以木薯干片,木薯淀粉,玉米淀粉的水解糖为原料,采用好氧深层发酵法进行甘油发酵。以木薯干片为原料时,甘油产率最高为11-12%,全糖转化率为47-49%,发酵周期为60-66h;而以玉米淀粉为原料,甘油产率仅8-9%,全糖转化率38-40%,发酵周期却需72小时以上  相似文献

随首市场对甘油(丙三醇)的需求量不断增加,且皂化甘油产量又不断下降,为缓解供需矛盾,开发甘油新来源,以淀粉为原料经酶法液化、糖化后,以亚硫酸盐诱导进行甘油发酵,再经处理分离、精制,能获得较好的甘油产品。本文介绍淀粉发酵法制取甘油的工艺及化学原理。  相似文献

对玉米芯稀硫酸水解条件及糖化液发酵L-乳酸进行了初步研究。结果表明,玉米芯木聚糖最适水解条件为2%H2SO_4、120℃、30 min、固液比1:10,糖化液还原糖含量可达40.8 g/L,主要成分为木塘。细菌A-19可以利用水解液中的葡萄糖和木糖产酸,最适发酵条件为45℃、pH 6.5,从45℃~51℃、pH 5.5~pH 6.5产量均较高。用未浓缩的水解液发酵24 h,L-乳酸产量为30.6g/L,残糖为1.6 g/L,糖酸转化率为82.6%;用浓缩1倍的水解液发酵48 h,L-乳酸产量为41.4 g/L,残糖4.1g/L,糖酸转化率为68.2%,在发酵48 h后继续补料发酵至72 h(补料液为浓缩3倍的水解液),L-乳酸产量为50.9 g/L,残糖6.3 g/L,糖酸转化率为71.8%。该研究为利用木质纤维素生产L-乳酸奠定了一定基础。  相似文献

生物质是一种可再生资源,生物质发酵可产生高端化工产品.本文主要探讨蒸汽爆破处理玉米秸秆及水解可发酵单糖,考察酵母发酵玉米秸秆糖化液产麦角甾醇的应用研究.实验结果表明:当固液比10%,盐酸浓度1.5%,90℃水解反应3 h,还原糖含量达到53.3%,纤维素转化率79%.发酵工艺参数为玉米秸秆糖化液6.0°Bx,玉米浆4%,pH 7.5,接种量10%,28℃摇床振荡培养32 h,细胞生物量达8.5 g/L,麦角甾醇含量可达2.35%.同时对玉米秸秆发酵产麦角甾醇晶体进行结构表征.  相似文献

玉米皮作为玉米淀粉加工的副产物,是一种可用于生产液体燃料的潜在廉价优质的生物质资源。本文以玉米皮为原料,对拜氏梭菌发酵生产丁醇进行了研究。实验结果表明,玉米皮首先在最优的预处理温度140℃下使用0.5%硫酸水溶液以固液比1∶8处理20 min,再添加200 IU/g底物糖化酶、1.0 IU/g底物木聚糖酶进行酶解,可以使原料中的淀粉和半纤维素转化为可发酵糖,此时水解液中的总糖浓度为50.46 g/L。然后使用1.0%的活性炭对水解液进行脱毒处理以去除发酵抑制物,再进行丁醇发酵,丁醇产量为9.72 g/L,总溶剂产量可达14.09 g/L,糖醇转化率为35.1%。上述研究结果证明玉米皮作为一种粮食加工废弃物用于液体燃料丁醇的生产在技术上是完全可行的。  相似文献

