The redox potential of the cell, as well as the antioxidant status of the tissue, are considered to be important regulatory constituents in an adaptive response in plants. Here the involvement of active antioxidants ascorbic acid (AA), reduced glutathione (GSH) and α - and β -tocopherols in reactive oxygen species scavenging, and the effect of anoxic stress on their reduction state were studied in 4 anoxia-tolerant and -intolerant plant species: Iris germanica L., Iris pseudacorus L., wheat ( Triticum aestivum L. cv. Leningradka) and rice ( Oryza sativa L. cv. VNIIR). The initial antioxidant content (both AA and GSH) was higher in the rhizomes of the more anoxia-tolerant Iris spp., as compared with that of the roots of the cereals. The predominant form of ascorbate was dehydroascorbic acid (DHA) in the cereals and AA in the Iris spp. Imposition of anoxia with subsequent reoxygenation resulted in an overall depletion of the reduced forms of antioxidants. No concurrent increase in oxidised forms (DHA and conjugated glutathione) was observed in anoxic samples. α -tocopherol content in Iris spp. was in the range 1–2 μg g−1 fresh weight, while β -tocopherol content was higher in the anoxia-intolerant I. germanica (7.2 μg g−1 fresh weight) as compared with the tolerant I. pseudacorus (1.5 μg g−1 fresh weight). In I. pseudacorus , a significant decrease in α - and β -tocopherol levels was observed only after long-term (45 days) anoxia. The results suggested exclusion of AA and GSH from the redox cycling under prolonged anoxia, and a concomitant decrease in the redox state, as well as an anoxia-induced depletion of α - and β -tocopherols.  相似文献

Coniferyl and sinapyl alcohols were isolated, identified and quantitatively determined as unbound (or weakly bound) phenylpropanoids in neutral hot-water extracts of Nicotiana tabacum L. callus tissue. This is the first identification of these alcohols in cultured tobacco callus. Scopoletin was also detected in these extracts, and it was the most abundant of these three phenylpropanoids with concentrations that ranged from 50–119 μg/g dry wt. Coniferyl alcohol (17–34 μg/g dry wt.) and sinapyl alcohol (23–35 μg/g dry wt.) were present in nearly equimolar concentration ratios and at levels which were about half those determined for unbound (or weakly bound) scopoletin. The amount of scopoletin extracted increased about 10 times when 1 M HCl-50% methanol - 0.3% ascorbic acid was used as the extractant. This indicated that most scopoletin moieties were strongly bound, perhaps by acid-hydrolyzable linkage. Coniferyl alcohol and sinapyl alcohol were not found in the acid extracts, presumably because they were acid-labile. In general, the concentration of each endogenous unbound (or weakly bound) phenylpropanoid appeared to remain relatively constant throughout the growth phase of the subculture. The only exceptions to this were the relatively higher concentrations of scopoletin and coniferyl alcohol present during the initial 0–2 weeks of subculture.  相似文献

Aromatic L-amino acid decarboxylase activity was measured in brain, heart, intestine, kidney, liver, muscle, pyloric caeca, spleen and stomach of skipjack, using L-3,4–dihydroxyphenylalanine as the substrate. Aromatic L-amino acid decarboxylase activity was found to be present in all of the organs studied. The highest activity was found in the intestine (1774 μmol min −1 g−1 wet wt of tissue). The influence of dietary copper, iron, and ascorbic acid on iron utilization was examined in a 2×2×2 factorial experiment. Male Sprague-Dawley weanling rats were fed copper-deficient (Cu-, 0.42 μg Cu/g) or copper-adequate (Cu+, 5.74 μg Cu/g) diets that contained one of two levels of iron (38 or 191μg Fe/g) and ascorbic acid (0 or 1% of the diet). These eight diets were fed for 20 d, and rats received an oral dose of 4 μCi iron-59 on d 15. Compared to Cu+ rats, the Cu− rats had 27% lower hemoglobin levels with 45, 59, and 65% lower cytochrome c oxidase (CCO) activities in the liver, heart, and bone marrow, respectively (p<0.0001). High dietary iron or ascorbic acid did not alter hemoglobin in Cu+ rats. However, hemoglobin was 23% lower in Cu− rats fed the highest, rather than the lowest levels of iron and ascorbic acid. Liver CCO was decreased (p<0.02) in Cu− rats fed high iron. Among Cu− rats, ascorbic acid did not influence CCO but decreased hemoglobin by 17% (p<0.001), reduced the percentage of absorbed iron-59 in the erythrocytes by 91% (p<0.05) and depressed the percentage apparent absorption of iron (p<0.05). These results suggest that the effects of elevated dietary iron and ascorbic acid on iron utilization are influenced by copper status.  相似文献

