Summary Shoot regeneration from leaf discs and leaf mesophyll protoplasts of 11 genotypes of Lycopersicon esculentum (the cultivated tomato), were compared. In both regeneration procedures genotypic differences were observed between inbred lines, and also between F1 hybrids and their parental lines. In the tested hybrid genotypes no heterosis effect with respect to shoot regeneration capacity was observed. A correlation between shoot regeneration from leaf discs and from leaf mesophyll protoplasts was apparent in the tested genotypes. This suggests that using the described procedure, shoot regeneration from leaf discs can be usef for rapid pre-screening for regeneration capacity from protoplasts of tomato genotypes.  相似文献

The leaf disc agroinoculation system was applied to study tomato yellow leaf curl virus (TYLCV) replication in explants from susceptible and resistant tomato genotypes. This system was also evaluated as a potential selection tool in breeding programmes for TYLCV resistance. Leaf discs were incubated with a head-to-tail dimer of the TYLCV genome cloned into the Ti plasmid ofAgrobacterium tumefaciens. In leaf discs from susceptible cultivars (Lycopersicon esculentum) TYLCV single-stranded genomic DNA and its double-stranded DNA forms appeared within 2–5 days after inoculation. Whiteflies (Bemisia tabaci) efficiently transmitted the TYLCV disease to tomato test plants following acquisition feeding on agroinoculated tomato leaf discs. This indicates that infective viral particles have been produced and have reached the phloem cells of the explant where they can be acquired by the insects. Plants regenerated from agroinfected leaf discs of sensitive tomato cultivars exhibited disease symptoms and contained TYLCV DNA concentrations similar to those present in field-infected tomato plants, indicating that TYLCV can move out from the leaf disc into the regenerating plant. Leaf discs from accessions of the wild tomato species immune to whitefly-mediated inoculation,L. chilense LA1969 andL. hirsutum LA1777, did not support TYLCV DNA replication. Two diploid clones of self-incompatible Solanum chacoense Bitt. with androgenetic ability were tested for anther and leaf disc culture response together with eight of their reciprocal F1 hybrids. Large differences were found among genotypes in frequency of anther induction as well as in the phase of plant regeneration. Anthers harvested in June showed a significantly higher percentage of response (17.5%) at the induction phase than those collected in July (13.8%) or August (12.7%). The lowest induction frequency was observed in May (7.3%). By contrast, plant regeneration from induced anthers did not vary during this time. Genotypic differences were also observed in leaf disc response. The two parental clones and two of their hybrids failed to produced any shoots. Among the remaining genotypes, two had only sporadic occurrence of shoot formation, two gave an intermediate response (15% and 24% of their discs carried shoots), whereas the discs of the two remaining genotypes responded well to culture (68% and 77%). The genetic analysis performed on the reciprocal hybrids revealed that a positive significant correlation existed between anther induction and leaf disc response (Spearman's r=0.82; p=0.01). This suggests that, under our conditions, these two aspects of tissue culture might share a common system of genetic control. Estimates of broad sense heritabilities, for leaf disc culture, 83% were obtained and the number of effective factors involved in the control of tissue culture response, indicated a relatively simple genetic control. Finally, considering the potentialities opened by the use of RFLP analysis, it might be possible to find probes that are linked with genes involved in tissue culture competence.  相似文献

A modified Sehwarzbach jet spore trap was used to sample Phytophthora infestans populations in small crops of potato within polyethylene tunnels and in the field. Airborne sporangia were eaptured in the trap and deposited upon potato leaf discs, of a cultivar lacking race-specific resistance genes, contained within the trap. Virulence characters of isolates captured in the spore trap were determined by subculturing the isolates on leaf discs of a range of potato genotypes with race-specific resistance (R-genes), incubating them and then recording compatible and mcompatible reactions, reactions. For four isolates of P. infestans the levels of sporulation on the differential leaf discs were noted and although cultivar × isolate interactions were inconsistent, variable light and temperature conditions had no significant effect on the reactions of the cultivars.  相似文献