从北京西郊森林土壤中分离到产生糖化生淀粉的葡糖化酶的特异腐质霉Humicola insolens HG‐618菌株。优化的产酶培养基如下:6%麦麸、2%玉米粉、0.7%磷酸氢二铵和1%玉米浆,p H 8.0,250m L三角瓶装40m L培养基,在40℃和280r/min培养90h。以生玉米淀粉为底物,发酵液中葡糖化酶活力为180U/m L。酶学性质试验表明,该酶的最适反应p H为6.0,最适反应温度为65℃;在p H 6.0–9.0范围酶活力稳定;酶热稳定性强,在60℃和p H 6.0保温8h,酶活力仍保留83%。一步糖化生玉米粉试验表明,在50m L 30%浓度生玉米粉浆和270U HG‐618葡糖化酶的反应体系中,采用自然p H 6.7,在55℃和100r/min条件下糖化40h。获得的糖化液还原糖浓度为17.1%(由酶产生的还原糖浓度为15.9%),生玉米粉中所含淀粉的糖化率为90%,且产生的糖全部为葡萄糖。因此,该葡糖化酶具有良好的一步糖化生玉米粉前景,它也可以用于生大米粉、生小麦粉和生马铃薯粉的直接糖化。  相似文献

本研究利用国内常用的糖化酶制剂糖化生玉米面中的淀粉,同时接种酵母菌,在30℃下,探讨了玉米淀粉的高浓度酒精发酵工艺。选择到了一株产高浓度酒精酵母菌,H0菌株。发酵温度为30℃、pH4一s、加糖化酶量为每克原料300单位、酵母接种量3%(v/v)和原料加量为33.0%(w/v)时,这株酵母菌在70小时内可产生17.5%(v/v)的乙醇。如果原料加量为36.0%(w/v)时,该菌株在96小时内可以产生18.O%(v/v)的乙醇。在前一种加料情况下,成熟发酵醪中的pH为5、残还原糖为O.19%、残总糖为3.5“。在后一种加料情况下,成熟发酵醪中的pH为5、残还原糖为0.81%、残总糖为5.1%。  相似文献

本文报道综合利用野生薯芋属植物盾叶薯芋(Dioscorea zingibensisC.H.wright)和穿龙薯芋(Dioscorea nipponica Makino)中的淀粉生产肌苷(Inosine)的方法。将薯芋原料带水磨碎,在水中筛分得到皂甙淀粉浆,用稀酸水解该物质使淀粉糖化,分离后得到糖液和糖渣。糖液加氮源和无机盐,发酵生产肌苷。糖渣再水解提取薯芋皂甙元。  相似文献

用中温蒸煮工艺生产高浓度酒精,首先利用耐高温α-淀粉酶在9s~97℃下同时糊化和液化淀粉.接着在60℃下加高转化率的糖化酶进行糖化,最后在30℃下加酵母菌悬液进行发酵。酵母菌w4在60h内可以产生18.3%的乙醇,在成熟发酵醪中的残还原糖和总糖分别为1.2%和4.1%,细胞存活率为68.5%。如果在发酵培养基中添加一定量的硫酸铵,可进一步改进这一工艺,使成熟发酵醪的乙醇浓度提高到18.9%,发酵周期缩短到50h,残还原糖和总糖分别减少到0.27%和3.1%。  相似文献

Corn hulls and corn germ meal were both evaluated as feedstocks for production of ethanol for biofuel. Currently, these fibrous co-products are combined with corn steep liquor and the fermentation bottoms (if available) and marketed as cattle feed. Samples were obtained from wet and dry corn mills. The corn hulls and germ meal were evaluated for starch and hemicellulose compositions. Starch contents were 12 to 32% w/w and hemicellulose (arabinoxylans) contents were 23 to 64% w/w. Corn fibrous samples were hydrolysed, using dilute sulphuric acid, into mixed sugar streams containing arabinose, glucose and xylose. Total sugar concentrations in the hydrolysate varied from 8.4 to 10.8% w/v. The hydrolysates were fermented to ethanol using recombinant E. coli strains K011 and SL40. Ethanol yields were 0.38 to 0.41g ethanol produced/g total sugars consumed and fermentations were completed in 60h or less. However, residual xylose was detected for each hydrolysate fermentation and was especially significant for fermentations using strain SL40. Strain K011 was a superior ethanologenic strain compared with strain SL40 in terms of both ethanol yield and maximum productivity.  相似文献

