The maturing egg of Histiopteris incisa has many features incommon with that of Pteridium, with which the species may berelated. The nuclear evaginations are however conspicuous inrespect of the extensive development of internal membrane systems,involving dilatation of the perinuclear space. In some in stancesthe inner membrane of the nuclear envelope, the proliferationof which gives rise to the dilatations, was strikingly tenuous,possibly indicating an extensive interchange between the perinuclearspace and that part of the nucleoplasm sequestered in the evaginations.The evaginations frequently contain an aggregate of electron-opaquematerial resembling structurally and in staining propertiessimilar aggregates in Pteridium, but in contrast to Pteridiumthere was no evidence that these aggregates were organized inor near the nucleolus. At an early stage of growth gametophytes support the sporophytes of ferns. Young sporophytes become independent of gametophytes when the first leaves develop. Although large fern gametophytes produce multiple archegonia simultaneously, only one sporophyte is typically established on one gametophyte. The number of sporophytes is believed to be controlled in two possible directions, from gametophyte to sporophyte or from preceding sporophyte to another sporophyte. To investigate the effects of gametophytes on their sporophytes, we studied the relationship between organic matter production by gametophytes and the growth of young sporophytes of Thelypteris palustris. We cut gametophytes in half (CGs) to reduce the gametophytes’ production of matter. There was no significant difference between the growth of sporophytes on intact gametophytes (IGs) and that on CGs. According to our estimates, based on the rate of organic matter production, the large gametophyte was able to produce two or more sporophytes. The resources required for CGs to make similar-sized sporophytes was twice that for IGs. In polyembryony each of the multiple sporophytes was similar in size to the single sporophytes. Resource limitation does not seem to explain why fern gametophytes establish single sporophytes.  相似文献

 稻田施用的除草剂可能会污染邻近水域,对非靶标水生植物产生影响。国家二级重点保护野生植物粗梗水蕨(Ceratopteris pteridoides)主要分布于稻田及其周围水域,其有性繁殖敏感阶段与除草剂施用时间一致,配子体易暴露在除草剂污染中。为了评价除草剂污染对水生态环境特别是对非靶标国家重点保护野生水生植物的影响,保护生物多样性和水生生态环境,该文研究了除草剂苄嘧磺隆(Bensulfuron-methyl)对粗梗水蕨配子体生长和性器官分化的影响。结果表明,苄嘧磺隆对粗梗水蕨孢子萌发没有影响,但对其配子体生长有抑制作用。苄嘧磺隆浓度和粗梗水蕨配子体面积呈明显的剂量效应关系,苄嘧磺隆对粗梗水蕨配子体生长抑制的EC50(半数效应浓度)为0.086μg&;#8226;L-1,低于已报道的环境背景浓度。随着苄嘧磺隆浓度升高,粗梗水蕨的雌雄同体配子体逐渐减少,且配子体上颈卵器形成时间推迟。在苄嘧磺隆为10μg&;#8226;L-1时,粗梗水蕨没有雌雄同体配子体形成。以上研究结果表明,低浓度苄嘧磺隆可抑制粗梗水蕨配子体的生长并影响其性器官的分化,使粗梗水蕨有性繁殖能力减弱。  相似文献

Examination of two species of osmundaceous ferns, two speciesof marattialean, and one dipterid, all representative of fernswith a long fossil history, has shown that the egg cells possessnumerous large amyloplasts. The nuclei of these egg cells alsoproduce no vesicular or sheet-like protrusions during maturation.In these respects the egg cells of the ferns examined differsharply from those of more recent ferns such as Pteridium andDryopteris. The meiotic process of sporogenesis in Pteridium is accompaniedby a regular cycle of ultrastructural events. These includethe dedifferentiation and redifferentiation of organelles, andthe elimination and restoration of ribosomes. Although similarin nature to corresponding stages in other land plants, sporogenesisin Pteridium provides several unique features. During prophase the plastids pass through a stage during whichno envelope can be resolved at the periphery of the stroma.They appear to survive this period and there is no evidencethat plastid lineage is broken from primary archesporium tospore. The plastid cycle is accompanied by the presence of theiron-protein complex phytoferritin. The repopulation of the spore cytoplasm with ribosomes, followingthe prophase elimination, is achieved through the agency ofnucleoloid-like bodies. These bodies, however, are not synthesizedwithin the nucleus, but form within membrane-bound regions ofthe early prophase cytoplasm. Prophase is also characterized by the development of nuclearvacuoles, expansions of the perinuclear space, which progressivelyramify through the karyoplasm prior to the dissolution of thenuclear envelope at metaphase I. Early gametophyte ontogeny was quantitatively distinct for Olympic Peninsula, Alaskan, and disjunct Idaho populations of the homosporous fern Blechnum spicant (L.) J. Sm. Although variable, gametophyte sex expression was shown to have a genetic component. Statistically different patterns of sex expression characterize each population. The Olympic Peninsula populations were distinct from each other but consistent in having a predominantly unisexual pattern. The disjunct Idaho population was predominantly bisexual at the time when comparable field collected gametophytes bear sporophytes. Preliminary experiments suggest that an antheridogen operates in this species. Increased sowing density favors maleness, and