A study for screening and selection of mutants of Pseudomonas corrugata (NRRL B-30409) based on their phosphate solubilization ability, production of organic acids, and subsequent effect on plant growth at lower temperatures under in vitro and in situ conditions was conducted. Of a total 115 mutants tested, two (PCM-56 and PCM-82) were selected based on their greater phosphate solubilization ability at 21°C in Pikovskaya’s broth. The two mutants were found more efficient than wild-type strain for phosphate solubilization activity across a range of temperature from psychotropic (4°C) to mesophilic (28°C) in aerated GPS medium containing insoluble rock phosphate. High-performance liquid chromatography analysis showed that phosphate solubilization potential of wild-type and mutant strains were mediated by production of organic acids in the culture medium. The two efficient mutants and the wild strain oxidized glucose to gluconic acid and sequentially to 2-ketogluconic acid. Under in vitro conditions at 10°C, the mutants exhibited increased plant growth as compared to wild type, indicating their functionality at lower temperatures. In greenhouse trials using sterilized soil amended with either soluble or rock phosphate, inoculation with mutants showed greater positive effect on all of the growth parameters and soil enzymatic activities. To the best of our knowledge, this is the first report on the development of phosphate solubilizing mutants of psychotropic wild strain of P. corrugata, native to the Indian Himalayan region.  相似文献

Five strains of a pigment mutant were isolated following UV irradiation and N-methyl-N′-nitro-N-nitrosoguanidine (MNNG) mutagenesis from a non-nitrogen fixing mutant of the cyanobacteriumGloeotrichia ghosei. Two of them (B-1 and V-1) were isolated by UV mutagenesis and other three (B-3, B-7 and Br-6) by MNNG mutagenesis. Among the five strains cultures of three strains (B-1, B-3 and B-7) were typically blue-green in colour. Culture of strain V-1 was found to be violet-pink and of Br-6 was brownish in colour. The parent strain of these mutants was dark-blue in colour. Blue-green mutants showed the predominance of phycocyanin (610 nm) whereas violet-pink and brown strains showed the predominance of phycoerythrin (550 nm) in the absorption spectra of water-soluble pigments. In contrast to these strains their parent strain showed both the absorption peaks (at 550 and 610 nm). Occurrence of stable pigment mutants of a filamentous cyanobacterium indicates that the synthesis of water-soluble pigments is genetically controlled in these mutant strains.  相似文献

We compared the stability of antibiotic-resistance markers in strains derived from Bacillus cereus UW85 in culture media and in the soybean rhizosphere in a growth chamber and in the field. We studied two independent, spontaneous mutants resistant to neomycin, three independent, spontaneous mutants resistant to streptomycin, and strains carrying plasmid pBC16, which encodes tetracycline resistance. Antibiotic-resistance markers were maintained in populations of all UW85 derivatives in culture and in the rhizosphere of soybeans grown in soil in a growth chamber. In two field experiments, antibiotic resistance was substantially lost in rhizosphere populations of B. cereus as early as 14 or as late as 116 days after planting. To distinguish between death of the inoculated strain and loss of its marker, we tested populations of B. cereus for other phenotypes (orange pigmentation, plasmid-borne resistance to tetracycline, and biocontrol activity) that are typical of UW85-derivatives used as inoculum, but atypical of the indigenous populations of B. cereus , and these phenotypes were maintained in populations from which the marker was lost. In general, neomycin-resistance markers were maintained at a higher frequency than streptomycin-resistance markers, and maintenance of antibiotic-resistance markers varied with position on the root and with the year of the experiment. In a semi-defined medium, the UW85 derivatives grew at the same rate as the wild type at 28°C, but most grew more slowly than the wild type at 16°C, demonstrating that antibiotic resistance can affect fitness under some conditions. The results suggest that the stability of antibiotic-resistance markers should be assessed in the ecosystems in which they will be studied.  相似文献

