Summary Patch-clamp techniques were used to study the permeability to ions of an ATP-sensitive channel in membranes from the pancreatic B-cell line (RINm5F). With patches in the outside-out configuration, theI-V curves for different Na+–K+ mixtures in the bath and 140 mM K+ in the pipette were almost linear, and crossed the zero-current axis at voltages that indicated a variable permeability ratio. When K+ was added symmetrically, the plot of the conductancevs. K+ activity exhibited saturation, with aG max of about 160 pS and a half-maximal activity of 216 mM. TheI-V behavior for different K+–Na+ mixtures in the bath could be accurately described with a model based on Eyring theory, assuming two sites and one-ion occupancy. For K+, the dissociation constants (KK) of the two sites were 290 and 850 mM, the lower value pertaining to the site close to the intracellular medium. In experiments with inside-out patches, both Na+ and Mg2+, when present in the bath, induced a voltagedependent block of the outward current. Fitting the data with the model suggested that for these ions only one of the two sites binds significantly, the corresponding dissociation constants being (mM): 46 for Na+ and 34 for Mg2+. Blocking by Na+ and Mg2+ may account for the low outward current seen in intact cells. This hypothesis is consistent with the observation that such current is further reduced by addition of 2,4-DNP, since metabolism inhibitors are expected to lower the ATP level, thereby liberating Mg2+ from the Mg2+-ATP complex, as well as inducing accumulation of Na+ by decreasing the rate of the Na+–K+ pump.  相似文献

ATP-dependent Ca2+ uptake distinct from that of the mitochondria is found in both plasma membrane and microsomal membranes of rat kidney. Activity attributed to these fractions is enhanced by ammonium oxalate and is apparently insensitive to NaN3. In contrast, rat kidney mitochondrial Ca2+ uptake is blocked by NaN3. The pH of optimal activity is significantly higher for the mitochondrial fraction. Microsomal membrane Ca2+ uptake differs from that of the plasma membrane. Microsomal membranes are four times as active as the plasma membrane at high (5 mM) ATP levels. Apparent Km values for Mg2+-ATP differ in the two preparations with a higher affinity for Mg2+-ATP found in the plasma membrane Ca2+ uptake activity of the plasma membrane preparation is readily inhibited by Na+. Sucrose gradient density fractionation indicates that the observed microsomal membrane Ca2+ pump activity is associated with membrane vesicles derived from the endoplasmic reticulum. Ca2+ pump activity of both plasma membrane and microsomal fraction is depressed din the adrenalectomized rat. This activity is not restored by a single natriuretic dose of aldosterone.  相似文献

LowK m GTPase activity was studied in the purified membranes from theLymnaea stagnalis CNS. Stimulation of GTPase activity of G proteins by dopamine in membrane preparations is transient; it is followed by reduction of the activity. Magnesium ions regulate activity of G proteins; thus, the value of [Mg2+] i is an important factor to be taken into account in investigations of functioning of G proteins. In order to determine the mean physiological level of the neuronal [Mg2+] i , neuronal elements of the molluscan CNS were separated from other cell types. The values of 1.16±0.19 and 1.49±0.08 mM were obtained for [Mg2+] i of the neuronal cells with Mag-fura-2 and Mag-fura-red, respectively. An inhibitory effect of dopamine was displayed within the range of free Mg2+ concentrations from 3 μM to 3 mM. GTPase activity was stimulated with Na+ in a dose-dependent manner. However, GTPase activity became independent of Na+ in the presence of dopamine. Replacing of ATP by 5′-adenylyl imidodiphosphate suppressed the inhibitory effect of dopamine, but did not completely prevent the dopamine influence. The effect of dopamine was dose-dependent and did not decrease after PTX treatment. The inhibitory influence of dopamine on the GTPase activity was reversed by a selective antagonist of the dopamine D2 receptors, S(−)-sulpiride.  相似文献

