Naturally occurring bioaerosol environments may present a challenge to biological detection-identification-monitoring (BIODIM) systems aiming at rapid and reliable warning of bioterrorism incidents. One way to improve the operational performance of BIODIM systems is to increase our understanding of relevant bioaerosol backgrounds. Subway stations are enclosed public environments which may be regarded as potential bioterrorism targets. This study provides novel information concerning the temporal variability of the concentration level, size distribution, and diversity of airborne bacteria in a Norwegian subway station. Three different air samplers were used during a 72-h sampling campaign in February 2011. The results suggested that the airborne bacterial environment was stable between days and seasons, while the intraday variability was found to be substantial, although often following a consistent diurnal pattern. The bacterial levels ranged from not detected to 103 CFU m−3 and generally showed increased levels during the daytime compared to the nighttime levels, as well as during rush hours compared to non-rush hours. The airborne bacterial levels showed rapid temporal variation (up to 270-fold) on some occasions, both consistent and inconsistent with the diurnal profile. Airborne bacterium-containing particles were distributed between different sizes for particles of >1.1 μm, although ∼50% were between 1.1 and 3.3 μm. Anthropogenic activities (mainly passengers) were demonstrated as major sources of airborne bacteria and predominantly contributed 1.1- to 3.3-μm bacterium-containing particles. Our findings contribute to the development of realistic testing and evaluation schemes for BIODIM equipment by providing information that may be used to simulate operational bioaerosol backgrounds during controlled aerosol chamber-based challenge tests with biological threat agents.  相似文献

A microbiological hood was designed to reduce the danger of airborne infection of laboratory workers. The hood uses absolute filters to deliver sterile air in a laminar flow to the work area. An air curtain across the hood opening permits easy access but separates the worker from aerosols produced in the hood, and protects material inside the hood from contamination by room air. Tests with bacterial and viral aerosols showed that the air curtain is at least 99.96% effective in preventing airborne particles from entering the work area.  相似文献

Besides the well known allergens, several other risk factors may exist for health in a laboratory animal unit. The exposure to these factors may be significant in animal units with poor general or local ventilation systems. Moreover, means to prevent the distribution of airborne contaminants may be limited in animal units housing rabbits or other bigger laboratory animals. Airborne contaminants in conventional laboratory rabbit rooms were sought to evaluate the occupational exposure of animal care personnel. Concentrations of airborne dust, bacteria, fungi, ammonia and endotoxins were measured during 2 days in three phases: before working activities began, during them and afterwards. Both stationary and some personal samples were taken. All of the contaminants sought were found in the rabbit room air. When compared to reported levels in farm animal production areas, the concentrations measured were generally low. However, moderate or high levels of airborne bacteria and fungi were found occasionally during work routines. Airborne contaminants should be considered as a potential occupational health risk for persons working with laboratory animals.  相似文献

Viable airborne microbial counts are commonly used in indoor air quality (IAQ) assessment, but studies linking the microbial counts to a specific type of indoor microbial contamination are limited. We hypothesize that the airborne microbial counts can differentiate air-cooling units with and without complaints of urine and body odors. The keratinolytic property of some isolated bacteria prompts to the hypothesis that keratinase is present in the units to break down keratins, structural proteins that form human skin scales, as sources of amino acids and ammonium to produce the odors. Seven bacterial species and four fungal species were identified in the units and room air. Airborne Staphylococcus haemolyticus and Methylobacterium organophilum counts contributed the most to the microbial dissimilarities of units with and without odor complaints. Keratinolytic bacteria and a methylotrophic bacterium were abundant in the units. All the units contained ammonium, and keratinase activity was higher in the units with odor complaints. Extracellular keratinase activity was more effective at 20 °C than at 30 or 4 °C. Keratinolytic bacteria produced high levels of ammonium in the culture with skin cells. Viable airborne microbial counts can help IAQ inspectors to identify potential odor-causing air-cooling units. Keratins may be broken down in the units and associated with the odor complaints.  相似文献

