Abstract Different types of synaptic contacts between bipolar, amacrine and ganglion cells were scored on random electron micrographs and on montages comprising the entire thickness of the inner synaptic layer. Currently accepted criteria were used when classifying the different cell processes. The percental distribution of dyads was estimated to 56 % amacrine-amacrine dyads, 34 % amacrine-ganglion dyads and 10 % ganglion-ganglion dyads. The ratio of amacrine conventional synapses to bipolar ribbon synapses was 6.8 : 1. The density per unit area of conventional synapses (0.035/μm2) and ribbon synapses (0.005/ μm2) was found markedly low as compared with other vertebrate species except the carp. The inner synaptic layer of the river lamprey is suggested to be of the intermediate type in which both simple and complex ganglion cell receptive fields may be expected.  相似文献

During the last few years many facts have been revealed about the neural structure in the retina of several vertebrate groups, particularly with regard to the monoamine containing neurons. A number of investigations have been made on mammals, birds and teleost fishes by the Falck-Hillarp method for the cellular demonstration of such amines. Abstract The pro-adenohypophysis (pro-AH) of migrating adult river lamprey, Lampetra fluviatilis, has been examined from October, when the gonads are developing slowly, until May, when the sexual maturation is completed. Two granulated cell types are dominating. One is chromophobic (C2, 95 nm), the other is basophilic (B1-B3) and probably gonadotropic. The mean diameter of the basophil secretory granules increases from 150 nm in October to 220 nm in April and May. The staining affinity of a rare, granulated cell type (D, 115 nm) has not been established. No acidophil cells are found in the pro-AH. The ultrastructural characteristics of the lead hematoxylin positive cells found sometimes in paraffin sections are unknown. Non-glandular stellate cells are common. They contain microfilaments, are probably contractile, and transform to phagocytes during the last months before spawning. This phagocytosis involves only the basophil cells. It is suggested that the stellate cells in this way destroy excess hormone. All cell types in the pituitary accumulate large irregular lipid droplets during the last month before spawning. These lipid droplets seem to be expelled from residual bodies as an end product of autolysis and phagocytosis.  相似文献

The retinal pigment epithelium of Lampetra fluviatilis was studied by electron microscopy. The epithelial cells differ in many details from those of gnathostomes. The lateral cell membranes are difficult to distinguish. The smooth endoplasmic reticulum is well developed; in some animals undulated membrane complexes, comprising systems of tightly fused double membrane plates, are related to the endoplasmic reticulum. Myeloid bodies are common and well developed, but pigment granules are comparatively sparse. The intercellular space between pigment epithelium and photoreceptors is rather wide. There are only a few inclusion bodies with membranous contents. The importance of the pigment epithelium in the retinal metabolic exchange is discussed in view of the fine structure of the cells. Compared with that of hagfishes, the lamprey retina is well developed. However, any comparison must be made against the background of a diphyletic development of the two groups.  相似文献

Tissues were processed for fluorescence microscopy of biogenic amines according to the method of Falck and Hillarp. Normal animals, and animals injected with α-methylnoradrenaline or 5,6-dihydroxytryptamine were used. Catecholamine containing neurons (junctional cells) occur in the innermost rows of cell bodies of the inner nuclear layer (INL) and close to the vitreous surface. Catecholamine containing fibers occur in three layers: (1) an outer layer around the innermost perikarya of the INL, which is a condition not found in retinas of gnathostome chordates; (2) a middle layer within the outer third of the inner synaptic layer (ISL), separated from the outer layer by ganglion cell axons; (3) a sparse inner layer within the innermost third of the ISL. A few catecholamine containing fibers were seen to extend from the innermost region of the INL to the outer synaptic layer. The position of the junctional cells in the lamprey corresponds to that in gnathostome chordates, but whereas all catecholamine containing fiber layers in gnathostomes are located sclerally to the optic fiber layer and within the ISL, the middle and the inner fiber layers in the lamprey occur vitreally to the optic fiber layer. Indoleamine accumulating neurons occur in the innermost row of perikarya of the INL and close to the vitreous surface. Those of the INL send fine, varicose branches to the ISL forming a network which is somewhat denser at the inner and outer borders of the ISL than in its middle. The indoleamine accumulating terminals do not ramify within the INL in contrast to the catecholamine containing terminals.  相似文献

