Food‐storing birds use spatial memory to find previously cached food items. Throughout winter, pinyon jays (Gymnorhinus cyanocephalus) rely heavily on cached pinyon pine (Pinus edulis) seeds. Because of a recent severe drought, pinyon pine trees had not produced a significant seed crop for several years. Therefore, 1‐ and 2‐yr‐old birds never had the opportunity to cache and recover seeds and birds 4 or more years of age had not recovered seeds in 3 yr. This study examined whether natural but extreme variability in experience might result in differences in abstract spatial memory ability during a non‐cache recovery test of spatial memory. Three groups of jays were tested for spatial memory ability in an open room analog of the radial arm maze. Two of the groups were 8 mo old: young/minimally experienced birds which had 2 mo of experience in the wild, while young/experienced birds had 5 mo of experience in their natural habitat. The third group, adult, consisted of birds more than 3 yr old, with at least 3 yr of experience in their natural habitat. This was the only group with experience caching pine seeds. All three groups performed equally and well above chance. This suggests that spatial memory is fully developed by 8 mo of age and is not affected by extensive experience in the wild.  相似文献

Enterobacter cloacae A-11 is a prototrophic, glycolytic mutant of strain 501R3 with a single transposon insertion in pfkA. The populations of strain A-11 on cucumber and radish seeds were smaller than the populations of strain 501R3 in natural soil, but the populations of these two strains on pea, soybean, sunflower, and sweet corn seeds were similar (D. P. Roberts, P. D. Dery, I. Yucel, J. Buyer, M. A. Holtman, and D. Y. Kobayashi, Appl. Environ. Microbiol. 65:2513-2519, 1999). The net effect of the mutation in pfkA in vitro was a shift from rapid growth on certain carbohydrates detected in seed exudates to much slower growth on other carbohydrates, amino acids, and organic acids. The impact of the mutation in pfkA was greatest on the growth rate of E. cloacae on the seeds that released the smallest quantities of fructose, other carbohydrates, and amino acids. Corn, pea, soybean, and sunflower seeds released total amounts of carbohydrates and amino acids at rates that were approximately 10- to 100-fold greater than the rates observed with cucumber and radish seeds for the first 24 h after inhibition began. The growth rate of strain A-11 was significantly less (50% less) than the growth rate of strain 501R3 on radish seeds, and the growth rate of strain A-11 was too low to estimate on cucumber seeds in sterile sand for the first 24 h after inhibition began. The growth rate of strain A-11 was also significantly lower on soybean seeds, but it was only 17% lower than the growth rate of strain 501R3. The growth rates of strains 501R3 and A-11 were similar on pea, sunflower, and corn seeds in sterile sand for the first 30 h after imbibition began. Large reductions in the growth rates of strain A-11 on seeds were correlated with subsequent decreased levels of colonization of seeds compared to the levels of colonization of strain 501R3. The strain A-11 populations were significantly smaller than the strain 501R3 populations only on radish and cucumber seeds. The mutation in pfkA appears to decrease the level of colonization by E. cloacae for seeds that release small quantities of reduced carbon compounds by decreasing the size of the pool of compounds that support rapid growth by this bacterium.  相似文献

Ubiquitin is a highly conserved, 76-amino acid polypeptide with several important regulatory functions in both plants and animals that all arise from its covalent ligation to other cellular proteins. Here, we demonstrate that higher plants have the capacity to conjugate ubiquitin to other plant proteins in vitro. Using 125I-labeled human ubiquitin as a substrate, conjugating activities were observed in crude etiolated tissue extracts from all species tested, including oats, rye, barley, corn, zucchini squash, pea, soybean, and sunflower. The reaction has a soluble distribution, is specific for ATP, and requires the protease inhibitor, leupeptin, to protect ubiquitin from inactivation during the assay. Conjugation is inhibited by N-ethylmaleimide and high concentrations of 2-mercaptoethanol suggesting that the mechanism of ubiquitin ligation in plants involves a similar thiolester intermediate to that found in the mammalian pathway. The conjugating activity in etiolated oat extracts is extremely labile with a half-life of about 20 minutes at 30°C. Detectable but low ATP-stimulated, conjugating activities were also observed in extracts from dry seeds and green leaves of oats. In addition to this conjugating activity, crude plant extracts have the capacity to degrade ubiquitin-protein conjugates formed in vitro. These results demonstrate that higher plants contain several of the enzymic activities necessary for ubiquitin's functions and provide a method for assaying ubiquitin conjugation in vitro.  相似文献

