With the periodic acid-thiosemicarbazide-silver proteinate procedure for the detection of polysaccharides in thin sections, glycogen is localized in the cavities of centrioles and basal bodies, within the axoneme (and surrounding it), in mitochondria, and in the "packing" cytoplasm of the middle piece of spermatozoa of several invertebrate and vertebrate species. The cytochemical localization of glycogen is verified by extraction with α-amylase solution. These findings establish the existence of stored glycogen in sperm. The polysaccharide presumably serves as an endogenous source of energy in the absence of extracellular metabolites, under either aerobic or anaerobic conditions. Other hypotheses on the physiological significance of intracellular glycogen stores in sperm are discussed. Sperm that store glycogen contain some enzymes of glycogen metabolism. In the presence of glucose-1-phosphate, ATP, and Mg++ ions, an amylophosphorylase catalyzes the in vivo synthesis of glycogen. The newly formed product resembles γ-particles, and is digestible with α-amylase.  相似文献

The ultrastructure of the sperm of Epiphragmophora tucumanensis(Doering), a pulmonate land snail belonging to the family Xanthonychidae,was examined. The results showed that this spermatozoon presents a similarmorphology to the other advanced stylommatophoran sperm described.The most peculiar characteristic is the form of the nucleus,which is straight instead of helically coiled. An acrosome ispresent, composed of an apical vesicle and an acrosomal pedestal.The connecting piece or neck region is formed by a basal bodyplus a centriolar derivative. It is also the point of originof coarse fibres that accompany the axoneme. Thus, the axonemalcomplex exhibits a 9+9+2 pattern. The midpiece is an elongatedregion composed of paracrystalline and matrix materials (collectively,the mitochondrial derivative) enclosing a single glycogen helixand the axoneme. The mitochondrial derivative extends to theposterior tip of the spermatozoon. As observed in other stylommatophorans,both an annulus and a glycogen piece are absent. The fine structure of the modified sperm and spermatogenesisof four sympatric species of Siphonaria is described. The morphologyof the sperm of all species is very similar. The head, whichis about 6 µm long, is composed of a nucleus with fibrouschromatin capped by an acrosome (about 1 µm long) comprisedof an acrosomal pedestal and apical vesicle. The midpiece hasa mitochondrial derivative which surrounds a single glycogenhelix, posterior to which is a glycogen piece. Although differencesbetween each species exist, the value of sperm morphology forpurposes of taxonomy in this genus is questioned. Comparisonwith other basommatophorans however suggests that sperm morphologymay be of value at a higher taxo-nomic level. The morphologicalchanges that occur during spermatogenesis are similar to thosedescribed for other molluscs with modified sperm, except thatduring early spermiogenesis the Golgi body and smooth endoplasmicreticulum become highly developed. This proliferation of theSER and Golgi occurs at the same time as elongation of the spermatid.Throughout spermatogenesis, the germ cells are closely associatedwith a somatic cell which, because of structural similaritieswith the somatic cell of mammalian seminiferous epithelium,has been termed a Sertoli cell. After the spermatids have beenreleased from the Sertoli cells of the testis, maturation continuesin the hermaphrodite duct where the acrosome reaches its finalsize and glycogen accumulates in the glycogen compartment ofthe mid-piece. Summary

Sperm ultrastructure is described for the nudibranch gastropod Cadlinella ornatissima, type species of the genus Cadlinella (Thiele). Although C. ornatissima exhibits most of the sperm features characteristic of other Opisthobranchia and the Pulmonata (a small, rounded acrosomal vesicle, a complex, helical, mitochondrial derivative—partially paracrystalline, coarse fibres associated with the axoneme), it also possesses a number of previously undescribed and possibly unique features (a longitudinally inrolled acrosomal pedestal, an axial structure within the cavity of the acrosomal pedestal, an electron-dense collar at the anterior region of the acrosomal pedestal, the presence of crystalloid bodies within the glycogen helices of the mitochondrial derivative). To our knowledge this is the first report of crystalloid bodies in mature sperm of any mollusc. Collectively this evidence raises questions concerning the affinities and systematic position of Cadlinella within the Nudibranchia. The peculiar nature of the sperm differences, in comparison with other investigated nudibranchs, suggest that Cadlinella is not easily linked to either the Cadlinidae or Chromodorididae, and should be considered incertae sedis.  相似文献