研究了以玉米秸秆水解糖为碳源,不同氮源条件下琥珀酸放线杆菌Actinobacillus succinogenesSF-9的丁二酸发酵产酸能力。结果表明玉米浆可以替代酵母膏作为丁二酸发酵的廉价氮源。厌氧摇瓶丁二酸发酵单因素试验,得到在初糖浓度50 g/L时,玉米浆的较佳用量为20 g/L。在5 L搅拌罐上,考察了不同初始玉米秸秆水解糖浓度对A.succinogenes SF-9发酵生产丁二酸的影响,结果显示高初始秸秆糖浓度对琥珀酸放线杆菌的生长有抑制作用。采用补料分批发酵,发酵60 h丁二酸的产量达到42.7g/L,丁二酸产率82.7%,生产强度0.81 g/(L·h)。丁二酸的产量和生产强度较分批发酵有明显提高。  相似文献

Gluconobacter oxydans could be immobilized as a biocatalyst for the conversion of glycerol to dihydroxyacetone. To reduce the production cost, the cells were produced from agricultural byproducts. Corn meal hydrolysate and corn steep liquor were employed to replace of sorbitol and yeast extract as medium for G. oxydans cell production. The optimal medium contained 80 g/L reducing sugar, 25 g/L corn steep liquor, and 10 g/L glycerol. The cell mass was about 4.22 g/L and the glycerol dehydrogenase activity was about 5.23 U/mL. For comparison, the cell mass was about 4.0 g/L and the glycerol dehydrogenase activity was about 5.35 U/mL cultured in sorbitol and yeast extract medium. These studies shown the corn meal hydrolysate and corn steep liquor medium was similar in performance to a nutrient-rich medium, but the cost of production was only 15% of that cultured in sorbitol and yeast extract medium. It was an economical process for the production of G. oxydans cells as biocatalyst for the conversion of glycerol to dihydroxyacetone in industry.  相似文献

本文以玉米浆和木薯为原料,用机械搅拌式发酵罐制备细菌纤维素(BC),对发酵过程的纤维素产量、还原糖消耗、溶氧变化和茵浓变化进行了监测,并以葡萄糖一蛋白胨-酵母粉培养基为对照进行了比较。实验得出玉米浆作氮源时不溶BC的产量为9.2g/L,而氮源成本只是对照组的15%;木薯水解液作碳源时的不溶BC产量达到11.7g/L,比对照组(10.8g/L)高8%;而用玉米浆搭配木薯水解液发酵生产BC,产量也达到10.1g/L,验证了这两种天然原料的廉价高效性,用于工业生产细菌纤维素具有良好的前景。  相似文献

研究了产丙三醇假丝酵母(Candida glycerolgenesis)产丙三醇及副产物与氧供给的关系。摇瓶试验发现其它营养条件一定,玉米浆添加量决定酵母量。在0.4%的玉米浆和装液比0.08时产丙醇最高,副产物乙醇、乙酸和乙酸乙酯最小,玉米浆和装液比影响丙三醇和副产物的形成。在5L的反应器中以搅拌转速控制供氧水平,菌体生长阶段比耗氧速率为28mg/(g.h),在发酵阶段比耗氧速率16mg/(g.  相似文献

A strain identified as Corynebacterium fascians was found to accumulate extracellular D-alanine from glycerol. Cultural conditions for the accumulation of D-alanine were investigated and, as a result, a yield of 7 g of D-alanine per liter was obtained after a 96-h incubation in a medium containing 5% glycerol, 4% (NH(4))(2)HPO(4), and 0.3% corn steep liquor. Optical purity of D-alanine was dependent upon the concentration of corn steep liquor. At the optimal condition, almost optically pure D-alanine was formed and readily isolated (5 g/liter) from the fermentation broth. The product was not contaminated with any detectable amount of other amino acids, except for glycine which was present at a concentration of less than 1 percent.  相似文献