The daily phagocytosis of shed photoreceptor outer segments by pigment epithelial cells is critical for the maintenance of the retina. In a subtractive polymerase chain reaction analysis, we found that functional differentiation of human ARPE19 retinal pigment epithelial (RPE) cells is accompanied by up-regulation of annexin (anx) A2, a major Src substrate and regulator of membrane–cytoskeleton dynamics. Here, we show that anx A2 is recruited to the nascent phagocytic cup in vitro and in vivo and that it fully dissociates once the phagosome is internalized. In ARPE19 cells depleted of anx A2 by using small interfering RNA and in ANX A2−/− mice the phagocytosis of outer segments was impaired, and in ANX A2−/− mice there was an accumulation of phagocytosed outer segments in the RPE apical processes, indicative of retarded phagosome transport. We show that anx A2 is tyrosine phosphorylated at the onset of phagocytosis and that the synchronized activation of focal adhesion kinase and c-Src is abnormal in ANX A2−/− mice. These findings reveal that anx A2 is involved in the circadian regulation of outer segment phagocytosis, and they provide new insight into the protein machinery that regulates phagocytic function in RPE cells.  相似文献

A possibility of exogenous NADH oxidation via the external pathway has been shown on homogenates and isolated liver cells of the lamprey Lampetra fluviatilis in the presence of rotenone and antimycin A. The homogenates were incubated in isotonic and hypotonic sucrose media, while cells, in isotonic salt medium. At incubating the tissue preparations in isotonic media, digitonin was used to enhance membrane permeability to NADH and cytochrome c. In homogenates, the maximal rate of NADH oxidation via the external pathway in the presence of cytochrome c and digitonin was 5.3 nmol O2/min/10 mg wet weight. This value in the cells amounted to 12.6, while without addition of exogenous NADH and cytochrome c, to 11.0 nmol O2/min/10 million cells. Cyanide inhibited completely the NADH oxidation via the external pathway both in homogenates and in cells. The intact lamprey hepatocytes, unlike homogenates, are suggested to contain sufficient concentrations of cytochrome c and extramitochondrial NADH to provide maximal NADH oxidation rate in mitochondria through external pathway. This allows thinking that potential possibilities of NADH oxidation via the external pathway in Cyclostomata and mammals are qualitatively and quantitatively close.  相似文献

ABCR is an ABC transporter that is found exclusively in vertebrate photoreceptor outer segments. Mutations in the human ABCR gene are responsible for autosomal recessive Stargardt disease, the most common cause of early onset macular degeneration. In this paper we review our recent work with purified and reconstituted ABCR derived from bovine retina and from cultured cells expressing wild type or site-directed mutants of human ABCR. These experiments implicate all-trans-retinal (or Schiff base adducts between all-trans-retinal and phosphatidylethanolamine) as the transport substrate, and they reveal asymmetric roles for the two nucleotide binding domains in the transport reaction. A model for the retinal transport reaction is presented which accounts for these experimental observations.  相似文献

To study H+ transport, the lamprey red blood cells were acidified to pH 6.0 by a pretreatment with an ionophore, nigericin. Incubation of the acidified cells in NaCl-medium at pH 8.0 was accompanied by a rapid H+ efflux from the erythrocytes. There was a tenfold decrease of the H+ efflux rate on addition to NaCl-medium of dimethylamiloride or on replacing Na+ in the medium (KCl-medium, pH 8.0). A high rate of Na+ influx into the acidified erythrocytes occurred only in the presence of H+ gradient (pH medium 8.0), but not in its absence (pH medium 6.0). The Na+-dependent H+ efflux from the cells and H+-dependent Na+ influx into the cells were quantitatively similar (about 700 mmol/l cells/h). A rapid elevation of the intracellular Na+ concentration as measured by flame photometry was also observed during incubation of the acidified cells in NaCl-medium (pH 8.0). The H+-dependent Na+ influx and an increase of the Na+ content in the acidified cells were significantly inhibited by amiloride. The data obtained for the first time prove with certainty the presence of the Na+/H+ exchanger in erythrocytes of the river lamprey.  相似文献

