Uteroglobin, which is a progesterone-binding protein of the rabbit uterine secretion, has been crystallized and subjected to X-ray diffraction analysis. Two crystalline forms have been observed: a triclinic one (P1, Z = 2, a = 36.36 (4) A?, b = 37.40 (4) A?, c = 53.28 (4) A?, α = 104.6 (1) °, β = 97.0 (1) °, γ = 111.3(1) °); and an orthorhombic one for which the cell is C-centred with a = 50.86 (5)A?, b = 52.22 (5)A?, c = 47.28 (5) A?, space group C2221, Z = 4. Three isomorphous derivatives have been obtained. The crystals appear suitable for detailed study of the three-dimensional structure of the protein.  相似文献

Two crystal forms of lysozyme from the egg white of the embden goose (Anser anser) have been obtained, both of which are suitable for X-ray diffraction analysis. The monoclinic form has space group P21 with cell dimensions a = 38.3 A?, b = 65.7 A?, c = 45.2 a?, β = 116 ° and the triclinic form (space group P1) has cell dimensions of a = 39.9 A?, b = 42.2 A?, c = 57.9 A? and α = 98.8 °, β = 102.5 °, γ = 90.5 °.  相似文献

Crystals of plastocyanins from pea and corn leaves have been obtained. Both are suitable for X-ray structure analysis with a resolution up to 1.8 Å. The crystal form of plastocyanin from pea leaves belongs to the space group P212121 with unit cell dimensions: a = 49.0 A?, b = 53.3 A?, c = 82.6 A?. The assumed number of protein molecules per asymmetric unit of the unit cell is two. Crystals of the oxidized (Cu2+) and reduced (Cu+) forms are isomorphic. No essential differences in spot intensities for the main zone with a resolution of 3 Å were revealed. The crystal form of plastocyanin from corn leaves belongs to the space group P1 with unit cell parameters: a = 24.8 A?, b = 30.0 A?, c = 58.5 A? and α = 96° 10′, β = 87°08′, γ = 78°40′. The assumed number of protein molecules per asymmetric unit is two.  相似文献

Erysimum latent virus, a tymovirus, contains 180 protein subunits arranged in a T = 3 icosahedral surface lattice. A cubic crystal form (with space group P213 and a = 414 A?) and a monoclinic form (space group B2, a = 442 A?, b = 422 A?, c = 387 A?, γ = 95 °) have been observed. The asymmetric units of the two crystal forms contain one-third and one whole virus particle, respectively. Two possible packing arrangements of the virus particles in the monoclinic unit cell have been deduced from the low-angle diffraction patterns. X-ray diffraction data from the monoclinic crystals extend to at least 3·7 Å resolution.  相似文献

The “goose-type” lysozyme isolated from the egg-white of the black swan Cygnus atratus has been crystallized. The space group is P21 with one molecule of protein in the asymmetric unit. The cell parameters are a = 46.2 A?, b = 65.1 A?, c = 38.7 A?, β = 110 °. The crystals diffract to a resolution of 2.25 Å and a set of diffraction data for the native protein has been collected photographically.  相似文献

A hexagonal crystal form (P6322, a = b = 34.0 A?, c = 113.5 A?) and a monoclinic form (P21, a = 37.1 A?, b = 32.2 A?, c = 32.4 A?, β = 110 °) of neutrophil cationic protein NP2, isolated from rabbit leukocytes, have been characterized. The monoclinic form, containing two promoters (Mr = 3844) per asymmetric unit, diffracts to at least 1.8 Å and is suitable for high-resolution structural studies.  相似文献

The Fc fragment was prepared from rabbit immunoglobulin G by digestion with papain, both with the inter-chain disulphide bond intact, and after reduction and alkylation. These two types of Fc crystallized in different, yet related forms, each with one dimer in the asymmetric unit. The covalently linked dimer crystallized in space group P21; a = 68.85 ± 0.05 A?, b = 72.50 ± 0.05 A?, c = 60.40 ± 0.05 A? and β = 105.1 ± 0.2 °. The reduced, non-covalently linked dimer also crystallized in space group P21; a = 81.55 ± 0.05 A?, b = 55.65 ± 0.05 A?, c = 68.85 ± 0.05 A? and β = 1051 ± 0.2 °. A non-crystallographic 2-fold axis relating the two identical polypeptide chains is clearly visible in the h0l projection of the second crystal form.  相似文献

Three crystal forms of rabbit muscle creatine kinase have been grown, one of which seems suited to a high resolution X-ray diffraction study. The first form is of monoclinic space group P21 with a = 54 A?, b = 114 A?, c = 145 A?, β = 91 ° and has as the asymmetric unit two molecules of total molecular weight 160, 000. The second form, grown in the presence of mercurials, is of space group A2 with a = 52 A?, b = 165 A?, c = 237 A?, β = 91 ° and also has two molecules in the asymmetric unit. The third crystal form, grown in the presence of a high concentration of cysteine, is of apparent space group P212121, but evidence indicates that the true space group may be P21221. The dimensions of the orthorhombic unit cell are a = 47 A?, b = 86 A?, c = 125 A?, and the asymmetric unit contains a single protein subunit. Assuming the latter space group, then the creatine kinase molecule possesses a twofold axis relating two identical subunits.  相似文献

