  • 1.1. The mechanism of action of glyburide (a sulfonylurea) on muscle has been investigated by measuring glucose uptake and glucose transporter (GLUT4) protein levels after chronic glyburide treatment.
  • 2.2. A dietary induced insulin resistant rat model (4 wk of high-fat, high-sucrose feeding) was given glyburide (2mg/kg/day) for 10 days and glucose uptake was measured in a perfused hindquarter preparation.
  • 3.3. Protein levels of the GLUT4 glucose transporter were determined by Western analysis.
  • 4.4. After 7 days of treatment, rats fed glyburide had lower blood glucose concentrations 2 hr (72 ± 5 vs 103 ± 12 mg/dl) and 24 hr (97 ± 7 vs 123 ± 7 mg/dl) after glyburide administration with no difference in serum insulin levels compared to vehicle treated animals.
  • 5.5. Glucose uptake was approx doubled in basal state (0 insulin) in response to glyburide (2.8 + 0.4 vs 1.7 ± 0.2μ mol/g per hr).
  • 6.6. Maximal insulin (100 nM) stimulated glucose uptake tended to be higher in the glyburide treated group, but did not reach statistical significance (8.0 ± 0.7 vs 7.0 ± 0.6 μmol/g per hr).
  • 7.7. Western analysis revealed no significant effect of glyburide on the GLUT4 protein level in skeletal muscle.
  • 8.8. These results suggest that glyburide alters glucose uptake through some mechanism other than alterations in the level of the GLUT4 glucose transporter protein.

  • 3.3. Accumulated 2-deoxyglucose-6-phosphate did not inhibit glucose transport.
  • 4.4. Unlike in adipocytes, basal and stimulated 2-deoxyglucose transport showed no difference in sensitivity to N-carbobenzoxy-glycyl-l-phenylalaninamide.
  • 5.5. Phenylarsine oxide blocked contraction-enhanced 2-deoxyglucose uptake.
  • 6.6. These results suggest that the glucose transporter of the sartorius exhibits auto-regulation, and that basal transport is not regulated by the same process as in adipocytes.

  • 1.1. Since glucose is one of the main energetic substrates for general metabolic processes in crustaceans, analysis of carbohydrate levels can furnish information on the energy metabolism of intact animals during osmoregulation.
  • 2.2. Different groups of Chasmagnathus granulata were transferred to different salinities (0 and 40%), and the glucose and glycogen concentrations in blood, gills, muscle and hepatopancreas were determined at the beginning of the experiment and 24, 72, 168 and 360 hr after the salinity changes.
  • 3.3. Differences in tissues carbohydrate levels were observed between summer and winter, that reflected differences in reserve mobilization.
  • 4.4. In the summer, hypo- and hyperosmotic shocks induced an increase in carbohydrate levels in almost all tissues studied, indicating gluconeogenesis.
  • 5.5. In the winter, a carbohydrate mobilization occurred only in the gills and hepatopancreas after both osmotic shocks.
  • 6.6. Thus, the substrate reserve used for energy production required for osmoregulation seems to be dependent on the season and tissues.

  • 1.1. Over an 8-year period, 19 biochemical parameters have been determined at various ages in the blood serum of 92 clinically healthy Lechwe waterbucks (Kobus leche), 33 males and 59 females.
  • 2.2. Significant differences have been noted with age. In neonates, the lowest values of total proteins, glucose, creatinine, urea, AST, ALT and iron have been noted; the highest ones have been seen for cholesterol, alkaline phosphatase, calcium and phosphorus.
  • 3.3. With regard to sex, raised values of glucose, urea, alkaline phosphatase and ALT, and lowered values of cholesterol, have been noted in juvenile females compared with males of the same age.
  • 4.4. In adult females, higher levels of urea and cholesterol and lower levels of glucose, triglycerides and natrium have been recorded compared with males.
  • 5.5. With sex and age, no significant changes have been found in the levels of GGT, magnesium, chlorides and copper.
  • 6.6. Out findings are discussed with those abstracted from the literature for related species.

  • 1.1. We studied the haemoglobin content, erythrocyte indices, erythrocyte enzymes and haemoglobin electrophoresis patterns of the metallic skink Niveoscineus metallicus and compared them to the small amount of published data on other small lizards.
  • 2.2. Haemoglobin was much lower than that recorded for the salamander.
  • 3.3. Erythrocyte enzymes (glucose phosphate isomerase and glucose 6 phosphate dehydrogenase) were lower in the skink than in the salamander. Glyceraldehyde phosphate dehydrogenase, phosphoglycerate kinase and pyruvate kinase were much higher in the skink than in the salamander.
  • 4.4. A single, slow, haemoglobin component was identified by electrophoresis.