Aerosols of Mycoplasma pneumoniae were studied at several relative humidities at a controlled temperature of 27 C. Production of an experimentally reproducible aerosol required preatomization of the organism in its suspending fluid and was dependent on the type of fluid used in atomization as well as on the procedures used to produce an aerosol. The airborne particles studied were within the range of epidemiological significance, with most being 2 mum or less in diameter. Survival of the airborne mycoplasma in these particles was found to be best at very low and at very high humidities. The most lethal relative humidity levels were at 60 and 80%, at which levels fewer than 1% of the organisms survived over a 4-hr observation period. However, survival of the organism at most relative humidity levels was such that long-term infectivity could be expected from aerosols of M. pneumoniae. Because of the extreme sensitivity of M. pneumoniae at critical humidity levels, control of the airborne transmission of these organisms may be possible in selected spaces.  相似文献

Aims:  To use real-time PCR for the detection of bacterial bioterror agents in a liquid air sample containing potential airborne interferences, including bacteria, without the need for DNA extraction.
Methods and Results:  Bacteria in air were isolated after passive sedimentation onto R2A agar plates and characterized by 16S rRNA sequencing. Real-time PCR was used to identify different bacterial bioterror agents in an artificial air sample consisting of a liquid air sample and a mixture of miscellaneous airborne bacteria showing different colony morphology on R2A agar plates. No time-consuming DNA extraction was performed. Specifically designed fluorescent hybridization probes were used for identification.
Conclusions:  Fourteen different bacterial genera were classified by 16S rRNA gene sequencing of selected bacterial colonies showing growth on R2A agar plates. Real-time PCR amplification of all the bacterial bioterror agents was successfully obtained in the artificial air sample containing commonly found airborne bacteria and other potential airborne PCR interferences.
Significance and Impact of the Study:  Bacterial bioterror agents can be detected within 1 h in a liquid air sample containing a variety of commonly found airborne bacteria using real-time PCR. Airborne viable bacteria at Kjeller (Norway) were classified to the genera level using 16S rRNA gene sequencing.  相似文献

There are a number of possible links between airborne pollen and respiratory disease that remain underappreciated by both the scientific community and health care providers within the Latin American and Caribbean region. A few regional studies have assimilated some preliminary information on this association, and most recently research on interactions between airborne pollen and pediatric asthma on the Caribbean island of Trinidad has shown critical areas that require further investigation. Pollen samples were collected over a 2-year period using a Burkard 7 Day Recording Volumetric Spore Sampler. Corresponding pediatric asthma data were recorded at a nearby medical hospital. Results indicated that the wet season showed higher pollen concentrations (82 pollen per cubic meter/day) than the dry season (53 pollen per cubic meter/day), and several pollen taxa were observed. An association between airborne pollen concentrations and pediatric asthma emergency room visits was suggested incorporating a threshold and lag period.  相似文献

本研究分析肺炎肺实变患儿支气管肺泡灌洗液病原谱构成,旨在为临床经验性抗感染治疗提供指导。收集2019年1月-2020年1月复旦大学附属儿科医院纤维支气管镜室诊治的住院肺炎病例临床资料,其中肺实变的诊断基于影像学证据,依托国家儿童医学中心病原学检测平台,并参考感染传染科医生解读,本研究对肺炎伴肺实变患儿支气管肺泡灌洗液标本的病原学检测结果进行回顾性分析。结果显示,286例肺炎患儿的286份支气管肺泡灌洗液标本被纳入研究,平均年龄5.5(5.8±3.1)岁,其中195例(68.2%)存在肺炎伴肺实变。总病原体检出率为76.6%(219/286),肺实变和无肺实变肺炎患儿病原体检出率分别为77.9%(152/195)和73.6%(67/91),差异无统计学意义;检出的前5种病原体均为肺炎支原体、腺病毒、鼻病毒、呼吸道合胞病毒和副流感病毒3型。5周岁及以上患儿肺炎支原体检出率达最高77.0%(127/165),肺实变病例和无肺实变病例肺炎支原体检出率分别为67.2%(131/195)和61.5%(56/91),差异无统计学意义。本研究结果提示,肺炎支原体、腺病毒、鼻病毒、呼吸道合胞病毒和副流感病毒3型是导致本研究目标人群患病的主要病原体;病原体检出率高低与是否肺实变无关。  相似文献