Aim:  The influence of environmental (temperature and pH) and biological (strain) parameters on the inactivation of Campylobacter jejuni by high hydrostatic pressure (HHP) was investigated.
Methods and Results:  Two clinical strains harvested in stationary phase were pressurized at 20°C and 37°C within a range of 50–400 MPa, in a phosphate (pH 7·0) or a citrate phosphate buffer (pH 5·6), for 10 min. Treatment efficiencies were determined by logarithmic comparisons of culturable cells on blood agar before and after treatment. Results were statistically compared using an anova of culturable cells after treatment to evaluate the effect of all factors. At least a 7-log reduction in cell numbers was observed for both strains. The pH and the strains had no effect on HHP treatment at 20°C while at 37°C, both pH and strain influenced significantly the HHP treatment on C. jejuni .
Conclusions:  The pressure efficacy on C. jejuni eradication was affected by both environmental and biological factors.
Significance and Impact of the Study:  Depending on the treatment conditions, C. jejuni sensitivity to HHP can significantly vary. The determination of the inactivation treatment by HPP has to be normalized considering the interaction of environmental and biological factors.  相似文献

The biochemical properties, virulence for mice and trout, and the extracellular virulence factors at 28° and 37°C of 11 environmental and nine human strains of Aeromonas hydrophila were compared. All the environmental isolates and four of the human group were virulent for trout at 3 x 107 cfu, but only human strains were able to cause death or lesions in mice by the intramuscular route. Extracellular virulence factors such as haemolysins, cytotoxins and proteases were also investigated in supernatant fluids of cultures grown at 28°C and 37°C. The production of haemolysins, caseinases, elastases and growth yields of environmental strains decreased sharply during cultivation at 37°C but cytotoxins were produced to the same extent, or slightly less, than at 28°C. The human strains differed from the environmental strains in response to growth temperatures: protease activity decreased at 37°C, although growth yield was not affected, but more haemolysins and cytotoxins were produced by the virulent strains at this temperature than at 28°C. Sodium caseinate SDS-PAGE of culture supernatant fluids of selected human strains revealed that temperature selectively inhibited the production of certain proteases.  相似文献

The O:34-antigen lipopolysaccharide as an adhesin in Aeromonas hydrophila   总被引:4,自引:0,他引:4  
Abstract We compared the ability of different Aeromonas hydrophila strains from serogroup O:34 grown at different temperatures to adhere to Hep-2 cells. We found a high level of adhesion when the strains were grown at 20 °C but not when they were grown at 37 °C. We previously described that these strains were able to form the O-antigen lipopolysaccharide when they grow at low temperature but not at high temperature. We also obtained by transposon mutagenesis mutants only devoid of the O-antigen lipopolysaccharide ( rfb mutants), and they showed significantly lower levels of adhesion to Hep-2 cells than the smooth strains. All these results prompted us to conclude that the O-antigen LPS, in these strains, is an important adhesin.  相似文献

Seedlings of Phaseolus aureus ROXB were grown under 12/12 h light/dark cycles with the light period at 32.5°C and darkness at 10°C (normal conditions N) or with light at 10°C and darkness at 32.5°C (inverse conditions, I). I-conditions affected the level of chlorophyll and carotenoids (very low), monogalactosyldiacylgycerol (low) and phosphatidylinositol (high) in the leaves. Leaves of I-seedlings showed a sharp and durable decline of relative water content during the low temperature phase. For the N-seedlings, loss of water was restricted to the end of this period. The loss of water was accompanied by visible symptoms of wilting at specific times of day. Although the pigment content remained nearly unchanged, ABA content of leaves of both N-and I-seedlings increased during water stress. Upon return to the warm period, ABA level continued to increase after the leaves had regained turgor, this 'after stress'increase being more pronounced in the leaves of I-seedlings. Exogenous application of ABA induced a slight increase in the content of phospholipids in N- and I-leaves and a decrease in free fatty acids, whereas monogalactosyldiacylglycerol content was significantly reduced in N-leaves after application of ABA. Upon transfer of I-plants to 20°C for 12 h during the light period, pigment and chloroplastic lipid content increased rapidly whereas upon a further exposure to 10°C in light, pigments and especially monogalactosyldiacylglycerol were lost. The control of pigment and lipid metabolism and the role of ABA during chilling stress are discussed.  相似文献