Rose-ringed parakeets (Psittacula krameri) are the world’s most successful introduced parrots, and > 2000 individuals reside on Kauai, Hawaii. These birds destroy crops, but impacts to other native and non-native species are largely unknown. Our study objectives on Kauai were to determine: (1) diets of rose-ringed parakeets at five sites (n = 9–25 per site), by sex, through crop and gizzard analysis and carbon and nitrogen stable isotope analysis, and (2) whether birds are dispersing or depredating seeds. We found 100% of birds (n = 64) were eating plant material and 80% of their diet was seed; males had more food in their crops and gizzards than did females. Corn (Zea mays) was eaten by 67% of birds and averaged 31% of mass in crops and gizzards. Invasive yellow guava (Psidium guajava) was eaten by 97% of birds and averaged 30% of their diet. Parakeets are potentially dispersing yellow guava seeds, as 66% of birds had intact guava seeds, and each bird had an average of three intact seeds. Diets differed statistically among sites. Parakeets from Lihue Airport did not have any corn, and isotopic carbon values also supported low feeding on corn by birds at Lihue Airport. No seeds of native plants were identified in rose-ringed parakeet diets. Our findings of a diverse plant diet, frequent seed predation, and potential to disperse invasive species’ seeds implies that land managers in agricultural, urban, and natural areas should be concerned with the current expansion of these invasive birds on Kauai and elsewhere.  相似文献

The anatomic size of the capillary-to-fiber (C/F) interface plays an important role in O(2) flux from blood to tissue by determining the surface area available for diffusion and is maintained in relative proportion to fiber mitochondrial volume across a wide range of muscle aerobic capacity. In the present study, we examined an estimate of the anatomic size of the C/F interface [the quotient of the individual C/F ratio and fiber perimeter, C/F perimeter exchange (CFPE) index] and fiber oxidative capacity in different skeletal muscles, or muscle regions, to test the hypothesis that capillarization would be maintained in relative excess of reduced fiber oxidative capacity in aged muscles. The right gastrocnemius, plantaris, and soleus muscles from young adult (8 mo old) and late middle-aged (28-30 mo old) Fischer 344 x Brown Norway F1 hybrid rats were excised for evaluation of flux through electron transport chain complexes I-III and/or morphometric estimation of capillarization. Muscle mass was lower in the gastrocnemius muscles of the older animals (2,076 +/- 32 vs. 1,825 +/- 47 mg in young adult vs. late middle-aged, respectively; mean +/- SE) but not the plantaris or soleus muscles. Fibers were smaller in the white region of gastrocnemius muscles but larger in the red region of gastrocnemius muscles of the older animals. There was no difference in the number of capillaries around a fiber, the individual C/F ratio, or the CFPE index between groups for any muscle/region, whereas flux through complexes I-III was reduced by 29-43% in late middle-aged animals. Thus the greater quotient of indexes of anatomic capillarity (individual C/F ratio or CFPE index) and fiber oxidative capacity in soleus and the white region of gastrocnemius muscles, but not in the red region of gastrocnemius muscles of the older animals, shows that anatomic capillarity is maintained in relative excess of oxidative capacity in some muscle regions in late middle-aged rats.  相似文献