Proper mitochondrial morphogenesis is crucial for successful development of motile sperm. It was known that recessive Drosophila melanogaster mutation emm caused anomalies in the formation of a mitochondrial derivative—nebenkern and led to male sterility. Here we identified primary mutation effect and showed that emm is required for the formation and maintenance of inner mitochondrial structure starting from early spermatocytes. Abnormal mitochondria structure affects subsequent cellular processes in spermatogenesis such as meiotic cytokinesis and spermatid elongation.  相似文献

Intracytoplasmic sperm injection (ICSI) has become the method of choice to treat human male infertility. One of the outstanding problems associated with this technique is our current lack of knowledge concerning the effect of sperm capacitation and motility upon the subsequent development of oocytes following ICSI. In the present study, we first examined the capacitation state of sperm exhibiting normal motility, along with sperm that had been activated, and examined the effect of reactive oxygen species (ROS) produced by these sperm types upon embryogenesis following bovine in vitro fertilization (IVF) and ICSI. Data showed that activated sperm reduced the chromosomal integrity of IVF/ICSI embryos at the blastocyst stage, while capacitated sperm produced ROS in capacitation media. Secondly, we treated sperm with carbonyl cyanide m-chlorophenyl hydrazine (CCCP), a chemical known to uncouple cell respiration within the mitochondria, and investigated the effect of this treatment upon blastocyst formation and chromosomal integrity at the blastocyst stage. Activated sperm in which the mitochondria had been treated with CCCP reduced levels of chromosomal aberration at the blastocyst stage following ICSI, by reducing mitochondrial activity in activated sperm. In conclusion, these findings suggest that capacitated sperm exhibiting activated motility induced chromosomal aberration during development to the blastocyst stage following ICSI. The injection of sperm exhibiting normal motility, or activated sperm in which mitochondrial activity had been reduced, improved the quality of ICSI-derived embryos. Therefore, the selection of sperm exhibiting progressive motility may not always be better for early embryo development and fetal growth following human ICSI, and that the use of a bovine model may contribute to a deeper understanding of sperm selection for human ICSI embryo development.  相似文献

The structure and maturation of the spermatozoa of Sciara coprophila   总被引:3,自引:0,他引:3  
The axial filament of Sciara coprophila does not conform to the usual 9 + 2 filament pattern but consists, rather, of as many as 76 pairs of filaments which decrease in number from the anterior to the posterior region of the sperm. It is first seen at the base of the head in the shape of an indented oval. The axial filament varies in configuration along the remaining length of the sperm as one whorl or two connected whorls of filament pairs. The other structures of the sperm revealed by the light and electron microscopes are a homogeneous, dense, spear-shaped nucleus, a row of spherical dense bodies in the middle piece enclosed by the axial filament and of unknown nature and function and a single mitochondrial derivative. The mitochondrial nebenkern derivative consists of a large electron transparent region bordered by cristae and a smaller paracrystalline region located adjacent to the axial filament. The derivative arises as paracrystalline material in a medial nuclear indentation. The electron transparent material is first seen at the anterior end of the middle piece. Unlike other known insect sperm, but reminiscent of sperm capacitation in mammals, sperm maturation is completed in the spermathecae of Sciara 7 to 9 hours after insemination. It consists of the acquisition of sperm motility and elimination of the electron transparent region of the mitochondrial nebenkern derivative. The electron microscope reveals in mature sperm that the axial filament doublets have changed configuration and consist of a single whorl which encloses the paracrystalline rod. The process by which the major portion of the nebenkern derivative is eliminated occurs in four identifiable stages. Since sperm maturation does not appear to be intrinsically controlled, factors in the spermathecal fluid may play a role in its completion.  相似文献