The ionic influence and ouabain sensitivity of lymphocyte Mg2+-ATPase and Mg2+-(Na+ + K+)-activated ATPase were studied in intact cells, microsomal fraction and isolated plasma membranes. The active site of 5′-nucleotidase and Mg2+-ATPase seemed to be localized on the external side of the plasma membrane whereas the ATP binding site of (Na+ + K+)-ATPase was located inside the membrane.Concanavalin A induced an early stimulation of Mg2+-ATPase and (Na+ + K+)-ATPase both on intact cells and purified plasma membranes. In contrast, 5′-nucleotidase activity was not affected by the mitogen. Although the thymocyte Mg2+-ATPase activity was 3–5 times lower than in spleen lymphocytes, it was much more stimulated in the former cells (about 40 versus 20 %). (Na+ + K+)-ATPase activity was undetectable in thymocytes. However, in spleen lymphocytes (Na+ + K+)-ATPase activity can be detected and was 30 % increased by concanavalin A. Several aspects of this enzymic stimulation had also characteristic features of blast transformation induced by concanavalin A, suggesting a possible role of these enzymes, especially Mg2+-ATPase, in lymphocyte stimulation.  相似文献

The Mg2+ dependence of the kinetics of the phosphorylation and conformational changes of Na+,K+-ATPase was investigated via the stopped-flow technique using the fluorescent label RH421. The enzyme was preequilibrated in buffer containing 130 mM NaCl to stabilize the E1(Na+)3 state. On mixing with ATP, a fluorescence increase was observed. Two exponential functions were necessary to fit the data. Both phases displayed an increase in their observed rate constants with increasing Mg2+ to saturating values of 195 (± 6) s−1 and 54 (± 8) s−1 for the fast and slow phases, respectively. The fast phase was attributed to enzyme conversion into the E2MgP state. The slow phase was attributed to relaxation of the dephosphorylation/rephosphorylation (by ATP) equilibrium and the buildup of some enzyme in the E2Mg state. Taking into account competition from free ATP, the dissociation constant (Kd) of Mg2+ interaction with the E1ATP(Na+)3 state was estimated as 0.069 (± 0.010) mM. This is virtually identical to the estimated value of the Kd of Mg2+-ATP interaction in solution. Within the enzyme-ATP-Mg2+ complex, the actual Kd for Mg2+ binding can be attributed primarily to complexation by ATP itself, with no apparent contribution from coordination by residues of the enzyme environment in the E1 conformation.  相似文献

Two non mitochondrial systems involved in ATP-dependent Ca2+ accumulation have been described and characterized in two membrane fractions from pea internodes purified on a metrizamide-sucrose discontinuous gradient. In the lighter membrane fraction an ATP-dependent Ca2+ accumulation system, which shows the characteristics of an ATP-dependent H+/Ca2+ antiport, predominates. This system is inhibited by FCCP and nigericin and stimulated by 50 mM KCl. It is saturated by 0.8–1.0 mM MgSO4-ATP, strictly requires ATP and is severely inhibited by an excess of free Mg2+ or Mn2+. A second system of ATP-dependent Ca2+ accumulation, recovered mainly in the heavier membrane fraction, is insensitive to FCCP, is saturated by 8–10 mM MgSO4-ATP, can utilize also ITP or other nucleoside triphosphates although at lower rate than ATP and is only scarcely affected by an excess of free Mg2+ or Mn2+. This system is interpreted as corresponding to the (Ca2+ + Mg2+)-ATPase described by Dieter, P. and Marmé, D. ((1980) Planta 150, 1–8).  相似文献