  • 1.1. Lipid, glucose and glycogen concentrations were measured in different tissues of the crab Chasmagnathus granulata during emersion.
  • 2.2. After 6 hr of emersion no reduction in the total amount of carbohydrates was found to occur, suggesting that a general metabolic arrest was taking place.
  • 3.3. A transitory increase in haemolymphatic glucose and lipid levels was observed. Possible causes are therefore discussed in relation to changes in the flux of substrates for energy production.

  • 1.1. In intact (control) crabs (Carcinus maenas) and crayfish (Orconectes limosus) a significant (P < 0.01) increase in both glucose and lactate concentrations in the blood was observed after exposure to air. Such changes were not observed in either eyestalk-less or eyestalk-less and saline injected animals (P > 0.05).
  • 2.2. Injections of Crustacean hyperglycemie hormone (CHH) into eyestalk-less animals before exposure to air were able to reverse the effects of eyestalk ablation, i.e., significant increases (P < 0.01) in blood glucose and lactate could again be observed.
  • 3.3. Significant hyperglycemia (P < 0.01), but no changes in lactate concentration (P > 0.05), was observed after injection of CHH in eyestalk-less submerged animals.
  • 4.4. These results suggest that the increase in glycolysis after air exposure is facilitated by CHH, possibly by increased substrate availability due to glycogen degradation.

The rates at which the pulmonate snail B. glabrata takes up soluble glucose and maltose from a defined medium were evaluated by measuring the buccal mass pulsation rate, the drinking rate, net changes in the concentrations of sugars in the medium and the rate of accumulation of 14C labelled maltose.
  • 2.2. It was demonstrated that B. glabrata was capable of net accumulation of maltose and glucose via the mouth and integument, respectively.
  • 3.3. Some of the maltose in the medium was also hydrolysed to glucose by exogenous snail enzymes.
  • 4.4. The mechanisms involved in the accumulation of the sugars and the relevance of the results to the biochemical ecology of the snails and other aquatic invertebrates are discussed.

  • 2.The water content in the tissues studied displayed only minor changes as the animals passed from one phase to the other.
  • 3.A significant increase in Na was recorded in the brain during the different phases indicating a depressed sodium pump, whereas the blood Na level showed a significant decrease during hibernation.
  • 4.K increased in blood serum, brain and kidney during hibernation, while a nonsignificant decrease was found in blood serum during arousal. The brain may act as a potassium reservoir.
  • 5.An increase in Ca and Mg concentration was recorded in blood serum, brain and kidney during prehibernation and hibernation. The data suggested a homeostatic function in the transport and metabolism of these cations.
  • 6.Aldosterone exhibited a highly significant decrease especially during hibernation. The aldosterone regulation of ionic composition is discussed.
  • 7.Na/K and Ca/Mg ratios in the brain may explain the decreased excitability during winter torpor.

  • 1.1. Oxygen equilibrium curves were measured on trout red blood cell suspensions at pH 7.8 and 8.4 at 15, 20 and 25 C. Normal red cells and red cells that had been depleted of their ATP content were used.
  • 2.2. The equilibrium data were fitted to the Adair's model and the enthalpy (ΔH) and entropy (ΔS) changes for the first and fourth steps of oxygenation and for overall oxygenation were calculated from the temperature dependencies of the Adair constants.
  • 3.3. For normal red blood cells, the apparent heat for the first oxygenation step, δh1, is close to zero.
  • 4.4. Temperature insensitivity of this step at physiological pH, combined with a large pH dependence, probably denotes a property of Hb4, the Root effect Hb of trout blood.
  • 5.5. At pH 7.8, ΔH4 is about —4kcal/mol, a small value which may be attributed to the large release of Bohr protons that occurs at the last oxygenation step and corresponds to an endothermic process which opposes to the exothermic oxygenation of the haem.
  • 6.6. The ΔH4 value appears to have a large influence on the enthalpy for overall oxygenation.
  • 7.7. Results for ATP-free red cells are consistent with a mere increase in the intracellular pH and suggest that ATP has no specific effect at and above pHi ~ 7.7.
  • 8.8. Effects of temperature and pH on trout red blood cell isotherms emphasize the primary importance of the major component of trout blood, namely Hb4, in trout blood functional properties.