Tissue Choline Studied Using a Simple Chemical Assay   总被引:1,自引:1,他引:0  
Abstract: An enzymatic-radioisotopic assay was used to measure free choline in unextracted tissue. The lowest concentration of free choline in any tissue studied was present in human cerebrospinal fluid (mean, 5.7 μM; range, 1.8–31.2 μM). A postmortem increase in concentration of free choline occurred in blood (O.2 nmol/min ml), kidney (13 nmol/min·g), and liver (22 nmol/min·g) of mice. The concentration of free choline in these tissues was estimated by extrapolation to be 5, 77, and 29 nmol/g (or ml), respectively. Several treatments were found to increase the concentration of free choline. For example, intraperitoneal administration of choline or 2-amino-2-methyl-propanol (a choline oxidase inhibitor) induced an increase in the level of choline in blood, kidneys, liver, and brain of mice, and administration of 2-dimethylaminoethanol (deanol) caused an increase in kidney and liver choline. The level of choline in blood was increased when rats were treated orally with either antibiotics or esters of choline such as phosphorylcholine, glycerylphos-phorylcholine, laroylcholine, or propionylcholine. The results show that the concentration of free choline may be regulated by intestinal metabolism, availability of esterified precursors, and activity of enzymes that metabolize choline.  相似文献

Yearling brown trout, Salmo trutta L., were exposed to various concentrations of inorganic aluminium (0–3.7 μM1−1) over a pH range of 4.3–6.5 in a flow-through bioassay apparatus using synthetic test media. Low pH, in the absence of aluminium, produced little effect on growth or survival except at the lowest pH tested (4.3). At pH less than 5.5, concentrations of total aluminium in excess of 1 μM 1−1 (27μg 1−1) were found to retard growth. The effects of a given aluminium concentration were markedly reduced at pH above 5.5.
The change in aluminium toxicity with pH must be related to changes in aluminium chemistry. When growth rates are correlated with the different aluminium species, calculated using thermodynamic equilibrium constants given in the literature, it appears that the Al(OH)2 + species is the most toxic, with a small contribution also coming from polymeric complexes.  相似文献

Mercury concentrations in the axial muscle tissue of most (243) of the 254 Anguilla australis and most (20) of the 27 A. reinhardtii collected from 30 sites in coastal rivers and lakes in Victoria, Australia, during 1975–78 were well below the Australian statutory health limit (0.5 μg g−1 wet weight). For A. australis the mean mercury concentration was 0.17 μg g−1 (±0.16 s.d. , range 0.01–1.60 μg g−1); for A. reinhardtii the values were 0.37 ± 0.23 μg g−1 (range 0.12–1.10 μg g−1). Statistical analyses showed that variation in mercury concentration due to total length accounted for only 13% of the total variation in A. australis and 2% in A. reinhardtii whereas locality accounted for 54 and 68%, respectively. Both species are thus considered suitable as indicators of mercury pollution.  相似文献