Leaf discs from Nicotiana tabacum L., N. glauca Grahm., and a series of interspecific periclinal chimeras were cultured on Murashige and Skoog (MS) medium containing either 2.5 mg/1 6-benzylaminopurine(BA) or 3.0 mg/16-furfurylaminopurine (kinetin). Most shoots regenerated from chimeral leaf discs were non-chimeral but 51 of 658 shoots were chimeral. The histogenic composition of plants regenerated from leaf discs of periclinal chimeras indicated that any cell layer in a leaf can be the ultimate source of shoots, and that shoots can be multicellular in origin. Certain periclinal arrangements were recovered more frequently and their chimeral nature was more stable during subsequent shoot growth. While N. tabacum leaf discs regenerated shoots on MS medium supplemented with either BA or with kinetin, N. glauca leaf discs did not form shoots on the medium containing kinetin. However, chimeras which possessed cells of both species arose on medium containing BA or kinetin, indicating that the morphogenetically competent (i.e., able to produce shoots in culture), N. tabacum cells either interacted with N. glauca cells leading to their competence or incorporated non-competent N. glauca cells into nascent shoot apical meristems.  相似文献

Changes in the level of endogenous formaldehyde (HCHO), some N-methylated compounds (choline and trigonelline) and peroxidase activity were examined in the leaves of bean genotypes (Phaseolus vulgaris L.) with different disease-sensitivity during ontogenesis in the stressfree condition and after natural infection by Pseudomonas savastanoi pv. phaseolicola (until the appearance of lesions). HCHO, as its dimedone adduct, and fully N-methylated compounds were determined by overpressured layer chromatography (OPLC) in different developmental stages and in the infected leaves/leaf discs. Peroxidase activity was measured by a spectrophotometric method. HCHO level decreased with ageing of the primary leaf and accordingly in the leaves at different developmental stages, then increased again in both cases due to the demethylation and methylation processes. Concentration of choline and trigonelline as potential HCHO generators decreased considerably while peroxidase activity increased with ageing of the plants. Comparing the symptomless and the Pseudomonas infected leaf discs (with watersoaked lesions) we found a decrease in the level of HCHO, choline and trigonelline and there was detectable increase in the peroxidase activity in the infected leaf tissues. Our findings are in accordance with previously published results that peroxidases play an important role in oxidative demethylation processes. Our hypothesis is that the high level of HCHO in the old leaves can originate from methylated components as the result of peroxidase activity and this high level may lead to the old leaf being resistant to pathogen. This conclusion is supported by the fact that the leaves of susceptible bean genotypes became resistant to Pseudomonas while growing older.  相似文献

Leaf discs from vegetative plants greatly increase their phenolic content when cultivated in vitro. Under long days the values remained constant, and were higher when compared with short days cultures. Under short days total phenolics decreased after 10 d, corresponding to the induction and expression of in vitro flowering. The effect of photoperiod and chlorogenic acid (0.01 mM) on leaf discs cultured from induced and non-induced plants, were analyzed regarding the neo-formation of roots, as well as vegetative and flower buds. Chlorogenic acid enhances the regeneration of roots in all treatments tested, with the highest stimulation on induced leaf discs cultivated in short days. The flowering was not affected by chlorogenic acid, but an inhibitory effect was observed on the neo-formation of vegetative buds in non-induced explants maintained in short days. Vegetative buds were reduced by 50% in flower-induced leaf discs cultivated under short days.  相似文献

We studied whether volatiles released by putative host plants affect the antipredator response of an herbivorous mite, Tetranychus urticae, when the patch was invaded by Phytoseiulus persimilis. Tetranychus urticae laid a lower number of eggs on tomato leaves than on lima bean leaves, suggesting that lima bean is a preferred host food source for T. urticae. In addition, T. urticae preferred lima bean plant volatiles to tomato plant volatiles in a Y-tube olfactometer test. To investigate the antipredator response of T. urticae, we examined the migration of T. urticae from a lima bean leaf disc to a neighbouring leaf disc (either a tomato or lima bean leaf disc) when ten predators were introduced into the original lima bean disc. A Parafilm bridge allowed for migration between the leaf discs. No migrations occurred between leaf discs when there were no predators introduced to the original leaf disc. However, when predators were introduced migrations did occur. When the neighbouring leaf disc was upwind of the original disc, the migration rate of the mite from original lima bean leaf disc to a neighbouring tomato leaf disc was significantly lower than that to a neighbouring lima bean leaf disc. By contrast, when the neighbouring leaf disc was downwind of the original leaf disc, there was no difference in the migration rates between lima bean leaf discs and tomato leaf discs. The number of T. urticae killed by P. persimilis for each treatment was not different, and this clearly shows that the danger was the same in all treatments regardless of the decision made by T. urticae. From these results, we conclude that T. urticae change their antipredator response by evaluating the difference in host plant volatiles in the patch they inhabit.  相似文献

