Fungal flora of 20 samples of lentil seeds collected from Assiut governorate, Egypt, were studied. Seventeen genera and 13 species were isolated on glucose- (15 genera and 27 species), cellulose-(15 genera and 25 species)Czapek's agar media at 28 °C. The most common species were as follows: on glucose-Czapek's agar, Aspergillus fumigatus, A. niger, A. flavus, A. terreus, Penicillium notatum and Rhizopus oryzae and on cellulose agar, A. fumigatus, A. niger, A. flavus and P. notatum. Thin layer chromatographic (TLC) analysis and a biological test (Artemia salina) indicated the presence and the toxicity of aflatoxin in the extract of one sample (aflatoxins B1, B2, G1 and G2, at 20 mg/kg, total). This is the first report of the natural occurrence of aflatoxins in lentils.  相似文献

A wide range of moulds representing several genera and species, was recorded in this study from 5 seed samples of each almond, cashew nut, chestnut, hazelnut, pistachio nut and walnut collected from different markets in Ar' Ar, Saudi Arabia. The total counts of fungi were widely fluctuated between 1960–7704 and 1948–7434 colonies/g dry seeds on glucose-Czapek's and glycerol agar media at 28°C, respectively, and represented twenty genera, 53 species and 2 varieties of fungi. The prevalent fungi on the 2 agar media wereAspergillus flavus, A. niger andPenicillium chrysogenum. On glucose-Czapek's agar,Rhizopus stolonifer andAspergillus flavus var.columnaris were isolated from all 6 kinds of nut,A. parasiticus from 5 kinds andA. fumigatus from 4 kinds with high frequencies.Eurotium species were completely absent on glucose-Czapek's agar but they were isolated in high frequency from all kinds of nut on glycerol agar medium. The different nut samples were analyzed by thin layer chromatography for the presence of aflatoxins B1, B2, G1 & G2, citrinin, ochratoxins, patulin, sterigmatocystin, diacetoxyscirpenol, T-2 toxin and zearalenone. Aflatoxins B1 & G1 were detected in 3 out of the 5 samples tested of chestnut at concentrations ranging between 20 to 60 µg/kg. All other samples of almond, cashew nut, hazelnut, pistachio nut, and walnut that were analyzed were mycotoxin free.  相似文献

Thirty-nine species and 16 fungal genera were isolated from Egyptian cotton seeds, cotton seed meal and cotton seed cake on 1% glucose-Czapek's agar medium incubated at 28 °C. Aspergillus was the most frequent genus and it emerged in 87–100% of the samples contributing 70–98% of total fungi in the three substrates tested. The most common species were A. niger, A. flavus, A. fumigatus, A. terreus and Rhizopus stolonifer; A. niger, A. fumigatus and Penicillium corylophilum; and A. niger, A. flavus, A. terreus, A. nidulans and Rhizopus stolonifer, respectively. Cotton seeds and cotton seed products were naturally contaminated by aflatoxin B1 and B2. About 16% of the different substrates tested were positive for aflatoxin contamination. No citrinin, ochratoxin A, patulin, sterigmatocystin, diacetoxyscirpenol, T-2 toxin or zearalenone were detected in the samples assayed.  相似文献

From 40 peanut seed samples collected in Egypt, forty-three species and one variety of fungi, belonging to 16 genera, were collected. The most dominant genera were Aspergillus (11 species + one variety), Penicillium (11 species) and Fusarium (4 species). From the preceding genera A. fumigatus, A. flavus, A. niger, P. chrysogenum and F. oxysporum were the most frequent species.Forty-nine isolates belonging to 12 species and one variety were tested for production of mycotoxins, after growth on liquid medium containing two carbon sources (sucrose or cellulose). Thin layer chromatographic analysis revealed that the quality and quantity of mycotoxins was higher on sucrose than cellulose. Mycotoxins identified were aflatoxins B1, B2, G1 & G2, citrinin; fumagillin; diacetoxyscirpenol T-2 toxin; satratoxin H; and zearalenone.  相似文献