an extract from soil cultures of gametophytes shifts cultures to an exclusively male pattern after a dramatic suppression of growth. Mating experiments revealed that all populations are interfertile, although fertility was highest when the test Idaho population underwent intergametophytic-selfing. The Idaho population evidenced a low level of genetic load consistent with predictions based on its sex expression. Although Olympic Peninsula populations evidenced apparent high genetic load in some experiments, failure to produce abundant sporophytes in other experiments suggested that additional cultural factors operated to reduce sporophyte formation. Moderate density mating experiments produced single sporophytes that were comparable to field collections. Isolated gametophytes underwent polyembryony after a time delay and gametophyte proliferation. Cultural conditions which allow sporophyte formation on isolated gametophytes without this delay or proliferation must be sought before further genetic analysis is undertaken.  相似文献

The present report is a part of our study on the reproductivebiology of a traditional Indian medicinal plant,Commiphora wightii,a source of guggul steroids. Field examination showed a predominantlylarge number of isolated and groups of female individuals. Onlyone andromonoecious and two exclusively male plants were recorded.Female plants set seed irrespective of the presence or absenceof pollen. Hand-pollination experiments and embryological studieshave confirmed the occurrence of non-pseudogamous apomixis,nucellar polyembryony and autonomous endosperm formation forthe first time in this plant, which is presently threatenedby over-exploitation. Under greenhouse conditions, Epidendrum nocturnum Jacq. plantsproduce fruits by both self-fertilization and cleistogamy. Biochemical and fine-structural studies were undertaken on theembryo and female gametophyte of seeds of Podocarpus henkeliiduring the 24-week period of post-fertilization growth and development. Although the d. wt of the embryo and female gametophyte showeda steady increase during development, the levels of sugars,amino acids, proteins and lipids differed between seed partsand showed changes during the four sampling intervals. Onlystarch was seen to increase steadily throughout development.Lipid levels were high in the mature embryo and free amino acidsshowed a steady increase until the seeds were shed. At thisstage seeds were fully hydrated, possessed abundant reservesand all organelles appeared fully functional. This was interpretedas part of the development strategy of neotonous (recalcitrant)seeds, namely, the maintenance of full metabolic competencefor continued growth in the absence of development arrest whichfollows drying in orthodox seeds. The isolation and growth in sterile culture through sixteenpassages, each of 4 to 6 weeks' duration, of an abnormal growthisolated from a gametophyte of Pteridium aquilinum var. latiusculumis reported, and the nutritional requirements described. The cultures consist of green, photosynthetic, predominantlyfilamentous, but coherent, masses. Prothalli were regeneratedon five occasions during the first two passages only. Outgrowthsof whitish or pale green parenchymatoua tissue occur. Thesepossess a central strand of elongated cells, some of which becomedifferentiated into a core of tracheides. Prothalli of the strain in culture are haploid. When examinedsoon after the initial explantation, the filamentous parts ofthe cultures were diploid, but there has been a subsequent increasein the number of chromosomes, and each cell is now irregularlyaneuploid, with a chromosome number between triploid and tetraploid.  相似文献

Abstract. Atmospheric greenhouse gas concentrations will continue to rise for the foreseeable future, and, due to radiative forcing, surface temperatures will also increase. If soils warm sufficiently, this will result in increased litter decomposition rates, while nitrogen availability will also rise due to increased deposition. A field-based experiment manipulating air temperatures using polythene tents and nitrogen availability by adding 50 kg N ha-1 yr-1 has shown that this will result in further encroachment of Pteridium into Calluna moorland. Although this experiment, treating plots on and around the boundary between Pteridium and Calluna, did not show any statistically significant effect on the movement of the boundary, there were clear indications of Calluna dieback at the boundary. In addition, the frequency of Pteridium fronds increased in the Calluna-dominated areas, while Calluna became less abundant in the pure Pteridium plots. Calluna dieback was not due to litter accumulation as is frequently assumed, but the dieback is likely to be caused by shading of Calluna by the increased growth of Pteridium fronds. Botanical diversity was higher in the Pteridium plots, but considering the importance of grouse shooting for the maintenance of Calluna moorland, displacement of Calluna by Pteridium may result in a lower conservation and economic value of moorlands.  相似文献