Comparative sperm ultrastructure within the molluscan nudibranch genus Halgerda (Discodorididae) was examined for the first time using transmission electron microscopy (TEM), based on 17 of the 35 known species. In addition, observations on two other discodorids are made to facilitate outgroup comparison with Halgerda, including one species of Discodoris (D. boholiensis) and Asteronotus cespitosus (currently accepted as the closest sister taxon to Halgerda). Comparison was also made with some genera of the Chromodorididae in view of sperm similarities. Spermatozoa of all species examined were of the complex, helical, elongate ( approximately 300-400 micro m) type characteristic of most heterobranch gastropods. These cells exhibit the following discrete regions (in anteroposterior sequence) : an acrosomal complex (composed of a rounded, membrane-bound vesicle and a column-like pedestal); a solid, helical nucleus; an elongate, helical midpiece (composed of an axoneme and associated nine coarse fibers, an enveloping mitochondrial derivative of matrix, and paracrystalline materials and glycogen helix); an annular complex; and a short glycogen piece. Of these regions, the midpiece is by far the longest, occupying over 90% of the total sperm length. Comparison with other members of the radula-bearing cryptobranch dorids reveals several sperm similarities to other genera in the clade, particularly those of other Discodorididae and also with the Chromodorididae. Comparison with previously studied genera reveals noteworthy sperm differences within the Discodorididae. The most notable differences are the internal structure of the acrosomal pedestal (long and homogeneous in Halgerda, Discodoris; short and homogeneous in Asteronotus; long and finely striated in Rostanga; oblong with angular electron-lucent striations in Jorunna) and the internal structure of the glycogen piece. The pronounced helical keels of most Halgerda and Discodoris nuclei contrast with the weakly helical nucleus of Asteronotus. Sperm features alone do not provide a means of defining the genus Halgerda or the family Discodorididae nor do they support the monophyletic status of the caryophyllidia-bearing dorids. Important sperm characters such as the acrosome, nucleus, and midpiece can often still be determined from specimens that have been initially fixed in formalin, then stored in ethanol for extended periods of time (i.e., museum material). Of all sperm features, the mitochondrial derivative of the midpiece is the most resistant to long-term fixation : the survival of acrosomal, nuclear, and axonemal components is variable, presumably a factor of prefixation autolysis, varied primary fixation times and temperatures, formalin quality, and duration of alcohol storage.  相似文献

Starch degradation inNicotiana tabacum cv. Samsun infected with PVY was studied with respect to the formation of Glc-6-P which serves as a starting substrate for glycolysis and pentosephosphate cycle. Enhanced activity of these two metabolic pathways at the time of maximal reproduction of the virus requires an increased content of this substrate which is probably produced mainly by the hexokinases. The results of this paper indicate that Gle-1-P, synthesized during starch degradation, is not the main intermediate enabling an enhanced activity of the pentosephosphate cycle, because despite a high starch content in the tissues only a decreased amylophosphorylase activity was found. However, an appreciable amount of Glc-6-P at the time of maximal reproduction of the virus results from enhanced phosphoglucomutase activity. As there is not enough Gle-1-P for an enhanced activity of this enzyme due to the lowered activity of amylophosphorylase and because its curve resembles activity curves of the hexokinases (published in the previous paper), this result could be explained by the presence of galactose and by the presence of an enzyme system which would be able to metabolize it to Gle-1-P.Additional index words: Nicotiana tabacum, respiration, glycolysis, pentosephosphate cycle.  相似文献

Gryon pennsylvanicum is a platygastrid hymenopteran that has lately received increasing attention in Europe due to its possible use in biological control of the conifer seed bug pest Leptoglossus occidentalis. Here the male reproductive system and the spermatogenesis of this species, along with those of Gryon muscaeformis, are examined ultrastructurally for the first time. The male genital system is formed by a pair of testes, each containing only one follicle, a pair of accessory glands and deferent ducts connected to a single ejaculatory duct. All the stages of spermatogenesis are described in detail. Characteristic features of the Gryon spp. sperm, which are 100 μm long, are the presence of a polygonal nucleus, only one mitochondrial derivative, the occurrence of the centriole adjunct and a typical insect 9 + 9 + 2 flagellar axoneme. The single derivative, however, results from a process in which one of the two mitochondria is lost during spermiogenesis. Unlike in other insects, two centrioles occur in spermatids as a consequence of the ameiotic parthenogenesis. These characteristics stand as a valuable tool for phylogenetic inferences. Furthermore this study suggests a useful strategy for laboratory mass rearing.  相似文献