产甘油假丝酵母 (Candidaglycerolgenesis)发酵产生的有机酸对丙三醇产品质量和产率均有影响。发现在发酵其它条件恒定 ,装液比和玉米浆浓度增加时 ,发酵液总酸是递增的。在装液比为 0 2和玉米浆浓度为 8g L时 ,丙酮酸和乳酸在细胞生长期可分别积累达 4 1g L和 1 0g L ,比正常发酵时增加 2倍以上 ,丙三醇产率也低 ;然而 ,装液比为 0 0 8和玉米浆浓度为 4g L时 ,丙酮酸和乳酸产生较低 ,丙三醇产率较高 ,但乙酸积累比供氧不足时高 ,可达 2 6g L。发酵过程中有机酸被细胞代谢 ,含量逐渐下降 ,如在初糖浓度为 1 0 0g L时 ,有机酸在细胞生长期积累至高峰后 ,丙三醇和有机酸随之均降低至较低含量 ,并且丙酮酸或乳酸可以转化为乙酸。此外 ,在外加一些添加剂时对其产生有机酸也有影响 ,如添加 1 %油酸和VB1时可以降低乙酸的积累 ,同时增加丙酮酸的含量 ,丙三醇产量也有所增加 ;而丙酮酸结构类似物氟代丙酮酸和亚硫酸盐促进乙酸的产生 ,使酮戊二酸合成减少 ,丙三醇产量约增加 2 0 %。  相似文献

Production of glycerol by Hansenula anomala in molasses-corn steep liquor based media was studied. The accumulation and yield of glycerol was dependent on the medium composition and aeration rate; pH control did not affect the yield. Summary The aim of this research was to develop methods to use low-cost carbon compounds for rhizobial inoculant production. Five raw starch materials; steamed cassava, sticky rice, fresh corn, dry corn and sorghum were tested for sugar production by an amylase-producing fungus. Streamed cassava produced the highest amount of reducing sugar after fermentation. Bradyrhizobium japonicum USDA110, Azorhizobium caulinodans IRBG23, Rhizobium phaseoli TAL1383, Sinorhizobium fredii HH103, and Mesorhizobium ciceri USDA2429 were tested on minimal medium supplemented with reducing sugar obtained from cassava fermentation. All strains, except B. japonicum USDA110, could grow in medium containing cassava sugar derived from 100 g steamed cassava per litre, and the growth rates for these strains were similar to those in medium containing 0.5 (w/v) mannitol. The sugar derived from steamed cassava was further used for production of glycerol using yeast. After 1 day of yeast fermentation, the culture containing glycerol and heat-killed yeast cells, was used to formulate media for culturing bradyrhizobia. A formulation medium, FM4, with a glycerol concentration of 0.6 g/l and yeast cells (OD600 = 0.1) supported growth of B. japonicum USDA110 up to 3.61 × 109 c.f.u./ml in 7 days. These results demonstrate that steamed cassava could be used to provide cheap and effective carbon sources for rhizobial inoculant production.  相似文献

实验以商品化的马铃薯葡萄糖液体培养基为基础培养基,以胞外粗多糖产量为考察指标,运用响应面分析法考察玉米浆浓度、KH2PO4浓度和发酵时间3个因素对胞外多糖发酵产量的影响,以获得黑根霉胞外多糖发酵最优工艺,建立高产、稳定、可控的胞外多糖发酵生产工艺技术方案。经响应面分析,各因素按照对响应值的影响顺序为:玉米浆浓度>发酵时间> KH2PO4浓度,且玉米浆浓度、发酵时间对胞外多糖产量的影响极显著,KH2PO4浓度对胞外多糖产量的影响不显著。胞外多糖发酵最优工艺为:玉米浆3.2mg/mL、KH2PO4 1.5mg/mL和发酵时间132h,在此条件下胞外多糖的最大预测产量为0.824mg/mL。实验重复性好,是一个高产、稳定、可控的胞外多糖发酵生产工艺技术方案,可以指导黑根霉胞外多糖发酵。  相似文献