Intra- and interspecific experimental pollinations were madeto determine the mating systems and the interspecific crossingpotential in individuals from 24 populations of five Pleurothallisspecies. Pleurothallis johannensis, P. ochreata and P. teresshow weak or partial self-incompatibility while P. adamantinensisand P.fabiobarrosii are more strictly self-incompatible. Wefound no differences in fruit set between intra- and interpopulationintraspecific crosses, and there was no correlation betweenfruit set and genetic variability or inbreeding in these species.All species are interfertile and showed no correlation betweengenetic similarity and crossing potential. We scored fruitsfor seed viability and observed a strong inbreeding depressionin all populations; however, there was no difference in seedviability among intrapopulation, interpopulation intraspecificand interspecific cross-pollinations. These species are pollinatedby flies with a behavioural pattern that facilitates self-pollination.Self-incompatibility and inbreeding depression are apparentlyimportant in the maintenance of the unusually high levels ofgenetic variability found in these species. As generally observedin other orchid species, barriers to hybridization between thesePleurothallis species are at the level of pollination. The female gametophyte of Ephedra foliata was used as an explantfor the production of haploids as it is composed of haploidcells, all of the same genotype. The regeneration of roots wasdependent upon the presence of NAA, while BAP had a modifyingeffect. At lower concentrations (0.05 parts 10–6 and 3.5parts 10–6) BAP enhanced the root promotion of NAA (0.05–4.0parts 10–6). At higher concentrations of BAP (1–6parts 10–6), roots and shoot buds were formed. Kinetinat 4.0 parts 10–6 with 0.5 parts 10–6 2, 4-D wasoptimal for shoot bud production in explants at the archegonialstage and 2, 4-D at 2.0 parts 10–6 with 0.5 parts 10–6kinetin was optimal for root formation. Cells of the callusand root tip had the haploid number of chromosomes, n = 7. Meristemoidswere located on the surface or embedded in the callus tissue.The deep seated meristemoids organized only root primordia,but the peripheral ones gave rise to root as well as shoot budprimordia. Analyses have been made of the K, Ca and Mg in separated sporophytes,spores and gametophytes of the moss Funaria hygrometrica duringsporophyte development. Degeneration of the gametophyte wasaccompanied by loss of K and a gain in Ca while the K contentof the developing sporophyte increased more rapidly than thatof Ca. The presence of an air–gap in the expanding capsulesignificantly influenced the observed cellular location of ions.Relative to the sporophyte the spores were shown to have a higherK and a lower Ca concentration. Spores of Dryopteris marginalis were germinated on a definednutrient medium at varying pH levels (3.2–8.2). Sporegermination in all treatments ranged from 65.5 to 75.0 per centand the effect of pH on germination is concluded to be minimal.Normal gametophyte development, as determined by the numberof cells produced and the pattern of development, is stronglyaffected by the pH of the medium and developmental abnormalitieswere observed below pH 5.8. High levels of plantlet regeneration can be achieved from shoot-tipcultures of the Andean potato Solanum goniocalyx thawed fromliquid nitrogen. A rapid rate of cooling and the presence ofdimethyl sulphoxide as a cryoprotectant appear to be necessary.Microscopical examination of surviving shoot-tip cultures showsthem to contain considerable numbers of damaged cells afterthawing. Structural aspects of this damage have been detailedusing transmission electron microscopy. Taxus brevifolia(Nutt.), commonly known as Pacific or westernyew, is a conifer native to the Pacific northwest of North America.Contrary to other Taxus species, T. brevifolia staminate strobiliare usually located on 2-year-old foliage although they mayoccur on foliage from 1 to 5-years-old. This delayed staminatestrobilus development may be an adaptation to the low lightenvironment where T. brevifolia grows. Microsporogenesis occurredin the autumn preceding pollination. Successive divisions producedisobilateral tetrads visible as early as mid-October. Over-winteringstaminate strobili usually contained separate microspores. In1996 to 1999, pollination occurred in March and April in twonatural forest sites on southern Vancouver Island, British Columbia,Canada. The low amounts of airborne pollen and prolonged pollinationperiod indicated low pollination success within T. brevifolia.Female receptivity was measured by the presence of a pollinationdrop. Protandry up to 18 d was observed. In vitro pollen germinationwas moderate to good, ranging from 65 to 88% depending on thetree and year. DAPI fluorescence staining showed successfulmale gametophyte development in vitro. The microspore