Studies on one of the protein rich pulses, horsegram (Dolichos biflorus L.) were carried out to know how far these low risk pulses are free from aflatoxin contamination under severe insect infestation in storage. A total of 150 stored seed samples of horsegram were analyzed for the presence of aflatoxins by collecting 25 samples each of undamaged and insect damaged seeds of all the three varieties (PDM-1, PHG-1 and HG-96). More than 33% of insect damaged seed samples were contaminated with aflatoxin B1 and B2, whereas less than 8% of the undamaged seed samples contain only low levels of aflatoxin B2. Higher levels of aflatoxin B1 (up to 130 μg/kg) were reported in insect damaged seed samples of all the three varieties under study. The levels of aflatoxin B2 were always lower than aflatoxin B1 of the corresponding seed samples with insect damage. Aflatoxin B1 was reported in both the undamaged and insect damaged seed samples of all the three varieties of horsegram. It is evident from the varietal response studies that PDM-1 and HG-96 varieties of horsegram are highly vulnerable to aflatoxin contamination whereas, PHG-1 variety is relatively less susceptible to it. In general, insect infestation leads to increase in fungal invasion (including aflatoxigenic fungi) and this further enhances the levels of aflatoxin contamination in horsegram seeds.  相似文献

Seed sample of lentil collected from the Swabi district were treated with NaCl and KCl at 0.01, 0.1 and 1.0% (w/w). Seed-borne mycoflora was observed at 0, 20, 40, 60 and 80 day intervals. Seed treatment with both the salts was found to be effective against storage fungi; however, KCl was more efficient against storage mycoflora such as Aspergillus species when compared with NaCl. With the passage of time, the incidence of deep-seated fungi was observed in salt-treated seed samples while untreated seed sample showed heavy infestation by the species of Aspergillus. Seed viability also remained unaffected in storage, except in seeds heavily infested with seed-borne mycoflora. Aspergillus spp. were the main cause of seed rot. Surface sterilisation of seeds with 1% Na(OCl)2 reduced the fungal infestation of seeds. Among various concentrations of salts, 0.1% (w/w) of both salts were better in controlling seed-borne mycoflora.  相似文献

Twenty-six genera and Sixty-six species, in addition to two varieties of Aspergillus nidulans and one variety of Humicola grisea were isolated from 80 peanut seed (uncovered or covered within shell at the time of sampling) samples and 40 samples of shells collected from different places in Egypt.At 28 °C, the broadest spectrum of genera and species was recorded in the shells (21 genera and 53 species), followed by the naked seeds (16 genera and 38 species) and the covered seeds (17 genera and 33 species). Aspergillus and Penicillium were regularly the most common genera in the three substrates. A. niger, A. fumigatus, A. flavus A. terreus, A. ochraceus and P. funiculosum were generally the most common fungal species recovered from the three substrates. A. flavus was represented in 80%, 60% and 80% of the samples constituting 16.1%, 8.4% and 27.2% of the total count of fungi of the shells, covered and uncovered seeds respectively.At 45 °C, nine genera and sixteen species were collected of which Aspergillus fumigatus was extremely dominant in the three substrates, followed by A. terreus and A. niger. A total of 121 seed samples of cabbage, cauliflower, cress, radish and turnip collected from five localities in Upper Egypt were assayed for their fungal flora. The highest count of fungi was recorded on cabbage seeds (75%), whereas the lowest count was observed on turnip seeds (33%). Thirty-five fungal species and two varieties belonging to 16 genera were identified. The broadest number of species (22 species + 1 variety) were isolated from cress seeds. However, only five species,viz. Aspergillus flavus, A. niger, Penicillium chrysogenum, P. funiculosum andRhizopus stolonifer, were found to be associated with seeds of the five plants. The relative efficacy of aqueous seed extracts against these five fungal species was tested. The antifungal drug Trosyd (tioconazole) was taken as a standard inhibitor. The aqueous extract of cabbage seeds inhibited three fungal species, cress extract two and cauliflower and turnip one species; radish seed extract was ineffective.  相似文献