HAWKSWORTH, D. L. & McMANUS, P. M., 1989. Lichen recolonization in London under conditions of rapidly falling sulphur dioxide levels, and the concept of zone skipping. The lichen flora on trees and wood at 50 sites in north-west London was examined in 1988 to determine what changes had taken place since 1980, a period when mean winter SO2 levels had fallen from around 130 μg m-3 to within the range 29–55 μg m-3 in the study area. Forty-nine species were found. The fruticose lichens Evernia prunaslri and Ramalina farinacea now occur in many central London parks, and the yellow green foliose Parmelia caperata was discovered at 12 sites (previously only at one, Ruislip, in 1980). Comparable numbers and growth forms of species occurred throughout the area. An assemblage of zone 4–5 species (Hawksworth & Rose, 1970 Nature, 227: 145–148) tolerant of mean winter SO2 levels of 60–70 μg m-3 failed to penetrate central London, although more sensitive species of zones 6 and 7 had; this phenomenon, which has not previously been reported, is referred to as 'zone skipping' and is attributed to the speed at which mean sulphur dioxide levels in London have fallen. Species returning are those to be expected on the basis of previously-published field correlations with mean winter SO2 levels. Twenty-five species not seen within 16 km of the centre of London on trees or wood during this century were found; of these, eight had not been seen during the last 200 years.  相似文献

Heavy metals were assessed in four species of sea turtles from the Baja California Peninsula, Mexico, representing the first report of heavy metal concentrations in tissues of post-yearling sea turtles from the Eastern Pacific. Concentrations of Cd measured in C. mydas kidney (653 μg/g dry wt) were the highest ever reported for any sea turtle species. Cd accumulated preferentially in kidney and the ratios of kidney to liver Cd in Baja California turtles were among the highest reported for sea turtles globally. Zn, Ni, and Mn concentrations were also significantly higher in kidney than other tissues, while Cu and Fe were greatest in liver, and all metals were lowest in muscle. With the exception of one value (69.9 μg/g in kidney of C. caretta), Pb was low in all tissues from Baja California. In comparisons across species, kidney of C. mydas had greater Zn and Ni concentrations as compared to other species, although there was no difference in liver metal levels among the species. Positive correlations were detected in the concentrations of Cd, Cu and Ni with the straight carapace length of C. caretta.  相似文献

The sediments and various organisms in Lake Päijänne were examined for contaminants. The average mercury content of water plants was 9, of plankton 14, of sediment 114, of zoobenthic predators 83, of fish 332–1510 and of birds 240–13685 μg kg−1 (wet weight). The average PCB content of plants was 3, of plankton 21, of the zoobenthos 44, of fish 36-117 and of birds 219–13490 μg kg−1. The average ΔDDT content of plants was 0.5, of plankton 6, of the zoobenthos 14, of fish 7–42 and of birds 144-8262 μg kg−1. Regional differences in mercury content were most pronounced in sediment and fish. PCB concentration was highest near a town. ΔDDT was quite evenly distributed. Water plant species did not differ from each other, nor did the plankton fractions. The zoobenthic predators contained more chlorinated hydrocarbons than did the herbivores. There were clear differences between most species of fish and the chlorinated hydrocarbon content was highest in vendace. In adult birds levels of all residues were significantly higher than in juveniles.
In most cases PCB content was positively correlated with ΔDDT and in birds PCB, ΔDDT and mercury levels were correlated. DDT residues occurred mostly as DDE, but in vendace the proportion of DDT was high. At most trophic levels, ΔDDT/PCB was 0.15-0.40 but in birds it reached 1–2.  相似文献

The distribution of selenium in a marine wader, the Oystercatcher (Haematopus ostralegus) is given by the levels in 15 tissues and plasma. Red blood cells (RBC) contain the highest level (23 mg/kg dry wt) followed by liver, lung, and kidney (17–19 mg/kg). Most other tissues range from 3–10 mg/kg. The average kidney and liver concentrations of the Oystercatcher belong to the concentrations characteristic in birds. However, the Oystercatcher's tissue selenium concentrations are in general four-to fivefold mammalian levels, but in liver and lung, 11- to 13-fold and in the RBC, 12- to 33-fold. The selenium plasma and RBC levels of the Oystercatcher vary during the year from 280 to 410 μg/L and 13 to 30 mg/kg dry wt, respectively; the plasma concentrations are positively correlated with the RBC selenium concentrations. An overview of literature data shows that the selenium kidney and liver concentrations of birds do not vary with geographical latitude and size (length) of the birds. In species of the ordersCharadriiformes andProcellariiformes, high selenium kidney, and to a lesser extent liver, concentrations may occur. A function of selenium in antioxidation is suggested.  相似文献