Since the host-specific toxins of Alternaria alternata f. sp. lycopersici play an important role in pathogenesis, they potentially could be applied as selective agents in in vitro selection at the cellular level for disease resistance. Prerequisite for this is that sensitivity to the Alternaria alternata f.sp. lycopersici pathotoxins is manifest at the cellular level. To gain insight into cellular effects of AAL-toxins and into the mechanisms of plant insensitivity to AAL-toxins, effects of AAL-toxins on leaves, leaf discs, roots, calli, suspension cells, minicalli and protoplasts of susceptible and resistant tomato genotypes were studied. In leaves of susceptible genotypes, toxins cause severe necrosis, while in leaves of resistant genotypes necrosis was never observed. Inhibition effects of toxins were observed at all other levels in susceptible and resistant genotypes: toxins inhibited shoot induction on leaf discs, root growth and growth of calli, suspension cells and protoplasts. This indicates a cellular site for AAL-toxins. Differences in sensitivity to AAL-toxins between susceptible and resistant genotypes were observed in leaves and roots, but were not observed during shoot induction on leaf discs, in calli, suspension cells and protoplasts. However, differences in sensitivity to AAL-toxins in roots were at least 20 times less than in leaves. Therefore insensitivity seems related to a higher level of tomato plant differentiation and is most pronounced in leaves.  相似文献

The area covered by visible mycelium of E. cichoracearum on the upper surface of leaves 4, 8, 12 and 16 of tobacco plants in field plots in Rhodesia was expressed as percentages of the proximal and distal halves at weekly intervals. Free amino nitrogen and carbohydrate in discs from proximal and distal halves of the same leaves were analysed when each leaf was expanding rapidly and was not infected, and several weeks later, when the rate of expansion had slowed down and there was slight infection. On two other occasions, similar leaf discs were inoculated with conidia, to measure the percentage germination and hyphal length from individual conidia after incubation for 2–3 days at constant temperature and humidity; duplicate discs were chemically analysed. Leaves were not susceptible until at least 6 weeks after they had emerged from the bud. Soluble carbohydrate increased and free amino nitrogen decreased during the change from resistance to susceptibility. Proximal parts of leaves were usually infected first; they initially contained less amino nitrogen and soluble carbohydrate than distal parts. All parts of the leaf seemed to be equally susceptible later, when there were no differences in their amino nitrogen or soluble carbohydrate. Upper leaves of intact plants had more natural infection than those from corresponding leaves from topped plants. More conidia germinated on discs from them and produced longer hyphae. The discs from intact plants contained less free amino nitrogen and more soluble carbohydrate than those from topped plants. The accuracy of visual assessments of susceptibility was, generally, confirmed by measurements of percentage germination and length of hyphae from individual conidia on leaf discs. Regressions of hyphal length on leaf composition showed that susceptibility was apparently related inversely to free amino nitrogen and water content and directly to insoluble carbohydrate per unit dry matter.  相似文献

This study investigates behavioural responses of adult western flower thrips (Frankliniella occidentalis Pergande; Thysan., Thripidae) females to direct contact with repellent phenylpropanoid plant compounds (salicylaldehyde and methyl salicylate) applied on bean and cucumber leaves. The residence time of F. occidentalis females until take off was significantly shorter on bean or cucumber leaf discs treated with salicylaldehyde at 1% concentration compared with control leaf discs. A methyl salicylate (1%) treatment of cucumber resulted in shorter time periods until thrips took off the treated leaf discs compared with the control leaf discs. In a choice experiment thrips avoided to settle on a 1% salicylaldehyde treatment of bean and cucumber leaf discs for a maximum of 3 h, on a 1% methyl salicylate treatment for a 5‐h period. Within a 24‐h period neither the egg‐laying nor the feeding activity of F. occidentalis was affected after salicylaldehyde application (0.1%, 1%) on bean or cucumber. In contrast, methyl salicylate (1%) applied on bean and cucumber significantly prevented thrips females from oviposition and reduced the percentage of damaged area caused by their feeding activity for 24 h. As olfactory repellent plant volatiles applied on crop plants may elicit diverse post‐landing responses of F. occidentalis, short‐ and long‐term effects should be considered when evaluating the factual applicability of secondary plant compounds in a successful thrips management strategy.  相似文献