A survey was carried out to obtain data on the occurrence of mycotoxins and the mycotoxin-producing potential of fungi isolated from nuts (almonds, peanuts, hazelnuts, pistachio nuts) and sunflower seeds in Spain. Thin-layer chromatography was used to separate the toxins. Aflatoxins were detected in one sample of almonds (95 ppb aflatoxin B1 and 15 ppb aflaxtoxin B2) and in one sample of peanuts at a level below 10 ppb of aflatoxin B1. 100% of samples showed variable incidence of fungal contamination. The predominant fungi present in samples were Penicillium spp, Aspergillus niger, A. flavus, A. glaucus and Rhizopus spp. The results showed that isolates of different species were able to produce aflatoxins B1, B2, G1, and G2, sterigmatocystin, ochratoxin A, patulin, citrinin, penicillic acid, zearalenone, and griseofulvin.  相似文献

A study has been carried out in Argentina on samples of corn genotypes from a breeding station as well as in commercially available corn meal. All samples were analyzed for fungal infection and aflatoxin B1.Mycological analysis of corn genotypes showed the presence of three principal genera of filamentous fungi Fusarium (100%), Penicillium (67%) and Aspergillus (60%). In the genus Fusarium three species were identified, F. moniliforme (42%), F. nygamai (56%) andF. proliferatum (1.8%). Eight species ofPenicillium were identified, the predominant species isolated were P. minioluteum, P. funiculosum and P. variabile. In the genus ranked third in isolation frequency, two species were identified, A. flavus and A. parasiticus, the percentage of infection was 78% and 21%, respectively. Only one corn genotype was contaminated with aflatoxin B1 at a level of 5 ppb. The cornmeal samples showed great differences in fungal contamination, the values ranging from 1 × 101 to 7 × 105 cfu g–1. Fusarium (68%), Aspergillus (35%) and Penicillium (21%) were the most frequent genera isolated. Among the genus, Aspergillus, A. parasiticus (38%) was the most frequent species isolated. All the samples of corn meal were negative to aflatoxin B1. These results indicate a low degree of human exposure to aflatoxins in Argentina through the ingestion of maize or corn meal.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

A PCR-based marker technique was developed to discriminate between morphologically similar split seed of vetch (Vicia sativa) and lentil (Lens culinaris subsp.culinaris). Sequence tagged microsatellite site (STMS) markers were more discriminatory than markers produced from the nontranscribed spacer (NTS) region of the 5S ribosomal RNA gene. A sequence characterized amplified region (SCAR) marker, developed from the 5S rRNA NTS region, was sensitive when resolved on agarose. However, the fluorescent-labeled 5S rRNA SCAR marker was unable to discriminate between vetch and lentil, probably because of the low copy number of the marker, and was not visualized on agarose. An STMS primer-pair (PSMPSAD123), developed from field pea, was able to discriminate split red cotyledon vetch from split red cotyledon lentil because it produced specific markers at 563 bp for lentil and 353 and 474 bp for vetch. The vetch-specific STMS marker was conserved among all species of theVicia genus used in this study and was sensitive enough to discriminate both on agarose gels and on polyacrylamide gel-based fluorescent systems. The fluorescent-tagged STMS analysis revealed peaks for vetch and lentil at the expected sizes in admixtures of milled vetch and lentil seeds, and it was sensitive enough to detect one vetch seed in 1999 lentil seeds. The development of PCR-based tests for detecting the level of vetch seed contamination in lentil export seed may provide a method for quality assurance of export lentil seed.  相似文献

Using the dilution plate method, 70 species and 31 genera were collected from 20 dust samples on glucose (28 genera and 64 species) and cellulose Czapek's agar (22 genera and 46 species) at 28° C. The most common fungi were Aspergillus niger, A. flavus, A. flavus var. columnaris, Phoma glomerata, Fusarium oxysporum, Penicillium chrysogenum and Mucor racemosus; and A. nidulans, Phoma humicola, Drechslera spicifera and Stachybotrys chartarum on the two media, respectively.Toxicity test showed that about 85% of the isolates tested were toxic to brine shrimp (Artemia salina). Thin layer chromatographic analysis revealed that 13 out of 23 toxic isolates produced known mycotoxins. Toxins identified were: aflatoxins B1 and B2, Kojic acid and trichodermine.  相似文献