During epididymal transit, redox remodeling protects mammalian spermatozoa, preparing them for survival in the subsequent journey to fertilization. However, molecular mechanisms of redox regulation in sperm development and maturation remain largely elusive. In this study, we report that thioredoxin-glutathione reductase (TXNRD3), a thioredoxin reductase family member particularly abundant in elongating spermatids at the site of mitochondrial sheath formation, regulates redox homeostasis to support male fertility. Using Txnrd3−/− mice, our biochemical, ultrastructural, and live cell imaging analyses revealed impairments in sperm morphology and motility under conditions of TXNRD3 deficiency. We find that mitochondria develop more defined cristae during capacitation in wildtype sperm. Furthermore, we show that absence of TXNRD3 alters thiol redox status in both the head and tail during sperm maturation and capacitation, resulting in defective mitochondrial ultrastructure and activity under capacitating conditions. These findings provide insights into molecular mechanisms of redox homeostasis and bioenergetics during sperm maturation, capacitation, and fertilization.  相似文献

A mouse t-complex-specific DNA probe was used to determine the ratio of t-carrying and (+)-carrying sperm in epididymal, vas deferens, and postejaculatory sperm cell populations from heterozygous (+t) mice with transmission ratios of greater than 95%. No detectable degeneration of (+)-carrying sperm was observed. In this respect, mouse t haplotypes differ from Drosophila melanogaster SD chromosomes. High transmission of t haplotypes must be a consequence of differential transport and/or differential sperm function during the fertilization process itself.  相似文献

The mature sperm of Ptinella aptera is a helically coiled, flagellate gamete ca. 1.4 mm long—twice the length of the beetle itself. The rod-like acrosome, comprising the anterior part of the sperm, is 0.4 μm thick but is expanded as a flange around the nucleus and the base of the tail, increasing the diameter of the sperm to 2 μm. The bulk of the tail consists of a pair of bodies with a characteristic ultrastructure of longitudinal tubules beneath a lamellar cortical layer. These bodies are probably homologous with the mitochondrial derivatives of other insect sperm. The axoneme is helically coiled and is flanked by a single accessory body. One of the ‘structured bodies’ is connected to the acrosome and the other to the accessory body. The sperm move actively in the female reproductive tract. The functional significance of this behaviour and of the evolution of the large gamete itself is discussed in relation to existing hypotheses.  相似文献

The elongated spermathecal duct of bumblebees has been studied in hibernating queens, queens shortly after hibernation, mature egg-laying queens and uninseminated queens captured during summer, and workers. Only rather small size differences are found when comparing spermathecae of queens and workers. Clear differences between bumblebee queens and workers are found when comparing the histochemistry of the spermathecal ducts. Adult queens, regardless of age and reproductive status have spermathecal ducts that contain PAS positive material, whereas workers do not. It is suggested that the polysaccharides in the spermathecal ducts of queens are necessary as a source of energy for the rapid activation of spermatozoa passing through the duct prior to oocyte fertilization. An ultrastructural investigation revealed the presence of high glycogen content in the cells lining the duct of queens. Assuming that sperm cells are kept in a rather inactive state in the reservoir, the carbohydrate (glycogen) probably serves as an energy source for the sperm. The comparatively increased spermathecal duct length of bumblebees may increase the retention time of sperm inside the lumen.  相似文献

The mitochondrial derivative of the sperm of the gastropod pulmonate Biomphalaria glabrata was studies to ascertain succinic dehydrogenase localization cytochemically. Two techniques were compared. One technique depends on a tetrazolium salt that yields an osmiophilic formazan upon reduction. The other technique is dependent on the reduction of copper ferricyanide. The effects of several electron transport inhibitors were studied. The reaction product observed in the matrix of the mitochondrial derivative using the former technique is sensitive to rotenone and is believed to be nicotinamide adenine dinucleotide-dependent. The reaction product observed in the intracristal spaces using the copper ferricyanide method is insensitive to rotenone and is believed to cytochemically demonstrate succinic dehydrogenase in this material.  相似文献