Leaf discs obtained from mature leaves of Xerosicyos danguyi were found to contain appreciable levels of stachyose throughout an 8-h nocturnal period during which this plant performs Crassulacean acid metabolism (CAM). In contrast, in mesophyll tissues obtained from paradermal sections of these same leaf discs and which were devoid of vascular tissues, stachyose pools were rapidly depleted during the nocturnal phase. The pattern of this depletion followed closely the depletion pattern observed for starch, indicating that mesophyll stachyose was possibly involved in nocturnal CAM processes and was not necessarily being used for export. Pulse-labelling of intact X. danguyi leaves prior to excision of leaf discs and mesophyll samples also indicated that, while labelled stachyose had turned-over completely in the mesophyll tissues by the end of the nocturnal period, substantial levels of labelled stachyose were always recovered from the leaf discs from which these mesophyll samples were derived. The data indicate the existence of two separate pools of stachyose in the X. danguyi leaf, one a mesophyll pool which turns over rapidly at night and which may be involved to a small extent in nocturnal CAM processes, and the other a pool associated with and possibly synthesized by the vascular tissues and which presumably represents the phloem-transport pool.Abbreviation CAM Crassulacean acid metabolism This work was supported by National Science Foundation Grant DCB 8901785.  相似文献

The ozone-sensitive tobacco variety Bel W3 was compared with the tolerant cv. Bel B using amphidiploid and amphihaploid genotypes of both. In search of the first genotypical differences, their reaction to acute ozone treatments was investigated with systems of decreasing degree of complexity: whole plants, grown under field, greenhouse and sterile conditions, excised tissues, calli, CCP, MCP and subcellular reactions. It was common to all systems that a fumigation, which clearly exceeds the threshold of the most tolerant material, led to equal reactions in all genotypes in respect of visible injury and membrane leaching. With whole plants and leaf discs growing conditions were found to influence the ozone threshold more than the genotype. Because the most resistant field-grown plants vary widely in their reaction, only sterile or greenhouse grown genotypes were compared. With the exception of whole sterile plants (no genotypical threshold differences), amphihaploids were more susceptible to ozone than their respective amphidiploids as to threshold and sensitivity spectra in all systems investigated. Higher ozone thresholds were detected for Bel B in all systems with one exception: MCP exhibited a lower threshold but also a lower degree of damage in the first buffer range of the sensitivity spectrum than those of Bel W3. Post-fumigation starch accumulation in mesophyll chloroplasts was the most prominent subcellular ozone reaction.  相似文献

Percentage germination, and growth of hyphae from single conidia of Erysiphe cichoracearum DC., were measured on leaf discs from topped and intact tobacco plants, grown in aerated nutrient solutions consisting of basal medium plus large or small amounts of potassium. The effect of supplying sodium was also studied. Discs were incubated on water and on 10% sucrose solution. Changes in free amino nitrogen and carbohydrate in comparable uninfected leaf discs, before and after incubation, were also measured. Potassium deficiency resulted in more free amino nitrogen and soluble carbohydrate and less insoluble carbohydrate, per cm.2 of leaf. Spore germination was not greatly affected by treatments, though it was usually less on discs from potassium-deficient leaves. The pathogen grew slower on potassium-deficient leaf discs, whether they were incubated on water or on sucrose. Incubating discs from some leaves on sucrose, compared with water, gave greatly increased sugar content and less fungal growth; discs from other leaves had a much smaller increase in sugar, and hyphal length was similar to that on discs incubated on water. Sodium, when potassium was scarce, increased potassium deficiency symptoms, free amino nitrogen and sugar content, and resistance to powdery mildew.  相似文献

A sterile culture nitrate of Penicillium expansum was shown to induce pisatin synthesis in pea leaf discs. The amount of pisatin produced by pea leaves was shown to decrease as they underwent senescence. N6-benzyladenine delayed senescence, and at the same time maintained the production of pisatin at a high level. In darkness, leaf discs maintained on either benzyl-adenine solution or distilled water produced greater amounts of pisatin than leaf discs which were not treated in this way. Benzyladenine also increased pisatin production by leaf discs kept in the light. The relevance of these results to disease resistance and possible mechanisms involved are discussed.  相似文献