Wild type Gracilaria tikvahiae, a macrophytic red alga, and fourteen genetically characterized pigment mutants were analyzed for their biliprotein and chlorophyll contents. The same three biliproteins, phycoerythrin, phycocyanin, and allophycocyanin, which are found in the wild type are found in all the Mendelian and non-Mendelian mutants examined. Some mutants overproduce R-phycoerythrin while others possess only traces of phycobiliprotein; however, no phycoerythrin minus mutants were found. Two of the mutants are unique; one overproduces phycocyanin relative to allophycocyanin while the nuclear mutant obr synthesizes a phycoerythrin which is spectroscopically distinct from the R-phycoerythrin of the wild type. The phycoerythrin of obr lacks the typical absorption peak at 545 nanometers characteristic of R-phycoerythrin and possesses a phycoerythrobilin to phycourobilin chromophore ratio of 2.6 in contrast to a ratio of 4.2 found in the wild type. Such a lesion provides evidence for the role of nuclear genes in phycoerythrin synthesis. In addition, comparisons are made of the pigment compositions of the Gracilaria strains with those of Neoagardhiella bailyei, a macrophytic red alga which has a high phycoerythrin content, and Anacystis nidulans, a cyanobacterium which lacks phycoerythrin. The mutants described here should prove useful in the study of the genetic control of phycobiliprotein synthesis and phycobilisome structure and assembly.  相似文献

Abstract 23 Strains of diazotrophic root-associated bacteria isolated from various parts of Finland were tested for nitrogenase activity during growth at various temperatures. Nitrogenase activity was optimal at 20–37°C in cultures of Klebsiella pneumoniae , and at 14–20°C in cultures of Klebsiella terrigena and Enterobacter agglomerans . Strains of K. terrigena and E. agglomerans showed no activity at 37°C, and K. pneumoniae only minimal or no activity at 14°C. Azospirillum lipoferum exhibited high nitrogenase activity at both 28–37°C, but less than 25% of optimal activity at 20°C and no activity at 14°C. Pseudomonas sp. expressed nitrogenase activity at 14–28°C. None of the strains manifested nitrogenase activity at 4 or 42°C. There were only small local variations within a species between strains isolated at different locations.  相似文献

Six strains of Rhizobium , three temperature-tolerant (U1, C13 and A19) and three temperature-sensitive strains (U10, C10 and A4) selected on the basis of previous study were used to screen the synergestic effect of different relative humidities (r.h. 50%, 65% and 90%) and temperatures (28°, 35°, 40° and 45°C) on the survival of rhizobia in inoculant carrier. At a particular temperature all the three r.h.'s were maintained. At a storage temperature of 28° and 35°C, the r.h. had little effect on the population of any of the rhizobial strains tested, but at 40° and 45°C, marked differences were observed and it was concluded that higher r.h. in conjunction with higher temperature resulted in low viable counts. The effect was similar but less obvious with the temperature-tolerant strains. It was found that 50% r.h. at different storage temperatures extended shelf life of rhizobial strains of blackgram, cowpea and arhar crops which were tested in this study in inoculant carrier.  相似文献