In contrast to everted mitochondrial inner membrane vesicles and eubacterial plasma membrane vesicles, the ATPase activity of chloroplast ATP synthase in thylakoid membranes is extremely low. Several treatments of thylakoids that unmask ATPase activity are known. Illumination of thylakoids that contain reduced ATP synthase (reduced thylakoids) promotes the hydrolysis of ATP in the dark. Incubation of thylakoids with trypsin can also elicit higher rates of ATPase activity. In this paper the properties of the ATPase activity of the ATP synthase in thylakoids treated with trypsin are compared with those of the ATPase activity in reduced thylakoids. The trypsin-treated membranes have significant ATPase activity in the presence of Ca2+, whereas the Ca2+-ATPase activity of reduced thylakoids is very low. The Mg2+-ATPase activity of the trypsinized thylakoids was only partially inhibited by the uncouplers, at concentrations that fully inhibit the ATPase activity of reduced membranes. Incubation of reduced thylakoids with ADP in Tris buffer prior to assay abolishes Mg2+-ATPase activity. The Mg2+-ATPase activity of trypsin-treated thylakoids was unaffected by incubation with ADP. Trypsin-treated membranes can make ATP at rates that are 75–80% of those of untreated thylakoids. The Mg2+-ATPase activity of trypsin-treated thylakoids is coupled to inward proton translocation and 10 mM sulfite stimulates both proton uptake and ATP hydrolysis. It is concluded that cleavage of the γ subunit of the ATP synthase by trypsin prevents inhibition of ATPase activity by the ε subunit, but only partially overcomes inhibition by Mg2+ and ADP during assay.  相似文献

An ATPase activity stimulated by divalent ions (Mg2+, Ca2+, Mn2+, Zn2+) has been observed in intact hamster fibroblasts cultured in vitro (BHK line). Such activity has been determined by the incubation (30 min at 37°C) of washed cell suspensions (about 1 mg of proteins) in a medium containing 100 mM NaCl, 20 mM KCl, 15 mM Tris—HCl (pH 7.4), 10 mM NaHCO3, 5 mM glucose and equimolar concentrations of ATP and divalent cation. Mg2+-ATPase activity is insensitive to ouabain and lacks specificity towards nucleoside triphosphate substrates. AMP and ADP are not hydrolyzed under these conditions. Apparent Km of 0.76 mM and Vmax of 1.46 μmol Pi · mg proteins?1 · h?1 have been calculated for Mg-ATP complex. This ATPase is an ectoenzyme, therefore its activity could be used as a suitable index of the action of chemicals like chromium compounds known for their cytotoxic effects on membrane functions.Salts of trivalent (CrCl3) and hexavalent (K2Cr2O7) chromium at concentrations ranging from 1 mM to 5 mM inhibit Mg2+-ATPase. The inhibition by K2Cr2O7 is observed after pretreatment of the cells with this compound followed by its absence from the assay medium “per se” for Mg2+-ATPase, and it is referred to the alterations of membrane bound enzyme structures by the oxidizing hexavalent chromium. The inhibition by CrCl3 is mainly evident when this compound is present in the incubation medium, and is referred to the interaction of trivalent chromium with Mg2+-ATP as it is partially reversed by increasing Mg2+-ATP concentration.  相似文献

 The membrane-bound F1 sector of the H+–ATPase complex (F-type ATPase) in dark-adapted photosynthetic chromatophores is endowed with MgATP- and CaATP-dependent ATPase activities, both sensitive to inhibitors such as oligomycin and venturicidin. Because of contatamination of free Mg2 + and Ca2+ ions in chromatophore preparations, kinetic characterization of the two hydrolitic reactions can be performed only in the presence of both substrates, using a model for two alternative substrates. The two activities are characterized by similar maximal rates and affinity constants [VMgATP and VCaATP: 13±1 and 10±1 nmol s–1 ATP hydrolyzed (μmol BChl)–1; KMgATP and KCaATP: 0.22±0.06 and 0.20±0.05 mm]. However, only the MgATP-dependent ATPase is coupled to Δ*H + generation. In this process CaATP acts as an alternative substrate and a competitive inhibitor of the proton pump, with a KI coincident with KCaATP for the hydrolytic activity. This finding highlights the central role that the coordination chemistry of the ion-nucleotide complex plays in determining the proton gating mechanism at the catalytic site(s) of the enzyme complex. These results are discussed on the basis of the coordination properties of the ions and of the available information on the protein structure. Electrophoretic measurements on membrane coated particles were performed with a Zytopherometer. Tris-HCl buffer 0.2 M pH 7.0 at 37°C with addition of different combinations of Na+, K+, Mg2+ and ATP was used as test medium. The membranes were of two types, an untreated preparation with low NaK ATPase activity and a deoxycholate treated preparation with high NaK ATPase activity. There was no marked difference in reaction between the two types of membranes. To both types of membranes Mg2+ gave a strong positive and ATP a slight negative addition to the membrane charge. In the presence of ATP Na+ gave a higher charge contribution than did K+ or a combination of Na+ and K+. This implies that K+ gives a higher affinity for ATP than Na+ does and or that ATP mediates a higher affinity for Na+ than for K+.  相似文献

