. This paper exhibits the comparison of the adsorption properties of two aquatic plants Myriophyllum spicatum and Ceratophyllum demersum for lead, zinc, and copper. The data obtained from batch studies conformed well to the Langmuir Model. Maximum adsorption capacities (qmax) were obtained for both plant species and each metal. The maximum adsorption capacities (qmax) achieved with M. spicatum were 10.37 mg/g for Cu2+, and 15.59 mg/g for Zn2+ as well as 46.49 mg/g for Pb2+ and with C. demersum they were 6.17 mg/g for Cu2+, 13.98 mg/g for Zn2+ and 44.8 mg/g for Pb2+. It was found that M. spicatum has a better adsorption capacity than C. demersum for each metal tested. Gibbs free energy and the specific surface area based on the qmax values were also determined for each metal.  相似文献

O3 concentrations in the troposphere are rising and those in the stratosphere decreasing, the latter resulting in higher fluxes of solar ultraviolet-B (UV-B) radiation to the earth's surface. We assessed whether the fluxes of CO2 and CH4 are altered by enhanced UV-B radiation or elevated tropospheric O3 concentrations in boreal peatland microcosms (core depth 40 cm, diameter 10.5 cm) with different vegetation cover. At the end of the UV-B experiment which lasted for a growing season, net CO2 exchange (NEE) and dark ecosystem respiration (R TOT) were sevenfold higher, and CH4 efflux 12-fold higher, in microcosms with intact vegetation dominated by Eriophorum vaginatum L. and Sphagnum spp., compared to microcosms from which we removed E. vaginatum. Vegetation treatment had minor effects on CH4 production and consumption potentials in the peat, suggesting that the large difference in CH4 efflux is mainly due to efficient CH4 transport via the aerenchyma of E. vaginatum. Ambient UV-B supplemented with 30% and elevated O3 concentrations (100 and 200 ppb, for 7 weeks) significantly increased R TOT in both vegetation treatments. Elevated O3 concentrations reduced NEE over time, while UV-B had no clear effects on the fluxes of CO2 or CH4 in the cloudy summer of the study. Field experiments are needed to assess the significance of increasing UV-B radiation and elevated tropospheric O3 concentration on peatland gas exchange in the long-term.  相似文献

Phytostabilization strategies may be suitable to reduce the dispersion of uranium (U) and the overall environmental risks of U-contaminated soils. The role of Glomus intraradices, an arbuscular mycorrhizal (AM) fungus, in such phytostabilization of U was investigated with a compartmented plant cultivation system facilitating the specific measurement of U uptake by roots, AM roots and extraradical hyphae of AM fungi and the measurement of U partitioning between root and shoot. A soil-filled plastic pot constituted the main root compartment (CA) which contained a plastic vial filled with U-contaminated soil amended with 0, 50 or 200 mg KH2PO4−P kg–1soil (CB). The vial was sealed by coarse or fine nylon mesh, permitting the penetration of both roots and hyphae or of just hyphae. Medicago truncatula plants grown in CA were inoculated with G. intraradices or remained uninoculated. Dry weight of shoots and roots in CA was significantly increased by G. intraradices, but was unaffected by mesh size or by P application in CB. The P amendments decreased root colonization in CB, and increased P content and dry weight of those roots. Glomus intraradices increased root U concentration and content in CA, but decreased shoot U concentrations. Root U concentrations and contents were significantly higher when only hyphae could access U inside CB than when roots could also directly access this U pool. The proportion of plant U content partitioned to shoots was decreased by root exclusion from CB and by mycorrhizas (M) in the order: no M, roots in CB > no M, no roots in CB > M, roots in CB > M, no roots in CB. Such mycorrhiza-induced retention of U in plant roots may contribute to the phytostabilization of U contaminated environments.  相似文献

The responses of the early development of Laminaria japonica collected from Kiaochow Bay in China to enhanced ultraviolet-B radiation (UV-B, 280–320 nm) were studied in the laboratory. The low UV-B radiations (11.7–23.4 J·m−2·d−1) had no significant effects on zoospores attachment, but when the UV-B dose > 35.1 J·m−2·d−1 the attachment decreased significantly compared with the control. Germination of embryospores was >93% under the low (11.7–35.1 J·m−2·d−1) doses, and in the range of 78.5%–88.5% under the high (46.8–70.2 J·m−2·d−1) UV-B doses, indicating a significant radiation effect. Under the higher UV-B exposure (35.1–70.2 J·m−2·d−1), all of the few gametophytes formed from embryospores died 120 h post-release. After exposure to the low UV-B radiation (11.7–23.4 J·m−2·d−1), the formation of sporophytes decreased and the female gametophyte clones increased compared with the control. However, the sex ratio and the relative growth of female gametophytes/sporophytes had not significantly changed. According to the results, enhanced UV-B radiation has a significant effect on the early development of L. japonica under laboratory conditions, suggesting that the UV-B radiation could not be overlooked as one of the important environmental factors influencing the ontogeny of macroalgae living in marine ecosystems.