The pigment and quinone content of wild-type barley ( Hordeum vulgare L., cv. Svalöfs Bonus) and of two photosynthetic mutants was assayed. Wild type plants and the photosystem Hacking mutant viridis zb63 contained chlorophyll a and b. whereas chlorina-f2 contained only chlorophyll a The inability of the mutant chlorina-f2 to convert chlorophyll a into chlorophyll a appears to he the primary effect of the mutation. In both mutants, the carotenoid composition was virtually identical to that of the wild type. As compared to the wild type. chlorina-f2 contained less lutein and neoxanthin. The mutant viridis-zb63 contained less β-carotene but more antheraxanthin and xeaxanthin than the wild type. The quinone content and composition of the wild type and the photosynthetic mutants was similar, and both mutants biosynthesized plastid quinones and chromanols starting from [14C]-labeled tyrosine. The data indicate that carotenoid and quinone biosynthesis are not altered in the two mutants as compared to the wild type.  相似文献

A demographic study of the Nile crocodile Crocodylus niloticus at Lake Ngezi, Zimbabwe, revealed that females predominated in all size classes and among embryos. The sex of C. niloticus was shown to be determined by the temperature of egg incubation in constant temperature laboratory experiments. At 31 °C and below only females were produced. The threshold temperature for maleness was between 31 ° and 34 °C, but appeared to vary between clutches. The duration of the incubation period varied with temperature and was 110 days at 28 °C, falling to 85 days at 34 °C. Incubation temperature affected hatchling length, but not mass. Hatchlings from incubation at 34 °C were shorter on average than those from incubation at 28 °C and 31 °C, but by three months had outgrown them. There was no sex-related difference in length in a random sample of 200 two-year-old C. niloticus on a crocodile farm. Mean temperatures in wild nests were consistently lower than 31 °C and therefore the male threshold as determined in the laboratory. Embryonic development was slow and hatching success poor. The shallowest eggs in a nest had higher mean temperatures and more advanced embryos than the deepest eggs. They also experienced daily temperature fluctuations of up to 10 °C during which the maximum occasionally rose to 35 °C. Constant temperature incubation was not a good model of field conditions, but the correlation between nest temperatures and embryonic sex is consistent with temperature-dependent sex determination in the wild.  相似文献

Four mutant strains of Euglena gracilis have been isolated after treatment of wild type cells with ultraviolet light or the chemical mutagen nitrosoguanidine. None of the mutants is capable of autotrophic growth or photosynthetic carbon dioxide fixation.The mutant strains contain normal amounts of the enzymes of the reductive pentose phosphate cycle and are qualitatively similar to the wild type in pigment composition, but are unable to carry out the Hill reaction (light induced reduction of 2,6-dichlorophenol indophenol). Isolated mutant plastids cannot photoreduce NADP with water as the electron donor but can carry out this reaction when the electron donating system is ascorbate and 2,6-dichlorophenol indophenol. Whole cells of the mutants show the light induced oxidation of cytochrome f by light reaction I but are unable to bring about cytochrome f reduction by light reaction II. The mutants appear to be blocked at or near light reaction II in the photosynthetic electron transport chain.The mutants may represent alterations of the chloroplast genome since the mutation isolation was carried out under conditions where chloroplast viability was severely impaired, but cell viability was unaffected.  相似文献

ATP content (per g fresh weight) and energy charge were higher during incubation at 8°C than at 28°C in the early stages of callus induction of potato tuber discs ( Solanum luberosum L. cv. Bintje). After a transfer from 28°C to 8°C, ATP content increased while a sharp decline in ATP content was observed after a transfer from 8°C to 28°C. ADP and AMP pools did not increase correspondingly. When the callus discs had entered the logarithmic growth phase, the energy charge was maintained within relatively narrow limits (0.77–0.80) at all culture temperatures.  相似文献