Allantoinase catalyzes the hydrolysis of allantoin to allantoic acid, a reaction important in both biogenesis and degradation of ureides. Ureide production in cotyledons of germinating soybean (Glycine max L.) seeds has not been studied extensively but may be important in mobilizing nitrogen reserves. Allantoinase was purified approximately 2500-fold from a crude extract of soybean seeds by differential centrifugation, heat treatment, ammonium sulfate fractionation, ethanol fractionation, and fast protein liquid chromatography (Pharmacia) with Mono-Q and Superose columns. The purified enzyme had a subunit size of 30 kD. Polyclonal antibodies produced against the purified protein titrated allantoinase activity in a crude extract of seed proteins. Antibodies recognized the 30-kD band in western blot analysis of crude seed extracts, indicating that they were specific for allantoinase.  相似文献

The cerambycid Dectes texanus texanus LeConte that infests soybean, Glycine max (L.) Merr., is thought to be the same Dectes that damages sunflower, Helianthus annuus L. However, morphometric characteristics and mating compatibility between individuals from soybean and sunflower fields have not been assessed. Eight hundred eighty stalks of soybean and sunflower residues infested with Dectes were collected from Texas and Kansas. Outside stalk diameter of sunflower (22.6 mm) was 3.2 times greater than that of soybean. Diameters of tunnels by Dectes in sunflower and soybean were 7.0 and 3.4 mm, respectively. Widths of head capsules and pronotums were approximately 1.3 times greater for Dectes larvae from sunflower than soybean. Larval, pupal, and adult Dectes from sunflower were approximately 2.5 times heavier and 1.3 times longer than from soybean. Adults reared from residues of soybean and sunflower were paired to evaluate mating compatibility. Although individuals collected and reared from different crop residues differed significantly in size, percentage of mating success by Dectes from the two crops (26.2%) did not differ significantly from that by adults from the same kind of crop (34.9%). Results indicated that Dectes that infest soybean and sunflower are the same species and reconfirmed that farmers should not plant soybean and sunflower in rotation or in adjacent fields.  相似文献

Freshly emerged adults of Zygogramma bicolorata Pallister (Coleoptera: Chrysomelidae), introduced for biological control trials against the noxious weed Parthenium hysterophorus , were seen to feed and lay eggs on sunflower leaves under confined conditions in the laboratory. The time taken for initiation of feeding was found to be positively correlated with adult age, while survival after feeding on the crop was negatively correlated. However, adults that had fed on sunflower even for 1 day on P. hysterophorus were incapable of ovipositing, and young larvae could not feed on the crop. Under field conditions, newly emerged adults were only attracted to feed on plants that were treated either with a crude aqueous extract of leaves or pollen grains of P. hysterophorus , indicating that the chances of Z. bicolorata becoming a pest of sunflower are negligible. Analysis of Leucaena leucocephala from various localities in Western Nigeria shows that young leaves, pods and seeds contain more crude protein but lower crude fibre and ether extract than mature ones. The crude fat component was higher in the seeds (4.6% and 5.5% for green and ripe seeds, respectively) than in any other edible part of the plant, while the green and brown seed coats contain the least amount of ether extract and total ash. The mineral composition of the dry matter of mature leaves is 2.8% calcium, 0.26% phosphorus, 0.37% magnesium, 1.78% potassium, 0.21% sodium and 0.12% iron.  相似文献