The fungi of 40 samples of barley grains collected from Upper Egypt were estimated using the dilution- and the grain-plate methods. 33 genera and 109 species, in addition to 2 varieties of Aspergillus nidulans, one variety of Aspergillus flavus and one variety of Penicillium cyclopium were identified. The spectrum of fungal genera and species collected by the grain-plate method (29 genera and 94 species+3 varieties) was broader than by the dilutionplate method (27 genera and 71 species+3 varieties), but the order of frequency of occurrence of fungal genera and species was basically similar in the two methods. The most frequent genera were Aspergillus (25 species+3 varieties), Penicillium (32 species+1 variety), Rhizopus (2 species), Alternaria (2 species), Fusarium (6 species) and Drechslera (5 species) and were represented (as assayed by the grainplate method) in 100 %, 77.5 %, 85 % 62,5 %, 60 % and 52.5 % of the samples constituting 60.4 %, 10.2 %, 7.7 %, 3.7 %, 5 % and 2.3 % of the gross total count of fungi respectively. From the preceding genera A. niger, A. fumigatus, A. flavus, A. sydowii, P. notatum, P. citrinum, F. oxysporum, F. moniliforme, F. solani, R. stolonifer, A. alternata and D. spicifera were the most frequent.  相似文献

Presence of Neotyphodium-like endophytes in European grasses   总被引:1,自引:0,他引:1  
In order to improve knowledge of Neotyphodium‐likc fungi in European grasses, microscopic detection of endophytic mycelium was achieved in living grasses and in seeds. Samples of grasses were gathered near permanent pastures or along paths in France (93 samples, 13 genera, 22 species). Typical convoluted mycelium was found inside leaf sheaths of four genera, especially in Festuca and Lolium spp. The results suggest that endophytes were present most often in the southern regions where plants may suffer from summer drought. Endophytes were looked for in 489 seed samples (63 genera, 237 species) received from 24 European botanical gardens. Neotyphodium was found in seeds of only six genera (22 species), especially in Festuca (15 species) and Lolium (three species). No endophytic mycelium was found in the other 215 species, although other work had demonstrated the presence of the fungus in 39 of the species analysed. Neotyphodium‐likc mycelium was detected for the first time in Festuca juncifolia, F. trachyphylla, F. vaginata, F. pseudodalmatica, F. rupicaprina, F. arenaria, Vulpia ciliata and Micropyrum tenellum. Further studies are needed for the correct identification of the Neotyphodium species observed.  相似文献

