细胞的有丝分裂与细胞的增殖、分化及胚胎发育、组织器官形成、损伤组织的修复和疾病的发生有关.各种物理因素、细胞所处的微环境(包括细胞外基质、细胞粘附)等,以及胞内的多种信号因子均能对细胞的有丝分裂方向产生影响.大量文献表明,应力纤维的排列为有丝分裂中心粒分离和定位提供轨道,最终影响纺锤体和有丝分裂的定向.本实验室的micro-pattern和静态单轴拉伸应变实验进一步提示了应力纤维的排布方式是影响有丝分裂方向的重要因素.本文围绕着应力纤维的排布对有丝分裂方向的影响这一研究观点,综述分析了整合素介导的细胞外粘附-黏着斑的组装-应力纤维的排布-有丝分裂纺锤体定向等一系列影响贴壁哺乳动物细胞有丝分裂定向的过程.并根据酵母模型,对哺乳动物细胞有丝分裂定向过程的分子机制进行了介绍;在该过程中肌球蛋白、动力蛋白和kar9等蛋白质起到重要作用.  相似文献

哺乳动物细胞的有丝分裂过程与细胞的增殖、分化以及生物体发育、组织器官形成、损伤组织的修复和疾病的发生有关.广泛存在的力学刺激能否对细胞有丝分裂方向产生影响,以及其影响有丝分裂定向的途径尚未完全阐明.采用小鼠成纤维细胞作为模型,研究周期性单轴拉伸力学刺激对细胞应力纤维排布和有丝分裂方向的影响.结果表明,周期性单轴拉伸诱导细胞有丝分裂与应力纤维垂直于拉伸方向排布.而阻断应力纤维的两种基本组成成分(微丝和肌球蛋白Ⅱ),会造成在周期性单轴拉伸条件下的应力纤维和有丝分裂方向重排.特别是,Y27632 (10 μmol/L) 和低浓度的ML7 (50 μmol/L)、Blebbistatin (50 μmol/L)可以诱导细胞有丝分裂与应力纤维平行于拉伸方向排布.统计结果表明,在不同实验条件下,应力纤维排布和有丝分裂方向均具有高度相关性.Western blot实验表明,肌球蛋白轻链磷酸化水平与周期性单轴拉伸刺激下的应力纤维排 布和有丝分裂方向密切相关.上述结果提示:周期性单轴拉伸力学刺激通过诱导应力纤维的排布,决定了细胞的有丝分裂方向.  相似文献

研究香兰素衍生物中的6.溴异香兰素(BVAN08)对细胞纺锤体结构的影响及诱发灾变死亡的相关机制,为开发该化合物为新的抗癌药物提供理论依据.通过光学显微镜观察BvAN08作用后细胞形态学变化,流式细胞术检测细胞周期,纺锤体功能检测点实验和原位免疫荧光杂交实验分析细胞有丝分裂进程和纺锤体结构.western印记检测BVAN08作用后相关蛋白质的变化.结果表明20~60 μmol/L BVAN08作用后,HepG2细胞变圆不再贴壁生长、随后脱落死亡,具有浓度依赖性量效关系;明显诱导细胞G2/M期阻滞、导致细胞有丝分裂指数升高,并出现大量的非二倍体和多倍体细胞;破坏细胞纺锤体的结构,多中心体细胞显著增加;该化合物促使细胞周期转录调节因子FoxM1及其下游靶分子细胞周期蛋白B1和CdK1的降解、阻止有丝分裂过程而导致有丝分裂灾变死亡.研究揭示BVAN08通过破坏纺锤体结构、诱发M期阻滞,导致细胞有丝分裂灾变死亡,FoxM1失活可能参与其作用机制.  相似文献

力学刺激对细胞发育具有重要意义,它如何对细胞分化及组织形态的发生产生影响是一个尚未完全阐明的问题.细胞的有丝分裂过程与细胞增殖、分化以及胚胎发育、组织器官形态形成和损伤组织的修复再生等特性密切相关,例如,细胞的有丝分裂方向就是影响细胞极性分化,乃至组织形态发生的因素之一.那么,力学刺激是否通过改变细胞有丝分裂方向从而影响细胞的分裂分化呢?以小鼠成骨细胞系MC3T3为模型,探讨了静态单轴拉伸应变刺激对细胞形态、应力纤维排布方向和有丝分裂方向的影响.结果显示,在4%及8%静态单轴拉伸应变条件下,48 h之内细胞形态发生明显变化,细胞呈梭状,长轴沿应变方向排列,细胞骨架微丝呈束状平行排列,方向与应变方向相关.统计学分析表明,4%应变刺激48 h后、8%应变6 h后、8%应变12 b后、8%应变24 h后,及8%应变48 h后,分别有49%,43%,54%,54%,和62%的细胞应力纤维排列方向与单轴拉伸应变方向的夹角在300以内,以及50%,48%,56%,53%和62%的细胞有丝分裂方向与单轴应变方向夹角在300以内.统计学分析表明,细胞形态、应力纤维排布及有丝分裂方向与拉伸方向相关,且应力纤维排列方向和有丝分裂方向之间呈现高的相关性,这种相关性在拉伸刺激48 h后表现很明显,由此推测,存在力学刺激影响细胞形态及细胞应力纤维排布方向,控制有丝分裂方向的机制.  相似文献