Mutant strains of Rhodopseudomonas spheroides have been isolated which contain 5–50 times more bacteriochlorophyll and carotenoids than the wild type when grown under highly aerobic conditions in the dark. Their pigment content is similar to the wild type when grown in the light. One of the mutants (TA-R) grew more slowly than its parent strain under aerobic conditions but formed pigments at about 60% of the rate observed under photosynthetic conditions. The other mutants grew at rates similar to the wild type under all conditions. Synthesis of bacteriochlorophyll by suspensions of the mutants began without delay upon transfer from conditions of high to low aeration. In contrast to the wild type, magnesium protoporphyrin-S-adenosylmethionine methyltransferase (EC 2.1.1.11) activity in particulate preparations from the mutants was not repressed by growth under aerobic conditions in the light or dark. Ribulose diphosphate carboxylase (EC 4.1.1.39) activity was repressed by O2 in the mutants as in the wild type. Other enzyme activities were compared in mutant TA-R and its parent strain grown under the same conditions. NADH oxidase activity in particles from aerobically grown TA-R was about one third that found in the parent strain. However, the respiration rates of the intact cells did not differ. Light inhibited the respiration of aerobically grown TA-R, indicating that the bacteriochlorophyll formed under these conditions had photochemical activity. It is concluded that the insensitivity of the mutants to O2 repression is due to defects in the regulatory system which controls formation of the enzymes concerned in pigment synthesis.  相似文献

SYNOPSIS. Several substrains of Euglena gracilis var. bacillaris made chlorotic by treatment with pyribenzamine or streptomycin, or by growth at high temperature (35–36°C.), have been examined for their carotenoid content. They differ from the normal green strain both qualitatively and quantitatively. Some strains produce no detectable carotenoids while the carotenoid concentration in the strains producing most is at best only one-fifth that of the normal strain. In all substrains producing carotenoids, the carotene fraction consists of β-carotene accompanied by some members of the phytofluene series. In only two of these substrains, HB-G and PBZ-G3, are xanthophylls produced in significant amounts. In HB-G, the main pigment is echinenone, and in PBZ-G3 it is zeaxanthin. The significance of these findings is briefly discussed.  相似文献

Abstract The effects of heat shock upon the expression of several developmentally regulated genes of Myxococcus xanthus were examined. No effects were observed on levels or timing of developmentally regulated β-galactosidase expression in eight randomly selected Tn5lac insertion mutants. However, heat shock significantly affected the fruiting behavior of temperature-sensitive aggregation ( tag ) mutants of M. xanthus . The tag mutant phenotype exhibits the normal aggregation of cells to form fruiting bodies at temperatures < 34°C, but cells fail to aggregate at temperatures ⩾ 34°C. Heat shock administered to tag mutant strains prior to starvation prohibited fruiting body formation at permissive temperatures. Additionally, tag mutant strains were found to be extremely sensitive to killing at 40°C. Heat shock was also found to increase tagA and tagE expression by 22 and 47%, respectively. ABSTRACT. We have characterized a novel, temperature-sensitive mutation affecting motility in Tetrahymena thermophila . Mutants grew and divided normally at the restrictive temperature (38°C), but became nonmotile. Scanning electron microscopic analysis indicated that nonmotile mutants contained the normal number of cilia and that the cilia were of normal length. Transmission electron microscopic analysis indicated that axonemes isolated from nonmotile mutants lacked outer dynein arms, so the mutation was named oad I ( outer arm defficient ). Motile mutants shifted to 38° C under conditions that prevent cell growth and division (starvation) remained motile suggesting that once assembled into axonemes at the permissive temperature (28° C) the outer arm dyneins remain functional at 38° C. Starved, deciliated mutants regenerated a full complement of functional cilia at 38° C, indicating that the mechanism that incorporates the outer arm dynein into developing axonemes is not affected by the oad I mutation. Starved, nonmotile mutants regained motility when shifted back to 28° C, but not when incubated with cycloheximide. We interpret these results to rule out the hypothesis that the oad I mutation affects the site on the microtubules to which the outer arm dyneins bind. Axonemes isolated from mutants grown for one generation at 38° C had a mean of 6.0 outer arm dyneins, and axonemes isolated from mutants grown for two generations at 38° C had a mean of 3.2 outer arm dyneins. Taken together, these results indicate that the oad I mutation affects the synthesis of outer arm dyneins in Tetrahymena .  相似文献