ATP and adenylylimidodiphosphate (AdoPP[NH]P) bind to (Na+ + K+)-ATPase in the absence of Mg2+ (EDTA present) with a homogeneous but 15-fold different affinity, the Kd values being 0.13 μM and 1.9 μM, respectively. The binding capacities of the two nucleotides are nearly equal and amount to 3.9 and 4 nmol/mg protein or 1.7 and 1.8 mol/mol (Na+ + K+)-ATPase, respectively. The Kd value for ATP is equal to the Km for phosphorylation by ATP (0.05–0.25 μM) and the binding capacity is equivalent to the phosphorylation capacity of 1.8 mol/mol (Na+ + K+)-ATPase. Hence, the enzyme contains two high-affinity nucleotide binding and phosphorylating sites per molecule, or one per α-subunit. Additional low-affinity nucleotide binding sites are elicited in the presence of Mg2+, as shown by binding studies with the non-phosphorylating (AdoPP[NH]P). The Kd and binding capacity for AdoPP[NH]P at these sites is dependent on the Mg2+ concentration. The Kd increases from 0.06 mM at 0.5 mM Mg2+ to a maximum of 0.26 mM at 2 mM Mg2+ and the binding capacity from 1.5 nmol/mg protein at 0.5 mM Mg2+ to 3.3 nmol/mg protein at 4 mM Mg2+. Extrapolation of a double reciprocal plot of binding capacity vs. total Mg2+ concentration yields a maximal binding capacity at infinite Mg2+ concentration of 3.8 nmol/mg protein or 1.7 mol/mol (Na+ + K+)-ATPase. The Kd for Mg2+ at the sites, where it exerts this effect, is 0.8 mM. The Kd for the high-affinity sites increases from 1.5–1.9 μM in the absence of Mg2+ to a maximum of 4.2 μM at 2 mM Mg2+ concentration. The binding capacity of these sites (1.8 mol/mol enzyme) is independent of the Mg2+ concentration. Hence, Mg2+ induces two low-affinity non-phosphorylating nucleotide binding sites per molecule (Na+ + K+)-ATPase in addition to the two high-affinity, phosphorylating nucleotide binding sites.  相似文献