The hypothesis that vitamin C interacts with vitamin E in vivo was investigated in juvenile lake sturgeon. Ten-month old lake sturgeon were fed diets supplemented with either 0 or 1250 mg ascorbic acid/kg diet concomitantly with either 0 or 200 mg α tocopherol/kg diet for 7 weeks at 17°C. Dietary vitamin C supplement resulted in significant increases of ascorbate concentrations in the posterior kidney and liver of sturgeon. Dietary vitamin E omission affected liver concentrations of α-tocopherol (10.0 ± 4.5 μg/g) in comparison to sturgeon fed a diet supplemented with vitamin E and vitamin C (99.5 ± 22.9 μg/g). Dietary vitamin C supplement decreased liver α-tocopherol concentration in vitamin E-deprived sturgeon. Also, vitamin E supplement lowered posterior kidney and liver ascorbic acid concentrations in vitamin C-deprived sturgeon. Gulonolactone oxidase and dehydroascorbic acid reductase activities were stimulated in groups fed vitamin C. Thiobarbituric acid-reactive substances concentrations (an indicator of lipid peroxidation) were higher in sturgeon fed either of vitamins as compared to sturgeon deprived of both vitamins. The results suggested that large doses of vitamins C and E may be prooxidant in vivo.  相似文献

The effects of light and temperature on cell size and cellular composition (chlorophyll, protein, carbohydrate) of two freshwater cryptophytes were studied with batch cultures. Neither of the species had a constant cell size but the size varied with growth conditions. At each temperature the smallest cells were recorded at the lowest experimental photon flux density. The smallest cells of Cryptomonas 979/67 had an average volume of 232 μm3 and the largest ones 1 020 μm3. In Cryptomonas 979/62 the smallest and largest cells measured 4 306 μm3 and 12 450 μm3. Both species increased their cellular chlorophyll content when PFB dropped below 110–120 μmol m-2 s-1. The highest and lowest chlorophyll contents of 979/67 were 7.45 fg μm-3 and 0.55 fg μm-2 respectively. For 979/62 the corresponding values were 10.23 fg μm-3 and 0.93 fg μm-3. In both species the protein content remained stable at PFDs higher than 110–120 μmol m-2 S-1. The highest content of protein measured in 979/67 was 638 fg μm-3 and the lowest 147 fg μm-3. For 979/62 these values were 1 036 fg μm-3 and 148 fg μm-3 respectively. The carbohydrate results were less clear and no pattern either in response to photon flux density or temperature was obvious. The lowest and highest contents recorded for 979/67 were 62 fg μm-3 and 409 fg μ-3 and for 979162, 36 fg μm-3 and 329 fg μm-3  相似文献

Comparison of epilithic and epixylic biofilm development in a boreal river   总被引:4,自引:0,他引:4  
SUMMARY. 1. We assessed substratum effects on lotic biofilm development by placing glass and white pine sampling units in a fourth-order boreal river, and analysing, at 6-week intervals, upper-surface biofilms for ATP, chlorophyll, ergosterol, and the activities of nine exoenzymes.
2. All parameters, except chlorophyll standing stock (range 80–320 μg dm−2) and β-xylosidase activity (range 0.4–4.8 μmol h−1 dm−2), were significantly greater for epixylic biofilms than for epilithic ones, but the magnitude of the increases varied from 2 to 5 fold, showing that, even under similar hydrodynamic conditions, epilithic and epixylic biofilms are structurally and functionally distinct. For example, ergosterol concentrations ranged from undetectable to 0.93 μg dm−2 for epilithon and from 11–49 μg dm−2 for epixylon; corresponding ranges for ATP were 1.6–3.7 (epilithon) and 4.2–7.7 μg dm−2 (epixylon), for acid phosphatase activity: 2.3–4.9 and 20–41 μmolh−1dm−2, and for alkaline phosphatase activity: 1.9–8.1 and 29–150 μmol h−1dm−2, respectively.
3. The more extensive epixylic development was attributed to utilization of the wood substratum as a supplemental carbon source and to a higher density of microbial attachment sites.  相似文献