Dark Ionic Flux and the Effects of Light in Isolated Rod Outer Segments   总被引:23,自引:9,他引:14  
We have determined the permeability properties of freshly isolated frog rod outer segments by observing their osmotic behavior in a simple continuous flow apparatus. Outer segments obtained by gently shaking a retina are sensitive but nonideal osmometers; a small restoring force prevents them from shrinking or swelling quite as much as expected for ideal behavior. We find that Na+, Cl-, No3-, glycerol, acetate, and ammonium rapidly enter the outer segment, but K+, SO4=, and melezitose appear impermeable. The Na flux is rectified; for concentration gradients in the physiological range, 2 x 109 Na+ ions/sec enter the outer segment, but we detect no efflux of Na+, under our conditions, when the gradient is reversed. Illumination of the outer segment produces a specific increase in the resistance to Na+ influx, but has no effect on the flux of other solutes. This light-dependent Na+ resistance increases linearly with the number of rhodopsin molecules bleached. We find that excitation of a single rhodopsin molecule produces a transient (~1 sec) "photoresistance" which reduces the Na+ influx by about 1%, thus preventing the entry of about 107 Na+ ions. At considerably higher light levels, a stable afterimage resistance appears which reduces the Na influx by one-half when 106 rhodopsin molecules are bleached per rod. We have incorporated these findings into a model for the electrophysiological characteristics of the receptor.  相似文献

7S RNA sequences from the hagfish (Myxiniformes) and lamprey (Petromyzontiformes) were cloned and analyzed. In both species, 7S L RNA (also designated SRP RNA, since it represents the RNA constituent of the signal recognition particle) was clearly detectable. The sequence similarity between the two species was 86%, compared with about 75% similarity between either of these species and mammals. 7S K RNA was also cloned from the lamprey. The similarity between the 7S K RNA of the lamprey and that of mammals was 68%. Interestingly, several interspersed elements were found with nearly 100% similarity compared with mammals. In contrast to the lamprey, no 7S K RNA-related sequences were detectable among hagfish RNA, neither in northern blots nor with the PCR assay. In view of the significant conservation between the 7S K RNA of lamprey and that of mammals (human), this unexpected result clearly separates lamprey and hagfish. In addition, the lack of detectable 7S K RNA sequences in an outgroup, such as amphioxus, indicates that these results do not reflect an autapomorphy of hagfish. Therefore, our data provide additional support to the notion of a sister group relationship between Petromyzontiformes and gnathostomous vertebrates to the exclusion of Myxiniformes. Serial sections, injections with india ink and latex, and observations on fresh material, have been used to determine the pattern of blood circulation within the intestine of larval and adult lampreys. Attention has also been paid to resolving the variable terms previously applied to many of the blood vessels, and to the possible functional significance of the differences found between the two life cycle stages. In the larva, the main arterial supply to the intestine consists of a typhlosolar artery, while the venous return is comprised of a posterior and a left and right anterior intestinal vein that usually unite before entering the liver. Although a typhlosolar artery is also present in the adult, the main venous return of the ammocoete is replaced at metamorphosis by a newly formed typhlosolar vein. Moreover, in the ammocoete a considerable amount of blood is discharged into the haemopoietic sponge-work of the typhlosole and the arterial supply to the intestine is poorly developed. By contrast, the typhlosolar sponge-work is lost in the adult and a more efficient arterial supply is developed within the lamina propria of the various intestinal regions. Furthermore, vascular couples are developed in the adult which facilitate the flow of blood in opposite directions in the intestinal wall. Since, during both life cycle stages, the arterial blood passes into tissue spaces, there is no true capillary network in the intestine and no evidence was found for the presence of a lymphatic system. It is suggested that the changes which take place in the intestinal blood supply and the internal structure of the gut during metamorphosis result in improvements both to the vascular system and to the assimilation efficiency.  相似文献