The ADP-dependent phosphofructokinase (PFK) from Thermococcus zilligii has been purified 950 fold; it had a specific activity of 190 U mg−1. The enzyme required Mg2+ ions for optimal activity and was specific for ADP. The forward reaction kinetics were hyperbolic for both cosubstrates (pH optimum of 6.4), and the apparent K m values for ADP and fructose-6-phosphate were 0.6 mM (apparent V max of 243 U mg−1) and 1.47 mM (apparent V max of 197 U mg−1), respectively. Significantly, the enzyme is indicated to be nonallosteric but was slightly activated by some monovalent cations including Na+ and K+. The protein had a subunit size of 42.2 kDa and an estimated native molecular weight of 66 kDa (gel filtration). Maximal reaction rates for the reverse reaction were attained at pH 7.5–8.0, and the apparent K m values for fructose-1,6-bisphosphate and AMP were 0.56 mM (apparent V max of 2.9 U mg−1) and 12.5 mM, respectively. The biochemical characteristics of this unique ADP-dependent enzymatic activity are compared to ATP and pyrophosphate-dependent phosphofructokinases. The action of ATP and its analogs as well as the effects of alkali ions were studied in their action on the ouabain receptor. One single ouabain receptor with a dissociation constant (KD) of 13 nM was found in the presence of (Mg2+ + Pi) and (Na+ + Mg2+ + ATP). pH changes below pH 7.4 did not affect the ouabain receptor. Ouabain binding required Mg2+, where a curved line in the Scatchard plot appeared. The affinity of the receptor for ouabain was decreased by K+ and its congeners, by Na+ in the presence of (Mg2+ + Pi), and by ATP analogs (ADP-C-P, ATP-OCH3). Ca2+ antagonized the action of K+ on ouabain binding. It was concluded that the ouabain receptor exists in a low affinity (Rα) and a high affinity conformational state (Rβ). The equilibrium between both states is influenced by ligands of (Na+ + K+)-ATPase. With 3 mM Mg2+ a mixture between both conformational states is assumed to exist (curved line in the Scatchard plot).  相似文献

A model of the active transport of ions through internal membranes of mitochondria is proposed. If concentrations of ions in a cell are known, this model allows calculating concentrations of all main ions (H+, Ca+2, K+, Mg2+, Na+, Cl) in the mitochondrion matrix and the resting potential across the membrane. The theoretical values satisfactorily agree with available experimental data on the concentrations and the potentials, including different operating regimes of the adenosine triphosphate (ATP) synthetase (the main regime, short circuiting or ATP synthetase blocking). The active transport of Mg2+ ions in exchange for protons was assumed. In accordance with the model, the ATP synthetase operation is possible only if the stoichiometric coefficient of protons is 3.  相似文献

The gene encoding homodimeric β-galactosidase (lacA) from Bacillus licheniformis DSM 13 was cloned and overexpressed in Escherichia coli, and the resulting recombinant enzyme was characterized in detail. The optimum temperature and pH of the enzyme, for both o-nitrophenyl-β-d-galactoside (oNPG) and lactose hydrolysis, were 50°C and 6.5, respectively. The recombinant enzyme is stable in the range of pH 5 to 9 at 37°C and over a wide range of temperatures (4–42°C) at pH 6.5 for up to 1 month. The K m values of LacA for lactose and oNPG are 169 and 13.7 mM, respectively, and it is strongly inhibited by the hydrolysis products, i.e., glucose and galactose. The monovalent ions Na+ and K+ in the concentration range of 1–100 mM as well as the divalent metal cations Mg2+, Mn2+, and Ca2+ at a concentration of 1 mM slightly activate enzyme activity. This enzyme can be beneficial for application in lactose hydrolysis especially at elevated temperatures due to its pronounced temperature stability; however, the transgalactosylation potential of this enzyme for the production of galacto-oligosaccharides (GOS) from lactose was low, with only 12% GOS (w/w) of total sugars obtained when the initial lactose concentration was 200 g/L.  相似文献

A method for the isolation of guinea pig ileum smooth muscle cell membranes is described. The plasma membrane fraction possessed a (Na+, K+)-ATPase which was inhibitied by ouabain. The Mg2+-dependent ATPase of the membrane fraction was stimulated by 1 μM Ca2+. A basal ATPase, not dependent on Mg2+, was directly stimulated by Ca2+ in the range of 1 μM to 1 mM.The isolated membranes contracted in response to the following substances: ATP, angiotensin II and some of its analogs, bradykinin, acetylcholine and histamine. The contractility was inhibited by ouabain and chlorambucil-angiotensin II, but not by cytochalasin B. No contraction was produced by AMP, angiotensin I and adrenaline.  相似文献