Abstract: Bioactive brain-derived neurotrophic factor (BDNF) and neurotrophin-3 were produced using the baculovirus expression system and purified to homogeneity using ion-exchange and reversed-phase chromatography. Yields of purified neurotrophin-3 (300–500 μg/L) were similar to levels reported for baculovirus-expressed nerve growth factor (NGF), whereas initial yields of BDNF were significantly lower (20–50 μg/L). Improved production of BDNF (150–200 μg/L) was achieved by expressing BDNF from a chimeric prepro-NGF/mature BDNF construct using the Trichoplusia ni insect cell line, Tn-5B1-4. Examination of the distribution of BDNF protein from both the nonchimeric prepro-BDNF and the chimeric prepro-NGF/mature BDNF viruses in Sf-21-and Tn-5B1-4-infected cells suggests a specific deficiency in the Tn-5B1-4 cells in processing the nonchimeric precursor. In addition, the vast majority of the BDNF protein at 2 days after infection was intracellular and insoluble. N-terminal amino acid sequencing of purified recombinant BDNF and neurotrophin-3 demonstrated that the insect cells processed their precursors to the correct N-terminus expected for the mature protein. Bioactivity was characterized in vitro on primary neuronal cultures from the CNS and PNS.  相似文献

Sphaerospora epinepheli n. sp. is described from grouper, Epinephelus malabaricus , in cage-cultured and wild fish collected from both coastal lines of southern Thailand. Subspherical to spherical spores and mono- or disporous pseudoplasmodia were observed in the lumen of kidney tubules. Pseudoplasmodia were round to elongate, size range 15.6–22.9 μm (length) × 8.4–21.6 μm (width). Spores were 7.8–10.0 μm (length) × 12.3–14.5 μm (thickness), and 7.0–9.5 μm (width) with two spherical polar capsules of equal size measuring 2.9–4.4 μm in diameter and containing polar filaments with six or seven windings. Two uninucleate sporoplasms showed iodine vacuoles. Blood stages, similar to C-blood protozoans observed from freshwater fish in Europe, were found from peripheral blood smears of grouper. Ultrastructural studies of blood stages showed a similar structure to unidentified mobile protozoans from the blood of carp. Electron dense bodies were observed in the cytoplasm of the primary cell blood stages. Infected proximal-tubular epithelial cells showed highly vacuolated cytoplasm and pycnotic nuclei.  相似文献

Samples from a natural population of pike (Esox lucius L.) from the River Danube were used in a 12-month study to determine seasonal variations in biochemical parameters of pike blood sera, hepatosomatic index (HSI) and gonadosomatic index (GSI). The ranges of enzyme activities for sample means were: aspartate aminotransferase (ASAT) 252.0–583.8 U 1−1, alanine aminotransferase (ALAT) 4.9–11 -2 U1 and alkaline phosphatase (AP) 39.5–91.8 U1−1. The ranges of other parameters analysed in serum were total protein 27.7–40.1 g 1 1, urea 0.57–l.52 mmol 1 1 and creatinine 21.2–118.6 μmol 1 1. The range of sample means for HSI and GSI were 1.28–4.16 and 0.07–20.2 respectively. Temperature ranged from 4.5 to 23.5°C. The activity of serum AP was positively correlated to water temperature in males only, while urea and creatinine showed a positive correlation to water temperature in individuals of both sexes. GSI was correlated significantly with HSI in females. Total protein reached the lowest values during the spawning period, while creatinine levels depended on both the sex and season.  相似文献