Crystals of the periplasmic ribose binding protein of Salmonella typhimurium have been subjected to X-ray analysis. The crystals grow as rectangular parallelopipeds with the symmetry of space group P21. Unit cell dimensions are a = 64·4 A?, b = 60·6 A?, c = 62·8 A?, and β = 91·25 °. Two closely related crystal forms of dimeric cytochrome c5 from Azotobacter rinelandii have been grown. The crystals belong to space groups (C2 with a = 45·0, b = 38·4, c = 41·3 A? and β = 101 ° 0′; and C1 (a centered triclinic cell) with a = 46·0, b = 37·6, c = 49·4 A?, α = 87 ° 20′, β = 96 ° 40′ and γ = 90 ° 0′. In C2 the 24,000 molecular weight dimer lies on a Crystallographic 2-fold axis; in C1 the entire dimer occupies the asymmetric unit.  相似文献

The lectin from the seeds of Abrus precatorius has been crystallized and the crystals subjected to study by X-ray diffraction and electron microscopy. Three closely related crystal forms were obtained, of orthorhombic space group P212121 with a = 138 A?, b = 142 A?, and c = 173 A?, of tetragonal space group P41212 with a = b = 136 A?, c = 176 A?, and a twinned intermediate of the first two. From electron microscopy and two-dimensional spatial filtering of electron micrographs of the crystals, the molecule appears to consist of four similar domains grouped in a roughly planar diamond-shaped arrangement having a local intramolecular dyad axis. The average diameter of the Abrus lectin molecule is 50 to 60 Å and the individual domains appear to have a diameter of about 25 Å.  相似文献

Crystals of crambin, a plant seed protein of molecular weight 5000, diffract X-rays strongly to the interplanar spacing limit of 0.88 Å. These diffraction data should allow a definition of atomic structure that is on a par with that typically obtained from crystals of small organic molecules. The crystals are in space group P21 and have unit cell dimensions a = 41.1 A?, b = 18.7 A?, c = 22·7 A?, and β = 90.6 °. The asymmetric unit contains one protein molecule.  相似文献

Reassembled alfalfa mosaic virus coat protein was partially digested with trypsin to remove the first 26 amino acids (Bol et al., 1974). These particles are empty icosahedral protein shells built with 60 alfalfa mosaic virus protein subunits. This aggregate has been crystallized in two different crystal forms, one of which diffracts X-rays to at least 3.4 Å resolution. The type I crystals (space group P63, a = 200 A?, c = 314 A?) contain two particles per cell separated by 195 Å with each sitting on a 3-fold axis. The type II crystals contain three particles per cell in space group P31or P32 (a = 201 A?, c = 485 A?). Other T = 1 viral particles have very similar diameters.  相似文献

The mitochondrial isoenzyme of aspartate aminotransferase (E.C. 2.6.1.1) has been isolated from chicken heart in an electrophoretically and immunologically homogeneous form. Large, well-diffracting single crystals of this enzyme, a dimeric molecule with a molecular weight of 90,000, have been grown by vapour phase diffusion against polyethylene glycol solutions. The crystals belong to space group P1. The unit cell, with the dimensions a = 55.6 A?, 6 = 58.7 A?, c = 76.0 A?, α = 85.3 °, β = 109.2 °, γ = 115.6 °, contains a single dimer. The diffraction pattern extends to at least 2.1 Å resolution.  相似文献

New data have been collected on the crystalline structure of collagen fibrils in tendon. The unit cell in decrimped tendon has been determined by measurements of the Bragg reflections in the X-ray diffraction pattern. The results are consistent with a triclinic cell with b = 75.5 A?, β = 93 °, a = bsinβ, a = 90 °, c = n × 668 A?, where n is probably 4 and γ = 90 °. A selection rule observed for prominent reflections is explicable either in terms of a specific orientation of the microfibrils on the lattice, or by a helical distortion of the microfibril axis. The cell parameter β can be varied by changing the ionic envirionment.  相似文献

The crystals of ribonuclease St, the extracellular ribonuclease from Streptomyces erythreus, have been obtained from (NH4)2SO4 solution with acetate buffer (pH 4.1). The crystals belong to a monoclinic space group C2 with dimensions a = 88.4 A?, b = 33.0 A?, c = 69.0 A?, β = 98.4 °. There are two protein molecules per asymmetric unit. Superoxide dismutase from spinach leaves was salted out with ammonium sulphate. The resulting crystals were monoclinic, space group C2, with unit cell dimensions a = 166.2 A?, b = 46.1 A?, c = 85.6 A? and β = 99.3 °. Wheat germ agglutinin crystallizes in two monoclinic space groups, P21 and C2, under identical crystallization conditions. Unit cell dimensions are a = 73.8 A?, b = 51.2 A?, c = 90.8 A?, γ = 90 ° for P21; a = 51.31 A?, b = 73.35 A?, c = 91.45 A?, β = 97.75 ° for C2, both with eight subunit molecules in the unit cell. The C2 crystals were chosen as suitable for investigating the three-dimensional structure to high resolution, because of their smaller asymmetric unit (containing the dimer), and also because they display better diffraction patterns.  相似文献

A [4Fe4S] ferredoxin from Bacillus thermoproteolyticus has been crystallized. The space group is P1 with two molecules in the unit cell, with the dimensions a = 32.96 A?, b = 37.83 A?, c = 39.82 A?, α = 118.1 °, β = 104.2 ° and γ = 89.7 °.