Prehension is essential for animal survival and fitness. It is involved in locomotion and feeding behavior and subject to physical and physiological constraints. Studies of prehension in primates have explored the importance of food properties and of the environment, but aging has rarely been studied although prehensile capacity may deteriorate with age in humans. To test the hypothesis that aging affects grasping abilities and to reveal possible behavioral adaptations to this, we quantified behavioral grasping strategies and pull strength in 10 young adult (2–3 yr old) and 10 aged (7–8 yr old) gray mouse lemurs (Microcebus murinus). We assessed grasping strategies in an experimental cage by quantifying grip types used to grasp static and mobile foods. We measured strength using a Kistler triaxial force platform. Our results show that 1) mobile and static foods affected individuals of different ages in similar ways; 2) older individuals used more mouth grasps than young ones; 3) aged individuals made twice as many attempts as young ones when grasping mobile food items but this difference was not significant; and 4) there were no differences in hand grip strength between age classes but young individuals showed a higher foot pull strength compared to old ones. These data suggest that the observed differences in behavior may be due to a decrease in foot grip strength, which in turn influences stability on narrow branches, forcing animals to use their hands to maintain stability and preventing them from using their hands for food-related tasks.  相似文献

Cucurbitacins are feeding stimulants for corn rootworm used in baits to control the adults of this insect pest. Corn rootworm larvae also feed compulsively on cucurbitacins. Cucurbitacins are reported to be gibberellin antagonists that may preclude their use as seed treatments for these soil-dwelling insects. The crude extract of a bitter Hawkesbury watermelon containing cucurbitacin E-glycoside significantly inhibited germination of watermelon, squash, and tomato seeds. Although the germination of corn seed was not significantly inhibited, root elongation was inhibited by crude extracts, but not by high-performance liquid chromatography-purified cucurbitacin E-glycoside. Therefore, the effects of the major components in the bitter watermelon extract (e.g., sugars) on seed germination and root elongation were determined. Pure sugars (glucose and fructose), at concentrations found in watermelon extract, mimicked the inhibition of seed germination and root elongation seen with the crude bitter Hawkesbury watermelon extract. Removal of these sugars may be necessary to use this extract as a bait for corn rootworm larvae as a seed or root treatment.  相似文献

The current study sought to examine the effects of chronic endurance treadmill running on oxidative capacity and capillary density in specific diaphragm muscle fiber types in young (5 mo) and senescent (greater than or equal to 23 mo) female Fischer 344 rats. Both young and senescent animals trained at approximately 75% of maximal O2 consumption for 1 h/day 5 days/wk for 10 wk. Plantaris citrate synthase activity was significantly increased (P less than 0.01) in both young and old trained groups. Densitometric analysis of succinate dehydrogenase (SDH) activity in diaphragm type I, IIa, and IIb muscle fibers was done using a computerized image-processing system. There were no age-related differences in SDH activity between the young and old groups for any of the fiber types. In addition, SDH activity was found to be significantly increased (P less than 0.05) in all three fiber types in both the young and senescent trained animals compared with their sedentary counterparts. Fiber size and capillary density did not differ between young and senescent rats, nor did exercise affect this measure. Each fiber, irrespective of type, had an average of approximately four capillaries in contact with it. However, type IIb fibers had a significantly lower capillary density per unit area than type I or IIa muscle fibers. The results indicate that the senescent costal diaphragm maintains its ability to adapt to an increased metabolic demand brought about by locomotor exercise. Of further interest is the finding that training adaptations occurred in all three fiber types, suggesting that increased work of breathing from moderate exercise leads to recruitment of all three fiber types.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The purpose of this study was to investigate the effects of mild therapeutic exercise (treadmill) in preventing the inactivity-induced alterations in contractile properties (e.g., power, force, and velocity) of type I soleus single fibers in three different age groups. Young adult (5- to 12-mo-old), middle-aged (24- to 31-mo-old), and old (32- to 40-mo-old) F344BNF1 rats were randomly assigned to three experimental groups: weight-bearing control (CON), non-weight bearing (NWB), and NWB with exercise (NWBX). NWB rats were hindlimb suspended for 2 wk, representing inactivity. The NWBX rats were hindlimb suspended for 2 wk and received therapeutic exercise on a treadmill four times a day for 15 min each. Peak power and isometric maximal force were reduced following hindlimb suspension (HS) in all three age groups. HS decreased fiber diameter in young adult and old rats (-21 and -12%, respectively). Specific tension (isometric maximal force/cross-sectional area) was significantly reduced in both the middle-aged (-36%) and old (-23%) rats. The effects of the mild therapeutic exercise program on fiber diameter and contractile properties were age specific. Mild treadmill therapeutic exercise attenuated the HS-induced reduction in fiber diameter (+17%, 93% level of CON group) and peak power (μN·fiber length·s(-1)) (+46%, 63% level of CON group) in young adult rats. In the middle-aged animals, this exercise protocol improved peak power (+60%, 100% level of CON group) and normalized power (kN·m(-2)·fiber length·s(-1)) (+45%, 108% level of CON group). Interestingly, treadmill exercise resulted in a further reduction in shortening velocity (-42%, 67% level of CON group) and specific tension (-29%, 55% level of CON group) in the old animals. These results suggest that mild treadmill exercise is beneficial in attenuating and preventing inactivity-induced decline in peak power of type I soleus single fibers in young adult and middle-aged animals, respectively. However, this exercise program does not prevent the HS-induced decline in muscle function in the old animals.  相似文献