A tissue culture method was developed to investigate the production of cotton (Gossypium hirsutum L. cv. Texas Marker-1) fibers in vitro. Ovules were excised from 3, 5, 7 and 9 days preanthesis ovaries and placed on an agar-solidified, modified Murashige and Skoog medium containing 2.3 μ M kinetin and 0.45 μ M –2,4–dichlo-rophenoxyacetic acid or 2.3 μ M kinetin and 10.7 μ M naphthaleneacetic acid. Ovules formed fibers and callus tissue. Fibers formed in vitro were up to 10 mm long, 10–22 μ wide and the cell wall was 1–3 μ M thick. Callus tissue cells were subcultured for over 25 weeks and their degree of elongation was monitored. The ability of ovule-derived cells to direct expansion in a longitudinal direction diminished, while lateral expansion increased with time in culture.  相似文献

Abstract— A method is described for quantifying the GABA distribution in cat spinal cord at 200–500 μn resolution. Isolated spinal cord (L5–S1) was frozen and sectioned at about 150 μm thickness. The frozen tissue section was cut into 200 or 500 μm square blocks. The GABA content of each square tissue block was determined by enzymic micromethods and GABA distribution was mapped quantitatively. Average GABA concentrations were: 0·4 mmol/l. in white matter, 1·2 mmol/l. in ventral horn and 1·7 mmol/l. in dorsal horn. The highest concentrations of GABA (2–3 mmol/l.) were found in the dorsolateral part of dorsal horn. In order to destroy the interneurons of dorsal horn, the blood vessels supplying the dorsal horn of the lumbar enlargement were unilaterally cauterized. Seven to 30 days after operation, both the size of dorsal root potential and the GABA level in the dorsal horn were markedly reduced on the cauterized side. These results suggest that GABA is highly concentrated in the interneurons of dorsal horn and functions as a transmitter of presynaptic inhibition.  相似文献

Muscle, liver, and kidney tissues from 38 polar bears (Ursus maritimus) caught in the Scoresby Sound area, Central East Greenland, were analysed for zinc, cadmium, mercury and selenium. In general, cadmium concentrations were low in muscle, liver and kidney tissue, with geometric means (g.m.) of 0.022 (range: <0.015–0.085), 0.841 (range: 0.092–3.29) and 13.1 (range: 1.04–115) g Cd/g wet weight (ww) respectively. This finding can be explained by low cadmium levels in the blubber of ringed seals. The concentration of mercury in muscle tissue was low (g.m. 0.071; range: 0.039–0.193 g Hg/g ww), whereas concentrations in liver and kidney tissue were relatively high (liver: g.m. 7.87; range: 1.35–24.8 g Hg/g ww, and kidney: g.m. 15.2; range: 1.59–66.6 g Hg/g ww). Mercury and cadmium were positively correlated with age in liver and kidney. Zinc was positively correlated with age in kidney, and selenium was correlated with age in liver. Contrary to other marine mammals, polar bears had higher mercury levels in the kidneys than in the liver. In all three tissues polar bears had significantly lower cadmium levels than ringed seals from the same area. Mercury levels were likewise significantly lower in the muscle tissue of polar bears than in ringed seals, whereas levels in the liver and kidney were significantly higher. The previous geographic trend for cadmium and mercury found in Canadian polar bears could be extended to cover East Greenland as well. Hence cadmium levels were higher in Greenland than in Canada, while the opposite was the case for mercury. Greenland polar bears had higher mercury and cadmium contents in livers and kidneys than polar bears from Svalbard. The mercury levels in muscle and liver tissue from polar bears from East Greenland were twice as high as found in bears from western Alaska, but half the levels found in northern Alaska. Cadmium and zinc were partially correlated in kidney tissue, and this was found for mercury and selenium as well. Cadmium and zinc showed molar ratios close to unity with the highest concentrations occurring in kidney tissue, while the levels of zinc exceeded cadmium in muscle and liver tissue by up to several decades. Mercury and selenium showed molar ratios close to unity in liver and kidneys.  相似文献