Airborne pollen and spores, as well as airflow directions, were continuously monitored during a cruise across the East Mediterranean from Tel Aviv, Israel, to Istanbul, Turkey. In spite of the fact that a high-altitude dust cloud moved, at that, time from North Africa, across the East Mediterranean, only a few dust particles were monitored on the boat. The numbers of counted airborne pollen along the cruise path were rather small. This is, in part, because the trip was taken after the main flowering season in the East Mediterranean region. Nevertheless, airborne pollen grains were still found, either as a result of remnant pollen releases by late-flowering plants or because of secondary lift-up of previously settled pollen. The presented pollen counts are average pollen counts /m3 air /6 h. The counts ranged between ∼5 pollen/m3 of air in mid-sea (July 16th–July 17th) or ∼6 pollen/m3 of air on the Israeli coast (July 16–July 17th), and 30 pollen/m3 of air near the coasts of Turkey and of the Greek Islands (July 18th–July 19th) and some 18 taxa of pollen were identified, most of them at the family level. Some 30 taxa of different spores were recorded. The numbers of airborne spores were relatively low in mid-sea (300–750 spores/m3 air), but were high near the coasts of Turkey (1,200–2,400 spores/m3 air) and of Israel (340–1,695 spores/m3 air).  相似文献

A unique agar drum sampler is described which indicates, continuously, the number of viable, bacterial particles per unit volume of air at the time and point of sampling. By selection of the timer and the sampling rate the sampler is suitable for quite a wide range of concentration and time. An impaction line of 484 in. greatly increases the capacity of this device over slit samplers and other instruments designed to give time-concentration data for viable airborne particles. This sampler should prove useful for: (i) monitoring airborne bacteria in hospitals, public places, and food and industrial plants; (ii) decay rate studies of bacterial aerosols; (iii) evaluation of aerial germicides; (iv) determination of effectiveness of air conditioning systems in removing airborne bacteria; and (v) many other studies in aerobiology.  相似文献

目的探讨儿科肺炎支原体(MP)感染特点,辅助临床医师早期诊断,合理用药。方法测定我院一年来2013例儿科呼吸道疾病患儿的肺炎支原体抗体(IgM)。结果2013例呼吸道感染患儿,检出肺炎支原体抗体(IgM)阳性者769例,占38.2%,769例阳性分别表现为肺炎369例(48%),支气管炎238例(31%),咽炎92例(12%),哮喘70例(9%)。其中,肺炎组与各组相比较,具有统计学意义。MP IgM感染的检出率明显高于其他各组(P〈0.01)。结论MP感染是患儿不可忽视的病原体,检测患儿血清MP抗体能够及早诊断,指导治疗。  相似文献

Objective

An Asian dust storm (ADS) contains airborne particles that affect conditions such as asthma, but the mechanism of exacerbation is unclear. The objective of this study was to compare immune adjuvant effects and airway inflammation induced by airborne particles collected on ADS days and the original ADS soil (CJ-1 soil) in asthma model mice.

Methods

Airborne particles were collected on ADS days in western Japan. NC/Nga mice were co-sensitized by intranasal instillation with ADS airborne particles and/or Dermatophagoides farinae (Df), and with CJ-1 soil and/or Df for 5 consecutive days. Df-sensitized mice were stimulated with Df challenge intranasally at 7 days after the last Df sensitization. At 24 hours after challenge, serum allergen specific antibody, differential leukocyte count and inflammatory cytokines in bronchoalveolar lavage fluid (BALF) were measured, and airway inflammation was examined histopathologically.

Results

Co-sensitization with ADS airborne particles and Df increased the neutrophil and eosinophil counts in BALF. Augmentation of airway inflammation was also observed in peribronchiolar and perivascular lung areas. Df-specific serum IgE was significantly elevated by ADS airborne particles, but not by CJ-1 soil. Levels of interleukin (IL)-5, IL-13, IL-6, and macrophage inflammatory protein-2 were higher in BALF in mice treated with ADS airborne particles.

Conclusion

These results suggest that substances attached to ADS airborne particles that are not in the original ADS soil may play important roles in immune adjuvant effects and airway inflammation.  相似文献