The accessory olfactory organ of Lampetra fluviatilis was found to consist of clusters of interconnected vesicles in tenuous connection with the exterior medium via the cavity of the olfactory organ. The walls of the vesicles are composed of two types of cells. One type are primary sense cells that resemble the olfactory sense cells in that their nucleus is situated peripherally and their axons pass directly into the brain. They differ from the olfactory sense cells in the size and number of cilia they bear, and also in the internal structure of the cilia. The second cell type are supporting and/or secretory cells. It is concluded that this sense organ is capable of responding to a “special kind” of chemical stimulus and its possible function is discussed.  相似文献

The cytology of the interstitial cells of the testis of the river lamprey has been studied from the autumn when the upstream migration begins up to the completion of sexual maturation in the following spring. In the early stages of spermatogenesis the interstitial tissue consists of isolated interlobular connective tissue cells. During the winter these appear to increase in numbers and form compact groups sometimes showing an acinar type of structure. In the earlier stages of the secretory cycle the cytoplasm contains small osmiophilic and sudanophilic granules, which appear to aggregate to form much larger lipid positive masses. In the later stages of spermatogenesis in early spring, the interstitium accumulates large areas of lipid and cholesterol positive droplets and in fixed material the cytoplasm shows extensive vacuolization. In the terminal stages the cells undergo complete disintegration. It seems probable that the stage when the interstitium shows masses of lipid and cholesterol positive droplets represents a phase of degeneration and that the secretion of hormone occurs in earlier stages. No convincing evidence has been obtained for the presence of the enzyme steroid-3β-ol-dehydrogenase during the later stages of the secretory cycle. There is evidence that following the degeneration of the interstitium there may be a replacement of the secretory tissue by the transformation of residual connective tissue elements, which may then re-enter the secretory phase. The changes in the interstitium are paralleled by hypertrophy and evidence of lipid metabolism in the modified fibroblasts of the lobular wall and this tissue may also be involved in the secretory activity of the testis.  相似文献

Resting cysts and the excystment process in the freshwater ciliate Euplotes muscicola were studied by both light and scanning electron microscopy. Groups of distinctly crested resting cysts adhere to the substrate. Silver-stained preparations reveal surface conservation of dorsal kinetosomes and dorsal argyrome while ventral organelles are directed inward. Excystment involves the development of an expanding excystment vacuole concurrent with a localized thinning on the dorsal cyst wall surface. Cells exit through the pre-formed ostiole, mid-dorsal region first, initially by the force of cytoplasmic streaming, but later aided by cirral movement. Newly emerged cells retain the excystment vacuole and show no dorsal ridging. As the cell expels its excystment vacuole and partially unfolds, normal trophont morphology is re-established. Both cyst structure and cyst typology have implications for hypotrich taxonomy.  相似文献

Abstract Each pigment-cup eye of Mesostoma ehrenbergi consists of two photoreceptor cells, the anterior cell being bilobate. the posterior almost linear, and of a multicellular pigment cup. The nuclei of the photoreceptor cells are located inside the medial region of the brain. Thin cytoplasmic photoreceptor projections provided with neurosecretory-like granules are interposed between the inner surface of the eye cup and the distal extremity of the microvilli. The breakdown and renewal of microvillar membranes was analysed. Membrane turnover is a continuous process. At dusk and during the night abscission of photoreceptive membranes occurs. At dawn the membrane fragments are degraded to granular material, which is then endocytosed into the submicrovillar cytoplasm as coated vesicles. These vesicles form multivesicular bodies. The degradation of multivesicular body content occurs during the following light hours. The dark period is correlated with membrane synthesis for elongation of reticular membranes, which are converted into ellipsoid bodies. The formation of new microvillar membranes occurs at the base of the microvillar border, and involves the fusion with the old microvillar membranes of small vesicles detached from the tubular endoplasmic membranes and from the flattened concentric cisternae of ellipsoid bodies. The correlations with daily cycles of other invertebrates are discussed.  相似文献

Demonstration of the synaptonemal complex for light microscopy has until now been based on staining with silver. After fixation at pH 9-10 it is also possible to visualize synaptonemal complexes with several nonspecific protein stains such as Coomassie brilliant blue, Giemsa, fast green, light green and Stains All. Although staining with silver gives the best contrast between synaptonemal complexes and the background, the other dyes have a number of advantages, such as more even staining, easy extractability, and lower cost than silver.  相似文献