ATPases of cardiac cells are known to be among the most important enzymes to maintain the fluxes of vital cations by hydrolysis of the terminal high-energy phosphate of ATP. Biochemically the activities of Ca2+-pump ATPase, Ca2+/Mg2+-ecto ATPase, Na+,K+-ATPase and Mg2+-ATPase are determined in homogenates and isolated membranes as well as in myofibrillar and mitochondrial fractions of various purities. Such techniques permit estimation of enzyme activitiesin vitro under optimal conditions without precise enzyme topography. On the other hand, cytochemical methods demonstrate enzyme activityin situ, but not under optimal conditions. Until recently several cytochemical methods have been employed for each enzyme in order to protect its specific activity and precise localization but the results are difficult to interpret. To obtain more consistent data from biochemical and cytochemical point of view, we modified cytochemical methods in which unified conditions for each ATPase were used. The fixative solution (1% paraformaldehyde –0.2% glutaraldehyde in 0.1 M Tris Base buffer, pH 7.4), the same cationic concentrations of basic components in the incubation medium (0.1 M Tris Base, 2mM Pb(NO2)3, 5 mM MgSO4, 5 mM ATP) and selective stimulators or inhibitors were employed. The results reveal improved localization of Ca2+-pump ATPase, Na+–K+ ATPase and Ca2+/Mg2+-ecto ATPase in the cardiac membrane.  相似文献

The chemical composition of rainwater is altered upon its passage through tree canopies. In order to investigate how rainwater chemistry is affected by canopy-dependent processes in characteristic forest types of Northwest German sandy lowland regions – oak–birch-forests, Betula pubescens Ehrh. swamp forests, and stands of Pinus sylvestris L. – comparative studies on the chemical composition of throughfall were carried out at seven forest sites, situated in close proximity within a nature reserve. Additionally, rainwater was sampled at three heathland sites for analysis of open-field precipitation and at three sites along an oak–birch-forest edge. Throughfall concentrations of most of the parameters analysed were significantly higher than open-field concentrations, especially with regard to electric conductivity, NH4-N, K+, and KMnO4-index. Ion concentrations in throughfall were the lowest in a 10-year-old stand of Betula pendula Roth. and Pinus sylvestris and in a Betula pubescens swamp forest and were highest beneath a stand of Pinus sylvestris. Except for Na+, Cl, and NO3, ion concentrations in both throughfall and open-field precipitation increased during the growing season (May–October). In throughfall, Ca2+, Mg2+, K+, and Mn2+ were strongly correlated. Enrichment ratios between throughfall and open-field deposition varied among sites and elements and were the highest for K‰+, Mg2‰+, and Mn2‰+. Estimates of canopy leaching indicated high leaching rates of K‰+ and Mn2‰+ and moderate leaching of Mg2‰+. The contribution of foliar leaching to throughfall deposition was higher at the deciduous than at the coniferous stands.  相似文献

Isolated hepatocytes in physiological [Na+] 0 tightly maintain [Mg2+] i . Upon β-adrenergic stimulation or in the presence of permeable cAMP, hepatocytes release 5–10% (1–3 mM Mg2+) of their total Mg2+ content. However, isolated basolateral liver plasma membranes (bLPM), release Mg2+ in the presence of [Na+] o even in the absence of catecholamine stimulation. The data indicate that a physiological brake for Mg2+ efflux is present in the hepatocyte and is removed upon cellular signaling. In contrast, this regulation “brake” is absent in purified bLPM thus rendering them fully active. The present study was carried out to reconstruct the missing regulatory component. Activation of Mg2+ extrusion in intact cells is consistent with cAMP dependent phosphorylation of the transporter or a regulatory protein. Treatment of bLPM with a non-specific phosphatase such as alkaline phosphatase (AP), decreased Mg2+ efflux by 70% compared to untreated bLPM. When AP-treated bLPM were loaded with protein kinase A (PKA), and stimulated with permeable cAMP, Mg2+ transport fully recovered. These data suggest that phosphorylation of the Na+/Mg2+ exchanger or a nearby protein activates the Mg2+ transport mechanism in hepatocytes.  相似文献