Extracellular ice formation in winter oilseed rape leaf discs (Brassica napus L. var. oleifera L. cv. Jantar) at different temperatures resulted in a transient membrane depolarization, which was followed by a decrease in membrane electric potential. In discs which underwent supercooling (no extracellular ice was formed), no membrane depolarization was observed. The inhibitors of calcium ion channels, gadolinium and lanthanum, decreased to some extend the amplitude of the frost-induced (−6 °C) depolarization and completely eliminated the decrease in membrane potential. Changes in membrane potential were associated with the increased electrolyte efflux, measured after thawing of the discs. No efflux from supercooled discs was observed. Application of calcium channel blockers decreased the level of the efflux induced by freezing at −6°C. It is suggested that membrane depolarization is one of the primary events induced by ice formation at a leaf surface. The possible reasons for changes in the membrane electric potential and their physiological consequences are discussed.  相似文献

Photoinhibition has been often evaluated with leaf discs floated on water or placed on wet papers to prevent desiccation. Under these conditions, there is a possibility that CO2 diffusion is blocked by water, which may lead to reduction in photosynthetic CO2 assimilation. Using Chenopodium album L. grown at two irradiances, photosynthesis, quantum yield of Photosystem II (ΔF/F m′), non-photochemical quenching (qN), and photoinhibition were compared between detached leaves and leaf discs. In low-light-grown plants, photoinhibition was greater in leaf discs than in detached leaves, while in high-light-grown plants, there was little difference. Leaf discs showed lower rates of photosynthesis and ΔF/F m′, and higher qN. The ΔF/F m′ in leaf discs increased when leaf discs were exposed to high concentration of CO2, suggesting that CO2 diffusion to chloroplasts was limited in leaf discs floated on water. Tobacco (Nicotiana tabacum L., cv. Samsun) leaf discs inoculated with tobacco mosaic virus (TMV) were treated with auxin-like herbicides 2,4-dichlorophenoxyacetic acid (2,4-D), 2-methyl-4-chlorophenoxyacetic acid (MCPA), 3-amino-1,2,4-triazol (Amitrol) and 6-chloro-2-ethylamino-4-isopropylamino-1,3,5-triazine (Atrazin). All herbicides in the concentration of 10–7 M enhanced the virus content (MCPA to 227.4 %, Amitrol to 218.1 % and Atrazin to 257.3 % of values found in TMV-infected, herbicide untreated discs). The 2,4-D alone did not affect the activity of the glucose-6-phosphate dehydrogenase and ribonucleases, but the 2,4-D treatment together with TMV infection raised their activities twice as high as in the untreated control discs. Polyacrylamide gel electrophoresis of acidic extracellular proteins washed from leaf discs treated with 2,4-D did not prove the induction of PR-proteins.  相似文献

The influence of varied concentrations of sucrose and ammonical (NH4+) nitrogen on in vitro induction and expression of anthocyanin pigments from Rosa hybrida cv. ‘Pusa Ajay’ was investigated. Of two explants (petal and leaf discs) selected and cultured under two different conditions (light and dark), leaf discs were found to be most suitable for callus initiation. Profuse and early callus induction was observed when leaf discs of rose were cultured under total dark conditions on solid Murashige and Skoog (MS) medium supplemented with 4.0 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D). Early pigment initiation, enhancement and maximum anthocyanin production from calluses were recorded when leaf discs were cultured on Euphorbia millii (EM) medium supplemented with 7% sucrose compared with calluses cultured at 4% sucrose concentration under 16/8 h (light/dark) photoperiod regime. Reducing the concentration of NH4+ nitrogen in the solid MS medium led to slight improvement in anthocyanin production in rose leaf calluses.  相似文献

Leaf discs fromSaintpaulia ionantha xconfusa hybrids (African violet) were transferred between basal medium (BM) containing no hormones and shoot-inducing medium (SIM) containing 2.0 mg 1–1 indole acetic acid and 0.08 mg l–1 6-benzylaminopurine to determine whether there is a window of competence for shoot regeneration. Leaf discs precultured on BM prior to transfer to SIM formed buds 3 days earlier than the controls (leaf discs not precultured) regardless of whether the discs were placed upside down or right side up on the medium. This suggests that cultured leaf cells were not competent for shoot induction during the first 3 days of culture. Leaf discs cultured right side up (abaxial surface to the medium) did not form buds on BM alone, unlike discs cultured upside down. Leaf disc survival was affected by a delay in hormonal exposure, but surviving leaf discs produced as many shoots as control leaf discs. This suggests that in the absence of exogenous plant hormones, cellular competence to regenerate shoots is not lost in excised leaf discs of African violet.Abbreviations BM Basal medium - SIMSho Shoot-inducing medium  相似文献