dividedforming a tube nucleus and generative cell within 3 d of culture.The generative cell then divided forming a sterile nucleus andspermatogenous nucleus after 17 d. The spermatogenous nucleusacquired a cell wall then divided forming two equal sperm after24 d. Biochemical and ultrastructural studies were undertaken on theembryo and female gametophyte of neotonous (recalcitrant) seedsof Podocarpus henkelii over a 9-d period following scarificationand incubation on a moist substrate. After 3 days incubationat 25 °C, cells of the root tip were characterized at theelectron microscope level by increased vacuolation, numerousamyloplasts and lipid mobilization. By d 6 measurable embryonicgrowth was noted and ultrastructural evidence of synthetic activitywas suggested by abundant endoplasmic reticulum (ER), ribosomesand dictyosomes. Fine-structural changes suggestive of reservemobilization were also observed in the female gametophyte. Biochemicalstudies indicated a gradual decline in lipid and protein inboth the embryo, and female gametophyte, over the 9 d of incubation.A decline in embryonic starch levels contrasted with the increaseseen in the female gametophyte at d 6. Small changes in thesugar levels of the female gametophyte contrasted with increasesin the embryonic tissues between d 3 and 6, a period coincidingwith the first records of germination. The maintenance of highmoisture contant and the evidence of metabolic activity obtainedfrom biochemical and ultrastructural observations suggest that,following scarification, the transition between maturation andgermination is characterized by a continuation of earlier syntheticactivity and reserve interconversions. Abstract. Vegetation development after conventional bracken control with herbicide in the uplands is often slow and rarely results in the development of the target community. Two experiments were established on contrasting sites to investigate cost‐effective methods for the re‐establishment of vegetation dominated by Calluna vulgaris (heather), which is the usual target community seen as the desired replacement for Pteridium aquilinum in the uplands by many land managers. At one site, where grazing intensity was low and shelter was sufficient to prevent rapid Pteridium litter loss, the necessary treatments to establish Calluna were to disturb the litter and add a source of Calluna seeds. A difference between continued Pteridium control or a relaxation of control was not observed over the time‐scale of the experiment. At the second site, where grazing intensity was high and shelter minimal, vegetation dominated by Rumex acetosella had developed on a thin but dense litter layer. To enhance Calluna regeneration it was necessary to prevent sheep grazing, disturb the remaining litter layer and add a source of Calluna propagules. A nurse crop hindered Calluna establishment due to its vigorous growth. At neither site was an initial application of fertilizer an aid to Calluna establishment. It is possible to devise methods for the re‐establishment of Calluna at sites where previous attempts at Pteridium control alone have not had the desired effect on vegetation development. However, the precise choice of methods used is dependent on the grazing management at the site. High levels of livestock grazing prevented Calluna establishment, but some trampling may aid litter breakdown. The ability to adjust stocking rates and access to the site by machinery are critical factors to take into account in producing an effective strategy for long‐term Pteridium control and its replacement by target vegetation.  相似文献

Plastid-dividing rings (PDs) are described for the first timein the ferns Ophioglossum, Gleichenia, Hymenophyllum, Trichomanes,Athyrium, Dryopteris, Ceratopteris and Pteridium. They are foundin the constricted isthmuses of undifferentiated plastids, amyloplasts,amylochloroplasts and chloroplasts in gametophytes and in arange of sporophytic tissues including apices, differentiatingleaf mesophyll and vascular parenchyma from roots, stems andleaves. Fern PDs comprise two concentric structures; a densegranular ring 60-100 nm wide adhering to the stromal face ofthe constriction and a smaller cytoplasmic annulus only 20-40nm wide. The diameters of the PD-containing constrictions, particularlythose in vascular parenchyma, where the outer component is usuallylacking, are extremely uniform (means of 177-188 nm in differentspecies and tissues) and closely in line with those in angiosperms.PDs in ferns differ from those previously described in othergroups of lands plants in that they are far more persistentand in multilobed plastids one is present at each constriction.The occurrence of multiple PDs in the giant plastids found inup to 30% of vascular parenchyma cells may be associated withorganelle fusion rather than division.Copyright 1993, 1999 AcademicPress Chloroplast division, division cycle, ferns, plastid-dividing rings, pteridophytes, transmission electron microscopy  相似文献