In Korscheltellus lupulinus (Lepidoptera : Hepialidae), both eusperm and parasperm can be found, whereas Micropterix calthella (Lepidoptera : Micropterigidae) produces only eusperm. The eupyrene sperm in K. lupulinus is 250 μm long. Spermatozoa found in K. lupulinus are very similar to those found in higher Lepidoptera, but the eusperm of M. calthella do not possess similar features. As in other Lepidoptera, the axoneme pattern of K. lupulinus is 9 + 9 + 2. Anteriorly, the axoneme is replaced by a fibrous stem similar to an acrosomal structure; posteriorly it becomes progressively uneven. The sperm of K. lupulinus has other original features. The anterior region of the sperm is rigid and contains the nucleus, which has a longitudinal groove adjacent to the axoneme. The 2 mitochondrial derivatives extend from the posterior end of the nucleus. There are no accessory bodies. Many peripheral cytoplasmic vesicles are present. A thin sheath surrounds the cell. It is thick in the parasperm and made of concentric or spiralling lamellae; it does not have any lacinate appendages.The axoneme of the sperm of M. calthella extends over the entire length of the cell and spirals in a helix around the nucleus. The nucleus is also almost as long as the cell. The 2 mitochondrial derivatives, flanked by accessory bodies, follow this spiralling. The axoneme has a 9 + 2 pattern. The plane, which contains the 2 central microtubules, is at an angle with the median plane of the nucleus.  相似文献

Mitochondria, important energy centers in the cell, also control sperm cell morphogenesis. Drosophila spermatids have a remarkably large mitochondrial formation called the nebenkern. Immediately following meiosis during sperm development, the mitochondria in the spermatid fuse together into two large aggregates which then wrap around one another to produce the spherical nebenkern: a giant mitochondrion about 6 micrometers in diameter. The fused mitochondria play an important role in sperm tail elongation by providing a structural platform to support the elongation of sperm cells. We have identified a novel testis-specific protein, Spermitin (Sprn), a protein with a Pleckstrin homology-like (PH) domain related to Ran-binding protein 1 at its C-terminus. Fluorescence microscopy showed that Sprn localizes at mitochondria in transfected Kc167 cells, and in the nebenkern throughout spermatid morphogenesis. The role of Sprn is unclear, as sprn mutant males are fertile, and have sperm tail length comparable to the wild-type.  相似文献

  • 1.1. Glycogen and galactogen contents of the albumen gland of the freshwater snail Lymnaea stagnalis were determined under different conditions, known to influence these polysaccharides viz egg laying, photoperiod and starvation.
  • 2.2. After oviposition, the galactogen content is restored within 32 hr, whereas glycogen remains constant during this period. Short-day photoperiods favour accumulation, long-day photoperiods induce depletion of glycogen. In contrast, the galactogen content is not affected by the photoperiod.
  • 3.3. Since glycogen and galactogen are present in the same cells of the albumen gland, the independent variation of these polysaccharides would imply the presence of separate intracellular regulation mechanisms.

Over the past several decades, research on the synthesis and organization of the cell wall polysaccharides of Aspergillus fumigatus has expanded our knowledge of this important fungal structure. Besides protecting the fungus from environmental stresses and maintaining structural integrity of the organism, the cell wall is also the primary site for interaction with host tissues during infection. Cell wall polysaccharides are important ligands for the recognition of fungi by the innate immune system and they can mediate potent immunomodulatory effects. The synthesis of cell wall polysaccharides is a complicated process that requires coordinated regulation of many biosynthetic and metabolic pathways. Continuous synthesis and remodeling of the polysaccharides of the cell wall is essential for the survival of the fungus during development, reproduction, colonization and invasion. As these polysaccharides are absent from the human host, these biosynthetic pathways are attractive targets for antifungal development. In this review, we present recent advances in our understanding of Aspergillus fumigatus cell wall polysaccharides, including the emerging role of cell wall polysaccharides in the host-pathogen interaction.  相似文献