油菜是我国重要的绿肥作物,除增加土壤肥力外,对后茬作物也有一定的化感作用。本文研究了不同浓度油菜浸提液(0.025、0.05和0.1 g·mL-1)对3种作物(燕麦、玉米和向日葵)种子萌发、幼苗生长及生理生化的影响。结果表明: 油菜浸提液对3种作物种子发芽率的影响不显著,但对燕麦有“低促高抑”的趋势,对向日葵有抑制的趋势。油菜浸提液高浓度处理对燕麦幼苗根和茎长、玉米幼苗茎长以及向日葵幼苗根长起抑制作用,低浓度处理对玉米幼苗根茎长起促进作用。燕麦和向日葵幼苗在浸提液浓度为0.05 g·mL-1时蛋白质含量最大,玉米在0.025 g·mL-1时蛋白质含量最大,显著高于对照。3个浓度下,向日葵脯氨酸含量均显著降低,玉米脯氨酸含量在浓度为0.05 g·mL-1时显著高于对照。玉米过氧化物酶(POD)活性在浸提液浓度为0.05 g·mL-1时最大,向日葵POD活性在浓度为0.025 g·mL-1时最大,与对照有显著差异,其余浓度处理与对照无显著差异。油菜浸提液浓度为0.025 g·mL-1时显著降低了燕麦超氧化物歧化酶(SOD)活性,3种作物幼苗过氧化氢酶(CAT)活性与对照均无显著差异。加入油菜浸提液后,向日葵幼苗丙二醛(MDA)含量在浓度为0.1 g·mL-1时显著增加,玉米幼苗MDA含量在不同浓度处理下均显著低于对照。综合分析,油菜浸提液对向日葵的化感抑制作用最强,其次为玉米、燕麦。  相似文献

The development of immunologic methods to reduce transmission of human lymphatic filariasis depends on measures that will enhance the host's ability to eliminate infective larvae, adult worms, or blood-borne microfilariae (mf). The present study was designed to assess the capacity of a crude extract of Brugia malayi mf to decrease the level of microfilaremia and adult worm burden in jirds inoculated with infective larvae, and to identify the filarial antigens that elicit antibody responses in these animals. Thirty weeks after subcutaneous inoculation with 75 infective larvae, 100% of control jirds were patent (i.e., had microfilaremia) compared with 60% of the group immunized with 10 micrograms of crude microfilarial extract (p less than 0.05). In addition, microfilaremia was lower in patent immunized animals compared with controls (p less than 0.05). The mean total number of adult female B. malayi per jird recovered at necropsy in control animals was 16.0 vs 7.0 in immunized jirds (p less than 0.05). Serum of immunized jirds contained anti-mf antibodies with an end titer of 1:8000, a value similar to that of animals with chronic B. malayi infection. Microfilarial antigens of Mr approximately 150,000, 75,000, 42,000, and 25,000 were identified in immunoblotting studies by reactivity with antibodies in sera of immunized jirds. Antibodies induced by immunization with microfilarial extract were not specific for this stage of the parasite life cycle, as jird anti-mf antibodies reacted with a Mr approximately 150,000 and several Mr 50,000 to 110,000 antigens derived from immature and mature adult parasites of both sexes. These data indicate that immunization of jirds with a water soluble microfilarial extract enhances the host's ability to eliminate adult worms and blood-borne mf. The filarial antigens that induce antibodies in immunized jirds have been identified.  相似文献