Bacterial and viral upper respiratory infections (URI) produce highly variable clinical symptoms that cannot be used to identify the etiologic agent. Proper treatment, however, depends on correct identification of the pathogen involved as antibiotics provide little or no benefit with viral infections. Here we describe a rapid and sensitive genotyping assay and microarray for URI identification using standard amplification and hybridization techniques, with electrochemical detection (ECD) on a semiconductor-based oligonucleotide microarray. The assay was developed to detect four bacterial pathogens (Bordetella pertussis, Streptococcus pyogenes, Chlamydia pneumoniae and Mycoplasma pneumoniae) and 9 viral pathogens (adenovirus 4, coronavirus OC43, 229E and HK, influenza A and B, parainfluenza types 1, 2, and 3 and respiratory syncytial virus. This new platform forms the basis for a fully automated diagnostics system that is very flexible and can be customized to suit different or additional pathogens. Multiple probes on a flexible platform allow one to test probes empirically and then select highly reactive probes for further iterative evaluation. Because ECD uses an enzymatic reaction to create electrical signals that can be read directly from the array, there is no need for image analysis or for expensive and delicate optical scanning equipment. We show assay sensitivity and specificity that are excellent for a multiplexed format.  相似文献

We examined the kinetics of airborne levels of mite allergen particles in a house by combined use of an indoor Burkard air sampler and immunoblotting. Airborne mite allergens collected on the Burkard sampling tape were transferred onto a nitrocellulose membrane, reacted with mouse monoclonal anti-mite allergen (Der pI) antibody, then treated with alkaline phosphatase conjugated anti-mouse IgG. Finally, the blotted allergen on the membrane was reacted with BCIP/NBT phosphatase, and purple spots visible by the naked eye were produced. The shape of the spots was observed under a microscope, and the spot area was measured by an image processor. This technique might be useful for analyzing the behavior of airborne allergen particles in indoor environments.  相似文献

Particles were more efficiently cleared at the higher ventilation rate in both the straw and paper stalls. Ammonia measurements reflected an accumulation over time. In the stalls with low ventilation, ammonia levels were significantly higher than in those stalls with high ventilation regardless of bedding type. The etiological structure of acute pneumonia and acute respiratory diseases was studied with a view to establishing the proportion of L. pneumophila among other causative agents of such diseases. A total of 299 patients were examined over time. The etiological diagnosis based on the data of serological examination was made in 70.6% of the patients with acute pneumonia and in 65% of the patients with acute respiratory viral infections and influenza. In the etiology of pneumonia, the leading role was found to belong to influenza A (H3N2) and B viruses, as well as to adenovirus, while in the etiology of acute respiratory viral infections and influenza, to influenza B virus, adenovirus and Mycoplasma pneumoniae. The importance of L. pneumophila in the etiology of acute pneumonia and acute respiratory diseases was shown. The proportion of L. pneumophila proved to be, on the average, 9.9% in acute pneumonia and 9.8% in acute respiratory diseases. L. pneumophila occurred most frequently in mixed infections in combination with adenovirus and influenza B virus. Diseases of Legionella etiology were found to have a seasonal character, occurring mostly in winter and spring.  相似文献

Air cleanliness in the working area of an animal room equipped with a conventional turbulent flow air distribution systems was compared with that in a similar room fitted with a one-way-flow air distribution system; in this, the supply air flowed from the working area through the racks of cages and was removed from the exhaust side. Before the introduction of animals, the air in the working and exhaust areas of both rooms was ascertained to be Class 100. With animals in situ, however, whereas in the turbulent airflow room both the workspace and exhaust air reached about Class 10,000 (with particle counts, bacterial counts and ammonia levels being almost the same) in the one-way-flow room, the air in the work space only went up to about Class 1000. With the addition of sliding doors or curtains in front of the rack in the one-way-flow room the work space air was maintained at Class 100 with almost no dust particles over 1 microns in size, airborne bacteria or ammonia being detectable. A comparison of all factors measured showed that whereas in the turbulent flow room the contamination of the work space air was 91% of that of the exhaust air, in the one-way-flow room it was only 47%, with curtains added this was reduced to 7% and with sliding doors to only 2%. In the latter case, contamination levels increased markedly on both sides during and immediately after cage changing, but recovered to the pre-cage changing levels within 30 min in the personnel working area and within 60 min on the exhaust side.  相似文献

The effectiveness of commercially available domestic air purifiers to reduce airborne Aspergillus fumigatus conidia and mouldy hay dust was investigated. It was found that the rate of particle clearance is a function of the volume of air passing through the purifiers but that the low throughflow of air makes their use of little value in clearing particles from a normal sized room. Vacuum cleaners were more effective than air purifiers because of their higher air throughput, so too were high volume fan systems in conjunction with simple filtration units. Ionizers had no effect but steam condensation was very efficient at clearing airborne particles.  相似文献