用0.05%和0.1%秋水仙素溶液对洋葱Allium cepa根尖进行控时处理,研究不同秋水仙素浓度和不同处理时间对洋葱根尖细胞有丝分裂畸变的影响。结果显示,秋水仙素有促进细胞有丝分裂染色体停滞在中期的能力;不同固定时间对洋葱根尖细胞有丝分裂有影响;秋水仙素对洋葱根尖细胞染色体有畸变效应,诱导产生畸变的能力受溶液浓度和处理时间的影响。  相似文献

细胞死亡是多细胞生物生命过程中重要的生理或病理现象,可分为坏死和程序性细胞死亡,而后者根据死亡细胞的形态学和发生机制的不同又可分为凋亡、自吞噬和mitotic catastrophe,其中mitotic catastrophe是近年来才被揭示报道,是指细胞在有丝分裂过程中死亡的现象,是一种发生在细胞有丝分裂期由于异常的细胞分裂而导致的细胞死亡,它常常伴随着细胞有丝分裂检查点的异常和基因或纺锤体结构的损伤而发生。现对mitotic catastrophe及相关的调控机制进行综述。  相似文献

二甲戊灵对离体大蒜染色体的加倍研究   总被引:2,自引:0,他引:2  
在MS分化培养基中分别添加不同浓度的秋水仙素和二甲戊灵,以 "改良蒜" 的蒜瓣基部为外植体诱导愈伤组织染色体变异并再生植株,观察根尖细胞染色体数和叶片下表皮保卫细胞特征检测诱变效果.结果表明:大蒜染色体数变异受诱变剂浓度和处理持续时间的影响,诱变剂高浓度和短时间处理均有利于大蒜愈伤组织的分化;添加0.1%秋水仙素处理5 d,愈伤组织存活率为80.7%,分化率为78.1%,四倍体的诱导率为4%;添加 100 μmol/L二甲戊灵处理5 d,愈伤组织存活率为100%,四倍体的诱导率为6%.对根尖细胞染色体和叶片下表皮气孔数目和大小观察显示,诱变株具有四倍体的基本特征.说明二甲戊灵能在离体条件下有效诱导大蒜产生四倍体,可替代秋水仙素对大蒜染色体的加倍作用.  相似文献

The entry of labelled calf-thymus histone, lysozyme, and poly-L-lysine into barley root tips was studied at concentrations which strongly inhibit root elongation. The macromolecules were suitably labelled and at these concentrations it was found, by autoradiography and fluorescence microscopy, that histone and lysozyme readily entered the roots and appeared to bind mainly to cell walls of the epidermis and cortex and to penetrate the cytoplasm occasionally. Except in cap cells, nuclei were rarely penetrated. Poly-L-lysine readily permeated cell walls and invaded cytoplasm and nuclei throughout the root tip. Some cells were damaged by contact with basic macromolecules, as evidenced by a change in appearance of protoplasts under phase contrast and by the inability of these same protoplasts to exclude labelled β-lactoglobulin. Such damage was restricted to cells in contact with the outer solution. Interior to the epidermis, development of many cells was inhibited without visible signs of damage. Evidence supports the conclusion that in the presence of polybasic polymers the integrity of cell membranes is altered, thereby allowing leakage of some cell constituents essential for normal development.  相似文献

用农杆菌介导法将嵌合基因GFP-mTn(mTn是微丝结合蛋白Talin的微丝结合域,可以显示活体细胞中微丝的结构)导入蓝猪耳.经激光共聚焦显微镜观察了转基因植株的各种不同组织中融合蛋白的表达和分布情况.在叶片的表皮细胞、保卫细胞、根部的皮层细胞中有融合蛋白的不同程度表达.但仅在保卫细胞中微丝标记状况良好,显示基因表达的组织特异性.经光诱导处于开放态的气孔的保卫细胞微丝呈网状结构,在细胞内无规则分布;经黑暗诱导处于关闭态的气孔保卫细胞中微丝束沿保卫细胞纵轴排列,呈卷曲状分布,并观察到螺旋和环状的微丝结构.在转基因植株的其他部位,例如茎表皮细胞、根毛细胞和花粉粒中,未检测到目的基因的表达.本研究获得的转基因植株为研究气孔运动过程中微丝动态变化提供了有用的材料.  相似文献

An investigation was made of the meristematic activity of the apical cell, its immediate derivatives (merophytes), and of other selected cell populations of the root of Equisetum scirpoides Michx. The plane of the first division of a derivative of the apical cell is radiallongitudinal, which provides evidence that merophytes immediately adjacent to the apical cell cannot be the ultimate root initials. The apical cell is as active mitotically in roots 20–40 mm long as it is in roots that are 0.25–1 mm in length. The mitotic activity of the apical cell and of other cell populations was determined from the mitotic index, and from determination of the durations of the cell cycle and of mitosis of the apical cell by using the colchicine method of metaphase accumulation. Microspectrophotometric measurements of DNA content indicated that there was no consistent increase in DNA (endopolyploidy) in the apical cell or in the other meristematic cells as roots increased in length. Conclusion: there is no evidence that the apical cell becomes quiescent or undergoes endopolyploidy as a root increases in length.  相似文献