Aflatoxin contamination in five varieties of pearl millet (ICMH-451, ICMP-50I, ICTP-8203, WCC-75 and ICMV-155) was studied from field and storage conditions in three districts of Andhra Pradesh State, India and the inter-relationships between various parameters such as stage of grain maturation in the field and insect pest infestation in storage in relation to aflatoxin production were evaluated. Aflatoxin contamination was more frequent in the seed samples collected from the fields during rainy season than winter season. All major aflatoxins were isolated from one or the other varieties of pearl millet, whereas aflatoxin G2 was not commonly observed in the seed samples collected during winter. Among all the varieties tested, ICMH-451 was vulnerable to aflatoxin contamination whereas ICMV-155 was the least susceptible variety. The higher amount of aflatoxins was observed in the matured seed samples followed by pre-matured and milky stage. Among all the toxins reported in the field, aflatoxin B1 was found in higher concentration (185 (μg/kg) followed by B2 (105 μg/kg). The four major types of aflatoxins with higher levels (35, 40, 140, 190 μg/kg of G1, G2, B2, B1 were reported in the rainy season seed samples after six months of storage, whereas aflatoxin G1 was not observed in any variety of stored seed sample from winter. Statistical analysis revealed that the aflatoxin incidence in relation to different parameters studied was significantly different for each factor. The relationship between aflatoxin contamination and insect damaged-grain clearly indicated that the seed samples with 16-40% of insect damage contained higher amounts of aflatoxins (758 μg/kg). Fungi of 19 genera, 30 species, and one variety were isolated from 25 samples of sheep-, cattle- and camel feedstuffs collected from different farms in the Beida Governorate, Libya.Aspergillus, Penicillium andFusarium were the most common genera in the three substrates tested. TLC was used to establish the identity of aflatoxins in the chloroform extract of all samples and the ability to produce aflatoxins byAspergillus flavus in a synthetic liquid medium. Twenty samples out of 25 tested were naturally contaminated and 21 isolates ofA. flavus out of 30 produced at least one of the following aflatoxins: B1; B1, G1; and B1, B2, G1, G2. Sixty-four species and 2 varieties which belong to nineteen genera of fungi were identified from 40 peanut seed samples collected from different places in Egypt by using a dilution-plate method on glucose-Czapek's medium. The most frequent genera were Aspergillus (21 species & 2 varieties), Penicillium (16 species) and Fusarium (6 species). A. flavus, A. fumigatus, A. niger, P. chrysogenum and F. oxysporum were the most common fungal species. Forty seven percent of the samples proved to be toxic to brine shrimp (Artemia salina) larvae. Thin-layer Chromatographic analysis revealed that peanuts were contaminated by aflatoxins (11 samples), thrichothecene-toxins (10 samples), zearalenone (one sample) and citrinin (one sample). Brewing and consumption of traditional beer have social–economic significance in most African countries including Botswana. Traditional sorghum malt, wort, and beer samples were collected from three villages around Gaborone, Botswana. Forty-six malt samples were analyzed for fungi on three different media and developing colonies were subcultured for identification. Rhizopus, Fusarium, Mucor, and Aspergillus were the most common genera isolated. Out of the 46 malt samples, 72% contained Rhizopus stolonifer, 63% Fusarium verticillioides (syn. Fusarium moniliforme), and 37% Aspergillus flavus. Although Aspergillus flavus was isolated from malt samples, aflatoxins (B1, B2, G1, and G2) were not detected in any of the samples analyzed. When the malt, wort, and beer samples were analyzed for fumonisin Bl and zearalenone, fumonisin B1 was detected in 3 malt samples, with concentrations ranging from 47 to 1316 μg/kg, while zearalenone was detected in 56%, 48% and 48% of the malt, wort and beer samples, respectively. Zearalenone concentration in samples ranged from 102 to 2213 μg/kg in malt, 26 to 285 μg/l in wort and 20 to 201 μg/l, in beer. Zearalenone carry-over from wort to beer ranged from 23 to 403%. Therefore, although aflatoxins and fumonisin B1 do not appear to be major contaminants, zearalenone is common and could pose a potential problem in traditional beer in Botswana.  相似文献

High incidence of diseases associated with the use of seeds saved from previous harvests as desire of maintaining local varieties with special attributes is of increased concern in wheat industry worldwide. Prevalent of seed-borne fungi in farmer-saved seeds and seed dressing fungicides to prevent infection from seeds to seedlings was studied in Northern Tanzania. One hundred and thirty five untreated farmer-saved seed lots were collected randomly from farmers. Alternaria alternata, Bipolaris sorokiniana, Drechslera tritici, Fusarium graminearum, Fusarium moniliforme, Aspergillus flavus, Cladosporium sphaerospermum, Epicoccum purpurascens, Pyricularia oryzae and Penicillium corylophilum were fungi isolated in farmer-saved seeds. Mean seed infection was 29% causing average grain yield loss of 1.2 mt/ha?1. Seed dressing with Metalaxy plus (Methyl carboxaitide), Mancozeb (Manganase-zinc salt) and Baytan (Chlorophenoxy ethanol) increased seed germination by 14, 13 and 17%, respectively, and grain yield by 28, 20 and 18%, respectively. Farmer-saved seeds were heavily infected by fungi with low grain yield performance.  相似文献

Forty-four fungal species belonging to 20 genera were isolated from 30 samples of qat leaves. The most frequent genera wereAspergillus, Alternaria, Penicillium, andCladosporium followed byFusarium, Drechslera, Chœtomium, andMucor. The most prevalent species in above genera wereAspergillus niger, A. flavus, A fumigatus, Alternaria alternata, Penicillium chrysogenum, P. citrinum, Cladosporium cladosporioides, andFusarium verticillioides. From these fungi, 17 species (39%) related to 7 genera (35%) proved to be true endophytes. Eleven out of 75 isolates were mycotoxigenic.A. alternata produced alternariol and alternariol monomethyl ether whereasA. flavus produced aflatoxins B1 and B2. Ochratoxin A, sterigmatocystin, citrinin and T-2 toxin were produced byA. ochraceus, A. versicolor, P. citrinum andF. oxysporum, respectively. The presence of such toxigenic fungi associated with qat leaves is considered to be a threat to public health.  相似文献