Seeds of oat, coconut, soybean, sunflower, rice, millet, kidney bean, buckwheat, wheat, and corn and vegetative tissue of oat, pea, and corn were assayed for free indole-3-acetic acid (IAA), esterified IAA, and peptidyl IAA. Three conclusions were drawn: (a) all plant tissues examined contained most of their IAA as derivatives, either esterified or as a peptide; (b) the cereal grains examined contained mainly ester IAA; (c) the legume seeds examined contained mainly peptidyl IAA. We evaluated thein vitro effect of norepinephrine (NE), over the range of concentrations between 10-12 M and 10-3 M, on adherence (to plastic surfaces) and chemotaxis (in a Boyden chamber) of peritoneal macrophages from BALB/c mice of different ages: young (12 weeks), adult (22 weeks), mature (48 weeks) and old (72 weeks). Increased adherence was induced by 10-12 M of NE in macrophages from young, adult, mature and old mice. Also, 10-9 M stimulated adherence in old animals, 10-5 M in mature mice, and 10-3 M in both young and old mices. With respect to chemotaxis, the low concentration of NE (10-12 M) was stimulatory only in young and adult animals, higher concentrations (10-5 M and 10-7 M) were inhibitory for macrophages from mature and old animals, and the highest concentration of NE (10-3M) stimulated this capacity of macrophages only in young and mature animals. The conclusion is that while the mobility of macrophages to the focus of infection (i.e. chemotaxis) is stimulated by low concentrations of NE (10-12, M) only in young-adult animals, this neurotransmitter induces a decline in this capacity in mature and old mice at high concentrations (10-5 M - 10-7 M). Also, macrophages from old animals have lost the capacity to respond to pharmacological (10-3 M) concentrations of NE. The lower capacity of response to NE by macrophages from old animals possibly contributes to immunosenescence.  相似文献

Mating by young males or low male‐to‐female ratios can decrease pregnancy rates and postpone birthdates in ungulates, thereby hindering population growth. Young (2.5–3.5 yr old) male bighorn (Ovis canadensis) behave differently than older males, and age, horn size, mating behavior, and social rank help determine reproductive success. We estimated birthdates in two populations of bighorn sheep in Utah, USA, to determine if mating by young males or low male‐to‐female ratios resulted in fewer young born per female, a shift in mean timing of births, or asynchronous births. When reintroduced, the Rock Canyon population consisted of four males (two each of 2.5 yr old and 1.5 yr old) and a 1 to 7.5 ratio of males (>2 yr old) to adult females (≥3.5 yr old); the Mount Nebo population consisted of four males ≤1.5 yr old and a 0 to 12 ratio of males to adult females. For both populations, the number of young born per female did not differ between the first parturition period after reintroduction (where females were impregnated by males from their source populations) and the second period of parturition (where females were impregnated by young, reintroduced males). Mean birthdates and synchrony (SD) of births did not differ for Rock Canyon (May 12, 2001 ± 4.5 d, May 14, 2002 ± 3.2 d) or Mount Nebo (May 23, 2005 ± 8.1 d, May 22, 2006 ± 10.2 d) between the first and second years following reintroduction. Mating by young males or low male‐to‐female ratios had no demonstrable effect on the number of young born per female or timing and synchrony of births in these populations.  相似文献