Effects of white, blue, red and far-red lights on the elongationgrowth of intact primary roots in rice seedlings were investigated.White light inhibited elongation of root cells. Blue light inhibitedboth cell elongation and cell multiplication, but red lightinhibited only cell elongation. The effect of far-red lightwas almost the same as that of the red. The lights exerted;the same effects on the growth of primary roots irrespectiveof age of the seedlings. The inhibitory effects of the lightswere also observed when the root of the deshooted seedlingswas irradiated, but not when only the shoot of the seedlingswas irradiated. It was inferred that it is not the shoot butthe root which responds to the light in this phenomenon. DCMUhardly affected root growth at a concentration so high as toinhibit strongly photosynthesis in the shoot. 富集分离了除草剂二甲戊灵降解真菌,并研究了其降解特性,结果表明,真菌可以降解二甲戊灵,利用富集培养的方法从环境中分离到16株能降解二甲戊灵的真菌。其中10株真菌5d内对100mg·L^-1二甲戊灵的降解率大于60%,以其中3株生理耐受能力强、降解能力高的真菌为例,研究了外加碳源浓度、初始pH值、二甲戊灵浓度和培养温度对真菌生长量和降解能力的影响,此3株真菌经鉴定分别属于土生曲霉组(Aspergillus terreus)、长梗串孢霉属(Monilochaetes)和烟色曲霉组(Aspergillus furnigatus),在外加碳源浓度为0.5%~1.0%的范围内,真菌生长量和降解率达到最大;在中性培养液中,3株真菌的生长量大,降解能力强;在浓度为100mg·L^-1时降解率和生长量都比较大,而绝对去除量随二甲戊灵浓度的提高而增加,在500mg·L^-1时达到最大;真菌的生长和降解需要适宜的温度,20~30℃培养时,降解率和生长量最大,可为农药污染治理及生产污水处理提供理论依据。  相似文献

In order to quantify the ultrastructural changes associated with cellular differentiation, we have performed a morphometric analysis of the ultrastructure of the calyptrogen, columella, and peripheral cells of the root cap of Zea mays. The relative volumes of the nucleus, nucleolus, and mitochondria in the protoplasm gradually decrease as a cell moves through the root cap. The relative volume of plastids increases 240% during the differentiation of calyptrogen cells into columella cells. This increase is transient, however, since the relative volume of plastids as well as starch in plastids decreases markedly as columella cells differentiate into peripheral cells. Dictyosomes and spherosomes increase more gradually than plastids, peaking in relative volume in the innermost peripheral cells (PCI). The relative volume of the vacuome decreases as calyptrogen cells differentiate into columella cells, after which it increases during the differentiation of peripheral cells. By the time the outermost peripheral cells (PCIII) are sloughed from the cap, the relative volume of the vacuome has almost tripled. These results indicate that each cell type comprising the root cap of Zea mays is characterized by a distinctive ultrastructure. Furthermore, the ultrastructural changes associated with the differentiation of these cells are organelle specific. The results of this study are discussed relative to the function of the various cell types of the root cap.  相似文献

Plasmolytic studies utilizing a graded series of mannitol solutions (0.1–1.4 M in 0.1 M increments) were conducted on adventitious roots of Zea mays to determine solute concentrations of cell types at various locations in the root. Results indicated that mature sieve-tube members had the highest solute concentration as determined by their C50 (the estimated mannitol concentration plasmolyzing an average of 50% of a given cell type) of any cell type in the root. In tissue 12 cm from the tip, C50 values calculated for proto- and metaphloem sieve-tube members were 1.15 and 1.19 M, respectively, while in tissue 0.5 cm from the root tip, values for the same cell types were 0.68 and 0.46 M, respectively. The C50 values for sieve elements in tissue 5 cm from the tip were intermediate (1.08 and 1.11 M). Although the companion cells generally plasmolyzed at nearly the same concentrations of mannitol as the sieve elements, their C50 values were slightly lower than adjacent mature sieve elements. The lowest C50 (0.35 M) for any cell type examined was associated with meristematic cells in tissue 0.1 cm from the root tip. Taken collectively, the results indicate that positive concentration gradients exist between mature sieve tubes and meristematic cells of the root apex of maize.  相似文献

Roots of Marsilea vestita ranging from 1–120 mm in length, as well as root primordia, were analyzed to determine mitotic activity and ploidy levels in the apical cell, five well-defined regions of the root proper, and two regions in the root cap. The mitotic index of the apical cell tended to be above the overall mean mitotic index for the entire apical meristem. No diurnal rhythm in mitotic index was apparent. The cell-cycle duration of the apical cell ranged from 12.1–25.2 hr, that of other regions of the root from 16.1–41.5 hr. There was no indication of polyploidy in any part of the apical meristem except in a few procambial cells. Thus, the results support the classical concept that the apical cell is the ultimate source of cells in the root.  相似文献
